In situ and in vitro degradation parameters of elephant grass silage with sugarcane bagasse

The objective of this study was to evaluate the chemical composition and in situ degradability of elephant grass silage dry matter with increasing levels of sugarcane bagasse. A completely randomized design was adopted with five treatments: 0, 5, 10, 15, and 20% inclusion of sugarcane bagasse on an as-fed matter basis, with five replications, totalling 25 experimental units. The increasing proportion of sugarcane bagasse analysed was favourable only in increasing the concentration of dry matter and fibre of silages and in maintaining a considerable crude protein content. In the in situ degradation assay, the addition of sugarcane bagasse in silage decreased the ruminal degradability of dry matter, and as the passage rates increased, the effective degradability decreased. In the in vitro gas production assay, the total production of gases was decreased with the inclusion of sugarcane bagasse, as did the degradability of organic matter and neutral detergent fibre. The recommendation for the inclusion of sugarcane bagasse should be analysed according to the desired objective, emphasizing that values higher than 5% can greatly compromise the final nutritive value of elephant grass silage.

Abstract In the current study, two euglossine species, Exaerete smaragdina and Eulaema nigrita, a cleptoparasite bee and its host, respectively, were used as models to: (i) access the genetic diversity and population structure of both species, sampled along a wide latitudinal range of Atlantic Forest, where the distribution of El. nigrita and Ex. smaragdina co-occurs; (ii) investigate the evolutionary history of these species through the Atlantic Forest, and in a wider scenario, to examine the evolutionary history of these species across others forest domains. Analyses involved males of El. nigrita and Ex. smaragdina sampled through Brazilian territory, including 19 sites in the Atlantic Forest. Bayesian Skyline Plot (BSP) was used to infer possible climate oscillations on population of both species over time. The BSP revealed stability in effective population size for both species in most of the Plio-Pleistocene period. However, BSP results aligned to the starlike configuration in the haplotype network, neutrality test, and population diversity patterns indicated population expansion of the two species during the late Pleistocene. Our findings suggest areas of potential refugia to the climatic oscillations of the Pleistocene in the Atlantic Forest in the Brazilian states of Espírito Santo for El. nigrita and Pernambuco for Ex. smaragdina.

Uterine secretory activity and histology of prepubertal female dogs in an animal model of partial ablation of endometrial glands development

RESUMO: A exposição inadequada do sistema reprodutor feminino a esteróides em períodos do desenvolvimento uterino pode inibir parcialmente o desenvolvimento das glândulas endometriais em cães. Entretanto, não se conhece os efeitos dos esteróides sobre a funcionalidade das glândulas formadas, bem como a possível ocorrência de fibrose endometrial. Objetivou-se avaliar a atividade secretória das glândulas endometriais de cadelas pré-púberes submetidas a protocolo de ablação parcial da adenogênese uterina. Foram utilizadas 16 fêmeas, sem-raça-definida, distribuídas nos grupos MPA (n=8), fêmeas que receberam aplicações de acetato de medroxiprogesterona a cada 3 semanas, e C (n=8), fêmeas controle não tratadas. Aos seis meses de idade, foi realizada ovariohisterectomia em todos os animais, e avaliados os cornos uterinos pelo exame histológico e de histoquímica. Para avaliar a intensidade de secreção (graus 1-4), foram utilizadas periodic acid-Schiff e alcian blue (AB) pH 2,5. Para a avaliação histológica foram utilizados tricrômico de Masson e azul de toluidina. Apenas marcações graus 1 e 2 foram observadas para PAS em ambos os grupos, sem diferença na intensidade de secreção uterina entre grupos com relação aos graus encontrados. Entretanto, o grupo MPA apresentou maior intensidade de secreção uterina em relação ao grupo C (p<0,05). Com relação ao AB pH 2,5, em ambos os grupos também foram encontradas apenas marcações de graus 1 e 2, sem diferença estatística entre grupos. Não foram observadas marcações para a coloração de tricrômico de Masson na região periglandular, em ambos os grupos. Foi observada maior quantidade de mastócitos presentes no útero na região do miométrio, em ambos os grupos. Conclui-se que cadelas pré-púberes com ablação parcial da adenogênese uterina apresentam mínima atividade secretória uterina, ausência de fibrose periglandular e maior presença de mastócitos no miométrio em relação ao endométrio.
ABSTRACT: Inadequate exposure of the female reproductive system to steroids in uterine developmental periods can partially inhibit the development of endometrial glands in dogs. However, the effects of steroids on the formed glands functionality remain unknown, as well as the possible occurrence of endometrial fibrosis. This study aimed to evaluate the secretory activity of endometrial glands in prebubertal female dogs submitted to a protocol of partial ablation of the uterine adenogenesis. Sixteen females of non-specific breed were distributed into two groups; MPA (n=8), females that received applications of medroxyprogesterone acetate every 3 weeks; and C (n=8) untreated control females. Ovariohysterectomy was performed in all animals at the age of 6 months and evaluated the uterine horns by histological and histochemistry exams. The secretion intensity (degrees 1-4) was evaluated using periodic acid-Schiff (PAS) and alcian blue (AB) pH 2.5. Histological evaluation was performed using Masson’s trichrome and toluidine blue. Only degree 1 and 2 marks for PAS were observed in both groups, with no difference of uterine secretion intensity between the groups regarding the degrees found. However, the MPA group revealed higher intensity of uterine secretion compared to group C (p<0.05). Staining with AB pH 2.5 also revealed only degree 1 and 2 marks in both groups, with no statistically significance between them. Masson’s trichrome staining revealed no marks in the periglandular region in both groups. A higher among of mast cells was observed in the myometrial region of the uterus in both groups. Prepubertal female dogs with partial ablation of the uterine adenogenesis present minimal uterine secretory activity, absence of periglandular fibrosis and increased presence of mast cells in the myometrium compared to endometrium.

