ABSTRACT. The Discussion is the hardest section of a scientific article to write, as cognitive skills must be used to properly contextualize the findings of a study. In this article, we guide scientific writers, particularly unexperienced ones, on how to structure a Discussion section based on an article by Docherty and Smith (1999). According to these authors, a discussion should be prepared by organizing information in the following order: (a) statement of principal findings; (b) strengths and weaknesses of the study; (c) strengths and weaknesses in relation to other studies, discussing particularly any differences in results; (d) meaning of the study: possible mechanisms and implications; and (e) unanswered questions and future research. Each component of this sequence is discussed in detail with examples drawn from the literature.

Resumo: O objetivo deste trabalho foi avaliar genitores com potencial de hibridação para o pré-melhoramento do tomateiro (Solanum lycopersicum) quanto à resistência à requeima. Foram utilizados seis acessos de tomateiro (BGH-2102, BGH-2117, BGH-2127, BGH-2130, BGH-2332 e BGH-2343) como genitores resistentes e 15 híbridos F1 originários destes genitores. Utilizou-se o delineamento de blocos ao acaso, com três repetições. As plantas foram inoculadas com uma mistura de esporângios de Phytophthora infestans, agente etiológico da requeima, na concentração de 5x103 esporângios mL-1. A área abaixo da curva de progresso da doença foi utilizada para avaliar a resistência. Realizou-se a análise dialélica, tendo-se considerado o efeito de genótipos como fixo. Estimou-se a capacidade geral e específica de combinação dos acessos. O padrão de resistência dos genitores e da maioria dos F1 foi o mesmo que o das testemunhas resistentes. Foram observados: variabilidade genética aditiva entre os genitores, predominância de efeitos gênicos não aditivos e desvios de dominância bidirecional no controle do caráter. A frequência de alelos favoráveis e divergentes para resistência à requeima é maior nos acessos BGH-2117, BGH-2127 e BGH-2343.

Abstract: The objective of this work was to evaluate parents with hybridization potential for pre-breeding of tomato (Solanum lycopersicum) for resistance to late blight. Six tomato accessions (BGH-2102, BGH-2117, BGH-2127, BGH-2130, BGH-2332, and BGH-2343) were used as resistant parents and 15 F1 hybrids originated from these parents. The design was a randomized complete block with three replicates. The plants were inoculated with a mixture of Phytophthora infestans sporangia, the etiological agent of late blight, at a concentration of 5x103 sporangia mL-1. The area under the disease progress curve was used to evaluate resistance. Diallelic analysis was performed, considering the effect of genotypes as fixed. The general and specific combining ability of the accessions was estimated. The resistance pattern of the parents and of most of the F1 was the same as that of the resistant controls. The following were observed: additive genetic variability among the parents, predominance of nonadditive gene effects, and bidirectional dominance deviations in the control of the trait. The frequency of favorable and divergent alleles for late blight resistance is higher in the BGH-2117, BGH-2127, and BGH-2343 accessions.

O objetivo deste trabalho foi selecionar acessos resistentes à pinta-preta (Alternaria tomatophila) por meio da análise de agrupamento das curvas de progresso da doença em tomateiro (Solanum lycopersicum). Foram avaliados 134 acessos de tomateiro do Banco de Germoplasma de Hortaliças da Universidade Federal de Viçosa (BGH-UFV), no delineamento de blocos ao acaso, além das testemunhas suscetíveis 'Débora' e 'Santa Clara'. As plantas foram inoculadas com uma mistura de conídios de diferentes isolados de Alternaria spp. e avaliadas regularmente quanto à severidade da doença a cada três dias após a inoculação, no total de seis avaliações. Ajustou-se o modelo logístico aos dados de severidade da pinta-preta, e as estimativas obtidas para a incidência final da doença (B1) e a taxa de progresso da doença (B3) foram submetidas à análise de variância multivariada (Manova). As médias dessas estimativas, para cada acesso, foram submetidas à análise de agrupamento. Foram formados 24 grupos distintos com base no agrupamento das curvas de progresso da doença, o que possibilitou identificar os acessos BGH-2143, BGH-2235, BGH-2270 e BGH-2118 de tomateiro como potenciais fontes de resistência à pinta-preta.

