Abstract Background Alpha-kinase 1 (ALPK1) is a master regulator in inflammation and has been proved to promote renal fibrosis by promoting the production of IL-1β in diabetic nephropathy (DN) mice. Pyroptosis is involved in high glucose (HG)-induced tubular cells injury, characterized by activation of Gasdermin D (GSDMD) and the release of IL-1β and IL-18, resulting in inflammatory injury in DN. It is reasonable to assume that ALPK1 is involved in pyroptosis-related tubular injury in DN. However, the mechanism remains poorly defined. Methods Immunohistochemistry (IHC) staining was performed to detect the expression of pyroptosis- and fibrosis-related proteins in renal sections of DN patients and DN mice. DN models were induced through injection of streptozotocin combined with a high-fat diet. Protein levels of ALPK1, NF-κB, Caspase-1, GSDMD, IL-1β, IL-18 and α-SMA were detected by Western blot. HK-2 cells treated with high-glucose (HG) served as an in vitro model. ALPK1 small interfering RNA (siRNA) was transfected into HK-2 cells to down-regulate ALPK1. The pyroptosis rates were determined by flow cytometry. The concentrations of IL-1β and IL-18 were evaluated by ELISA kits. Immunofluorescence staining was used to observe translocation of NF-κB and GSDMD. Results The heat map of differentially expressed genes showed that ALPK1, Caspase-1 and GSDMD were upregulated in the DN group. The expression levels of ALPK1, Caspase-1, GSDMD and CD68 were increased in renal biopsy tissues of DN patients by IHC. ALPK1expression and CD68+ macrophages were positively correlated with tubular injury in DN patients. Western blot analysis showed increased expressions of ALPK1, phospho-NF-κB P65, GSDMD-NT, and IL-1β in renal tissues of DN mice and HK-2 cells, accompanied with increased renal fibrosis-related proteins (FN, α-SMA) and macrophages infiltration in interstitial areas. Inhibition of ALPK1 attenuated HG-induced upregulation expressions of NF-κB, pyroptosis-related proteins Caspase-1, GSDMD-NT, IL-1β, IL-18, α-SMA, and pyroptosis level in HK-2 cells. Also, the intensity and nuclear translocation of NF-κB and membranous translocation of GSDMD were ameliorated in HG-treated HK-2 cells after treatment with ALPK1 siRNA. Conclusions Our data suggest that ALPK1/NF-κB pathway initiated canonical caspase-1-GSDMD pyroptosis pathway, resulting in tubular injury and interstitial inflammation of DN.

Abstract Objectives To investigate the changes in the coagulation function and hemodynamic parameters in patients with Hemorrhagic Traumatic Shock (HTS) after restrictive fluid resuscitation. Methods A total of 139 patients with HTS admitted to our hospital were enrolled, among which 69 HTS patients were divided into the control group and the remaining 70 HTS patients as the observation group. Patients in the control group underwent regular fluid resuscitation, while those in the observation group underwent restrictive fluid resuscitation. Results During treatment, 70 patients in the observation group had a lower bleeding amount, infusion amount, and blood transfusion volume than those in the control group (p < 0.05). After treatment, patients in the observation group had better hemodynamic parameters and blood coagulation than those in the control group (p < 0.05), and the incidence rate in the observation group was only 12.9%, which was significantly lower than 60.87% in the control group, while the cure rate in the observation group was 100%, which was significantly higher than that in the control group (p < 0.05). Conclusions Restrictive fluid resuscitation could remarkably increase the cure rate and reduce the bleeding amount during HTS treatment, thereby benefiting the recovery of the patient's blood coagulation.

Abstract Under the influence of the COVID-19, people's awareness of physical health and immunity has increased significantly. Chitooligosaccharide is an oligomer of β-(1, 4)-linked D -glucosamine, furthermore, is one of the most widely studied immunomodulators. Chitooligosaccharide can be prepared from the chitin or chitosan polymers through enzymatically, chemically or physically processes. Chitooligosaccharide and its derivatives have been proven to have a wide range of biological activities including intestinal flora regulation, immunostimulant, anti-tumor, anti-obesity and anti-oxidation effects. This review summarizes the latest research of the preparation methods, biological activities in immunity and safety profiles of Chitooligosaccharide and its derivatives. We recapped the effect mechanisms of Chitooligosaccharide basing on overall immunity. Comparing the effects of Chitooligosaccharide with different molecular weights and degree of aggregation, a reference range for usage has been provided. This may provide a support for the application of Chitooligosaccharide in immune supplements and food. In addition, future research directions are also discussed.

