ABSTRACT: Lettuce and arugula are the most cultivated leafy vegetables in hydroponic systems in the world, and the little information about Azospirillum brasilense effects in research in this system makes it necessary to verify the best method of inoculation of these bacteria. Therefore, this study aimed to verify the beneficial effects of inoculation methods of A. brasilense on plant growth and nutrition of lettuce and arugula grown in the hydroponic system. The study was conducted in a greenhouse with 30% shading. A randomized block design with five replicates in a 4 × 2 factorial scheme was used. Four A. brasilense inoculation methods (non-inoculated, foliar application, nutrient solution, and nutrient solution + foliar application) and two leafy vegetable species - arugula and lettuce - were evaluated. All inoculation methods improved plant height, shoot fresh and dry mass, root fresh and dry mass, fresh leaf yield, leaf chlorophyll index, accumulation of nitrogen, potassium, calcium, sulfur, phosphorus, iron, manganese, and copper and reduced nitrate accumulation in shoot and root of arugula and lettuce. Inoculation via nutrient solution + foliar application of A. brasilense is the most suitable for hydroponic arugula cultivation because it provides greater fresh leaf yield. All inoculation methods increased leaf yield in hydroponic lettuce. Hydroponic arugula and lettuce plants showed greater accumulation of nutrients and plant growth with less nitrate accumulation in the shoot under inoculation with A. brasilense.

RESUMO A alface e a rúcula são as hortaliças folhosas mais cultivadas em sistemas hidropônicos no mundo, e as poucas informações sobre os efeitos do Azospirillum brasilense em pesquisas neste sistema torna-se necessário verificar o melhor método de inoculação dessas bactérias. Portanto, o objetivo deste estudo foi verificar os efeitos benéficos de métodos de inoculação de A. brasilense em alface e rúcula no crescimento e nutrição das plantas em sistema hidropônico. O estudo foi conduzido em casa de vegetação com 30% de sombreamento. Foi usado o delineamento em blocos ao acaso com 5 repetições em esquema fatorial 4 × 2, com quatro métodos de inoculação de A. brasilense (não inoculado, foliar, solução nutritiva e solução nutritiva + inoculação foliar) e o segundo fator foi a aplicação em rúcula e alface. Todos os métodos de inoculação melhoraram a altura da planta, massa fresca e seca da parte aérea, massa fresca e seca da raiz, rendimento da planta, índice de clorofila foliar, acúmulo de nitrogênio, potássio, cálcio, enxofre, fósforo, ferro, manganês, cobre e redução do acúmulo de nitrato na parte aérea e raízes de rúcula e alface. A inoculação via solução nutriente + inoculação foliar de A. brasilense é a mais indicada para o cultivo hidropônico de rúcula, pois proporciona maior rendimento de folhas, e todos os métodos de inoculação foram capazes de aumentar o rendimento de folhas em alface hidropônica. Plantas hidropônicas de rúcula e alface apresentaram maior acúmulo de nutrientes e crescimento das plantas com menor acúmulo de nitrato na parte aérea sob inoculação com A. brasilense.

This work describes a green synthesis, the characterization, and biological evaluation of silver nanoparticles (AgNPs). The AgNPs suspension was synthesized using aqueous leaf extract of Eucalyptus grandis, which presented a characteristic band at 407 nm in the UV-Vis spectrum. The AgNPs presented a spherical shape and size of 9.7 ± 0.3 nm. The nanoparticles were stable over a month, indicating that E. grandis’ extract is suitable for their preparation and stabilization. The X-ray analysis showed that the crystallinity of AgNPs corresponded to the centered face phase of silver. The antibacterial and leishmanicidal activities of AgNPs were evaluated. The AgNPs presented antibacterial activity on the Gram-negative bacteria Escherichia coli at 53.9 µg mL-1. The leishmanicidal activity evaluation against promastigote forms of Leishmania infantum, Leishmania amazonensis, and Leishmania braziliensis showed that the biological response is dependent on the volume of AgNP suspension. It was demonstrated that L. infantum was more sensitive to the nanoparticle’s treatment than L. amazonensis and L. braziliensis. The treatment of L infantum promastigotes with 150 µL of AgNP suspension reduced parasite growth by 67.9%, a result which was similar to the treatment with 1 (66.7%) or 2 µL (70.6%) of amphotericin B used as a positive control.

