ABSTRACT L-carnitine perform a major role in transporting long-chain fatty acids into the mitochondria, where they are oxidized. It has been used in animal diets to decrease fat and increase muscle protein. The aim of this study was to evaluate the zootechnical performance, degree of steatosis in the liver, and genotoxic potential in Astyanax lacustris fed with different levels of L-carnitine (LC). Yellowtail tetra juveniles (n = 140) were distributed in 20 tanks of 70 L, with seven fish in each, in a water recirculation system with controlled temperature (27±0.1⁰C). The treatments with different levels of L-carnitine supplementation were: 0 (control), 250, 500, 750, and 1000 mg of LC per kg of food. The diets were provided twice a day for 60 days. The results showed that the different levels of LC did not affect (P>0.05) weight gain, survival, viscerosomatic index, and the liver hepatocytes showed a normal appearance. However, the use of LC supplementation showed genotoxic potential with a significant difference (P<0.05) for cell alterations when compared to the control at concentrations above 500mg kg-1.

RESUMO A L-carnitina exerce um papel importante no transporte de ácidos graxos de cadeia longa até a mitocôndria para serem oxidados e tem sido incorporada em rações para animais com o objetivo de diminuir a deposição de gordura e aumentar a proteína muscular. O objetivo deste trabalho foi avaliar o desempenho zootécnico, o grau de esteatose no fígado e o potencial genotóxico em Astyanax lacustris alimentados com diferentes níveis de L-carnitina (LC). Juvenis de lambari-do-rabo-amarelo (n=140) foram distribuídos em 20 caixas de 70L, sete peixes em cada, em um sistema de recirculação de água com temperatura controlada (27±0,1⁰C). Os tratamentos com os níveis de suplementação foram: 0 (controle), 250, 500, 750 e 1000 mg de LC kg-1 de ração. As dietas foram fornecidas duas vezes ao dia, durante 60 dias. Os resultados mostraram que os diferentes níveis de LC não influenciaram (P>0,05) o ganho de peso; a sobrevivência, o índice viscerossomático e os hepatócitos do fígado apresentaram-se com aparência normal. No entanto, a suplementação com LC apresentou potencial genotóxico com diferença significativa (P<0,05) para alterações celulares quando comparada ao controle em concentrações superiores a 500mg kg-1.

Abstract We evaluated the influence of a 32-day camping in Antarctica on physical performance and exercise-induced thermoregulatory responses. In Brazil, before and after the Antarctic camping, the volunteers performed an incremental exercise at temperate conditions and, two days later, an exercise heat stress protocol (45-min running at 60% of maximum aerobic speed, at 31°C and 60% of relative humidity). In Antarctica, core temperature was assessed on a day of fieldwork, and average values higher than 38.5°C were reported. At pre- and post-Antarctica, physiological (whole-body and local sweat rate, number of active sweat glands, sweat gland output, core and skin temperatures) and perceptual (thermal comfort and sensation) variables were measured. The Antarctic camping improved the participants’ performance and induced heat-related adaptations, as evidenced by sweat redistribution (lower in the chest but higher in grouped data from the forehead, forearm, and thigh) and reduced skin temperatures in the forehead and chest during the exercise heat stress protocol. Notwithstanding the acclimatization, the participants did not report differences of the thermal sensation and comfort. In conclusion, staying in an Antarctic camp for 32 days improved physical performance and elicited physiological adaptations to heat due to the physical exertion-induced hyperthermia in the field.

RESUMO Avaliou-se o valor nutricional do capim-elefante verde colhido aos 56, 84 e 112 dias de idade, por meio do consumo e da digestibilidade em ovinos, da degradabilidade in situ e da cinética da fermentação ruminal. O capim-elefante colhido aos 56 dias proporcionou maiores valores de consumo da MS (1204,81g/dia), da EB (4910,41kcal/dia), da ED (3436,21kcal/dia) e da PB (130,90g/dia). Para o capim-elefante colhido aos 56 dias, ainda foram observados os maiores valores de digestibilidade da MS (70,64%), da EB (70,11%), da PB (68,96%), da FDN (71,4%) e do conteúdo ED (2,86Mcal/kg MS). Com o avanço da idade de corte, houve redução da degradabilidade potencial da MS (77,99% x 68,33%), da PB (84,80% x 56,52%) e da FDN (72,18% x 64,33%), para as idades de corte de 56 e 112 dias, respectivamente. O capim cortado aos 56 dias apresentou maior valor de produção cumulativa de gases (210,50mL/g de MS) e de degradabilidade da matéria seca (63,9%) após 96 horas de fermentação. O capim-elefante verde deve ser colhido aos 56 dias de rebrota em razão das características nutricionais apresentadas.

ABSTRACT The nutritional value of fresh elephant grass harvested at 56, 84 and 112 days of age was evaluated by intake and digestibility in sheep, in situ degradability, and ruminal fermentation’s kinetics. The elephant grass harvested at 56 days provided higher DM intake (1204.81g/day), GE (4910.41kcal/day), DE (3436.21kcal/day), and CP (130.90g/day) values. Furthermore, the highest DM (70.64%), GE (70.11%), DE (2.86%) and CP (68.96%) digestibility values were observed at 56 days. With the increase in the harvest age there was a reduction in the DM (77.99 x 68.33%), CP (84.80 x 56.52%) and NDF (72.18 x 64.33%) potential degradability in the treatments 56 and 112 days, respectively. Higher values of cumulative gas production (210,50mL/g DM) and DM degradability for elephant grass were observed at 56 days, after 96 hours of fermentation. The fresh elephant grass should be harvested at 56 days of regrowth due to the nutritional characteristics found in this study.

