RESUMO Doenças infecciosas são as maiores responsáveis por falhas reprodutivas (FR) em cadelas, causando aborto, morte fetal e natimortalidade. Este estudo teve como objetivo investigar a associação entre agentes infecciosos, FR inexplicáveis e anemia em cadelas. Todas as amostras maternas e fetais foram negativas para a presença dos principais agentes infecciosos causadores de FR: herpes vírus canino 1, Neospora caninum, Brucella spp. e B. canis, enquanto agentes como o de Leishmania spp., parvovírus canino, Ehrlichia canis e Anaplasma platys foram encontrados em sangue materno. Coinfecções de A. platys/E. canis e A. platys/Leishmania spp. foram diagnosticadas. Os resultados indicam que os animais com anemia causadas por doenças transmitidas por vetores podem ser mais suscetíveis a sofrerem FR do que animais com valores hematológicos normais.

Mycoplasma hyopneumoniae is the etiologic agent of enzootic pneumonia in pigs and causes large economic losses in the swine industry. There is little data on the positivity of this disease in Brazil. The objective of this study was to evaluate the seropositivity for this agent in 200 serum samples collected from pigs in a slaughterhouse located in the central region of São Paulo. A high percentage (52%) of positivity was found indicating the presence of the agent and the need to implement control measures.

Avaliou-se a infecção por Brucella abortus em cavalos de carroça de Curitiba e São José dos Pinhais-PR. Um total de 123 amostras foi submetido ao teste do antígeno tamponado acidificado (ATA), soroaglutinação lenta em tubos (SAL) e prova do 2-mercaptoetanol (2-ME) para confirmação dos resultados. Oito (6,5%) equinos foram positivos para o ATA e um animal permaneceu positivo ao teste confirmatório. Existem evidências da presença de brucelose entre os cavalos de carroça.

The aim of the present study was to evaluate a species-specific nested PCR based on a previously described species-specific PCR for detection of B. ovis in semen and urine samples of experimentally infected rams. The performance of the species-specific nested PCR was compared with the results of a genus-specific PCR. Fourteen rams were experimentally infected with the Brucella ovis REO 198 strain and samples of semen and urine were collected every week up to 180 days post infection. Out of 83 semen samples collected, 42 (50.6%) were positive for the species-specific nested PCR, and 23 (27.7%) were positive for the genus-specific PCR. Out of 75 urine samples, 49 (65.3%) were positive for the species-specific nested PCR, whereas 11 (14.6%) were genus-specific PCR positive. Species-specific nested PCR was significantly more sensitive (P<0.001) than the genus-specific PCR in semen and urine from experimentally infected rams. In conclusion, the species-specific nested PCR developed in this study may be used as a diagnostic tool for the detection of B. ovis in semen and urine samples from suspected rams.

O presente estudo objetivou avaliar uma técnica de nested PCR espécie-específica delineada a partir de PCR espécie-específica descrita anteriormente para detecção de B. ovis em sêmen e urina de carneiros infectados experimentalmente. O desempenho da nested PCR espécie-específica foi comparado com os resultados de uma PCR gênero-específica. Quatorze carneiros foram infectados experimentalmente com Brucella ovis REO 198 e amostras de sêmen e de urina foram colhidas semanalmente até 180 dias após a infecção. De 83 amostras de sêmen, 42 (50,6%) foram positivas pela nested PCR espécie-específica, e 23 (27,7%) foram positivas pela PCR gênero-específica. De 75 amostras de urina, 49 (65,3%) foram positivas pela nested PCR espécie-específica, enquanto 11 (14,6%) foram positivas em PCR gênero-específica. A técnica de nested PCR espécie-específica foi significativamente mais sensível (P<0,001) do que a PCR gênero-específica no sêmen e na urina de carneiros infectados experimentalmente. Em conclusão, a nested PCR espécie-específica desenvolvida neste estudo pode ser utilizada como ferramenta de diagnóstico para detecção de B. ovis em sêmen e urina de carneiros suspeitos.

