Multiparasitism in Crab-eating fox (Cerdocyon thous) (Carnivora: Canidae), including new records, in the Brazilian Pampa - case report

ABSTRACT Cerdocyon thous is considered the most common wild canid in Rio Grande do Sul, and can serve as a host for several parasites, playing an important role in maintaining the biological cycle. Despite being a widespread and abundant species, they have a relatively little-known fauna of endo and ectoparasites, mainly focusing on the different regions of their occurrence. With this work, the objective was to report the multiparasitism in a specimen of C. thous in the Brazilian Pampa. Using modified Centrifuge-Flotation and Spontaneous Sedimentation techniques, a high taxonomic diversity of parasites was observed, and the finding of eggs compatible with Lagochilascaris spp. in C. thous in Brazil, in addition to Uncinaria spp. eggs and Cystoisospora spp. oocysts in the same host in the south of the country. The presence of eggs of different helminths in the feces of C. thous is extremely important, mainly for public health, but also for the knowledge of the biodiversity of parasites present in wild populations of canids, until then little studied, thus allowing the implementation of control programs.
RESUMO Cerdocyon thous é considerado o canídeo silvestre mais comum no Rio Grande do Sul e pode servir como hospedeiro de diversos parasitos, desempenhando papel importante na manutenção do ciclo biológico. Mesmo sendo uma espécie difundida e abundante, têm uma fauna de endo e ectoparasitos relativamente pouco conhecida, principalmente enfocando as diferentes regiões de sua ocorrência. Com este trabalho, objetivou-se relatar o multiparasitismo em um espécime de C. thous no Pampa brasileiro. Por meio das técnicas de centrifugo-flutuação modificada e sedimentação espontânea, observou-se uma alta diversidade taxonômica de parasitos, sendo relatado, pela primeira vez, o achado de ovos compatíveis com Lagochilascaris spp. em C. thous no Brasil, além de ovos de Uncinaria spp. e oocistos de Cystoisospora spp. no mesmo hospedeiro, no sul do país. A presença de ovos de diferentes helmintos nas fezes de C. thous é de extrema importância, principalmente na saúde pública, mas também para o conhecimento da biodiversidade de parasitos presentes em populações silvestres de canídeos, até então pouco estudados, permitindo, assim, a implementação de programas de controle.

Resumo Moluscos infectados por trematódeos podem aumentar ou diminuir sua atividade reprodutiva em resposta à presença da infecção. Nosso objetivo foi verificar as alterações reprodutivas de Subulina octona após a infecção por Paratanaisia bragai. Os moluscos infectados foram individualmente expostos durante 24 horas a 20 ovos do parasito e quatro grupos foram formados (10, 20, 30 e 40 d.p.i. – dias pós infecção), com respectivos grupos controle. A cada 10 dias, o número de ovos no trato reprodutivo, o número de ovos eclodidos, o conteúdo de galactogênio e alterações histopatológicas eram avaliadas. O padrão reprodutivo nos moluscos controle e infectados evidenciou um comportamento alternante, nos quais períodos de alta produção de ovos e filhotes foram seguidos por períodos de baixa produção. No entanto, em relação ao conteúdo de galactogênio, moluscos controle e infectados seguiram o mesmo padrão de variação. Na histologia, observamos a presença de gametas masculinos e femininos com acentuada redução no número de óocitos. Os resultados indicam que o desenvolvimento intramolusco do parasito afeta a biologia reprodutiva do hospedeiro.
Abstract Snails infected by trematodes may increase or decrease their reproductive activity in response to the presence of infection. Our aim was to verify the reproductive alterations in Subulina octona after infection by Paratanaisia bragai. The infected snails were individually exposed for 24 hours to 20 parasite eggs and four groups were formed (10, 20, 30 and 40 d.p.i.- days after infection), along with control groups. Every 10 days, the number of eggs in the reproductive tract, number of eggs hatched, galactogen content and histopathological changes were evaluated. The reproductive in the control and infected snails presented an alternating pattern, where periods of high production of eggs and newly hatched were followed by periods of low production. However, in relation to the amount of galactogen, both control and infected groups followed the same pattern of variation. In the histology, we observed the presence of male and female gametes with marked reduction in the number of oocytes. The results indicate that the intra-snail development of the parasite affects the reproductive biology of the host.

