Abstract This study aimed to investigate the antimicrobial activity of kefir fermented fruit by-products against strains of Alicyclobacillus spp., and determine their chemical characterization and antioxidant activity. According to the results, extracts fermented for a longer period (72 h) showed greater inhibition, and the extract of acerola by-product fermented for 72 h achieved the best results. For all strains, the Minimal Inhibitory Concentration (MIC) was 0.78%, except for A. acidocaldarius subsp. rittmannii, (1.56%). The same applies to the Minimal Bactericidal Concentration (MBC), in which 1.56% of the extract was capable of inactivating A. cycloheptanicus and A. acidocaldarius. In addition, damages to the structure of the microorganisms caused by the extracts were detected by Scanning Electron Microscopy. Metabolite identification through liquid chromatography (UHPLC-Qtof-MS) showed that the fermented extracts had a greater number of compounds compared to the non-fermented ones, such as glucuronic, succinic and glutaric acids. In conclusion, the extracts of fruit by-products showed to have bioactive properties, such as antibacterial potential and antioxidant activity against the Alicyclobacillus strains tested in this study, not to mention the value added by the use of a food by-product.

This study aimed to investigate a miniaturized extraction methodology with analysis by GC-MS for simultaneous detection and quantification of cocaine (COC), benzoylecgonine (BZE) and cocaethylene (CE) in hair to monitor drug addicts. The process was done following the international guide of the Scientific Task Force on Forensic Toxicology. After sample extraction, derivatizing solution was added, clean-up was done by the Hollow Fiber Liquid Phase Microextraction (HF-LPME) and adapted to use the ultrasonic bath, a simple and easy-to-handle device. Isobutylchloroformate was first used as derivatization reagent to convert benzoylecgonine to isobutylbenzoylecgonine. Analytes quantification occurred within a linear range of 0.1 to 20 ng mg-1 for COC and 0.05 to 5 ng mg-1 for BZE and CE, with a correlation coefficient of r > 0.99. Limits of detection were 0.05; 0.03 and 0.03 ng mg-1, whereas limits of quantification were 0.1; 0.05 and 0.05 ng mg-1 for COC; BZE and CE, respectively. There was no matrix effect interference. Intra and inter-day precision and accuracy were acceptable according to international guidelines. The analytical method HF-LPME-GC-MS was successfully applied to 14 hair samples from patients admitted in drugs detoxification programs. All samples resulted positive for cocaine (0.80-> 20 ng mg-1) and benzoylecgonine (0.20-> 5 ng mg-1), 11 samples were positive for cocaethylene (0.10-> 0.60 ng mg-1).

Abstract The present study describes a method for simultaneous analysis of cocaine, benzoylecgonine and cocaethylene in urine samples. After solid phase extraction, Gas Chromatography-Mass Spectrometry was used for identification and quantification. The calibration curves were linear at 20 - 3000 ng/mL, r2 0.9997 for benzoylecgonine, 15 - 2000 ng/mL, r2 0.9985 and r2 0.9993 for cocaine and cocaethylene, respectively. Accuracy values: cocaine: 93,5 - 102,1%; benzoylecgonine: 97,5 - 104,8%; cocaethylene: 90,6 - 101,5%. Precision values: cocaine: C.V 5,4 - 14,6%; benzoylecgonine: C.V 7,8 - 12,3%; cocaethylene: C.V 5,9 - 12,3%. Detection and quantification limit values: cocaine and cocaethylene:10 ng/mL and 15 ng/mL, respectivalty; benzoylecgonine:15 ng/mL and 20 ng/mL, respectivaley. Recovery values: cocaine: 78,0 - 85,8%; benzoylecgonine: 74,0 - 79,8%; cocaethylene: 83,0 - 91,5%. The method described is advantageous compared to others, as it simultaneously detects the major analytes found in urine samples due to cocaine use and has been successfully validated.

