Resumo Nas regiões Centro-Oeste e Sul do Brasil, a cultura da erva-mate (Ilex paraguariensis) constitui-se uma importante fonte de extrativismo florestal, além de diversificar a renda, principalmente na agricultura familiar. A erva-mate, como outras espécies florestais, é infectada por diversos patógenos, como o gênero Fusarium, causador da podridão-de-raízes. Os patógenos possuem ampla variabilidade genética, mesmo dentro de um único gênero e dentro de uma única espécie, resultado de adaptações ao meio em que vivem. Este estudo teve como objetivo avaliar o sequenciamento das regiões do genoma ITS e β-tubulina na identificação em nível de espécie dos isolados de Fusarium spp. patogênicos à erva-mate, e identificar a formação de grupos de compatibilidade vegetativa (VCG) entre os isolados de Fusarium spp. patogênicos à erva-mate. O sequenciamento das regiões ITS e β-tubulina consolidou a separação entre os sete isolados patogênicos em F. solani e F. oxysporum. Para esses isolados, a técnica para obtenção de mutantes nit foi adequada para a classificação fenotípica, fundamentais nos testes de VCG para espécies de Fusarium, sendo obtidos cinco VCG.

Abstract In the central-western and southern regions of Brazil, the cultivation of yerba mate (Ilex paraguariensis) is considered an important source of forest extraction, besides providing an alternative income, especially for families. The yerba mate, like other forest species, is infected by several pathogens, such as fungi of the genus Fusarium, causing root rot. Pathogens have wide genetic variability, even within a single genus and species, resulting from adaptations to their environments. This study aimed at characterizing molecularly isolated Fusarium spp. through the sequencing of the ITS and β-tubulin genome regions, identifying the species level as well as the formation of vegetative compatibility groups (VCG) among these isolates of the root rot pathogen in yerba mate. The sequencing of the ITS and β-tubulin regions, defined the separation between the seven pathogenic isolates of F. solani and F. oxysporum. For the same isolates, the technique to obtain nit mutants was suitable for the phenotypic classification, obtaining five VCGs for species of Fusarium, root rot in yerba mate.

RESUMO Objetivou-se avaliar o controle genético integrado ao controle químico da ferrugem asiática da soja (FAS) e os efeitos dessas medidas sobre a produtividade da cultura. O experimento foi conduzido em Erechim/RS, nas safras 2016/17 e 2017/18, sob delineamento de blocos casualizados, em esquema de parcela subdividida (cultivares, nas parcelas; e fungicidas, nas subparcelas), com quatro repetições. Foram utilizadas as seguintes cultivares: BMX Vanguarda (sem tolerância à FAS); TMG 7062; TMG 7262; e TMG 7161, tolerantes à FAS (cultivares com tecnologia Inox™). Os fungicidas utilizados foram: T1) testemunha (sem aplicação de fungicidas); T2) azoxistrobina + benzovindiflupyr; T3) difenoconazol + ciproconazol; T4) trifloxistrobina + protioconazol; e T5) epoxiconazol + fluxapiroxade + piraclostrobina. Foram realizadas quatro aplicações de fungicidas nos estádios V6; R1; R5.1; e R6. Durante a condução do experimento, para o cálculo da área abaixo da curva de progresso da doença (AACPD), avaliou-se a severidade da doença, com intervalos de 7 dias, em uma amostra aleatória de 10 trifólios por parcela. Após a colheita, determinou-se os componentes de rendimento: número de grãos por planta, peso de mil grãos (g), e a produtividade (kg ha-1). Na safra 2017/18 o fungicida difenoconazol + ciproconazol não foi eficiente para o controle da FAS. As cultivares de soja TMG 7062, TMG 7161 e TMG 7261 retardaram o progresso da doença, entretanto somente a cultivar TMG 7161 apresentou tolerância sob a presença do inóculo nas safras 2016/17 e 2017/18. A associação entre controle químico e genético mostra-se eficiente no controle da FAS.

