RESUMO: No Brasil, pelo menos 16 espécies de plantas tóxicas podem afetar o sistema nervoso central dos bovinos. Recentemente no estado do Piauí, no Nordeste do Brasil, Brunfelsia uniflora foi relatada como causadora de sinais nervosos em jumentos, bovinos e pequenos ruminantes. Para avaliar a toxicidade de B. uniflora, 20 camundongos foram distribuídos em quatro grupos de cinco animais recebendo diferentes extratos da planta. Os extratos de alcalóides, flavonóides, saponinas ou solução salina (grupo controle) foram administrados por gavagem. Após a administração dos extratos, em dose única (5g kg-1), todos os animais foram observados diariamente. Os camundongos que receberam os extratos de B. uniflora apresentaram sinais clínicos moderados a graves, incluindo piloeração, vocalização e convulsões. Todos os camundongos que receberam extrato de saponinas morreram entre 10 e 20 minutos após. A avaliação bioquímica sérica dos animais que receberam saponinas, revelou discretos aumentos nos níveis de proteína total e diminuição nos níveis de magnésio e cloro. Conclui-se que, as saponinas presentes nas folhas de B. uniflora induzem efeitos neurológicos tóxicos agudos e, morte em camundongos.

ABSTRACT: In Brazil, at least 16 poisonous plant species can affect the central nervous system of livestock. Recently in the state of Piauí, Northeastern Brazil, Brunfelsia uniflora was reported as a cause of nervous signs in donkeys, cattle, and small ruminants. In order to assess the toxicity of B. uniflora extracts, 20 Swiss mice were distributed into four groups (n=5) receiving by gavage alkaloids, flavonoids, saponins, or saline (control group). After administration of the extracts in a single dose (5g kg-1), all mice were observed daily for clinical signs. Mice that received the extracts showed moderate to severe clinical signs, including piloerection, vocalization, and seizures. All mice dosed with saponins died between 10 and 20min after administration. Serum biochemical evaluation of animals that received saponins revealed slight increases in total protein levels and decreased magnesium and chlorite levels. In conclusion, saponins of B. uniflora leaves induced acute toxic neurological effects and death in mice.

RESUMO: O objetivo deste estudo foi avaliar a atividade acaricida in vitro de Amburana cearensis contra fêmeas ingurgitadas de Rhipicephalus (Boophilus) microplus. O extrato etanólico bruto das folhas de A. cearensis foi submetido à partição com solventes orgânicos (hexano, clorofórmio e acetato de etila) em ordem crescente de polaridade. Foram realizadas três diluições, nas concentrações de 5, 10 e 25mg ml-1. Para avaliar a atividade carrapaticida, fêmeas teleóginas foram submetidas ao teste de imersão de fêmeas ingurgitadas. Os parâmetros analisados foram peso das fêmeas, peso da massa de ovos, porcentagem de eclosão, eficiência reprodutiva, índice de reprodução esperada e eficácia. Para cada extrato, fração e concentração, os testes foram realizados em triplicata. Dentre as frações, a hexânica (2,5%) apresentou os melhores resultados para todos os parâmetros analisados, alcançando 52,7% de inibição da oviposição; 39,0% de porcentagem de eclosão; 3.271 de índice de eficiência reprodutiva; e 67% de eficácia. O extrato obteve uma dose resposta gradual com o aumento da concentração; podendo ser utilizado como terapia auxiliar no controle do R. (B.) microplus.

ABSTRACT: This experiment was carried out to study the bioacaricidal activity of Amburana cearensis against engorged females of Rhipicephalus (Boophilus) microplus. The crude ethanolic extract from the leaves of A. cearensis was submitted to partition with organic solvents (hexane, chloroform and ethyl acetate) in order of increasing polarity; and evaluated using three treatments at concentrations of 5, 10 and 25mg ml-1. To evaluate the acaricidal activity, engorged females were submitted to the adult immersion test. Parameters analyzed were the weight of females; weight of egg mass; percentage of hatch; reproductive efficiency; expected rate of reproduction; and efficacy. For each extract and concentrating fraction, tests were performed in triplicate. Among all fractions, the hexane (2.5%) achieved the best results in all parameters analyzed: 52.7% of inhibition of oviposition; 39% of hatching rate; 3,271 index of reproductive efficiency; and 67.0% effectiveness. The extract obtained gradual dose response with increasing concentration; and could be used as an aid in the control of R. (B.) microplus therapy.