Abstract This work describes the circulation over the continental slope and the São Paulo Plateau in the Santos Basin during the SANSED winter 2019 survey. The cruise consisted of four legs in the period between June, 11 and August, 03 2019. The observed circulation is dominated by the Atlantic southwestern boundary current regime and remotely-generated anticyclones and cyclones. The former is composed by the Brazil Current, the Intermediate Western Boundary Current and their mesoscale meanders; the latter are 300km vortical rings with origin in the eastern side of the South Atlantic Basin. A Lagrangian scheme applied over satellite altimeter maps indicate that the origin of these rings is primarily the Cape Basin of South Africa. The interaction between the boundary currents, their cyclonic meanders, and the anticyclonic rings is complex, and varies widely. During the SANSED winter 2019 survey period, three anticyclones interacted with the Brazil Current, instabilizing it, forming dipoles with the current cyclonic meanders, leading to their downstream propagation. Ancienter cyclonic eddies within Santos Basin may interfere with the propagation of the large anticyclones further south. In addition, the continuous arrival of remotely-originated anticyclones, the larger portion over the São Paulo Plateau presented a tendency of counter-clockwise circulation during the whole cruise period.

ABSTRACT Background: Antiretroviral therapy use has led to a decline in HIV-related mortality yet disparities by gender and/or sexual orientation may exist. In this study, we estimated hazards of death in people living with HIV (PLWH) according to gender and sexual orientation. Methods: We included PLWH ≥ 18 years enrolled between 2000 and 2018 at INI/Fiocruz, Rio de Janeiro, Brazil. Participants were grouped as cisgender or transgender women, cisgender men who have sex with men (MSM) or men who have sex with women, or cisgender men with unknown sexual orientation. We assessed disparities in the hazard of death using Cox proportional hazards models. Results: Among 5,576 PLWH, median age at enrollment was 35 years, 39% were MSM, 28% cisgender women, 23% men who have sex with women, 5% transgender women, and 5% men with unknown sexual orientation. A total of 795 deaths occurred in 39,141 person-years of follow-up. Mortality rates per 1,000 person-years were: 82.4 for men with unknown sexual orientation, 24.5 for men who have sex with women, 18.3 for cisgender, 16.6 for transgender women, and 15.1 for MSM. Compared to MSM, men with unknown sexual orientation had the highest death hazard ratio (adjusted hazard ratio [aHR] 2.93, 95% confidence interval [CI] 2.35–3.81), followed by men who have sex with women (aHR 1.17, 95%CI 0.96, 1.43); death hazard ratios for cisgender and transgender women were not statistically different. Conclusion: We observed disparities in the hazard of death for men with unknown sexual orientation and men who have sex with women despite universal access to antiretroviral therapy in Brazil. Future work should characterize and assist men with unknown sexual orientation with tailored policies and interventions. Increased hazard of death was not observed for transgender women, which probably results from interventions implemented in our service to reach, engage, retain, and support this population.

