ABSTRACT To explore the functions of Txnip and its mechanism in adipocyte differentiation, the preadipocytes were isolated from subcutaneous adipose tissue of three-day-old piglets and induced into adipogenic differentiation. The expression of Txnip and ChREBP was silenced, and the Txnip overexpression was achieved in the cells with transfection of the recombinant lentivirus strategies. Txnip silencing promoted the differentiation of porcine preadipocytes and PPARγ expression, and a PPARγ inhibitor reduced this facilitation. Instead, Txnip overexpression exerted a suppressive effect on the cell differentiation and PPARγ expression, and the PPARγ agonist offset this inhibition. High glucose stimulated the preadipocyte differentiation and expressions of ChREBP and Txnip. In contrast, Txnip expression was reduced by ChREBP silencing, suggesting glucose-regulated Txnip expression through the mediation of ChREBP. Moreover, the expressions of ChREBP and Glut4 induced by high glucose and glucose uptake of the cells were reduced by Txnip-overexpression, but increased by Txnip silencing, while these changes of Txnip did not alter their expressions under low glucose. Collectively, Txnip could be an inhibitor of porcine preadipocyte differentiation, which attenuated the adipogenesis through the negative feedback regulation on PPARγ. ABSTRACT Objectives: to develop and assess a serious game on healthy eating and physical activity to promote health and tackle childhood obesity. Method: a descriptive, applied and methodological study.For the development of the game, the following steps were taken: conceptualization, pre-production with the development of the game documentation, prototyping, production and assessment of thecomputer and health experts. Results: a prototype has been developed up to beta version. The game was positively assessed both in terms of gameplay and mechanics, and in relation to the content presented, standing out as a powerful strategy for health promotion. The information from the assessment phase contributed to the settings in the software in order to make it available in the future for the target population of this research. The greatest advantage of the proposed game is the fact that it is an open educational resource. Conclusions: the expert assessments showed that the game has great educational potential and it is considered suitable for future application to the target audience.The serious game can become a technological teaching resource available for use in schools and health facilities, and can also be reused for the production of other educational games by accessing its source code.

Two new end-to-end azido-bridged polymeric copper(II) complexes, [CuL¹(µ13-N3)Jn (1) and [CuL²(µ13-N3)Jn (2), where L¹ and L² are the monoanionic form of 5-methoxy-2-[(2-methylaminoethylimino)methyl]phenol and 4-chloro-2-[(3-dimethylaminopropylimino)methyl]phenol, have been prepared and structurally characterized by elemental analysis, IR spectra, and single crystal X-ray determination. Complex (1) crystallizes in the orthorhombic space group P2(1)2(1)2(1), with unit cell dimensions a = 7.6651(9) Å, b = 8.3213(9) Å, c = 20.149(2) Å, V = 1285.2(2) A³, Z = 4, R1 = 0.0296, and w׳R2 = 0.0697. Complex (2) crystallizes in the orthorhombic space group Pbca, with unit cell dimensions a = 10.353(1) Å, b = 13.069(2) Å, c = 22.265(2) Å, V = 3012.6(7) A³, Z = 8, R1 = 0.0400, and w׳R2 = 0.1014. Each Cu atom in the complexes is in a square pyramidal coordination, with three donor atoms of the Schiff base ligand and one azido N atom defining the basal plane, and with another azido N atom occupying the apical position. The Cu-Cu distances are 5.173(2) Å in (1) and 5.277(2) Å in (2). Hydrogen bonds can influence the bridging mode of the azide ligand.

