Abstract South American opossums (Didelphis spp.) are definitive hosts of Sarcocystis neurona, Sarcocystis speeri, Sarcocystis lindsayi and Sarcocystis falcatula. In Brazil, diverse studies have demonstrated a high frequency of Sarcocystis falcatula-like in sporocysts derived from opossums, and high genetic diversity has been observed in surface antigen-encoding genes (SAGs). In this study, genetic diversity of Sarcocystis spp. derived from Didelphis albiventris and Didelphis aurita from the cities of Campo Grande and São Paulo, was accessed by sequencing SAG2, SAG3, SAG4, the first internal transcribed spacer (ITS-1) and cytochrome c oxidase subunit I (cox1). Molecular identification was performed for 16 DNA samples obtained from sporocyst or culture-derived merozoites. The ITS-1, cox1, and SAG3 fragments were cloned, whereas SAG2 and SAG4 were sequenced directly from PCR products. Four alleles variants were found for SAG2, 13 for SAG3 and seven for SAG4, from which four, 13 and four, respectively, were novel. Twenty-seven allele variants were found for ITS-1, all phylogenetically related to S. falcatula-like previously described in Brazil. Sarcocystis sp. phylogenetically related to Sarcocystis rileyi was evidenced by cox1 in three opossums. Further studies are needed to clarify the role of Didelphis spp. as definitive hosts of Sarcocystis spp. other than that previous described.

Resumo Gambás sul-americanos (Didelphis spp.) são hospedeiros definitivos de Sarcocystis neurona, Sarcocystis speeri, Sarcocystis lindsayi e Sarcocystis falcatula. No Brasil, diversos estudos têm demonstrado alta frequência de Sarcocystis falcatula-like em esporocistos derivados de gambás, com grande diversidade nos genes que codificam antígenos de superfície (SAGs). Neste estudo, a diversidade genética de Sarcocystis spp., oriundos de Didelphis albiventris e Didelphis aurita, dos municípios de Campo Grande e São Paulo, foi acessada por meio do sequenciamento de SAG2, SAG3 e SAG4, da primeira região espaçadora interna transcrita (ITS-1) e citocromo c oxidase subunidade I (cox1). Identificação molecular foi realizada em 16 amostras de DNA, obtidas de esporocistos ou merozoítos derivados de cultivo. Os fragmentos de ITS-1, cox1 e SAG3 foram clonados, enquanto SAG2 e SAG4 foram sequenciados diretamente dos produtos de PCR. Quatro alelos foram observados em SAG2, 13 em SAG3 e sete em SAG4, sendo novos quatro, 13 e quatro, respectivamente. Em ITS-1, 27 alelos foram observados, todos filogeneticamente relacionados à S. falcatula-like, previamente detectados no Brasil. Sarcocystis sp. filogeneticamente relacionado à Sarcocystis rileyi foi evidenciado por cox1 em três gambás. Mais estudos são necessários para entender o papel de Didelphis spp. como hospedeiro definitivo de Sarcocystis spp. diferentes daqueles previamente descritos.

Abstract The genus Neorickettsia comprises trematode-associated bacteria that can cause diseases in animals and humans. Despite detection of Neorickettsia antigens in the intestine of coatis kept in captivity in southern Brazil through immunohistochemistry, the molecular identity of the bacteria in South American procyonids remains elusive. The aim of the present study was to investigate the occurrence of Neorickettsia sp. in blood samples from coatis in central-western Brazil. Between March 2018 and January 2019, animals were captured and recaptured in two areas of the Cerrado (Parque Estadual do Prosa, PEP; and Vila da Base Aérea, VBA) located in the city of Campo Grande, state of Mato Grosso do Sul, central-western Brazil. All captures were performed according to convenience. DNA from 97 blood samples was subjected to nested PCR (nPCR) targeting a fragment of the 16S rRNA gene of Neorickettsia sp. Six samples (3.6%; five from VBA and one from PEP) from different coatis were positive in nPCR based on the 16S rRNA. The sequences obtained (~500 bp) showed ˃ 99% similarity to N. risticii. Phylogenetic analysis clustered the sequences detected in the present study in a clade with N. risticii. This is the first molecular detection of Neorickettsia sp. in coatis in Brazil.