Infectious diseases in wild animals have been increasing as a result of their habitat alterations and closer contact with domestic animals. Canine distemper virus (CDV) has been reported in several species of wild carnivores, presenting a threat to wildlife conservation. We described the first case of canine distemper virus infection in lesser grison (Galictis cuja). A free-ranging individual, with no visible clinical sigs, presented sudden death after one day in captivity. Molecular diagnosis for CDV infection was performed using whole blood collected by postmortem intracardiac puncture, which resulted positive. The virus phylogeny indicated that domestic dogs were the probable source of infection.

Doenças infecciosas em animais selvagens têm aumentado devido às alterações em seu habitat e ao maior contato com animais domésticos. A cinomose já foi descrita em diversas espécies de carnívoros selvagens, representando uma ameaça à conservação da vida selvagem. Nesse estudo é descrito o primeiro caso de infecção pelo vírus da cinomose em um furão (Galictis cuja). Um indivíduo de vida livre, sem sinais clínicos aparentes, apresentou morte súbita após um dia em cativeiro. Foi realizado o diagnóstico molecular para detecção do vírus da cinomose canina, sendo o resultado positivo. A filogenia do vírus indicou que cães domésticos foram a provável fonte de infecção.

A retrospective study of major clinical and epidemiological findings was performed in 11 cases of tetanus observed in eight sheep and three goats from 1998 to 2008. The cases occurred predominantly in animals aging from five days to two years, in winter season, with history of recent wounds or surgical procedures. Muscle rigidity and muscular tremors (72.0%), tachycardia/dyspnea (54.5%), change of placement of ears (45.4%), protusion of the third eyelid (27.2%), hypersensitivity to external stimulus (18.1%), and elevation of ears (18.1%) were the most common clinical signs.

O presente estudo relata a ocorrência de co-infecção entre o vírus da cinomose canina (CDV) e Toxoplama gondii em cães com sinais neurológicos. Amostras de soro e tecido nervoso (pos-mortem) de 21 cães, suspeitos de cinomose canina foram analisadas pela Reação de Imunofluorecência indireta (RIFI) para pesquisa de anticorpos contra T. gondii e N. caninum e por RT-PCR para CDV. Dezessete (80,9%) cães foram positivos para o CDV pela RT-PCR e 8 (38,1%) foram positivos para anticorpos contra T. gondii. Sete cães (41,1%) apresentaram-se positivos para ambos agentes, caracterizando processo de co-infecção. Somente 1 (4,7%) cão foi soropositivo para N. caninum (RIFI=100), entretanto este mesmo animal foi positivo para T. gondii (RIFI=4096) e para CDV (RT-PCR).

O objetivo do estudo foi adaptar e avaliar a PCR para detecção de Brucella ovis e comparar os resultados com aqueles obtidos por cultivo microbiológico do sêmen, urina e dos órgãos de carneiros infectados experimentalmente. Dos 31 animais infectados experimentalmente, amostras de PCR do sêmen apresentaram maior sensibilidade (21,6%) do que o cultivo (8,0%). Em amostras de urina, a sensibilidade das técnicas foi semelhante (10,1% para a cultivo e 12,7% para PCR). PCR detectou a presença do agente em 21,5% das amostras testadas, enquanto os órgãos de cultivo detectaram em apenas 3,3% das amostras. PCR detectou um maior número de amostras positivas do que o cultivo microbiológico.

Condensation of 3-formylchromone (1) with thiazolidine-2,4-dione (2) afforded 5-[4-oxo-4H-chromen-3-yl)methylene]-1,3-thiazolidine-2,4-dione (3). Reaction of 3 with hydrazine hydrate, phenyl hydrazine and hydroxylamine hydrochloride gave the corresponding pyrazole and isoxazole derivatives 4-7. Compound 3 was subjected to react with thiourea, guanidine and cyanoguanidine to give the corresponding pyrimidine derivatives 8-10. Pyrimido[1,2-a]pyrimidine 12, benzo[1,5]diazepine 15, pyrido[1,2-b][1,2,4]triazepine 16, 1,2,4-triazolo[3,4-b][1,3,4]thiadiazepine 19 and 1,2,4-triazino[3,2-b][1,3,4]thiadiazepine 20 linked thiazolidine-2,4-dione were prepared from the reaction of 3 with N,N- and N,S- bifunctional nucleophiles. The chemical reactivity of 3 towards carbon nucleophiles gave new heterocyclic moieties linked thiazolidine-2,4-dione 22‑25. The synthesized compounds were screened in vitro for their antimicrobial activities against Staphylococcus aureus, Proteus vulgaris and Candida albicans.