RESUMO: O objetivo deste estudo foi avaliar o efeito da carga microbiana inicial, temperatura e tempo de contato na formação de biofilme de Alicyclobacillus acidoterrestris em aço inoxidável e borracha natural de qualidade alimentar utilizando suco de laranja como meio de cultura. A baixa carga inicial de A. acidoterrestris (2 log UFC/mL) levou à formação de biofilme na superfície do aço inoxidável após 48 h de contato a 28 ºC e após 24 h a 45 ºC, e na superfície natural de borracha de qualidade alimentar após 48 h de contato nas duas temperaturas. A alta carga microbiana inicial (5 log UFC/mL) levou à formação de biofilme em aço inoxidável após 4 h de contato a 28 °C e 45 °C, enquanto o biofilme foi formado em borracha natural de qualidade alimentar após 8 h de contato a 28 °C e 4 h a 45 °C. A carga microbiana também afetou a presença de esporos no biofilme, o que foi observado em ambas as superfícies apenas com altas cargas iniciais de A. acidoterrestris.

ABSTRACT: The objective of this study was to evaluate the effect of the initial microbial load, temperature and contact time on the biofilm formation of Alicyclobacillus acidoterrestris on stainless steel and natural food-grade rubber using orange juice as culture medium. The low initial load of A. acidoterrestris (2 log CFU/mL) led to biofilm formation on the stainless steel surface after 48 h of contact at 28 ºC and after 24 h at 45 ºC, and on natural food-grade rubber surface after 48 h of contact at both temperatures. The high initial microbial load (5 log CFU/mL) led to biofilm formation on stainless steel after 4 h of contact at 28 °C and 45 °C, while biofilm was formed on natural food-grade rubber after 8 h of contact at 28 °C and 4 h at 45 °C. The microbial load also affected the presence of spores in biofilm, which was observed on both surfaces only at high initial loads of A. acidoterrestris.

ABSTRACT The present study evaluated the antibacterial and antibiofilm activity of carvacrol against Salmonella Typhimurium. The minimum inhibitory concentration (MIC) and minimum bactericidal concentration (MBC) were determined and the time-kill curve and scanning electron microscopy (SEM) were performed to evaluate antibacterial activity. Antibiofilm activity was evaluated by quantifying total biomass using crystal violet assay, and metabolic activity was determined using MTT [3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide] assay. The action of carvacrol against preformed biofilm on polypropylene and stainless steel was also evaluated by colony counting and SEM. The MIC and MBC was 312 µg mL-1. Carvacrol at MIC and 2 x MIC eliminated cells after 6 and 1 h of treatment, respectively, as exhibited in the time-kill curve. The greatest reduction in biofilm biomass and metabolic activity was 1,719 OD550 and 0,089 OD550 respectively, both at 4 x MIC of carvacrol. In carvacrol treated biofilms of S. Typhimurium on polypropylene, a reduction of 5.12 log was observed with 4 x MIC, while on stainless steel, carvacrol at 4 x MIC reduced bacterial counts by 5 log. The results showed that carvacrol exhibits antibacterial activity and can be used as an alternative for the control of S. Typhimurium biofilms.

RESUMO: O amendoim é um alimento nutritivo e muito consumido no mundo todo. Algumas espécies de fungos do gênero Aspergillus, produtores de aflatoxinas, colonizam o amendoim. A exposição humana as aflatoxinas ocorre por meio do consumo de alimentos contaminados. O objetivo deste estudo foi avaliar a ocorrência e exposição da população as aflatoxinas em amendoim comercializado no município de Maringá, Brasil, durante o período de maio de 2013 a abril de 2017. Foram avaliadas 104 amostras de amendoim por análise cromatográfica e confirmadas por derivação com ácido trifluoroacético. A frequência de contaminação foi de 24,0% em concentração média de 13,4µg/kg. Vinte amostras positivas (19,2%) apresentaram-se acima do limite máximo permitido no Brasil para a somatória das quatro aflatoxinas. A ingestão diária provável média encontrada foi de 1,28µg/kg peso corpóreo/dia, superando a ingestão diária máxima tolerável provisória (0,001µg/kg peso corpóreo/dia). De acordo com os elevados teores de aflatoxinas encontrados em amendoim, há necessidade de um maior monitoramento da presença de aflatoxinas em alimentos, tendo em vista a redução nos níveis de contaminação do amendoim in natura.

