Abstract In this work, an in silico study and evaluation of the cytotoxicity of 4-(4-chlorophenyl)thiazole compounds against mouse splenocytes and the chloroquine-sensitive Plasmodium falciparum 3D7 strain are reported. The in silico results showed that the compounds have important pharmacokinetic properties for compounds with potential drug candidates. Regarding cytotoxicity assays against splenocytes, the compounds have low cytotoxicity. In addition, they were able to promote activation of these cells by increasing nitric oxide production without promoting cell death. Finally, they were able to promote cell proliferation. Regarding the in vitro anti-P. falciparum activity assays, it was observed that the compounds were able to inhibit the parasite’s growth, presenting IC50 values ranging from 0.79 to greater than 10 µM. These results are promising when compared to chloroquine. Therefore, this study showed that 4-(4-chlorophenyl)thiazole compounds are promising candidates for antimalarials.

Cancer cell lines are widely used as in vitro models of tumorigenesis, facilitating fundamental discoveries in cancer biology and translational medicine. Currently, there are few options for glioblastoma (GBM) treatment and limited in vitro models with accurate genomic and transcriptomic characterization. Here, a detailed characterization of a new GBM cell line, namely AHOL1, was conducted in order to fully characterize its molecular composition based on its karyotype, copy number alteration (CNA), and transcriptome profiling, followed by the validation of key elements associated with GBM tumorigenesis. Large numbers of CNAs and differentially expressed genes (DEGs) were identified. CNAs were distributed throughout the genome, including gains at Xq11.1-q28, Xp22.33-p11.1, Xq21.1-q21.33, 4p15.1-p14, 8q23.2-q23.3 and losses at Yq11.21-q12, Yp11.31-p11.2, and 15q11.1-q11.2 positions. Nine druggable genes were identified, including HCRTR2, ETV1, PTPRD, PRKX, STS, RPS6KA6, ZFY, USP9Y, and KDM5D. By integrating DEGs and CNAs, we identified 57 overlapping genes enriched in fourteen pathways. Altered expression of several cancer-related candidates found in the DEGs-CNA dataset was confirmed by RT-qPCR. Taken together, this first comprehensive genomic and transcriptomic landscape of AHOL1 provides unique resources for further studies and identifies several druggable targets that may be useful for therapeutics and biologic and molecular investigation of GBM.

Abstract The use of creep feeding for preweaning piglets is important to improve the performance of the piglets. The objective of this experiment was evaluate the effect of using or altering the position of piglet’s creep feeder during lactation on piglet’s performance and on behavior of piglets and sows kept in a hot climate environment. Forty-five sows and their litters at 10 days of lactation were randomly distributed into three treatments: front feeder (FF) - near the side of the sow’s head; back feeder (BF) - near the side of the rump of the sow; and no feeder (NF). All piglets were weighed individually to evaluate the average weight, weight gain and coefficient of variation of the weight. Behavior assessments of the piglets and sows were recorded in 3 period. At 15 and 21 d, piglets of the FF treatment were heavier (P ≤ 0.0001) than piglets of the other treatments. At 10-21d piglets of FF treatment had 76.2% less belly nosing behavior than the NF piglets (P=0.015). The treatments had no impact on behavior of the sows. The creep feeders positioned in the front of the farrowing crate increased piglet growth rate and decreased frequency of belly nosing behavior.

Nonalcoholic fatty liver disease (NAFLD), characterized by hepatosteatosis and steatohepatitis, is intrinsically related to obesity. Our previous study reported on the anti-obese activity of α,β-amyrin (AMY), a pentacyclic triterpene isolated from Protium heptaphyllum. This study investigated its ability to prevent fatty liver and the underlying mechanism using the mouse model of NAFLD. NAFLD was induced in male Swiss mice fed a high fat diet (HFD) for 15 weeks. The controls were fed a normal chow diet (ND). The mice were simultaneously treated with AMY at 10 and 20 mg/kg or fenofibrate at 50 mg/kg. Lipid levels along with metabolic and inflammatory parameters were assessed in liver and serum. The liver sections were histologically examined using H&E staining. RT-qPCR and western blotting assays were performed to analyze signaling mechanisms. Mice fed HFD developed severe hepatic steatosis with elevated triglycerides and lipid droplets compared with ND controls. This was associated with a decrease in AMP-activated protein kinase (AMPK) activity, an increase of mechanistic target of rapamycin complex 1 (mTORC1) signaling, and enhanced sterol regulatory element binding protein 1 (SREBP1) expression, which have roles in lipogenesis, inhibition of lipolysis, and inflammatory response. AMY treatment reversed these signaling activities and decreased the severity of hepatic steatosis and inflammatory response, evidenced by serum and liver parameters as well as histological findings. AMY-induced reduction in hepatic steatosis seemed to involve AMPK-mTORC1-SREBP1 signaling pathways, which supported its beneficial role in the prevention and treatment of NAFLD.

