RESUMO OBJETIVO Investigar a presença das mutações no gene Angiopoietina (ANGPT1) e gene Plasminogênio (PLG) em pacientes com Angioedema Hereditário (AEH) com inibidor C1 esterase (C1-INH) normal e negativos para mutação do gene F12. MÉTODOS Foram avaliados pacientes com diagnóstico clínico de AEH sem deficiência ou disfunção de C1-INH e negativos para mutação do gene F12. Realizou-se extração, quantificação e diluição do DNA a uma concentração de 100 ng/uL, em seguida amplificação do DNA (PCR) para avaliação molecular do exon 2 do gene ANGPT1 e do exon 9 do gene PLG para identificação das mutações c.807G>T.p.A119S e c.988A>G p.K330E, respectivamente. O produto da PCR foi avaliado em eletroforese em gel de agarose 1%. Foi realizado o sequenciamento pelo método de Sanger. As análises dos eletroferogramas foram realizadas pelo programa FASTA®. RESULTADOS Foram sequenciadas amostras de 15 mulheres, idade entre 10 e 60 anos, com níveis séricos de inibidor de C1 esterase e C4 normais variando de 22 a 39mg/dL e 10 a 40mg/dL, respectivamente. Não foram identificadas mutações nos éxons analisados dos genes ANGPT1 e PLG. Entretanto no gene PLG foram encontrados polimorfismo de nucleotídeo único (SNP), em duas pacientes homozigotas e cinco heterozigotas. CONCLUSÃO Mais estudos sobre SNP são necessários para esclarecer estes achados pois eles podem ser utilizados como marcadores moleculares do AEH e alvo para novos tratamentos.

SUMMARY OBJECTIVE To investigate the presence of the Angiopoietin 1 (ANGPT1) and Plasminogen (PLG) mutations in patients with Hereditary Angioedema (HAE) and normal C1 esterase inhibitor (C1-INH) levels, who do not harbor the F12 gene mutation. METHODS Patients clinically diagnosed with HAE but without C1-INH deficiency or dysfunction and F12 gene mutation were evaluated. DNA extraction, quantification, and dilution were performed at a concentration of 100 ng/µL, followed by a DNA amplification (PCR) for molecular evaluation of exon 2 of the ANGPT1 gene and exon 9 of the PLG gene for identification of mutations c.807G>T / p.A119S and c.988A>G / p.K330E, respectively. The PCR product was evaluated in 1% agarose gel electrophoresis. Sequencing was performed using the Sanger method. The electropherograms were analyzed using the FASTA® program. RESULTS DNA samples from 15 women were sequenced. Their ages ranged from 10 to 60 years and the normal C1 esterase and C4 inhibitor serum levels ranged from 22 to 39 mg/dL and from 10 to 40 mg/dL, respectively. No mutations were detected in the analyzed exons of ANGPT1 and PLG. However, a single-nucleotide polymorphism (SNP) was detected in two homozygotic and five heterozygotic patients. CONCLUSION Further studies are needed to evaluate these SNPs and scrutinize their potential for use as molecular markers of HAE and as novel therapeutic targets.

ABSTRACT Extracts of the seeds of Tephrosia vogelii Hook. f. were studied in relation to its chemical composition and toxicity to the brown stink bug Euschistus heros (F.). The extracts were obtained in ethyl acetate and ethanol in the sequence according to the polar nature of the solvents. Extracts were sprayed in concentration of 1.0, 2.5, 5.0, 7.5 and 10% on third-instars nymphs and adults, and mortality was recorded. Presence two rotenoids in ethyl acetate was detected, with analyzed with gas chromatography-mass spectrometry (GC-MS). Crude fraction analyses confirmed the presence of these rotenoids (tephrosin - 2.71% in ethyl acetate and 3.66% in methanol; and deguelin - 10.46% in ethyl acetate and 1.22% in methanol) and three other rotenoids in small amounts. Eight days after applications, ethyl acetate caused more stink bugs mortality and on less time than ethanol extract, because great quantity of rotenoids, as polarity. Concentrations above to 1 and 2.5% of the ethyl acetate extracts caused mortality above 80% of the nymphs and adults of E. heros, respectively. Concentration were considered high, thus chemist analyzes demonstrated high rotenoids presence. In conclusion, seed T. vogelli extracts, rich in deguelin and tephrosin (3:1), cause mortality of E. heros, however, high concentration are necessary.

