The Xingu River crosses several indigenous territories along its course, among which are one of the most extensive and populous in Brazil. There lives Mebêngôkre (Kayapó) people, who speak the language of the same name, from the Jê family, Macro-Jê trunk. Its reserves occupy areas covering the states of Mato Grosso and Pará. Among them is the Kayapó TI with more than 30 schools in the region, but the Mebêngôkre still suffer the typical pressures of minority languages. In 2019, the language was made co-official in São Félix do Xingu (PA). This fact has not yet changed the local linguistic reality. In order to admit and forward the demands of the Mebêngôkre regarding their linguistic rights, the Extension Project Actions to Combat Linguistic Prejudice (UFRJ), in partnership with the municipality's Department of Indigenous School Education, created the action "Me kuni umari — in network for the rights language”. That is the experience reported here.

O Rio Xingu atravessa diversos territórios indígenas ao longo de seu curso, dentre os quais, estão um dos mais extensos e populosos no Brasil. Ali vive o povo Mebêngôkre (Kayapó), que fala a língua de mesmo nome, da família Jê, tronco Macro-Jê. Suas reservas ocupam áreas que abragem os estados do Mato Grosso e Pará. Entre elas está a TI Kayapó com mais de 30 escolas na região, mas os Mebêngôkre sofrem as pressões típicas de línguas minorizadas. Em 2019, a língua foi cooficializada em São Félix do Xingu (PA), mas esse fato ainda não modificou a contento a realidade linguística local. Para recolher e encaminhar as demandas dos Mebêngôkre quanto a seus direitos linguísticos, o Projeto de Extensão Ações de Combate ao Preconceito Linguístico (UFRJ), em parceria com o Departamento de Educação Escolar Indígena do município, criaram a ação “Me kuni umari — em rede pelos direitos linguísticos”, da qual trata este relato de experiência.

Mari-mari (Cassia leiandra Benth.) is an Amazonian native fruit that is highly appreciated in the northern region of Brazil, but there are few studies regarding its chemical composition and/or biological properties. A phytochemical study was conducted on the extracts of the pulp, peel and seeds; and their antioxidant, cytotoxic and antimicrobial potential were also determined. Methanol and acetone extracts from different fruit parts were subjected to total phenolic, 2,2-diphenyl-1 picrylhydrazyl (DPPH) radical scavenging and FeIII reducing capacity (FRAP) assays. From the peel extracts, the engeletin and astilbin could also be isolated and characterized based on nuclear magnetic resonance spectroscopy and high-resolution mass spectrometry. A quantification using ultra-fast liquid chromatography of acetone dry crude extracts (DCE) of the peel showed that engeletin was the main compound (532.77 mg g-1 DCE). Astilbin content was also determined (131.53 mg g-1 DCE). The antibacterial and antiproliferative activities were evaluated and the methanol extract of the peel showed higher growth inhibition activity against all tested bacteria. Engeletin showed a relevant antimicrobial effect against Staphylococcus aureus, Staphylococcus epidermidis and Escherichia coli. Acetone and methanol peel extracts exhibited a relevant inhibitory effect on the proliferation of breast carcinoma cells (MCF-7), with no toxicity to the normal cell line (NIH/3T3).

RESUMO: O pequi (Caryocar spp.) é um fruto nativo do cerrado brasileiro e Caryocar brasiliense Camb. é uma das espécies mais prevalentes neste bioma. O consumo da polpa de pequi tem sido associado a benefícios à saúde, como efeitos antioxidantes, anti-inflamatórios, hipolipidêmicos, hepatoprotetores e anticarcinogênicos. Esses benefícios têm sido atribuídos à sua alta densidade em nutrientes e compostos bioativos. No entanto, há evidências de considerável variação no perfil nutricional entre polpas de pequi de diferentes espécies, e entre polpas da mesma espécie, mas de diferentes áreas do cerrado. Além disso, não há informações na literatura sobre o perfil de polifenóis da polpa de C. brasiliense, tampouco sobre a influência da área nativa do fruto na composição desses fitoquímicos. Assim, este estudo analisou a composição de nutrientes e identificou os compostos fenólicos na polpa de frutos de C. brasiliense nativos de quatro diferentes áreas do Cerrado. Houve notável variação nos teores de energia (176,3-387,2 kcal/100 g) e lipídios (13,0-37,5 g/100 g) entre as amostras. Em contrapartida, não foi encontrada diferença na composição em polifenóis, uma vez que ácido gálico, ácido protocatecuico, catequina, epicatequina, ácido p-cumárico e ácido elágico foram identificados em todas as polpas de pequi analisadas. A polpa de C. brasiliense tem potencial para ser utilizada como ingrediente funcional rico em compostos bioativos, porém com diferentes teores de energia e lipídios de acordo com a área nativa do fruto para atender a distintas alegações de saúde do produto.