Perioperative central venous oxygen saturation and its correlation with mortality during cardiac surgery: an observational prospective study

Resumo Justificativa: A cirurgia cardíaca pode produzir déficit persistente na razão entre oferta de oxigênio (DO2) e consumo de oxigênio (VO2). A Saturação venosa central de Oxigênio (SvcO2) é uma medida acessível e indireta da razão DO2/VO2. Objetivo: Monitorar a SvcO2 perioperatória e avaliar sua correlação com a mortalidade em cirurgia cardíaca. Método: Este estudo observacional prospectivo avaliou 273 pacientes submetidos a cirurgia cardíaca. Coletamos amostras de sangue para medir a SvcO2 em três momentos: T0 (após indução anestésica), T1 (final da cirurgia) e T2 (24 horas após a cirurgia). Os pacientes foram divididos em dois grupos (sobreviventes e não sobreviventes). Os seguintes desfechos foram analisados: mortalidade intra-hospitalar, tempo de permanência na Unidade de Terapia Intensiva (UTI) e de internação hospitalar, e variação na SvcO2. Resultados: Dos 273 pacientes, 251 (92%) sobreviveram e 22 (8%) não. Houve queda significante da SvcO2 perioperatória nos sobreviventes (T0 = 78% ± 8,1%, T1 = 75,4% ± 7,5% e T2 = 68,5% ± 9%; p< 0,001) e nos não sobreviventes (T0 = 74,4% ± 8,7%, T1 = 75,4% ± 7,7% e T2 = 66,7% ± 13,1%; p< 0,001). No T0, a porcentagem de pacientes com SvcO2< 70% foi maior no grupo não sobrevivente (31,8% vs. 13,1%; p = 0,046) e a regressão logística múltipla mostrou que a SvcO2 é um fator de risco independente associado ao óbito, OR = 2,94 (95% IC 1,10 − 7,89) (p = 0,032). O tempo de permanência na UTI e de hospitalização foi de 3,6 ± 3,1 e 7,4 ± 6,0 dias, respectivamente, e não foi significantemente associado à SvcO2. Conclusões: Valores precoces de SvcO2 intraoperatória < 70% indicaram maior risco de óbito em pacientes submetidos à cirurgia cardíaca. Observamos redução perioperatória da SvcO2, com altos níveis no intraoperatório e mais baixos no pós-operatório.
Abstract Background: Cardiac surgery can produce persistent deficit in the ratio of Oxygen Delivery (DO2) to Oxygen Consumption (VO2). Central venous oxygen Saturation (ScvO2) is an accessible and indirect measure of DO2/VO2 ratio. Objective: To monitor perioperative ScvO2 and assess its correlation with mortality during cardiac surgery. Methods: This prospective observational study evaluated 273 patients undergoing cardiac surgery. Blood gas samples were collected to measure ScvO2 at three time points: T0 (after anesthetic induction), T1 (end of surgery), and T2 (24 hours after surgery). The patients were divided into two groups (survivors and nonsurvivors). The following outcomes were analyzed: intrahospital mortality, length of Intensive Care Unit (ICU) and hospital stay (LOS), and variation in ScvO2. Results: Of the 273 patients, 251 (92%) survived and 22 (8%) did not. There was a significant perioperative reduction of ScvO2 in both survivors (T0 = 78% ± 8.1%, T1 = 75.4% ± 7.5%, and T2 = 68.5% ± 9%; p< 0.001) and nonsurvivors (T0 = 74.4% ± 8.7%, T1 = 75.4% ± 7.7%, and T2 = 66.7% ± 13.1%; p < 0.001). At T0, the percentage of patients with ScvO2< 70% was greater in the nonsurvivor group (31.8% vs. 13.1%; p= 0.046) and the multiple logistic regression showed that ScvO2 is an independent risk factor associated with death, OR = 2.94 (95% CI 1.10−7.89) (p= 0.032). The length of ICU and LOS were 3.6 ± 3.1 and 7.4 ± 6.0 days respectively and was not significantly associated with ScvO2. Conclusions: Early intraoperative ScvO2 < 70% indicated a higher risk of death. A perioperative reduction of ScvO2 was observed in patients undergoing cardiac surgery, with high intraoperative and lower postoperative levels.