The objective of this work was to select accessions resistant to early blight (Alternaria tomatophila) through cluster analysis of the disease progress curves in tomato (Solanum lycopersicum). One hundred and thirty-four tomato accessions from the Banco de Germoplasma de Hortaliças of Universidade Federal de Viçosa (BGH-UFV) were evaluated in a randomized complete block design, as well as the susceptible controls 'Débora' and 'Santa Clara'. The plants were inoculated with a mixture of spores from different Alternaria spp. isolates and evaluated regularly regarding disease severity every three days after inoculation, totalizing six evaluations. The logistic model was adjusted to data on early blight severity, and the estimates obtained for final disease incidence (B1) and rate of disease progression (B3) were subjected to multivariate analysis of variance (Manova). The means of these estimates, for each access, were subjected to cluster analysis. Twenty-four distinct groups were formed based on disease-progress curve clusters, which allowed the identification of the BGH-2143, BGH-2235, BGH-2270, and BGH-2118 accessions as potential sources of resistance to early blight.

Requeima causada por Phytophthora infestans é uma doença destrutiva de tomate no Brasil e em outras regiões tropicais e subtropicais. O objetivo do estudo foi o de analisar a herança da resistência à requeima e determinar os fatores genéticos que contribuem para a resistência na linhagem '163A'. A linhagem '163A' resultou de cruzamento interespecífico entre Solanum lycopersicum e S. habrochaites f. glabratum. As plantas foram avaliadas por infecção naturale por inoculaçãocom uma mistura de isolados do patógeno com 1000 esporângios mL-1. A expressão de '163A' contra múltiplos isolados do patógeno foi estável. A análise genética suportou a hipótese de dois genes recessivos controlando a resistência. O teste do modelo aditivo-dominante mostrou bom ajuste para dados que confirmam a ausência ou negligenciam epistasia.

Late blight caused by the oomycete Phytophthora infestans is a destructive disease of tomato in Brazil and other tropical and subtropical regions. The purpose of the present study was to analyses the inheritance of resistance to late blight and determine the genetic factors contributing to the resistance in the inbred line in '163A'. The Line '163A' resulted from interspecific cross between Solanum lycopersicum and S. habrochaites f. glabratum. Inoculated field with mixture isolates of pathogen with 1000 spores mL-1 and naturally infested field trials showed that the expression of '163A' against multiple isolates of the pathogen was stable. The genetic analysis supported the hypothesis of two recessive genes controlling the resistance. The scaling test of additive-dominance model showed that is a good fit for the data confirming the absence or neglect of epistasis.

O objetivo deste trabalho foi caracterizar 50 subamostras de tomateiro do Banco de Germoplasma de Hortaliças da Universidade Federal de Viçosa (BGH-UFV). Foram avaliadas juntamente com as testemunhas Débora, Fanny e Santa Clara, em experimento em blocos ao acaso, com duas repetições. O experimento ocorreu na Horta de Pesquisas da UFV, no período de fevereiro a maio de 2007. Avaliou-se a severidade da doença, que é a porcentagem da área foliar lesionada. Os valores de severidade foram transformados em área abaixo da curva de progresso da doença (AACPD), possibilitando uma melhor visualização dos resultados. Foi realizada a análise de variância, seguido do agrupamento das médias de AACPD pelo teste de Scott-Knott, a 5% de significância. As subamostras BGH-2081, BGH-2034, BGH-700, BGH-2057, BGH-2035, BGH-2054, BGH-2018, BGH-2065, BGH-2008 e BGH-2032 apresentaram menor média de AACPD em relação às testemunhas, podendo ser utilizadas em futuros programas de melhoramento.

