ABSTRACT The fertile land in Heilongjiang Province of China is suitable for rice cultivation, but this area is susceptible to low temperature and chilling injury, which is prevented by planting rice varieties suitable for GP that is an important measure. However, selection based on rice traits is vulnerable to environmental influences and takes a long time, and selection based on molecular markers may result in progeny recombination and lack of reliability. Therefore, an efficient accurate and intelligent identification method for rice growth period is urgently needed. In this study, machine learning and deep learning methods in Python were used to analyze the Raman spectra of 6 rice varieties in three accumulated temperature region of Heilongjiang Province. 1) In machine learning, Principal Component Analysis (PCA) was adopted for feature extraction, in combination with Support Vector Machine (SVM) classification models suitable for nonlinear data sets for identification, the identification rate was 93.33% and the type of this experimental data set was determined to be discrete. 2) In deep learning, Continuous Wavelet Transform (CWT) methods was adopted for data preprocessing, combined with the Convolutional Neural Networks (CNN) model with its own feature extraction, with the highest accuracy of 94.82%, which was higher than the PCA+SVM identification model. 3) Based on the method mentioned in 2), in order to improve the feature extraction ability of the model as a whole, Convolutional Block Attention Module (CBAM) was used to improve the CNN identification model for the first time for one-dimensional data sets, and the highest identification rate was 98.28%, which was better than the PCA+SVM identification model. 3) In the verification test, Raman spectral information of 4 rice varieties was brought into the constructed CWT+CNN-CBAM identification model for identification, and the identification results were as high as 94.79%. The experimental results showed that the CWT visualization data processing method based on Raman technology combined with the CNN identification model of CBAM with improved feature extraction ability in deep learning achieved the best identification results, which could provide an efficient, accurate and intelligent method for the identification of different growth period of rice varieties in Heilongjiang Province.

ABSTRACT Direct seeded canola (Brassica napus L.) has become increasingly popular in China’s key canola-producing regions. The studies on the K response of direct-seeded canola performance are limited. This research aimed to investigate the K utilization efficiency and canopy radiation interception rate of direct seeded winter canola at various K application rates. Two seasons field trials were done with five K levels (0, 75, 150, 225, and 300 kg K2O ha-1) utilizing two key local cultivars (Huayouza no. 9 and Zhongshuang no. 11) in Southwest China. Canola yield rose by 17.9%-82.6% in 2017-2018 and 21.1%-73.6% in 2018-2019 when K was applied, compared to K-unfertilized. Canola yield go up rapidly at first and then gradually when K ingestion grew, demonstrating the phenomena of K luxury consumption. As K levels rose, internal utilization efficiency, agronomic efficiency, and physiological efficiency all decreased. In canola pod development and maturation period, 150 kg K2O ha-1 intercepted the most solar radiation (73.8%), while K-unfertilized treatment intercepted the least (61.6%) on average. The K supply rates had a significant effect on canopy radiation interception and K absorption in winter canola. When K application exceeded a specific rate, luxury consumption occurred. Canola’s luxury K consumption was mostly stored in the pericarp and plant stem.

Abstract Long non-coding RNA AK001796 was initially identified altered in lung cancer. Recent research showed it could participate in the prognosis of hepatocellular carcinoma (HCC). However, the general biological role of AK001796 and its underlying mechanisms in HCC remain unclear. Here we demonstrated that the expression level of AK001796 in HCC tissues and cell lines was up-regulated. Silencing AK001796 suppressed the proliferation ability of HCC cells. Through dual luciferase reporter assays and loss/gain of functions studies, we identified that AK001796 could bind to miR-150, a star microRNA, promoting HCC proliferation. Furthermore, it was reported that growth factor receptor binding protein 2-associated binder 1 (GAB1) is a target gene of miR-150. Owing to AK001796 being a decoy for miR-150 and binding the same putative sites of miR-150 as GAB1, we presented that inhibition of miR-150 in AK001796 silencing cells reversed the reduction in GAB1. Subsequently, our findings demonstrated that silencing AK001796 can impair phospho-ERK1/2 and phospho-AKT. In conclusion, our investigation revealed that AK001796 promoted proliferation by enhancing phospho-ERK1/2 and phospho-AKT through AK001796/miR-150/GAB1 axis in HCC. These results provided further evidence for the critical roles of AK001796 accumulating HCC and suggested that AK001796 might act as an HCC biomarker in clinical treatment.