Lawsonia intracellularisé uma bactéria intracelular obrigatória responsável pela enteropatia proliferativa, uma doença entérica endêmica em suínos e comum em potros. Em cães, no entanto, existem poucos estudos sobre essa doença e inexistem relatos de cães infectados por L. intracellularisna América Latina. O objetivo deste estudo foi avaliar a presença de L. intracellularisem amostras de fezes de cães diarréicos e aparentemente saudáveis em Minas Gerais, Brasil. Foram incluídos 58 cães, sendo 18 aparentemente saudáveis e 40 diarréicos. Submeteu-se o DNA bacteriano fecal a uma reação de PCR nested para L. intracellularis. Três das 40 (7,5%) amostras de cães diarréicos foram positivas para L. intracellularis, estando todos esses animais com aproximadamente 3 meses de idade. Estes resultados salientam a necessidade de mais estudos para confirmar o papel deste patógeno como uma possível causa de doença entérica em cães

Lawsonia intracellularisis an obligate intracellular bacterium that is responsible for proliferative enteropathy, an enteric disease endemic in swine and common in foals. However, few studies have investigated this disease in dogs, and there are no reports of dogs infected with L. intracellularisin Latin America. The aim of this study was to evaluate the fecal shedding of L. intracellularisin diarrheic and non-diarrheic dogs in Minas Gerais, Brazil. A total of 58 dogs, 18 apparently healthy and 40 diarrheic, were examined in this study. DNA extracted from feces was analyzed using a nested PCR reaction to detect L. intracellularis.Three out of 40 (7.5%) diarrheic samples, all from 3-month-old puppies, were positive for L. intracellularis. These results highlight the need for additional studies to examine the role of this pathogen as a possible cause of enteric disease in dogs

Clostridium perfringens is a gram-positive, anaerobic, fermentative, spore-forming bacillus that may be found in the environment but is more commonly found as part of the microbiota of humans and animals. The bacterium is considered a common enteric pathogen; however, the pathogenesis and predisposing factors of the disease can differ among species. Therefore, specific studies are necessary for understanding the role of this pathogen, how to diagnose it and which control measures are applicable. The aim of this paper is to review the current knowledge regarding C. perfringens infections in pigs, horses and broiler chickens

O objetivo do presente trabalho foi avaliar a sensibilidade antimicrobiana de estirpes de Clostridium difficile isoladas de animais e humanos no Brasil. Foram utilizados 54 estirpes de C. difficile isoladas de fezes de leitões (n=16), cães (n=13), seres humanos (n=13), potros (n=8), bezerros (n=2), jaguatirica (n=1) e um lobo-guará (n=1). A sensibilidade antimicrobiana foi determinada pelo método de diluição seriada em ágar para penicilina, florfenicol, oxitetraciclina, eritromicina, vancomicina, metronidazol e tilosina. Todos os isolados foram sensíveis ao metronidazol e á vancomicina. Resistência ao florfenicol foi rara, sendo que 52 (96,4%) das estirpes foram sensíveis a esse antimicrobiano. Cinco (9,3%), cinco (9,3%), 14 (25,9%) e 20 (37,0%) foram resistentes a oxitetraciclina, penicilina, tilosina e eritromicina, respectivamente.

The objective of this study was to evaluate antimicrobial susceptibility in Clostridium difficile strains isolated from animals and humans in Brazil. The 54 C. difficile strains used were isolated from stool samples from piglets (n=16), dogs (n=13), humans (n=13), foals (n=8) calves (n=2), an ocelot (n=1) and a maned wolf (n=1). Antimicrobial susceptibility was determined using the serial plate agar dilution method for penicillin, florfenicol, oxytetracycline, erythromycin, vancomycin, metronidazole and tylosin. The C. difficile strains assessed were susceptible to metronidazole and vancomycin. Florfenicol resistance was rarely observed; 52 (96.4%) strains were sensitive to this antimicrobial. Five (9.3%), five (9.3%), 14 (25.9%) and 20 (37.0%) strains were resistant to oxytetracycline, penicillin, tylosin and erythromycin respectively.

O objetivo do presente trabalho foi padronizar um modelo de infecção por Clostridium difficile (ICD) em hamsters sírios (Mesocricetus auratus). Para seleção dos isolados capazes de causar letalidade, cinco animais por grupo receberam uma dose de clindamicina (30mg kg-1) por gavagem. Após 48 horas, administraram-se 107 unidades formadoras de colônia (UFC), por animal, de quatro diferentes isolados toxigênicos de C. difficile. Selecinou-se um dos isolados capazes de causar diarreia e letalidade e administrou-se 4x102; 4x104; 4x106; 4x108UFC por animal, novamente com cinco hamsters por grupo. Em todas as diluições testadas, foi possível observar a ocorrência de diarreia e morte. A maior concentração testada (4x108UFC por animal) causou óbito de 100% dos hamsters do grupo. Todos os animais que vieram a óbito apresentaram tiflite hemorrágica, foram positivos para as toxinas A/B e foi possível isolar C. difficile do conteúdo intestinal, confirmando a reprodução experimental da doença. A dose letal para 50% da população foi estabelecida em 6,3x104UFC por animal. O modelo de indução de ICD em hamsters descritos no presente estudo passa a ser uma ferramenta valiosa para estudos relativos à patogenia, tratamento e controle dessa doença.