ABSTRACT: Pequi (Caryocar spp.) is a fruit native to the Brazilian Savannah (Cerrado), and Caryocar brasilienseCamb. is one of the most prevalent species in this biome. The consumption of the pequi pulp has been associated with health benefits, such as antioxidant, anti-inflammatory, hypolipidemic, hepatoprotective, and anticarcinogenic effects. These benefits have been attributed to its high density in nutrients and bioactive compounds. However, there is evidence about considerable variation in the nutritional profile among pequi pulps of different species, and among pulps of the same species, but from different areas of the Cerrado. In addition, there is no information in literature regarding the polyphenols profile of C. brasiliense pulp, neither about the influence of the native area of the fruit on the composition of these phytochemicals. So, this study analyzed the nutrient composition and identified the phenolic compounds in the pulp of C. brasiliense fruits native to four different areas of the Cerrado. There was a remarkable variation in the contents of energy (176.3-387.2 kcal/100 g) and lipids (13.0-37.5 g/100 g) among samples. In contrast, no difference was observed in the polyphenols composition, since gallic acid, protocatechuic acid, catechin, epicatechin, p-coumaric acid, and ellagic acid were identified in all pequi pulps. C. brasiliense pulp shows potential to be used as a functional ingredient rich in bioactive compounds, but with different contents of energy and lipids according to the fruit’s native area to attempt distinct health allegations of the product.

ABSTRACT Microbial contamination of the wort during the fermentation process causes significant losses in ethanol production worldwide and creates a dependence of the industry on chemicals and antibiotics to control contamination. Therefore, this study used electron beam (e-beam) to disinfect wort from sugarcane (Saccharum officinarum L.) molasses and investigate the bioethanol fermentation. Four treatments (T0 – T3) were carried out using ionizing doses of radiation through the electron accelerator: 0 (control), 10, 20, and 40 kGy. Total mesophiles, total bacteria, sucrose, glucose, fructose, phenolics, flavonoids, hydroxymethylfurfural (5-HMF), and Furfural were measured. An alcoholic fermentation assay was performed after the irradiation process. The irradiated treatments showed no inversion of sugars and formation of the inhibitory by-products flavonoids, furfural and 5-HMF, except for the phenolic compounds. The lower dose tested (10 kGy) reduced more than 99.9 % of the total mesophiles and more than 99.99 % of the total bacteria in the substrate. In the fermentation, the irradiated worts presented similar (p > 0.05) yields (92, 93, and 94 %) and ethanol productivity levels (0.89, 0.88, and 0.87 g L–1 h–1, for T1, T2, and T3 respectively). However, all treatments presented higher yields and productivity (p < 0.05) when compared to the control (88 % and 0.85 g L–1 h–1), highlighting the possible use of e-beam in wort fermentation at a lower dose (10 kGy). This allows reduction in losses caused by microbial contamination, besides increasing fermentation yield and productivity with lower energy consumption.