Resumo O gênero Neorickettsia compreende bactérias associadas a trematódeos que podem causar doenças em animais e humanos. Apesar da detecção de antígenos de Neorickettsia por imuno-histoquímica no intestino de quatis mantidos em cativeiro no sul do Brasil, a identidade molecular da bactéria em procionídeos da América do Sul permanece indefinida. O presente trabalho teve como objetivo investigar a ocorrência de Neorickettsia sp. em amostras de sangue de quatis do Centro-Oeste do Brasil. Entre março de 2018 e janeiro de 2019, os animais foram capturados em duas áreas de Cerrado (Parque Estadual do Prosa PEP e Vila da Base Aérea VBA) localizadas na cidade de Campo Grande, Mato Grosso do Sul, Centro-Oeste do Brasil. Todas as capturas e recapturas foram realizadas por conveniência. O DNA de 97 amostras de sangue foi submetido a "nested" (nPCR), baseada em um fragmento do gene 16S rRNA de Neorickettsia sp. Seis (3,6% - 5 de VBA e 1 de PEP) amostras de quatis diferentes foram positivas na nPCR, baseada no rRNA 16S. As sequências obtidas (~500 pb) apresentaram ˃99% de identidade com N. risticii. A inferência filogenética agrupou as sequencias detectadas no presente estudo em um clado com N. risticii. Esta é a primeira detecção molecular de Neorickettsia sp. em quatis do Brasil.

Resumo Foi avaliada a epidemiologia, hematologia e patologia das infecções por Leishmania spp., Toxoplasma gondii, Platynosomum illiciens, vírus da imunodeficiência felina (FIV) e vírus da leucemia felina (FeLV) e das coinfecções em gatos não domiciliados em uma área endêmica para leishmaniose. O diagnóstico foi realizado por exames sorológicos e necropsia. Foram descritas lesões macroscópicas e achados histopatológicos. Usaram-se imuno-histoquímica e hibridização cromogênica in situ para detecção de L. infantum. Encontrou-se infecção em 27 dos 50 gatos amostrados, dentre eles, 14 apresentavam coinfecções. Foi observada forte correlação entre esplenomegalia e linfadenomegalia com FeLV, e associação entre lesões hepáticas e caquexia com parasitismo por P. illiciens. Além disso, foi encontrado um aumento significativo na contagem de monócitos nos animais infectados pelo FeLV e uma diminuição na contagem de hemácias nos animais infectados pelo FIV. Formas amastigotas de Leishmania spp. e alterações teciduais foram detectadas em órgãos linfoides de um animal coinfectado com P. illiciens, T. gondii e FIV. O poliparasitismo registrado em gatos errantes do Centro-Oeste brasileiro deve ser considerado nas estratégias de controle de zoonoses. Além disso, gatos errantes de Campo Grande podem ser fontes de infecção de FIV, FeLV e P. illiciens para populações de gatos domiciliados.

Abstract We evaluated the epidemiological, hematological, and pathological data of Leishmania spp., Toxoplasma gondii, Platynosomum illiciens, feline immunodeficiency virus (FIV), and feline leukemia virus (FeLV) infections and the coinfections in stray cats of an endemic area for leishmaniasis. The diagnosis was performed by serological tests and necropsy. We described gross lesions and histopathological findings. We used immunohistochemistry and chromogenic in situ hybridization for L. infantum detection. We found infection in 27 out of 50 sampled cats, among them, 14 presented coinfections. A strong correlation between splenomegaly and lymphadenomegaly with FeLV, and an association between hepatic lesions and cachexia with parasitism due to P. illiciens were observed. Moreover, we found a significant increase in the monocyte count in the FeLV-infected and a decrease in the red blood cell count in the FIV-infected animals. Amastigote forms of Leishmania spp. and tissue changes were detected in lymphoid organs of an animal coinfected with P. illiciens, T. gondii, and FIV. Polyparasitism recorded in stray cats of the Brazilian Midwest should be considered in effective control strategies for public health diseases. Moreover, stray cats of Campo Grande may be a source of infection of FIV, FeLV and P. illiciens for populations of domiciled cats.

ABSTRACT Background: Rickettsia of the spotted fever group (SFG) has been reported in ticks and domestic animals in Campo Grande (CG), Midwest Brazil. Methods: We searched for Rickettsia in the SFG in capybaras and their ticks in an urban park in the CG. Results: The seropositivity rate was 88.2% (15/17). Although 87.7% of the capybaras sampled showed infestations with Amblyomma sculptum, A. dubitatum, and Amblyomma spp., no molecular results were detected in ticks. Conclusions: Since Rickettsia from the SFG circulates among capybaras in the urban parks of Campo Grande, this large rodent species should be monitored within the One Health Agenda.