Abstract This study compares local ecological knowledge (LEK) of fishers from the Southwest Atlantic Ocean (SWAO), Brazil, related to the franciscana dolphin (Pontoporia blainvillei). We conducted 330 ethnographic interviews in ten fishing communities in southern and southeastern Brazil between 2012 and 2018. Boolean or Classic Logic was used to identify 95 fishers who were able to recognize the franciscana dolphin accordingly to the taxonomic entity P. blainvillei: 23 in northern Espírito Santo state, one in southern Espírito Santo, 20 in northern Rio de Janeiro state, and 51 in northern Paraná state. Among these 95 fishers, 87.4% (n = 83) reported incidental captures in fishing nets. Among these, 52 (54.7%) did not know any solution to this problem. Interviews revealed that the fishers usually discard carcasses in the sea after fat and muscle tissue are removed so that they can be used as bait for shark fishing or as food. In Southeastern Brazil, fishers LEK related to their ability to identify franciscana dolphin varied from ‘no identification’ and ‘extremely low identification’ to ‘partial’ and ‘good identification,’ while in southern Brazil, fishers mainly presented a ‘good identification’ of the dolphins. We propose comanagement actions to conserve the franciscana dolphin in the SWAO.

Abstract The Santos Basin (SB) is the main petroliferous basin in the Brazilian continental margin and one of the most studied marine areas in Brazil. However, historical data suggest that new efforts should be carried out to acquire quantitative biological data, especially in the deep sea, to establish the baseline of essential ocean variables in different ecosystems for future monitoring programs. The Brazilian energy company Petrobras planned and executed 24 oceanographic cruises over a period of 2 years to assess the benthic (SANSED cruise) and pelagic (SANAGU cruise) systems of the SB (356 days at sea in 2019 and 2021/2022). These efforts were part of the Santos Project, which comprised a comprehensive environmental study aimed at investigating benthic and pelagic variables to characterize ecology, biogeochemistry, thermohaline properties of water masses, and ocean circulation patterns, geomorphology, and sedimentology, as well as organic and inorganic chemistry. Here we present the detailed sampling designs and the field methods employed on board, during the SB scientific cruises. All sampling protocols were based on standardized approaches. For the benthos analyses, triplicate sediment samples were performed using a GOMEX-type box corer (0.25 m²) or a large modified Van Veen grab (0.75 m²) at 100 stations ranging from 25 to 2400 m depth. At each station, 25 geochemical and physico-chemical parameters were analyzed in addition to micro-, meio-, and macrofauna and living foraminifera samples. For the pelagic system, 60 stations were selected to investigate the plankton community, ranging in size from pico- to macroplankton, through vertical, horizontal, and oblique net hauls (20, 200, and 500 μm mesh size), as well as 25 biogeochemical parameters collected with an aid of a CTD-rosette sampler. Part of this scientific information also serves the Regional Environmental Characterization Project (PCR-BS) in support of Petrobras’ Santos Basin drilling licensing process led by the Brazilian Environmental Agency – IBAMA. This project contributes to the sustainable development of the SB, in line with the guidelines of the United Nations Decade of Ocean Science for Sustainable Development.

Abstract The presence of fungi in healthcare settings, including hemodialysis units, represents a significant risk for immunocompromised patients. This study aimed to investigate the occurrence of fungi in the air and water of a hemodialysis unit located in a tertiary public hospital in Maceió, Alagoas, Brazil. Over a period of three consecutive months, monthly air samples were collected and analyzed using the spontaneous sedimentation technique on Petri dishes containing Sabouraud Dextrose Agar (SDA). Simultaneously, water samples (100 mL) were collected from four specific water distribution points and subjected plating on SDA. Fungi were phenotypically identified based on their macroscopic and microscopic characteristics. In total, 498 colony-forming units (CFUs) of fungi were isolated, with 86 CFUs originating from the air and 412 CFUs from the water. Regarding the water samples, a higher concentration of fungal CFUs was observed in the potable water from the supply network (229 CFUs). Unexpectedly, 23 CFUs were identified in the reverse osmosis samples and 11 CFUs in the storage tank, which are post-treatment points where the presence of microorganisms is not desired. The fungus Cladosporium spp. was the most prevalent in both air and water samples, followed by Penicillium spp. in the air and Rhodotorula spp. in the water. These findings underscore the need to implement effective control and monitoring measures for fungi in the hemodialysis unit to ensure patient safety.

