Resumo: A recente pandemia de COVID-19 levou muitos países a adotarem políticas emergenciais de transferência de renda como forma de conter a crise econômica e humanitária. Tais iniciativas foram desenvolvidas em um contexto de agudização das desigualdades de gênero, raça, etnia e classe em decorrência de medidas de isolamento físico e social. O artigo analisa, a partir de um estudo exploratório e da chamada avaliação baseada na teoria do programa, as premissas teóricas dos programas Bolsa Família (2003), Auxílio Emergencial (2020) e Auxílio Brasil (2021) e suas respectivas dinâmicas de implementação. Na medida em que os programas de transferência de renda ocupam lugar central na agenda pública contemporânea, conclui-se que a avaliação de limites e avanços, tanto em sua concepção teórica como em relação aos mecanismos disparados em cada contexto, contribui para revelar evidências sobre sua efetividade no enfrentamento das desigualdades duráveis e daquelas que surgem em cenários de emergência sanitária.

El artículo analiza, a partir de un estudio exploratorio y de la llamada evaluación basada en la teoría del programa, los supuestos teóricos de los programas Bolsa Familia (2003), Ayuda de Emergencia (2020) y Ayuda Brasil (2021), y sus respectivas dinámicas de implementación. Dado que los programas de transferencias monetarias ocupan un lugar central en la agenda pública contemporánea, se concluye que la valoración de los límites y avances, tanto en su concepción teórica como en relación con los mecanismos desencadenados en cada contexto, ayuda a revelar evidencias sobre su efectividad para enfrentar las desigualdades duraderas y las que surgen en escenarios de emergencia sanitaria.

ABSTRACT OBJECTIVE To evaluate the quality of information on gestational syphilis (GS) and congenital syphilis (CS) on the Sistema de Informação de Agravos de Notificação (SINAN-Syphilis Brazil – Notifiable Diseases Information System) by compiling and validating completeness indicators between 2007 and 2018. METHODS Overall, care, and socioeconomic completeness scores were compiled based on selected variables, by using ad hoc weights assigned by experts. The completeness scores were analysed, considering the region and area of residence, the pregnant woman’s race/colour, and the year of case notification. Pearson’s correlation coefficients were used to validate the scores obtained by the weighted average method, compared with the values obtained by principal component analysis (PCA). RESULTS Most selected variables presented a good or excellent degree of completeness for GS and CS, except for clinical classification, pregnant woman’s level of education, partner’s treatment, and child’s race/colour, which were classified as poor or very poor. The overall (89.93% versus 89.69%) and socioeconomic (88.71% versus 88.24%) completeness scores for GS and CS, respectively, were classified as regular, whereas the care score (GS-90.88%, and CS-90.72%) was good, despite improvements over time. Differences in the overall, care and socioeconomic completeness scores according to region, area of residence, and ethnic-racial groups were reported for syphilis notifications. The completeness scores estimated by the weighted average method and PCA showed a strong linear correlation (> 0.90). CONCLUSION The completeness of GS and CS notifications has been improving in recent years, highlighting the variables that form the care score, compared with the socioeconomic scores, despite differences between regions, area of residence, and ethnic-racial groups. The weighted average was a viable methodological alternative easily operationalised to estimate data completeness scores, allowing routine monitoring of the completeness of gestational and congenital syphilis records.

ABSTRACT Objective: To report the experience of creating an online extension course, focusing on Advanced Nursing Practice in the promotion ofchild continence. Method: An experience report on the creation of a course for nursing students, carried out in the second half of 2021 at a federal university in Brazil. It was based on the Meaningful Learning Theory, Instructional Design, and on the Digital Storytelling strategy. Results: The course was planned to be offered online, including knowledge about childhood continence, Advanced Nursing Practice, urinary and intestinal symptoms, and nurses’ work in the context of pediatric urology. Final considerations: The authors proposed an innovative online course based on their experience to promote the teaching of child urological care in nursing education.