ABSTRACT The objective of this study was to evaluate the genetic control integrated to the chemical control of Asian soybean rust (ASR) and the effects of these measures on crop yield. The experiment was conducted in Erechim, Rio Grande do Sul State, Brazil, in 2016/17 and 2017/18 crop years, under a randomized block design, in a subdivided plot scheme (cultivars in the plots and fungicides in the subplots), with four replicates. The following cultivars were used: BMX Vanguarda (without ASR tolerance); TMG 7062; TMG 7262, and TMG 7161, tolerant to ASR (Inox™ Technology cultivars). The fungicides used were: T1) control (without application of fungicides); T2) azoxystrobin + benzovindiflupyr; T3) difenoconazole + cyproconazole; T4) trifloxystrobin + prothioconazole, and T5) epoxiconazole + fluxapyroxad + pyraclostrobin. Four fungicide applications were carried out at the V6, R1, R5.1 and R6 stages. During the experiment, for the calculation of the area under disease progress curve (AUDPC), disease severity was assessed at 7-day intervals in a random sample of 10 trifolia per plot. After harvest, yield components were determined: number of grains per plant, thousand grain weight (g), and yield (kg ha-1). In 2017/18 crop year, the fungicide difenoconazole + cyproconazole was not efficient for ASR control. The soybean cultivars TMG 7062, TMG 7161 and TMG 7261 delayed the disease progression; however, only TMG 7161 presented tolerance in the presence of the inoculum in 2016/17 and 2017/18 crop years. The association between chemical and genetic control is shown to be efficient for ASR control.

RESUMO: Na busca por melhores rendimentos, o tratamento de sementes por microbiolização tem sido utilizado como alternativa ao tratamento químico. Objetivou-se verificar a qualidade fisiológica e sanitária de sementes de feijão crioulo, var. Chumbinho, após microbiolização com doses de um produto comercial (p.c.) com Trichoderma harzianum (cepa ESALQ-1306). Os tratamentos foram: T1) 100 mL p.c./100 kg de sementes; T2) 150 mL p.c.; T3) 200 mL p.c.; T4) 200 mL tratamento químico (p.c., 250 g L-1 fipronil + 25 g L-1 piraclostrobina + 225 g L-1 tiofanato metílico); e T5) controle (sem revestimento de sementes). Os testes foram: sanidade (blotter test), germinação, primeira contagem, envelhecimento acelerado, teste de frio sem solo, índice de velocidade de germinação (IVG), comprimento de parte aérea e de raiz das plântulas e emergência em casa de vegetação. T. harzianum controlou Aspergillus spp., Penicillium spp. e Fusarium oxysporum. A dose 100 mL de p.c. teve melhores resultados para germinação e vigor, sendo que essa dose é uma alternativa ao tratamento químico em sementes de feijão crioulo.

ABSTRACT: In the search for improved yields, seed treatment by microbiolization has been used as an alternative to chemical treatment. The objective was to verify the physiological and sanitary quality of creole bean seeds, var. Chumbinho, after microbiolization with doses of a commercial product (c.p.) with Trichoderma harzianum (strain ESALQ-1306). The treatments were: T1) 100 mL c.p./100 kg seeds; T2) 150 mL c.p.; T3) 200 mL c.p.; T4) 200 mL of chemical treatment (c.p., 250 g L-1 fipronil + 25 g L-1 pyraclostrobin + 225 g L-1 thiophanate-methyl); and T5) control (without coating of seeds). The tests were: sanitary test (blotter test); germination and first count; accelerated aging, cold germination without soil, speed of germination rate (SGR), seedling shoot and root lengths, and emergence of seedlings in a greenhouse. T. harzianum controlled Aspergillus spp., Penicillium spp. and Fusarium oxysporum. With 100 mL c.p. of T. harzianum dose had better results for the germination and vigor, and this dose it is an alternative to chemical treatment in creole bean seeds.

RESUMO A ferrugem da folha do trigo (Puccinia triticina) é uma das doenças mais frequentes em lavouras de trigo. Para um controle efetivo dessa doença é recomendado o controle foliar através da aplicação de misturas de fungicidas triazóis e estrobilurinas. O objetivo deste estudo foi avaliar a eficácia das diferentes formulações fungicidas e do número de aplicações para o controle da ferrugem da folha do trigo na cultivar TBIO Pioneiro 2010. O delineamento experimental utilizado foi inteiramente casualizado, fatorial 2x2, consistindo em duas formulações fungicidas e dois números de aplicações, com quatro repetições. Os tratamentos foram: azoxistrobina + tebuconazol com três e quatro aplicações; trifloxistrobina + protioconazol com três e quatro aplicações; e um tratamento testemunha sem aplicação de fungicida. Nos tratamentos com três aplicações, os fungicidas foram aplicados nos estádios fenológicos de: alongamento, emborrachamento e florescimento. Os tratamentos com quatro aplicações foram realizados nos estádios fenológicos de: perfilhamento, alongamento, emborrachamento e florescimento. A eficiência dos fungicidas no controle da ferrugem foi analisada através da avaliação da severidade da doença, calculando-se a Área Abaixo da Curva de Progresso da Doença (AACPD). Os dados de severidade foram obtidos através de notas da porcentagem da área foliar com sintomas/sinais da doença segundo a escala diagramática de Cobb. Fungicidas com azoxistrobina + tebuconazol e trifloxistrobina + protioconazol apresentaram eficiência no controle de ferrugem da folha de trigo (todos acima de 85%). Em componentes de rendimento, não houve diferença estatística entre o número de aplicações, mas, houve diferença estatística entre as formulações de fungicidas, exceto para produtividade. Além disso, para os componentes do rendimento, todos os tratamentos fungicidas diferiram significativamente da testemunha.

