Imbalance between the cellular proliferation and death in harderian gland of female mice with hyperprolactinemia

SUMMARY OBJECTIVE: To assess the impact of the metoclopramide-induced hyperprolactinemia in cellular death and proliferation in the harderian gland of female mice. METHODS: Twenty female mice were divided into two groups of 10 animals each and treated: 0.2 mL of saline solution (controls, Ctr) and 200 µg of metoclopramide (experimental, hyperprolactinemia), both for 50 consecutive days and at 12:00 a.m. On the 50th day, the female were euthanized, and the harderian glands were removed and processed for immunohistochemistry for detected ki67 and TUNEL method. Data were statistically analyzed by unpaired Student’s t test (p<0.05). RESULTS: The harderian gland of the hyperprolactinemia group showed increase in the immunoexpression of Ki67 and TUNEL compared to the Ctr group (p<0.05), and there was no significant difference in the amount of porphyrin in the HPrl group compared to the Ctr group. CONCLUSION: The hyperprolactinemia led to increased cell death in the acini the harderian gland and cell proliferation in the stroma glandular, fact that suggesting a reduction process of cellular activity and fibrosis, which suggests impairment in the functioning of the lacrimal harderian.

RESUMO A doxorrubicina (dox) é um medicamento antineoplásico que induz cardiotoxicidade por estresse oxidativo. Os flavonoides são antioxidantes extraídos de plantas como Camellia sinensis e Arrabidaea chica (Fridericia chica). Esta pesquisa objetivou avaliar efeitos protetores do extrato de A. chica (AC), comparado ao de C. sinensis (CS), frente ao estresse oxidativo induzido pela dox, no coração. Cardiomiócitos e células neoplásicas MDA-MB 231 foram incubados com AC e CS. Depois, adicionou-se dox e avaliaram-se taxas de viabilidade e morte celular. A citometria de fluxo para o ensaio de iodeto de propídeo (IP) em cardiomiócitos mostrou as seguintes taxas de morte celular: controle 53%; dox 78% (maior que controle, P=0,015); AC_12,5μg/mL + dox 65% (menor que dox, P=0,031); AC_25μg/mL + dox 62% (menor que dox, P=0,028); AC_50μg/mL + dox 63% (menor que dox, P=0,030); CS_12,5μg/mL + dox 71% (menor que dox, P=0,040); CS_25μg/ml + dox 69% (menor que dox, P=0,037); CS_50μg/mL + dox 74% (menor que dox, P=0,044). Resultados das células MDA-MB 231 mostraram que nenhum extrato interferiu na atividade antitumoral da dox. Os dados de IP foram corroborados pelos de MTT. Este estudo reporta promissora utilização de A. chica na prevenção da cardiotoxicidade induzida pela dox.

RESUMO Avaliou-se o valor nutricional do capim-elefante verde colhido aos 56, 84 e 112 dias de idade, por meio do consumo e da digestibilidade em ovinos, da degradabilidade in situ e da cinética da fermentação ruminal. O capim-elefante colhido aos 56 dias proporcionou maiores valores de consumo da MS (1204,81g/dia), da EB (4910,41kcal/dia), da ED (3436,21kcal/dia) e da PB (130,90g/dia). Para o capim-elefante colhido aos 56 dias, ainda foram observados os maiores valores de digestibilidade da MS (70,64%), da EB (70,11%), da PB (68,96%), da FDN (71,4%) e do conteúdo ED (2,86Mcal/kg MS). Com o avanço da idade de corte, houve redução da degradabilidade potencial da MS (77,99% x 68,33%), da PB (84,80% x 56,52%) e da FDN (72,18% x 64,33%), para as idades de corte de 56 e 112 dias, respectivamente. O capim cortado aos 56 dias apresentou maior valor de produção cumulativa de gases (210,50mL/g de MS) e de degradabilidade da matéria seca (63,9%) após 96 horas de fermentação. O capim-elefante verde deve ser colhido aos 56 dias de rebrota em razão das características nutricionais apresentadas.
ABSTRACT The nutritional value of fresh elephant grass harvested at 56, 84 and 112 days of age was evaluated by intake and digestibility in sheep, in situ degradability, and ruminal fermentation’s kinetics. The elephant grass harvested at 56 days provided higher DM intake (1204.81g/day), GE (4910.41kcal/day), DE (3436.21kcal/day), and CP (130.90g/day) values. Furthermore, the highest DM (70.64%), GE (70.11%), DE (2.86%) and CP (68.96%) digestibility values were observed at 56 days. With the increase in the harvest age there was a reduction in the DM (77.99 x 68.33%), CP (84.80 x 56.52%) and NDF (72.18 x 64.33%) potential degradability in the treatments 56 and 112 days, respectively. Higher values of cumulative gas production (210,50mL/g DM) and DM degradability for elephant grass were observed at 56 days, after 96 hours of fermentation. The fresh elephant grass should be harvested at 56 days of regrowth due to the nutritional characteristics found in this study.