RESUMO Objetivou-se com este trabalho avaliar o efeito do primeiro desbaste sobre a morfometria de um plantio de Tectona grandis L.f. (teca). O delineamento experimental foi o de blocos ao acaso, com quatro tratamentos e três repetições sendo: T1 = testemunha (sem desbaste), T2, T3 e T4 = remoção da área basal de 20%, 30% e 40% respectivamente. As variáveis morfométricas avaliadas foram: proporção de copa (PC), índice de abrangência (IA), grau de esbeltez (GE), índice de saliência (IS), formal de copa (FC) e comprimento de copa (CC). A aplicação de desbastes em teca influenciou significativamente em todas as variáveis morfométricas avaliadas, com exceção de proporção de copa (PC) e índice de saliência (IS). As intensidades de desbastes (20%, 30% e 40%) não apresentaram diferença significativa para as variáveis: altura comercial do fuste (HF), comprimento de copa (CC), diâmetro de copa (DC), proporção de copa (PC) e forma de copa (FC) da Tectona grandis até o quarto ano pós-desbaste.

ABSTRACT The aim of this study was to evaluate the effect of first thinning on the morphometry of trees in a forest plantation of Tectona grandis Lf (teak). The experimental design used was randomized block with four treatments and three replications were: T1 = control treatment (no thinning), T2, T3 end T4 with the removal of basal area the 20, 30 and 40% respectively. The evaluated morphometric variables were: crown proportion (PC), coverage index (AI), slenderness degree (GE), salience index (IS), crown form (FC) and crown length (CC). The application of thinning in teak, significantly influenced in all morphometric variables analyzed, except for the proportion of room (PC) and the projection index (SI). The thinning intensities (20%, 30% and 40%) presented no significant differences for variables: commercial bole height (HF), crown length (CL), crown diameter (DC), crown proportion (PC) and crown form (FC) of teak to the fourth post-thinning year.

Bactérias do gênero Staphylococcus estão entre os principais agentes causadores da mastite ovina. Um dos maiores entraves ao tratamento dos animais doentes são cepas resistentes aos antimicrobianos empregados. A pesquisa do gene mecA nos estafilococos é um instrumento auxiliar para a determinação de aspectos epidemiológicos da doença. Este trabalho teve por objetivo investigar a resistência à oxacilina em estafilococos coagulase-negativos isolados no leite de ovelhas com mastite subclínica. Foram analisadas 448 amostras de leite de dois rebanhos. Os micro-organismos isolados foram submetidos previamente a testes de sensibilidade a antibióticos in vitro a partir da técnica de difusão em disco. Naqueles resistentes à oxacilina nestes testes efetuou-se a pesquisa do gene mecA, com a extração do DNA cromossômico por meio da técnica de extração fenol-clorofórmio. Os estafilococos coagulase-negativos apresentaram resistência à oxacilina e a presença do gene mecA foi detectada em quatro isolados, que também apresentaram características de multirresistência. Tais achados reforçam a importância deste grupo de micro-organismos na etiologia da mastite subclínica em ovinos e abre perspectivas para futuras pesquisas para a investigação da epidemiologia da doença.

Staphylococcus bacteria are among the main agents of ovine mastitis. One of the greatest barriers to treatment of sick animals are strains resistant to antimicrobial agents. The research of the mecA gene in staphylococci is an auxiliary tool for the determination of epidemiological aspects of disease. The present study aimed to investigate oxacillin resistance in coagulase-negative staphylococci isolated in the milk from ewes with subclinical mastitis. A total of 448 samples from 2 flocks were analyzed. Colonies were previously submitted to testing for susceptibility to antibiotics in-vitro using the technique of disk diffusion. Using the oxacillin-resistant strains in these tests, the research of mecA gene was conducted with the extraction of chromosomal DNA by way of the technique of phenol-chloroform extraction. Coagulase-negative staphylococci were resistant to oxacillin, and the mecA gene was detected in 4 isolates, which also showed characteristics of multidrug resistance. These findings reinforce the importance of these microorganisms in the etiology of subclinical mastitis in ewes, and open perspectives for future research to investigate the epidemiology of this disease.

The objective of the present study was to determine the frequency of somatic chromosomal anomalies and Y chromosomal microdeletions (azoospermia factor genes, AZF) in infertile males who seek assisted reproduction. These studies are very important because the assisted reproduction techniques (mainly intracytoplasmic sperm injection) bypass the natural selection process and some classical chromosomal abnormalities, microdeletions of AZF genes or some deleterious genic mutations could pass through generations. These genetic abnormalities can cause in the offspring of these patients male infertility, ambiguous external genitalia, mental retardation, and other birth defects. We studied 165 infertile men whose infertility was attributable to testicular problems (60 were azoospermic, 100 were oligospermic and 5 were asthenospermic). We studied 100 metaphases per patient with GTG banding obtained from temporary lymphocyte culture for chromosomal abnormality detection and performed a genomic DNA analysis using 28 Y chromosome-specific sequence-tagged sites for Y AZF microdeletion detection. Karyotyping revealed somatic anomalies in 16 subjects (16/165 = 9.6%). Of these 16, 12 were in the azoospermic group (12/60 = 20%) and 4 were in the oligospermic group (4/100 = 4%). The most common chromosomal anomaly was Klinefelter syndrome (10/165 = 6%). Microdeletions of AZF genes were detected in 12 subjects (12/160 = 7.5%). The frequencies detected are similar to those described previously. These results show the importance of genetic evaluation of infertile males prior to assisted reproduction. Such evaluation can lead to genetic counseling and, consequently, to primary and secondary prevention of mental retardation and birth defects.
