RESUMO: Testes in vitro são realizados para avaliar a eficácia de aditivos antimicotoxinas (AAMs); entretanto, tais experimentos apresentam uma baixa correlação com ensaios in vivo, que também são exigidos para determinar a eficácia de AAMs. Em busca de um método alternativo, o presente estudo investigou o uso de uma técnica ex vivo. A capacidade de seis AAMs (AAM1 a AAM6) de reduzir a absorção intestinal de aflatoxina B1 (AFB1) foi avaliada. Explantes jejunais foram coletados de frangos de corte e submetidos a dois tratamentos por AAM em câmaras de Ussing: T1 (controle) - 2,8 mg/L AFB1, e T2 - 2.8 mg/L AFB1 + 0,5% AAM. Os AAMs também foram testados in vitro para verificar a adsorção de AFB1 em fluido intestinal artificial. Nos ensaios ex vivo, AAM1 ao AAM6 diminuíram a absorção intestinal de AFB1 em 67,11%, 73,82%, 80,70%, 85,86%, 86,28% e 82,32%, respectivamente. Quanto aos achados in vitro, AAM1 ao AAM6 apresentaram adsorção de 99,72%, 99,37%, 99,67%, 99,53%, 99,04% e 99,15%, respectivamente. O modelo ex vivo avaliado mostrou-se eficiente na avaliação de AAMs. Não houve correlação entre os resultados ex vivo e in vitro. Estudos adicionais são necessários para definir a correlação entre achados ex vivo e in vivo na tentativa de reduzir os testes em animais.

ABSTRACT: In vitro tests are performed to evaluate the efficacy of antimycotoxins additives (AMAs); nevertheless, such assays show a low correlation with in vivo trials, which are also required to determine AMAs’ efficacy. In search of an alternative method, the current study investigated the use of an ex vivo technique. Six AMAs (AMA1 to AMA6) had their ability to reduce intestinal absorption of aflatoxin B1 (AFB1) evaluated. Jejunal explants were obtained from broilers and subjected to two treatments per AMA in Ussing chambers: T1 (control) - 2.8 mg/L AFB1, and T2 - 2.8 mg/L AFB1 + 0.5% AMA. AMAs were also tested in vitro to assess adsorption of AFB1 in artificial intestinal fluid. In the ex vivo studies, AMA1 to AMA6 decreased intestinal absorption of AFB1 by 67.11%, 73.82%, 80.70%, 85.86%, 86.28% and 82.32%, respectively. As for the in vitro results, AMA1 to AMA6 presented an adsorption of 99.72%, 99.37%, 99.67%, 99.53%, 99.04% and 99.15%, respectively. The evaluated ex vivo model proved useful in the assessment of AMAs. No correlation was reported between ex vivo and in vitro findings. Further studies are needed to elucidate the correlation between ex vivo and in vivo results seeking to reduce animal testing.

RESUMO: O milho roxo é um alimento importante para o povo peruano. Suas características nutricionais e antioxidantes únicas fazem com que seja amplamente exportado para outros países. No entanto, quando contaminado por fungos, pode desencadear inúmeros problemas de saúde nos consumidores. Este estudo teve como objetivo avaliar a presença de 27 micotoxinas em 63 amostras de milho roxo coletadas no Peru. A frequência de ocorrência e a concentração média das seguintes micotoxinas foram determinadas: alternariolmetileter (AME), alternariol (AOH), tentoxina, neosolaniol, nivalenol, wortmanina, desoxinivalenol, 3-acetil desoxinivalenol, 15-acetil desoxinivalenol, zearalenona, aflatoxina B1, aflatoxina B2 , aflatoxina G1, aflatoxina G2, fumonisina B1, fumonisina B2, fumonisina B3, ocratoxina A, ocratoxina, toxina T-2, toxina HT-2, fusarenona x, ácido ciclopiazonico, gliotoxina, agroclavina e citreoviridina. As principais micotoxinas encontradas no milho roxo foram AME e AOH, com frequência de ocorrência de 14,3% e 7,9% e concentração média de 23,3% e 1,8%, respectivamente. AME e AOH não possuem níveis de orientação na legislação brasileira. Contrastantemente, os níveis de micotoxinas que estão dentro dos padrões das regulamentações do país estavam abaixo do limite de quantificação. Os presentes resultados sugerem que o milho roxo é uma matéria-prima com grande potencial para a produção e industrialização de produtos especiais.