Resumo Este estudo teve como objetivo determinar a co-soropositividade para agentes transmitidos por carrapatos, como Anaplasma marginale, Babesia bovis, Babesia bigemina, Coxiella burnetii, Anaplasma phagocytophilum, e Trypanosoma vivax em bovinos de corte do Pantanal Brasileiro. Amostras de soro foram colhidas de 400 animais (200 vacas; 200 bezerros) e submetidas a Ensaios Imunoenzimáticos Indiretos (iELISA) para detecção de anticorpos IgG anti- A. marginale, anti- B. bovis, anti- B. bigemina e anti- T. vivax, e à Reação de Imunofluorescência Indireta (RIFI) para detecção de anticorpos IgG anti -C. burnetii e anti- A. phagocytophilum. Ausência de correlação foi vista entre os animais soropositivos para C. burnetii e A. phagocytophilum com os outros agentes e correlação moderada ocorreu entre A. marginale x B. bigemina x B. bovis. Vacas foram mais soropositivas que bezerros para T. vivax e bezerros mais soropositivos que vacas para B. bovis e B. bigemina. Maior número de animais soropositivos para um único agente foi visto para T. vivax (15,2%). Vacas demonstraram maior co-soropositividade para T. vivax + A. marginale (25,5%) e bezerros para T. vivax + B. bovis + B. bigemina + A. marginale (57,5%). A alta correlação entre a soropositividade para A. marginale x B. bovis x B. bigemina é provavelmente devida à presença do vetor biológico, o carrapato Rhipicephalus microplus, nas fazendas estudadas. As vias de transmissão comuns, mediadas por dípteros hematófagos e fômites, podem explicar a alta co-soropositividade das vacas para A. marginale e T. vivax. A baixa soropositividade para C. burnetii é provavelmente devida ao tipo de sistema de criação empregado (extenso). A soropositividade para A. phagocytophilum em apenas um animal sugere a ocorrência de reação sorológica cruzada com outro agente do gênero Anaplasma.

Abstract This study investigated the seropositivity for five different tick-borne agents, namely Anaplasma marginale, Babesia bovis, Babesia bigemina, Coxiella burnetii, Anaplasma phagocytophilum, and Trypanosoma vivax in beef cattle in the Brazilian Pantanal. The serum samples collected from animals (200 cows; 200 calves) were used in indirect enzyme-linked immunosorbent assays (iELISA) to detect IgG antibodies against A. marginale, B. bovis, B. bigemina, and T. vivax, and Indirect Fluorescent Antibody Test (IFAT) for detecting IgG antibodies against C. burnetii and A. phagocytophilum. No correlation was observed between seropositivity for C. burnetii and A. phagocytophilum with other agents whereas moderate correlation was observed for A. marginalexB. bigemina x B. bovis. Cows were more seropositive for T. vivax whereas calves were more seropositive for B. bovis and B. bigemina. The highest number of seropositive animals by a single agent was observed for T. vivax (15.2%). Co-seropositivity for T. vivax + A. marginale was higher in cows (25.5%) and for T. vivax + B. bovis + B. bigemina + A. marginale was higher in calves (57.5%). The high seropositivity correlation for A. marginale x B. bovis x B. bigemina is probably due to the presence of the tick biological vector, Rhipicephalus microplus, in the studied farms. Common transmission pathways, mediated by hematophagous dipterans and fomites, may explain the high co-seropositivity of cows for A. marginale and T. vivax. Low seropositivity to C. burnetii is probably due to the type of breeding system employed (extensive). Seropositivity for A. phagocytophilum in only one animal suggests the occurrence of a cross-serological reaction with another agent of the genus Anaplasma.