RESUMEN Objetivo: Relatar la experiencia de creación de un curso de extensión online, cuyo foco es la Práctica Avanzada de Enfermería en la promoción de la continencia infantil. Método: Informe de experiencia sobre la creación de un curso para estudiantes de enfermería, realizado en el segundo semestre de 2021 en una universidad federal de Brasil. Se basó en la Teoría del Aprendizaje Significativo, en el Diseño Instruccional, y en la estrategia de Digital Storytelling. Resultados: El curso fue planificado para ser ofrecido online, incluyendo conocimiento sobre la continencia infantil, la Práctica Avanzada de Enfermería, los síntomas urinarios e intestinales, y la actuación de los enfermeros en el contexto de la uropediatría. Consideraciones finales: Se propuse un curso innovador basado en la experiencia de los autores, con el objetivo de promover la enseñanza del cuidado urológico infantil en la formación de enfermeros.

Objectives: Gene-environment interactions increase the risk of psychosis. The objective of this study was to investigate gene-gene and gene-environment interactions in psychosis, including single nucleotide variants (SNVs) of dopamine-2 receptor (D2R), N-methyl-d-aspartate receptor (NMDAR), and cannabinoid receptor type 1 (CB1R), lifetime cannabis use, and childhood trauma. Methods: Twenty-three SNVs of genes encoding D2R (DRD2: rs1799978, rs7131056, rs6275), NMDAR (GRIN1: rs4880213, rs11146020; GRIN2A: rs1420040, rs11866328; GRIN2B: rs890, rs2098469, rs7298664), and CB1R (CNR1: rs806380, rs806379, rs1049353, rs6454674, rs1535255, rs2023239, rs12720071, rs6928499, rs806374, rs7766029, rs806378, rs10485170, rs9450898) were genotyped in 143 first-episode psychosis patients (FEPp) and 286 community-based controls by Illumina HumanCoreExome-24 BeadChip. Gene-gene and gene-environment associations were assessed using nonparametric Multifactor Dimensionality Reduction software. Results: Single-locus analyses among the 23 SNVs for psychosis and gene-gene interactions were not significant (p > 0.05 for all comparisons); however, both environmental risk factors showed an association with psychosis (p < 0.001). Moreover, gene-environment interactions were significant for an SNV in CNR1 and cannabis use. The best-performing model was the combination of CNR1 rs12720071 and lifetime cannabis use (p < 0.001), suggesting an increased risk of psychosis. Conclusion: Our study supports the hypothesis of gene-environment interactions for psychosis involving T-allele carriers of CNR1 SNVs, childhood trauma, and cannabis use.

ABSTRACT Objective: to analyze the spatial distribution of violence against older adults in the Brazilian South region before and in the first year of the COVID-19 pandemic. Method: an ecological study that analyzed reports of violence against older adults between 2019 and 2020, in Southern Brazil. Local empirical Bayesian rates, Moran’s Local Autocorrelation Index and Getis Ord Gi analysis were calculated. Results: there was a reduction in the smoothed rate of violence against older adults. The high-high and hot-spot cities in 2019 became more violent in 2020; and cities with low prevalence became less violent in 2020. The highest COVID-19 fatality rates were also in those cities with the highest violence rates. Conclusion: the data reinforces the complexity of violence and its worsening due to the pandemic. In addition, they contribute to decision-making in Nursing, highlighting the need to encourage new research studies on this theme.