The aim of this study was to standardize a model of Clostridium difficile infection (CDI) in Syrian hamsters (Mesocricetus auratus). In order to evaluate strains capable of causing lethality, five hamsters per group received clindamycin (30mg kg-1) by gavage. After 48 hours, 107 colony forming units (CFU) of spores' solution of four strains were administered per animal. One strain capable of causing diarrhea and death was selected and administered at the following concentrations: 4x102; 4x104; 4x106; 4x108 CFU per animal. All dilutions tested were able to cause diarrhea and death. The highest concentration showed 100% of mortality. Post mortem evaluation revealed hemorrhagic typhlitis in all death animals. In addition, all intestinal contents were positive for A/B toxins, and toxigenic C. difficile strains were isolated, confirming the induction of infection by this microorganism. The dose lethal to 50% of the population was calculated: 6.3x104 CFU per animal. The standardized model of CDI in hamster is now available for studies on pathogenesis, treatment and control of this disease.

This study investigated the adjuvant potential of W/O/W multiple emulsions and microemulsions, comparing them with traditional aluminum hydroxide and oil-in-water emulsion adjuvants against bluetongue vaccine (BTV). Local inflammatory reactions were assessed in rabbits by measuring the temperature of the animals and the skin thickness at the site of application. Antibodies titers were determined by serum-neutralization test. Histological analyses of lesions at the site of adjuvants application were done. Results showed that multiple emulsion and microemulsion maintained their stability even in the presence of complex components and presented adequate characteristics for subcutaneous administration. They were able to induce immune response against BTV, but it was smaller than the traditional adjuvants. Despite microemulsion adjuvant showed lower antibodies titre, it was easier to prepare more stable at 4°C and it was the only one that did not induce any local reaction.

Clostridium difficile é considerado um enteropatógeno emergente que acomete humanos e animais domésticos. A presença da doença em equinos e cães já foi relatada no Brasil e trabalhos sugerem que atualmente C. difficile seja um dos principais causadores de diarreia neonatal em suínos no país. Além disso, relatos recentes sugerem a transmissão do agente entre o homem e animais, gerando a hipótese de C. difficile ser um agente zoonótico. Com isso, o presente trabalho tem como objetivo revisar as principais características da doença, além de fornecer dados recentes sobre a ocorrência no Brasil da infecção por C. difficile nas principais espécies de animais domésticos.

Clostridium difficile is an emerging enteropathogen of humans and domestic animals. The bacterium was recently confirmed to be present in foals and dogs in Brazil, with some recent studies suggesting that C. difficile is one of the most important causes of piglet diarrhea in the country. Moreover, some reports also suggest the transmission of this microorganism between animals and humans, raising the possibility that C. difficile is a zoonotic disease. Therefore, the aim of the present review is to describe the main features of C. difficile infection in domestic animals and outline the occurrence of the disease in horses, dogs and pigs in Brazil.

The minimum inhibitory concentration (MIC) was determined for 13 antibiotics against Clostridium perfringens isolated from Brazilian piglets. The collection of isolates was performed in June to October 2010. All isolates were susceptible to amoxicillin and ceftiofur, whereas most were resistant to tetracycline and lincomycin. Avilamycin and narasin were more effective against isolates from non-diarrheic than from diarrheic piglets. The other antimicrobials were less active in need of high concentrations to inhibit the growth of the C. perfringens type A. These results suggest the need for further studies evaluating molecular factors related to the antimicrobial resistance of C. perfringens.

