Resumo As armadilhas extracelulares de neutrófilos (NETs) foram relatadas pela primeira vez como uma estratégia microbicida para neutrófilos ativados. Por meio de uma resposta imunológica contra vários estímulos, os neutrófilos liberam seu DNA ligado a proteínas de grânulos, núcleo e citoplasma (por exemplo, elastase e mieloperoxidase). Até o momento, os NETs têm sido implicadas em danos aos tecidos durante processos inflamatórios intensos, principalmente quando sua liberação depende da geração de radicais de oxigênio. Os flavonóides são agentes antioxidantes e anti-inflamatórios, e destes, a quercetina é comumente encontrada em nossa dieta diária. Portanto, a quercetina pode exercer alguma atividade protetora contra o dano tecidual induzido por NETs. Em nossos ensaios in vitro, a quercetina reduziu a liberação das NETs, mieloperoxidase (MPO) e elastase a partir de neutrófilos estimulados com forbol 12-miristato 13-acetato (PMA). A atividade dessas enzimas também foi diminuída na presença de quercetina. A quercetina também reduziu o efeito citotóxico dos NETs sobre células alveolares (linha celular A549). Além disso, os ensaios in silico indicaram interações favoráveis entre a quercetina e as proteínas da NET (MPO e elastase). No geral, nossos resultados demonstram que a quercetina diminui os efeitos deletérios das NETs, reduzindo sua liberação à partir de neutrófilos ativados e diminuindo a atividade enzimática de MPO e elastase, possivelmente por meio de interação direta.

Abstract Neutrophil extracellular traps (NETs) were first reported as a microbicidal strategy for activated neutrophils. Through an immunologic response against several stimuli, neutrophils release their DNA together with proteins from granules, nucleus, and cytoplasm (e.g., elastase and myeloperoxidase). To date, NETs have been implicated in tissue damage during intense inflammatory processes, mainly when their release is dependent on oxygen radical generation. Flavonoids are antioxidant and anti-inflammatory agents; of these, quercetin is commonly found in our daily diet. Therefore, quercetin could exert some protective activity against tissue damage induced by NETs. In our in vitro assays, quercetin reduced NETs, myeloperoxidase (MPO), and elastase release from neutrophils stimulated with phorbol 12-myristate 13-acetate (PMA). The activity of these enzymes also decreased in the presence of quercetin. Quercetin also reduced the cytotoxic effect of NETs on alveolar cells (A549 cell line). Further, in silico assays indicated favorable interactions between quercetin and NET proteins (MPO and elastase). Overall, our results demonstrate that quercetin decreases deleterious cellular effects of NETs by reducing their release from activated neutrophils, and diminishing the enzymatic activity of MPO and elastase, possibly through direct interaction.

Resumo Em uma escala global o Sorghum bicolor (L.) é uma das culturas mais importantes, ocupando o quinto lugar entre todos os cereais. É cultivada na África, Ásia, Oceania e Américas, onde serve como fonte de alimento e ração para humanos e animais, respectivamente. No entanto, a produção é limitada por vários fatores, incluindo pragas e doenças. Entre as pragas que atacam o sorgo, Schizaphis graminum (Rondani), comumente chamado de pulgão verde dos cereais, é a praga mais destrutiva do sorgo. O dano de S. graminum ao sorgo é agravado pelo déficit hídrico que favorece sua ocorrência. Informações limitadas sobre a distribuição espacial da praga e seus inimigos naturais impedem o desenvolvimento de estratégias de manejo ecologicamente corretas. Portanto, o objetivo desta pesquisa foi determinar a distribuição espaço-temporal de S. graminum e seu inimigo natural Coccinella septempunctata (L.) durante a fase vegetativa e reprodutiva da cultura por meio de análise geoestatística. A distribuição espaço-temporal de S. graminum foi influenciada pelo estágio de desenvolvimento do sorgo, paisagem circundante e presença de seu principal predador C. septempunctata. Além disso, a abundância de C. septempunctata foi influenciada pela densidade de S. graminum. Os resultados deste estudo são necessários para o desenvolvimento de estratégias sustentáveis de manejo de pragas contra S. graminum.

