Resumo Bemisia tabaci é um complexo de espécies que causa danos a uma ampla gama de hospedeiros vegetais por meio de alimentação séria. Ele transmite vírus de plantas de diferentes grupos para importantes safras agrícolas. Algumas safras comerciais importantes do Paquistão são cana-de-açúcar, arroz, tabaco e óleo de semente. Apresenta alta variabilidade genética e é diferenciado em raças ou biótipos. Os biótipos são: biótipo Q, biótipo B, biótipo B2, biótipo M, biótipo L, biótipo A, biótipo H, biótipo C, biótipo K, biótipo N, biótipo R, biótipo E, biótipo P, biótipo J, biótipo S, biótipo AN . Embora o relatório atual baseado no estudo bayesiano das sequências de DNA do gene 1 da oxidase do citocromo mitocondrial (CO1) tenha classificado as diferentes populações de moscas-brancas em doze grupos genéticos, que são Mediterrâneo, África Subsaariana com folha de prata, Oceano Índico, Ásia II, Ásia I, Austrália, Novo Mundo, Itália, China, África Subsaariana sem folha prateada, Batata-doce Mediterrâneo / Ásia Menor / África e Uganda. Os begomovírus são o maior grupo de vírus transmitidos por B. tabaci e causam as principais doenças de culturas, como a doença do cacho do tomate e da pimenta-malagueta, doença do mosaico da mandioca, doença do mosaico amarelo de leguminosas e doença do enrolamento da folha do algodão. O principal objetivo do presente estudo é inculpar conhecimento sobre a diversidade genética da mosca-branca em campos de algodão em todo o Paquistão por meio da análise da sequência parcial de DNA do gene mitocondrial Citocromo Oxidase I (mtCO1).

Abstract Bemisia tabaci is a species complex that causes damage to its broad range of plant hosts through serious feeding. It transmits plant viruses of different groups to important agricultural crops. Some important cash crops of Pakistan are sugar cane, rice, tobacco and seed oil. It shows high genetic variability and is differentiated as races or biotypes. Biotypes are, biotype Q, biotype B, biotype B2, biotype M, biotype L, biotype A, biotype H, biotype C, biotype K, biotype N, biotype R, biotype E, biotype P, biotype J, biotype S, biotype AN. Although the current report based on the Bayesian study of mitochondrial cytohrome oxidase gene1 (CO1) DNA sequences has classified the different populations of whiteflies into twelve genetic groups which are Mediterranean, Sub-Saharan Africa silverleafing, Indian Ocean, Asia II, Asia I, Australia, New World, Italy, China, Sub-Saharan Africa non-silverleafing, Mediterranean/Asia Minor/Africa and Uganda sweet potato. Begomoviruses is largest group of viruses transmitted by B. tabaci and cause major diseases of crops such as tomato and chili leaf curl disease, cassava mosaic disease; yellow mosaic disease of legumes and cotton leaf curl disease. The main objective of current study is to inculpate knowledge regarding genetic diversity of whitefly in cotton fields across Pakistan via analysis of partial DNA sequence of mitochondrial gene Cytochrom Oxidase I (mtCO1).

Resumo O estudo foi desenhado para investigar o efeito da suplementação de mananoligossacarídeo (MOS) na histomorfologia intestinal, imunidade contra o vírus da doença de Newcastle (NDV) e parâmetros produtivos de frangos de corte. Um total de 1.800 pintos de corte de um dia da linhagem Cobb-500 foram selecionados e distribuídos aleatoriamente em 6 grupos de tratamento: T1 (dieta basal sem antibióticos como controle negativo); T2 (dieta basal mais antibióticos como grupo controle positivo); T3 (dieta basal mais 200g/ton MOS); T4 (dieta basal mais 400g/ton MOS); T5 (dieta basal mais 600g/ton MOS) e T6 (dieta basal mais 800g/ton MOS). Cada tratamento tinha 6 repetições e o consumo de ração, ganho de peso corporal e conversão alimentar foram registrados semanalmente. Os resultados mostraram que as aves suplementadas com MOS apresentam consumo de ração, ganho de peso e CA significativamente maiores (P < 0,05). Da mesma forma, a suplementação de MOS mostrou efeito positivo na altura das vilosidades e na profundidade das criptas tanto no jejuno quanto no íleo. A densidade de células caliciformes não foi afetada pela adição de MOS (P < 0,05). Além disso, as aves alimentadas com dietas contendo MOS apresentaram melhor desempenho produtivo em comparação aos grupos controle positivo e negativo. Em conclusão, o MOS pode substituir os promotores de crescimento de antibióticos (AGPs) como defensores de alimentos não microbianos que melhoram o desempenho.

