Resumo Beagles são menos suscetíveis ao carrapato Rhipicephalus sanguineus sensu lato devido à produção de benzaldeído e 2-hexanona. Nosso trabalho anterior já publicado mostrou que esses compostos podem reduzir a carga de carrapatos em cães suscetíveis. Aqui testamos a hipótese de que um aumento na dose destes repelentes e na taxa de liberação poderia aumentar a eficácia e a persistência do efeito repelente. As formulações de liberação lenta destes compostos, com doses e taxas de liberação mais elevadas foram testadas em cães infestados artificialmente. Dez cães foram distribuídos aleatoriamente em dois grupos com cinco cães cada. O grupo tratado recebeu colares contendo formulações de liberação lenta dos compostos, enquanto o grupo controle recebeu colares com formulações limpas. Cinco infestações ambientais foram realizadas, com o número de carrapatos (em todas as fases) nos cães sendo contados, uma vez ao dia, por 40 dias. Não se observou aumento significativo na eficácia do repelente com doses e taxas de liberação mais elevadas e, enquanto observou-se maior persistência na atividade repelente. O tratamento com as formulações resultou em uma redução de duas a três vezes no número de carrapatos dos estágios imaturos, por até três semanas. No entanto, o número de adultos foi semelhante em ambos os grupos. A perda de atividade repelente após a terceira semana de teste coincidiu com uma mudança nas taxas de liberação relativa para os dois compostos. A hipótese é que as quantidades relativas, ao invés das quantidades absolutas de liberação lenta, são importantes para a atividade repelente. Então, a hipótese é de que a repelência apresentada por hospedeiros menos susceptíveis aos carrapatos depende da percepção pelos carrapatos de misturas específicas de voláteis liberados por estes hospedeiros.

Abstract Beagles are less susceptible to Rhipicephalus sanguineus sensu lato tick due to the production of the allomones benzaldehyde and 2-hexanone. Our previous published work showed that these compounds can reduce tick burden on susceptible dogs. Here we tested the hypothesis that an increase in repellent dose and release rate could increase repellent efficacy and persistence. Slow-release formulations of these compounds, with higher doses and release rates, were tested on artificially-infested dogs. Ten dogs were randomly assigned to two groups with five dogs each. The treated group received collars with slow-release formulations of the compounds attached, while the control group received collars with clean formulations attached. Five environmental infestations were performed, with the number of ticks (at all stages) on the dogs being counted once a day for 40 days. No significant increase in repellent efficacy was observed with the higher doses and release rates, whereas a greater persistence in repellent activity was observed. Treatment with the formulations resulted in a two-to-three-fold reduction in the number of immature stage ticks for up to three weeks. However, the number of adults was similar in both groups. Loss of repellent activity after the third week of testing coincided with a marked change in the relative release rates for the two compounds. It is hypothesized that relative amounts, rather than absolute amounts, of repellent release from slow-release formulations are important for repellent activity. We also hypothesize that the avoidance of less-preferred hosts by ticks relies on olfactory-mediated perception of specific blends of volatile cues from less preferred hosts.

The toxicological impact of chemical pesticides on fungal entomopathogens and their use in tank-mixing can be directly measured through in vitro compatibility tests. This study reports the in vitro toxicity of eight insecticides, four fungicides and five herbicides in the conidial germination, vegetative growth and conidiation of Metarhizium anisopliae (strain CG 168). A conidial suspension containing the pesticide at recommended field dosage was subjected to constant agitation in a rotary shaker for 3h to simulate a tank mixing. Then, aliquots of each suspension were used to determine conidial germination, vegetative growth and conidiation on potato dextrose agar (PDA). The fungicides difenoconazole (69 mL ha-1), propiconazole (75 mL ha-1), trifloxystrobin (313 g ha-1) and azoxystrobin (56 mL ha-1) were the most harmful products to all biological stages of M. anisopliae and they should not be applied together with this fungus in tank mixing. The insecticides exhibited the least degree of toxicity to this fungal pathogen, whereas the herbicides had the greatest impact on mycelial growth. The agrochemicals compatible with M. anisopliae were the insecticides methyl parathion (240 mL ha-1), thiamethoxam (31 g ha-1), and lambda-cyhalothrin (6.3 mL ha-1) and the herbicides glyphosate (1560 mL ha-1), bentazon (720 mL ha-1), and imazapic+ imazapyr (84 g ha-1). The compatible pesticides could be simultaneously used with this bio-control agent for integrated pest management in rice production systems.