Resumo O gene msp4 de A. marginale é unicodon, estável e pouco heterogêneo, sendo considerado como um marcador útil para caracterização filogeográfica desta bactéria. Este trabalho teve como objetivo analisar a filogeografia de A. marginale com base no gene msp4 em bovinos de corte do Pantanal Brasileiro, comparativamente a outra regiões do mundo. Alíquotas de sangue foram colhidas de 400 bovinos (200 vacas e 200 bezerros) em cinco propriedades de cria e recria extensiva. Como resultado, 56,75% (227/400) mostraram-se positivas para A. marginale pela qPCR para o gene msp1β e destas, 8,37% (19/227) amostras foram positivas na PCR convencional para o gene msp4. Na análise de distância Network, 14 sequências do Pantanal brasileiro foram agrupadas em um único grupo com as da Thailândia, Índia, Espanha, Colômbia, Paraná (Brasil), México, Portugal, Argentina, China, Venezuela, Austrália, Italia e Minas Gerais (Brasil). Dentre 68 sequências do Brasil e do mundo, constatou-se a presença de 15 genótipos, sendo o genótipo número um (#1) o mais distribuído. As sequências msp4 de A. marginale detectadas em bovinos de corte no Pantanal brasileiro apresentaram baixo polimorfismo com formação de dois genogrupos filogeneticamente relacionados àqueles encontrados em ruminantes de países das América do Sul e Central, Europa e Ásia.

Abstract The msp4 gene of A. marginale is unicodon, stable and mostly homogeneous, being considered as a useful marker for phylogeographic characterization of this bacterium. The objective of this work was to analyze the phylogeography of A. marginale based on the msp4 gene in beef cattle from the Brazilian Pantanal, compared to those found in other regions worldwide. The blood samples investigated were collected from 400 animals (200 cows and 200 calves) reared in five extensive breeding farms in this region. The results indicated that of the evaluated samples, 56.75% (227/400) were positive for A. marginale based on the msp1β gene by quantitatitve PCR (qPCR), while 8.37% (19/227) were positive for the msp4 gene in the conventional PCR. In the Network distance analysis, 14 sequences from the Brazilian Pantanal were grouped into a single group with those from Thailand, India, Spain, Colombia, Parana (Brazil), Mexico, Portugal, Argentina, China, Venezuela, Australia, Italy and Minas Gerais (Brazil). Among 68 sequences from Brazil and the world, 15 genotypes were present while genotype number one (#1) was the most distributed worldwide. Both Splitstree and network analyses showed that the A. marginale msp4 sequences detected in beef cattle from the Brazilian Pantanal showed low polymorphism, with the formation of one genogroup phylogenetically related to those found in ruminants from South and Central America, Europe, and Asia.

Resumo O objetivo do trabalho foi avaliar a influência do sexo, da sazonalidade e de infecções por Trypanosoma cruzi e Trypanosoma evansi nos valores hematológicos de lobinhos (Cerdocyon thous) que habitam o Pantanal Sul-Mato-Grossense. Entre novembro de 2015 e outubro de 2016, foram amostrados 48 lobinhos. Os valores hematológicos mensurados foram as contagens de eritrócitos e leucócitos, volume globular e volume corpuscular médio. Consideramos como parasitados os animais positivos para T.cruzi e T. evansi em qualquer um dos testes diagnósticos utilizados. Observamos que sete (14.5%) lobinhos encontraram-se parasitados somente por T.cruzi, sete por T. evansi, e nove (19%) estavam coinfectados. Os animais parasitados por T.cruzi mostraram um aumento significativo das contagens de leucócitos (14.7 x 103) em relação aos animais não parasitados (10.4 x 103), parasitados por T. evansi (12.4 x 103) e coinfectados (12.9 x 103). Observamos diferença significativa em relação aos valores médios de eritrócitos e volume corpuscular médio entre os períodos de cheia e seca: 2.6 x 106 e 165, e 3.4 x 106 e 132, respectivamente. Todos apresentaram macrocitose não confirmada morfologicamente.

Abstract The aim of this work was to evaluate the influence of sex, seasonality and infections by Trypanosoma cruzi and Trypanosoma evansi on the hematological values of crab-eating foxes (Cerdocyon thous) that inhabit the Pantanal from Mato Grosso do Sul. Between November of 2015 and October of 2016, 48 animals were sampled. The hematological values measured were erythrocyte and leukocyte counts, globular volume and mean corpuscular volume. We considered as parasitized the positive animals for T.cruzi and T. evansi in any of diagnostic tests used. We observed that seven (14.5%) of the crab-eating foxes were parasitized only by T.cruzi, seven by T. evansi, and nine (19%) were coinfected. The parasitized animals by T.cruzi showed a significant increase in leukocyte counts (14.7 x 103) compared to non-parasitized animals (10.4 x 103), parasitized by T. evansi (12.4 x 103) and coinfected (12.9 x 103). We observed a significant difference in relation to mean erythrocyte and mean corpuscular volume values between wet and dry periods: 2.6 x 106 and 165, and 3.4 x 106 and 132, respectively. All presented morphologically unconfirmed macrocytosis.