RESUMO Descreve-se a viabilidade da técnica cirúrgica de transposição uretral pré-púbica em cadáveres de cães machos, adultos, advindos de um hospital veterinário, a fim de se avaliar a manutenção da luz uretral a partir de um desvio uretral pré-púbico, descrevendo-se detalhadamente a abordagem e as estruturas anatômicas envolvidas. A técnica consistiu inicialmente da orquiectomia, seguida de celiotomia retroumbilical, tração vesical cranial, secção transversa da uretra membranosa a 1cm caudal à próstata, espatulação e reparo da borda livre. A seguir, divulsão e secção do pênis 1,5cm caudal ao osso peniano, espatulação da borda uretral peniana e transposição desta em direção à cavidade abdominal, fazendo-se anastomose por meio de sutura interrompida simples à uretra membranosa. Foi mensurado o comprimento uretral desde o meato externo até o início da uretra prostática em dois momentos: pré e pós-transposição. Ao final, avaliou-se, por meio de uretrografia retrógrada de contraste positivo, o diâmetro e o selamento na anastomose uretral e o fluxo do contraste até a bexiga. A transposição uretral pré-púbica é uma técnica cirúrgica exequível, anatomicamente compatível com a espécie canina e capaz de manter o diâmetro uretral e o fluxo do contraste de forma satisfatória, com manutenção estética externa idêntica a um cão macho orquiectomizado.
ABSTRACT We describe the viability of the surgical technique of pre-pubic urethral transposition in cadavers of adult male dogs, coming from a veterinary hospital to evaluate the maintenance of urethral light from a prepubic urethral deviation, describing in detail the approach and anatomical structures involved. The technique consisted initially of orchiectomy, followed by retroumbilical celiotomy, cranial vesical traction, transverse section of the membranous urethra at 1cm caudal to prostate, spatulation and free edge repair. Next, the penis divulsion and section 1.5cm caudal to the penile bone, spreading the penile urethral border and transposing it towards the abdominal cavity, making an anastomosis of simple interrupted suture to the membranous urethra. The dogs had the urethral length measured from the external meatus to the beginning of the prostatic urethra at two moments: pre-transposition and post-transposition. Postoperative retrograde urethrography was used to evaluate the diameter and sealing of the urethral anastomosis and contrast flow to the bladder. In conclusion, the prepubic urethral transposition is a feasible surgical technique, anatomically compatible with the canine species and able to maintain the urethral diameter and contrast flow satisfactorily, with an external esthetic maintenance identical to an orchiectomized male dog.

RESUMO Objetivou-se avaliar os efeitos do zinco ou do propilenoglicol sobre o desempenho e nas características da carcaça e da carne de ovinos sob pastejo em pastagem nativa da Caatinga. Utilizaram-se 24 ovinos, mestiços Santa Inês, machos, não castrados, peso inicial de 19,3±2,52kg, com quatro meses de idade, oito repetições por tratamento, distribuídos em um delineamento inteiramente ao acaso. Os tratamentos foram: sem aditivo, controle (CT), adição de sulfato de zinco (ZnSO4.7H2O) (Zn) para fornecimento de 300mg de Zn dia-1 via sal mineral, e propilenoglicol (PG), 2,5mL kgPC0,75-1 animal-1 dia-1 misturado no concentrado. Os animais foram suplementados com concentrado constituído de milho (54,0%), farelo de soja (45,1%) e calcário (0,911%) em 0,7% do PC médio de cada tratamento. O ensaio foi conduzido durante quatro meses (março a junho/2014). Avaliaram-se o desempenho e, após o abate dos animais, as características de carcaça e os componentes não carcaça, os parâmetros físico-químicos e organolépticos na carne. O Zn e o propilenoglicol não afetaram o desempenho e os parâmetros da carcaça em termos absoluto e relativo, bem como os pesos dos cortes e seus rendimentos (P>0,05). Não houve efeito dos aditivos nas características organolépticas e nos aspectos físico-químicos da carne (P>0,05), exceto cinzas (P<0,05). O zinco e o propilenoglicol não melhoram o desempenho e as características da carcaça de animais terminados em pastagem nativa da Caatinga. O baixo suprimento energético devido à má qualidade do pasto entre os meses de abril a junho contribui para a ineficiência no aproveitamento dos constituintes proteicos, implicando menor consumo e consequente diminuição do desempenho.
ABSTRACT The aim was to evaluate the effects of zinc sulfate or propylene glycol on performance and carcass and meat characteristics from sheep under grazing in native Caatinga (Brazilian savannah) pasture during the rainy season. Twenty-four intact Santa Ines crossbreed male lambs, with an initial weight of 19.3±2.52kg, and 4-mo old, were distributed in a completely randomized design with eight replications per treatment. The treatments were composed of additives, as follows: no additive - control (CT); addition of zinc sulfate (ZnSO4.7H2O) (Zn) to supply a total of 300mg Zn day−1 on mineral salt; and addition of propylene glycol (PG), 2.5mL kg LW0.75−1 animal−1 day−1 mixed directly in the concentrate. Afterwards, they were supplemented with concentrate at 0.7% of the average LW of each batch according to the treatments. The concentrate was based on corn (54.0%), soybean meal (45.1%), and limestone (0.911%). The trials were carried out during the rainy season (March to June 2014), and evaluated the performance, and before of slaughter, carcass characteristics and non-carcass components were evaluated and the physical-chemical aspects and the organoleptic attributes on the meat. The additives did not affect the performance, parameters of carcass, in terms absolute and relative, carcass morphometric measurements, weight of the commercial cuts and yields (P>0.05). The additives did not affect the organoleptic characteristics and physical-chemical aspects of the meat (P>0.05), except, ash (P<0.05). The zinc sulfate and propylene glycol did not improve on performance and in carcass characteristics in lambs finished on a native Caatinga (Brazilian savannah) pasture. The low supply energetic due to poor quality of pasture between the months of April to June, which contributed to inefficiency in the utilization of proteics constituents, implying in lower intake, and decrease in performance.