Abstract At the global level, Sorghum bicolor (L.), is one of the most important crops, which ranks fifth among all cereals. It is cultivated in Africa, Asia, Oceania and the Americas, where it serves as a source of food and feed for humans and animals, respectively. However, production is constrained by several factors including pests and diseases. Among the pests that are known to attack Sorghum, Schizaphis graminum (Rondani), commonly called the green cereal aphid, is the most destructive pest of sorghum. S. graminum damage to sorghum is worsen by water deficit which favors its occurrence. Limited information on the spatial distribution of the pest and its natural enemies impedes the development of ecologically friendly management strategies. Therefore, the objective of this research was to determine the spatiotemporal distribution of S. graminum and its natural enemy Coccinella septempunctata (L.) during the vegetative and reproductive stage of the crop using geostatistical analysis. The spatiotemporal distribution of S. graminum was influenced by Sorghum development stage, surrounding landscape, and presence of its main predator C. septempunctata. Moreover, the abundance of C. septempunctata was influenced by the density of S. graminum. The findings from this study are required for developing sustainable pest management strategies against S. graminum.

ABSTRACT Sweet potato is among the most important food crops in the world. Due to its high genetic diversity, this species has cultivars that can respond in different ways depending on mineral fertilization. Among the nutrients, phosphorus is related to physiological and biochemical processes that can influence the final quality of the product. This study aimed to assess the influence of P doses on the quality and bioactive compounds of sweet potato cultivars - Paraná, BRS Amélia, and BRS Cuia. The pH, soluble solids (SS), titratable acidity (TA), SS/TA ratio, soluble sugars, and the bioactive compounds - flavonoids, anthocyanins, and carotenoids - were evaluated. The P doses influenced sweet potato cultivars. The dose of 120 kg ha-1 promoted increments in the parameters pH, SS, TA, and SS/TA ratio for the cultivar BRS Cuia. The same dose promoted an increase in total soluble sugars for the cultivar BRS Amélia. P dose of 240 kg ha-1 promoted higher levels of flavonoids and anthocyanins in the cultivar Paraná, and there was an increase in carotenoid content at the dose of 60 kg ha-1.

RESUMO A batata-doce está entre as culturas alimentícias mais importantes do mundo. Por apresentar alta diversidade genética, essa espécie possui cultivares que podem responder de maneiras diferentes em função da adubação mineral. Dentre os nutrientes, o fósforo está relacionado com processos fisiológicos e bioquímicos que podem influenciar na qualidade final do produto. Este estudo teve como objetivo avaliar a influência de doses de P na qualidade e compostos bioativos de cultivares de batata-doce - Paraná, BRS Amélia e BRS Cuia. Foram avaliados o pH, sólidos solúveis (SS), acidez titulável (AT), relação SS/AT, açúcares solúveis e os compostos bioativos - flavonoides, antocianinas e carotenoides. As doses de P influenciaram as cultivares de batata-doce. A dose de 120 kg ha-1 promoveu incrementos nos parâmetros pH, SS, AT e relação SS/AT para a cultivar BRS Cuia. A mesma dose promoveu incrementos nos açúcares solúveis totais para a cultivar BRS Amélia. A dose de 240 kg ha-1 promoveu maiores teores de flavonoides e antocianinas na cultivar Paraná, e houve aumento no teor de carotenoides na dose de 60 kg ha-1.

Leachate from a composting plant was characterized and applied as a biofertilizer in lettuce crops. Characteristics of untreated (UE) and anaerobically treated (TE) leachate samples were compared. The pH of TE (8.2) increased in relation to the UE (5.2) due to an increase in ammonia nitrogen in TE (1197 mg L-1) compared to UE (859 mg L-1). Anaerobic treatment was efficient in the removal of organic matter from the leachate, evaluated by the decrease of dissolved organic carbon, biochemical oxygen demand, total organic carbon and total solids. K presented the highest concentration (1743 mg L-1) in TE, followed by Mg (135 mg L-1). Cd, Pb and Cr were present at low concentrations in the samples, 0.047, 0.206 and 0.081 mg L-1, respectively. Salmonella, thermotolerant coliforms and viable helminth eggs were not present in TE, which was applied as a biofertilizer in a lettuce crop and compared to mineral fertilization based on fresh matter and dry matter production. Lettuce production using TE was statistically equivalent to mineral fertilization. Toxic metals were not detected in the lettuce shoots. It was concluded that the anaerobically treated leachate from the composting company has the potential to be used as a biofertilizer.