Epidermal growth factor receptor regulates fibrinolytic pathway elements in cervical cancer: functional and prognostic implications

Epidermal growth factor receptor (EGFR) signaling and components of the fibrinolytic system, including urokinase-type plasminogen activator (uPA) and thrombomodulin (TM), have been implicated in tumor progression. In the present study, we employed cBioPortal platform (http://www.cbioportal.org/), cancer cell lines, and an in vivo model of immunocompromised mice to evaluate a possible cooperation between EGFR signaling, uPA, and TM expression/function in the context of cervical cancer. cBioPortal analysis revealed that EGFR, uPA, and TM are positively correlated in tumor samples of cervical cancer patients, showing a negative prognostic impact. Aggressive human cervical cancer cells (CASKI) presented higher gene expression levels of EGFR, uPA, and TM compared to its less aggressive counterpart (C-33A cells). EGFR induces uPA expression in CASKI cells through both PI3K-Akt and MEK1/2-ERK1/2 downstream effectors, whereas TM expression induced by EGFR was dependent on PI3K/Akt signaling alone. uPA induced cell-morphology modifications and cell migration in an EGFR-dependent and -independent manner, respectively. Finally, treatment with cetuximab reduced in vivo CASKI xenografted-tumor growth in nude mice, and decreased intratumoral uPA expression, while TM expression was unaltered. In conclusion, we showed that EGFR signaling regulated expression of the fibrinolytic system component uPA in both in vitro and in vivo settings, while uPA also participated in cell-morphology modifications and migration in a human cervical cancer model.

RESUMO A criação de equinos brasileira movimenta anualmente cerca de R$ 16,15 bilhões e impulsiona o setor econômico primário do país. O mercado de animais da raça Crioula vem se expandindo devido ao seu alto potencial zootécnico. No Rio Grande do Sul, a criação é tipicamente extensiva, aliada, muitas vezes, à alta densidade animal, o que favorece as constantes infecções por diversos parasitos. O objetivo deste estudo foi analisar retrospectivamente a frequência de parasitos gastrointestinais em cavalos da raça Crioula naturalmente infectados na região sul do Rio Grande do Sul. Foram utilizadas 585 amostras de fezes de equinos da raça Crioula, adultos, de ambos os sexos, recebidas de criatórios e centros reprodutivos localizados na região. Do total de amostras, 89,74% (525/585) foram positivas para algum helminto. Em 57,60% (337/585) das amostras, observaram-se somente ovos da família Strongylidae. Infecções por Parascaris spp. e Strongyloides sp., apresentaram frequências de 2,22% (13/585) e 1,53% (9/585), respectivamente. Assim, pode-se concluir que os parasitos mais frequentes na população equina da região sul do RS pertencem à família Strongylidae.

RESUMO O objetivo do presente estudo foi avaliar o valor nutricional e energético da torta de mamona destoxificada por diferentes métodos para suínos em crescimento. Foram utilizados 24 suínos machos, castrados, distribuídos em seis tratamentos, sendo uma ração referência e cinco rações testes (75% da dieta referência e 25% das tortas de mamona) contendo a torta de mamona destoxificada pelos métodos do hidróxido de cálcio, da autoclavagem, do hidróxido de cálcio+ensilagem, da autoclavagem+ensilagem e da extrusão. As tortas de mamona submetidas aos processos de destoxificação com hidróxido de cálcio e autoclavagem resultaram em maior teor de proteína digestível. Para o valor de energia metabolizável, os processos de destoxificação com hidróxido de cálcio, autoclavagem, hidróxido de cálcio+ensilagem e extrusão não diferiram entre si. Considerando-se a valoração nutricional e energética da torta de mamona por meio dos processos de destoxificação, os métodos com uso de hidróxido de cálcio e autoclavagem são os mais eficientes.
ABSTRACT The aim of this study was to evaluate the nutritional and energy value of castor bean cake detoxified by different methods for growing pigs. A total of 24 barrows was distributed in six treatments, being in a reference diet and 5 test diets (75% of the reference diet and 25% of castor beam cake) containing castor bean cake detoxified by the method of calcium hydroxide, autoclaving, calcium hydroxide + silage, autoclaving + silage and extrusion. Castor bean cake subjected to detoxification processes with calcium hydroxide and autoclaving resulted in higher content of digestible protein. For the metabolizable energy value, the process of detoxification with calcium hydroxide, autoclaving, calcium hydroxide+silage, and extrusion did not differ. Considering the nutritional and energy valuation of castor bean cake detoxified by different procedures, the methods using calcium hydroxide and autoclaving were the most efficient.