Effects of aerobic and resistance training of long duration on pro- and anti-inflammatory cytokines in rats

Abstract Objective: To determine possible changes in serum concentrations of pro- and anti-inflammatory cytokines of eutrophic rats subjected to aerobic or resistance physical training. Methods: This study examined serum concentrations of TNF-α, IFN-γ, IL-6, IL-10 and IL-1-β in rats that performed aerobic or resistance training for 16 weeks. Thirty-five Wistar rats (male adult) were divided into three groups: Control Group (CG), Aerobic Group (AG) and Resistance Group (RG). Rats were sacrificed 48 h after the final training session. Serum concentrations of cytokines were analysed by ELISA. Results: TNF-α levels were higher in the RG, followed by the AG and CG groups (p < 0.001). IFN-γ and IL-10 levels were not significantly different between groups (p = 0.097 and p = 0.17, respectively). The levels of IL6 and IL1-β were higher in AG compared to RG and CG (p = 0.0004 and p = 0.003, respectively). In general, our results indicate a higher pro-inflammatory profile in AG and probably RG animals. Conclusion: Further studies are required in order to better clarify the effects of aerobic and resistance exercise training on pro- and anti-inflammatory cytokines. Additionally, future studies should address the metabolic and molecular pathways involved in these responses.
Resumo Objetivo: Determinar as possíveis alterações nas concentrações séricas de citocinas pró e anti-inflamatórias de ratos eutróficos submetidos a treinamento físico aeróbio ou resistência. Método: Este estudo examinou a concentração sérica de TNF-α, IFN-γ, IL-6, IL-10 e IL-1-β em ratos que realizaram treinamento aeróbio e de resistência durante 16 semanas. Trinta e cinco ratos Wistar (macho adulto) foram divididos em 3 grupos: grupo controle (GC), grupo aeróbico (GA) e grupo de resistência (GR). Os ratos foram sacrificados 48 horas após a sessão de treino final. As concentrações séricas de citocinas foram analisadas por ELISA. Resultados: Os níveis de TNF-a foram maiores no GR, seguindo-se os grupos GA e GC (p < 0.001). IFN-γ e os níveis de IL-10 não foram significativamente diferentes entre os grupos (p = 0.097 e p = 0.17, respectivamente). Os níveis de IL-6 e IL-1-β foram maiores em GA, quando comparados com GC e GR (p = 0.0004 e p = 0.003, respectivamente). Em geral, os nossos resultados indicam um perfil pró-inflamatório maior nos animais do GA e, provavelmente, no GR. Conclusão: Mais estudos são necessários para melhor esclarecer os efeitos do treinamento físico aeróbico e resistência sobre as citocinas pró e anti-inflamatórias. Além disso, estudos futuros devem abordar as vias metabólicas e moleculares envolvidas nestas respostas.
Resumen Objetivo: Determinar posibles cambios en las concentraciones séricas de citoquinas pro y antiinflamatorias de ratas eutróficas sometidas a entrenamiento físico aérobico y de resistencia. Método: Se examinaron las concentraciones séricas de TNF-α, IFN-γ, IL-6, IL-10 e IL-1-β en ratas sometidas a entrenamiento aeróbico o de resistencia de 16 semanas de duración. Treinta y cinco ratas Wistar (macho adulto) fueron divididas en 3 grupos: Grupo Control (GC), Grupo Aeróbico (GA) y Grupo Resistencia (GR). Las ratas se sacrificaron 48 horas después de la sesión de entrenamiento final. Las concentraciones séricas de las citoquinas se analizaron por ELISA. Resultados: Los niveles de TNF-α fueron mayores en el GR, seguido por los grupos de GA y GC (p < 0.001). Los niveles de IFN-γ e IL-10 no fueron significativamente diferentes entre los grupos (p = 0.097 y p = 0.17, respectivamente). Los niveles de IL-6 y IL-1-β fueron mayores en GA comparado con GR y GC (p = 0.0004 y p = 0.003, respectivamente). En general, nuestros resultados indican mayor perfil antiinflamatorio en GA y probablemente en GR. Conclusión: Se necesitan estudios adicionales para aclarar mejor los efectos de un entrenamiento aeróbico o de resistencia en las citoquinas pro y antiinflamatorias. Además, los estudios futuros deben abordar las vías metabólicas y moleculares involucradas en estas respuestas.