This is an initial study using infrared (IR) in silico data as a standard database in preliminary method for new synthetic seized drugs. For this purpose, ten of the most common synthetic illicit seized substances on the Brazilian market were compared and computational chemistry was used as a tool for theoretical standard database. Infrared data from standard electronic library, experimental data from seized samples and data simulated by Density Functional Theory (DFT) were evaluated. The feasibility of the method was based on the degree of correlation of evaluated data. The results suggest that the computational methodology can be a viable way to analyze the structures of new synthetic drugs and obtain preliminary infrared profiles. Correlations data indicated a presumptive identification of the analyzed samples and it was also possible to observe a preliminary identification of drugs in five classes. However, it was observed that some synthetic cathinones and phenylethylamines were confused with amphetamines. Therefore, new studies must be developed to optimize the use of these data, because the simulated IR spectra can be advantageous to evaluate the profile of possible substances that may be synthesized in the future. ABSTRACT Papaya cultivation is widespread in Brazil, particularly in the states of Bahia and Espírito Santo, where most commercial plantations are concentrated. Owing to the economic and social importance of papaya, the present study aimed to determine the explanatory variables between the genotypes of two cultivars: Golden (from the Soil group) and Tainung Nº 1 hybrid (from the Formosa group), cultivated under high temperatures and hydric stress. The genotypes containing more desirable agronomic characteristics were identified for use in plant genetic improvement programs. Principal component analysis (PCA) was applied to select the desirable genotypes for Golden and Tainung Nº 1 cultivars based on specific variables analyzed for two groups of variables; for group 1, the plant height, stem diameter, leaf length, leaf width, and leaf number were analyzed, whereas for group 2, the leaf and root dry mass, stem dry mass and fresh mass of 10 discs, fresh mass, and stem and root fresh mass were analyzed. When exposed to hydric and thermal stress, the Tainung Nº 1 cultivar outperformed the Golden cultivar for the evaluation characteristics selected for use in genetic improvement programs.

RESUMO No Brasil, o cultivo do mamão é difundido, principalmente nos estados da Bahia e Espírito Santo, onde se concentra a maior parte dos plantios comerciais. Devido à importância econômica e social do mamão, este estudo teve como objetivo determinar variáveis explicativas entre os genótipos de mamoeiro, cultivados sob altas temperaturas e condições de estresse hídrico, para definir quais contêm características agronômicas mais desejáveis para serem utilizadas em programas de melhoramento genético de plantas. Para isso, a análise de componentes principais foi aplicada para selecionar genótipos desejáveis para as cultivares Golden e Tainung Nº 1, com base em dois grupos de variáveis: grupo 1 - altura da planta, diâmetro do caule, comprimento da folha, largura da folha e número de folhas; e grupo 2 - massa seca da folha, massa seca da raiz, massa seca do caule, massa seca de 10 discos, massa fresca de 10 discos, massa fresca, massa fresca do caule e massa fresca da raiz. A cultivar Tainung Nº 1 do grupo Formosa apresentou vantagens em relação à cultivar Golden, quando submetida a condições de estresse hídrico e térmico para as características de avaliação sendo mais indicada para ser utilizada em programas de melhoramento genético.

Criminal minds create new psychoactive substances (NPS) to dribble the Drug Regulatory System. Early Warning Systems (EWA) were instituted worldwide to combat this practice. Brazilian government has started establishing EWA at a national level since August 2021. The role of drug analysis laboratories in EWA is very important and experts working at forensic laboratories are in the main position to detect NPS and any changes in the molecular structures. However, for this practice, it is necessary to use reliable analyses techniques for unequivocal identification. The goal of this work is to use single-crystal X-ray diffraction (XRD) to detect the presence of NPS and elucidate its molecular structure using single crystals from a seized blotter paper. XRD is also useful to prepare reference substances to produce entries to be included in Fourier transformed infrared spectroscopy (FTIR) and mass spectrometry databases, which are some of the most relevant routine techniques in forensic laboratories. This approach was used to study the NPS 25R-NBOH and 25I-NBOMe using XRD, FTIR, Raman, and nuclear magnetic resonance spectroscopies. The results showed that molecular structure methods must be used to elucidate NPS in an unambiguous manner and the potential of XRD use in the forensic area as a reference method.