RESUMEN Objetivo: analizar la distribución espacial de la violencia contra las personas mayores en la región sur de Brasil antes y durante el primer año de la pandemia de COVID-19. Método: estudio ecológico que analizó denuncias de violencia contra las personas mayores, entre 2019 y 2020, en la región sur de Brasil. Se calcularon las tasas bayesianas empíricas locales, el índice de autocorrelación local de Moran y el análisis Getis Ord Gi. Resultados: hubo reducción en la tasa suavizada de violencia contra las personas mayores. Las ciudades alto-alto y hotspot de 2019 se volvieron más violentas en 2020; y las ciudades con baja prevalencia se volvieron menos violentas en 2020. Las tasas de mortalidad por COVID-19 más altas también se registraron en las ciudades con mayores tasas de violencia. Conclusión: los datos confirman la complejidad de la violencia y el agravamiento debido a la pandemia; además contribuyen a la toma de decisiones en enfermería y destacan la necesidad de fomentar nuevas investigaciones sobre este tema.

RESUMO: Objetivo: analisar a distribuição espacial da violência contra a pessoa idosa na região Sul do Brasil antes e no primeiro ano de vigência da pandemia de COVID-19. Método: estudo ecológico que analisou as notificações de violência contra o idoso, entre 2019 e 2020, no Sul do Brasil. Foram calculadas as taxas bayesianas empíricas locais, o Índice de Autocorrelação Local de Moran e a análise de Getis Ord Gi. Resultados: houve redução da taxa suavizada de violência contra o idoso. As cidades alto-alto e hot-spot em 2019, se tornaram mais violentas em 2020; e as cidades com baixa prevalência, se tornaram menos violentas em 2020. As maiores taxas de letalidade por COVID-19 também foram naquelas cidades com maiores taxas de violência. Conclusão: os dados reforçam a complexidade da violência e seu agravamento pela pandemia. E contribuem com as tomadas de decisões na enfermagem, desatacando-se a necessidade de suscitar novas pesquisas sobre esta temática.

Abstract Background RNA extraction is a step that precedes several molecular techniques. The fibrous tissue, more specifically the dura mater, has several limitations in routine protocols, and lacks optimization protocols to overcome these problems. Objective To test stock reagents and purification kits, optimizing commercial kit protocols for RNA extraction from the dura mater. Methods Dura mater samples were obtained from eight Wistar rats and maintained in two different stabilizers. The samples were purified using four different protocols, and the RNA was evaluated for the yield and purity in NanoDrop 2000 (Thermo Scientific, Wilmington, DE, United States). Beta-actin gene was used for analyzing gene expression, since is one of the most used reference genes. Results The RNA preservation was similar in both stabilizers. The addition of an incubation step prior the purification protocols allowed better tissue digestion and RNA recovery. The RNA purified using the protocols membrane-based showed higher quality than liquid-liquid purification. This impact was observed in the 3-week evaluation using RT-qPCR. Conclusion Stabilizers are efficient for RNA preservation and membrane-based purification protocols are more suitable for RNA recovery from dura mater tissue, allowing the evaluation of gene expression in this type of tissue. Adaptations in the dura mater RNA extraction protocol differ from the pre-established protocols because it takes into account the peculiarity of fibrous tissue and low cellularity. In addition to providing a low-cost mechanism, based on techniques that are part of the laboratory routine, it is possible to improve the quality of the extracted material, ensuring greater efficiency in the use of subsequent techniques.

Resumo Antecedentes A extração de RNA é uma etapa que antecede várias técnicas moleculares. O tecido fibroso, mais especificamente a dura-máter, apresenta várias limitações nos protocolos de rotina e carece de protocolos de otimização para superar estes problemas. Objetivo Testar reagentes de estoque e kits de purificação, otimizando protocolos de kits comerciais para extração de RNA da dura-máter. Métodos Amostras de dura-máter foram obtidas de oito ratos Wistar e mantidas em dois estabilizadores diferentes. As amostras foram purificadas em quatro protocolos diferentes e o RNA foi avaliado quanto ao rendimento e pureza no NanoDrop 2000 (Thermo Scientific, Wilmington, DE, United States). O gene da beta-actina foi utilizado para analisar a expressão gênica, uma vez que é um dos genes de referência mais utilizados. Resultados A preservação do RNA foi semelhante em ambos os estabilizadores. A adição de uma etapa de incubação antes dos protocolos de purificação permitiu uma melhor digestão do tecido e recuperação de RNA. O RNA purificado pelos protocolos baseados em membrana apresentou qualidade superior ao da purificação líquido-líquido. Este impacto foi observado na avaliação de três semanas usando RT-qPCR. Conclusão Os estabilizadores são eficientes para preservação do RNA e os protocolos de purificação baseados em membrana são mais adequados para recuperação de RNA do tecido da dura-máter, permitindo a avaliação da expressão gênica neste tipo de tecido. As adaptações no protocolo de extração de RNA da dura-máter diferem dos protocolos preestabelecidos porque leva em consideração a peculiaridade do tecido fibroso e com baixa celularidade. Além de fornecer um mecanismo de baixo custo, baseado em técnicas que fazem parte da rotina laboratorial, é possível melhorar a qualidade do material extraído, garantindo maior eficácia no uso de técnicas subsequentes.