Isovitexin as marker and bioactive compound in the antinociceptive activity of the Brazilian crude drug extracts of Echinodorus scaber and E. grandiflorus

Abstract Echinodorus scaber Rataj and Echinodorus grandiflorus (Cham. & Schltdl.) Micheli, Alismataceae, are popularly used to relieve inflammatory complaints and as diuretic. A study on the antinociceptive effect and selected marker compounds in eleven extracts from different locations was undertaken and their antinociceptive effect was assessed. The fingerprints were compared by HPLC-DAD and the content of vitexin, isovitexin, isoorientin and vitexin-2-O-rhamnoside were determined. All samples presented antinociceptive activity reducing the writhes by 36.4-62.5% and 47.4-79.8% at 10 and 50 mg/kg, respectively; indomethacin (5 mg/kg) reduced writhes by 82.6-90.1%. The content of the flavonoids C-glycosides, however, presented a strong variation. Isovitexin and isoorientin were found in all the samples, with content ranging from traces to 14.70 µg/mg and 2.12-84.27 µg/mg extract, respectively, while vitexin-2-O-rhamnoside occurred in quantifiable amounts only in 3 out of 11 samples ranging from 5.43 to 33.13 µg/mg extract; vitexin was not detected at all or detected in trace amounts. According to the fingerprints, the samples could be arranged in four main groups. All eleven extracts showed antinociceptive activity. Isovitexin was the only flavonoid present in all samples and can be regarded, acting in synergy with the other compounds or not, as the responsible for the antinociceptive activity. Therefore, isovitexin is a good choice as chemical marker when the antinociceptive activity of E. scaber and E. grandiflorus is investigated.

RESUMO O objetivo deste estudo foi avaliar o efeito antibacteriano e tripanocida in vitro do extrato hidroalcóolico das raízes de Tradescantia sillamontana Matuda (Commelinaceae), conhecida popularmente como veludo branco. Foi avaliada a atividade antibacteriana in vitro frente às bactérias Streptococcus mitis (CIM = 100 µg/mL; CMB = 150 µg/mL), Streptococcus mutans (CIM = 200 µg/mL; CMB = 220 µg/mL), Streptococcus sanguinis (CIM = 400 µg/mL; CMB = 425 µg/mL), Streptococcus sobrinus (CIM = 400 µg/mL; CMB = 420 µg/mL) e Bacteroides fragilis (CIM = 400 µg/mL; CMB = 430 µg/mL) pelo método de diluição em caldo. Os protozoários da família tripanossomatídeo causam doenças tropicais que costumam ser negligenciadas que costumam ser como a tripanossomíase, para a qual estão disponíveis poucos medicamentos. Neste contexto, o extrato hidroalcóolico das raízes de T. sillamontana também foi avaliado frente às formas tripomastigotas da cepa Y de Trypanosoma cruzi, com promissora atividade frente a este protozoário (IC50 = 2,4 µg/mL). Quando avaliada a atividade citotóxica frente a fibroblastos da linhagem LLCMK2, o extrato apresentou moderada citotoxicidade (CC50 = 480,37 µg/mL). Os resultados ora apresentados para o extrato hidroalcóolico das raízes de Tradescantia sillamontana Matuda demonstraram promissoras atividades antibacteriana e tripanocida, sendo uma fonte alternativa de produtos naturais com atividades contra T. cruzi e algumas bactérias do gênero Streptococcus e Bacteroides.
ABSTRACT The aim of this study was to investigate the in vitro, antibacterial and trypanocidal effect of the hydroalcoholic extract from the roots of Tradescantia sillamontana Matuda (Commelinaceae), commonly known as Veludo branco. The in vitro antibacterial activity against the standard bacteria Streptococcus mitis (MIC = 100µg/mL; MBC = 150 µg/mL), Streptococcus mutans (MIC = 200µg/mL; MBC = 220 µg/mL), Streptococcus sanguinis (MIC = 400µg/mL; MBC = 425 µg/mL), Streptococcus sobrinus (MIC = 400µg/mL; MBC = 420 µg/mL) andBacteroides fragilis (MIC = 400µg/mL; MBC = 430 µg/mL), using microdilution broth methods. Protozoans from the trypanosomatid family cause neglected tropical diseases such as trypanosomiasis, for which few drugs are available. In this context, the hydroalcoholic extract of the Tradescantia sillamontana roots was also investigated with regards to the in vitro effects against the trypomastigote forms of theY strain of Trypanosoma cruzi, showing strong activity against this parasite (IC50 = 2.4 µg/mL). When performing cytotoxic activity against fibroblasts LLCMK2 line, the extract showed moderate cytotoxicity (CC50 = 480.37 mg/mL). The results presented for the hydroalcoholic extract of the roots of Tradescantia sillamontana Matuda demonstrated effective antibacterial and trypanocidal activities and were shown to be an alternative source of natural products with activity against T. cruzi and some bacteria of the genus Streptococcus and Bacteroides.