RESUMO As gestações múltiplas em humanos correspondem a apenas 3% das gestações, sendo 97-98% dessas gestações gemelares. Sabe-se que a morbimortalidade fetal é maior em gêmeos monocoriônicos do que nos dicoriônicos. A espécie canina é naturalmente multípara, mas o diagnóstico gestacional de gêmeos monocoriônicos não é comum. O objetivo deste relato é descrever o diagnóstico ultrassonográfico de gêmeos monocoriônicos em duas cadelas, sendo uma da raça Pug (caso 1) e outra da raça Shih-Tzu (caso 2). Foi possível verificar a presença de uma única vesícula gestacional contendo dois fetos em cada cadela, por meio da visibilização de duas cabeças ou de dois corpos dentro de uma mesma placenta. Esses fetos apresentavam viabilidade e organogênese adequadas e o grau de desenvolvimento era similar aos demais fetos da ninhada. No caso 1, os gêmeos nasceram mortos e de tamanho menor que os outros cinco fetos nascidos vivos. Os gêmeos do caso 2 nasceram vivos, mas também eram pequenos em relação aos irmãos da ninhada. Os riscos gestacionais associados a essa condição em cadelas ainda não são conhecidos, no entanto já existem relatos de morte fetal em gestação monocoriônica nessa espécie. Portanto, o exame ultrassonográfico durante a gestação permite um diagnóstico precoce da monocorionicidade, e a monitorização da viabilidade fetal pode trazer benefícios para a saúde da matriz, assim como para o restante da ninhada.
ABSTRACT Multiple pregnancies in humans account for only 3% of pregnancies, 97-98% of which are twin pregnancies and the morbimortality is higher in the monochorionic twins when compared to dichorionic ones. The canine species is naturally multiparous, but the diagnosis of monochorionic twin pregnancy is not common. The objective of this report was to describe the ultrasonographic diagnosis of monochorionic twin pregnancies in two bitches [Pug (case 1) and Shih tzu (case 2)]. It was possible to verify the presence of one gestational vesicle containing two fetuses in each female by observing two heads or two bodies within the same placental site. These fetuses presented adequate viability and normal organogenesis. Their development was similar to the other fetuses. In case 1 they were stillborn and smaller than the other five live-born fetuses. The twins in case 2 were born alive, but they also appeared smaller when compared to the littermates. The gestational risks associated with this condition in pregnant bitches are still unknown, however, there are reports of fetal death in monochorionic pregnancies in this species. Therefore, ultrasonographic exam during pregnancy allows an early monochorionic diagnosis and monitoring the fetal viability could bring health benefits to both the female and the littermates.

Ropivacaína isolada ou associada à metadona ou à morfina, pela via epidural, em cadelas submetidas à ovário-histerectomia

RESUMO Objetivou-se avaliar os efeitos fisiológicos, sedativos e analgésicos da administração peridural de ropivacaína isolada ou associada à morfina ou à metadona. Para tal, 24 cadelas submetidas à ovário-histerectomia receberam acepromazina, e a anestesia foi induzida e mantida com propofol e isoflurano (FiO2 = 1,0), respectivamente. De acordo com o protocolo peridural, formaram-se três grupos de igual número: GR (ropivacaína - 2,0mg/kg); GRMETA (ropivacaína - 2,0mg/kg e metadona - 0,3mg/kg) e GRMORF (ropivacaína - 2,0mg/kg e morfina - 0,1mg/kg). Registraram-se os parâmetros fisiológicos intraoperatórios e os graus de sedação e analgesia pós-operatórios. No GR constataram-se maiores médias de pressões arteriais 30 minutos após a anestesia epidural em relação ao GRMETA (sistólica e média) e ao final do procedimento cirúrgico comparativamente ao GRMORF (sistólica, diastólica e média). Não foram observadas diferenças significativas entre os grupos relativamente à analgesia e ao grau de sedação pós-operatórios. A administração epidural de ropivacaína é segura e eficaz e proporciona boa analgesia, independentemente da sua associação com morfina ou metadona.
ABSTRACT The aim of this study was to evaluate the physiological, sedative and analgesic effects of epidural administration of ropivacaine sole or associated to morphine or methadone. Twenty-four bitches were submitted to ovariohysterectomy and received acepromazine and after, propofol and isoflurane (FiO 2 = 1.0) for anesthesia induction and maintenance, respectively. Based on established epidural protocol (L7-S1), three groups were formed: GR (ropivacaine - 2.0mg/kg); GRMETA (ropivacaine - 2.0mg/kg and methadone - 0.3mg/kg) and GRMORF (ropivacaine - 2.0mg/kg and morphine - 0.1mg/kg). Intraoperative physiological parameters and degrees of postoperative sedation and analgesia were recorded. In the GR, the means of arterial pressures, 30 minutes after epidural anesthesia, were higher compared with GRMETA (systolic and mean) and, at the end of the clinical procedure, compared to GRMORF (systolic, diastolic and mean). Differences between groups were not observed for postoperative analgesia and degree of sedation. Epidural administration of ropivacaine is safe and effective and provides good analgesia regardless of its association with morphine or methadone.