ABSTRACT Cowpea crops have high socioeconomic importance in the Northeast region of Brazil. These crops generate employment and income; in addition, it is an excellent source of protein, minerals, vitamins, and dietary fiber, and contributes to the food security of thousands of people. Biofortification of cowpea with iron, zinc, and proteins can contribute to prevent the higher hunger. The objective of this work was to assess iron, zinc, and protein contents in cowpea grains of different cultivars and the effect of grain size and color on the contents of these nutrients. Twenty-four cultivars with variation in size and color of the seed coat were evaluated. Iron contents ranged from 7.12 to 8.60 mg 100 g-1, with an overall mean of 7.75 mg 100 g-1. Zinc contents ranged from 4.46 to 4.93 mg 100 g-1, with an overall mean of 4.71 mg 100 g-1. Protein contents ranged from 31.50 to 36.24 g 100 g-1, with an overall mean of 33.57 g 100 g-1. Grain size ranged from 13.55 to 37.88 g, with an overall mean of 21.50 g. The cultivars Caldeirão, BRS-Guariba, and Pingo-de-Ouro-1-2 have higher iron contents; BRS-Aracê and BRS-Imponente have higher zinc contents; BR-14-Mulato, BRS-Guariba, BRS-Aracê, and BRS-Inhuma, have higher protein contents; and BR-3-Tracuateua and BRS-Imponente have larger grain sizes. The size and color of the seed coat do not influence the iron, zinc and protein contents of the evaluated cowpea cultivars.

RESUMO O feijão-caupi é uma cultura de grande importância socioeconômica na região Nordeste do Brasil. Além de ser um gerador de emprego e renda, é uma excelente fonte de proteínas, minerais, vitaminas e fibras alimentares, contribuindo para a segurança alimentar de milhares de pessoas. A biofortificação do grão de feijão-caupi com ferro, zinco e proteínas pode contribuir na prevenção da fome oculta. Este trabalho teve como objetivo caracterizar os teores de ferro, zinco e proteínas nos grãos de cultivares de feijão-caupi e verificar a influência da cor e tamanho do grão nos teores desses nutrientes. Foram avaliadas 24 cultivares com variação no tamanho e cor do grão. O teor de ferro variou de 7,12 a 8,60 mg 100g-1 e média geral de 7,75 mg 100g-1. O teor de zinco apresentou variação de 4,46 a 4,93 mg 100 g-1 e média geral de 4,71 mg 100 g-1. O teor de proteínas variou de 31,50 a 36,24 g 100g-1, com média geral de 33,57 107 g 100g-1. O tamanho do grão variou de 13,55 a 37,88 g e apresentou média geral de 21,50 g. As cultivares Caldeirão, BRS Guariba e Pingo de Ouro 1-2 apresentam maior teor de ferro; BRS Aracê e BRS Imponente, maior teor de zinco; BR 14-Mulato, BRS Guariba, BRS Aracê e BRS-Inhuma, maior teor de proteínas; e BR 3-Tracuateua e BRS Imponente, maior tamanho de grão. O tamanho e a cor do tegumento grão não influenciam nos teores de ferro, zinco e proteínas das cultivares de feijão-caupi avaliadas.

Resumo Allium sativum L. é uma erva da família Alliaceae com sabor e aroma específicos e propriedades medicinais e nutracêuticas amplamente comercializada em diversos países. O Brasil é um dos maiores importadores de alho do mundo, apesar da sua produção ser restrita e limitada ao consumo interno. Assim, explorar a diversidade genética do alho comercial conservado em bancos de germoplasma é essencial para fornecer informações genéticas adicionais acerca dessa cultura economicamente importante. Uma ferramenta adequada para esse fim é a caracterização citogenética desses acessos. Este estudo teve como objetivo caracterizar a diversidade citogenética entre sete acessos de alho de um Banco de Germoplasma no Brasil. Os cariótipos foram obtidos por coloração convencional e com os fluorocromos de cromomicina A3 (CMA) e 4,6-diamidino-2-fenilindol (DAPI). Todos os acessos analisados apresentaram número cromossômico 2n = 16, fórmula cariotípica 6M + 2SM, cariótipos simétricos, núcleos reticulados em intérfase e cromossomos com condensação uniforme da cromatina da prófase para a metáfase. A coloração com fluorocromos mostrou diferenças na quantidade e distribuição de heterocromatina ao longo dos cromossomos e entre os acessos estudados. Com base no padrão de distribuição desses pequenos polimorfismos, foi possível separar os sete acessos em três grupos. Também foi possível diferenciar individualmente alguns dos acessos. Um dos resultados obtidos mostrou distensão heteromórfica da região organizadora nucleolar observada nos pares dos cromossomos 6 ou 7 com características peculiares. Foi sugerido, por exemplo, que o bloco heteromórfico de heterocromatina (CMA +++ / DAPI-) no cromossomo 6 do acesso “Branco Mineiro Piauí” pode ser usado como um marcador para identificar esse genótipo ou pode estar associado a algum caráter de interesse econômico.