Clostridium difficile tem sido relatado como o principal causador de colite neonatal em suínos. Apesar da crescente importância deste agente, não há dados sobre infecções causadas por C. difficile em suínos no Brasil. O objetivo do presente estudo foi detectar as toxinas A/B e isolar C. difficile a partir de 60 amostras de fezes de leitões diarreicos ou apararentemente saudáveis, com no máximo sete dias de vida, e oriundos de 15 granjas diferentes. As toxinas A/B foram detectadas por ELISA e uma PCR multiplex foi utilizada para detecção dos genes responsáveis pela codificação das toxinas A, B e toxina binária. As toxinas A/B de C. difficile foram detectadas em dez amostras de fezes (16.7%). Dessas, sete eram de animais diarreicos e três de leitões aparentemente saudáveis. Foi possível isolar C. difficile em 12 das 60 (20%) amostras trabalhadas. Dessas, três estirpes eram não-toxigênicas (A-B-) e nove eram toxigênicas, sendo quatro caracterizadas como A+B+ e cinco como A-B+. O gene responsável pela codificação da toxina binária foi encontrado em 50% das estirpes isoladas, proporção superior ao relatado em estudos anteriores. Todas as estirpes não toxigênicas foram isoladas de animais não diarreicos. Os resultados encontrados sugerem a ocorrência de diarreia por C. difficile em granjas no Brasil.

Clostridium difficile has emerged as a major cause of neonatal colitis in piglets, displacing classic bacterial pathogens. However, there is no information regarding the distribution of this microorganism in pig farms in Brazil. In the present study, the presence of toxins A/B and of C. difficile strains in stool samples from 60 diarrheic or non-diarrheic newborn piglets (one to seven days old), from 15 different farms, was studied. The presence of toxins A/B was detected by ELISA and PCR was used to identify toxin A, toxin B and binary toxin gene in each isolated strain. C. difficile A/B toxins were detected in ten samples (16.7%). Of these, seven were from diarrheic and three were from non-diarrheic piglets. C. difficile was recovered from 12 out of 60 (20%) fecal samples. Of those, three strains were non-toxigenic (A-B-) and nine were toxigenic. Of the nine toxigenic strains, four were A+B+ strains and five were A-B+ strains. The presence of binary toxin observed in the present study was much higher (50%) than in previously reported studies. All three non-toxigenic strains were isolated from otherwise healthy piglets. The results suggest the occurrence of neonatal diarrhea by C. difficile in farms in Brazil.

A síndrome da dilatação intestinal suína (PIDS) é causa importante de morte súbita em suínos em fase de crescimento e terminação. Acomete animais aparentemente saudáveis, em boa conformação corporal e sem histórico prévio de doença gastrointestinal, cursa com intensa dilatação abdominal aguda, de evolução rápida, levando à dispnéia e morte por asfixia em até duas horas. A patogênese não está plenamente esclarecida, porém há hipóteses sobre fatores de risco associados ao manejo, aos hábitos alimentares e tipo de alimentação fornecida aos suínos. São discutidos nesta revisão os principais fatores predisponentes para a ocorrência de PIDS, sua fisiopatologia e o diagnóstico adequado dessa afecção. As causas de morte súbita devem ser devidamente avaliadas e os fatores predisponentes à PIDS devem ser determinados para melhor efetividade de diagnóstico e controle de mortalidade.

The porcine intestinal distension syndrome (PIDS) is a major cause of sudden death in growing and finishing pigs. It affects apparently healthy animals in good body conformation and without prior evidence of gastrointestinal disease. PIDS presents as an acute intense abdominal distension with rapid evolution, leading to respiratory distress and death by asphyxiation within two hours. The pathogenesis is not fully understood, but there are a few hypotheses about risk factors associated with management, feed intake and type of feed offered to the pigs. In this review, the main predisposing factors for the occurrence of PIDS, its physiopathology and the proper diagnosis of this condition are discussed. Causes of sudden death must be properly assessed and risk factors must be determined for better effectiveness of diagnosis and mortality control.

Este experimento foi realizado com o objetivo de testar a eficiência da leucomicina no controle da EPS. Para isso, utilizaram-se 90 leitões de cinco semanas de idade, provenientes de granja sem histórico clínico de EPS. Os animais foram divididos em três grupos (tratamentos) de 30, sendo T1 o grupo dos inoculados e não medicados, T2 e T3 os inoculados e medicados com 90ppm e 180ppm de leucomicina, respectivamente. No dia zero do experimento, os 90 suínos foram inoculados com aproximadamente 4,5x10(9) L. intracellularis. A medicação foi utilizada nas rações dos grupos T2 e T3 somente do dia anterior até 13 dias após a inoculação com L. intracellularis. Ganho de peso, consumo de ração e sinais clínicos foram avaliados durante todo o experimento. Todos os leitões foram eutanasiados 23 dias pós-inoculação (pi). Os sinais clínicos foram mais evidentes nos animais do grupo T1 que nos do T2 e do T3 entre os dias 16 e 21. Ganho de peso diário, consumo diário de ração e conversão alimentar foram melhores nos grupos medicados com leucomicina. A extensão total das lesões intestinais por grupo (T1, T2 e T3) foram de 869, 473 e 331cm, respectivamente. A maioria dos animais (84,4%) teve marcação positiva para L. intracellularis à IHC nas seções de íleo. Leucomicina nas doses de 90 e 180ppm por 14 dias foi eficiente para a melhora do desempenho de leitões inoculados com homogeneizado intestinal contendo L. intracellularis.