ABSTRACT Introduction: Participants in cardiac rehabilitation programs have low adherence to their sessions, which makes extremely important to recognize the barriers that cause non-adherence, identifying whether the type of service and level of adherence influence these barriers. Methods: This is a cross-sectional observational study, in which 220 individuals (66.80±11.59 years) of both genders who are members of public and private exercise-based cardiac rehabilitation programs participated. The volunteers were divided according to the level of adherence, considering patients with low adherence (PLA) those with < 70% of attendance and high adherence (PHA) those with > 70%. Then, initial evaluation, Cardiac Rehabilitation Barriers Scale, analysis of socioeconomic level, Hospital Anxiety and Depression Scale, and Mini-Mental State Examination were applied. Results: Higher total barriers were found in PLA in the public service compared to PHA in the private service (P=0.023). In the subscale “perceived need”, PHA in the public service showed higher values than PLA and PHA in the private service (P≤0.001). The “access” barrier was higher for PHA in the public service when compared to PHA in the private service (P=0.024). PHA in the public service exhibited a higher barrier regarding questions about distance, transportation problems, cost, and time constraints. Conclusion: The public program presents higher barriers in the questions and categories compared to the private program, mainly the PHA. Furthermore, there are differences in the profile of the participants regarding socioeconomic and anxiety levels, treatment time, ethnicity, and city where they live.

Abstract Austrodiplostomum Szidat & Nani, 1951 is a genus of parasitic digenetic trematodes widely distributed in the Neotropical region. Infects a wide variety of species, families and requests for freshwater fish. We identify samples of Austrodiplostomum sp, based on metacercariae isolates from eyes of tambaqui (Colossoma macropomum), a fish of high commercial importance in Brazil, and widely consumed by the population of the northern region. The sequences obtained clustered with A. compactum. This is the first report of the occurrence of diplostomids in farmed tambaqui in Amazonia.

RESUMO Objetivos: construir e validar instrumento relacionado à prevenção de lesão por pressão para avaliação do conhecimento, da atitude e da prática de cuidadores de idosos institucionalizados. Métodos: estudo metodológico realizado em três fases: construção do instrumento, análise dos juízes e análise semântica e aparente, com 78 participantes, seguindo as etapas do processo de validação de instrumentos psicométricos para os critérios clareza e pertinência. Resultados: na rodada Delphi I, o índice de validade de conteúdo global do critério “clareza” foi de 0,66, “pertinência” 0,85 e valor de Kappa > 0,76. Em Delphi II, o critério de “clareza” foi de 0,95, “pertinência” 1,00 e valor de Kappa > 0,97. Conclusões: dispõe-se de instrumento válido quanto a conteúdo e aparência para dar seguimento à análise da sua confiabilidade em medir os construtos aos quais se propõe, para que possa servir de ferramenta para gestão do cuidado na prevenção de lesão por pressão.

ABSTRACT Objectives: to construct and validate an instrument to assess the knowledge, attitudes, and practices related to pressure injury prevention among caregivers of institutionalized older people. Methods: this is a three-stage methodological study that consisted of instrument construction, analysis by experts, and semantic and appearance analysis, with 78 participants, observing the validation process steps for psychometric instruments in the criteria of clarity and relevance. Results: in the Delphi I round, the validity index of the general content in the clarity criterion was 0.66, in relevance 0.85, and the Kappa value was >0.76. In Delphi II, clarity was 0.95, relevance 1.00, and the Kappa value was >0.97. Conclusions: this is a valid instrument in terms of content and appearance, which allows further analysis of its reliability for the measurement of the constructs for which it is intended. Therefore, it can be considered a tool for care management in pressure injury prevention.

RESUMEN Objetivos: construir y validar un instrumento pera evaluación del conocimiento, actitud y práctica relacionado a la prevención de lesiones por presión entre cuidadores de ancianos institucionalizados. Métodos: estudio metodológico realizado en tres fases: construcción del instrumento, análisis de los evaluadores y análisis semántico y de apariencia, con 78 participantes siguiendo las etapas del proceso de validación de instrumentos psicométricos para los criterios claridad y pertinencia. Resultados: en la ronda Delphi I el índice de validez de contenido general del criterio “claridad” fue de 0,66, “pertinencia” 0,85, y valor de Kappa > 0,76. En Delphi II, el criterio de “claridad” fue 0,95, “pertinencia” 1,00 y valor de Kappa > 0,97. Conclusiones: se tiene un instrumento válido respecto de contenidos y apariencia, para continuar analizando su confiabilidad para medir los constructos a los cuales se propone, para que sirva como herramienta de gestión del cuidado en la prevención de lesiones por presión.