ABSTRACT We evaluated the influence of host preference, mating, and release density on Telenomus remus (Nixon, 1937) (Hymenoptera: Platygastridae) parasitizing eggs of Spodoptera frugiperda (Smith, 1797) (Lepidoptera: Noctuidae). First, we tested host preference of T. remus (free choice test) offered a choice between eggs of Corcyra cephalonica (Stainton, 1865) (Lepidoptera: Pyralidae) and S. frugiperda. Parasitism capacity and host preference (S. frugiperda) of T. remus reared on either of the two hosts did not differ. Secondly, we evaluated the influence of mating behavior of T. remus females on its parasitism. Only the offspring sex ratio differed between treatments, indicating that the species reproduces by parthenogenesis of the arrhenotoky type. Finally, we evaluated the influence of release density on T. remus parasitism. This was tested by releasing different numbers of the parasitoid per S. frugiperda egg using T. remus reared for different numbers of generations on C. cephalonica eggs. The regression analysis between percentage of parasitism and density of released T. remus females showed a quadratic effect for all tested parasitoid generations (F35, F40, and F45) with maximum parasitism from 65.07% to 71.69%. Our results allow the conclusion that (a) T. remus prefers S. frugiperda eggs, regardless of the host on which this parasitoid was reared, showing no preimaginal conditioning; (b) Mating does not affect the number of eggs parasitized by T. remus or the development of its offspring; and (c) The optimal T. remus release density when reared on C. cephalonica is between 0.133 and 0.150 females/S. frugiperda.

RESUMO: Este estudo investigou os aspectos biológicos de Dichelops melacanthus (Hemiptera: Pentatomidae) em três temperaturas (19±1°C, 25±1°C e 31±1°C), alimentados com grãos imaturos de milho 2B688Hx (expressa a proteína inseticida Cry1F, para o controle de lepidópteros-alvo) e 2B688. Foi observado que a temperatura de 19±1°C prejudicou o desenvolvimento de D. melacanthus, pois, nessa condição, foi registrada a maior taxa de mortalidade das ninfas, menor peso de adultos e as fêmeas não ovipositaram. Por outro lado, a duração da fase jovem, mortalidade, peso de adultos, fecundidade e viabilidade de ovos não diferiu entre as temperaturas 25±1°C e 31±1°C. Também foi demonstrado que os aspectos biológicos dos percevejos alimentados com grãos imaturos de milho 2B688Hx não diferiram dos que se desenvolveram com a isolinha 2B688. Este estudo comprova que D. melacanthus desenvolve-se melhor em condições de temperaturas mais elevadas (até 31±1°C), e o fato de se alimentar com grãos imaturos de milho 2B688Hx não afeta o seu desenvolvimento.

ABSTRACT: This study investigated the development of Dichelops melacanthus (Hemiptera: Pentatomidae) at different temperatures (19±1°C, 25±1°C and 31±1°C) reared on 2B688Hx (expressing Cry1F insectice protein to control lepidopterans target) immature corn grains. The temperature of 19±1°C impaired the development of D. melacanthus and recorded the highest mortality of nymphs, lower weight and adult females laid no eggs. Moreover, the length of the nymphal biology, adult weight, fecundity and viability of eggs did not differ between temperatures 25±1°C and 31±1°C. It was also shown that the biological aspects of green belly stink bugs fed with 2B688Hx immature grains of corn do not differ from that develop with 2B688 isoline. This study showed that the biological cycle of D. melacanthus is favored at warmer temperature (until 31±1°C), and the consume of immature corn grain 2B688Hx does not affect its development.