Abstract Allium sativum L. is an herb of the Alliaceae family with a specific taste and aroma and medicinal and nutraceutical properties that are widely marketed in several countries. Brazil is one of the largest importers of garlic in the world, despite of its production is restricted and limited to internal consumption. Thus, explore the genetic diversity of commercial garlic conserved at germplasm banks is essential to generate additional genetic information about its economically important crop. A suitable tool for this purpose is the cytogenetic characterisation of these accessions. This study aimed to characterise the cytogenetic diversity among seven accessions of garlic from a Germplasm Bank in Brazil. The karyotypes were obtained by conventional staining and with chromomycin A3 (CMA) and 4,6-diamidino-2-phenylindole (DAPI) fluorochromes. All accessions analysed showed chromosome number 2n = 16, karyotype formula 6M+2SM, symmetrical karyotypes, reticulate interphase nuclei, and chromosomes with uniform chromatin condensation from prophase to metaphase. The fluorochromes staining showed differences in the amount and distribution of heterochromatin along the chromosomes and between accessions studied. Based on the distribution pattern of these small polymorphisms, it was possible to separate the seven accessions into three groups. It was also possible to differentiate some of the accessions individually. One of the results obtained showed a heteromorphic distension of the nucleolar organiser region observed on the chromosome pairs 6 or 7 with peculiar characteristics. It was suggested for example, that the heteromorphic block of heterochromatin (CMA+++/DAPI-) on chromosome 6 of the “Branco Mineiro Piauí” accession can be used as a marker to identify this genotype or may be associated with some character of economic interest.

Abstract Human Respiratory Syncytial Virus (hRSV) infection results in death and hospitalization of thousands of people worldwide each year. Unfortunately, there are no vaccines or specific treatments for hRSV infections. Screening hundreds or even thousands of promising molecules is a challenge for science. We integrated biological, structural, and physicochemical properties to train and to apply the concept of artificial intelligence (AI) able to predict flavonoids with potential anti-hRSV activity. During the training and simulation steps, the AI produced results with hit rates of more than 83%. The better AIs were able to predict active or inactive flavonoids against hRSV. In the future, in vitro and/or in vivo evaluations of these flavonoids may accelerate trials for new anti-RSV drugs, reduce hospitalizations, deaths, and morbidity caused by this infection worldwide, and be used as input in these networks to determine which parameter is more important for their decision.

Resumo A infecção pelo Vírus Sincicial Respiratório Humano (hRSV) resulta na morte e hospitalização de milhares de pessoas em todo o mundo a cada ano. Infelizmente, não existem vacinas ou tratamentos específicos para tais infecções. A testagem de centenas, ou mesmo milhares, de moléculas promissoras é um desafio para a ciência. Neste trabalho, nós integramos propriedades biológicas, estruturais e físico-químicas para treinar e aplicar o conceito de inteligência artificial (IA) capaz de prever flavonoides com potencial atividade anti-hRSV. Durante as etapas de treinamento e simulação, a IA produziu resultados com taxas de acerto superiores a 83%, sendo capaz de prever flavonoides ativos ou inativos contra o hRSV. No futuro, avaliações in vitro e/ou in vivo desses flavonoides poderão acelerar os testes de novas drogas anti-RSV, reduzir hospitalizações, mortes e morbidade causadas por essa infecção. Além disso, a validação futura destes dados poderá determinar qual parâmetro tem maior peso na decisão da inteligência.