RESUMO A mandioca tem sido amplamente utilizada para alimentação animal e humana. Também tem sido demonstrado que possui propriedades anti-neoplásicas e anti-helmínticas. A toxicidade devido ao consumo da mandioca tem sido relatada em ruminantes e animais de laboratório. Assim, este estudo teve como objetivo investigar os efeitos tóxicos da acetona cianidrina, um metabólito da linamarina que está presente na mandioca, em ratos Wistar. Foram utilizados seis grupos de cinco animais cada para avaliar os efeitos tóxicos da acetona cianidrina administrada a 25 (G1), 50 (G2), 75 (G3), 100 (G4) e 125 (G5) umol / kg como uma dose oral única. O grupo controle recebeu água acidificada (pH 3,5). Os animais foram monitorados após a administração de acetona cianidrina e os sintomas clínicos foram registrados. Os níveis séricos das enzimas foram medidos para avaliar a função renal e hepática. Durante a necropsia, amostras teciduais foram coletadas para análise histopatológica. Após a administração, alguns animais dos grupos G2, G4 e G5 apresentaram sintomas neurológicos como convulsões, contração muscular involuntária, andar cambaleante, incordenação motora, prostração e midríase. Todos os animais do G5 e quatro animais do G4 morreram após sete minutos da administração de acetona cianidrina. Animais dos outros grupos, particularmente do G2, se recuperaram da fase aguda. A análise bioquímica revelou lesões e disfunção hepáticas. A histopatologia revelou lesões severas no fígado e cérebro. Os resultados permitem inferir que a acetona cianidrina tem efeitos tóxicos no fígado, pulmão e sistema nervoso central de ratos. No entanto, em concentrações de até 25 umol / kg os animais podem sobreviver à fase aguda.

ABSTRACT Cassava has been widely used for animal and human nutrition. It has also been demonstrated to have antineoplastic and anthelmintic properties. Toxicity due to cassava consumption has been reported in ruminants and laboratory animals; therefore, this study aimed to investigate the toxic effects of acetone cyanohydrin, a metabolite of linamarin that is present in cassava, in Wistar rats. Six groups of five animals each were used to evaluate the toxic effects of acetone cyanohydrin administered at 25 (G1), 50 (G2), 75 (G3), 100 (G4) and 125 (G5) µmol/kg as a single oral dose. The control group received acidified water (pH 3.5). The animals were monitored after administration of acetone cyanohydrin, and clinical symptoms were recorded. Serum enzyme levels were measured to assess the kidney and liver function. During necropsy, tissue samples were collected for histopathological examination. After administration, some animals in the G2, G4, and G5 groups presented neurological symptoms such as convulsions, involuntary muscle contraction, staggering gait, motor coordination disability, prostration, and mydriasis. All of the animals in the G5 and four animals in the G4 group died seven minutes after the administration of acetone cyanohydrin. Animals in the other groups, particularly in G2, recovered from the acute phase. Biochemical analysis revealed hepatic lesions and liver dysfunction. Histopathology revealed severe lesions in both the liver and brain. In conclusion, acetone cyanohydrin has toxic effects in the liver, lung, and central nervous system in rats; however, at concentrations up to 25 µmol/kg, the animals could survive the acute phase.

ABSTRACTINTRODUCTION: The transmission cycle of Trypanosoma cruzi in the Brazilian Pantanal region has been studied during the last decade. Although considerable knowledge is available regarding the mammalian hosts infected by T. cruzi in this wetland, no studies have investigated its vectors in this region. This study aimed to investigate the presence of sylvatic triatomine species in different habitats of the Brazilian Pantanal region and to correlate their presence with the occurrences of vertebrate hosts and T. cruzi infection.METHODS: The fieldwork involved passive search by using light traps and Noireau traps and active search by visual inspection. The light traps were placed at five selected points along forested areas for seven nights during each of the nine excursions. At each point where a light trap was set, eight Noireau traps were placed in palm trees and bromeliads.RESULTS: In all, 88 triatomine bugs were collected: two and one individuals from light traps and Noireau traps, respectively; three from peridomestic areas; 23 in coati nests; and 59 in thornbird nests. In this study, active search in microhabitats showed higher efficiency than passive search, since 95% of the triatomine bugs were caught in nests. Further, triatomine bugs were only found to be infected by T. cruzi in coati nests.CONCLUSIONS: Coati nests might act as a point of convergence and dispersion for triatomine bugs and mammal hosts infected by T. cruzi, thereby playing an important role in the sylvatic cycle of T. cruziin the Pantanal region.