ABSTRACT: Purple maize is an important foodstuff for the Peruvian people. Its unique nutritional and antioxidant characteristics makes it widely exported to other countries. However, when contaminated by fungi, it can trigger numerous health problems in the consumers. This study aimed to evaluate the presence of 27 mycotoxins in 63 samples of purple maize collected in Peru. Frequency of occurrence and mean concentration of the following mycotoxins were determined: alternariolmetileter (AME), alternariol (AOH), tentoxin, neosolaniol, nivalenol, wortmannin, deoxynivalenol, 3-acetyl deoxynivalenol, 15-acetyl deoxynivalenol, zearalenone, aflatoxin B1, aflatoxin B2, aflatoxin G1, aflatoxin G2, fumonisin B1, fumonisin B2, fumonisin B3, ochratoxin A, ochratoxin α, T-2 toxin, HT-2 toxin, fusarenon x, cyclopiazonic acid, gliotoxin, agroclavin and citreoviridin. The main mycotoxins reported in purple maize were AME and AOH, with a frequency of occurrence of 14.3 and 7.9%, and mean concentration of 23.3% and 1.8%, respectively. AME and AOH do not have guidance levels in the Brazilian legislation. Contrastingly, levels of mycotoxins which are within the standards of the country’s regulations were below the limit of quantification. The present results suggested that purple maize is a raw material with a great potential for the production and industrialization of special products.

RESUMO: Nos últimos anos, o Brasil tem incentivado o cultivo de pecãs para atender a demanda do mercado nacional e internacional de nozes. Novas variedades genéticas de pecãs foram selecionadas nos últimos anos, mas informações científicas disponíveis sobre a ocorrência de fungos e aflatoxinas na literatura internacional estão desatualizadas. Portanto, o presente estudo objetivou quantificar e identificar a microbiota fúngica e a presença de aflatoxinas em nozes cultivadas no sul do Brasil. Cinquenta e dois lotes de nozes (variedade Barton) foram obtidos de produtores de 19 municípios do Estado do Rio Grande do Sul e analisados por meio de Ágar Dicloran Glicerol 18% (DG18) e Aspergillus Flavus e Parasiticus Agar (AFPA), após incubação em 25 °C durante 7 dias. Análises de aflatoxinas foram realizadas usando HPLC acoplado a um espectrômetro de massa. Os resultados revelaram pelo menos 10 gêneros diferentes de fungos. Aspergillus, Penicillium, Fusarium e Cladosporium foram predominantes. Espécies xerofílicas de Aspergillus (A. wentii, A. ruber, A. pseudoglaucus e A. chevalieri) foram comumente encontradas nas amostras. Nenhuma espécie potencial produtora de aflatoxinas foi isolada e nenhuma aflatoxina foi detectada (LOQ=1 μg/kg e LOD=0,1 μg/kg para AFB1 e AFB2; e 0,3 μg/kg para AFG1 e AFG2) nas amostras avaliadas. A ausência desse grupo carcinogênico de micotoxinas é altamente positiva e pode impulsionar os investimentos no setor, além de estimular a comercialização e o consumo dessa variedade de nozes.

ABSTRACT: In recent years, Brazil has encouraged the cultivation of pecans to meet both demands of domestic and international market of nuts. New genetic varieties of pecans have been selected in recent years, but available scientific information on the occurrence of fungi and aflatoxins in the international literature is out of date. Therefore, the present study aimed to quantify and identify fungal microbiota and the presence of aflatoxins in pecan nuts cultivated in southern Brazil. Fifty-two pecan nut lots (Barton variety) were obtained from producers from 19 cities of Rio Grande do Sul State and analyzed by direct plating in Agar Dichloran Glycerol 18% (DG18) and Aspergillus Flavus and Parasiticus Agar (AFPA), following incubation at 25 °C for 7 days. Aflatoxins analyses were carried out using HPLC coupled with a mass spectrometer. Results revealed at least 10 different genera of fungi. Aspergillus, Penicillium, Fusarium, and Cladosporium were predominant. Xerophilic species of Aspergillus (A. wentii, A. ruber, A. pseudoglaucus, and A. chevalieri) were commonly reported in the samples. No potential aflatoxin-producing species was isolated and no aflatoxins were detected (LOQ=1 μg/kg and LOD=0.1 μg/kg for AFB1 and AFB2, and 0.3 μg/kg for AFG1 and AFG2) in the evaluated samples. The absence of this carcinogenic group of mycotoxins is highly positive and could boost the investments in the sector, as well as stimulate the commercialization and consumption of this variety of nut.