RESUMO: A caprinocultura na região Nordeste do Brasil desempenha importante função socioeconômica e estratégica. A rusticidade desta atividade nesta região, aliada a sua rápida expansão em outros estados, tem gerado perdas na sua cadeia produtiva a nível regional e nacional, causadas por doenças infecciosas, destacando-se a Linfadenite Caseosa (LC), afecção amplamente difundida nos rebanhos caprinos, acarretando sérios prejuízos econômicos à atividade. Embora LC tenha sido detectada como um problema endêmico no nordeste brasileiro, uma análise abrangente e mais recente desta prevalência nos rebanhos caprinos brasileiros se faz necessária. O objetivo desse estudo foi determinar a frequência de anticorpos anti-Corynebacterium pseudotuberculosis em cinco dos nove estados que compõem a região Nordeste do Brasil. Foram processadas amostras de soro de 2571 caprinos provenientes de 218 propriedades rurais oriundas de cinco estados da região Nordeste do Brasil, coletadas entre os anos de 2010 a 2012. O diagnóstico da infecção por C. pseudotuberculosis foi realizado pela técnica de ELISA-indireto. Em 88,5% (193/218) das propriedades investigadas, pelo menos um caprino foi soropositivo para C. pseudotuberculosis, sugerindo que o agente se encontra disseminado nos rebanhos do Nordeste, com a maior prevalência encontrada no Rio Grande do Norte (94,5%) e a menor no estado de Sergipe (70,3%). Foram identificados 783 (30,4%; IC 95%=28,7-32,2%) caprinos soropositivos, com a maior prevalência entre animais encontrada no Piauí (41,4%) e a menor no estado de Sergipe (22,5%). De um total de 279 reprodutores avaliados, 106 (37,9%) resultaram soropositivos, destacando-se o Rio Grande do Norte, com 45,30%, e Piauí com 46,8% dos reprodutores positivos para LC. Das 1420 matrizes testadas, 599(42,1%) apresentaram sorologia positiva para C. pseudotuberculosis. Entre os estados esta distribuição também se manteve semelhante, destacando-se o Rio Grande do Norte, com 47,5% e Piauí com 59,5% das matrizes positivos para LC. Entre os 872 caprinos jovens avaliados, 78 (8,9%) foram soropositivos para LC, observando-se uma diferença estatística na frequência de sopositividade obtidas entre os adultos e os jovens (P<0,001). Reforça-se a necessidade do diagnóstico da enfermidade em rebanhos caprinos da região Nordeste para possível implementação de programas de controle da doença e medidas mais precisas no manejo da LC junto aos criadores de caprinos.
ABSTRACT: Goat farming in the Northeast region of Brazil plays an important socioeconomic and strategic role. The rusticity of this activity in this region, allied with its rapid expansion in other states, has caused losses in its production chain in regional and national levels, caused by infectious diseases, especially caseous lymphadenitis (CL), a widespread goat herds disease which has been causing serious economic loss to this activity. Although CL has been detected as an endemic problem in the Brazilian Northeast, a comprehensive and more recent analysis of this prevalence in Brazilian goats is necessary. The objective of this study was to determine the frequency of anti-Corynebacterium pseudotuberculosis antibodies in five of nine states of the northeast region of Brazil. Serum samples were collected from 2571 goats from 218 farms in five states in the Northeast region of Brazil, collected between 2010 and 2012. The diagnosis of C. pseudotuberculosis infection was made using the ELISA-indirect technique. In 88.5% (193/218) of the investigated properties, at least one goat was seropositive for C. pseudotuberculosis, suggesting that the agent is widespread in Northeast herds, with the highest prevalence found in Rio Grande do Norte (94.5%) and the lowest one in the state of Sergipe (70.3%). A total of 783 (30.45%; 95% CI = 28.71-32.26%) seropositive goats were found, and the highest prevalence among animals was found in Piauí (41.4%) and the lowest in the state of Sergipe (22.5%). Out of a total of 279 breeding herds, 106 (37.99%) were seropositive, standing out the Rio Grande do Norte, with a total of 45.30%, and Piauí 46.8% of positive breeding for CL. A total of the 1420 tested matrices, 599 (42.1%) presented positive serology for C. pseudotuberculosis. Among the States, this distribution also remained similar, standing out Rio Grande do Norte with 47.5%, and Piauí with 59.5% of positive matrices for CL. Among 872 young goats evaluated, 78 (8.9%) were seropositive for CL, observing a statistical difference in the frequency of soropositivity obtained between adults and young adults (P<0.001). The necessity to diagnose the disease in goat herds in the Northeast region is reinforced for the possible implementation of disease control programs and more precise measures to manage CL along with goat farmers.