Previous studies have reported on the glucose and lipid-lowering effects of ferulic acid (FA) but its anti-obesity potential has not yet been firmly established. This study investigated the possible anti-obesitogenic effects of FA in mice fed a high-fat diet (HFD) for 15 weeks. To assess the antiobesity potential of FA, 32 male Swiss mice, weighing 20–25 g (n=6–8 per group) were fed a normal diet (ND) or HFD, treated orally or not with either FA (10 mg/kg) or sibutramine (10 mg/kg) for 15 weeks and at the end of this period, the body weights of animals, visceral fat accumulation, plasma levels of glucose and insulin hormone, amylase and lipase activities, the satiety hormones ghrelin and leptin, and tumor necrosis factor-α (TNF-α) and monocyte chemoattractant protein-1 (MCH-1) were analyzed. Results revealed that FA could effectively suppress the HFD-associated increase in visceral fat accumulation, adipocyte size and body weight gain, similar to sibutramine, the positive control. FA also significantly (P<0.05) decreased the HFD-induced elevations in serum lipid profiles, amylase and lipase activities, and the levels of blood glucose and insulin hormone. The markedly elevated leptin and decreased ghrelin levels seen in HFD-fed control mice were significantly (P<0.05) reversed by FA treatment, almost reaching the values seen in ND-fed mice. Furthermore, FA demonstrated significant (P<0.05) inhibition of serum levels of inflammatory mediators TNF-α, and MCH-1. These results suggest that FA could be beneficial in lowering the risk of HFD-induced obesity via modulation of enzymatic, hormonal and inflammatory responses.

RESUMO Avaliou-se a macroscopia, a microestrutura, a resposta clínica e histopatológica de implantes de acrilonitrila butadieno estireno (ABS) e poliamida em coelhos submetidos à evisceração. Para o estudo experimental, os implantes foram elaborados por meio de prototipagem rápida, com 12mm de diâmetro, pesados e tiveram suas superfícies avaliadas macroscopicamente e por microscopia eletrônica de varredura. Adicionalmente, foi realizado ensaio de compressão para determinar a força necessária para fraturar os implantes. Após a evisceração do olho esquerdo, nove coelhos da raça Nova Zelândia receberam implantes de poliamida e nove outros receberam implantes de ABS. Os animais foram avaliados diariamente nos primeiros 15 dias após a cirurgia e a cada sete dias até o fim do período experimental (90 dias). Avaliação histopatológica foi realizada aos 15, 45 e 90 dias após a cirurgia. Os implantes de ABS pesaram 0,44g, e os de poliamida 0,61g. A microscopia eletrônica de varredura demonstrou que os implantes de ABS eram formados por microporos equidistantes, enquanto os de poliamida apresentavam microporos de vários tamanhos. A força necessária para fraturar os implantes de ABS foi de 14.39±0.60 Mpa, enquanto para os de poliamida foi de 16.80±1.05Mpa. Quinze dias após a cirurgia, foi observada infiltração fibrovascular centrípeta. Os implantes podem ser utilizados para correção de cavidades anoftálmicas por se mostrarem inertes, biocompatíveis e permitirem a infiltração tecidual.
ABSTRACT The purpose of this study was to evaluate acrylonitrile butadiene styrene (ABS) and polyamide implants in rabbits submitted to evisceration at the macroscopic and microstructure level and to assess clinical response and histopathological changes as well. For the experimental study implants of 12mm diameter were prepared by rapid prototyping, weighed and the outer and inner surfaces evaluated macroscopically and by electron microscopy. In addition, a compression test was performed and ultrastructural damage was then determined. After evisceration of the left eyeball, nine New Zealand rabbits received ABS implants and nine others received polyamide implants. The animals were assessed daily for 15 days after surgery and every seven days until the end of the study (90 days). Histopathological evaluation was performed at 15, 45 and 90 days after surgery. The ABS implants weighed approximately 0.44g, while the polyamide ones weighed 0.61g. Scanning electron microscopy demonstrated that the ABS implants had regular-sized, equidistant micropores, while the polyamide ones showed micropores of various sizes. The force required to fracture the ABS implant was 14.39 ±0.60 Mpa, while for the polyamide one, it was 16.80 ±1.05 Mpa. Fifteen days after surgery, we observed centripetal tissue infiltration and scarce inflammatory infiltrate. Implants may be used in the filling of anophthalmic cavities, because they are inert, biocompatible and allow tissue integration.