ABSTRACT: Peanuts are a nutritious food consumed worldwide. Some species of the genus Aspergillus, producers of aflatoxins, colonize peanuts. Human exposure to aflatoxins occurs by ingesting contaminated foods. The objective of this paper was to assess the occurrence and dietary exposure of the population to aflatoxins in peanuts commercially available in Maringa, Brazil, from May 2013 to April 2017. A total of 104 peanut samples were evaluated by thin-layer chromatography and confirmed by derivatization with trifluoroacetic acid. The contamination rate detected was 24.0% in average concentration of 13.4µg/kg. Twenty positive samples (19.2%) exceeded the maximum level permitted in Brazil for the sum of four aflatoxins. Estimated probable daily intake was 1,28 µg/kg body weight/day, exceeding the Provisional Maximum Tolerable Daily Intake (0.001μg/kg body weight/day). According to the high levels of aflatoxins found in peanuts, there is need for further monitoring the presence of aflatoxins in peanuts in natura to reduce the levels of contamination.

Abstract Aflatoxins are mutagenic, carcinogenic, and teratogenic mycotoxins. The objective of this work was to study the presence of aflatoxigenic Aspergillus in commercial Bulgur wheat in the city of Maringá, Paraná, Brazil. Thirty samples of commercial Bulgur wheat, acquired in the period of August 2011 to January 2012, were evaluated. The enumeration analysis showed that samples had up to 273.3 CFU of molds and 133.3 CFU of aflatoxigenic Aspergillus per gram of wheat. Forty-two monosporic isolates were obtained and identified as Aspergillus flavus. The isolates were analyzed regarding their aflatoxigenic potential by culture in coconut milk agar; hydroxide vapor exposure; chromatography; and polymerase chain reaction (PCR) targeting genes that code enzymes of the aflatoxins synthesis pathway. Some of the isolates were confirmed to be aflatoxin producers and several of them presented a genetic profile of aflatoxin synthesis. The obtained results demonstrated that Bulgur wheat A. flavus contamination is concerning.

Realizou-se um estudo transversal controlado, com hortifruticultores, durante o período de outubro de 2009 a outubro de 2010, a fim de caracterizar o uso de praguicidas, estabelecer o perfil sócio demográfico e analisar a atividade das colinesterases em trabalhadores rurais do sul do Brasil. Os dados foram obtidos de 173 trabalhadores e 179 controles. Um formulário estruturado foi aplicado obtendo informações sócio demográficas, conhecimento e práticas de trabalho relacionadas com o uso de praguicidas. Os valores de referência para colinesterases totais (ChEs) e butirilcolinesterase (BuChE) foram obtidos a partir da média da atividade enzimática do grupo controle. A idade média da população exposta foi de 40,0±11,2 anos. Os sinais/sintomas foram significativamente diferentes (p<0,05) entre a população exposta e não exposta. Quarenta trabalhadores (23,1%) apresentaram sinais/sintomas relacionados com a exposição aos praguicidas. O valor médio do grupo controle foi de 6,3 μmol/mL/min para o gênero masculino e 5,6 μmol/mL/min para o gênero feminino na ChEs; 2,4 μmol/mL/min para o gênero masculino e 2,0 μmol/mL/min para o gênero feminino na BuChE. Do total de trabalhadores (n=173), 08 (4,6%) apresentaram inibição elevada (>30%) das atividades das ChEs, e para BuChE nenhum trabalhador apresentou alta inibição (>50%). Dentre os fatores que poderiam estar envolvidos destacam-se o gênero, escolaridade, orientação para trabalhar com praguicidas, exposição e medidas de higiene.