RESUMO Este trabalho teve como objetivo avaliar o valor nutritivo e a força de cisalhamento da cultivar de Urochloa brizantha (syn Brachiaria brizantha) cv Xaraés submetida a três intensidades luminosas e quatro cortes. O experimento foi conduzido na FMVZ - Unesp de Botucatu, com delineamento experimental em blocos ao acaso, sendo os tratamentos: luminosidade natural, redução de 30% e 60% de luz, com quatro cortes e três repetições. As análises realizadas foram: composição bromatológica, digestibilidade e a força de cisalhamento. Não houve diferença na digestibilidade entre os tratamentos em nenhum dos cortes, mas a qualidade forrageira foi influenciada pelos níveis de intensidade de luz, tendo o tratamento com 60% de redução de luminosidade apresentado maiores concentrações de proteína bruta e cinzas, menores teores de fibra em detergente neutro, hemicelulose, celulose e força de cisalhamento. Em relação aos cortes estudados, o primeiro teve o menor intervalo de corte e produziu forragem com qualidade superior em comparação ao último, pois obteve menor teor de fibra em detergente ácido, lignina, hemicelulose, celulose e consequente menor força de cisalhamento. Portanto, a redução de 60% de luminosidade é benéfica à qualidade e à força de cisalhamento da cultivar Xaraés.
ABSTRACT This study aimed to evaluate the nutritive value and shear strength of the Xaraés grass (Urochloa brizantha) under the three intensities of light and four cuts. The experiment was conducted at FMVZ - UNESP, Botucatu, with a randomized block design, with the following treatments: natural luminosity, 30% and 60% light reduction, with four cuts and three replications. The analyzes were bromatological composition, digestibility, and shearing strength. There was no difference in digestibility between the treatments in any of the cuts, forage quality was influenced by the light intensity levels, and the treatment with 60% of light reduction produced higher concentrations of crude protein and ash, lower levels of neutral detergent fiber, hemicellulose, cellulose and shear strength. According to the studied cuts, the first one had the lowest cut interval and produced superior forage compared with the last one, as it obtained lower fiber content in acid detergent, lignin, hemicellulose, cellulose and consequent lower shear force. Therefore, the reduction of 60% of luminosity is beneficial to the quality and shear force of the Xaraés palisade grass.