Effect of Glyphosate on Guineagrass Submitted to Different Soil Water Potential

RESUMO: A ação de herbicidas sobre plantas daninhas que se desenvolvem sob déficit hídrico pode ser comprometida, pois as rotas de penetração de herbicidas hidrofílicos são reduzidas em razão da menor hidratação da cutícula nessas plantas. Além disso, compostos hidrofóbicos presentes na camada de cera epicuticular em plantas estressadas hidricamente dificultam sobremaneira a penetração de compostos hidrofílicos. Avaliou-se neste estudo a eficiência de controle do herbicida glyphosate em plantas de capim-colonião quando submetidas a distintos déficits hídricos. O estudo foi conduzido em casa de vegetação, sendo os tratamentos compostos por plantas de capim-colonião submetidas a três condições hídricas do solo [baixo déficit hídrico (13%), déficit hídrico intermediário (10%) e alta restrição hídrica (8%)], três doses do herbicida glyphosate (0,0, 270,0 e 540,0 g e.a. ha-1) e dois estádios fenológicos de desenvolvimento da planta (4-6 folhas e 1-3 perfilhos). O manejo hídrico teve início quando as plantas apresentaram duas folhas desenvolvidas. Foram realizadas avaliações visuais de controle aos 7, 14, 21 e 35 dias após a aplicação do herbicida; analisaram-se ainda os parâmetros morfofisiológicos área foliar específica, condutância estomática e a diferença de temperatura do ambiente com a temperatura foliar no dia da aplicação do herbicida, bem como a matéria seca da parte aérea e da raiz ao final do estudo. Com o aumento da restrição hídrica, houve decréscimo nos parâmetros morfofisiológicos analisados, bem como no acúmulo de matéria seca da parte aérea e das raízes das plantas estudadas. O controle das plantas foi mais eficiente quando ocorreu a aplicação da dose de 540 g ha-1 de glyphosate, quando elas foram controladas no estádio vegetativo de 1-3 perfilhos, e no manejo hídrico de 13%. Pode-se afirmar que plantas de capim-colonião sob restrição hídrica têm menor eficácia de controle quando tratadas com glyphosate.
ABSTRACT: The action of herbicides on weeds that develop under water deficit can be compromised, because the routes of penetration of hydrophilic herbicides are reduced due to the lower hydration of the cuticle in these plants. Moreover, hydrophobic compounds found in the epicuticular wax coating of plants under water stress, hinder the penetration of hydrophilic compounds. This study evaluated the control efficiency of glyphosate on guineagrass plants when submitted to different water deficits. The study was conducted in a greenhouse, and treatments were composed of guineagrass plants submitted to three soil water conditions [low water deficit (13%), intermediate water deficit (10%) and high water restriction (8%)], three doses of glyphosate (0.0, 270.0 and 540.0 g ha-1) and two phenological stages of plant development (4-6 leaves and 1-3 tillers). The water management started when plants presented two developed leaves. Visual evaluations were performed 7, 14, 21 and 35 days after the application of the herbicide; the morpho-physiological parameters of the specific leaf area, stomatal conductance and the difference between the environment temperature and the leaf temperature on the day of the herbicide application were also analyzed at the end of the study, as well as the dry matter of shoot and root. With the increase of the water restriction, there was a decrease in the analyzed morpho-physiological parameters, as well as in the dry matter accumulation of the shoot and roots of the studied plants. Plant control was more efficient when a 540 g ha-1 dose of glyphosate was applied, and when they were controlled at their vegetative stage of 1-3 tiller, and with a water management of 13%. It is possible to state that guineagrass under water restriction have less control efficacy when treated with glyphosate.

Ocorrência de carrapatos em Tamandua tetradactyla (Linnaeus, 1758) na Caatinga, Nordeste do Brasil