O coccídio Caryospora bigenetica foi descrito na serpente Crotalus horridus (Viperidae) nos Estados Unidos da América. Este estudo representa o primeiro registro da ocorrência de C. bigenetica em serpentes da América do Sul. Amostras de fezes foram obtidas, entre novembro de 2013 e maio de 2014, de 256 serpentes silvestres mantidas em um criatório científico nos Estados do Mato Grosso do Sul (MS; n = 214) e Rio de Janeiro (RJ; n = 42), Brasil. Caryospora bigenetica foi encontrada em 14 (5,6%) serpentes, todas pertencentes à família Viperidae. Dez Bothrops moojeni e duas Crotalus durissus de MS estavam infectadas. O coccídio também foi encontrado em uma C. durissus e uma Bothrops jararacussu do Estado do Rio de Janeiro. Os oocistos foram esféricos, com parede dupla, sendo a externa ligeiramente mamilonada, estriações aparentes transversalmente, 13.0 µm (12 – 14); grânulo polar junto à parede interna. Esporocisto oval ou piriforme, 10.0 × 8.0 µm (9 – 11 × 8 – 9); corpo de Stieda discóide; sub-Stieda presente; resíduo do esporocisto presente, formado por um grupo de corpos esféricos entre os esporozoítos. Este estudo aumenta o número de hospedeiros viperídeos de C. bigenetica e expande a distribuição geográfica para a América do Sul.

The coccidian Caryospora bigenetica was first described in the snake Crotalus horridus (Viperidae) from United States of America. This study represents the first record of the occurrence of C. bigenetica in snakes in South America. Feces were sampled between November 2013 and May 2014 from 256 wild snakes maintained in scientific breeding facilities in the states of Mato Grosso do Sul (MS; n = 214) and Rio de Janeiro (RJ; n = 42), Brazil. Caryospora bigenetica was found in 14 (5.6%) snakes, all belonging to the family Viperidae. Ten Bothrops moojeni and two Crotalus durissus from MS were infected. The coccidian was also found in one C. durissus and in one Bothrops jararacussu from the state of RJ. The oocysts were spherical with a double wall, the exterior lightly mammillated, striations apparent in transverse view, 13.0 µm (12 – 14); polar granule fixed in the internal wall. Sporocysts oval or pyriform, 10.0 × 8.0 µm (9 – 11 × 8 – 9); Stieda body discoid; sub-Stieda body present; sporocyst residuum present, formed by a group of spheroid bodies between sporozoites. This study increases the number of viperid hosts of C. bigenetica and expands the geographical distribution to South America.

A infecção simultânea por parasitas de diferentes espécies pode resultar em alterações imprevisíveis. O presente estudo avaliou a patologia de cães naturalmente coinfectados por Leishmania infantum e Ehrlichia canis. A saúde dos cães foi investigada pelas análises histopatológicas, hematológicas e bioquímicas de 21 cães infectados somente por L. infantum e 22 cães coinfectados por L. infantum e E. canis. Observou-se uma reação inflamatória crônica, predominantemente linfohistioplasmocítica, na pele dos dois grupos. A plasmocitose, encontrada nos tecidos linfóides, provavelmente estava relacionada com a hipergamaglobulinemia observada em todos os cães amostrados. A desorganização da matriz extracelular da derme da região inguinal e da orelha, demonstrada pela substituição das fibras de colágeno espessas por fibras finas, foi relacionada com o grau de reação inflamatória, independente da presença de parasitas. Ainda, observamos duas vezes mais animais do grupo coinfectado apresentando formas amastigotas na pele de orelha pela histopatologia comparado ao número de cães infectados apenas por Leishmania, tornando-os desta forma mais infectivos aos vetores. Nossos resultados ressaltam que a saúde de cães coinfectados estava severamente comprometida devido aos altos níveis de proteína plasmática total, globulinas, fosfatase alcalina, creatina quinase e anemia acentuada.