Abstract Objective: This study aimed to perform a meta-analysis to investigate the diagnostic safety and accuracy of Ultrasound-Guided Core Needle Biopsy (US-CNB) Axillary Lymph Nodes (ALNs) region in patients with Breast Cancer (BC). Methods: The authors searched the electronic databases PubMed, Scopus, Embase, and Web of Science for clinical trials about US-CNB for the detection of ALNs in breast cancer patients. The authors extracted and pooled raw data from the included studies and performed statistical analyses using Meta-DiSc 1.4 and Review Manager 5.3 software. A random effects model was used to calculate the data. At the same time, data from the Ultrasound-guided Fine-Needle Aspiration (US-FNA) were introduced for comparison with the US-CNB. In addition, the subgroup was performed to explore the causes of heterogeneity. (PROSPERO ID: CRD42022369491). Results: In total, 18 articles with 2521 patients were assessed as meeting the study criteria. The overall sensitivity was 0.90 (95% CI [Confidence Interval], 0.87‒0.91; p = 0.00), the overall specificity was 0.99 (95% CI 0.98‒ 1.00; p = 0.62), the overall area under the curve (AUC) was 0.98. Next, in the comparison of US-CNB and US-FNA, US-CNB is better than US-FNA in the diagnosis of ALNs metastases. The sensitivity was 0.88 (95% CI 0.84‒ 0.91; p = 0.12) vs. 0.73 (95% CI 0.69‒0.76; p = 0.91), the specificity was 1.00 (95% CI 0.99‒1.00; p = 1.00) vs. 0.99 (95% CI 0.67‒0.74; p = 0.92), and the AUC was 0.99 vs. 0.98. Subgroup analysis showed that heterogeneity may be related to preoperative Neoadjuvant Chemotherapy (NAC) treatment, region, size of tumor diameter, and the number of punctures. Conclusion: US-CNB has a satisfactory diagnostic performance with good specificity and sensitivity in the preoperative diagnosis of ALNs in BC patients.

ABSTRACT Objective: To explore the factors affecting short-term prognosis of circulatory failure patients undergoing venoarterial extracorporeal membrane oxygenation (VA-ECMO) treatment. Methods: A total of 136 patients undergoing VA-ECMO were enrolled in this study and subsequently divided into the death group (n=35) and the survival group (n=101) based on whether death occurred during hospitalisation. Extracorporeal membrane oxygenation (ECMO) running time, length of intensive care unit stay, length of hospital stay, costs, and ECMO complications were then compared between the two groups. Results: The average age of all patients undergoing ECMO was 47.64±16.78 years (53.2±16.20 years in the death group and 45.713±16.62 years in the survival group) (P=0.022). Patients in the survival group exhibited a clear downward trend in lactic acid value following ECMO treatment compared to those in the death group. Total hospitalisation stay was longer in the survival group (35 days) than in the death group (15.5 days) (P<0.001). In the analysis of ECMO complications, the incidence of neurological complications, renal failure, limb complications, and infection were higher in the death group than in the survival group (P<0.05 for all). Specifically, as a risk factor for patient survival and discharge, the occurrence of infection will lead to increased hospitalisation stays and costs (P<0.05 for both). Conclusion: Complications such as kidney failure and infection are associated with in-hospital death, and ECMO-related complications should be actively prevented to improve the survival rate of VA-ECMO treatment.