The purpose of this study was to characterize 50 tomato genotypes of the Vegetable Genebank of the Federal University of Viçosa. They were evaluated together with the controls Débora, Fanny and Santa Clara, in a randomized block design with two replications. The experiment was conducted in a research field of the UFV, from February to May 2007. We evaluated the disease severity, which is the percentage of diseased leaf area. The severity values were transformed into area under the disease progress curve (AUDPC), improving the result visualization. The analysis of variance and grouping of AUDPC means by the Scott-Knott test at 5 % significance were performed. The accessions BGH-2081, BGH-2034, BGH-700, BGH-2057, BGH-2035, BGH-2054, BGH-2018, BGH-2065, BGH-2008, and BGH-2032 had a lower mean AUDPC than the controls and are therefore indicated for future breeding programs.

O objetivo deste trabalho foi estimar curvas de progresso de requeima, em genótipos de tomateiro, e identificar grupos de genótipos resistentes à doença. Foram avaliados 25 híbridos de tomateiro, originados de cruzamentos entre quatro variedades comerciais, um acesso do Banco de Germoplasma de Hortaliças (BGH), da Universidade Federal de Viçosa (UFV), e cinco linhagens F8 (Solanum lycopersicum x Solanum habrochaites), estas últimas selecionadas como fonte de resistência à requeima. As plantas foram inoculadas com uma mistura de esporângios de Phytophthora infestans e, em seguida, foram realizadas seis avaliações quanto à severidade de requeima, a intervalos de três dias. Ajustou-se o modelo exponencial aos dados de percentagem de severidade de requeima, e as estimativas obtidas quanto à incidência inicial da doença (y o) e taxa de progresso da doença (r) foram submetidas à análise de variância multivariada (Manova). As médias dessas estimativas, para cada genótipo, foram submetidas à análise de agrupamento. Observou-se um número ótimo de oito grupos distintos, o que possibilitou identificar genótipos resistentes e suscetíveis. Os híbridos experimentais Ikram x 73 A, Nemo-Netta x 133 A, Ikram x 163 A e Nemo-Netta x 163 A apresentaram a menor taxa de progresso de requeima e, portanto, maior resistência à doença.

The objective of this work was to estimate the progress curves of late blight in tomato genotypes, and to identify tomato genotype groups resistant to this disease. Twenty-five hybrid tomatoes, originated from crosses between four fresh market cultivars, one access from the Banco de Germoplasma de Hortaliças (BGH) of Universidade Federal de Viçosa (UFV), and five F8 lines (Solanum lycopersicum x Solanum habrochaites), selected as late blight resistance source, were evaluated. Plants were inoculated with a sporangium mixture of Phytophthora infestans, and then, six evaluations for late blight severity were carried out every three days. The exponential model was fitted to the data of late blight severity percentage, and the obtained parameter estimates - initial incidence of disease (y o )and progress rate of disease (r) - were submitted to the multivariate analysis of variance (Manova). The means of these estimates, obtained for each genotype, were submitted to the cluster analysis. An optimal number of eight distinct groups was observed, which made it possible to identify the genotypes belonging both to the resistant and susceptible groups. The experimental hybrids Ikram x 73 A, Nemo-Netta x 133 A, Ikram x 163 A and Nemo-Netta x 163 A showed the smaller rate of disease progress and, therefore, higher late blight resistance.