This research investigated the predictive value of combined detection of brain natriuretic peptide (BNP) and cystatin C (Cys C) in heart failure after percutaneous coronary intervention (PCI) in patients with acute myocardial infarction (AMI). Sixty-five AMI patients complicated by heart failure (HF) after PCI and 79 non-heart failure (non-HF) patients were involved in this research. The levels of Cys C and BNP were measured. Risk factors for heart failure in AMI patients after PCI were analyzed by multivariate logistic regression analysis. Efficacy of BNP and Cys C on predicting heart failure were analyzed by receiver operating characteristic (ROC) curve. Cys C and BNP levels were significantly higher in the HF group than in the non-HF group. BNP and Cys C levels were the independent influencing factors causing heart failure within one year after PCI. The area under the predicted curve (AUC) of Cys C, BNP, and combined Cys C and BNP were 0.763, 0.829, and 0.893, respectively. The combined detection of Cys C and BNP was highly valuable in predicting heart failure in AMI patients after PCI, which can be regarded as the serum markers for diagnosis and treatment of heart failure.

ABSTRACT Objective: To determine the absolute number of serum T lymphocytes and cytokine levels and the characteristics of patients with active pulmonary tuberculosis and to assess their effect on the immune status of these patients and their diagnostic and predictive value for tuberculosis. Methods: We included 1,069 patients with active tuberculosis, 51 patients with latent tuberculosis infection, and 600 health individuals. Absolute serum T-lymphocyte counts and cytokine levels were quantified. Results: T lymphocytes were significantly reduced in patients with active tuberculosis when compared with healthy individuals. The immune function of patients gradually decreased with age and was stronger in female patients than in males. Th1 cells expressed higher levels of cytokines than did Th2 cells. Logistic regression analysis showed that reduced CD3+ T, CD8+ T, and NK cell counts, as well as reduced IL-4 and IFN-g expression, were independent influencing factors for active tuberculosis. ROC analysis showed that the sensitivity and specificity of absolute CD3+ T and CD8+ T lymphocyte counts and combined factors were significantly higher than were those of IL-4 and IFN-g for diagnosing active tuberculosis. Conclusions: Serum T-lymphocyte counts and cytokine levels can assess the immune status of tuberculosis patients; they are also useful biomarkers for predicting and diagnosing tuberculosis.

RESUMO Objetivo: Determinar o número absoluto de linfócitos T séricos e os níveis de citocinas séricas, bem como as características, de pacientes com tuberculose pulmonar ativa e avaliar o efeito desses no estado imunológico desses pacientes e seu valor diagnóstico e preditivo para tuberculose. Métodos: Foram incluídos 1.069 pacientes com tuberculose ativa, 51 pacientes com tuberculose latente e 600 indivíduos saudáveis. Foram realizadas a contagem absoluta de linfócitos T séricos e a quantificação de citocinas séricas. Resultados: Os linfócitos T estavam significativamente reduzidos nos pacientes com tuberculose ativa em comparação com os indivíduos saudáveis. A função imunológica dos pacientes diminuiu gradativamente com a idade e mostrou-se mais forte nas mulheres do que nos homens. As células Th1 expressaram maiores níveis de citocinas do que as células Th2. A análise de regressão logística mostrou que contagens reduzidas de células T CD3+, T CD8+ e NK e expressão reduzida de IL-4 e IFN-g foram fatores de influência independentes para tuberculose ativa. A análise ROC mostrou que a sensibilidade e especificidade dos valores absolutos de linfócitos T CD3+ e T CD8+ e de fatores combinados foram significativamente maiores do que as da IL-4 e do IFN-g para o diagnóstico da tuberculose ativa. Conclusões: A contagem de linfócitos T séricos e os níveis de citocinas séricas podem avaliar o estado imunológico de pacientes com tuberculose; também são biomarcadores úteis na predição e diagnóstico da tuberculose.

Abstract This study investigated the effect of Lactobacillus isolated from Chinese fermented food on antibiotic-induced intestinal microflora disorder during the early life of mice. The experimental strain Lactobacillus fermentum (LF) CQPC04 was isolated from naturally fermented pickles in Chongqing, China. Lactobacillus plantarum (LP) KFY02 was isolated from naturally fermented yogurt from Korla in the Xinjiang Uygur Autonomous Region. The results showed that LF-CQPC04 and LP-KFY02 alleviated the decrease in bacterial diversity caused by the antibiotics, maintained the abundance of beneficial bacteria, and reduced the abundance of harmful bacteria. Abstract Glutathione S-transferases (GSTs) are proteins synthesized in plants and responsible for their tolerance to environmental stresses. However, little information is available on the GST gene family of sweet potato, a globally important crop. The genetic evolution of GSTs in sweet potato remains unclear. The present study investigated the GST gene family in sweet potato by transcriptomic and comparative genomic analyses. A total of 51 GSTs were identified. Gene expression analysis showed differential expression patterns of the GSTs between two investigated varieties. Some GST expression levels were either up- or downregulated under oxidative, salinity and drought stresses. The results of the investigation provided new insights on the GST gene family in sweet potato, which may further the understanding of the roles of these genes in regulating abiotic stresses.