Resumo A presença de fungos em ambientes de saúde, incluindo unidades de hemodiálise, representa um risco significativo para pacientes imunocomprometidos. Este estudo teve como objetivo investigar a ocorrência de fungos presentes no ar e na água de uma unidade de hemodiálise localizada em um hospital público terciário na cidade de Maceió, Alagoas, Brasil. Ao longo de três meses consecutivos, amostras mensais de ar foram coletadas e analisadas por meio da técnica de sedimentação espontânea em placas de Petri contendo Ágar Sabouraud Dextrose (ASD). Paralelamente, amostras de água (100 mL) foram coletadas em quatro pontos específicos de distribuição de água e submetidas ao plaqueamento por esgotamento em meio ASD. Os fungos foram identificados fenotipicamente por meio de associação de suas características macroscópicas e microscópicas. No total, foram isoladas 498 unidades formadoras de colônias (UFC) de fungos, sendo 86 UFC provenientes do ar e 412 UFC da água. Em relação à água, observou-se uma maior concentração de UFC de fungos na água potável proveniente da rede de abastecimento (229 UFC). Inesperadamente, foram identificados 23 UFC nas amostras da osmose reversa e 11 UFC no reservatório de armazenamento, pontos de pós-tratamento nos quais a presença de microrganismos não é desejada. O fungo Cladosporium spp. foi o mais prevalente tanto no ar quanto na água, seguido por Penicillium spp. no ar e Rhodotorula spp. na água. Esses achados enfatizam a necessidade de implementar medidas eficazes de controle e monitoramento de fungos na unidade de hemodiálise, visando garantir a segurança dos pacientes.

Resumo Barragens são umas das principais ameaças para peixes potamódromos Neotropicais, causando bloqueios de rotas migratórias e perdas de áreas de reprodução em um número crescente de sistemas de drenagem. Neste cenário, dados atuais defendem que a manutenção de tributários de fluxo livre em bacias hidrográficas fragmentadas seria uma importante estratégia na conservação de peixes migradores, tanto em situações de panmixia quanto em cenários com diferentes estoques coexistindo, tal como já relatado para Prochilodus lineatus. Este estudo analisou locos microssatélites e haplótipos mitocondriais em amostras de P. lineatus obtidas em diferentes anos ao longo da bacia do rio das Cinzas, no reservatório de Capivara (maior reservatório na calha principal do rio Paranapanema, alto rio Paraná) e no rio Tibagi, todos conectados por trechos livres, visando investigar as contribuições da bacia do rio das Cinzas na dinâmica populacional de peixes migradores na região do reservatório de Capivara. Ambos os marcadores detectaram altos níveis de diversidade genética e indicaram diferentes estoques de P. lineatus ao longo da bacia do rio das Cinzas, sugerindo alguma estruturação genética temporal, bem como a mistura destes estoques na amostra do reservatório de Capivara e de outros pontos, corroborando as relações de estoques neste reservatório e seus tributários. Abstract Dams are one of the main threats to Neotropical potamodromous fish, causing blockages of migratory routes and loss of breeding areas in an increasing number of drainage systems. In this scenario, current data argue that the maintenance of free-flowing tributaries along fragmented watersheds would be an important strategy to the migratory fish conservation, both in panmixia situations and in scenarios with different coexisting stocks, as already reported for Prochilodus lineatus. This study analyzed microsatellite loci and mitochondrial haplotypes in samples of P. lineatus obtained in different years along the Cinzas River basin, as well as in the Capivara Reservoir (largest Reservoir in the Paranapanema main channel, upper Paraná River) and in the Tibagi River, all connected by free stretches, aiming to investigate the contributions of the Cinzas River basin to the population dynamics of migratory fish in the Capivara reservoir region. Both markers detected high genetic diversity levels and indicated different genetic stocks of P. lineatus along the Cinzas River basin, suggesting some temporal genetic structuring, as well as the mixture of these stocks in the sample from the Capivara reservoir and from other points, corroborating the relationships among stocks in this reservoir and its tributaries. systems scenario freeflowing flowing conservation P largest channel stretches region structuring points