RESUMO Objetivo: O futebol é o esporte preferido pelos estudantes universitários masculinos entre muitos esportes na China. Métodos: De acordo com a virilidade e as características competitivas dos estudantes universitários masculinos, um curso eletivo de futebol pode exercitar seus corpos e melhorar sua qualidade física. Embora os cursos eletivos afetem positivamente a saúde física dos estudantes universitários, diferentes projetos têm funções diferentes devido a seus outros conteúdos didáticos, formas de ensino, material didático, e métodos de ensino. Resultados: a implementação dos padrões nacionais da China para o físico dos estudantes universitários apresenta novas exigências para o nível de aptidão física dos estudantes universitários masculinos. No entanto, a pesquisa mostra que o nível geral de aptidão física dos estudantes universitários chineses está diminuindo, e a intensidade de exercício da maioria dos estudantes não pode atender às exigências. Conclusões: Para estudar a influência do esporte sobre o físico e a personalidade dos estudantes de acordo com o Student Physique and Health Standard, este documento estuda e analisa o impacto dos cursos eletivos de futebol universitário no corpo e na personalidade dos estudantes universitários e discute suas estratégias de treinamento. ABSTRACT Objective: Soccer is a favored sport for male college students among many sports in China. Methods: According to male college students’ virility and competitive characteristics, an elective soccer course can exercise their bodies and improve their physical quality. Although elective courses positively affect college students’ physical health, different projects have different functions due to their other teaching content, teaching forms, teaching material, and teaching methods. Results: the implementation of China’s national standards for college students’ physique presents new requirements for the fitness level of male college students. However, the research shows that the overall fitness level of Chinese college students is decreasing, and the exercise intensity of most students cannot meet the requirements. Conclusions: To study the influence of sports on students’ physique and personality according to the Student Physique and Health Standard, this paper studies and analyzes the impact of college soccer elective courses on college students’ bodies and personalities and discusses their training strategies. Abstract Coronary heart disease (CHD) is one of the main diseases causing morbidity and mortality globally, with oxidative damage and lipid peroxidation associated with oxidative stress and atherosclerosis. Cinnamomum migao is a Miao ethnomedicine that has been shown to provide relief from CHD symptoms. A gas chromatograph-mass spectrometer (GC-MS) was used to determine the chemical components of ten batches of migao fatty oil (MFO), yielding a total of 35 chemical constituents. The antioxidant activity of MFO in vitro was assessed using the DPPH· and ABTS·+ assay methods, and both indicated good antioxidant activity. The network pharmacology predicted the active constituents of MFO and their therapeutic influence on the mechanism of oxidative damage-induced coronary heart disease (OD-CHD). Six compounds were discovered in MFO, including tau-cadinol acetate and dodecanoic acid, which may play a role in OD-CHD by acting on lipids and atherosclerosis, the PI3K-Akt signalling pathway, and other pathways. In conclusion, our study lays the foundation for investigating the use of MFO in the treatment of OD-CHD.

RESUMEN La alta concentración de sal de cromo ha causado una grave contaminación al medio ambiente desde su producción. El residuo de cromo de larga data ha contaminado el suelo y la concentración total de cromo de algunos suelos contaminados ha alcanzado los 30000 mg / kg. Para la remediación de suelos contaminados con cromo se propuso el método electrodinámico mejorado. ABSTRACT High concentration of chromium salt has caused serious pollution to the environment since its production. The long-standing chromium residue has polluted the soil, and the total chromium concentration of some polluted soil has reached 30000 mg / kg. For the remediation of chromium contaminated soil, the enhanced electrodynamic method was proposed. In order to improve the efficiency of electrokinetic remediation of chromium contaminated soil, two enhanced electrokinetic remediation technologies were proposed: Electrokinetic oxidation enhanced remediation technology and electrokinetic enhanced remediation technology. Cr(III) in soil was polluted by oxidant chromium oxide to increase the content of dissolved Cr(VI),so as to improve the efficiency of electric repair, in order to find a suitable PRB medium as a breakthrough to improve the repair efficiency. The experimental results show that compared with the traditional electrodynamic technology, the enhanced electrodynamic method can effectively improve the removal rate of total chromium in the soil, and provide technical support for the enhanced electrodynamic remediation of chromium contaminated soil. The pH control system makes the pH of anode and cathode electrolyte always maintain acidic, and the H+ migration speed is faster than that of OH-, and the pH of soil near the anode is lower than that near the cathode.

The etiology of polycystic ovary syndrome (PCOS) is complex and the pathogenesis is not fully understood. Some studies have shown that dysregulation of ovarian granulosa cells may be related to abnormal follicles and excessive androgen in women with PCOS. Our team has also confirmed the high expression status of H19 in PCOS patients in the early stage. However, the relationship between H19 and miR-19b in the development of PCOS is still unknown. Therefore, we used bioinformatics to predict the binding sites of human H19 and miR-19b, and of miR-19b and CTGF genes. After the silencing and overexpression of H19, real-time polymerase chain reaction (PCR) was used to detect the expressions of H19, miR-19b, and CTGF. Western blotting was used to detect CTGF protein. Proliferation of KGN cells after H19 silencing was detected by CCK8. Flow cytometry was used to detect the apoptosis of KGN cells after H19 silencing. After the overexpression of H19, it was found that the expression of miR-19b gene decreased and the expression of CTGF increased, whereas silencing of H19 did the opposite. In addition, H19 could promote cell proliferation and decrease cell apoptosis. Finally, luciferase reporter assays showed that the 3′-end sequences of lncRNA H19 and CTGF contained the binding site of miR-19b. In conclusion, our study indicated that lncRNA H19 acted as a ceRNA to bind to miR-19b via a “sponge” to regulate the effect of CTGF on KGN cells, which may play a vital role in PCOS.