Different parasites that commonly occur concomitantly can influence one another, sometimes with unpredictable effects. We evaluated pathological aspects of dogs naturally co-infected with Leishmania infantum and Ehrlichia canis. The health status of the dogs was investigated based on histopathological, hematological and biochemical analyses of 21 animals infected solely with L. infantum and 22 dogs co- infected with L. infantum and E. canis. The skin of both groups showed chronic, predominantly lymphohistioplasmacytic inflammatory reaction. The plasmacytosis in the lymphoid tissues was likely related with the hypergammaglobulinemia detected in all the dogs. The disorganization of extracellular matrix found in the reticular dermis of the inguinal region and ear, characterized by the substitution of thick collagen fibers for thin fibers, was attributed to the degree of inflammatory reaction, irrespective of the presence of parasites. In addition, the histopathological analysis revealed that twice as many dogs in the co-infected group presented Leishmania amastigotes in the ear skin than those infected solely with Leishmania, increasing the possibility of becoming infected through sand fly vectors. Our findings highlight the fact that the health of dogs infected concomitantly with L. infantum and E. canis is severely compromised due to their high levels of total plasma protein, globulins, alkaline phosphatase and creatine kinase, and severe anemia.

O presente estudo teve como objetivo investigar a ocorrência de anticorpos anti-Toxoplasma gondii, Leishmania infantum e Neospora caninum, em 151 gatos, por meio da Reação de Imunofluorescência Indireta (RIFI). Entre os meses de janeiro e abril de 2013, amostras de soro foram coletadas de 151 gatos domésticos (65 gatos errantes e 86 gatos domiciliados; 55 machos e 96 fêmeas), de Campo Grande, Mato Grosso do Sul, Brasil. Anticorpos IgG anti-T. gondii, anti-L. infantum e anti-N. caninum foram encontrados em 49 (32,5%), 34 (22,5%) e 10 (6,6%) gatos amostrados, respectivamente. Verificou-se uma associação estatisticamente significativa entre as infecções por T. gondii e N. caninum, T. gondii e L. infantum e N. caninum e Leishmania infantum (p <0,05). Além disso, foi observada uma interação significativa entre sexo, área de atividade na probabilidade de infecção por T. gondii (p = 0,0324). No entanto, não foi observada interação significativa entre sexo e área de atividade nas infecções por N. caninum e L. infantum. Este estudo mostrou que os gatos de uma área endêmica brasileira para leishmaniose visceral são expostos a três diferentes protozoários, sendo dois deles importantes agentes zoonóticos.

An investigation was made into the occurrence of antibodies to Toxoplasma gondii, Leishmania infantum and Neospora caninum in 151 domestic cats, based on the indirect fluorescent antibody test (IFAT). Serum samples were collected from 151 domestic cats (65 free-roaming and 86 domiciled cats; 55 males and 96 females) in Campo Grande, Mato Grosso do Sul, Brazil between January and April 2013. IgG antibodies to T. gondii, L. infantum and N. caninum were found, respectively, in 49 (32.5%), 34 (22.5%) and 10 (6.6%) sampled cats. A positive correlation was found between T. gondii and N. caninum, T. gondii and L. infantum, and N. caninum and L. infantum (p <0.05) infections. Also, a significant interaction was identified between gender and area of activity on the probability of T. gondii (p = 0.0324) infection. However, no significant interaction was observed between gender and area of activity on infections by either N. caninum or L. infantum. This study showed that cats from an area endemic for visceral leishmaniasis in Brazil are exposed to three different protozoans, two of which are causal agents of important zoonosis.

Hemoplasmas são bactérias encontradas aderidas aos eritrócitos de felinos. A hemoplasmose felina está frequentemente associada a gatos velhos machos, sem raça definida e com acesso à rua. O presente estudo objetivou realizar a detecção molecular de espécies de hemoplasmas em gatos domiciliados e errantes em Campo Grande, estado do Mato Grosso do Sul (MS), Brasil. Entre janeiro/2013 e abril/2013, amostras de sangue foram colhidas de 151 gatos domésticos (65 errantes e 86 domiciliados) e avaliadas por PCR frente à presença de sequências do gene do 16S rRNA de hemoplasmas, seguidas de sequenciamento, análise pelo BLAST e análise filogenética. Os resultados deste estudo mostraram uma ocorrência de 36,4%. Vinte e três (15,2%) gatos mostraram-se positivos na PCR para ‘Candidatus Mycoplasma haemominutum’, 17 (11,2%) para Mycoplasma haemofelis, e 15 (9,9%) para ‘Candidatus Mycoplasma turicensis’. A co-infecção por dois ou três hemoplasmas ocorreu em 25 gatos (16,6%). Não foi observada diferença estatística significativa entre sexo e estilo de vida dos gatos amostrados e a presença de hemoplasmas. O estudo mostrou que diferentes espécies de hemoplasmas circulam na população de gatos (domiciliados e errantes) na cidade de Campo Grande, MS, Brasil. Sugere-se o diagnóstico diferencial para hemoplasmose felina em gatos que apresentam sinais clínicos inespecíficos de doença com manifestação sistêmica.

