Abstract Introduction The isolation of captured peripheral blood mononuclear cells (PBMNCs) from leukoreduction filters (LRFs) can be of great importance in terms of bringing the lost cells back into use. Objective The aim of this study was to evaluate various methods based on their potential to recover the peripheral blood cells from LRFs with a focus on mononuclear cells (MNCs). Method For cell isolation from LRFs, three distinct methods (back-flushing, direct and vacuum pump) were compared through the calculation of the yield of isolated MNCs. The viability of extracted cells was determined by the flow cytometry technique. Moreover, the recovered MNCs were characterized regarding the presence of blood stem cell purification. The cell culture, microscopic observation, and immunophenotyping were employed to characterize the blood stem cells (hematopoietic, mesenchymal and progenitor endothelial stem cells). Results The yield of isolation obtained in the back-flushing, direct and vacuum pump methods were 17.7 ± 1.28, 17.3 ± 0.96 and 21.2 ± 0.90 percent, respectively. Although the highest potential for total blood cell recovery belonged to the vacuum pump method, the lowest cell viability (85.73 ± 4.84%) was observed in this method. However, the isolation process of the back-flushing and direct methods had less effect on cell viability. The characterization of the isolated MNCs displayed that the dominant positive phenotype was for CD34/CD45, indicating hematopoietic stem cells. In addition, the endothelial stem/progenitor cells were significantly detected as CD31/CD133 positive cells. Conclusion According to our results and considering the safety and efficiency potential of each of the applied methods, the back-flushing in comparison with the other methods can be considered a suitable procedure for MNC isolation from LRFs.

Abstract Objective: To systematically review the hyalinization of experimental tooth movement in humans and animals. Material and Methods: The electronic databases of MEDLINE, PubMed, Cochrane Library, Embase, Institute for Scientific Information, and Google Scholar were searched for performing a systematic review of the related literature published until 2019. Moreover, Endnote X9 software was utilized to manage electronic titles. The searches were fulfilled using keywords of "hyalinization," "orthodontic tooth movement" OR "OTM" AND "periodontal ligament" OR "PDL," "molecular pathways," AND "mechanical cell." Therefore, this systematic review was conducted based on the key consideration of the Preferred Reporting Items for Systematic Reviews and Meta-Analyses (PRISMA) statement. Results: Forces ranged between 2.3 and 50 CN had been applied. According to the research design, incisors had been included in two experiments, whereas maxillary molars had been displaced by tipping forces in another rat examination. However, a majority of investigations had stated that hyalinization had firstly appeared within the first 24 hours. ABSTRACT Stress can inhibit gonadal activity via Hypothalamus-Pituitary-Gonad (HPG) axis activity suppression. In the present study, effects of intermittent feeding (IF) on gonadal function under stress in male and female mice were evaluated. Twenty eight male and twenty eight female mice's were divided into four groups. The control group received adequate food and water without stress. The second group received four days of electric shock without food deprivation. The third group was deprived of food two hours/day for a week, and the fourth group was deprived of food (2 hours/day for seven consecutive days) and then electric foot shock stress was applied to them for four days. Blood samples were collected from all animals for plasma testosterone, estrogen and/or Interlukin-6 (IL-6) evaluation. The animals’ gonads were also removed and fixed for the measure of their weight. Results showed that stress reduces both testosterone and estrogen levels, whereas IF did not change the hormone levels. In addition, stress increases blood IL-6 concentration. The combination of IF and stress, increased the hormone levels in animals. Stress and IF alone had no significant effect on gonadal weight in the male mice, whereas stress decreased gonadal weight in the females. Combination of stress with IF increased gonadal weight in both male and female mice. In conclusion stress showed a negative effect on gonadal function in both animals with more effect on females. Intermittent feeding inhibits the stress effect and even promotes the gonadal function in both sexes. The effect may be due to IL-6 reduction.

Solid-phase peptide synthesis of dipeptide (histidine-β-alanine) as a chelating agent examined by common N-9-fluorenylmethyloxycarbonyl-N-trityl-L-histidine and tert-butyloxycarbonyl-β-alanine-OH amino acid derivatives. Trityl chloride resin was used as a carrier resin. The molecular structure of the dipeptide was definite by using different methods such as ultraviolet visible (UV-Vis), Fourier transform infrared (FTIR), proton (1H) nuclear magnetic ressonance (NMR) and liquid chromatography-mass spectrometry (LC-MS) and the chelating property of synthesized dipeptide was investigated for removing of metal ions Al3+, Cu2+, Hg2+ and Pb2+in vitro. In addition, the pharmacological and biological activities of dipeptide were examined by prediction of activity spectra for substances (PASS) program.

No Irã, um número grande de espécies de Rosa são expostas à extinção. Por causa disto, as técnicas de preservação são necessárias para garantir o futuro destas mesmas espécies. Nesta investigação, os objetivos foram aperfeiçoar a micropropagação e preservar durante médio prazo uma destas espécies, Rosa pulverulenta. No estádio de proliferação, o número máximo de novas folhas (9,60) foi produzido no meio contendo 4 μM BAP + 0 μM GA3, mas o número máximo de gemas axilares foi observado no meio com 4 μM BAP + 3 μM GA3. Os resultados obtidos permitiram inferir que o maior aumento na altura de haste (49,5 mm) durante o estádio de climatização foi observado nos explantes tratados com 1 μM IBA + 0,5 μM NAA durante o enraizamento. Observa-se que embora a geladeira tenha proporcionado uma menor taxa de crescimento, a sobrevivência em médio prazo foi maior (100,00%) do que no phytotron (87,50%). Além disto, a taxa de crescimento dos explantes que foram mantidos na geladeira foi acelerada durante o periodo de recuperação e ficaram próximas daquelas mantidas no phytotron.

In Iran, a large number of Rosa species have been exposed to extinction and therefore preservation techniques are necessary to safeguard their future. In the present investigation, the objectives were to optimize the micropropagation and medium-term conservation of one of these wild species, Rosa pulverulenta. At proliferation stage, the maximum number of new leaves (9.6) were produced on the medium containing 4 μM BAP + 0 μM GA3, whereas the maximum number of axillary shoots (4.1) were observed in the medium containing 4 μM BAP + 3 μM GA3. The results for rooting experiments suggested that the highest increase in stem height (49.5 mm) at the acclimatization stage was observed in plantlets treated with 1 μM IBA + 0.5 μM NAA during the rooting stage. Comparing the refrigerator and phytotron conditions for medium-term conservation of in vitro plantlets indicated that although the refrigerator conditions resulted in lower growth rate compared with the phytotron, the survival rate in refrigerator (100%) was significantly higher than phytotron (87.5%). Furthermore, the growth rate of the plantlets from the refrigerator was accelerated during the recovery period and verged on to the ones stored in the phytotron.