Lavouras de morango recebem várias aplicações de agrotóxicos e resíduos nos frutos são recorrentes. O desenvolvimento e a aplicação de métodos de manejo que evitem a aplicação de pesticidas, principalmente durante o período de frutificação, é uma necessidade iminente. Compararam-se as populações do ácaro rajado Tetranychus urticae Koch. em plantas adubadas com fertilizantes orgânicos e minerais (como o usados por produtores) e tratadas com produtos alternativos. As plantas foram cultivadas em vasos, em casa de vegetação infestada de população local do ácaro. Extratos de Tephrosia voggelii (Hook), óleo de nim (Azadirachta indica A. Juss.) e calda sulfocálcica foram pulverizados três vezes em intervalos de 10 dias. As avaliações foram realizadas três vezes após cada pulverização. A interação entre a fertilização e produtos alternativos em pulverização foi também avaliada, mas não foi significativa. A população do ácaro foi cerca de 44% menor em plantas fertilizadas com fertilizante orgânico do que aquelas com fertilizante mineral. Extrato de T. voggelii reduziu em grande quantidade a população do ácaro; o óleo de nim reduziu em algumas avaliações e redução intermediária foi observada com calda sulfocálcica. Adubação orgânica e extrato de T. voggeli são estratégias potenciais para manejar a população de ácaros e estudos adicionais são propostos para permitir ampla utilização

Strawberry fields receive several pesticides spraying and residues in fruits are recurrent. Development and application of methods that avoid applying pesticides mostly during fructification period are imminent needs. Comparison of population of two-sppoted spider mite (TSSM) Tetranychus urticae Koch. in organic and mineral fertilized plants (as used by growers) and treated with alternative products were done. Plants were grown in vessels in a greenhouse infested with local population of TSSM. Extracts of Tephrosia voggeli (Hook), neem oil (Azadirachta indica A. Juss.), and lime sulfur were applied three times against TSSM in ten days intervals. Assessments were done three times after each spraying. Interaction between fertilization and spraying alternatives was also assessed but was not significant. TSSM population was about 44% lesser in organic fertilized plants than those in inorganic fertilized. T. voggeli extracts reduced in large amounts TSSM; neem oil reduced in some assessments and an intermediate reduction was found for sulfur lime. Organic fertilization and T. voggeli extracts are potential strategies to manage TSSM and additional studies are proposed to enable wide utilization

Captures of the coffee berry borer (CBB) Hypothenemus hampei (Ferrari) were assessed in traps in the field. IAPAR designed traps [plastic bottles (2 L) lured with methanol:ethanol (1:1) in a vessel] were placed either at 0.5, 1.0 and 1.5m high from the ground or simultaneously tested in the 2004 fructification season. Traps placed at the three heights trapped 5.5 times more CBB than the others, mostly at the traps placed at 0.5 m (75%). Treatments using the IAPAR designed trap placed at 1.2 m high; IAPAR trap with a white plastic plate above (IAPAR modified I) at 1.2 m high; IAPAR at 0.5 m high and two additional vessels at 1.0 and 1.5m high (IAPAR modified II) and T-163 trap [three red plastic cups (300 ml) and a red plastic plate as a cover] lured with M:E (1:1) at 1.2m height were compared in the vegetative (2005) and fructification (2006) periods. IAPAR modified II (dispenser vessels placed at 0.5, 1.0 and 1.5 m) trapped more beetles than the remaining types (2.72 times more beetles than IAPAR design); and IAPAR modified I traps trapped more beetles than T 163 and IAPAR traps in the vegetative period. In the reproductive period, IAPAR modified II trapped less beetles than IAPAR and IAPAR modified I. In 2007 vegetative season, IAPAR modified II trap were compared with IAPAR trap and trapped 2.8 times more beetles. The positive responses to a vertical distribution of the volatile attractants in the vegetative period of the planting allow the development of more efficient trapping systems for CBB.

O caruncho-do-feijão Zabrotes subfasciatus (Coleoptera: Chrysomelidae: Bruchinae) é uma importante praga de grãos de feijão armazenado nas regiões tropicais. A eficiência do caulim [com ou sem óleo de nim (Azadirachta indica)] e terra diatomácea (TD) (tratamento padrão) foi estudada em laboratório com o intuito de obter alternativas para o controle químico deste inseto. Insetos foram confinados em frascos de plástico com feijão tratado com caulim (2, 4 e 8g kg-1), caulim + nim [2g kg-1(5% óleo de nim)], terra diatomácea (1g kg-1) e controle. Mortalidade de insetos adultos, número de ovos e emergência da geração F1 foram avaliados. Caulim causou a mortalidade de Z. subfasciatus, porém foram necessários maiores períodos e doses que a TD para promover elevada mortalidade (100% ou aproximadamente). Os tratamentos com caulim também afetaram o comportamento da fêmea, pois muitos ovos foram colocados nas paredes dos frascos. O número de adultos emergidos (F1) foi similar entre a TD e o caulim; portanto, caulim constitui uma ferramenta promissora para o manejo de Z. subfasciatus. A mistura de caulim e óleo de nim não foi eficiente no controle de Z. subfasciatus.