Resumo Um levantamento atualizado das plantas com sementes e análises relevantes acerca desta biodiversidade são apresentados. Este trabalho se iniciou em 2010 com a publicação do Catálogo de Plantas e Fungos e, desde então vem sendo atualizado por mais de 430 especialistas trabalhando online. O Brasil abriga atualmente 32.086 espécies nativas de Angiospermas e 23 espécies nativas de Gimnospermas e estes novos dados mostram um aumento de 3% da riqueza em relação a 2010. A Amazônia é o Domínio Fitogeográfico com o maior número de espécies de Gimnospermas, enquanto que a Floresta Atlântica possui a maior riqueza de Angiospermas. Houve um crescimento considerável no número de espécies e nas taxas de endemismo para a maioria dos Domínios (Caatinga, Cerrado, Floresta Atlântica, Pampa e Pantanal), com exceção da Amazônia que apresentou uma diminuição de 2,5% de endemicidade. Entretanto, a maior parte das plantas com sementes que ocorrem no Brasil (57,4%) é endêmica deste território. A proporção de formas de vida varia de acordo com os diferentes Domínios: árvores são mais expressivas na Amazônia e Floresta Atlântica do que nos outros biomas, ervas são dominantes no Pampa e as lianas apresentam riqueza expressiva na Amazônia, Floresta Atlântica e Pantanal. Este trabalho não só quantifica a biodiversidade brasileira, mas também indica as lacunas de conhecimento e o desafio a ser enfrentado para a conservação desta flora.
Abstract An updated inventory of Brazilian seed plants is presented and offers important insights into the country's biodiversity. This work started in 2010, with the publication of the Plants and Fungi Catalogue, and has been updated since by more than 430 specialists working online. Brazil is home to 32,086 native Angiosperms and 23 native Gymnosperms, showing an increase of 3% in its species richness in relation to 2010. The Amazon Rainforest is the richest Brazilian biome for Gymnosperms, while the Atlantic Rainforest is the richest one for Angiosperms. There was a considerable increment in the number of species and endemism rates for biomes, except for the Amazon that showed a decrease of 2.5% of recorded endemics. However, well over half of Brazillian seed plant species (57.4%) is endemic to this territory. The proportion of life-forms varies among different biomes: trees are more expressive in the Amazon and Atlantic Rainforest biomes while herbs predominate in the Pampa, and lianas are more expressive in the Amazon, Atlantic Rainforest, and Pantanal. This compilation serves not only to quantify Brazilian biodiversity, but also to highlight areas where there information is lacking and to provide a framework for the challenge faced in conserving Brazil's unique and diverse flora.

Replacement of soybean meal by cottonseed meal 38% in multiple supplements for grazing beef heifers