ABSTRACT Leaf rust (Puccinia triticina) is one of the most common diseases in wheat crops. For effective control of such disease, leaf application of mixtures of triazole and strobilurin fungicides is recommended. The aim of this study was to evaluate the efficacy of different fungicide formulations and numbers of applications in controlling wheat leaf rust in the cultivar TBIO Pioneiro 2010. Experimental design was completely randomized, 2x2 factorial arrangement, consisting of two fungicide formulations and two numbers of applications, as well as four replicates. Treatments were: azoxystrobin + tebuconazole, three and four applications; trifloxystrobin + prothioconazole, three and four applications; and a control treatment without fungicide application. For treatments with three applications, fungicides were applied at the phenological stages of: elongation, booting and flowering. For treatments with four applications, fungicides were applied at the phenological stages of: tillering, elongation, booting and flowering. The efficiency of fungicides on leaf rust control was analyzed based on the disease severity assessment by calculating the Area Under Disease Progress Curve (AUDPC). Severity data were obtained based on percentage scores of leaf area with visible disease symptoms/signs according to Cobb’s diagrammatic scale. Fungicides with azoxystrobin + tebuconazole and trifloxystrobin + prothioconazole showed efficient control of wheat leaf rust (higher than 85%). For yield components, there was no statistical difference between numbers of applications, but there was a statistical difference between fungicide formulations, except for grain yield. Furthermore, for yield components, all fungicide treatments significantly differed from control.

RESUMO Cordia americana (L.) Gottschling & J. S. Mill. (guajuvira) é uma espécie florestal amplamente distribuída no território brasileiro e que apresenta importância econômica e ambiental devido à qualidade de sua madeira e utilização em reflorestamentos. Com o objetivo de avaliar a patogenicidade de Fusarium sp. associado às sementes, foram caracterizados, morfológica e molecularmente, quatro isolados do fungo que foram inoculados nas sementes de guajuvira. A caracterização morfológica foi realizada com o auxílio de uma chave de identificação para o gênero Fusarium e para a identificação molecular foram sequenciadas as regiões genômicas Internal Transcribed Spacer (ITS) e Fator de elongação 1 - alpha (TEF1-α). Os resultados do sequenciamento de DNA concordaram com os da caracterização morfológica e permitiram identificar as espécies Fusarium acuminatum e Fusarium verticillioides. No teste de patogenicidade, verificou-se que ambas as espécies do fungo foram patogênicas a Cordia americana, causando tombamento em pré e pós-emergência, além de apodrecimento de raízes e necrose no hipocótilo.

ABSTRACT Cordia americana (L.) Gottschling & J. S. Mill. (guajuvira) is a forest species widely distributed in Brazil and represents environmental and economic importance due to the quality of its timber and use in reforestation. In order to evaluate the pathogenicity of Fusarium sp. associated with seeds, characterizing the isolates morphologically and molecularly in order to identify them on the species level, was obtained four fungal isolates that were inoculated in seeds to demonstrate their pathogenicity. The morphological characterization was performed using an identification key for the genus Fusarium and, for the molecular identification, genomic regions Internal Transcribed Spacer (ITS) were sequenced with its elongation factor 1 alpha (TEF1-α). The results of the molecular identification agreed with the morphological characterization and allowed to identify the species Fusarium acuminatum and Fusarium verticillioides, both being pathogenic to Cordia americana, causing damping-off in pre and post emergence, and rotting of roots and necrosis of hypocotyl.