The aim of this study was to evaluate the efficacy of orally administered leucomycin at 90 and 180ppm for the prevention of porcine proliferative enteropathy (PPE) in experimentally infected pigs. A total of 90 commercial five-week-old pigs were randomly assigned to receive leucomycin in feeding at 90 (T2), 180ppm (T3), or untreated (T1). All animals in the treated groups received medicated feed for 14 days starting one day before inoculation. Each pig was inoculated intragastrically with approximately 4.5x10(9) Lawsonia intracellularis in the form of porcine intestinal mucosal homogenate. Body weight, feed consumption and clinic signs were evaluated throughout the study. Necropsies and gross evaluation of intestines were performed in all animals on day 23 post-inoculation (pi) or at death, and ileum samples were collected for immunohistochemistry (IHC) for L. intracellularis. Clinical presentation of the disease was more evident in the non-medicated group (T1) than in the medicated ones (T2, T3) between days 16 and 21pi. Average daily gain, average daily feed consumption and feed conversion efficiency were better in groups treated with either dose of leucomycin. The total intestine lesion length per group (T1, T2 and T3) was 869, 473 and 331cm, respectively. The majority of the animals (84.4%) were positive for L. intracellularis antigen in ileum sections stained by IHC. Under the conditions of this study, leucomycin administered in feed at 90 and 180ppm for 14 days was effective in improving performance of pigs inoculated with intestinal homogenate containing L. intracellularis.

A maioria dos estudos epidemiológicos envolvendo Haemophilus parasuis isolados de rebanhos suínos brasileiros se baseia em sorotipagem. Entretanto, uma alta porcentagem de amostras não é sorotipável. Técnicas moleculares têm sido utilizadas com sucesso para caracterizar a diversidade dos isolados de H. parasuis e a epidemiologia das infecções por esse agente dentro e entre rebanhos. Esta revisão enfoca aspectos gerais da infecção por H. parasuis, principalmente em relação às técnicas de epidemiologia molecular.

The majority of epidemiological studies involving Haemophilus parasuis from Brazilian herds are based on serotyping. However, a high percentage of isolates are non-typable. Recently, molecular-based techniques were successfully used to characterize the diversity of H. parasuis isolates and the epidemiology of H. parasuis infections within and among herds. This review tackles general aspects of H. parasuis infection, mainly regarding to techniques of molecular epidemiology.

A diarréia é a principal manifestação clínica das infecções entéricas. O desequilíbrio entre os processos de absorção e secreção intestinal decorrentes de alterações no transporte de água e eletrólitos é determinante na patogênese dos processos diarréicos. Os mecanismos fisiopatológicos estão associados às características particulares de cada patógeno. Dessa forma, a interação específica entre os patógenos entéricos e o epitélio intestinal resulta em diferentes tipos de diarréias. Na produção de suínos, o conhecimento da fisiopatologia das infecções entéricas é fundamental no desenvolvimento de métodos de diagnóstico e, em um futuro próximo, na elaboração de estratégias de prevenção e controle dessas afecções. Com base nos processos fisiológicos característicos do trato gastrointestinal, esta revisão tem como objetivo descrever aspectos relevantes e atuais dos mecanismos fisiopatológicos das diarréias em suínos. Serão enfatizadas as alterações patológicas no transporte intestinal de íons e nutrientes decorrentes da ação de patógenos entéricos importantes na produção suinícola.

Diarrhea is the main clinical manifestation of enteric infections. Imbalance between absorption and intestinal secretion process due to changes in the water and electrolytes transport is critical in the diarrheal diseases pathogenesis. The pathophysiological mechanisms are associated to specific characteristics of each pathogen. Therefore, specific interaction between enteric pathogens and intestinal epithelium result in different types of diarrhea. In swine production, the knowledge about the enteric infections pathophysiology is important for the development of diagnostic methods and, in a near future improvement of prevention and control strategies. The aim of this review is to describe different relevant aspects of the swine diarrhea pathophysiology based on the physiological processes typical of gastrointestinal tract. Pathological changes in the intestinal transport of ions and nutrients due to infectious relevant for swine production will be emphasized.