The genetic diversity and genetic structure of three Chinese silkworm species Bombyx mori L., Antheraea pernyi Guérin-Meneville and Samia cynthia ricini Donovan were comparatively assessed based on RAPD markers. At the species level, A. pernyi and B. mori showed high levels of genetic diversity, whereas S. cynthia ricini showed low level of genetic diversity. However, at the strain level, A. pernyi had relatively highest genetic diversity and B. mori had lowest genetic diversity. Analysis of molecular variance (AMOVA) suggested that 60% and 72% of genetic variation resided within strains in A. pernyi and S. cynthia ricini, respectively, whereas only 16% of genetic variation occurred within strains in B. mori. In UPGMA dendrogram, individuals of A. pernyi and B. mori formed the strain-specific genetic clades, whereas those of S. cynthia ricini were distributed in a mixed way. The implications of these results for the conservation and utilization in breeding programs of three silkworm species are discussed.

Bombyx mori and Bombyx mandarina are morphologically and physiologically similar. In this study, we compared the nucleotide variations in the complete mitochondrial (mt) genomes between the domesticated silkmoth, B. mori, and its wild ancestors, Chinese B. mandarina (ChBm) and Japanese B. mandarina (JaBm). The sequence divergence and transition mutation ratio between B. mori and ChBm are significantly smaller than those observed between B. mori and JaBm. The preference of transition by DNA strands between B. mori and ChBm is consistent with that between B. mori and JaBm, however, the regional variation in nucleotide substitution rate shows a different feature. These results suggest that the ChBm mt genome is not undergoing the same evolutionary process as JaBm, providing evidence for selection on mtDNA. Moreover, investigation of the nucleotide sequence divergence in the A+T-rich region of Bombyx mt genomes also provides evidence for the assumption that the A+T-rich region might not be the fastest evolving region of the mtDNA of insects.

A cepa Actinomyces A01 foi isolada do solo de um campo agrícola no subúrbio de Beijing, China. De acordo com as características morfológicas, culturais, fisiológicas e bioquímicas, e análise da sequência 16S rDNA , a cepa A01 foi identificada como Streptomyces lydicus. Nos testes de espectro antimicrobiano, a cepa A01 apresentou atividade inibitória intensa e estável contra vários fungos patogênicos para plantas, como Fusarium oxysporum, Botrytis cinerea, Monilia laxa, etc. Entretanto, não foi encontrada atividade antibacteriana. Em experimentos em estufas, o desenvolvimento do fungo cinza do tomate foi fortemente inibido pelo tratamento com o caldo de fermentação da cepa A01, com eficiência superior à do pyremethanil e polyoxin. Por cromatografia em coluna e HPLC, obteve-se um composto fúngico (pureza 99,503%), cuja análise estrutural por UV, IR, MS e NMR revelou ser natamicina, um antibiótico polienico produzido por S. chattanovgensis, S. natalensis e S.gilvosporeus, empregado como conservador biológico natural em alimentos. O presente estudo relata a detecção de uma nova cepa produtora de natamicina e sua aplicação potencial como um agente de controle biológico de doenças fúngicas em plantas.

Actinomyces strain A01 was isolated from soil of a vegetable field in the suburb of Beijing, China. According to the morphological, cultural, physiological and biochemical characteristics, and 16S rDNA sequence analysis, strain A01 was identified as Streptomyces lydicus. In the antimicrobial spectrum test strain A01 presented a stable and strong inhibitory activity against several plant pathogenic fungi such as Fusarium oxysporum, Botrytis cinerea, Monilinia laxa, etc. However, no antibacterial activity was found. In pot experiments in greenhouse, the development of tomato gray mold was markedly suppressed by treatment with the fermentation broth of the strain A01, and the control efficacy was higher than those of Pyrimethanil and Polyoxin. A main antifungal compound (purity 99.503%) was obtained from the fermentation broth of strain A01 using column chromatography and HPLC. The chemical structural analysis with UV, IR, MS, and NMR confirmed that the compound produced by the strain A01 is natamycin, a polyene antibiotic produced by S. chattanovgensis, S. natalensis, and S. gilvosporeus, widely used as a natural biological preservative for food according to previous reports. The present study revealed a new producing strain of natamycin and its potential application as a biological control agent for fungal plant diseases.