A controlled cross-sectional study of family growers of fruit and vegetables was conducted between October 2009 and October 2010 to characterize the use of pesticides, establish the socio-demographic profile, and analyze cholinesterase activity in small-scale agricultural workers in Southern Brazil. Data was collected for 173 workers and 179 controls. A structured questionnaire was applied collecting socio-demographic information and determining knowledge and work practices in relation to pesticide use. The benchmarks for total cholinesterase (ChEs) and butyrylcholinesterase (BuChE) were obtained from the average enzymatic activity of the occupationally unexposed group (control). The mean age of the exposed population was 40.0 ± 11.2 years. The symptoms differed significantly (p<0.05) between the exposed and unexposed populations. Forty (23.1%) workers showed symptoms associated with exposure to pesticides. The average value of enzymatic activity in the occupationally unexposed group for ChEs was 6.3 μmol/mL/min among males and 5.6 μmol/mL/min among females while for BuChE was 2.4 μmol/mL/min among males and 2.0 μmol/mL/min among females. Eight (4.6%) workers had high inhibition (>30%) of ChEs activity, whereas no workers showed high inhibition (>50%) of BuChE. Potential factors involved include gender, education, pesticide orientation, exposure, and hygiene measures.

Aflatoxinas são metabólitos carcinogênicos produzidos pelo Aspergillus flavus, A. parasiticus e A. nomius. No presente estudo, amostras de amendoim foram coletadas em diferentes estágios fenológicos da planta durante as safras de 2007/2008 e 2008/2009, bem como no período de armazenamento do amendoim referente à safra de 2007/2008. A micoflora e os níveis de aflatoxinas foram determinados. Os resultados demonstraram a presença de Fusarium spp., Macrophomina spp., Trichoderma spp., Aspergillus spp. e Cladosporium spp. no período de maturação (39,8; 17,9; 8,2; 2,7 e 1,7%, respectivamente) e no armazenamento (49,8; 27,8; 12,5; 8,8 e 1,0%). As aflatoxinas não foram detectadas nas amostras coletadas. Dos 25 isolados de Aspergillus spp., 24 foram produtores de aflatoxina B1 (96%), 10 de B2 (40%), 17 de G1 (68%) e 10 de G2 (40%). O isolamento de Aspergillus spp. durante o armazenamento não foi influenciado pela temperatura, umidade relativa e atividade de água (p > 0,05). A detecção de Aspergillus spp. produtores de aflatoxinas nos diferentes estágios fenológicos: ginóforo aéreo, granação (sementes), frutos maduros (vagem) e frutos secos (colheita) demonstra a importância de boas práticas agrícolas da produção ao armazenamento do amendoim no sul do Brasil.

Aflatoxins are carcinogens produced by Aspergillus flavus, A. parasiticus and A. nomius. In the present study, peanut samples were collected at different phenological stages of the plant during the 2007/2008 and 2008/2009 seasons and from stored peanuts harvested in 2007/2008. The mycoflora and aflatoxins in the peanuts were evaluated. The results showed the presence of Fusarium spp., Macrophomina spp., Trichoderma spp., Aspergillus spp. and Cladosporium spp. during the period of peanut maturation (39.8, 17.9, 8.2, 2.7 and 1.7%, respectively) and storage (49.8, 27.8, 12.5, 8.8 and 1.0%, respectively). However, aflatoxins were not detected in the samples. Of the 25 Aspergillus spp. isolates, 24 (96%) were producers of aflatoxin B1 (96%), 10 (40%) of aflatoxin B2, 17 (68%) of aflatoxin G1, and 10 (40%) of aflatoxin G2. The isolation of Aspergillus spp. during storage was not influenced by the temperature, relative humidity or water activity (p > 0.05). The detection of aflatoxin-producing Aspergillus spp. in the samples analysed at different phenological stages, aerial gynophore, pod filling (seeds), mature fruits (pod), and dry fruits (harvest), indicates the importance of good agricultural practices from the cultivation to storage of peanuts in southern Brazil.

The productivity of wheat and corn crops depends on climatic conditions and resistance against phytopathogenic fungi such as those of the genus Fusarium. Some species of this genus produce zearalenone (ZEA), a mycotoxin with hyperestrogenic effects. The objective of this study was to investigate the presence of ZEA in samples of cracked wheat (n = 109), popcorn (n = 51) and corn grits (n = 50) commercialized in the State of Paraná, Brazil. Commercial samples of each crop were collected between September 2007 and June 2008 and analyzed by thin-layer chromatography. The method used for detection of the mycotoxin in wheat and corn derivatives presented a recovery rate of 94.5% and 99.5%, respectively, detection limit of 40 μg.kg-1 and quantification limit of 55 μg.kg-1. No contamination with ZEA was detected in cracked wheat samples. Among the corn derivatives, only one cracked corn sample was contaminated with ZEA (64 μg.kg-1). Despite the low contamination observed, monitoring the occurrence of mycotoxins in foods is important to ensure safety.