The Mexican bean weevil Zabrotes subfasciatus (Coleoptera: Chrysomelidae: Bruchinae) is an important pest of stored beans in tropical regions. The efficiency of kaolin [with or without neem (Azadirachta indica) oil] and diatomaceous earth (DE) (standard treatment) was studied in laboratory aiming to obtain alternatives for chemical control of this insect. Insects were confined in plastic vials containing beans treated with kaolin (2, 4 and 8g kg-1), kaolin + neem [2g kg-1(5% neem oil)], diatomaceous earth (1g kg-1) and control. Mortality of adult insects, number of eggs and F1generation beetles emergency were assessed. Kaolin caused mortality of Z. subfasciatus, however higher periods and doses than DE were necessary to promote high mortality (100% or close). Kaolin treatments also affected female behavior because many eggs were placed in the vials walls. Number of emerged adults (F1) was similar between DE and kaolin; hence, kaolin constitutes a promising tool to the management of Z. subfasciatus. The mixture of kaolin and neem oil was not efficient in the control of Z. subfasciatus.

O percevejo alidídeo Neomegalotomus parvus (Westwood) não é facilmente detectado no campo e a descoberta de atraentes pode ser uma forma adequada para monitorar a espécie. A atração de N. parvus a armadilhas (transparentes, verde transparente e amarela) iscadas com urina de vaca e amônia foi estudada em dois experimentos em campo. Armadilhas foram colocadas próximas a um terraço com plantas de feijão-guandu (Cajanus cajan L.). Água corrente, solução aquosa de NaCl 10% (p/v), urina de vaca e amônia (solução aquosa de NH4OH 1%) foram testadas como atraentes em armadilhas amarelas, transparentes e transparentes verdes foram utilizadas. Armadilhas verdes iscadas com urina capturaram mais insetos do que armadilhas amarelas e transparentes (3,2 e 7,2 vezes mais, respectivamente), e mais insetos do que armadilhas da mesma cor com apenas água (zero insetos) ou solução de NaCl (14,5 vezes mais). Armadilhas iscadas com amônia capturaram mais insetos do que armadilhas com água: 14,4; 4,7 e 6,7 vezes mais em verde transparente, amarela e transparente, respectivamente, ou solução de NaCl: 3,9; 7,6 e 6,2 vezes, respectivamente. Amônia ou urina de vaca são atrativas para N. parvus e podem ser testadas para monitorar o percevejo no campo.

The alydid bug parvus (Westwood) is not easily detected in the field and the discovery of attractants may be a suitable way to monitor this species. The attraction of N. parvus to traps (transparent, transparent green and yellow) baited with cow urine and ammonia was studied in two field trials. Traps were placed near a terrace with pigeon pea plants (Cajanus cajan L.). Tap water, NaCl 10% aqueous solution (w/v), cow urine, and ammonia (NH4OH 1% aqueous solution) were tested as attractants in yellow, transparent and transparent green traps. Green traps baited with cow urine caught more bugs than yellow and transparent traps (3.2 and 7.2 times more, respectively), and more bugs than traps of the same color with just water (no captures) or NH4OH solution (14.5 times more). Traps baited with ammonia caught more bugs than traps with water: 14.4, 4.7 and 6.7 times more than transparent green, yellow, and transparent traps, respectively, or NaCl solution: 3.9, 7.6 and 6.2 times, respectively. Both ammonia and cow urine are attractive to N. parvus and may be tested to monitor the bugs in the field.