ABSTRACT Introduction: Systemic Mastocytosis comprises a group of neoplastic diseases characterized by clonal expansion and infiltration of mast cells into several organs. The diagnosis and treatment of this disease may be challenging for non-specialists. Objective: Make suggestions or recommendations in Systemic Mastocytosis based in a panel of Brazilian specialists. Method and results: An online expert panel with 18 multidisciplinary specialists was convened to propose recommendations on the diagnosis and treatment of Systemic Mastocytosis in Brazil. Recommendations were based on discussions of topics and multiple-choice questions and were graded using the Oxford Centre for Evidence-Based Medicine 2011 Levels of Evidence Chart. Conclusion: Twenty-two recommendations or suggestions were proposed based on a literature review and graded according to the findings.

ABSTRACT Background: Some studies have suggested the importance of vitamin D [25(OH)D] in malaria clinical practice. The prevalence of 25(OH)D deficiency in the Amazon population is not well known, and there are few studies in patients with malaria. This study aimed to evaluate 25(OH)D serum levels in patients with malaria and determine their relationships with epidemiological, clinical, laboratory, and parasitemia data. Methods: An analytical cross-sectional study of 123 patients with malaria and 122 individuals without malaria was performed in Itaituba, Pará, Brazil, from January 2018 to October 2019, by evaluating sociodemographic, clinical-epidemiological, parasitological, and laboratory data and adopting a 5% significance level. Parametric tests (Student's t-test), non-parametric tests (Mann-Whitney U), and Spearman’s correlation ([rs], for non-parametric variables) were used according to the nature of the distribution of the variables. For the qualitative variables, Pearson's chi-square test, Fisher's exact test, and the G test were used. Spearman's correlation was used to compare the results of the 25(OH)D levels and blood counts performed among patients and the control group. Results: Malaria patients residing in a mining area had 25(OH)D serum levels that were significantly lower than those in the control group residing in the mining area, though both were within normal levels. Red blood cell counts had an inverse correlation with parasitemia (Plasmodium falciparum), and platelet levels had an inverse correlation with parasitemia (Plasmodium vivax). 25(OH)D deficiency was evidenced in Itaituba, in the state of Pará, which is an endemic area of malaria in the Amazon region.

Abstract The combination of flours from cereal and legume results in less caloric products, with high contents of fiber, proteins, ash and antioxidants. Thus, this study aimed at developing muffins made with different proportions of wholemeal rye flour (WRF) and wholemeal common black bean flour (WBF). Physicochemical and physical analyses were performed to characterize starches, flours and the muffins. Sensory analysis was also carried out. WRF had the highest values for L* value, moisture and ash contents, whereas WBF had more proteins and lipids, and heterogeneous particle size distribution. The formulation with the highest acceptability grade with total replacement of whole wheat flour, was that containing 70% WRF and 30% WBF (F2). This sample was characterized as high-quality muffin, chocolate flavor, chocolate odor, light and soft crumb. Its texture was defined as low hardness, chewiness and high cohesiveness. It was concluded that the F2 formulation had a high dietary fiber content (20%), low lipid content (8%), containing 32% resistant starch, 9% protein and 51.43 mg GAE 1001 g-1 of total phenolic content, being considered an excellent nutritional food option.