A condensação do 3-formilcromano (1) com tiazolidina-2,4-diona (2) produziu 5-[4-oxo-4H-croman-3-ila)metileno]-1,3-tiazolidina-2,4-diona (3). A reação de 3 com hidrato de hidrazina, fenil hidrazina e hidrocloreto de hidroxilamina produziu os derivados correspondentes pirazol e isoxazol 4-7. O composto 3 reagiu com tiouréia, guanidina e cianoguanidina para produzir os derivados correspondentes de pirimidina 8-10. Pirimido[1,2-a]pirimidina 12, benzo[1,5]diazepina 15, pirido[1,2-b][1,2,3]triazepina 16, 1,2,4-triazolo[3,4-b][1,3,4]tiadiazepina 19 e 1,2,4-triazino[3,2-b][1,3,4]tiadiazepina 20 ligados à tiazolidina-2,4-diona foram preparados a partir da reação de 3 com nucleófilos bifuncionais N,N- e N,S-. A reatividade química de 3 frente aos nucleófilos carbonados produziu novos derivados heterocíclicos de tiazolidina-2,4-diona 22-25. Os compostos sintetizados foram testados in vitro para verificar sua atividade antimicrobiana contra Staphylococcus aureus, Proteus vulgaris e Candida albicans

Current knowledge on bat lyssavirus infections in their native hosts is limited and little is known about the virulence, virus dissemination and transmission among free-living insectivorous bats. The present study is a brief description of rabies virus (RABV) dissemination in tissues of a naturally infected pregnant southern yellow bat (Lasiurus ega) and its fetuses, obtained by reverse-transcriptase polymerase chain reaction (RT-PCR). The RT-PCR was positive in samples from the brain, salivary gland, tongue, lungs, heart, kidneys and liver. On the other hand, the placenta, three fetuses, spleen, intestine and brown fat tissue tested negative. This research demonstrated the absence of rabies virus in the fetuses, thus, in this specific case, the transplacentary transmission was not observed.

The aim of the current study was to evaluate the infection rate by Brucella spp. in wild and in captive animals. Serum samples from 121 animals (94 free-ranging and 27 captive) of different mammal species were evaluated. Sera were submitted to rose Bengal test (RBT) for screening and serum agglutination tests (SAT) and 2-mercaptoethanol test (2-ME) for confirmatory results. Nine animals (five free-ranging and four captive) tested positive in RBT, but negative in the confirmatory tests. Several domestic animal diseases that have control programs are not focused on wild reservoirs, such as brucellosis in Brazil. The study of new reservoirs in wildlife is essential to prevent emerging diseases.

This study investigated the etiology of canine ehrlichiosis and possible clinical and epidemiological data associated with the infection in 70 dogs suspect of ehrlichiosis attended at the Veterinary Hospital of the São Paulo State University in Botucatu city during 2001 and 2002. Dogs were evaluated by clinical-epidemiological and hematological data and molecular analysis by partial amplification and DNA sequencing of the ehrlichial dsb gene. E. canis DNA was amplified and sequenced in 28 (40.0%) dogs. Dogs younger than 12 months old showed significantly higher infection rates (65.0%; P < 0.05). Diarrhea, apathy, and anorexia were the major clinical signs observed in 55.2% (P = 0.05), 47.0% (P > 0.05), and 42.4% (P > 0.05) of the PCR-positive dogs, respectively. Twenty-five anemic (<5.5 × 10(6) RBC.µL-1), and 8 leukopenic (<5.5 × 10³ WBC.µL-1) dogs were PCR-positive (P > 0.05). All 28 PCR-positive dogs showed thrombocytopenia (<175 × 10³ platelets.µL-1) and revealed statistical significance (P < 0.05). E. canis was the only Ehrlichia species found in dogs in the studied region, with higher infection rates in younger dogs, and statisticallyassociated with thrombocytopenia.