Abstract The study was designed to investigate the effect of mannan-oligosaccharide (MOS) supplementation on intestinal histomorphology, immunity against Newcastle disease virus (NDV) and productive parameters of broilers. A total of 1800, day old broiler chicks of Cobb-500 strain were selected and randomly assorted into 6 treatment groups: T1 (basal diet without antibiotics as negative control); T2 (basal diet plus antibiotics as positive control group); T3 (basal diet plus 200g/ton MOS); T4 (basal diet plus 400g/ton MOS); T5 (basal diet plus 600g/ton MOS) and T6 (basal diet plus 800g/ton MOS). Each treatment was having 6 replicates and the feed intake, body weight gain and feed conversion ratio (FCR) were recorded on weekly basis. Results showed that, MOS supplemented birds have significantly higher feed intake, weight gain and FCR (P < 0.05). Similarly, supplementation of MOS showed positive effect on villus height and crypt depth both in jejunum and ilium. Goblet cell density was unaffected by MOS addition (P < 0.05). Furthermore, birds fed with diets containing MOS, exhibited better productive performance in comparison to positive and negative control groups. In conclusion, MOS can replace antibiotic growth promoters (AGPs) as non-microbial performance-enhancing feed advocates.

This study sought to explore the effect of the administration of long-term, exogenous oxytocin on buffalo milk. In this study, newly-calved postpartum buffaloes (Nili-Ravi) (n = 24) were divided equally into three groups: (control without oxytocin - CON, 10 i.u. oxytocin - LOW, 30 i.u. oxytocin - HIGH). Oxytocin was injected twice a day before each 154-day milking. Milk production, milk composition, somatic cell counts, and progesterone (P4) were evaluated daily, weekly, and fortnightly, respectively. Results revealed that the HIGH group produced high milk production and pH values, but lower milk density than the CON and LOW groups. The HIGH group and LOW group had higher milk fat and solid-not-fat (SNF) compared to the CON group. SNF and protein were higher in the LOW group than the CON group. Freezing point, lactose, solids, somatic cell count, and P4 were similar in all three groups. Overall, it can be concluded that long-term, exogenous oxytocin has a positive effect on milk production, fat, SNF, and protein, with a negative effect on milk pH. None of the oxytocin treatments affected somatic cell counts and P4 concentration in postpartum buffalo milk.

Resumo O presente estudo teve como objetivo determinar os efeitos de diferentes níveis de Zingiber officinale como aditivo à base de plantas medicinais sobre o desempenho de crescimento, características da carcaça, bioquímica sérica, contagem bacteriana total (CBT), morfologia intestinal e parâmetros imunológicos de frangos de corte. Um total de 1.500 pintos de corte de um dia de idade (Hubbard) foram igualmente credenciados em cinco grupos de tratamento, cada um com seis repetições (50 aves/repetição). Cinco dietas experimentais foram preparadas usando dieta basal, ou seja, com controle positivo de antibióticos (PC), 3 g/kg de gengibre (grupo A), 6 g/kg de gengibre (grupo B), 9 g/kg de gengibre (grupo C) e sem controle negativo de antibióticos (NC). Os grupos A e C apresentaram consumo de ração (FI) significativamente (p < 0,05) maior do que os outros grupos. O grupo C apresentou contagem bacteriana total (CBT) significativamente menor (p < 0,05) seguido pelo grupo B em comparação com o NC. As características da carcaça apresentaram efeitos não significativos entre os diferentes tratamentos. O comprimento e largura médios das vilosidades foram significativamente (p < 0,05) maiores em todos os grupos suplementados com gengibre em comparação com os grupos de controle. Os parâmetros séricos do sangue, incluindo colesterol, triglicerídeos e lipoproteínas de baixa densidade (LDL), foram significativamente (p < 0,05) menores nos grupos B e C em comparação com os grupos controle. Enquanto as lipoproteínas de alta densidade (HDL) foram significativamente maiores no grupo B em comparação com os outros. Em conclusão, a suplementação de gengibre a 0,6% na dieta basal melhorou significativamente o desempenho de crescimento e a morfometria intestinal de frangos de corte. Ele também mostrou um impacto positivo sobre o colesterol, triglicerídeos e micróbios intestinais. Portanto, o gengibre pode ser um substituto melhor para os promotores de crescimento com antibióticos.