Abstract Objective To explore the molecular mechanism of neuropathologic damage induced by radiofrequency ablation at different temperatures. Methods This is basic research, and 36 SD rats were used to construct the neuropathological injury model. The rats were subjected to radiofrequency stimulation at different temperatures and were divided into 6 groups according to the temperature injury: 42°, 47°, 52°, 57°, 62°, and 67°C groups. Conduction time, conduction distance, and nerve conduction velocity were recorded after temperature injury. HE-staining was used to observe the histopathological morphology of the sciatic nerve. The expression of SCN9A, SCN3B, and NFASC protein in sciatic nerve tissue were detected by western blot. Results With the increase in temperature, nerve conduction velocity gradually decreased, and neurons were damaged when the temperature was 67°C. HE-staining showed that the degrees of degeneration of neurons in rats at 47°, 52°, 57°, 62°, and 67°C were gradually increased. The expression of SCN9A, SCN3B protein in 57°, 62°, 67°C groups were much higher than that of NC, 42°, 47°, 52°C groups. However, the expression of NFASC protein in 57°, 62°, 67°C groups was much lower than that of the NC, 42°, 47°, 52°C groups. Conclusion There was a positive correlation between temperature caused by the radiofrequency stimulation to neuropathological damage. The mechanism is closely related to the expression of SCN9A, SCN3B, and NFASC protein in nerve tissue caused by heat transfer injury.

ABSTRACT Introduction: This single-center study of propensity-matched data was performed to assess the effect of the no-touch saphenous vein (NTSV) harvesting technique on early- and long-term outcomes of patients after off-pump coronary artery bypass grafting (OPCABG) in China. Methods: A retrospective analysis of 767 patients who underwent OPCABG in the Beijing Anzhen Hospital (June 2017 to October 2021) was performed, and their data entered the conventional saphenous vein (CSV) harvesting technique group or the NTSV group. In-hospital and follow-up outcomes were evaluated by adjusting baseline characteristics using propensity score matching (1:1). Clinical outcomes and postoperative angiographic results were compared. Results: The saphenous vein graft patency rates at postoperative three months and one year for the NTSV group vs. CSV group were 99.6% vs. 96.2% (P<0.001) and 97.3% vs. 93.1% (P<0.001), respectively. The two matched groups received a significantly different cumulative incidence function of saphenous vein graft occlusion for the longer follow-up period in Kaplan-Meier curves (χ2=4.330, log-rank P=0.037). No difference in early- and long-term mortality or major adverse cardiac and cerebrovascular events (MACCE) were observed between the groups. The rate of MACCE was not statistically significant different between the groups, but there was a tendency favoring the no-touch technique (9.8% CSV vs. 4.8% NTSV; P=0.067). More patients in the NTSV group developed postoperative leg wound exudation (5.4% vs. 1.2%; P=0.032) and skin numbness (22.2% vs. 8.9%; P=0.001) than in the CSV group. Conclusion: The NTSV is an excellent conduit to be used in OPCABG. There remains a need to reduce leg wound complications.

SUMMARY: The Wnt pathway is essential for the initiation of lizard tail regeneration. The regenerated lizard tails exhibit obvious morphological differences compared to the original ones. The expression of Wnt1 and Wnt2b proteins in the regenerating tail of Scincella tsinlingensis was detected by immunohistochemistry and then comparatively analyzed for ultrastructural changes in the original and regenerated spinal cord. The ependymal layer of the original spinal cord was pseudostratified with multiciliated cells and primary monociliated cells, while the cells of the ependymal layer of the regenerated spinal cord were organized in a monolayer with a few biciliated cells. Immunolocalization indicated that Wnt1 and Wnt2b were mainly distributed in the dermis near the original tail stump, spinal cord, and clot-positive migratory cells during Stage I, 0-1 days post-amputation (dpa). Wnt1 and Wnt2b were predominantly detected in the epaxial and hypaxial musculature near the original tail stump, wound epithelium, and spinal cord in the original tail during Stage II, 1-7 dpa. Mesenchymal cells and wound epithelium showed immunostaining during Stage III and IV, 7-15 dpa. The ependymal tubes contained these signaling proteins during Stage V and VI, 20- 30 dpa. Labeling was mainly observed in nearby regenerative blood vessels, ependymal cells, epaxial and hypaxial musculature in the apical epithelial layer (AEC) after 45-160 dpa. These findings indicated that Wnt1 and Wnt2b proteins presented primarily in regenerating epidermis and nerve tissues were a critical signal for tail regeneration in S. tsinlingensis.