A exposição às aflatoxinas (AFs) na dieta é um fator de risco para o desenvolvimento do carcinoma hepatocelular (CHC) e a exacerbação da hepatite aguda em indivíduos portadores do vírus da hepatite B (VHB). O uso de biomarcadores, como a aflatoxina M1 (AFM1) na urina, produto de biotransformação da aflatoxina B1 (AFB1), permite avaliar se a população está exposta às AFs. O objetivo do presente estudo foi investigar ocorrência de AFM1 na urina de portadores e não portadores crônicos do VHB. Foi selecionado um grupo, de forma aleatória, representado por 43 portadores do VHB atendidos em dois hospitais da cidade de Maringá, Brasil, no período de Março a Junho/2008. O grupo controle foi composto por 29 voluntários adultos saudáveis anti-HBs positivo e HBsAg negativo. A determinação de AFM1 foi realizada por meio de detecção por fluorescência em sistema de cromatografia a líquido de alta eficiência com derivação pós-coluna utilizando Kobra Cell®. Das 72 amostras analisadas, 05/29 (17,2%) foram positivas para AFM1 em indivíduos não portadores do VHB, e 16/43 (37,2%) de pacientes portadores do VHB. Este estudo demonstrou a ocorrência de AFM1 na urina dos dois grupos estudados. Entretanto, há evidências de que os portadores do VHB possuam alto risco no desenvolvimento do CHC devido ao efeito aditivo pela interação entre aflatoxinas e VHB.

Exposure to aflatoxins (AFs) in the diet may favour the development of hepatocellular carcinoma (HCC) and the acute exacerbation of hepatitis in chronic hepatitis B virus (HBV) carriers. Measurement of biomarkers such as aflatoxin M1 (AFM1), a metabolite of aflatoxin B1 (AFB1), in urine allows for the assessment of populations exposed to aflatoxins. The aim of this study was to investigate the occurrence of aflatoxin M1 in the urine of HBV carrier and non-carrier patients. One group included 43 randomly selected HBV carriers treated at two hospitals in the city of Maringa, Brazil, from March to June 2008. Control group consisted of 29 healthy adult volunteers with anti-HBs positive and HBsAg negative test results. Detection of AFM1 was performed by fluorescence using high performance liquid chromatography (HPLC) and post-column derivation with the Kobra Cell®. Of the 72 samples analysed, 05/29 (17.2%) AFM1 positive samples were from HBV non-carriers, and 16/43 (37.2%) of samples were from chronic HBV carriers. This study showed AFM1 in the urine of the two surveyed population. However, there is evidence that the chronic HBV carriers have a higher risk of developing HCC due to additive interaction between AFs and HBV.

A detecção de fungos micotoxigênicos em alimentos é importante porque sua presença pode indicar uma possível contaminação com as micotoxinas associadas. Fusarium graminearum é um patógeno de trigo e um produtor de micotoxinas. A reação da polimerase em cadeia (PCR) é empregada na identificação específica de F. graminearum. No entanto, essa metodologia não tem sido comumente empregada na detecção de F. graminearum em alimentos. Assim, o presente trabalho teve como objetivo desenvolver uma metodologia molecular para detectar F. graminearum em amostras comerciais de trigo para quibe. Dois métodos foram testados. No primeiro, uma amostra desse cereal foi contaminada com micélio de F. graminearum. O DNA genômico foi extraído dessa mistura e utilizado em uma reação de PCR específica para F. graminearum. A espécie F. graminearum foi detectada somente em amostras altamente contaminadas. No segundo método, amostras de trigo para quibe foram inoculadas em um meio de cultura sólido e isolados com características culturais de F. graminearum foram obtidos. O DNA extraído desses isolados foi utilizado em reações de PCR específicas para F. graminearum. Um isolado obtido teve o genótipo de produção de tricotecenos determinado por PCR. A metodologia estabelecida poderia ser utilizada em levantamentos de contaminação de alimentos com F. graminearum.