Abstract The quality of the raw material for chocolate manufacture depends on the control of cacao seed fermentation. Practices affect the diversity and metabolic activities of spontaneous yeasts in fermentations. The control of the initial inoculum is important to standardize fermentations and to improve the final seed quality. We conducted fermentations using non-cleaned or cleaned fermentation boxes to evaluate the dynamics of the microorganism populations and their effect in seed composition in two consecutive years. The isolated yeasts were identified by sequencing the D1-domain of the 26S ribosomal genes. Pulp and cotyledon parameters were monitored during fermentations. Wickerhamomyces anomalus was the dominant yeast in the fermentations, but the species was less represented in cleaned boxes. In the cleaned-box fermentation, there was a delay in temperature rise of the fermentation, which retarded the suitable moment for seed-mass turning. The reduction in seed-pulp quality caused by restricted rainfall in one year limited the seed-mass heating and voided the seed turning. Thus, under substrate restriction, the initial reduced inoculum caused by cleaning the boxes impaired fermentation, mainly under environmental conditions that compromise the quality of the pulp. However, this variation had little effect on the desired degradation of cotyledon components, substrates for flavor precursors.

Abstract Based on a holistic concept of polymicrobial etiology, we have hypothesized that putative and candidate periodontal pathogens are more frequently detected in consortia than alone in advanced forms of periodontal diseases (PD). Objective To correlate specific consortia of periodontal pathogens with clinical periodontal status and severity of periodontitis. Methodology Subgingival biofilm was obtained from individuals with periodontal health (113, PH), gingivitis (91, G), and periodontitis (209, P). Genomic DNA was purified and the species Aggregatibacter actinomycetemcomitans (Aa), Aa JP2-like strain, Porphyromonas gingivalis (Pg), Dialister pneumosintes (Dp), and Filifactor alocis (Fa) were detected by PCR. Configural frequency and logistic regression analyses were performed to correlate microbial consortia and PD. Results Aa + Pg in the presence of Dp (phi=0.240; χ2=11.9, p<0.01), as well as Aa JP2 + Dp + Fa (phi=0.186, χ2=4.6, p<0.05) were significantly more associated in advanced stages of P. The consortium Aa + Fa + Dp was strongly associated with deep pocketing and inflammation (p<0.001). The best predictors of disease severity (80% accuracy) included older age (OR 1.11 [95% CI 1.07 – 1.15], p<0.001), Black/African-American ancestry (OR 1.89 [95% CI 1.19 – 2.99], p=0.007), and high frequency of Aa + Pg + Dp (OR 3.04 [95% CI 1.49 – 6.22], p=0.002). Conclusion Specific microbial consortia of putative and novel periodontal pathogens, associated with demographic parameters, correlate with severe periodontitis, supporting the multifactorial nature of PD.

RESUMO: Em programas de melhoramento genético, o uso de informações moleculares garantiu importantes avanços para a melhoria de características de interesse econômico, no âmbito da produção animal. O advento da tecnologia de painéis de SNPs aplicados à seleção genômica ampla (GWS) e associação genômica ampla (GWAS), aliado ao avanço computacional, com o uso de softwares e análises robustas, permitiram melhor compreensão sobre a arquitetura genética dos animais de produção e, consequentemente, maior eficiência na seleção. Nesse contexto, o método estatístico single-step GBLUP tem sido utilizado, frequentemente, na execução da GWS e, mais recentemente, em GWAS, possibilitando predições acuradas e detecção de QTLs, respectivamente. No entanto, em países em desenvolvimento e, em espécies como os ovinos e caprinos, que existe maior dificuldade para a aquisição de dados genômicos, o uso da simulação de dados tem se mostrado eficiente para estudar os principais fatores envolvidos no processo de seleção, como o tamanho da população de treinamento, densidade de chipde SNPs e estratégias de genotipagem, cujos efeitos estão diretamente associados à acurácia da predição de valores genéticos genômicos. Nesta revisão, serão abordados pontos importantes sobre o uso da genômica no melhoramento genético de características produtivas em animais, principais métodos de predição e estimação de efeitos de marcadores moleculares na atualidade, a importância da simulação de dados para a validação desses métodos, bem como as vantagens, os desafios e as limitações no cenário atual da produção animal com o uso da seleção e associação genômica ampla.