RESUMO A Caatinga é um bioma exclusivamente brasileiro e um dos mais diversificados do mundo, abrigando grande variedade faunística. Este estudo objetivou relatar a presença de carrapatos em tamanduás Tamandua tetradactyla (Linnaeus, 1758) atendidos no Centro de Conservação e Manejo de Fauna da Caatinga, Petrolina-PE, Brasil. De fevereiro de 2010 a fevereiro de 2016, 23 T. tetradactyla foram recebidos e inspecionados para avaliação da presença de carrapatos. A identificação foi realizada pela observação das características morfológicas e pela utilização de chaves taxonômicas. Aproximadamente 8,69% (2/23) dos animais apresentavam parasitismo por carrapatos. Os ixodídeos foram encontrados fixados nas regiões ventral e cervical, e identificados como Rhipicephalus sanguineus sensu lato (cinco fêmeas), Amblyomma auricularium (uma larva, 18 ninfas e uma fêmea) e Amblyomma sp. (seis larvas). Este estudo relata o parasitismo inédito de larva de A. auricularium em T. tetradactyla no semiárido pernambucano, bem como reafirma a maior afinidade por membros da superordem Xenarthra. O parasitismo de R. sanguineus s.l. em tamanduá-mirim pode estar relacionado ao compartilhamento do mesmo habitat com cães domésticos infestados. O parasitismo sobre animais silvestres em novas localidades onde não foi registrado previamente deve ser estudado para ampliação do conhecimento sobre a distribuição geográfica dos ixodídeos no Brasil.
ABSTRACT The Caatinga is an exclusively Brazilian biome and one of the most diverse in the world, housing a great variety faunal. Southern Tamandua (Linnaeus, 1758), known as collared anteater, can be found throughout the country. The present study aimed to report the presence of ticks on anteaters treated at the Conservation Center and Wildlife Management of Caatinga, Petrolina, Pernambuco, Brazil. From February 2010 to February 2016, 23 T. tetradactyla from the municipalities of Juazeiro (BA) Casa Nova (BA), Sento Sé (BA), Nonato (PI), Petrolina (PE), Great Pond (PE ) and Willow (PE), have been received and inspected to evaluate the presence of ticks, which were later collected with the help of anatomical tweezer and stored in tubes containing 70% alcohol. The identification was carried out by observation of morphological characteristics in stereomicroscope and use of taxonomic keys. Approximately 8.69% (2/23) of the animals had parasitism by ticks. The Ixodids were found fixed on ventral and cervical regions and were identified as Rhipicephalus sanguineus sensu lato (five female), parasitizing the anteater 1 coming from Petrolina-PE; Amblyomma auricularium (one larva, 18 nymphs and a female) and Amblyomma sp. parasitizing the anteater 2 coming from Salgueiro-PE. This study reports the novel parasitism by larva of A. auricularium in T. tetradactyla in semiarid of Pernambuco ; and reaffirms the higher affinity of this ixodid by members of the superorder Xenarthra. It was possible to determine the parasitism of R. sanguineus s.l. in collared anteater, the which may be related to the sharing of the same habitat with domestic dogs infested. The parasitism of wild animals in new locations where they were not previously registered must be stimulated to expand the knowledge of the geographical distribution of ixodids in Brazil.

RESUMO: As células tronco mesenquimais (MSC) residem em pequenas quantidades em muitos tecidos e órgãos adultos, desempenhando um papel ativo na homeostase destes locais. O isolamento de MSC já foi demonstrado em amostras de medula óssea, tecido adiposo e fluido amniótico de cabras. O sangue de cordão umbilical é considerado uma fonte importante desse tipo de células. No entanto, até o presente momento, não foi demonstrado o isolamento de MSC provenientes do sangue de cordão umbilical de cabras. Dessa forma, o objetivo do presente estudo foi isolar, cultivar e caracterizar células tronco mesenquimais provenientes do sangue do cordão umbilical caprino. As MSC foram isoladas utilizando o gradiente de densidade Ficoll-Paque e cultivadas em DMEM suplementado com 10% ou 20% de FBS. A caracterização desse tipo celular foi realizada através de análise por citometria de fluxo e diferenciação em linhagens celulares mesodermais. A analise no citômetro de fluxo demonstrou a presença de duas populações distintas, um grupo com células maiores e outro com células menores; observando expressão positiva de CD90, CD44 e CD105, e negativa para CD34 nas células maiores; enquanto que as menores foram positivas para CD90 e CD105, mas negativas para CD44 e CD34. As células isoladas demonstraram capacidade de se diferenciar em condrócitos e osteoblastos quando incubadas com meio de diferenciação específico. O presente estudo demonstrou que células tronco mesenquimais podem ser obtidas com sucesso do sangue do cordão umbilical caprino.
ABSTRACT: Mesenchymal stem cells (MSC) reside in small numbers in many adult tissues and organs, and play an active role in the homeostasis of these sites. Goat derived multipotent MSC have been established from bone marrow, adipose tissues and amniotic fluid. Umbilical cord blood (UCB) is considered an important source of these cells. However, the MSC isolation from the goat UCB has not been demonstrated. Therefore, the aim of the present study was to isolate, culture and characterize goat umbilical cord blood derived mesenchymal stem cells. MSC were isolated from UCB by Ficoll-Paque density centrifugation and cultured in DMEM supplemented with 10% or 20% FBS. FACS analysis was performed and induction lineage differentiation was made to characterize these cells. They exhibited two different populations in flow cytometry, and revealed the positive expression of CD90, CD44 and CD105, but negative staining for CD34 in larger cells, and positive stained for CD90 and CD105, but negative for CD44 and CD34 in the smaller cells. MSC from goat UCB showed capability to differentiate into chondrocytes and osteoblasts when incubated with specific differentiation medium. Present study established that goat mesenchymal stem cells can be derived successfully from umbilical cord blood.