Abstract The current study aimed to determine the effects of different levels of Zingiber officinale as a herbal feed additive on growth performance, carcass characteristic, serum biochemistry, total bacterial count (TBC), gut morphology, and immunological parameters of broilers. A total of 1500, day-old broiler chicks (Hubbard) were equally accredited to five treatment groups, each with six replicates (50 birds/replicate). Five experimental diets were prepared using basal diet i.e. with antibiotics positive control (PC), 3 g/kg ginger (group A), 6 g/kg ginger (group B), 9 g/kg ginger (group C) and without antibiotics negative control (NC). Group A and C showed significantly (p<0.05) higher feed intake (FI) as compared to other groups. Group C showed significantly (p<0.05) lower Total bacterial count (TBC) followed by group B as compared to NC. Carcass characteristics showed non-significant effects among different treatments. Mean villi length and width were significantly (p <0.05) higher in all ginger supplemented groups as compared to the control groups. Blood serum parameters including cholesterol, triglycerides, and low-density lipoproteins (LDL) were significantly (p<0.05) lower in groups B and C in comparison with the control groups. Whereas high-density lipoproteins (HDL) was significantly higher in group B as compared to the others. In conclusion, ginger supplementation @0.6% in the basal diet significantly improved growth performance and gut morphometry of broilers. It also showed a positive impact on cholesterol, triglycerides and gut microbes. Therefore, ginger could be a better substitute for antibiotic growth promoters.

Resumo O timerosal é um composto organomercurial, utilizado na preparação de imunoglobulina intramuscular, antivenenos, tintas de tatuagem, antígenos de teste cutâneo, produtos nasais, gotas oftálmicas e vacinas como conservante. Na maioria das espécies animais e nos humanos, o rim é um dos principais locais de deposição de compostos de mercúrio e órgãos-alvo de toxicidade. Assim, a presente pesquisa teve como objetivo avaliar a nefrotoxicidade induzida pelo timerosal em ratos machos. Vinte e quatro ratos albinos machos adultos foram categorizados em quatro grupos. O primeiro grupo era um grupo de controle. Ratos do Grupo II, Grupo III e Grupo IV receberam 0,5µg / kg, 10µg / kg e 50µg / kg de timerosal uma vez ao dia, respectivamente. A administração de timerosal diminuiu significativamente as atividades de catalase (CAT), superóxido dismutase (SOD), peroxidase (POD), glutationa redutase (GR), glutationa (GSH) e conteúdo de proteína, enquanto aumentou as substâncias reativas ao ácido tiobarbitúrico (TBARS) e peróxido de hidrogênio (H2O2) níveis dependentes da dose. Os níveis de nitrogênio ureico no sangue (BUN), creatinina, urobilinogênio, proteínas urinárias, molécula de lesão renal-1 (KIM-1) e lipocalina associada à gelatinase de neutrófilos (NGAL) aumentaram substancialmente. Em contraste, a albumina urinária e a depuração da creatinina foram reduzidas de forma dependente da dose nos grupos tratados com timerosal. Os resultados demonstraram que o timerosal aumentou significativamente os indicadores de inflamação, incluindo fator nuclear kappaB (NF-κB), fator de necrose tumoral-α (TNF-α), interleucina-1β (IL-1β), níveis de interleucina-6 (IL-6) e atividades da ciclooxigenase-2 (COX-2), DNA e danos histopatológicos dependentes da dose. Portanto, os presentes achados verificaram que o timerosal exerceu nefrotoxicidade em ratos albinos machos.