ABSTRACT: The use of molecular information in breeding programs contributed to important advances in the improvement of traits of economic interest in livestock production. The advent of single nucleotide polymorphism (SNP) panels applied to genome-wide selection (GWS) and genome-wide association studies (GWAS), along with computational advances (e.g., use of powerful software and robust analyses) allowed a better understanding of the genetic architecture of farm animals and increased the selection efficiency. In this context, the statistic method single-step GBLUP has been frequently used to perform GWS, and more recently GWAS analyses, providing accurate predictions and QTL detection, respectively. Nevertheless, in developing countries, species such as sheep and goats, whose genomic data are more difficult to be obtained, the use of data simulation has been efficient in the study of the major factors involved in the selection process, such as size of training population, density of SNP chips, and genotyping strategies. The effects of these factors are directly associated with the prediction accuracy of genomic breeding values. In this review we showed important aspects of the use of genomics in the genetic improvement of production traits of animals, the main methods currently used for prediction and estimation of molecular marker effects, the importance of data simulation for validation of those methods, as well as the advantages, challenges and limitations of the use of GWS and GWAS in the current scenario of livestock production.

RESUMO: A avaliação da qualidade na e da Educação Infantil constitui-se como um campo de conhecimentos e práticas mobilizadores de aspectos estruturantes para a área, que transitam da competência dos diferentes agentes nos processos avaliativos aos seus efeitos na efetivação da política pública. Nos marcos de uma avaliação alinhada à promoção da qualidade e no reconhecimento da Educação Infantil como uma política pública submetida ao controle social, propõe-se a discussão sobre as potencialidades e tensões presentes na avaliação mista, um modelo de avaliação que combina procedimentos e instrumentos internos e externos às instituições. A discussão parte de uma experiência de avaliação mista realizada junto com a equipe técnica do Núcleo de Assessoria Técnica (NAT) do Ministério Público do Estado de São Paulo (MPSP), no âmbito do Grupo de Atuação Especial de Educação - Núcleo Ribeirão Preto (GEDUC-RP). Os resultados indicam que a avaliação mista foi potente devido ao seu formato dialógico: a avaliação externa possibilitou um olhar técnico e multiprofissional alinhado às proposições da política pública de qualidade; a avaliação interna promoveu uma autorreflexão coletiva da qualidade, mas evidenciou poderes diferenciados e fragilidades no que se refere ao olhar crítico de algumas instituições. Conclui-se que é no cruzamento destes olhares que se criam possibilidades de superação dos limites de cada tipo de avaliação e também convida à reflexão sobre os desafios que se instauram na dinâmica das relações entre as pessoas e órgãos públicos.

ABSTRACT: The evaluation of the quality of Early Childhood Education constitutes a field of knowledge and practices that mobilize structural aspects into the area considering the competence of the different agents in the evaluation processes and their effects in the effectiveness of public policy. In the framework of an evaluation aligned with the promotion of quality and the recognition of Early Childhood Education as a public policy subordinate to social control, we propose to discuss the potentialities and tensions present in the mixed-method evaluation, a model that combines internal and external procedures and instruments inside and outside of institutions. The discussion starts from a mixed-method evaluation experience carried out in conjunction with the technical team of the Public Ministry of the State of São Paulo (MPSP), in the Special Education Group of Ribeirão Preto. The results indicate that the mixed evaluation was powerful because of its dialogic format: the external evaluation allowed a technical and multiprofessional look aligned with the public quality policy; the internal evaluation facilitated a collective self-reflection on quality but showed different strengths and weaknesses in terms of critical views. By combining these perspectives, possibilities are created for overcoming the limitations of each type of evaluation, and also for reflecting on the dynamics of relationships between individuals and public agencies.