The new coronavirus, called Severe Acute Respiratory Syndrome Coronavirus 2 (SARS-CoV-2), was discovered in late December 2019 after cases were reported in the city of Wuhan, China. In January 2020, the World Health Organization (WHO) officially declared the Coronavirus Disease 2019 (COVID-19) as a pandemic, which has an official record of around 500 million cases and more than 6.0 million deaths in worldwide. An important factor in controlling the pandemic is the development of more effective and efficient diagnostic tests. In this context, this review has as its main proposal to discuss the effective differences between the possible diagnostic tests, the implications of molecular and serological methods available on the market and the analytical and clinical parameters involved in the development and application of these methods.

Resumo O Questionário revisado de bullying de Olweus (rBVQ) é um instrumento internacionalmente utilizado para identificar bullying, mas devido às dificuldades de estudantes brasileiros na leitura e escrita, foi adaptado para uso áudio-guiado em smartphones. Investigou-se a validade do construto por meio de análise confirmatória fatorial em uma solução de dois fatores. Participaram 1742 e 2316 alunos de quintos e sétimos anos de trinta escolas públicas de São Paulo. Encontrou-se excelentes índices de adequação para ambos os anos escolares (quintos: CFI = 0,985; RMSEA = 0,020; sétimos: CFI = 0,990; RMSEA = 0,015) e cargas fatoriais maiores que 0,4 para todos os itens, indicando sua relevância para o construto. O rBVQ demonstrou excelentes propriedades psicométricas e pode ajudar estudos futuros que objetivam investigar bullying em populações com perfis semelhantes. Além disso, este estudo inova por avaliar um instrumento áudio-guiado e utilizando tecnologia móvel.

Abstract The revised Olweus Bully/Victim Questionnaire (rBVQ) is an internationally used instrument to identify bullying, but due to Brazilian students’ low proficiency in reading and writing, it had to be adapted for audio-guided use through smartphones. We investigated construct validity, using Confirmatory Factor Analysis (CFA), under a two-correlated factor solution. Participated 1,742 fifth graders and 2,316 seventh graders from 30 public schools in the city of São Paulo. We found excellent fit indices models for both grades (fifth grade: CFI=0.985, RMSEA=0.020; seventh grade: CFI=0.990, RMSEA=0.015), as well as factor loading higher than 0.4 for all items, indicating their relevance to the construct. The rBVQ demonstrated excellent psychometric properties and may help future studies that aim to investigate bullying in populations with similar profiles. In addition, this study innovates by evaluating an audio-guided instrument and using mobile technology.

Resumen El Cuestionario Revisado de Bullying de Olweus (rBVQ) es un instrumento utilizado internacionalmente para identificar bullying, pero debido a las dificultades de estudiantes brasileños en la lectura y la escritura, se ha adaptado su uso por medio de audioguías en smartphones. Se investigó la validez del constructo mediante el Análisis Factorial Confirmatorio, bajo un modelo de dos factores. Participaron 1742 y 2316 estudiantes de quinto y séptimo curso de 30 escuelas públicas de São Paulo. Excelentes tasas de adecuación fueron encontradas para ambos casos (quinto año: CFI=0,985, RMSEA=0,020; séptimo año: CFI=0,990, RMSEA=0,015), además de cargas factoriales superiores a 0,4 para todos los ítems, lo que indica su relevancia para el constructo. El instrumento rBVQ ha presentado excelentes propiedades psicométricas y puede ayudar a futuros estudios pretendan investigar el bullying en poblaciones con perfiles similares. Además, este estudio innova al evaluar un instrumento guiado por audio, empleando la tecnología móvil.