Abstract Aspergillus westerdijkiae is one of the most important spoilage and toxigenic fungi contaminating coffee beans and may produce ochratoxin A (OTA), a mycotoxin that characterize a health risk to the coffee consumers. Biological control strategies can be used for prevention of fungal invasion and decrease mycotoxin exposure. The aims of this study were to evaluate the in vitro effect of three Bacillus sp. biocontrol candidates on A. westerdijkiae mycelial growth, spore counts and OTA production. A green-coffee based medium was inoculated with A. westerdijkiae and Bacillus spp. (B. safensis RF69, B. amyloliquefaciens RP103 and B. subtilis RP242) and after incubation, the fungal growth, sporulation and mycotoxin production was evaluated. Mycelial growth rate was reduced in a range between 76-95% and conidial production was also significantly decreased. All isolates were capable of reducing OTA production in a range between 62-96%. The results showed that the biocontrol candidates may be an effective control method for A. westerdijkiae and OTA in coffee.

Abstract Fusarium head blight is an important disease occurring in wheat, caused mainly by the fungus Fusarium graminearum. In addition to direct damage to crops, reduced quality and yield losses, the infected grains can accumulate mycotoxins (toxic metabolites originating from prior fungal growth), especially deoxynivalenol (DON). Wheat crops harvested in 2014/2015 in southern Brazil were affected by high levels of Fusarium head blight. In this context, the aim of this study was evaluate the mycotoxicological quality of Brazilian wheat grains and wheat products (wheat flour and wheat bran for DON. DON contamination was evaluated in 1,504 wheat and wheat product samples produced in Brazil during 2014. It was determined by high performance liquid chromatograph fitted to a mass spectrometer (LC-MS / MS). The results showed that 1,000 (66.5%) out of the total samples tested were positive for DON. The mean level of sample contamination was 1047 µg.kg-1, but only 242 samples (16.1%) had contamination levels above the maximum permissible levels (MPL) - the maximum content allowed by current Brazilian regulation. As of 2017, MPL will be stricter. Thus, research should be conducted on DON contamination of wheat and wheat products, since wheat is a raw material widely used in the food industry, and DON can cause serious harm to public health.

RESUMO O objetivo deste trabalho foi avaliar a atividade anestésica do óleo essencial (EO) de Ocimum americanum L. em jundiás (Rhamdia quelen ). No primeiro experimento, os efeitos depressores e a composição química dos óleos obtidos a partir das folhas (LEO) e das inflorescências (IEO) desta espécie vegetal foram comparados. Para isto, juvenis de jundiás (n = 10) foram transferidos para aquários contendo diferentes concentrações de EO (25 - 500 mg L-1) a fim de determinar os tempos de indução aos estágios de anestesia e de recuperação. No experimento seguinte foram determinados os efeitos da exposição a 300 e 500 mg L-1 de LEO em parâmetros de estresse (cortisol, glicose e níveis de sódio plasmáticos) após os animais serem expostos durante 1 min ao ar. Os peixes (n = 10 por tempo de amostragem) foram coletados imediatamente ou transferidos para aquários livres de anestésico até amostragem (15, 30, 60 ou 240 min). LEO demonstrou ser composto majoritariamente por β-linalol e 1,8-cineol em proporções similares, enquanto que IEO apresentou β-linalol como constituinte majoritário. O estágio de anestesia foi verificado em jundiás com 200-500 mg L-1 entre 4-8 min para LEO e 6-16 min para IEO. Menores concentrações de ambos os EO não promoveram anesthesia até 30 min. O uso de LEO como anestésico preveniu o aumento de cortisol e a perda de sódio induzida pela exposição aérea. Já a glicemia encontrava-se aumentada em peixes expostos ao LEO em comparação ao grupo basal (não exposto ao ar) na maioria dos tempos de observação. Assim, EO de O. americanum obtido a partir das folhas foi considerado adequado para procedimentos anestésicos devido a sua rápida indução e capacidade de prevenir o estresse decorrente do manuseio.