SUMMARY: Matrigel is a basement membrane matrix extracted from the EHS mouse tumor containing extracellular matrix protein, its main components are laminin, type IV collagen, nestin, heparin sulfate, growth factor and matrix metalloproteinase.At room temperature, Matrigel polymerized to form a three dimensional matrix with biological activity. It can simulate the structure, composition, physical properties and functions of the cell basement membrane in vivo, which is beneficial to the culture and differentiation of the cells in vitro, and can be used for the study of cell morphology, biochemical function, migration, infection and gene expression. In this study, Matrigel three-dimensional culture model of bone marrow mesenchymal stem cells(BMSCs) was established, and its morphology, proliferation and survival were observed. BMSCs were isolated and cultured with whole bone marrow adherence method. The Second generation BMSCs with good growth condition were selected and mixed with Matrigel to form cell gel complexes. The morphology and proliferation of mesenchymal stem cells were observed by phase contrast microscope and HE staining,Live/Dead staining was used to evaluate the cell activity.Phase contrast microscopy showed that BMSCs were reticulated in Matrigel and proliferated well, After 7 days, the matrix gel gradually became soft and collapsed, a few cell reticular crosslinking growth was seen at 14 days; HE staining showed that the cytoplasm of the cells was larger on the fourth day and the cells were elongated and cross-linked on the seventh day; Live/dead staining showed that most cells showed green fluorescence with the prolongation of culture time, on the first, 4 and 7 days, the activity of bone marrow mesenchymal stem cells in Matrigel gradually increased, and the percentages were 92.57 %, 95.54 % and 97.37 %, respectively. Matrigel three-dimensional culture system can maintain the morphology, function and proliferation ability of bone marrow mesenchymal stem cells.

RESUMEN: Matrigel es una matriz de membrana basal extraída del tumor de ratón EHS que contiene proteína de matriz extracelular. Los componentes principales son laminina, el colágeno tipo IV, nestina, sulfato de heparina, factor de crecimiento y metaloproteinasa de matriz. A temperatura ambiente, Matrigel se polimerizó para formar una matriz tridimensional. Es posible simular la estructura, la composición, las propiedades físicas y las funciones de la membrana basal celular in vivo, lo que es beneficioso para el cultivo y la diferenciación de las células in vitro, y se puede utilizar para el estudio de la morfología celular, la función bioquímica, la migración, infección y expresión génica. En este estudio, se estableció el modelo de cultivo tridimensional Matrigel de células madre mesenquimales de médula ósea (BMSC), y se observó su morfología, proliferación y supervivencia. Las BMSC fueron aisladas y cultivadas con el método de adherencia de la médula ósea completa. Se seleccionaron las BMSC de segunda generación con buenas condiciones de crecimiento y se mezclaron con Matrigel para formar complejos de gel de células. La morfología y la proliferación de las células madre mesenquimales se observaron con microscopio de contraste de fase y se tiñó con Hematoxilina-Eosina (HE); para evaluar la actividad celular se usó la tinción Live/Dead. La microscopía de contraste mostró que las BMSC se reticularon en Matrigel y proliferaron bien. Después de 7 días, se observó que el gel de matriz gradualmente se volvió blando y colapsó, y se visualizó un cruce transversal de algunas células reticulares a los 14 días. La tinción mostró que la mayoría de las células mostraron una fluorescencia verde con la prolongación del tiempo de cultivo; en los primeros 4 y 7 días, la actividad de las células madre mesenquimales de la médula ósea en Matrigel aumentó gradualmente y los porcentajes fueron de 92,57 %, 95,54 % y 97,37 %, respectivamente. El sistema de cultivo tridimensional de Matrigel puede mantener la morfología, la función y la capacidad de proliferación de las células madre mesenquimales de la médula ósea.