O presente estudo investigou a etiologia da erliquiose monocítica canina em 70 cães atendidos no Hospital Veterinário da Universidade Estadual Paulista, na cidade de Botucatu, durante 2001 e 2002. Os cães foram avaliados segundo achados clínicos, epidemiológicos e laboratoriais e pela amplificação parcial e sequenciamento do gene dsb de Ehrlichia. DNA de Ehrlichia canis foi amplificado e sequenciado em 28 (40,0%) cães. Observou-se maior frequência deanimais positivos com idade até 12 meses (P < 0,05). Diarreia, apatia e anorexia foram os sinais clínicos mais frequentes observados em 55,2% (P = 0,05), 47,0% (P > 0,05) e 42,2% (P > 0,05) dos cães PCR positivos, respectivamente. Vinte e cinco cães com anemia (<5,5 × 10(6) eritrócitos.µL-1) e 8 com leucopenia (<5,5 × 10³ leucócitos.µL-1) foram positivos na PCR, porém não apresentaram associação estatística (P > 0,05) frente à infecção por E. canis. Todos os 28 cães positivos na PCR apresentaram trombocitopenia (<175 × 10³ plaquetas.µL-1; P < 0,05). E. canis foi a única espécie de Ehrlichia detectada em cães da região estudada, com alta taxa de infecção em cães jovens e, estatisticamente, associadaa cães trombocitopênicos.

Neosporosis is a parasitic disease caused by a protozoon Neospora caninum, which is recognized as an important cause of bovine abortion and canine neuropathy. Considering the isolation of N. caninum and the frequent serology in bovines of our region, the objectives of the present trial were to evaluate the occurrence of antibodies anti -N. caninum in dogs of the Botucatu range micro region, in the State of São Paulo, and its association with gender, age and origin (urban area - exclusively from the city; rural area - only farms and cottages; and periurban area - access to the urban and rural areas) of the dogs studied. A total of 963 dogs of defined breeds or non-defined breeds, of both genders and different ages, showing no clinical symptoms, were analyzed. Animals were randomly selected during the anti-rabies vaccination campaign in the region, from May to September 1998. Serum samples obtained from the animals were evaluated by means of the Indirect Fluorescent Antibody Test (IFAT) using as antigen N. caninum, standard strain NC-1. A total of 245 animals were reagent (25.4% positive results), with 161 (27.5%) males and 84 (22.3%) females. According to the origin - urban, rural and periurban areas - 223 (25.8%), 11(16.9%) and 11(33.3%) dogs were, respectively, reagent to the IFAT. All of the 11 cities in this region presented seropositive dogs with occurrence rates ranging from 53.5 to 8.9%. The lower positive percentage was observed in dogs below 1 year old (16.2%) when compared with animals from 1 to 4 years old and more than 4 years old (28.4 % and 28.0% respectively), which did not present any differences between them. Results obtained characterized the seropositivity for N. caninum in dogs from all Municipalities in the Botucatu micro region showing the wide distribution of the agent in the region.

Neosporose é uma enfermidade parasitária causada pelo protozoário Neospora caninum reconhecido como importante causa de abortamento bovino e neuropatia canina. Considerando o isolamento de N. caninum e a sorologia freqüente em bovinos em nossa região, os objetivos do presente trabalho foram avaliar a ocorrência de anticorpos anti-N. caninum em cães da Microrregião da Serra de Botucatu, Estado de SãoPaulo, e sua associação ao sexo, idade e procedência quanto à zona urbana (exclusivamente cidade), rural (somente chácaras e sítios) e peri-urbana (acesso à zona urbana e rural) dos cães estudados. Foram analisados 963 cães, com ou sem raça definida, de ambos os sexos e diferentes idades, sem apresentação de qualquer sintomatologia clínica. Os animais foram selecionados aleatoriamente durante a campanha de vacinação anti-rábica da microrregião da Serra de Botucatu, no período de maio a setembro de 1998. O soro obtido dos animais foi avaliado por meio da Reação de Imunofluorescência Indireta (RIFI) utilizando como antígeno a cepa padrão NC-1 deN. caninum. Observaram-se 245 animais reagentes (25,4% de positividade), sendo 161 (27,5%) machos e, 84 (22,3%) fêmeas. Dos animais de zona urbana, rural e mista 223 (25,8%),11(16,9%) e 11(33,3%), respectivamente, foram reagentes à prova de RIFI. Todos os 11 municípios apresentaram cães soropositivos com valores de ocorrência que variaram de 8,9% a 53,5%. Observou-se percentual de positividade menor em cães até um ano (16,2%) quando comparados àqueles entre 1 a 4 anos e superior a 4 anos (28,4 % e 28,0%, respectivamente) que não apresentaram diferença entre si. Os resultados obtidos caracterizaram soropositividade para N. caninum em cães pertencentes a todos os municípios da Microrregião da Serra de Botucatu evidenciando a ampla distribuição do agente na região.