Abstract Objectives: This study aims to explore the effect of silencing Beclin-1 gene on autophagy and apoptosis of Benign Prostatic Hyperplasia (BPH) (BPH-1) cells under the condition of Androgen Deprivation (AD) and Autophagy Inhibition (AI). Methods: Control group (BPH-1 group), empty carrier group (sh-RNA-BPH-1 group) and Beclin-1 silenced group (sh-Beclin1-BPH-1 group) were set. The Beclin-1 gene silencing efficiency was detected by RT-PCR and Western blot. Autophagic flux was monitored by GFP-LC3 cleavage assay and cell apoptosis was analyzed by flow cytometry. The protein expression levels of LC3, Caspase-3, PARP-1, Bcl-2, and Bax were detected by Western blot. Results: The transfection of sh-Beclin-1 obviously down-regulated the expression of Beclin-1 at both mRNA and protein levels. Under the conditions of AD and AI, silencing of Beclin-1 restrained the autophagy of BPH-1 cells, as evidenced by a decreased number of autophagosomes and down-regulation of LC3-II protein (p < 0.001). The results of flow cytometry showed that the apoptotic rate of sh-Beclin-1 group was elevated significantly compared to the other two groups (p < 0.01). Western blot results showed that silencing of Beclin-1 promoted 89 kd fragmentation of PARP-1 (p < 0.001) and Caspase-3 activation (p < 0.01). Moreover, silencing of Beclin-1 resulted in declined Bcl-2 and augmented Bax protein expression in BPH-1 cells (p < 0.01), which ultimately led to a decreased Bcl-2/Bax ratio. Conclusions: The results indicated that the silencing of Beclin-1 gene hampered autophagy while activating apoptosis in BPH-1 cells. Thus, Beclin-1 may participate in an antagonistic relationship between autophagy and apoptosis in BPH.