The detection of mycotoxigenic fungi in foodstuff is important because their presence may indicate the possible associated mycotoxin contamination. Fusarium graminearum is a wheat pathogen and a producer of micotoxins. The polymerase chain reaction (PCR) has been employed for the specific identification of F. graminearum. However, this methodology has not been commonly used for detection of F. graminearum in food. Thus, the objective of the present study was to develop a molecular methodology to detect F. graminearum in commercial samples of bulgur wheat. Two methods were tested. In the first method, a sample of this cereal was contaminated with F. graminearum mycelia. The genomic DNA was extracted from this mixture and used in a F. graminearum specific PCR reaction. The F. graminearum species was detected only in samples that were heavily contaminated. In the second method, samples of bulgur wheat were inoculated on a solid medium, and isolates having F. graminearum culture characteristics were obtained. The DNA extracted from these isolates was tested in F. graminearum specific PCR reactions. An isolate obtained had its trichothecene genotype identified by PCR. The established methodology could be used in surveys of food contamination with F. graminearum.

INTRODUÇÃO: A obesidade na adolescência é um fator preditivo da obesidade no adulto. A prevalência em crianças e adolescentes tem aumentado na maior parte dos países e tem-se traduzido em um dos mais significativos problemas nutricionais da atualidade. OBJETIVOS: Este estudo teve por objetivo verificar os perfis antropométrico, lipídico e glicêmico em adolescentes de baixo nível socioeconômico de uma instituição filantrópica no noroeste do Paraná. MATERIAL E MÉTODOS: A população estudada foi composta por 92 adolescentes, com idade entre 10 e 17 anos. Foi utilizado o índice de massa corporal (IMC) para verificar o estado nutricional dos adolescentes, considerando normal 24,9 kg/m² e sobrepeso > 25 kg/m². A razão cintura-quadril (RCQ) foi mensurada por meio do quociente da cintura pelo quadril, e as concentrações de glicose, triglicerídeos, colesterol total, frações HDL-C, LDL-C e VLDL-C foram determinadas segundo o método enzimático colorimétrico. RESULTADOS: Encontraram-se 9,79% de sobrepeso nos adolescentes e 9,78% de RCQ e colesterol inadequados para a idade. Baixa correlação foi encontrada entre IMC e sobrepeso, glicemia, hipercolesterolemia, hipertrigliceridemia, LDL-C, VLDL-C e HDL-C, enquanto idade (razão de chance [RC] = 49,16) e RCQ (RC = 13,99) apresentaram correlação significativa (p < 0,05) com IMC. CONCLUSÃO: O resultado demonstra a necessidade de políticas públicas de saúde no âmbito escolar como fator preventivo da obesidade e seus fatores de risco.

INTRODUCTION: Obesity in adolescence is a predictive factor of obesity in adulthood. The prevalence in children and adolescents has increased in most countries and has become one of the most significant nutritional problems nowadays. OBJECTIVES: This study aimed at verifying the anthropometric, lipid and glycemic profiles in lower class adolescents from a philanthropic institution located in the Northeast of Paraná State. MATERIAL AND METHOD: The studied population consisted of 92 adolescents, from 10 to 17 years of age. The Body Mass Index (BMI) was used to verify their nutritional status, considering < 24.9 kg/m² normal and > 25 kg/m² overweight. The waist-to-hip measurement was calculated through waist-to-hip ratio (WHR) and glucose concentrations, triglycerides, total cholesterol, HDL-c, LDL-c and VLDL-c fractions were determined through enzymatic methods. RESULTS: 9.79% were overweight and 9.78% of WHR and cholesterol levels were inadequate to the age group. BMI (overweight), blood glucose, hypercholesterolemia, hypertriglyceridemia, LDL-c, VLDL-c and HDL-c showed low correlation, whereas age (OR = 49.16) and WHR (OR = 13.99) showed significant correlation p < 0.05. CONCLUSION: The result evidenced the necessity of health public policies in educational scope in order to prevent obesity and its risks factors in adolescence.