RESUMO O objetivo deste estudo foi determinar os valores energéticos de rações expandidas, obtidas em diferentes temperaturas de expansão para frangos de corte em diferentes idades. As rações foram expandidas nas temperaturas: 80; 100; 120; 140ºC. Dois ensaios biológicos foram conduzidos utilizando-se o método tradicional de coleta total de excretas para determinar a energia metabolizável aparente corrigida (EMAn). Os ensaios metabólicos foram conduzidos com pintos machos Cobb, de 11 a 19 dias (fase inicial) e de 27 a 35 dias de idade (fase de crescimento), utilizando-se as mesmas aves do primeiro ensaio e, assim, preservando os tratamentos a que foram submetidas. Os valores da EMAn das rações da fase inicial foram: 2937; 2900; 2806 e 2751kcal/kg, e da fase de crescimento: 3045; 3031; 3115 e 2977kcal/kg, respectivamente. Os resultados mostraram uma redução linear dos níveis de EMn com o aumento da temperatura de expansão na idade de 11 a 19 dias. As perdas relativas entre as rações expandidas a 80 e 100ºC foram mínimas, enquanto nas temperaturas de 120 e 140ºC foram significativamente superiores. No ensaio de metabolismo para a fase de crescimento, verificou-se que as perdas relativas entre as rações expandidas a 80 e 100ºC foram pequenas (-14kcal). Para a ração expandida a 120ºC, o valor energético foi superior (84 kcal), enquanto para 140ºC foi significativamente inferior (-138kcal). Esses resultados mostram que, na fase de crescimento, os frangos de corte maximizaram o aproveitamento energético das rações na temperatura de expansão de 120ºC e que, em temperaturas acima desse nível, ocorrem altas perdas da EMAn das rações, que podem comprometer o consumo, a deposição de proteína e a conversão alimentar e, consequentemente, trazer grandes prejuízos econômicos pelo menos à idade de abate. As temperaturas de expansão de rações entre 80 e 100ºC apresentaram os melhores valores de EMAn para frangos com idade entre 15 e 10 dias, enquanto para idade de 31 a 35 dias foi de 120ºC.
ABSTRACT The aim of this study was to determine the energetic value of feed in different expansion temperatures for broilers of different ages. The feedexpanded in the following temperatures: 80; 100, 120 and 140ºC. Two biological assays were run to establish apparent metabolizable energy corrected by nitrogen balance (AMEN) using the traditional total excreta collection method. In the first assay Cobb chicks were used from 11 to 19 days of age (initial period), and 29 to 37 days of age (growth period), as well as the same treatments of the first assay. The AMEN values for the initial period were respectively: 2937, 2900, 2806 and 2751 kcal/kg; and broilers in growing period were respectively: 3045, 3031, 3115 and 2977 kcal/kg. The results showed a linear decrease of the levels EMAN with an increase of the temperature of expansion from 11 to 19 days old. The loss relation between feed expanded at 80 and 100ºC were minimal, while in temperatures between 120 and 140ºC they were significantly higher. In the metabolic assay for period growth, we observed that the relation of energy values between feed expanded at 80 and 100ºC were smaller (14 kcal), while for the expanded in 120ºC they were superior (84 kcal), and at 140ºC were inferior (138 kcal). These results suggest greater energy utilization efficiency in period growth at a temperature of 120 ºC, while in temperatures above of this level there was greater loss of EMAN in feeds, which can prejudice feed intake, protein deposition and feed conversion, and consequently damage economics by increasing slaughter age. The expansion temperatures in feed between 80 and 100ºC showed the best EMAN values for broilers with 15 - 19 days of age, while for 31 - 35 days old it was 120 ºC.