Uma série de compostos, sintéticos e comerciais, análogos ao 1,4-dimetoxibenzeno, um cairomônio de Diabrotica speciosa, juntamente com outros compostos comerciais atrativos para outras espécies de Diabrotica, tiveram suas atividades testadas frente à D. speciosa. Armadilhas de copos amarelos contendo os compostos foram instaladas em plantação de feijão. As avaliações foram realizadas 24 horas depois. Armadilhas com 1,4-dimetoxibenzeno capturaram significativamente mais insetos do que armadilhas testemunha. Armadilhas com análogos do 1,4-dimetoxibenzeno não capturaram mais insetos que as armadilhas testemunha. Os resultados mostraram que a posição e a natureza dos substituintes, juntamente com a aromaticidade verificada na estrutura do composto natural, são cruciais para a atratividade.

Several synthetic and commercial analogs of 1,4-dimethoxybenzene, a kairomone of Diabrotica speciosa, along with other compounds already shown to be attractive to other species of Diabrotica, were tested as attractant to D. speciosa. Yellow cup traps were lured with the compounds and installed in a common bean field. Assessments were conducted 24 h later. 1,4-dimethoxybenzene lured traps caught significantly more beetles than the control traps. Captures of traps lured with 1,4-dimethoxybenzene analogs did not differ from the control traps. Results showed that position and nature of the substituents on the aromatic ring played a crucial role in the activity of the natural compound. The aromatic ring was also very important to the activity of the kairomone.

Besouros Microtheca punctigera (Achard) são pragas sérias de plantas hospedeiras da família Brassicaceae. Efeitos das concentrações (0,00; 0,25; 0,50; e 1,00%) do óleo emulsionável de nim (Azadirachta indica Meliaceae) sobre parâmetros de comportamento e biologia de M. punctigera foram investigados em laboratório. Nabiça (Raphanus raphanistrum L.) foi a planta hospedeira utilizada. Ensaios de preferência alimentar com múltipla e sem chance de escolha foram conduzidos. Múltiplos efeitos do óleo de nim foram observados. Machos foram repelidos pelo óleo de nim em teste de múltipla escolha. Adultos (múltipla escolha) e larvas (múltipla e sem chance de escolha) sofreram deterrência. A mortalidade de larvas foi mais elevada nos tratamentos com óleo de nim que no tratamento controle. Futuras investigações são sugeridas para testar o óleo de nim no manejo de pragas no campo.

The effects of concentrations (0.00, 0.25, 0.50 and 1.00%) of the neem tree (Azadirachta indica - Meliaceae) oil emulsion on the behavioral and biological parameters of M. punctigera were investigated in the laboratory. Wild radish (Raphanus raphanistrum L.) host plant was used. Multiple and no-choice feeding preference assays were conducted which shown multiple effects. The males were repelled by the neem oil in multiple-choice assay. The adult (multiple-choice) and larvae (multiple and no-choice) feeding were deterred. The larvae mortality was higher in the neem oil treated than the control leaves. Further investigations are suggested to test neem oil in the management of the pest in the field.

Diabrotica speciosa (Germar, 1824) (Coleoptera: Chrysomelidae: Galerucinae) é uma importante praga polífaga na América Latina. Seu controle é realizado com inseticidas sintéticos, o que causa problemas ambientais. Desse modo, o objetivo do trabalho foi avaliar a isca amilácea de cucurbitacina (Lagenaria vulgaris L.) associada ao inseticida carbaril no controle de D. speciosa em laboratório. Foram realizados testes de múltipla escolha pareados e confinamento com insetos adultos. Os tratamentos foram: isca amilácea de cucurbitacina, isca + carbaril, apenas carbaril e a testemunha (água destilada). As soluções foram pulverizadas em plantas de milho mantidas em casa-de-vegetação. Folhas foram retiradas para realização dos ensaios em laboratório e sete avaliações (três, cinco, sete, dez, 12, 14 e 17 dias após a aplicação) foram realizadas. Foram avaliados o consumo foliar e a mortalidade. A isca de cucurbitacina exerceu efeito fagoestimulante aos adultos de D. speciosa. A adição da isca ao carbaril aumentou a eficiência do inseticida.