ABSTRACT The aim of this study was to evaluate the anesthetic activity of the essential oil (EO) of Ocimum americanum L. in silver catfish (Rhamdia quelen ). In the first experiment, the depressor effects and chemical composition of the leaf EO (LEO) and inflorescence EO (IEO) were compared. Juveniles (n = 10) were placed in aquaria containing different concentrations of EO (25 - 500 mg L-1) to determine the point at which anesthesia was induced and the length of the recovery period. In the following experiment, the effects of 300 and 500 mg L-1 LEO exposure on stress parameters (plasma cortisol, glucose and sodium levels) after air exposure for 1 min were assayed. Fish (n = 10 per sampling time) were sampled immediately or transferred to anesthetic-free aquaria until sampling (15, 30, 60 or 240 min). LEO was composed mainly of β-linalool and 1,8-cineole in similar proportions, whereas IEO showed β-linalool as major compound. Anesthesia was obtained in silver catfish with 200-500 mg L-1 between 4-8 min for LEO and 6-16 min for IEO. Lower EO concentrations did not reach anesthetic stage up to 30 min. LEO used as anesthetic prevented the cortisol increase and sodium loss induced by aerial exposure. Glucose levels were raised in catfish exposed to LEO compared to basal group (not air exposed) in almost all observation times. EO of O. americanum obtained from leaves was considered suitable to anesthetic procedures due to its fast induction and handling-induced stress prevention.

O objetivo do presente estudo foi avaliar o efeito da aspersão de extratos de Lippia alba na estabilidade lipídica de filés de carpa húngara armazenados a -18±2°C. Filés não tratados (controle sem aspersão) ou aspergidos (1mL 10g-1 de filé) com água destilada (controle água destilada) ou com extratos de L. alba (0,10g mL-1) hidrometanólico ou aquoso foram analisados durante o armazenamento nos dias zero, 90 e 180. Independente do tempo de congelamento, o extrato hidrometanólico reduziu os valores de dienos conjugados (DC) dos filés em relação aos demais tratamentos, além de reduzir os valores de ácidos graxos livres aos 90 dias (P<0,05). O extrato aquoso resultou em maior teor de peróxidos após 180 dias de congelamento comparado aos demais tratamentos (P<0,05). Os extratos hidrometanólico e aquoso reduziram os valores de substâncias reativas ao ácido tiobarbitúrico (TBARS) após 180 dias, comparados aos filés tratados com água destilada e sem aspersão (P<0,05). Ambos os extratos de L. alba retardaram a oxidação lipídica, sendo que o extrato aquoso retardou a degradação de produtos primários da oxidação lipídica (peróxidos) em produtos secundários (TBARS), enquanto o extrato hidrometanólico parece ser mais eficiente, pois inibiu de forma similar tanto a formação de DC e peróxidos, quanto a sua degradação em produtos secundários (TBARS)

The effect of sprinkling with Lippia alba extracts was evaluated on the lipid stability of common carp fillets stored at -18±2°C. Fillets that received no treatment (no spray control) or that were sprayed (1mL 10g-1 fillet) with distilled water (water control) or with hydro-methanolic or aqueous extract of L. alba (0.10g mL-1) were evaluated immediately (time zero) and after 90 and 180 days. Regardless of the storage time, the hydro-methanolic extract reduced the conjugated dienes (CD) values of fillets compared to the other treatments, and reduced the free fatty acid levels at 90 days (P<0.05). The aqueous extract caused higher peroxide value after 180 days of frozen storage compared to the other treatments (P<0.05). The hydro-methanolic and aqueous extracts reduced thiobarbituric acid reactive substances (TBARS) values after 180 days compared to the non-treated fillets or to the water-sprayed fillets (P<0.05). Both extracts have delayed lipid oxidation. While the aqueous L. alba extract delayed the degradation of primary oxidation products (peroxides) into secondary products (TBARS), the hydro-methanolic extract was more efficient as it inhibited both the CD and peroxide formation and its degradation into secondary products (TBARS)