Contractile profile of esophageal and gastric fundus strips in experimental doxorubicin-induced esophageal atresia

Esophageal atresia (EA) is characterized by esophageal and gastric motility changes secondary to developmental and postsurgical damage. This study evaluated the in vitro contractile profile of the distal esophagus and gastric fundus in an experimental model of EA induced by doxorubicin (DOXO). Wistar pregnant rats received DOXO 2.2 mg/kg on the 8th and 9th gestational days. On day 21.5, fetuses were collected, sacrificed, and divided into groups: control, DOXO without EA (DOXO-EA), and DOXO with EA (DOXO+EA). Strips from the distal esophagus and gastric fundus were mounted on a wire myograph and isolated organ-bath system, respectively, and subjected to increasing concentrations of carbamylcholine chloride (carbachol, CCh). The isolated esophagus was also stimulated with increasing concentrations of KCl. In esophagus, the concentration-effect curves were reduced in response to CCh in the DOXO+EA and DOXO-EA groups compared to the control group (P<0.05). The maximum effect values (Emax) for DOXO+EA and DOXO-EA were significantly lower than control (P<0.05), but the half-maximal effective concentration (EC50) values were not significantly different when the three groups were compared (P>0.05). In response to KCl, the distal esophagus samples in the three groups were not statistically different with regard to Emax or EC50 values (P>0.05). No significant difference was noted for EC50 or Emax values in fundic strips stimulated with CCh (P>0.05). In conclusion, exposure of dams to DOXO during gestation inhibited the contractile behavior of esophageal strips from offspring in response to CCh but not KCl, regardless of EA induction. The gastric fundus of DOXO-exposed offspring did not have altered contractile responsiveness to cholinergic stimulation.

Avaliaram-se a macroscopia e a microestrutura de esferas compostas por cimento ósseo de alfa-fosfato tricálcico de dupla pega munido de canais de interconexão (alfa-TCP-i) e a integração do implante ao tecido orbital de coelhos através de análise macroscópica e histopatológica. A superfície interna e externa do alfa-TCP-i foi avaliada macroscopicamente e por microscopia eletrônica. Após enucleação do bulbo ocular esquerdo, 12 coelhos da raça Nova Zelândia receberam implantes do alfa-TCP-i com 12mm de diâmetro. A avaliação clínica foi realizada diariamente nos primeiros 15 dias e depois a cada quinze dias até o fim do período de estudo. Para análise morfológica, seguiu-se a exenteração de três animais por período experimental (15, 45, 90 e 180 dias). As superfícies externa e interna do implante mostraram-se compactas, sólidas e lisas, com seis canais que se interconectavam centralmente. A microarquitetura caracterizou-se pela formação de colunas de cristais hexagonais. Sinais de infecção, exposição, deiscência de sutura ou extrusão não foram notados em nenhum dos animais durante o período de estudo. A análise morfológica revelou a presença de fina cápsula ao redor do implante de onde surgiram projeções fibrovasculares que o penetraram através dos canais de interconexão. Nos primeiros dias após a inserção das esferas, notou-se intensa reação inflamatória que se mostrou ausente aos 180 dias. O implante do cimento de alfa-fosfato tricálcico foi bem tolerado em coelhos, mostrou-se relativamente inerte e permitiu a infiltração de tecido fibrovascular através dos canais de interconexão.
The purpose of this study was to evaluate the macroscopy and microstructure of a double setting alpha-tricalcium phosphate bone cement sphere provided with interconnection channels (alpha-TCP-i), as well as the integration of the implant with the rabbits' orbital tissue, through macroscopic analysis and histopathology. The external and internal surfaces of the alpha-TCP-i were evaluated macroscopically and by electron microscopy. Twelve New Zealand rabbits received 12mm implants of alpha-TCP-i following enucleation of the left eye. The clinical assessment was undertaken daily during the first 15 days, followed by fortnightly assessment until the end of the study period. For the morphological analysis, exenteration was performed in 3 animals per experimental period (15, 45, 90 and 180 days). The external and internal surfaces of the implant appeared solid, smooth and compact, with six channels which interconnected centrally. The micro-architecture was characterized by the formation of columns of hexagonal crystals. No signs of infection, exposure, dehiscence of sutures or extrusion of the implant were noted in any of the animals during the entire period of the study. The morphological evaluation demonstrated the presence of a thin capsule around the implant, from whence appeared fibro-vascular projections, which penetrated it through the interconnecting channels. In the first days after the insertion of the implant, an intense inflammatory reaction was noted. At 180 days, however, there were no signs of inflammation. The alpha-tricalcium phosphate cement implant was well tolerated in this rabbit model and appeared to be relatively inert with some fibrovascular ingrowth through the large channels.