A transmissão horizontal de fungos entomopatogênicos tem papel importante na epizootiologia, pois está diretamente associada à disseminação do agente causal da doença de insetos. Em hospedeiros que apresentam restrita mobilidade, a presença de pragas secundárias no agroecossistema pode ajudar na formação de focos secundários da doença e, com isso, aumentar a mortalidade das pragas-chave. Para tanto, a disseminação de Beauveria bassiana, introduzida inoculativamente através de iscas tipo telha, foi avaliada em um bananal infestado por Cosmopolites sordidus (Piracicaba) e outro por C. sordidus e Metamasius hemipterus (Botucatu). Constatou-se que, em Piracicaba, não foram capturados insetos infectados na testemunha, sendo os índices de infecção nas parcelas que receberam B. bassiana de 8 e 15%. Em Botucatu, o índice de infecção dos insetos capturados na testemunha chegou a 17%, enquanto que, nas parcelas tratadas, variou de 28 a 54%. Os resultados obtidos neste trabalho demostram que a presença de M. hemipterus influenciou na disseminação de B. bassiana para a população de C. sordidus, praga-chave dessa cultura.

Horizontal transmission of entomopathogenic fungi plays an important role in the epizootiology, because it is directly associated with the spread of causal agents of insect diseases. In hosts with restricted mobility, the presence of secondary pests in the agroecosystem can help in the formation of secondary disease outbreaks and increase the mortality of the target pest. For this purpose, the dissemination of Beauveria bassiana inoculated using tile-type baits was evaluated in a banana plantation infested with Cosmopolites sordidus (Piracicaba) and one with C. sordidus and Metamasius hemipterus (Botucatu). In Piracicaba, insects contaminated with the pathogen were not found in the control group, and infection rates in the plots that received B. bassiana ranged between 8 and 15%. In Botucatu, the infection rate of insects in the control reached 17%, whereas in the fungal treatment plots, it varied from 28 to 54%. The present results demonstrate the influence of M. hemipterus in the dissemination of B. bassiana in populations of C. sordidus, a target pest of this crop.

Estudou-se a produção dos fungos Isaria fumosorosea (ESALQ1296) e Isaria farinosa (ESALQ1205) em fermentação líquida e sólida utilizando produtos e resíduos agroindustriais. Inicialmente, avaliaram-se combinações de substratos líquidos naturais e alternativos aos meios completo e batata-dextrose. Os meios líquidos mais promissores foram: melaço + caldo de arroz, caldo de arroz + levedura e melaço + levedura + caldo de arroz, pois resultaram nas maiores produções de propágulos viáveis. O meio melaço + caldo de arroz foi selecionado para etapa seguinte do trabalho, na qual se avaliou a produção dos fungos em grãos. A fermentação sólida foi mais eficiente quando se utilizaram os meios de farelo de soja e milho quirela para I. farinosa,e arroz inteiro e arroz quirela para I. fumosorosea. A fermentação bifásica para os fungos estudados a partir desses produtos agroindustriais selecionados proporcionou elevada produção de conídios em curto intervalo de tempo, podendo ser implantada em escala industrial.

This work investigated the production of the fungi Isaria fumosorosea and Isaria farinosa in biphasic fermentation using agro-industrial products and residues. Combinations of natural liquid substrates, alternative to the complete medium and potato dextrose medium, were evaluated. The best liquid media were sugarcane molasses + rice broth, rice broth + yeast and sugarcane molasses + yeast + rice broth, which resulted in the highest viable propagule concentration. The molasses + rice broth medium was selected for the next phase of the study in which the production of both fungal isolates was evaluated in solid grain substrates. In solid-state fermentation, the best conidia production was achieved with the soybean meal and broken corn for I. farinosa, and whole rice and broken rice for I. fumosorosea. Results demonstrated that the two fungal species could be rapidly produced with higher yield of conidia on agro-industrial resources by using biphasic fermentation techniques.