Malaria is responsible for thousands of deaths each year. Currently, artemisinin combination therapy (ACT) is used as first-choice medication against the disease. However, the emergence of resistant strains prompts the search for alternative compounds. The present study aimed to investigate the antiplasmodial activities of natural triterpenes (compounds 1 and 2), and semisynthetic derivatives 1a, 1b, 1c, and 1d. Antiplasmodial assays were carried out using the SYBR Green technique, whereas cytotoxicity was evaluated by the MTT (3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide) method. Hemolytic assays were performed on human erythrocytes. An in silico analysis of the compounds against PfENR (Plasmodium falciparum 2-trans-enoyl-reductase) was carried out by molecular docking. Experiments with 1, and its derivatives against P. falciparum showed that 1a was very similar in terms of biological activity to compound 1 (half maximal inhibitory concentration (IC50) ca. 4 µM), whereas 1b, 1c, and 1d had reduced antiplasmodial activities (IC50 between 8-103 µM). The selectivity indexes of 1 and 1d for HepG2, and Vero cells were > 10. Docking results partially agreed with the in vitro experiments, with 1 and 1c having the best and worst affinities with PfENR, respectively. In conclusion, the results showed that 1 and 1d may serve as biotechnological tools in the development of antimalarial drugs.

Considering the promising antitumor effects of compounds with dual anti-inflammatory and antiproliferative activities, thus benzophenones analogs (2-7) were evaluated on in vivo anti-inflammatory assay and molecular docking analysis. Those with the best molecular docking results were in vitro evaluated on cyclooxygenase (COX) enzymes and tested regarding antiproliferative activity. All derivatives displayed in vivo anti-inflammatory activity. Among them, the substances 2’-hydroxy-4’-benzoylphenyl-β-D-glucopyranoside (4), 4-hydroxy-4’-methoxybenzophenone (5) and 4’-(4’’-methoxybenzoyl)phenyl-β-D-glucopyranoside (7) showed the best values of Glide Score in COX-2 docking evaluation and 4 and 5 selectively inhibited COX-2 and COX-1 in vitro enzymatic assay, respectively. Thus, 4 and 5 were tested against breast cancer (MCF-7, MDA MB-231, Hs578T) and non-small-cell-lung cancer (A549) cell lines. The estrogen-positive MCF-7 cell line was more responsive compared to other tested cell lines. They induced cell cycle arrest at G1/S transition in MCF-7 cell line once there was an increase in G0/G1 population with concomitant reduction of S population. The antiproliferative activity of these substances on MCF-7 was associated with their ability to inhibit cyclin E expression, a critical regulator of G1/S transition. Taken together, the data indicate that 4 and 5 have dual anti-inflammatory and antiproliferative activities and support further studies to evaluate their antitumor potential.

Abstract Flow accelerated corrosion (FAC) is a steel degradation that occurs in heat recovery steam generators in the power industry. The mechanism of this corrosion comprises an electrochemical dissolution of the semi-protective magnetite layer (Fe3O4) that is formed within the pipes employed in the boilers. The FAC is influenced by different factors such as fluid velocity, pH and dissolved oxygen concentration. In this context, Rotating Cage tests were used to evaluate the influence of pH and dissolved oxygen content on FAC of A210, P11 and P22 steels. General corrosion was lower for P11 and P22 steels, an effect consistent with the presence of chromium in their compositions. General corrosion was most critical at pH 8.5. The corrosion intensity decreased for the three steels when the dissolved oxygen concentration increased, at 2 m/s. This behavior is coherent considering the most effective precipitation of Fe2O3 inside the pores of the Fe3O4 layer in a more oxidizing environment. For 3 m/s, the corrosion intensity increased for both materials when oxygen increased, showing the shear stress effect. These behaviors were more significant at pH 8.5. About pitting corrosion, the analyzes show localized attack in the majority of coupons, however with very low number of pits.