Diabrotica speciosa (Germar, 1824) (Coleoptera: Chrysomelidae: Galerucinae) is an important polyphagous pest in Latin America. The control is made using synthetic insecticides, which cause environmental problems. Then, objective this work was to study the cucurbitacin (Lagenaria vulgaris L.) starch based associated with carbaril insecticide for D. speciosa control, in the laboratory. Multiple-choice and no-choice tests with adult insects were realized. The treatments were: cucurbitacin starch based bait, cucurbitacin starch based bait plus carbaril insecticide, insecticide carbaril alone and control (distillated water). Corn plants were sprayed with insecticide solutions in greenhouse and leaves were removed. Leaves were offered to insects and seven evaluations (three, five, seven, ten, 12, 14 and 17 days after applied) were achieved. Feeding leaf area and mortality were evaluated. Cucurbitacin starch based bait elicited phagostimulant effect on adults of D. speciosa. Addition of cucurbitacin starch based bait on carbaril insecticide increased insecticide efficiency.

Preferência alimentar é informação básica importante para o desenvolvimento de estratégias de manejo. Experimentos de preferência alimentar com múltipla chance de escolha foram conduzidos em laboratório para o crisomelídeo Microtheca punctigera (Acherd). A alimentação foi avaliada 72h após o início dos experimentos. Com uma larva por placa de Petri, foram utilizadas discos de folhas de almeirão, Nasturtium officinale L., rúcula, Eruca sativa L., mostarda, Brassica juncea Cosson, couve chinesa, B. pekinensis (Lour.) Rupr., e nabiça (Raphanus raphanistrum L.). A alimentação em ordem decrescente foi em couve chinesa, mostarda, nabiça, rúcula e almeirão (7,97; 1,85; 0,98; 0,36 e 0,11 mm², respectivamente). A alimentação em couve chinesa foi de 4,31 vezes maior do que em mostarda. O mesmo experimento foi repetido com um adulto por placa. Respostas de machos e fêmeas foram similares. A alimentação foi maior em mostarda (87,2 e 142,8 para machos e fêmeas, respectivamente); em rúcula (51,5 e 132,7) e couve chinesa (51,8 e 89,0) foi intermediária. Agrião (22,96 e 39,3) e nabiça (12,03 e 28,4) foram os menos consumidos. Num terceiro experimento, dez larvas por placa foram usadas, incluindo-se também espinafre, Tetragonia expansa Murr., rabanete, Raphanus sativus L., e couve, B. oleracea var. acephala L. A freqüências diárias de larvas em cada alimento foram também medidas. A alimentação foi similar em couve chinesa e mostarda (47,89 e 53,78, respectivamente). O número de insetos foi maior em mostarda, couve chinesa e nabiça. Prováveis explicações da preferência e proposições de novas pesquisas são discutidos.

Host plant feeding preference is important basic information for the development of insect management strategies. Multiple-choice feeding preference assays were conducted in the laboratory for the chrysomelid beetle, Microtheca punctigera (Achard). Feeding was assessed 72 h after onset of experiments. With one larva per Petri dish, food items comprised watercress, Nasturtium officinale L., arugula, Eruca sativa L., mustard, Brassica juncea Cosson, Chinese cabbage, B. pekinensis (Lour.) Rupr. and wild radish (Raphanus raphanistrum L.). Feeding ranking preferences were Chinese cabbage, mustard, wild radish, arugula and watercress (7.97, 1.85, 0.98, 0.36 and 0.11 mm², respectively). Feeding on Chinese cabbage was 4.31 times more intense than on mustard. The same experiment was repeated with one adult per dish. Responses of males and females were quite similar. Feeding was higher on mustard (87.2 and 142.8 for males and females, respectively). Feeding on arugula (51.5 and 132.7) and Chinese cabbage (51.8 and 89.0) were intermediate. Watercress (22.96 and 39.3) and wild radish (12.03 and 28.4) were the least preferred host plants. In a third experiment, ten larvae per dish were used and spinach, Tetragonia expansa Murr., radish, Raphanus sativus L. and collard, B. oleracea var. acephala L., were also included. Daily larval frequencies on each food were also measured. Feeding was similar on Chinese cabbage and mustard (47.89 and 53.78, respectively). Number of insects was greater on mustard, Chinese cabbage and wild radish. Probable explanations for results and proposals for further investigations are discussed.