This study evaluated the seasonal influence on the yield and chemical composition of the essential oil (EO) of Nectandra megapotamica. Fresh young (YL) and old leaves (OL) obtained from three trees in each season (Nov/2010 to Sep/2011) collected in Santa Maria-RS were hydrodistilled in triplicate. The chemical composition was determined by the gas chromatography coupled to mass spectrometry (GC-MS) and the yield on dry basis was evaluated by two-way ANOVA (seasons, development stage). Spring (Sp) and summer (Su) showed higher average incomes (0.45 and 0.33%), which occurred when flowering, fruiting, and growth of YL and senescence of OL took place, while autumn (Au) presented the lowest yield (0.25%) during the rustification of OL. The highest yield was obtained for the YL in Sp (0.59%) and the lowest for the OL in Au (0.21%). The major constituents of the EO were independent from the season and were identified as α-pinene, bicyclogermacrene, β-pinene, germacrene D, and limonene. Seasonality and phenology influenced the production of EO probably due to morphological and metabolic alterations in the leaves as well as due to the needs of the tree, such as attraction and/or protection.

Variance and performance of two sampling plans for aflatoxins quantification in maize were evaluated. Eight lots of maize were sampled using two plans: manual, using sampling spear for kernels; and automatic, using a continuous flow to collect milled maize. Total variance and sampling, preparation, and analysis variance were determined and compared between plans through multifactor analysis of variance. Four theoretical distribution models were used to compare aflatoxins quantification distributions in eight maize lots. The acceptance and rejection probabilities for a lot under certain aflatoxin concentration were determined using variance and the information on the selected distribution model to build the operational characteristic curves (OC). Sampling and total variance were lower at the automatic plan. The OC curve from the automatic plan reduced both consumer and producer risks in comparison to the manual plan. The automatic plan is more efficient than the manual one because it expresses more accurately the real aflatoxin contamination in maize.

Este estudo analisou a composição química e o potencial anestésico do óleo essencial (OE) de Nectandra megapotamica em robalos-peva (Centropomus parallelus). Para a extração do OE por hidrodestilação, as folhas foram separadas em jovens (OE-J) ou velhas (OE-V) e a composição química foi determinada por CG-EM. O potencial anestésico foi acessado através da avaliação do tempo de indução e recuperação da anestesia e resposta ao estresse do procedimento anestésico e transporte. Foram realizados três experimentos: em primeiro lugar, quatro concentrações diferentes de cada OE foram testadas para avaliar o tempo de indução à anestesia e de recuperação; em segundo lugar, duas concentrações do OE-V foram testadas para avaliar os efeitos sobre os parâmetros de estresse (níveis plasmáticos de glicose, lactato, Na+ e K+) causados pelo procedimento anestésico; em terceiro lugar, os peixes foram transportados em sacos plásticos com duas concentrações do OE-V para avaliação da qualidade da água e mortalidade. Todos os experimentos foram realizados em peixes aclimatados à salinidade zero e 33. Os constituintes majoritários do OE-J e OE-V foram: biciclogermacreno (46,5/34,6%), α-pineno (26,8/26,2%), β-pineno (7,9/12,3%) e germacreno D (9,6/9,1%). Sedação leve foi alcançada com 30 °L L-1(1,3-3,2 min) e anestesia profunda a partir de 150 °L L-1(5,6-8,0 min) com ambos OEs. O tempo de recuperação variou entre 1-10 min.O OE-V não foi capaz de evitar o estresse do procedimento anestésico, evidenciado pelos elevados níveis plasmáticos de glicose e lactato observados em todos os grupos. Os níveis plasmáticos de Na+ e K+ não foram significativamente afetados pelos tratamentos. Durante o transporte, o OE-V não impediu a deterioração da qualidade da água e a mortalidade pós-transporte. Concluindo, o OE de Nectandra megapotamicaapresenta atividade anestésica em robalos-peva, mas não foi capaz de evitar o estresse do procedimento anestésico e transporte.