Abstract Thimerosal is an organomercurial compound, which is used in the preparation of intramuscular immunoglobulin, antivenoms, tattoo inks, skin test antigens, nasal products, ophthalmic drops, and vaccines as a preservative. In most of animal species and humans, the kidney is one of the main sites for mercurial compounds deposition and target organs for toxicity. So, the current research was intended to assess the thimerosal induced nephrotoxicity in male rats. Twenty-four adult male albino rats were categorized into four groups. The first group was a control group. Rats of Group-II, Group-III, and Group-IV were administered with 0.5µg/kg, 10µg/kg, and 50µg/kg of thimerosal once a day, respectively. Thimerosal administration significantly decreased the activities of catalase (CAT), superoxide dismutase (SOD), peroxidase (POD), glutathione reductase (GR), glutathione (GSH), and protein content while increased the thiobarbituric acid reactive substances (TBARS) and hydrogen peroxide (H2O2) levels dose-dependently. Blood urea nitrogen (BUN), creatinine, urobilinogen, urinary proteins, kidney injury molecule-1 (KIM-1), and neutrophil gelatinase-associated lipocalin (NGAL) levels were substantially increased. In contrast, urinary albumin and creatinine clearance was reduced dose-dependently in thimerosal treated groups. The results demonstrated that thimerosal significantly increased the inflammation indicators including nuclear factor kappaB (NF-κB), tumor necrosis factor-α (TNF-α), Interleukin-1β (IL-1β), Interleukin-6 (IL-6) levels and cyclooxygenase-2 (COX-2) activities, DNA and histopathological damages dose-dependently. So, the present findings ascertained that thimerosal exerted nephrotoxicity in male albino rats.

Resumo A cisplatina (CP) é uma droga antineoplásica poderosa, comumente usada, com vários efeitos colaterais. Casticin (CAS) é considerado um eliminador de radicais livres e um potente antioxidante. A presente pesquisa foi planejada para avaliar o potencial curativo da CAS em lesão renal induzida por PC em ratos albinos machos. Vinte e quatro ratos albinos machos foram distribuídos em quatro grupos iguais. O Grupo 1 foi considerado grupo controle. Os animais do Grupo 2 foram injetados com 5 mg / kg de PB por via intraperitoneal. O Grupo 3 foi cotratado com CAS (50 mg / kg) por via oral e injeção de CP (5 mg / kg). O Grupo 4 foi tratado com CAS (50 mg / kg) por via oral durante todo o experimento. A administração de CP reduziu substancialmente as atividades de catalase (CAT), superóxido dismutase (SOD), peroxidase (POD), glutationa S-transferase (GST), glutationa redutase (GSR), glutationa (GSH), enquanto aumentou as substâncias reativas ao ácido tiobarbitúrico (TBARS) e níveis de peróxido de hidrogênio (H2O2). Os níveis de ureia, creatinina urinária, urobilinogênio, proteínas urinárias, molécula 1 de lesão renal (KIM-1) e lipocalina associada à gelatinase de neutrófilos (NGAL) aumentaram substancialmente. Em contraste, a albumina e a depuração da creatinina foram significativamente reduzidas no grupo tratado com PC. Os resultados demonstraram que a CP aumentou significativamente os indicadores de inflamação, incluindo fator nuclear kappa-B (NF-κB), fator de necrose tumoral-α (TNF-α), interleucina-1β (IL-1β), interleucina-6 (IL-6) níveis e atividade da ciclooxigenase-2 (COX-2) e danos histopatológicos. No entanto, a administração de CAS apresentou um efeito paliativo contra a toxicidade renal gerada por CP e recuperou todos os parâmetros, trazendo-os a um nível normal. Estes resultados revelaram que o CAS é um composto eficaz com potencial curativo para combater o dano renal induzido por CP.