RESUMO A tartaruga-verde, Chelonia mydas, apresenta distribuição cosmopolita. No Brasil, ocorre na costa, porém desova em ilhas oceânicas. A helmintofauna de tartarugas-verdes é diversificada, podendo-se dizer que tem a maior diversidade comparada com outras espécies de tartarugas. Objetivou-se avaliar aspectos ecológicos da comunidade de helmintos gastrointestinais e relacionar com a condição corporal de tartarugas-verdes recolhidas no litoral do Espírito Santo. Foram utilizados 36 exemplares juvenis da espécie C. mydas. O trato gastrointestinal foi separado e dividido em porções: esôfago/estômago, intestino delgado e intestino grosso. Cada porção foi inspecionada à procura de parasitos, e os exemplares encontrados foram separados para posterior identificação. Das 36 tartarugas avaliadas, 34 estavam parasitadas por helmintos (94,44%), com um total de 10.734 helmintos. Foram encontradas 18 espécies de trematodas pertencentes a quatro famílias. A riqueza de espécies encontrada foi de 4,29±2,19 (1-10) e a intensidade média de infecção foi de 315,64±281,83 (2-994) helmintos. Os parasitos mais prevalentes foram Cricocephalus albus, Metacetabulum invaginatum e Neoctangium travassosi, ambos com 61,11% (22/36), Pronocephalus obliquus com 33,33% (12/36), e Glyphicephalus lobatus com 30,55% (11/36). O helminto mais abundante foi M. invaginatum com 70,63 helmintos/animal, seguido de C. albus com 58,77 helmintos/animal e N. travassosi com 41,75 helmintos/animal.
ABSTRACT The green turtle, Chelonia mydas has worldwide distribution. In Brazil, it is found on the coast, but spawning occurs on oceanic islands. The helminth fauna of green turtles is diverse and has the greatest diversity when compared with other species of turtles. This study aims to evaluate ecological aspects of gastrointestinal helminth community and connect to the body condition of green turtles collected on the coast of Espírito Santo. A total of 36 juvenile specimens of the species C. mydas were used. The gastrointestinal tract was removed and divided into portions: esophagus/stomach, small intestine and large intestine. Each portion was inspected looking for parasites and the specimens found were separated for later identification. Of the 36 turtles evaluated, 34 were parasitized by helminths (94.44%), with a total of 10,734 helminths. Results include findings of 18 species of trematodes belonging to four families. The species richness was 4,29 ± 2,19 (1-10) and the mean intensity of infection was 315,64 ± 281,83 (2-994) helminths. The prevalent parasites were Cricocephalus albus, Metacetabulum invaginatum and Neoctangium travassosi, both with 61,11% (22/36), Pronocephalus obliquus with 33,33% (12/36), and Glyphicephalus lobatus with 30,55% (11/36). The abundant helminth was Metacetabulum invaginatum with helminths 70,63/animal, followed by C. albus with helminths 58,77/animal and N. travassosi with helminths 41,75/animal.

Goat umbilical cord cells are permissive to small ruminant lentivirus infection in vitro

Abstract Small ruminant lentiviruses isolated from peripheral blood leukocytes and target organs can be propagated in vitro in fibroblasts derived from goat synovial membrane cells. These cells are obtained from tissues collected from embryos or fetuses and are necessary for the establishment of the fibroblast primary culture. A new alternative type of host cells, derived from goat umbilical cord, was isolated and characterized phenotypically with its main purpose being to obtain cell monolayers that could be used for the diagnosis and isolation of small ruminant lentiviruses in cell culture. To accomplish this goal, cells were isolated from umbilical cords; characterized phenotypically by flow cytometry analysis; differentiate into osteogenic, chondrogenic and adipogenic lineage; and submitted to viral challenge. The proliferation of goat umbilical cord cells was fast and cell monolayers formed after 15 days. These cells exhibited morphology, immunophenotype, growth characteristics, and lineage differentiation potential similar to mesenchymal stem cells of other origins. The goat umbilical cord derived cells stained positive for vimentin and CD90, but negative for cytokeratin, CD34 and CD105 markers. Syncytia and cell lysis were observed in cell monolayers infected by CAEV-Cork and MVV-K1514, showing that the cells are permissive to small ruminant lentivirus infection in vitro. These data demonstrate the proliferative competence of cells derived from goat umbilical cords and provide a sound basis for future research to standardize this cell lineage.