Objetivou-se avaliar o efeito do uso de suplementos múltiplos com diferentes níveis de farelo de algodão 38% o sobre o consumo, a digestibilidade aparente total de componentes da dieta, a eficiência de síntese microbiana e o desempenho produtivo de novilhas de corte em pastagem de Brachiaria decumbens no período das águas. Utilizaram-se 25 novilhas nelores e 10 novilhas mestiças com predominância de sangue zebu com 16 meses idade e 249,8 ± 0,6 kg de peso corporal. O delineamento foi inteiramente casualizado com cinco tratamentos e sete repetições. Os suplementos continham aproximadamente 30% de proteína bruta (PB) e substituição do farelo de soja pelo farelo de algodão 38% nos níveis de 0; 33; 67 e 100%, (CS0, CS33, CS67 e CS100, respectivamente), avaliados em relação a um grupo controle, que recebeu apenas mistura mineral ad libitum (MM). O ganho médio diário foi 397, 546, 570, 525 e 542 g, respectivamente, para os grupos MM, CS0, CS33, CS67 e CS100. Não houve efeito dos níveis de farelo de algodão 38% sobre o ganho médio diário, que diferiu, no entanto, entre os animais que receberam suplemento e os do grupo controle. Os consumos em kg/dia de proteína bruta e carboidratos não-fibrosos (CNF) foram maiores nos grupos sob suplementação. Os coeficientes de digestibilidade aparente totais da PB e dos CNF foram maiores nos animais que receberam suplemento em relação aos do grupo controle. O aumento do nível de substituição do farelo de soja pelo farelo de algodão teve efeito linear positivo sobre os coeficientes de digestibilidade aparente totais da MS, matéria orgânica e PB. Não houve diferença na eficiência microbiana, expressa em g de proteína bruta por kg de NDT consumido entre os animais sob suplementados e os do grupo controle, assim como não foi observada diferença entre os níveis de substituição do farelo de soja pelo farelo de algodão. O uso de farelo de algodão 38% em substituição ao farelo de soja na dieta não prejudica o desempenho de novilhas de corte em pastejo.
The objective of this work was to evaluate the effect of using multiple supplements with different levels of cottonseed meal 38% on intake, total apparent digestibility of dietary components, efficiency of microbial synthesis and productive performance of beef heifers in Brachiaria decumbens pasture during the rainy season. It was used 25 Nellore heifers and 10 crossbred heifers with predominance of Zebu breed at 16 months of age and with 249.8 ± 0.6 kg of body weight. It was used a complete random experimental design with five treatments and seven replicates. Supplements contained approximately 30% of crude protein (CP) and soybean meal replaced by cottonseed meal 38% at the levels 0; 33; 67 and 100%, (CS0, CS33, CS67 and CS100, respectively) evaluated in relation to a control group, which received only mineral mixture (MM) ad libitum. Daily weight gain was 397, 546, 570, 525 and 542 g, respectively, for the groups MM, CS0, CS33, CS67 and CS100. There was no effect of the levels of cottonseed meal 38% on daily weight gain, which differed among animals fed supplement and control group, however. Intakes of crude protein and non-fibrous carbohydrates (NFC) in kg/day were greater in groups under supplementation. Coefficients of total apparent digestibility of CP and NFC were greater in animals fed supplements in relation to the control group. Increase on the replacement level of soybean meal by cottonseed meal had a positive linear effect on coefficients of total apparent digestibility of DM, organic matter and CP. There was no difference on microbial efficiency, expressed as g of crude protein per kg of TDN consumed by the animals under supplements and the ones of the control group, neither it was observed difference among levels of replacement of soybean meal by cottonseed meal. The use of cottonseed meal 38% replacing soybean meal in the diet does not harm performance of grazing beef heifers.

RESUMO Avaliou-se o comportamento de genótipos de feijoeiro sob infestação de Bemisia tabaci (Genn.) biótipo B em condições de campo, nas épocas de cultivo “da seca” e “das águas”. Utilizaram-se os genótipos IAC-Tybatã, IAC-Una, FT-Nobre, Pérola, LP 98-122, LP 02-130, LP 01-38, LP 9979, BRS-Pontal, BRS-Requinte, BRS-Triunfo, BRS-Grafite, CV-48 e Z-28. Adotou-se o delineamento de blocos casualizados com 14 tratamentos e três repetições. As avaliações foram realizadas semanalmente, contando-se o número de ovos e de ninfas de B. tabaci biótipo B em dez folíolos por parcela. Na safra “da seca” os genótipos menos ovipositados por B. tabaci biótipo B foram IAC-Una e LP 02-130 e os mais ovipositados foram os genótipos Z-28 e Pérola. As menores presenças de ninfas de mosca-branca foram observadas em LP 98-122 e FT-Nobre e as maiores em LP 01-38, Pérola e Z-28. Para a safra “das águas” os genótipos não diferiram entre si em relação ao ataque de B. tabaci biótipo B. Os fatores da produção apresentaram diferença estatística somente na safra das secas.
ABSTRACT Bean genotypes were evaluated under the infestation of Bemisia tabaci (Genn.) biotype B under field conditions in the dry and rainy season. The genotypes used were IAC-Tybatã, IAC-Una, FT-Nobre, Pérola, LP 98-122, LP 02-130, LP 01-38, LP 9979, BRSPontal, BRS-Requinte, BRS-Triunfo, BRS-Grafite, CV-48 and Z-28. The experiment was arranged in randomized blocks with 14 treatments and 3 replications. The evaluations were made on a weekly basis by counting B. tabaci biotype B eggs and nymphs on 10 leaflets per plot. In the dry season the least oviposited genotypes by B. tabaci biotype B were IAC-Una and LP 02-130, while the most oviposited were Z-28 and Pérola. The least presence of nymphs of whitefly was observed on LP 98-122 and FT-Nobre, and the most on LP 01-38, Perola and Z-28. For the rainy season the genotypes did not differ in relation to the attack of B. tabaci biotype B. The yield factors presented a statistical difference only in the dry season.