Abstract The present study aimed to evaluate the effect of alkaline treatments with urea, NaOH and Ca(OH)2 on chemical composition and in situ ruminal degradability of dry matter, crud protein and neutral detergent fiber of sugarcane tip hay. Samples were incubated in the rumen of three cannulated cattle for up to 72 hours in a split plot randomized block design. Ammoniation with 6% urea increased (p<0.05) the crude protein content by 13% and reduced the neutral detergent fiber and insoluble nitrogen content of the hay. When treated with the highest doses of the compounds, there was a high potential degradability of dry matter, crude protein and neutral detergent fiber, and a shorter neutral detergent fiber lag time. Ammoniation with urea promotes a reduction in the content of NDF, hemicellulose and insoluble nitrogen, with an increase in the content of CP in the hay, with emphasis for the level of 6% urea. The ruminal degradation of sugarcane tip hay increases with alkaline treatments using 6% urea or 3% NaOH, however, ammoniation with urea is indicated for the treatment of hay, as this is low cost and can be easily adopted by farmers in the semiarid region.

Resumo O interesse por espécies vegetais com ação antiviral tem crescido exponencialmente e algumas tem sido relatadas como possuídoras de propriedades anti-HIV. Essa pesquisa tem como objetivo realizar o fracionamento fitoquímico biodirecionado pela atividade antirretroviral das cascas do caule da espécie Lafoensia pacari. Trata-se de um estudo experimental in vitro e a metodologia envolve preparo do material vegetal, obtenção do extrato etanólico, fracionamento, purificação, identificação e quantificação das frações, extração ácido-base, ressonância magnética nuclear, teste de inibição da TR do HIV-1 e estudos de docking molecular. A partir do fracionamento biodirecionado pela atividade antirretroviral verificou-se uma maior atividade nas subfrações acetanólica. Com destaque para a subfração acetanólica neutros com 60,98% de inibição de TR e o ácido elágico com 88,61% de inibição de TR e ausência de citotoxicidade. Verificou-se com o teste de citotoxicidade em linhagem de macrófagos que a fração clorofórmica foi mais tóxica que a fração acetanólica. A análise do espectro J-resolvido na região aromática apresentou um simpleto em 7.48 e 6.93 ppm que foram identificados como ácido elágico e ácido gálico respectivamente. A enzima 5TIQ obteve melhor parâmetro de afinidade com o ligante ácido elágico que foi confirmado pelos espectros HSQC-1H-13C. O ácido gálico mostrou-se também favorável a formar interação com a enzima 5TIQ, sendo confirmado através do espectro HSQC-1H-13C. Através da avaliação do PreADMET verificou-se que o ácido elágico é uma molécula promissora pela sua atividade de inibição da TR e parâmetros farmacocinéticos e de toxicidade.

Abstract Interest in antiviral plant species has grown exponentially and some have been reported to have anti-HIV properties. This research aims to perform the bio-guided phytochemical fractionation by antiretroviral activity of Lafoensia pacari stem barks. This in vitro experimental study involved the preparation of plant material, obtention of ethanolic extract, fractionation, purification, identification and quantification of fractions, acid-base extraction, nuclear magnetic resonance, HIV-1 RT inhibition test and molecular docking studies. From the bio-guided fractionation by the antiretroviral activity there was a higher activity in the acetanolic subfractions, highlighting the acetate subfraction – neutrals with 60.98% of RT inhibition and ellagic acid with 88.61% of RT inhibition and absence of cytotoxicity. The macrophage lineage cytotoxicity assay showed that the chloroform fraction was more toxic than the acetate fraction. The analysis of the J-resolved spectrum in the aromatic region showed a singlet at 7.48 and 6.93 ppm which was identified as ellagic acid and gallic acid, respectively. The 5TIQ enzyme obtained better affinity parameter with the ellagic acid ligand, which was confirmed by the HSQC-1H-13C spectra. Gallic acid was also favorable to form interaction with the 5TIQ enzyme, being confirmed through the HSQC-1H-13C spectrum. From the PreADMET evaluation it was found that ellagic acid is a promising molecule for its RT inhibition activity and pharmacokinetic and toxicity parameters.