RESUMO Os escorpiões de interesse médico no Brasil pertencem ao gênero Tityus. Dentre eles, Tityus stigmurus é o principal responsável por picadas de escorpião na região Nordeste. Após uma picada, a peçonha de escorpião se distribui rapidamente para os órgãos, atingindo os rins depressa. Entretanto, há poucos trabalhos sobre fisiopatologia renal do envenenamento por escorpiões. Neste trabalho, avaliamos os efeitos da peçonha de T. stigmurus (TsV) sobre parâmetros renais em rins de ratos isolados. Ratos Wistar (n = 6), pesando 250-300 g, foram perfundidos com solução de Krebs-Henseleit contendo 6 g/100 mL de albumina de soro bovino. TsV nas concentrações de 0,3 e 1,0 μg/mL foi testado e os efeitos sobre a pressão de perfusão (PP), resistência vascular renal (RVR), fluxo urinário (FU), taxa de filtração glomerular (TFG) e excreção de eletrólitos foram analisados. Apenas na concentração de 1,0 μg/mL de TsV foram observados efeitos que aumentaram PP (controlePP40' = 92,7 ± 1,95; TsVPP40' = 182,0 ± 4,70 mmHg*, *p < 0,05), RVR (controleRVR40' = 3,28 ± 0,23 mmHg; TstRVR40' = 6,76 ± 0,45 mmHg*, *p < 0,05), FU (controleFU50' = 0,16 ± 0,04; TstFU50' = 0,60 ± 0,10 mL/g/min*, *p < 0,05), a TFG e excreção de eletrólitos, com alterações histológicas que indicam de lesão renal tubular. Em conclusão, peçonha de T. stigmurus induz um aumento transiente da PP com lesão tubular, ambos os quais conduzem a uma excreção elevada de eletrólitos.
ABSTRACT Scorpions belonging to the Tityus genus are of medical interest in Brazil. Among them, Tityus stigmurus is the main scorpion responsible for stings in the Northeast region. After a sting, the scorpion venom distributes rapidly to the organs, reaching the kidneys quickly. However, there are few studies concerning the renal pathophysiology of scorpion poisoning. In this study, we evaluated the effects of T. stigmurus venom (TsV) on renal parameters in isolated rat kidneys. Wistar rats (n = 6), weighing 250-300 g, were perfused with Krebs-Henseleit solution containing 6 g/100 mL bovine serum albumin. TsV at 0.3 and 1.0 μg/mL was tested, and the effects on perfusion pressure (PP), renal vascular resistance (RVR), urinary flow (UF), glomerular filtration rate (GFR), and electrolyte excretion were analyzed. Effects were observed only at TsV concentration of 1.0 μg/mL, which increased PP (controlPP40' = 92.7 ± 1.95; TsVPP40' = 182.0 ± 4.70* mmHg, *p < 0.05), RVR (controlRVR40' = 3.28 ± 0.23 mmHg; TstRVR40' = 6.76 ± 0.45* mmHg, *p < 0.05), UF (controlUF50' = 0.16 ± 0.04; TstUF50' = 0.60 ± 0.10* mL/g/min,*p < 0.05), GFR and electrolyte excretion, with histological changes that indicate renal tubular injury. In conclusion, T. stigmurus venom induces a transient increase in PP with tubular injury, both of which lead to an augmented electrolyte excretion.

Resumo Um levantamento atualizado das plantas com sementes e análises relevantes acerca desta biodiversidade são apresentados. Este trabalho se iniciou em 2010 com a publicação do Catálogo de Plantas e Fungos e, desde então vem sendo atualizado por mais de 430 especialistas trabalhando online. O Brasil abriga atualmente 32.086 espécies nativas de Angiospermas e 23 espécies nativas de Gimnospermas e estes novos dados mostram um aumento de 3% da riqueza em relação a 2010. A Amazônia é o Domínio Fitogeográfico com o maior número de espécies de Gimnospermas, enquanto que a Floresta Atlântica possui a maior riqueza de Angiospermas. Houve um crescimento considerável no número de espécies e nas taxas de endemismo para a maioria dos Domínios (Caatinga, Cerrado, Floresta Atlântica, Pampa e Pantanal), com exceção da Amazônia que apresentou uma diminuição de 2,5% de endemicidade. Entretanto, a maior parte das plantas com sementes que ocorrem no Brasil (57,4%) é endêmica deste território. A proporção de formas de vida varia de acordo com os diferentes Domínios: árvores são mais expressivas na Amazônia e Floresta Atlântica do que nos outros biomas, ervas são dominantes no Pampa e as lianas apresentam riqueza expressiva na Amazônia, Floresta Atlântica e Pantanal. Este trabalho não só quantifica a biodiversidade brasileira, mas também indica as lacunas de conhecimento e o desafio a ser enfrentado para a conservação desta flora.
Abstract An updated inventory of Brazilian seed plants is presented and offers important insights into the country's biodiversity. This work started in 2010, with the publication of the Plants and Fungi Catalogue, and has been updated since by more than 430 specialists working online. Brazil is home to 32,086 native Angiosperms and 23 native Gymnosperms, showing an increase of 3% in its species richness in relation to 2010. The Amazon Rainforest is the richest Brazilian biome for Gymnosperms, while the Atlantic Rainforest is the richest one for Angiosperms. There was a considerable increment in the number of species and endemism rates for biomes, except for the Amazon that showed a decrease of 2.5% of recorded endemics. However, well over half of Brazillian seed plant species (57.4%) is endemic to this territory. The proportion of life-forms varies among different biomes: trees are more expressive in the Amazon and Atlantic Rainforest biomes while herbs predominate in the Pampa, and lianas are more expressive in the Amazon, Atlantic Rainforest, and Pantanal. This compilation serves not only to quantify Brazilian biodiversity, but also to highlight areas where there information is lacking and to provide a framework for the challenge faced in conserving Brazil's unique and diverse flora.