Técnicas de carburação em fornos rotativos a óleo de xisto e gás natural para a produção de ferros fundidos

A indústria metalúrgica tem buscado empregar processos de fusão mais eficientes. Os fornos rotativos a gás natural e óleo de xisto surgem como uma alternativa interessante para a produção de ligas ferrosas. Durante a fusão no forno rotativo acontece uma maior queima de elementos químicos, exigindo a utilização de técnicas de carburação adequadas. No presente trabalho foram avaliadas diferentes formas de adição de carburantes durante a fusão de ferros fundidos cinzentos e nodulares, utilizando fornos rotativos a óleo de xisto e gás natural. Dois tipos de carburantes, com diferentes granulometrias, foram adicionados em diferentes estágios do processo de fusão. Os resultados mostram que o rendimento de carburação é fortemente afetado pelo estado da carga metálica. O melhor rendimento foi obtido pelo carburante de coque de petróleo calcinado com granulometria de 1-3 mm, por meio da adição à carga no estado pastoso.
Metallurgic industry has sought for more efficient casting processes. Fuel oil and gas-fired rotary furnaces rise as an interesting alternative for cast irons production. It must be emphasized that during the fusion process in rotary furnaces, a higher burning of alloying elements occur, demanding the use of a suitable carbonizing technique in order to attend the chemical composition specifications. The present work aimed in the application and study of different carbonizing techniques during the production of gray and ductile cast irons using fuel oil and gas-fired rotary furnaces. Two carburant types with different particle sizes were added in different stages of fusion process. Results show that carbonizing efficiency is strongly affected by the state of the metallic charge. The best results were obtained by the calcined petroleum coke with a particle size of 1-3 mm, added to the pasty metal loading.

Cystic fibrosis (CF) is the most common genetic disease among Caucasians and is rare among sub-Saharan Africans. The Brazilian population is not ethnically homogeneous but it is the result of three-way ethnic admixture of Europeans, Africans and Amerindians in varying proportions, depending on the region. In the present study, we investigated 33 patients who had been diagnosed and are currently under treatment for CF at the University Hospital João de Barros Barreto, Belém, Pará State. The molecular analysis for G542X, G551D and R553X mutations was performed by PCR followed by RFLP using BstNI, HincII and MboI, respectively, in polyacrylamide gel eletrophoresis and stained with AgNO3. ThedeltaF508 mutation (a deletion of 3 bp) was only analyzed by polyacrylamide gel electrophoresis and stained with AgNO3. Each sample was analyzed for regions of interest in the CFTR gene using amplified by PCR and specific primers. The deltaF508 and G551D mutations presented frequencies of 22.7 and 3%, respectively. In 74.3% of the remaining patients, none of the mutations investigated was found. The present study characterized in a sample of patients with an established clinical diagnosis of CF (asthma, repeated bronchopneumonia, disorders of nutritional status, etc.) the most frequent mutation ( deltaF508) in the North region of Brazil and is also the first report of the G551D mutation. In spite of the wide spectrum of CF mutations and the heterogeneous ethnic origin of the Amazon population, the molecular diagnosis is a helpful additional tool for the diagnosis and treatment of CF patients.