O objetivo deste trabalho foi avaliar a incidência de fungos dos gêneros Aspergillus, Fusarium e Penicillium e as contaminações com micotoxinas em grãos de cinco híbridos comerciais de milho em função da umidade de colheita. O trabalho foi conduzido em Astorga, Estado do Paraná, durante o período da ‘safrinha’ de 2007 e da safra de ‘verão’ de 2007/2008. As amostras de grãos, colhidas em cinco épocas distintas e em cinco repetições, foram submetidas à determinação de umidade pelo método da estufa a 103 ± 1ºC por 72h. O teste de sanidade Blotter test e a quantificação micotoxinas (método da cromatografia em camada delgada) foram efetuados, utilizando-se um delineamento inteiramente casualizado. Os resultados obtidos foram submetidos à análise de variância e de regressão. O gênero Fusarium apresentou crescimento linear da incidência, em função do aumento da umidade, enquanto os gêneros Aspergillus e Penicillium tiveram suas incidências reduzidas. Observou-se que é possível a produção de aflatoxinas, associadas aos grãos de milho, ainda no campo, e que a antecipação da colheita, seguida de secagem imediata, assegura a qualidade sanitária dos grãos de milho.

The objective of this work was to evaluate the incidence of Aspergillus, Fusarium and Penicillium fungi, and the contaminations with mycotoxins in grains of five corn commercial hybrids due to harvest humidity. The work was conducted in Astorga, Paraná State, during the autumn/fall harvest of 2007 and the summer harvest of 2007/2008. The corn grain samples were collected in five distinguished periods and also in five replications, subjected to humidity content determination at an oven heated to 103 ± 1ºC for 72h. The blotter test and the mycotoxins presence were evaluated by thin layer chromatography, using a completely randomized design. The data were submitted to analyses of variance and regression. The genera Fusarium presented linear increasing incidence due to humidity increasing, whereas Aspergillus spp. and Penicillium spp. had their incidences decreased. It was observed that the possible production of aflatoxins, associated to corn grains, still in the field, and, that the anticipation of the crop harvesting, followed by immediate drying assures the sanitary quality of the corn grains.

Este trabalho teve por objetivos verificar as características físicas, mecânicas, contaminação por Fusarium sp e por zearalenona e suas relações, em vinte variedades crioulas de milho da região sul do Brasil. Verificou-se que das amostras analisadas, 60% foram consideradas como possuidoras de endosperma do tipo duro e 40% com endosperma do tipo intermediário. As variáveis físicas e mecânicas se correlacionaram, indicando que quanto maior a proporção de endosperma vítreo, mais denso e maior a força necessária até a ruptura do grão. A contaminação por Fusarium sp esteve entre 5,5% e 24,75% nos grãos analisados, correlacionando-se positivamente com o índice de flotação, indicando que as variedades crioulas de milho com uma maior proporção de endosperma macio podem apresentar uma maior contaminação por Fusarium sp, sugerindo-se a utilização de variedades crioulas com a textura do endosperma predominantemente vítrea. A presença de zearalenona foi verificada em 75% das amostras, em concentrações que variaram de 50 a 640 µg kg-1.

This work had as objectives the study of the physical and mechanical characteristics of maize kernels in relation to the contamination by Fusarium sp and by zearalenone in twenty landraces of maize from the southern region of Brazil. From the analyzed samples, 60 % has been considered to have a hard endosperm type and 40 % an intermediary one. A correlation between the physical and mechanical variables was observed as an indication that the higher is the proportion of hard endosperm, more dense will be the kernel and more force for its rupture will be necessary. The level of contamination by Fusarium sp was between 5.5 and 24.75% among the analyzed grains, correlating positively with the flotation index, indicating that the landraces of maize with a softer endosperm can present a higher contamination by this genus. The presence of zearalenone was verified in 75 % of the samples, in concentrations varying from 50 to 640 µg kg-1.