RESUMEN: La evaluación de la calidad en y de la Educación Infantil constituye un campo de conocimientos y prácticas que movilizan aspectos estructurales para el área, que pasan de la competencia de los diferentes agentes en los procesos de evaluación a sus efectos en la efectividad de las políticas públicas. En el marco de una evaluación alineada con la promoción de la calidad y en el reconocimiento de la Educación Infantil como política pública sujeta a control social, proponemos discutir las potencialidades y tensiones presentes en la evaluación de método mixto, un modelo de evaluación que conjuga procedimientos e instrumentos internos y externos a las instituciones. La discusión es parte de una experiencia de evaluación de método mixto realizada en conjunto con el equipo de Asesoramiento Técnico del Ministerio Público del Estado de São Paulo (MPSP), en el Grupo de Educación Especial de Ribeirão Preto. Los resultados indican que la evaluación mixta fue poderosa por su formato dialógico: la evaluación externa permitió una mirada técnica y multiprofesional alineada con los planteamientos de la política pública de calidad; la evaluación interna promovió una autorreflexión colectiva sobre la calidad, pero mostró diferentes facultades y debilidades en cuanto a la visión crítica de algunas instituciones. Se concluye que es en la intersección de estas perspectivas que se crean posibilidades para superar los límites de cada tipo de evaluación y también invita a reflexionar sobre los desafíos que se presentan en la dinámica de las relaciones entre las personas y los organismos públicos.

RESUMO Na experimentação agrícola, a utilização do tamanho ótimo de parcela é uma estratégia importante, para aumentar a precisão experimental, e trabalhos desta natureza são escassos para a cultura do grão-de-bico, leguminosa que vem conquistando o mercado e consumidores em todo o mundo. Assim, o objetivo deste trabalho foi determinar o tamanho ótimo de parcela, para avaliar experimentos com a cultura do grão-de-bico, em cenários formados por combinações de números de tratamentos, números de repetições e níveis de precisão. Dois experimentos em branco foram realizados, compostos por oito linhas de cultivo, de 7 m de comprimento, espaçadas em 50 cm entre linhas e 10 cm entre plantas; foram avaliadas as seis linhas centrais, totalizando 60 plantas por linha de cultivo. Foi determinado o índice de heterogeneidade do solo e o tamanho ótimo de parcela foi estimado por meio do método de Hatheway, em cenários formados pelas combinações de i tratamentos (i = 4, 8, 12 e 16), r repetições (r = 3, 4, 5, 6, 7 e 8) e níveis de precisão (DMS = 25%, 30% e 40%). O índice de heterogeneidade do solo foi superior a 0,7 para todas as variáveis avaliadas. O método de Hatheway possibilita estimar diferentes tamanhos de parcelas, de acordo com as condições e limitações da área experimental. Em experimentos com o grão-de-bico, com 4 a 16 tratamentos, DMS de 25% e seis repetições, as parcelas de 25 unidades básicas são suficientes para identificar, com probabilidade de 5%, diferenças significativas entre as médias dos tratamentos.

ABSTRACT In agricultural experimentation, use of the optimum plot size is an important way of increasing experimental precision; however, studies of this type are scarce for the chickpea, a legume that has been conquering both the market and consumers throughout the world. The aim of this study, therefore, was to determine the optimum plot size for evaluating experiments with the chickpea, in scenarios comprising combinations of the number of treatments, number of replications and levels of precision. Two blank experiments were carried out, comprising eight crop rows, 7 m in length, at a spacing of 50 cm between rows and 10 cm between plants; the six central rows were evaluated, giving a total of 60 plants per row. The index of soil heterogeneity was determined, and the optimum plot size was estimated using the Hatheway method in scenarios formed by a combination of i treatments (i = 4, 8, 12 and 16), r replications (r = 3, 4, 5, 6, 7 and 8) and levels of precision (LSD = 25%, 30% and 40%). The index of soil heterogeneity was greater than 0.7 for each of the variables under evaluation. The Hatheway method makes it possible to estimate different plot sizes based on the conditions and limitations of the experimental area. In experiments with the chickpea including 4 to 16 treatments, 25% LSD and six replications, plots of 25 basic units are sufficient to identify significant differences between the mean values of the treatments with a probability of 5%.