A piometra é doença que acomete as fêmeas da espécie canina de meia-idade a idosa durante o diestro. Aspectos hormonais, microbiológicos, hematológicos e bioquímicos são bem descritos. Entretanto, são poucos os estudos que avaliam o papel de cada um desses no prognóstico da piometra canina. O objetivo deste estudo foi identificar marcadores associados ao agravamento clínico de cadelas com piometra. Foram avaliadas prospectivamente 80 cadelas com piometra tratadas cirurgicamente. O grupo 1 foi composto por cadelas que receberam alta até 48 horas de pós-operatório, e o grupo 2 por aquelas que necessitaram de internamento prolongado ou morreram. Os achados hematológicos, bioquímicos e os níveis do lactato sanguíneo foram comparados entre os grupos, e variáveis como multirresistência bacteriana, síndrome da resposta inflamatória sistêmica (SIRS), hiperlactatemia e aumento da creatinina foram analisadas por meio da dispersão de frequências entre os grupos. Entre as variáveis estudadas, a presença de SIRS e a elevação da creatinina sérica >2,5mg/mL foram eficazes em predizer o agravamento da doença e podem ser utilizadas como marcadores prognósticos da piometra canina.
The pyometra is a disease that affects middle age and elderly female dogs during diestrus. Hormonal, microbiological, biochemical and hematological aspects are well described. However, few studies have evaluated the role of each in the prognosis of canine pyometra. The aim of this study was to identify markers associated with clinical worsening of dogs with pyometra. We prospectively evaluated 80 dogs with pyometra treated surgically. Group 1 consisted of dogs that were discharged within 48 hours after surgery and Group 2 consisted of those who required prolonged hospitalization or died. The findings of hematological, biochemical and blood lactate levels were compared between groups and variables such as bacterial multidrug resistance, systemic inflammatory response syndrome (SIRS), hyperlactatemia and increased creatinine were analyzed through the dispersion of frequencies between groups. Among the variables studied, the presence of SIRS and elevated serum creatinine >2.5mg/mL were effective in predicting the worsening of the disease and can be used as prognostic markers of canine pyometra.

A padronização de técnicas e o teste da associação do Plasma Rico em Plaquetas (PRP) e das Células-Tronco Mononucleares (CTMs) na consolidação de falhas ósseas corticais, por meio de avaliação clínica, biomecânica, radiológica e histológica, é avaliada em um estudo piloto. Foram utilizados seis cães adultos, fêmeas, sem raça definida, pesando entre 5 e 10kg, separados por sorteio aleatório em seis tratamentos. Foi confeccionada uma falha elíptica de 1,0x0,4cm na cortical medial diafisária da tíbia direita de cada animal, sendo preenchida de acordo com o tratamento proposto. No cão I, a falha foi preenchida com solução fisiológica (SF); no II, com o PRP; no III, com a fração total das células mononucleares (FTCM); no IV, com a fração vascular estromal (FVE); no V, com o PRP associado à FTCM; no VI, com a associação PRP e FVE. Foram realizadas avaliações: clínicas, diariamente; dos graus de claudicação, semanalmente; radiológica e perimetria da coxa, antes, no pós-operatório imediato, aos 7, 14, 21 e 30 dias; biomecânica, antes do procedimento, aos 10, 20 e 30 dias; e biópsias, aos 15 e 30 dias. A FTCM obteve uma contagem e viabilidade média de 2,0x108cél. e 90%, respectivamente, enquanto a FVE obteve 3x106cél. e 50%. O PRP concentrou, em média, sete vezes o número inicial de plaquetas do sangue total, de 250.000 µl-1 plaquetas no sangue total para 1.750.000 µl-1 plaquetas no PRP. Obteve-se padronização adequada de técnicas, possibilitando o teste da associação entre as células-tronco mononucleares (CTMs) e o plasma rico em plaquetas (PRP), assim como seu uso isolado, no reparo de falhas ósseas corticais, indicando a possibilidade de a associação FTCM e PRP ser o melhor tratamento.
The standardization of techniques and tests of the association of Platelet Rich Plasma (PRP) and Mononuclear Stem Cells (MSCs) in the consolidation of cortical bone defects by clinical, biomechanical, radiological, and histological analysis is evaluated in a pilot study. Six adult female dogs of mixed breed, weighing between 5 and 10kg, separated by random draw in six treatments were used. An elliptical failure of 1.0 x0.4cm was done in the medial diaphyseal cortical of the right tibia of each animal, that was filled according with the proposed treatment. In dog I, the failure was filled with saline (S), in dog II with PRP, in dog III with total mononuclear cell fraction (TMCF), in dog IV with stromal vascular fraction (SVF), in dog V with association of PRP and TMCF, and in dog VI with an association of PRP and SVF. Daily clinical evaluation, weekly degrees of lameness, radiological and girth before, immediate postoperative, 7, 14, 21 and 30 days, biomechanics before the procedure, at 10, 20 and 30 days, and biopsies at 15 and 30 days were performed. The TMCF got a count and viability of 2,0x108cells and 90% respectively, while for SVF it was 3x106cells and 50%, respectively. The PRP concentrated on average seven times the original number of platelets from whole blood, platelets from whole blood 250.000 μl-1to 1.750.000 μl-1platelets in PRP. This afforded adequate standardization of techniques, enabling the test of association between mononuclear stem cells (MSCs) and platelet-rich plasma(PRP), as well as their separate use to repair cortical bone defects, indicating the possibility of the association between FTCM and PRP to be the best treatment.