Hemoplasmas are bacteria living in feline red blood cells. Feline hemoplasmosis is frequently associated with old male cats that have access to the streets. This study aimed to detect the presence of hemoplasma speciess in domiciled and free-roaming cats in Campo Grande, state of Mato Grosso do Sul (MS), Brazil, using molecular techniques. Between January 2013 and April 2013, EDTA-whole blood samples were collected from 151 domestic cats (65 free-roaming and 86 domiciled cats). Samples were subjected to PCR assays targeting hemoplasmas 16S rRNA, followed by sequencing, BLAST analysis and phylogenetic analysis. Results show an occurrence of 36.4% for hemoplasmas. Twenty-three cats (15.2%) were positive for ‘Candidatus Mycoplasma haemominutum’, 17 (11.2%) for M. haemofelis and 15 (9.9%) for ‘Candidatus M. turicensis’, from PCR. Coinfection by two or three hemoplasmas was found in 25 cats (16.6%). No statistically significant difference between genders or between lifestyles was observed for the presence of hemoplasmas among the cats. Results show different hemoplasma species are present in cat population (Campo Grande, MS, Brazil). It is suggested that a differential diagnosis for feline hemoplasmosis should be made when cats show nonspecific clinical signs of disease with systemic manifestation.

Patógenos transmitidos por carrapatos atingem uma variedade de hospedeiros vertebrados. Para identificar os agentes patogênicos transmitidos por carrapatos entre cães soropositivos para Leishmania infantum no município Campo Grande-MS, foi realizado um estudo sorológico e molecular para a detecção de Ehrlichia canis, Anaplasma platys e Babesia vogeli em 60 amostras de soro e baço, respectivamente. Adicionalmente, foi realizado o diagnóstico confirmatório de L. infantum por meio de técnicas sorológicas e moleculares. Também foi realizado o alinhamento e análise filogenética das sequências para indicar a identidade das espécies de parasitas que infectam esses animais. Anticorpos IgG anti-Ehrlichia spp., anti-B. vogeli e anti-L. infantum foram detectados em 39 (65%), 49 (81,6%) e 60 (100%) dos cães amostrados, respectivamente. Vinte e sete (45%), cinquenta e quatro (90%), cinquenta e três (88,3%), dois (3,3%) e um (1,6%) cães mostraram-se positivos na PCR para E. canis, Leishmania spp., Leishmania donovani complex, Babesia sp. e Anaplasma sp., respectivamente. Após o seqüenciamento, os amplicons mostraram 99% de similaridade com isolados de E. canis, B. vogeli e A. platys e Leishmania chagasi. Os resultados deste estudo indicaram que os cães soropositivos para L. infantum de Campo Grande, MS, são expostos a vários agentes transmitidos por carrapatos, e, portanto, devem ser incluídos no diagnóstico diferencial em cães com suspeita clínica de leishmaniose.

Tick-borne pathogens affect a wide range of vertebrate hosts. To identify tick-borne pathogens among dogs from Campo Grande, MS, Brazil testing seropositive for Leishmania infantum (syn. L. chagasi), a serological and molecular study was conducted to detect Ehrlichia canis, Anaplasma platys and Babesia vogeli in 60 serum and spleen samples. A confirmatory diagnosis of L. infantum based on serological and molecular assays was also performed, as was sequence alignment and phylogenetic analysis to assess the identity of the parasite species infecting these animals. IgG antibodies to Ehrlichia spp., B. vogeli and L. infantum were found, respectively, in 39 (65%), 49 (81.6%) and 60 (100%) of the sampled dogs. Twenty-seven (45%), fifty-four (90%), fifty-three (88.3%), two (3.3%) and one (1.6%) dog were positive, respectively, for E. canis, Leishmania spp., Leishmania donovani complex, Babesia sp. and Anaplasma sp. in PCR assays. After sequencing, the amplicons showed 99% of identity with E. canis, B. vogeli, A. platys and Leishmania chagasi isolates. The findings of this study indicate that L. infantum-seropositive dogs from Campo Grande are exposed to multiple tick-borne pathogens, which should therefore be included in the differential diagnosis of dogs with clinical suspicion of leishmaniasis.