In this work, an iron/biochar composite was evaluated as a catalyst for 3,6-dichloro-2-methoxybenzoic acid (dicamba) herbicide degradation by heterogeneous Fenton-like process. The biochar was produced from pyrolysis of coffee husks. After FeII adsorption by biochar, the material was submitted to chemical reduction, via borohydride, producing zero-valent iron nanoparticles (BIO-Fe0). The material was characterized by Fourier transform infrared spectroscopy (FTIR), scanning electron microscopy and energy dispersive X-ray spectroscopy, Raman scattering spectroscopy, nitrogen adsorption/desorption analysis, among other techniques. The BIO-Fe0 is a heterogeneous material, and the main constituent elements are carbon (C), oxygen (O), silicon (Si), sulfur (S) and iron (Fe). The dicamba degradation was monitored by high-performance liquid chromatography and biological assays using beans (Phaseolus vulgaris) as indicator species. The commercial dicamba (500 mL; 250 mg L 1) was submitted to Fenton-like process (initial pH 3.20; BIO-Fe0: 3 or 5 g; 1.00 mL H2O2 at 3.00 mmol L 1; 1.00 mL Al2(SO4)3 at 1.00 mol L 1; 25 ºC and 5 h of reaction), presenting ca. 100% of degradation. These treated solutions were used in a biological assay. The intoxication levels were less 30%. These results show that BIO-Fe0 has the potential to be used in the dicamba degradation by Fenton-like processes.

Abstract Simultaneous analysis studies of several agronomic traits in cultivated plants make it possible to identify phenotypic and genotypic differences due to environmental variations, such as altitude. Therefore, the objective was to evaluate, through multivariate analysis of agronomic, physicochemical and physiological characters, passion fruit hybrids cultivated in different environments. The hybrids used were Gigante Amarelo, Rubi do Cerrado and Sol do Cerrado, cultivated in the southern region of Espírito Santo in four municipalities/environments: Marataízes (41 m), Jerônimo Monteiro (104 m), Alegre (711 m), and Ibitirama (1016 m). The agronomic characters of the plants, the physical-chemical characteristics of the fruits and the physical, biochemical and physiological qualities of the seeds were analyzed. The Singh method was used to determine the most important differentiating characters between hybrids growing in different environments. Based on these characters, a dissimilarity matrix was generated and a principal coordinate analysis was performed. It was observed that the pulp yield was influenced by altitude. The three hybrids showed greater performance in terms of agronomic characters at altitude (41 m) than at altitude (104 m). The Sol do Cerrado hybrid showed high performance in the physical-chemical characteristics of the fruits at altitude (104 m).

Resumo Estudos de análise simultânea de diversos caracteres agronômicos em plantas cultivadas permitem identificar diferenças fenotípicas e genotípicas devido a variações ambientais, como a altitude. Diante disso, objetivou-se avaliar através da análise multivariada de caracteres agronômicos, fisico-químicos e fisiológicos, híbridos de maracujazeiros cultivados em diferentes ambientes. Os híbridos utilizados foram Gigante Amarelo, Rubi do Cerrado e Sol do Cerrado, cultivados na região sul do Espírito Santo em quatro municípios/ambientes: Marataízes (41 m), Jerônimo Monteiro (104 m), Alegre (711 m), e Ibitirama (1016 m). Foram analisados os caracteres agronômicos das plantas, as características físico-químicas dos frutos e as qualidades físicas, bioquímicas e fisiológicas das sementes. O método Singh foi usado para determinar os caracteres diferenciadores mais importantes entre híbridos crescendo em diferentes ambientes. Com base nesses caracteres, uma matriz de dissimilaridade foi gerada e uma análise de coordenadas principais foi realizada. Observou-se que o rendimento de polpa foi influenciado pela altitude. Os três híbridos apresentaram maior desempenho em termos de caracteres agronômicos na altitude (41 m) do que na altitude (104 m). O híbrido Sol do Cerrado apresentou alto desempenho nas características físico-químicas dos frutos na altitude (104 m).