Quarenta isolados bacterianos endofíticos de plantas sadias de tomateiro foram avaliados quanto à sua potencialidade como agentes de biocontrole de doenças do tomateiro. Foi realizada, em casa de vegetação, uma seleção massal utilizando-se Pseudomonas syringae pv. tomato e Alternaria solani, como patógenos desafiantes. Com base na média do número de lesões por planta, quatro isolados foram selecionados como potenciais agentes de biocontrole dessas enfermidades fúngica e bacteriana do tomateiro. Esses isolados foram identificados, por meio do sequenciamento do gene 16S do DNA ribossômico, como Acinetobacter johnsonii (UFV-E05), Serratia marcescens (UFV-E13), Sinorhizobium sp. (UFV-E25) e Bacillus megaterium (UFV-E26). Os mesmos isolados selecionados para o biocontrole também foram avaliados quanto à sua capacidade de promover o crescimento em plantas e somente S. marcescens (UFV-E13) proporcionou aumento na altura das plantas.

Forty isolates of endophytic bacteria obtained from healthy tomato plants were tested for their potential as biocontrol agents of tomato diseases. A massal screening was performed at greenhouse using Pseudomonas syringae pv. tomato and Alternaria solani as challenging pathogens. Based on the average number of lesions per plant, four isolates were selected as potential agents of biocontrol of these tomato diseases caused by fungi and bacteria. These isolates were identified by 16S ribosomal DNA sequence analysis as Acinetobacter johnsonii (UFV-E05), Serratia marcescens (UFV-E13), Sinorhizobium sp. (UFV-E25) and Bacillus megaterium (UFV-E26). The four endophytes selected for biocontrol were also evaluated for their ability of promoting plant growth and only S. marcescens (UFV-E13) presented increase in the height of the plants.

In Brazil, no commercial tomato (Solanum lycopersicum L. formerly Lycopersicon esculentum Mill.) varieties are available which are resistant to the late blight, one of the most important tomato diseases, produced by the phytopathogenic oomycete Phytophthora infestans. The wild tomato (Solanum habrochaites Knapp & Spooner, formerly Lycopersicon hirsutum Dunal) shows resistance to P. infestans, because of which we investigated an interspecific cross between S. lycopersicum cv. Santa Clara and S. habrochaites accession BGH 6902 maintained at the Horticultural Germplasm Bank at the Federal University of Viçosa (Banco de Germoplasma de Horticultura (BGH), Universidade Federal de Viçosa (UFV), Viçosa, Minas Gerais, Brazil) The genitors, F1, F2, BC1 and BC2 were used to study the inheritance of resistance to P. infestans and to estimate the genetic parameters associated with resistance. Analysis of the area under the disease progress curve (AUDPC) indicated that inheritance is polygenic and that dominance controls character, whereas mean analysis showed that the additive effect was the most important. Although the character presents variability, the heritability is low which generates the need to better control the environment to obtain success with the selection.

As atividades amilolítica e pectinolítica de 45 isolados de Alternaria solani, provenientes de diferentes hospedeiros, foram estimadas por meio da difusão enzimática em meio sólido específico e mensuração do halo de degradação do substrato. Todos os isolados degradaram pectina. Apenas 17 isolados apresentaram atividade amilolítica, sendo nove isolados provenientes de batateira. Somente o isolado AS18 se destacou como bom produtor de ambas as enzimas. Uma vez que a atividade pectinolítica foi mais evidente, avaliou-se a influência de pectinases na agressividade de A. solani ao tomateiro. Para isso, cinco isolados (2 de berinjela, 2 de tomateiro e 1 de batateira) contrastantes quanto à produção de pectinases foram selecionados para testes em folíolos destacados e plantas inteiras. Quatro isolados foram utilizados no teste em folíolos destacados (AS6, AS7, AS12 e AS26), e constatou-se haver variabilidade patogênica. A correlação obtida entre o tamanho das lesões e a atividade pectinolítica foi de r = 0,963 (P = 0,087). Cinco isolados (AS6, AS7, AS12, AS25 e AS26) foram inoculados em plantas inteiras de tomate. Os isolados não diferiram quanto ao número de lesões/cm² de área foliar, porém variaram em agressividade. Houve correlação (r = 0,916; P = 0,042) entre a atividade de pectinases e o índice de doença, sugerindo possível papel para as enzimas pécticas durante a infecção de A. solani em tomateiro. É provável que as diferenças no perfil enzimático dos isolados estejam associadas ao hospedeiro original de onde os mesmos foram obtidos. Os resultados reforçaram evidências de especificidade por hospedeiro em populações de A. solani.