Abstract Pitaya is a rich source of bioactive compounds, such as polyphenols and betalains. Moreover, these compounds has been associated with reduced risk of type 2 diabetes mellitus. The aim of this study is to evaluate the effect of this fruit on glycemia and oxidative stress in aloxan-induced diabetic mice. Thus, considering that In the in vivo assays, the mice were divided into 5 groups (n = 6): (a) healthy group treated with water; (b) diabetic mice treated with metformin at 200 mg/kg body weight diluted in water; (c) untreated diabetic mice; (d) diabetic mice treated with pitaya at 200 mg/kg and (e) diabetic mice treated with pitaya at 400 mg/kg of body weight diluted in water. The results shows that pitaya was able significantly to reduce blood glucose (p < 0.05) (200 mg/kg); significantly reduce cholesterol (200 and 400 mg/kg) and significantly increase HDL-c (400 mg/kg) levels. In the oxidative stress experiment, Malondialdehyde levels in the liver were significantly reduced (p < 0.05) in the groups treated with pitaya, when compared to the other groups, suggesting lower lipid peroxidation. The consumption of pitaya reduced the blood glucose and cholesterol and increase the HDL, in addition, the lipid peroxidation - which is common in diabetic patients, was reduced.

ABSTRACT: Given the need for alternative therapies for bovine mastitis, an infectious disease of high prevalence and significant economic impact in dairy farms, this study evaluated the in vivo antimicrobial efficacy of an intramammary formulation based on soluble polypyrrole (PPy), a promising conductive polymer for biomedical applications, especially as an effective antimicrobial agent. In this assay, mammary quarters of healthy Holstein Friesian cows (n = 8) were inoculated with Staphylococcus aureus and treated, via the intramammary route, with an experimental formulation based on soluble PPy (5%) and a commercial formulation based on gentamicin sulfate. The effect of these treatments was evaluated based on milk cultures, Total Bacterial Count (TBC), Somatic Cell Count (SCC), and milk composition analysis of mammary quarter samples in seven experimental moments. In addition, hematological evaluations of the animals were also performed. The intramammary application of three levels of the experimental formulation based on soluble PPy resulted in higher log/mL of the TBC and SCC compared to a positive control group and a group that received gentamicin sulfate. The experimental formulation did not induce changes in milk composition or in hematological parameters. Certain factors related to pharmacokinetics, such as the type of carrier used in the formulation and the pharmacodynamics of soluble PPy, may have contributed to the antimicrobial ineffectiveness of the experimental formulation. The results of this study neither define nor decrease the antimicrobial potential of soluble PPy. Further research is required to develop formulations that enable the antibacterial action of soluble PPy in the intramammary environment of dairy cows.

RESUMO: Considerando a necessidade de terapias alternativas para a mastite bovina, uma inflamação, normalmente de causa infecciosa, de alta prevalência e de alto impacto econômico nas fazendas leiteiras, avaliou-se a eficácia antimicrobiana, in vivo, de uma formulação intramamária à base de polipirrol (PPy) solúvel, um polímero condutor promissor para aplicações biomédicas, especialmente como potente antibacteriano. Neste ensaio, quartos mamários de vacas holandesas sadias (n = 8) foram inoculados com Staphylococcus aureus e tratados, por via intramamária, com formulação experimental à base de PPy solúvel (5%) e com formulação comercial à base de sulfato de gentamicina. O efeito desses tratamentos foi avaliado com a realização de lactoculturas, da Contagem Bacteriana Total (TBC), da Contagem de Células Somáticas (SCC) e da análise da composição do leite das amostras obtidas de quartos mamários, em sete momentos experimentais. Avaliação hematológica dos animais também foi realizada. A aplicação intramamária de três doses da formulação experimental à base de PPy solúvel resultou em maiores log/mL da TBC e da SCC quando comparadas ao grupo controle positivo e ao grupo que recebeu sulfato de gentamicina. A administração da formulação experimental não induziu alterações na composição do leite e nos parâmetros hematológicos. Alguns fatores farmacocinéticos e farmacodinâmicos do PPy solúvel podem ser atribuídos a ineficácia antimicrobiana da pomada experimental. Outras pesquisas devem ser realizados em prol do desenvolvimento de formulações que permitam a atuação antibacteriana do PPy solúvel no ambiente intramamário de vacas leiteiras.