ABSTRACT Purpose: to describe the anthropometric parameters of children diagnosed with pediatric feeding disorder by a multiprofessional team. Methods: a retrospective clinical study conducted on 196 children on maternal complaints of feeding difficulties and a diagnosis of pediatric feeding disorder. Children with neurological diseases or having no pediatric feeding disorder diagnosis were excluded from the study. Weight and height data were obtained and parameters such as weight-for-age, length/height-for-age and body mass index for-age were analyzed, according to the recommendations of the World Health Organization. The two-proportion equality test was used and the level of significance was set at 0.05 (5%) in all analyses. Results: results showed mean (±Standard Deviation) values of 13.09±5.1 for weight (kg), 0.61±15.53 for height (cm) and 15.37±1.56 for body mass index (kg/m²). The results also showed that 87.2% of the children were within the adequate z-score for weight-for-age, 93.4% were within the adequate z-score for length/height-for-age, and that 88.8% had a normal z-score for-age for body mass index. Conclusion: the sample of children diagnosed with pediatric feeding disorder exhibited adequate anthropometric parameters for their age range.

Resumo O objetivo deste trabalho foi identificar locos de caracteres quantitativos (QTLs) associados à produtividade em uma população segregante de arroz (GYP). Uma população de 245 linhagens puras recombinantes de arroz, do cruzamento 'Epagri 108' (Oryza sativa subsp. indica) x 'IRAT 122' (O. sativa subsp. japonica), foi avaliada em diferentes locais e anos e genotipada por marcadores de polimorfismo de nucleotídeo único (SNPs). Obteve-se um mapa de 1.592,8 cM a partir de 9.831 SNPs, tendo-se identificado 25 QTLs. Os seguintes nove SNPs apresentaram estabilidade entre os diferentes ambientes: M1.37719614 e M6.9563117 para GYP; M4.29340056, M5.25588710, M7.29115624 e M12.4534450 para peso de 100 grãos (HGW); e M1.38398157, M4.28368337 e M7.25991230 para altura de plantas. Seis SNPs não estavam presentes nos blocos de ligação: M6.9563117 e M4.1077080 para GYP; M5.25588710 e M6.8886398 para HGW; e M2.34471005 e M8.5955948 para altura de plantas. Os SNPs M6.9563117 e M5.25588710 foram considerados ambientalmente estáveis e não estiveram presentes em blocos de ligação, o que indica seu alto potencial para uso na seleção assistida por marcadores de produtividade de grãos, em programas brasileiros de melhoramento de arroz.

Abstract The objective of this work was to identify the quantitative trait loci (QTLs) associated with grain yield in a rice segregant population (GYP). A population of 245 inbred recombinant rice lines from the 'Epagri 108' (Oryza sativa subsp. indica) x 'IRAT 122' (O. sativa subsp. japonica) cross was evaluated at different locations and years and genotyped by single nucletide polymorphism (SNP) markers. A map of 1,592.8 cM was obtained from 9,831 SNPs, identifying 25 QTLs. The following nine SNPs showed stability between the different environments: M1.37719614 and M6.9563117 for GYP; M4.29340056, M5.25588710, M7.29115624, and M12.4534450 for 100-grain weight (HGW); and M1.38398157, M4.28368337, and M7.25991230 for plant height (PH). Six SNPs were not present in the linkage blocks: M6.9563117 and M4.1077080 for GYP; M5.25588710 and M6.8886398 for HGW; and M2.34471005 and M8.5955948 for PH. The M6.9563117 and M5.25588710 SNPs were considered environmentally stable and were not present in the linkage blocks, showing their high potential for use in marker-assisted selection for grain yield in Brazilian rice breeding programs.