This study analyzed the chemical composition and anesthetic potential of essential oil (EO) of Nectandra megapotamica in fat snook (Centropomus parallelus). For the extraction of EO by hydrodistillation, leaves were separated in young (EO-Y) or old (EO-O), and the chemical composition of the EOs was determined by CG-MS. The anesthetic potential was assessed by the evaluation of induction and recovery time of anesthesia and stress response from anesthesia and transport. Three experiments were carried out: i) four different concentrations of each EO were tested to evaluate anesthesia induction and recovery time; ii) two concentrations of EO-O were tested for the evaluation of its effects on stress parameters (glucose, lactate, and Na+ and K+ plasma levels) caused by anesthesia; and iii) fish were transported in plastic bags, supplied with two concentrations of EO-O for the evaluation of water quality and mortality. All experiments were performed on fish acclimated to 0 and 33 ppt salinity. The main constituents of the Y and O-EOs were bicyclogermacrene (46.5/34.6%), α-pinene (26.8/26.2%), β-pinene (7.9/12.3%), and germacrene D (9.6/9.1%). Mild sedation was achieved at 30 °L L-1(1.3-3.2 min) and deep anesthesia at 150 °L L-1(5.6-8.0 min) with both EOs. The recovery time ranged from 1-10 min. The EO-O was not able to avoid the stress of anesthesia evidenced by elevated glucose and lactate plasma levels observed in all groups. Plasma levels of Na+ and K+ were not significantly affected by treatments. During transport, the use of EO-O did not prevent deterioration in water quality and the post-transport mortality. In conclusion, the EO of N. megapotamica has anesthetic activity in fat snook, but it was not able to prevent the stress of anesthesia and transport.

O objetivo deste estudo foi avaliar fontes de concentrados proteicos vegetais alternativos em substituição ao farelo de soja sobre o desempenho e composição centesimal de jundiás (Rhamdia quelen) e coloração dos filés. Foram utilizadas 20 unidades experimentais, com capacidade de 90L cada uma, onde 400 peixes permaneceram alojados durante 45 dias. Os juvenis de jundiá possuíam 11,5±5,0cm e peso inicial de 10,4±5,5g. Foram avaliadas quatro fontes de concentrados proteicos, sendo eles: dieta controle, tendo o farelo de soja com base proteica; CL: dieta contendo concentrado proteico de crambe + concentrado proteico de linhaça; CLGM: composta por concentrado proteico de crambe + concentrado proteico de linhaça + concentrado proteico de folha de mandioca + concentrado proteico de farelo de girassol e GM: constituída por concentrado proteico de farelo de girassol + concentrado proteico de folha de mandioca. Aos 30 e 45 dias, foram avaliados os parâmetros de crescimento (peso, fator de condição, taxa de crescimento específico, ganho em peso diário e relativo, taxa de eficiência proteica e conversão alimentar aparente) e avaliação centesimal do peixe inteiro e filé (umidade, proteína bruta, gordura e cinzas). Além disso, aos 45 dias, foi realizada a avaliação instrumental da cor nos filés. Ao final do período experimental, foram observadas diferenças significativas para os parâmetros de crescimento e avaliação centesimal, sendo que os animais alimentados com as dietas CL apresentaram maior crescimento (13,8cm), taxa de crescimento específico (2,43% dia-1), ganho em peso diário (0,41g dia-1), ganho em peso relativo (108,3%) e menor conversão alimentar (1,21), comparado às dietas contendo concentrado proteico de girassol e concentrado proteico de folhas de mandioca. Observou-se também que os animais alimentados com as dietas GM e CLGM apresentaram uma pigmentação amarelada nos filés. Conclui-se que os animais alimentados com a dieta CL obtiveram os maiores índices de desempenhos, podendo ser utilizada como alimento na formulação de dietas para esta espécie. As demais combinações não redundaram em resultados melhores que a dieta controle e CL.

The aim of this study was to evaluate vegetable protein concentrates to replace soybean meal (traditional protein) on performance and chemical composition of catfish (Rhamdia quelen) and its influence on the coloration of the fillets. We used 20 experimental units (EU), with a capacity of 90L each, where 400 fish were rearing for 45 days. Juvenile catfish had 11.5±5.0cm and an initial weight of 10.4±5.5g. We evaluated four diets with different sources of protein concentrates, which are: Control, conventional diet using soybean meal as source of protein; CL: diet containing protein concentrate crambe meal protein concentrate; CLGM: protein concentrate consisting of crambe meal protein concentrate protein concentrate cassava leaf protein concentrate and GM sunflower meal: protein concentrate consisting of sunflower meal protein concentrate cassava leaf. At 30 and 45 days were evaluated growth parameters (weight, condition factor, specific growth rate, daily weight gain and relative protein efficiency ratio and feed conversion) and proximate evaluation of the whole fish and fillet (humidity, crude protein, fat and ash). In addition, the 45-day evaluation was made color instrumental in fillets. At the end of the experiment, significant differences were observed for growth parameters and proximate, and the animals fed the LC diets had higher growth (13,8cm), specific growth rate (2,43% day-1), daily weight gain (0,41g day-1), relative weight gain (108,3%) and lower feed conversion (1,21) compared to diets containing protein concentrate of sunflower and cassava leaves. We also observed that animals fed on GM diets and CLGM showed a yellowish pigment in the fillets. It is concluded that animals fed the CL diet had the highest levels of performance and can be used as food in the formulation of diets for this species. All others combinations do not showed no better results than the control diet and CL.