RESUMEN A través de una gama de técnicas petrológicas se estudió la petrología, la diagénesis, las características de poro y los factores que controlan las variaciones regionales de la calidad del yacimiento de las areniscas Chang 7. ABSTRACT Through a range of petrological techniques, the petrology, diagenesis, pore characteristics, and controlling factors on the regional variations of reservoir quality of the Chang 7 sandstones were studied. These sandstones, mainly arkoses, lithic arkoses, and feldspathic litharenites, were deposited in a delta front and turbidites in semi-deep to deep lacustrine. The detrital constituents were controlled by the provenance and sedimentary condition, which resulted in a spatially variable composition; e.g., high biotite and feldspar contents in the northeast (NE) of the study area, and high contents of rock fragments, especially dolomite, matrix, and quartz in the southwest (SW). Diagenesis includes intense mechanical compaction, cementation, and dissolution of unstable minerals. Diagenetic minerals which were derived internally include quartz, ankerite, ferrous calcite, albite, illite, kaolinite, and chlorite. Thus the original sandstone composition hadfirm control over the development and distribution of cement. Mechanical compaction and late-stage cementations contribute to the porosity loss of sandstones of Chang7 member. The dissolution porosity in major sandstone, slightly higher than primary porosity is principally dependent on the accessibility of acid fluid. The high content of plastic component facilitated the reduction of primary porosity and limited the mineral dissolution. The best reservoir sandstones are found in W, and partly from NE, M districts, with porosity are primary. The relatively high textural maturity of these sandstones reduces the impact of compaction on primary pores, and commonly existed chlorite rims limited the precipitation of pore filling quartz and carbonate cementation in late stage.

Abstract The clustered regularly interspaced short palindromic repeat (CRISPR)/CRISPR-associated (Cas) technology has recently emerged as a powerful genomic editing tool with great potential for crop breeding. However, commonly used protocols for screening of CRISPR/Cas9-induced mutations are laborious, time-consuming, and costly. In the present study we examined the applicability of high resolution melting (HRM) analysis fast screening CRISPR/Cas9-induced mutations in T0 plants and subsequent genotyping of T1 populations. Comparative analysis demonstrated that HRM analysis could identify mutant T0 plants carrying various types of mutation, including single nucleotide substitutions and short insertion/deletions, with false positive/negative rates of 0-2.78%. Furthermore, T1 plants derived from single T0 plants could be correctly genotyped by HRM analysis using WT parents and T0 plants as controls. We hence recommend the adoption of HRM analysis in CRISPR/Cas9 mediated genetic studies and breeding in rice and other crop species.

ABSTRACT In this paper, the zinc carbonate hydroxide (Zn5(OH) 6(CO2)2, ZCHO) was prepared by a facile hydrothermal method assisted chitosan (CS). The as-prepared product was characterized by X-ray diffraction (XRD), scanning electron microscopy (SEM) and thermogravimetric analysis (TG). Results indicated that every micro-sized ZCHO particle had a multi-layered structure and was composed of many ZCHO nanoplates. The used CS played the role of the template in the formation of well multi-layered ZCHO particles. TG measurements had further indicated that the remained content of CS in the as-prepared ZCHO powders was about 8 Wt.%. The adsorption isotherm results showed that the ZCHO powders with CS exhibited massive removal of Cu2+ from aqueous solution at room temperature. The equilibrium adsorption was well described by the Freundlich isotherm model. Based on the above experimental results, the possible removal mechanism of Cu2+ by the multi-layered ZCHO was proposed.

The aim of this study was to find related pathogenic genes in cerebral autosomal dominant arteriopathy with subcortical infarcts and leukoencephalopathy in (CADASIL)-like patients. The direct sequencing and high-throughput multiplex polymerase chain reaction (PCR) was performed to screen for related genes. The clinical and imaging data of a CADASIL-like patient (the pro-band) and his family members were collected. At first, the known hereditary cerebral vascular genes of the pro-band were screened with direct sequencing to find candidate gene mutations. High-throughput multiplex PCR was then used to analyze the single nucleotide polymorphism of the candidate gene in the family members. The results showed that there was missense mutation of the high temperature requirement protease A1 (HTRA1) gene in the pro-band, which may be a pathogenic factor according to the biological software analysis. The following SNP results revealed that the other family members also had the HTRA1 gene mutation. Thus, the CADASIL-like family disease may be caused by heterozygous HTRA1 gene mutation, which leads to autosomal dominant hereditary cerebral small vessel disease.