Abstract Cisplatin (CP) is a commonly used, powerful antineoplastic drug, having numerous side effects. Casticin (CAS) is considered as a free radical scavenger and a potent antioxidant. The present research was planned to assess the curative potential of CAS on CP persuaded renal injury in male albino rats. Twenty four male albino rats were distributed into four equal groups. Group-1 was considered as a control group. Animals of Group-2 were injected with 5mg/kg of CP intraperitoneally. Group-3 was co-treated with CAS (50mg/kg) orally and injection of CP (5mg/kg). Group-4 was treated with CAS (50mg/kg) orally throughout the experiment. CP administration substantially reduced the activities of catalase (CAT), superoxide dismutase (SOD), peroxidase (POD), glutathione S-transferase (GST), glutathione reductase (GSR), glutathione (GSH) content while increased thiobarbituric acid reactive substances (TBARS), and hydrogen peroxide (H2O2) levels. Urea, urinary creatinine, urobilinogen, urinary proteins, kidney injury molecule-1 (KIM-1), and neutrophil gelatinase-associated lipocalin (NGAL) levels were substantially increased. In contrast, albumin and creatinine clearance was significantly reduced in CP treated group. The results demonstrated that CP significantly increased the inflammation indicators including nuclear factor kappa-B (NF-κB), tumor necrosis factor-α (TNF-α), Interleukin-1β (IL-1β), Interleukin-6 (IL-6) levels and cyclooxygenase-2 (COX-2) activity and histopathological damages. However, the administration of CAS displayed a palliative effect against CP-generated renal toxicity and recovered all parameters by bringing them to a normal level. These results revealed that the CAS is an effective compound having the curative potential to counter the CP-induced renal damage.

The study aimed to test whether the ultrasonographic morphometry and characteristic traits of udders and teats could be used to identify clinical mastitis in forty-eight adult Holstein-Friesian dairy cows. The inclusion and exclusion criteria were also declared with the help of the California Mastitis Test and Somatic Cell Count. Clinically, dairy cows (n = 48) were in their third lactation in order to undergo both ultrasonographic and phenotypic morphometry. Udder parenchyma as echotexture, teat width, teat width at the rosette of Furstenberg, teat cistern width, teat wall thickness, teat canal length, and ratios of TC/TWT, TWT/TW and TC/TW, as well as supramammary lymph node size, were evaluated, whereas phenotypic morphometric traits, i.e., udder circumference, udder depth, udder length, teat length, teat circumference, teat diameter, and the shortest distance from teat ends to floor were measured. There was substantial difference between morphological parameters of the teat such as, teat width, teat width at the rosette of Furstenberg, teat wall thickness, ratio of TC/TWT, ratio of TWT/TW, ratio TC/TW, teat canal length, echogenicity variables, and phenotypic morphometric traits, except for the teat cistern width. Pearson correlation indicated a strong correlation between lymph node length and lymph node width. Clinical mastitis in cattle is related to udder and teat morphometric traits. Incorporating these characteristics into the selection and breeding programme may aid in the selection of animals for mastitis resistance. Ultrasound is an effective imaging and morphological assessment tool for the diagnosis and prognosis of clinical mastitis in dairy cows.

ABSTRACT One of the most effective and cost-efficient methods to manage plant parasitic nematodes is to use resistant cultivars. However, little information on the resistance of commercial okra cultivars grown in the country was available. Therefore, nine okra cultivars were evaluated for their comparative resistance against Meloidogyne incognita, the most damaging root-knot nematode. None of the tested okra cultivars showed high level of resistance. However, three cultivars viz. Pusa Swami, PB Selection and Green Star appeared as moderately resistant. Sabz Pari, Neelum and Tulsi were found moderately susceptible to the nematode. Two cultivars i.e. Ikra-1 and Ikra-2 were susceptible and Arka Anamika was highly susceptible. The okra cultivars differed significantly in their growth parameters depending on their response to M. incognita. Moderately resistant cultivars sustained little damage as compared to moderately susceptible or susceptible cultivars while the highly susceptible cultivar suffered the maximum damage by the nematode. Significant differences in the numbers of galls, eggmasses and reproductive factors of M. incognita on the nine okra cultivars were also observed. The highest numbers were found on Arka Anamika, followed by Ikra-1 and Ikra-2. The lowest numbers were found on the moderately resistant cultivars, followed by the moderately susceptible ones. High and positive correlations between the number of galls and eggmasses and the decreases in growth metrics were found using linear and non-linear regression analysis.