Brucella abortus infection is reported in a dog from a rural area that presented at clinical evaluation left testicular enlargement and right testicular decrease. Serum resulted negative to rapid agglutination test and agar gel immunodifusion with Brucella ovis antigen but positive to buffered plate agglutination test, tube agglutination test and 2- Mercapthoetanol with B. abortus antigen. Brucella isolation was negative in blood, testicular material, semen and urine. Brucella DNA was detected in PCR from urine and blood.

The ring test (RT) was analyzed regarding its application for the individual and herd bovine brucellosis diagnoses. Individual samples of milk from 464 cows and 54 composite samples of milk bucket from these animals were evaluated. The results were analyzed considering the serological results obtained by the rose bengal (RBT), tube agglutination (TAT) and 2-mercaptoethanol (2-ME) tests. From the 464 individual milk samples analyzed by the RT, 123 (26.5%) presented positive results. From those, 30 were positive only to the RBT, 28 by the RBT/TAT/2-ME and 18 by the TAT. From the 123 positive samples by the RT, 95 came from the 2-ME seronegative animals, characterizing 77.2% of false-positive reactions and 4/341 negative reactions came from the seropositive animals, characterizing 1.2% of false-negative reactions in the individual RT. From the 54 samples of compositive milk analyzed by the RT, 17 were positive. From these positive samples, one was considered false positive since all the animals that composed it were negative by the 2-ME. Three of the composite milk samples that were negative to RT(3/37) were constituted by milk from positive animals, characterizing 8.1% of false-negative results by the RT. The individual RT showed high percentage of false-positive results, while the RT in samples from bucket detected 84.2% of the contaminated milk and 75% of the infected herds.

Analisaram-se 464 amostras individuais de leite e 54 amostras de leite de latões, oriundos de leite dos mesmos animais, por meio do teste do anel do leite (TAL) visando à sua aplicação no diagnóstico individual e de rebanhos da brucelose bovina. Foram também avaliadas 464 amostras de soro sangüíneo por meio de provas do antígeno tamponado acidificado (ATA), soroaglutinação lenta em tubos (SAL) e 2-mercaptoetanol (2-ME), todas para brucelose. Cento e vinte e três amostras (26,5%) testadas pelo TAL apresentaram resultados positivos. Dessas, 30 resultaram positivas ao ATA, 28 ao ATA, à SAL e ao 2-ME e 18 à SAL. Das amostras positivas ao TAL, 95 pertenciam a animais sorologicamente negativos ao 2-ME, caracterizando 77,2% (95/123) das reações falso-positivas; dos resultados negativos ao TAL, 4 pertenciam a animais sorologicamente positivos, caracterizando 1,2% (4/341) de reações falso-negativas no TAL individual. Das 54 amostras de leite de latões analisadas pelo TAL, 17 foram consideradas positivas, das quais uma foi caracterizada como falso-positivo, pois todos os animais que a compunham foram negativos ao 2-ME. De 37 latões considerados negativos ao TAL, três continham leite de animais positivos ao 2-ME, caracterizando 8,1% de falso-negativos. O TAL individual demonstrou elevado percentual de resultados falso-positivos, enquanto o TAL em amostras de leite obtidas em latões detectou 84,2% de latões contaminados e 75% de rebanhos infectados.