Amilolytic and pectinolytic activities of 45 isolates of Alternaria solani from distinct hosts were investigated. Enzyme secretion was evaluated in glucose-free medium amended with pectin or starch as carbon source. All isolates degraded pectin. Only 17 isolates had amilolytic activity, and nine of these isolates were isolated from potato. Only AS18 had good amilolytic and pectinolytic activities. The role of pectinases produced by A. solani in its aggressiveness on tomato was examined. Five isolates showing substantial differences regarding the ability to produce pectic enzymes were selected, two from eggplant, two from tomato plants, and one from potato plant. Four isolates (AS6, AS7, AS12, and AS26) were inoculated on detached leaflets of tomato and pectinolytic activity was correlated with lesion size (r = 0.963; P = 0.087). Pathogenicity test with five isolates (AS6, AS7, AS12, AS25, and AS26) was conducted in tomato plants. Variation in the number of lesions per cm² of leaf area was not detected, but isolates varied in aggressiveness. Pectinolytic activity and aggressiveness were correlated (r = 0.916; P = 0.042). In general, pectinases-producing isolates caused larger lesions on tomato plants. It is possible that enzymatic profile differences are associated with the host origin. The results contribute to cumulative evidences of host specificity in populations of A. solani.

Protoplastos são ferramentas biológicas importantes para pesquisas em fungos filamentosos, sendo empregados intensamente em transformação genética. O isolamento de protoplastos de Magnaporthe grisea foi facilitado com Novozym 234, contudo, este complexo enzimático encontra-se indisponível no mercado. Assim, objetivou-se comparar a eficiência de enzimas líticas disponíveis comercialmente na obtenção de protoplastos de M. grisea. Paralelamente, analisaram-se estabilizadores osmóticos, tempos de digestão e freqüência de regeneração. Maior produção de protoplastos foi obtida com o uso simultâneo de Lysing Enzymes e Cellulase Onozuka R-10. O uso de 10 ou 15 mg de cada complexo enzimático, em 3 mL de estabilizador osmótico, resultou em maior liberação de protoplastos. O melhor estabilizador osmótico foi MgSO4 1,2 M / NaH2PO4 0,01 M, pH 5,8, seguido por MgSO4 0,8 M / NaH2PO4 0,01 M, pH 5,8. O isolamento de protoplastos foi monitorado a cada 60 minutos, atingindo o máximo após incubação por 3 a 6 horas. No entanto, maior freqüência de regeneração (19,4%) foi registrada para protoplastos obtidos após 3 horas de hidrólise enzimática.

Protoplasts are important biological tools in filamentous fungi research. Fungal protoplasts have been extensively used in experiments with genetic transformation. Protoplastization of Magnaporthe grisea was accomplished with Novozym 234, however, this enzymatic complex is no commercially available for purchase. Thus, the efficiency of several other commercial enzymes in M. grisea protoplasts preparation was investigated. At the same time, osmotic buffer, digestion time and regeneration rate were also analyzed. The highest protoplasts production was obtained with Lysing Enzymes plus Cellulase Onozuka R-10. The use of 10 or 15 mg of each enzymatic complex in 3 mL of osmotic buffer was most effective for the protoplasts yields. The best osmotic buffer was MgSO4 1.2 M / NaH2PO4 0.01 M, pH 5.8, followed by MgSO4 0.8 M / NaH2PO4 0.01 M, pH 5.8. The M. grisea protoplasts yield, evaluated at each 60 min, increased with 3 to 6 hours of incubation. However, the highest regeneration frequency (19.4%) was recorded for protoplasts obtained with 3 hours of enzymatic hydrolyse.