RESUMO: Experimentos de campo foram conduzidos durante dois anos para investigar a fitotoxicidade da algaroba (Prosopis juliflora) no controle de plantas daninhas e no rendimento do trigo nos períodos de 2013-14 e 2014-15. Foram preparados extratos aquosos de folhas, caules e raízes de algaroba com concentração de 10, 20, 30 e 40%, os quais foram comparados com água corrente (controle). Utilizou-se o delineamento em blocos completamente casualizados com quatro repetições. Todos os parâmetros (densidade de plantas daninhas, biomassa fresca das plantas daninhas, biomassa seca das plantas daninhas, teor de clorofila, índice de área foliar, duração de área foliar, taxa de crescimento da cultura, taxa de assimilação líquida, altura da planta e número de perfilhos) sofreram redução significativa pela aplicação dos extratos de algaroba. Entre as diferentes concentrações, o nível máximo (40%) das três partes das plantas da algaroba reduziu os parâmetros do estudo, incluindo densidade das plantas daninhas, biomassa fresca e seca das plantas daninhas, índice de área foliar, duração da área foliar, taxa de crescimento da cultura, taxa de assimilação líquida, teores de clorofila, altura das plantas, número de perfilhos, comprimento da espiga, número de grãos, peso de mil grãos, rendimento de grãos, rendimento biológico, índice de colheita e teor de proteína do grão. Com base nos resultados, conclui-se que os extratos aquosos da algaroba são capazes de conter muitas substâncias fitotóxicas que podem exercer efeito negativo no crescimento e no rendimento das culturas agrícolas. Por essa razão, o trigo não deve ser plantado perto da algaroba.

ABSTRACT: Two-year field experiments were conducted to investigate the phytotoxicity of mesquite (Prosopis juliflora) on weed control and yield of wheat in 2013-14 and 2014-15. Mesquite aqueous extracts were prepared with 10, 20, 30 and 40% concentration of leaves, stems, and roots and were compared with tap water (control). These experiments were laid out in a completely randomized block design with four replications. All parameters (weed density, fresh weed biomass, dry weed biomass, chlorophyll content, leaf area index, leaf area duration, crop growth rate, net assimilation rate, plant height and number of tillers) were significantly reduced by applying the mesquite extracts. Among different concentrations, the maximum level (40%) of all three plant parts of mesquite subsequently reduced the study parameters including weed density, fresh and dry weed biomass, leaf area index, leaf area duration, crop growth rate, net assimilation rate, chlorophyll contents, plant height, number of tillers, spike length, number of grains, 1000-grain weight, grain yield, biological yield, harvest index and grain protein content. On the basis of the results, it is concluded that aqueous extracts of mesquite may contain many phytotoxic substances which can negatively affect the growth and yield of agricultural crops; therefore, wheat should not be planted close to mesquite.

MTT (3-(4,5-dimethyl thiazol-2-yl)-2,5-diphenyl tetrazolium bromide) assay is commonly used to validate the viability of metabolically active cells. The study was conducted to examine and validate the MTT test to assess the sperm viability of Nili-Ravi buffalo bulls and compare the efficiency of the test with the supra-vital staining technique (eosin-nigrosine) and hypo-osmotic swelling test. Fresh semen samples from breeding Nili-Ravi buffalo bulls (n = 20) were collected using an artificial vagina. After assessing the quality of semen for normal parameters, the MTT assay was carried out in phosphate buffer saline. Results revealed a high significant correlation (r = 0.995) between the viability of sperm and the rate of reduction of MTT. The other proportions of some semen samples showed a weak relationship between the eosinnigrosine method (r = -0.32), hypo-osmotic swelling test (r = -0.12) and motility (r = -0.08). However, the MTT assay was found to be superior to other tests as it was able to determine those sperm which were more than 90% viable. In conclusion, the MTT assay is a simple, robust test that can be used to select Nili-Ravi buffalo bulls on the basis of sperm quality.