Resumo Um levantamento atualizado das plantas com sementes e análises relevantes acerca desta biodiversidade são apresentados. Este trabalho se iniciou em 2010 com a publicação do Catálogo de Plantas e Fungos e, desde então vem sendo atualizado por mais de 430 especialistas trabalhando online. O Brasil abriga atualmente 32.086 espécies nativas de Angiospermas e 23 espécies nativas de Gimnospermas e estes novos dados mostram um aumento de 3% da riqueza em relação a 2010. A Amazônia é o Domínio Fitogeográfico com o maior número de espécies de Gimnospermas, enquanto que a Floresta Atlântica possui a maior riqueza de Angiospermas. Houve um crescimento considerável no número de espécies e nas taxas de endemismo para a maioria dos Domínios (Caatinga, Cerrado, Floresta Atlântica, Pampa e Pantanal), com exceção da Amazônia que apresentou uma diminuição de 2,5% de endemicidade. Entretanto, a maior parte das plantas com sementes que ocorrem no Brasil (57,4%) é endêmica deste território. A proporção de formas de vida varia de acordo com os diferentes Domínios: árvores são mais expressivas na Amazônia e Floresta Atlântica do que nos outros biomas, ervas são dominantes no Pampa e as lianas apresentam riqueza expressiva na Amazônia, Floresta Atlântica e Pantanal. Este trabalho não só quantifica a biodiversidade brasileira, mas também indica as lacunas de conhecimento e o desafio a ser enfrentado para a conservação desta flora.
Abstract An updated inventory of Brazilian seed plants is presented and offers important insights into the country's biodiversity. This work started in 2010, with the publication of the Plants and Fungi Catalogue, and has been updated since by more than 430 specialists working online. Brazil is home to 32,086 native Angiosperms and 23 native Gymnosperms, showing an increase of 3% in its species richness in relation to 2010. The Amazon Rainforest is the richest Brazilian biome for Gymnosperms, while the Atlantic Rainforest is the richest one for Angiosperms. There was a considerable increment in the number of species and endemism rates for biomes, except for the Amazon that showed a decrease of 2.5% of recorded endemics. However, well over half of Brazillian seed plant species (57.4%) is endemic to this territory. The proportion of life-forms varies among different biomes: trees are more expressive in the Amazon and Atlantic Rainforest biomes while herbs predominate in the Pampa, and lianas are more expressive in the Amazon, Atlantic Rainforest, and Pantanal. This compilation serves not only to quantify Brazilian biodiversity, but also to highlight areas where there information is lacking and to provide a framework for the challenge faced in conserving Brazil's unique and diverse flora.

A espécie Celtis iguanaea (Jacq.) Sargent é popularmente conhecida como esporão de galo ou grão de galo. As folhas são indicadas pelo uso popular para o tratamento de dores no corpo e no peito, para reumatismo, asma, cólicas, má digestão e como diurético; as raízes são utilizadas para infecções urinárias e as cascas para a febre. O presente trabalho objetivou contribuir para o estudo fitoquímico e atividade fitotóxica com enfoque alelopático das cascas de Celtis iguanaea. O extrato etanólico foi submetido à partição com os solventes hexano, clorofórmio e acetato de etila. As substâncias friedelina e epifriedelinol (triterpenos) foram isoladas da fração hexano e identificadas por meio de métodos espectroscópicos de RMN de ¹H e 13C. O extrato bruto na concentração de 0,1 mg mL-1 causou inibição acentuada do hipocótilo em 34,97% e estimulou o crescimento da radícula em 29,64% de plântulas de Lactuca sativa. No ensaio de toxicidade frente à Artemia salina o extrato bruto e frações apresentaram uma CL50 superior a 1000 μg mL-1, indicando que o mesmo não possui efeito tóxico.
The species Celtis iguanaea (Jacq.) Sargent is popularly known as "esporão de galo" or "grão de galo". Its leaves are recommended by the popular use for the treatment of body and chest aches, as well as for rheumatism, asthma, cramps, indigestion and as diuretic; its roots are used for urinary infections and its bark for fever. This study aimed to contribute to the phytochemical investigation of the toxic activity focused on the allelopathic effect of the bark of Celtis iguanaea. The ethanol extract was subjected to solvent partition with hexane, chloroform and ethyl acetate. The substances friedelin and epifriedelinol (triterpenes) were isolated from the hexane fraction and identified by spectroscopic methods ¹H and 13C NMR. The crude extract at a concentration of 0.1 mg mL-1 caused marked inhibition of hypocotyl in 34.97% and stimulated radicle growth in 29.64% seedlings of Lactuca sativa. In the toxicity test against Artemia salina the crude extract and fractions showed an LC50 higher than 1000 μg mL-1, indicating that it has no toxic effect.