Existe uma crescente demanda por anestésicos baratos e seguros capazes de reduzir o estresse em peixes produzido durante procedimentos como captura e manuseio. Neste contexto, o presente estudo avaliou o potencial como anestésico dos óleos essenciais (EO) de três espécies vegetais nativas (Hesperozygis ringens, Lippia sidoides e Ocotea acutifolia) em jundiás - Rhamdia quelen. Adicionalmente, a composição química desses óleos e suas influências sobre o estresse também foram avaliadas. Os EO de H. ringens e O. acutifolia foram efetivos como anestésicos sem efeitos adversos detectáveis. EO de O. acutifolia (150 µL L-1) promoveu um aumento na glicemia. Em relação a sua composição, pulegona correspondeu a 96,63% do EO de H. ringens, e óxido de cariofileno a 56,90% do EO de O. acutifolia. Dois quimiotipos, timol e carvacrol (68,40% e 67,89%, respectivamente) foram verificados para os EO de L. sidoides. Ambas as amostras de EO de L. sidoides apresentaram atividade anestésica em jundiás, contudo a exposição produziu perda de muco e mortalidade. Desta forma, somente os EO de H. ringens e O. acutifolia têm seu uso recomendável como anestésicos

There is an increasing demand for inexpensive and safe anesthetics that can reduce fish stress caused by some procedures such as capture and handling. In this context, the present study evaluated the potential of essential oils (EO) of three Brazilian native plants (Hesperozygis ringens, Lippia sidoides and Ocotea acutifolia) as anesthetics for the silver catfish - Rhamdia quelen. Moreover, an analysis was made of the chemical composition of these oils and their influence on stress parameter. EO of H. ringens and O. acutifolia were effective as anesthetics, without behavioral side effects. EO of O. acutifolia (150 µL L-1) promoted an increase in blood glucose level. Regarding to the composition, pulegone accounts for 96.63% of the EO of H. ringens, and caryophyllene oxide amounts to 56.90% of the EO of O. acutifolia. Two chemotypes, thymol and carvacrol (68.40% and 67.89%, respectively) were verified for EO of L. sidoides. Both samples of EO of L. sidoides showed anesthetic activity in silver catfish, but exposure also caused loss of mucus and mortality. Thus, only the EO of H. ringens and O. acutifolia are advised for anesthetic use

O objetivo deste trabalho foi avaliar a eficiência de dois planos de amostragem para análise de fumonisinas (B1+B2) em 11 lotes de milho. Empregaram-se o plano de amostragem manual em grãos inteiros, através do calador graneleiro e o plano de Amostragem Automático em Fluxo Contínuo em grãos moídos (AAFC). Para a comparação dos dois planos de amostragem, foram utilizadas duas medidas de dispersão: a variância e o coeficiente de variação. Em cada plano de amostragem, foram determinadas as variâncias associadas às três etapas do procedimento de detecção das fumonisinas (amostragem, preparação da amostra e análise) e os coeficientes de variação total. A correlação entre as médias de concentrações de fumonisinas e as variâncias de cada etapa do procedimento de detecção foi avaliada para cada plano amostral através da análise de regressão. Na AAFC de milho moído para análise de fumonisinas, obtiveram-se resultados inferiores (P<0,01) de variância na etapa de amostragem (0,0226mg kg-1)², assim como no coeficiente de variação total (6,37%), quando comparados com a variância da amostragem manual (0,0685mg kg-1)² e coeficiente de variação total do plano de amostragem manual em grãos inteiros (8,94%), apresentando, portanto, maior eficiência na detecção de fumonisinas em milho.