Abstract Aedes aegypti L. (Diptera: Culicidae) is the main transmitter of pathogens that cause human diseases, including dengue, chikungunya, zika and yellow fever. Faced with this problem, this study aims to select fungi with entomopathogenic potential against Ae. aegypti and develop formulations that optimize the control action of entomopathogenic fungi in the semi-field condition. 23 fungal strains native from Amazon were inoculated in Potato-Dextrose-Agar (PDA) culture medium for 14 days and then transferred by scraping to tubes containing 0.9% NaCl solution. To obtain the larvae, eggs were collected using traps in peridomestic environments for 7 days. 20 larvae of Ae. aegypti in 125 mL erlenmeyers containing 20 mL of conidial suspension at a concentration of 1x106 conidia/mL for initial selection and 1×104, 1×105, 1×106 and 1×107 conidia/mL for determination of LC50. Mortality was checked every 24 h for 5 days. The three fungi with the best virulence rates were identified using molecular techniques. The compatibility between fungi at a concentration of 1×106 conidia/mL and oily adjuvants, mineral oil and vegetable oil (andiroba, chestnut and copaiba) at concentrations of 0.1, 0.5 and 1% was evaluated. The germination capacity of 100 conidia per treatment was evaluated after incubation at 28 ºC for 24 h. To evaluate the entomopathogenic potential of the fungal formulations, conidial suspensions (1×106 conidia/mL) were added with 0.1% mineral and vegetable oil. The treatments were submitted to laboratory and semi-field conditions and mortality was verified every 24 h for 5 days. Beauveria sp. (4,458) (LC50 = 8.66× 103), Metarhizium anisopliae (4,420) (LC50 = 5.48×104) and M. anisopliae (4,910) (LC50 = 1.13×105) were significantly more effective in the larval control of Ae. aegypti, in relation to the other fungal morphospecies evaluated. Mineral oil was better compatible in all treatments evaluated. Beauveria sp. (4,458) was considerably less virulent under semi-field conditions. M. anisopliae (4,910) formulated with mineral oil increased larval mortality to 100% on the 4th day in the laboratory and on the 5th day in the semi-field. Fungal formulations developed from native Amazonian isolates represent a promising tool for the development of strategies to control Ae. aegypti.

Resumo Aedes aegypti L. (Diptera: Culicidae) é o principal transmissor de patógenos que causam doenças humanas, incluindo dengue, chikungunya, zika e febre amarela. Diante desse problema, este estudo tem como objetivo selecionar fungos com potencial entomopatogênico frente ao Ae. aegypti e desenvolver formulações que otimizem a ação de controle de fungos entomopatogênicos na condição de semi-campo. 23 cepas fúngicas nativas da Amazônia foram inoculadas em meio de cultura Potato-Dextrose-Agar (PDA) por 14 dias e depois transferidas por raspagem para tubos contendo solução de NaCl 0,9%. Para obtenção das larvas, os ovos foram coletados por meio de armadilhas em ambientes peridomiciliares por 7 dias. 20 larvas de Ae. aegypti em erlenmeyers de 125 mL contendo 20 mL de suspensão de conídios na concentração de 1×106 conídios/mL para seleção inicial e 1×104, 1×105, 1×106 e 1×107 conídios/mL para determinação de CL50. A mortalidade foi verificada a cada 24 horas durante 5 dias. Os três fungos com as melhores taxas de virulência foram identificados por meio de técnicas moleculares. Foi avaliada a compatibilidade entre fungos na concentração de 1×106 conídios/mL e adjuvantes oleosos, óleo mineral e óleo vegetal (andiroba, castanha e copaíba) nas concentrações de 0,1, 0,5 e 1%. A capacidade germinativa de 100 conídios por tratamento foi avaliada após incubação a 28 ºC por 24 h. Para avaliar o potencial entomopatogênico das formulações fúngicas, suspensões de conídios (1×106 conídios/mL) foram adicionadas de óleo mineral e vegetal a 0,1%. Os tratamentos foram submetidos a condições de laboratório e semi-campo e a mortalidade foi verificada a cada 24 horas durante 5 dias. Beauveria sp. (4.458) (LC50 = 8,66×103), Metarhizium anisopliae (4.420) (LC50 = 5,48×104) e M. anisopliae (4.910) (LC50 = 1,13×105) foram significativamente mais eficazes no controle larval de Ae. aegypti, em relação às demais morfoespécies fúngicas avaliadas. O óleo mineral foi melhor compatível em todos os tratamentos avaliados. Beauveria sp. (4.458) foi consideravelmente menos virulento em condições de semi-campo. M. anisopliae (4.910) formulado com óleo mineral aumentou a mortalidade larval para 100% no 4º dia no laboratório e no 5º dia no semi-campo. Formulações fúngicas desenvolvidas a partir de isolados nativos da Amazônia representam uma ferramenta promissora para o desenvolvimento de estratégias de controle de Ae. aegypti.