O objetivo deste trabalho foi verificar a reação de resistência de porta-enxertos e cultivares de tomateiro à raça 3 de F. oxysporum f. sp. lycopersici e a comprovação de sua resistência às raças 1 e 2. Os porta-enxertos testados foram: Guardião, Helper-M, Anchor-T, Dr. K, Kagemuscha, Block, Magnet e He-Man, e, como testemunhas, as cultivares 'Kada', 'Santa Clara', 'Gisele', 'BHRS 2,3'. O delineamento experimental utilizado foi o inteiramente casualizado, com parcelas subdivididas e 5 repetições, sendo os porta-enxertos e as cultivares, alocados na parcela principal e as 3 raças de F. oxysporum f. sp. lycopersici, nas subparcelas. As plantas foram inoculadas e mantidas em casa de vegetação com temperatura e umidade controladas. Os porta-enxertos foram avaliados pela severidade de infecção do fungo no xilema. Todos os porta-enxertos foram resistentes às raças 1 e 2 de Fusarium oxysporum f. sp. lycopersici e apenas Guardião e Block foram resistentes à raça 3.

The aim of this research was to verify the reaction of tomato rootstocks to race 3 of F. oxysporum f. sp. lycopersici and his resistance to races 1 and 2. It was tested the rootstocks: Guardião, Helper-M, Anchor-T, Dr K, Kagemusha, Block, Magnet and He-man and, as a control, the cultivars 'Kada', 'Santa Clara', 'Gisele', 'BHRS 2,3'. The experimental design was a completely randomized type, with split-plots and 5 replicates. The rootstocks and cultivars were allocated in principal plot, and the 3 races of F. oxysporum f. sp. lycopersici, in the sub-plots. The plants were inoculated and maintained in vegetation house with temperature and humidity controlled. The rootstock was assessed by the severity of disease infection in the xylem. All rootstock were resistant to races 1 and 2 of F. oxysporum f. sp. lycopersici and only Guardian and Block were resistant to race 3.

Extratos vegetais podem auxiliar no controle de doenças de plantas, por sua atividade antimicrobiana ou pela ativação de mecanismos de defesa. Neste trabalho, objetivou-se avaliar o efeito dos extratos aquosos de alho e alecrim autoclavados e não autoclavados sobre o crescimento micelial de Colletotrichum lindemuthianum, a produção de faseolina em feijoeiro e a severidade de antracnose em campo. O fungo foi incubado em meio de cultura contendo os extratos, e o crescimento foi mensurado por 18 dias. Para avaliação de faseolina, os extratos foram aplicados sobre hipocótilos de feijão, a extração foi feita em etanol e a leitura foi realizada em espectrofotômetro. A severidade da antracnose foi avaliada em plantas de feijão pulverizadas com os extratos e inoculadas com o fungo. Para o extrato de alho, a maior redução (57,6%) no crescimento micelial foi com a dose de 3,0% do extrato não autoclavado. Já para o extrato de alecrim, a maior redução (18,6%) foi com o extrato autoclavado. A autoclavagem não teve efeito sobre a indução de faseolina. À campo, os extratos não reduziram significativamente a severidade de antracnose. O extrato de alho não autoclavado mostrou-se mais eficiente na redução do crescimento de C. lindemuthianum, enquanto que o de alecrim, autoclavado ou não, foi mais efetivo na indução de faseolina.

Plant extracts may assist in controlling plants diseases due to its antimicrobial activity and activation of defense mechanisms. This study aimed to evaluate the effect of aqueous extracts of garlic and rosemary, autoclaved or non autoclaved, on the mycelial growth of Colletotrichum lindemuthianum, production of phaseollin by beans and severity of anthracnose in the field. The fungus was incubated in medium containing the extracts and its growth measured for 18 days. For phaseollin evaluation the extracts were applied on bean hypocotyls, the phytoalexin extracted in ethanol and read in spectrophotometer. The severity of anthracnose was evaluated in bean plants sprayed with the extracts and inoculated with the fungus. For the garlic extract, the greater reduction (57,6%) in micelial growth was at 3% in the non autoclaved extract, as for the rosemary extract, the greater reduction (18,6%) was in the autoclaved extract. Autoclaving had no effect on the induction of phaseollin. At field conditions, the extracts did not reduce significantly the severity of anthracnose. The non autoclaved garlic extract showed to be more efficient in reducing the mycelial growth of C. lindemuthianum, while rosemary, autoclaved or not, proved to be more effective in the induction of phaseollin.