The aim of this research was to evaluate the efficiency of two sampling plans for fumonisins (B1+B2) analysis in 11 lots of maize. For sample collection the two systems used were a manual system in whole grains, using sampling spear, and an automatic system in milled grains, using the continuous flow sampling system (CFSS). For comparison purpose, two dispersion measurements were used: variance and coefficient of variation. For each sampling plan the variance associated to the three steps of fumonisins quantification (sampling, sample preparation, and analysis), and the total coefficient of variation were determined. The correlation between the average fumonisins concentration and the variances for each phase of the detection procedure was evaluated for each sampling plan using analysis of regression. At the automatic sampling plan in milled grains for fumonisins analysis in maize both sampling variance (0.0226mg kg-1)² and total coefficient of variation (6.37%) were lower (P<0.01) than in the manual sampling plan in whole grains whit (0.0685mg kg-1)² of variance and 8.94% of coefficient of variation, showing so better efficiency for fumonisin detection on maize.

Das folhas de Senecio bonariensis Hook. & Arn. foram isolados três sesquiterpenóides. β-Cariofileno (1), óxido de β-cariofileno (2) e germacreno D (3) foram identificados por CG-IE-EM, IV, ¹H and 13C-RMN. A importância ecológica e quimiotaxionômica para o gênero Senecio é discutida, assim como a correlação biossintética entre as substâncias isoladas. As substâncias 1 e 2 tiveram sua atividade antimicrobiana avaliada pelo método de microdiluição em caldo contra cepas bacterianas e fúngicas. Ambas não inibiram o crescimento microbiano nas concentrações testadas.

From the leaves of Senecio bonariensis Hook. & Arn. three sesquiterpenoids were isolated. β-caryophyllene (1), β-caryophyllene oxide (2) and germacrene D (3) were characterized by GC-EI-MS, IR, ¹H and 13C-NMR data. Their ecological and chemotaxonomical significance for the genus Senecio are discussed, as well as the biosynthetic correlation between the isolated substances. The antimicrobial activity of substances 1 and 2 were evaluated by the microdilution method against bacterial and fungal strains. Both compounds did not inhibit the microorganism growth at the tested concentrations.

Neste trabalho foi avaliado o efeito de um adsorvente (aluminossilicato de cálcio e sódio) no desempenho de alevinos de jundiá (Rhamdia quelen) alimentados com rações contendo níveis crescentes de aflatoxina. Foram utilizados 360 peixes com peso inicial de 4,2 ± 0,6 g, criados em sistema de recirculação da água termorregulada, durante 90 dias, e alimentados com rações contendo um dos quatro níveis de aflatoxinas (0, 150, 250 e 350 µg/kg), com e sem inclusão de adsorvente (0, 0,3 e 0,6%), com três repetições. O uso de aflatoxina teve efeito negativo sobre o crescimento e o ganho de peso dos alevinos, que reduziram proporcionalmente aos níveis de aflatoxina, mas não ocasionou mortalidade. Os níveis de adsorvente na dieta não diminuíram os efeitos das aflatoxinas no desempenho dos peixes, mas diminuíram a aceitação da ração, em virtude da palatabilidade do produto. Não é recomendável a utilização de aluminossilicato de cálcio e sódio como adsorvente de aflatoxinas em rações para alevinos de jundiá.

The effects of aflatoxins and one adsorbent (Sodium and calcium aluminum silicate) on silver catfish (Rhamdia quelen) fingerlings were evaluated in this study. Three hundred and sixty fish, with initial weight of 4 g, were raised in a thermo-regulated recirculation water system, during 90 days. Twelve treatments with four aflatoxin levels in diet (0, 150, 250 and 350 µg/kg), with and without adsorbents (0, 0.3 and 0.6%) were tested in three replications. The results showed that the negative effect of aflatoxins significantly reduced the growth and weight gain of fingerlings, proportionately to the increasing levels of aflatoxin in diet, with mortality occurrence. The adsorbent levels in diet did not diminish the effects of aflatoxin action on fish performance due to the low inclusion and also little acceptance of ration in relation to the product palatability. It is concluded that silver catfish fingerlings fed diets containing aflatoxin are susceptible to negative effects, with high losses in growth and weight gain.