Abstract The impact of the Antarctic Ozone Hole Influence over Southern Brazil in October 2015 was analyzed using daily mean data of the Total Column Ozone (TCO), Ultraviolet Index (UVI) and Radiative Cloud Fraction (RCF) from the Ozone Monitoring Instrument satellite instrument. Vertical profiles and fields of ozone content and Potential Vorticity available from the European Centre for Medium-Range Weather Forecast reanalysis, air masses backward trajectories from the HYbrid Single-Particle Lagrangian Integrated Trajectory model and channel 3 water vapor images from the Geostationary Operational Environmental Satellite GOES-13 were also analyzed. The five identified events showed an -7.4±2.3% average TCO reduction, leading to an +16.6±54.6% UVI increase even with a predominance of partly cloudy days. Other impacts were observed in the ozone profiles, where the most significant anomalies occurred from 650 K reaching 1.2 ppmv at the 850 K level. In the ozone fields at 700 K, the presence of a polar origin tongue was observed causing negatives anomalies between -0.2 and 0.4 ppmv in a transient system format forced with eastward-traveling Rossby waves passing through the Southern of Brazil and Uruguay.

ABSTRACT Cover crop species have been used in mixes in order to improve soil conditions; however, it is important to evaluate the relationship of the employed cultivars with phytonematodes to identify those that do not multiply the pathogens in the cultivated area. This study aimed to assess the response of 11 plant species used in cover crop mix to Meloidogyne enterolobii, M. incognita and Pratylenchus brachyurus. Six experiments were conducted in a greenhouse, using a completely randomized design, with five replications. BMX Potência RR soybean and Santa Clara tomato were used as controls. In at least one of the experiments, IPR 91 Baili buckwheat was susceptible (reproduction factor - RF ≥ 1) to M. enterolobii, M. incognita and P. brachyurus. Common white lupin, URS Altiva white oat, Iapar 61 (Ibiporã) black oat and Embrapa 139 (Neblina) black oat were susceptible to M. enterolobii and M. incognita and resistant (RF < 1) and/or immune (RF = 0) to P. brachyurus. IPR 89 rye and IPR 116 radish were found susceptible to M. incognita and P. brachyurus and resistant to M. enterolobii. BRS 1501 millet and Embrapa 29 (Garoa) black oat were susceptible to M. incognita and resistant/immune to M. enterolobii and P. brachyurus. Brachiaria ruziziensis was considered resistant to M. enterolobii and M. incognita and susceptible to P. brachyurus. URS Corona white oat was the only cover crop considered to be resistant and/or immune to all the nematode species evaluated.

RESUMO Espécies de plantas de cobertura vêm sendo utilizadas em mixes, visando melhorar as condições do solo; porém, é importante avaliar a relação das cultivares utilizadas com fitonematoides, a fim de identificar aquelas que não multipliquem os patógenos na área cultivada. Objetivou-se avaliar a reação de 11 espécies vegetais utilizadas em mix de plantas de cobertura a Meloidogyne enterolobii, M. incognita e Pratylenchus brachyurus. Foram conduzidos 6 experimentos em casa-de-vegetação, em delineamento inteiramente casualizado, com 5 repetições, utilizando-se a soja BMX Potência RR e o tomate Santa Clara como testemunhas. Em pelo menos um dos experimentos, o trigo mourisco IPR 91 Baili foi suscetível (fator de reprodução - FR ≥ 1) a M. enterolobii, M. incognita e P. brachyurus. O tremoço-branco cv. comum, aveia branca URS Altiva, aveia preta Iapar 61 (Ibiporã) e aveia preta Embrapa 139 (Neblina) foram suscetíveis a M. enterolobii e M. incognita e resistentes (FR < 1) e/ou imunes (FR = 0) a P. brachyurus. O centeio IPR 89 e o nabo forrageiro IPR 116 foram suscetíveis a M. incognita e P. brachyurus e resistentes a M. enterolobii. O milheto BRS 1501 e a aveia preta Embrapa 29 (Garoa) foram suscetíveis a M. incognita e resistentes/imunes a M. enterolobii e P. brachyurus. A Brachiaria ruziziensis foi considerada resistente a M. enterolobii e M. incognita e suscetível a P. brachyurus. A aveia branca URS Corona foi a única planta considerada resistente e/ou imune para todas as espécies de nematoides avaliadas.