RESUMO Introdução: Lesões no esporte geralmente ocorrem durante o ensino e treinamento esportivo, o que diretamente afeta o desempenho da função do corpo humano e o desenvolvimento do desempenho no esporte. Objetivo: Estudar a viabilidade de melhorar o biorritmo em lesões por treino esportivo. Métodos: 120 jovens atletas que praticam atletismo em escolas de atletismo tradicionais na província de Liaoning são os objetos desta pesquisa. Os dados efetivos da hora e dos tipos de lesões por esporte, além da data de nascimento, mês e data dos atletas lesionados durante o treino entre 2005 e 2006 foram coletados. Resultados: Os resultados mostram que, na relação entre o ritmo três do corpo humano e a lesão do atleta no esporte, a probabilidade da lesão no salto triplo é menor; quanto ao ritmo biológico em um ou mais de um período ou período crítico, a probabilidade de dano é de 95%, o que demonstra que a maioria das lesões de atletas no esporte ocorre numa etapa de período baixo e crítico. Conclusões: O manejo do treinamento esportivo deve se adaptar ao ritmo biológico original do corpo humano e o plano de treinamento de ciclo mensal correspondente deve ser feito de acordo com mudanças que ocorrem nos ciclos físicos dos atletas para evitar lesões durante o treino. ABSTRACT Introduction: Sports injury often occurs in sports teaching and training, which directly affects the performance of the human body function and the improvement of sports performance. Objective: To study the feasibility of improving the biorhythm in sports training injury. Methods: 120 young athletes who are engaged in track and field training in traditional track and field sports schools in Liaoning Province are taken as the research objects. The effective data of the time and types of sports injuries, and the birth date, month and date of the injured athletes during training from 2005 to 2006 were collected. Results: Results show that in the relationship between human body three rhythm and the athlete's sports injury, the probability of sports injury in triple height is smaller; regarding the biological rhythm in one or more than one period or critical period, the damage probability is 95%, showing that most athletes sports injuries occur at a low and critical period stage. Conclusions: The arrangement of sports training must be adapted to the original biological rhythm of the human body, and the corresponding monthly cycle training plan should be made according to changes that occur in athletes’ physical cycles to avoid injury during training. RESUMEN Introducción: Lesiones en el deporte generalmente ocurren durante la enseñanza y entrenamiento deportivo, algo que directamente afecta el rendimiento de la función del cuerpo humano y el desarrollo del rendimiento en el deporte. Objetivo: Estudiar la viabilidad de mejorar el biorritmo en lesiones por entrenamiento deportivo. Métodos: Os objetos de esta investigación son 120 jóvenes atletas que practican atletismo en escuelas de atletismo tradicionales en la provincia de Liaoning. Se recogieron los datos efectivos de la hora y los tipos de lesiones por deporte, además de la fecha de nacimiento, mes y fecha de los atletas lesionados durante el entrenamiento entre 2005 y 2006. Resultados: Los resultados muestran que, en la relación entre el ritmo tres del cuerpo humano y la lesión del atleta en el deporte, la probabilidad de lesión en el salto triple es menor. Cuanto al ritmo biológico en uno o más de un periodo o periodo crítico, la probabilidad de daño es de 95%, lo que demuestra que la mayoría de las lesiones de atletas en el deporte ocurre en una etapa de periodo bajo y crítico. Conclusión: El manejo del entrenamiento deportivo debe adaptarse al ritmo biológico original del cuerpo humano y el plan de entrenamiento de ciclo mensual correspondiente debe hacerse de acuerdo a los cambios que ocurren en los ciclos físicos de los atletas para evitar lesiones durante el entrenamiento. OBJECTIVES: To investigate the role of miR-139-5p and the TLR4/MyD88/NF-κB signaling pathway in acute lung injury in septic mice. METHOD: A total of 140 healthy male SPF C57BL/6 mice were divided into seven groups, i.e., Normal, Control, NC, miR-139-5p mimic, miR-139-5p inhibitor, TAK-242, and miR-139-5p inhibitor+TAK-242 groups. The levels of miR-139-5p, proteins related to the TLR4/MyD88/NF-κB signaling pathway (TLR4, MyD88, and p-NF-κB p50), and MPO, SOD, GSH, and MDA in lung tissue were measured. The lung tissue wet-to-dry mass ratio (W/D), arterial oxygen partial pressure (PaO2), and carbon dioxide partial pressure (PaCO2) were measured. RESULTS: A web-based bioinformatic tool predicted that MyD88 was a target of miR-139-5p, which was verified by a dual luciferase reporter assay. Compared with those in the Normal group, the levels of miR-139-5p, PaO2, SOD, and GSH were significantly lower, while those of TLR4, MyD88, p-NF-κB p50, W/D, PaCO2, IL-1β, TNF-α, IL-6, MPO, and MDA were higher in all other groups. Moreover, compared with their levels in the Control group, these indicators exhibited contrasting results in the miR-139-5p mimic and TAK-242 groups, but were similar in the miR-139-5p inhibitor group. In the miR-139-5p inhibitor+TAK-242 group, acute lung injury, aggravated by miR-139-5p inhibitor, was partially rescued by TAK-242. CONCLUSION: miR-139-5p inhibits the TLR4/MyD88/NF-κB signaling pathway to alleviate acute lung injury in septic mice.

Engeletin is a natural derivative of Smilax glabra rhizomilax that exhibits anti-inflammatory activity and suppresses lipid peroxidation. In the present study, we sought to elucidate the mechanistic basis for the neuroprotective and pro-angiogenic activity of engeltin in a human umbilical vein endothelial cells (HUVECs) oxygen-glucose deprivation and reoxygenation (OGD/R) model system and a middle cerebral artery occlusion (MCAO) rat model of cerebral ischemia and reperfusion injury. These analyses revealed that engeletin (10, 20, or 40 mg/kg) was able to reduce the infarct volume, increase cerebral blood flow, improve neurological function, and bolster the expression of vascular endothelial growth factor (VEGF), vasohibin-2 (Vash-2), angiopoietin-1 (Ang-1), phosphorylated human angiopoietin receptor tyrosine kinase 2 (p-Tie2), and platelet endothelial cell adhesion molecule-1 (PECAM-1/CD31) in MCAO rats. Similarly, engeletin (100, 200, or 400 nM) markedly enhanced the migration, tube formation, and VEGF expression of HUVECs in an OGD/R model system, while the VEGF receptor (R) inhibitor axitinib reversed the observed changes in HUVEC tube formation activity and Vash-2, VEGF, and CD31 expression. These data suggested that engeletin exhibited significant neuroprotective effects against cerebral ischemia and reperfusion injury in rats, and improved cerebrovascular angiogenesis by modulating the VEGF/vasohibin and Ang-1/Tie-2 pathways.