RESUMO O objetivo deste trabalho foi avaliar o efeito do rolipram durante a maturação de oócitos bovinos, expressão gênica e embriões produzidos in vitro. Os ovários bovinos foram coletados no matadouro. Os COCs foram selecionados e divididos em cinco grupos: controle 0 tempo; controle: MIV por 24 horas; tratamentos rolipram com bloqueio MIV por 24 horas em meio de maturação contendo 100, 150 e 200µM. Após 24 horas, todos os grupos foram recolocados em MIV por mais 24 horas. Subsequentemente COCs foram submetidos ao mesmo sistema MIV e fertilizados, sendo avaliada a taxa de clivagem e de blastocisto, além da expressão dos seguintes genes: Mater, BMP15 e Bax. Nenhuma diferença foi observada na expressão gênica. Dos oócitos avaliados logo após a aspiração folicular, 79,0% estavam em GV, GVBD, MI, enquanto 13,40% estavam em MII, e 7,60% em D/NI. A diferença significativa foi observada em diferentes concentrações (T100, T200 e T150µM) em oócitos que atingiram a fase MII em comparação aos tratamentos de controle (P=0,3). Diferenças foram observadas nas taxas de clivagem (P<0,5) entre T150 e T200 quando comparadas com as taxas do grupo C/24. Uma grande diferença foi observada na taxa de blastocisto (P<0,1) entre os tratamentos em relação ao grupo controle.
ABSTRACT The aim of this work was to evaluate the effect of the Rolipram during the maturation of bovine oocytes and gene expression of embryos produced in vitro. Bovine ovaries were collected in slaughterhouse. The COCs were selected and divided into 5 groups: Control 0 time; Control: IVM for 24 hours; Rolipram treatments with IVM blocking for 24 hours in maturation medium containing (100, 150 and 200µM). After 24 hours all groups were reseated in IVM for another 24 hours. Subsequently COCs were subjected to the same IVM system and fertilized, being checked for cleavage post fertilization and for blastocyst. In addition, performed expression of the following genes: Mater, BMP15 and Bax. No difference was found in gene expression. Of oocytes evaluated shortly after follicular aspiration, 79.00% were in GV, GVBD, MI, while 13.40%, were in MII and 7.60%, D/NI. Significant difference was observed in different concentrations (T100, T200 and T150µM) in oocytes that have reached the MII phase compared to control treatments (P= 0.003). Differences were observed in cleavage rate (P< 0.05) between T150 and T200 when compared to the C/24 Group. A high difference was observed on blastocyst rate (P< 0.001) among treatments compared to the control group.

Objective: To present the essential guidelines for pharmacological management of patients with psychomotor agitation in Brazil. Methods: This is a systematic review of articles retrieved from the MEDLINE (PubMed), Cochrane Database of Systematic Reviews, and SciELO databases published from 1997 to 2017. Other relevant articles in the literature were also used to develop these guidelines. The search strategy used structured questions formulated using the PICO model, as recommended by the Guidelines Project of the Brazilian Medical Association. Recommendations were summarized according to their level of evidence, which was determined using the Oxford Centre for Evidence-based Medicine system and critical appraisal tools. Results: Of 5,362 articles retrieved, 1,731 abstracts were selected for further reading. The final sample included 74 articles that met all inclusion criteria. The evidence shows that pharmacologic treatment is indicated only after non-pharmacologic approaches have failed. The cause of the agitation, side effects of the medications, and contraindications must guide the medication choice. The oral route should be preferred for drug administration; IV administration must be avoided. All subjects must be monitored before and after medication administration. Conclusion: If non-pharmacological strategies fail, medications are needed to control agitation and violent behavior. Once medicated, the patient should be monitored until a tranquil state is possible without excessive sedation. Systematic review registry number: CRD42017054440.

Recommendations were summarized according to their level of evidence, which was determined using the Oxford Centre for Evidence-based Medicine system and critical appraisal tools. Results: We initially selected 1,731 abstracts among 5,362 articles. The final sample included 104 articles that fulfilled all the inclusion criteria. The management of agitated patients should always start with the least coercive approach. The initial non-pharmacological measures include a verbal strategy and referral of the patient to the appropriate setting, preferably a facility designed for the care of psychiatric patients with controlled noise, lighting, and safety aspects. Verbal de-escalation techniques have been shown to decrease agitation and reduce the potential for associated violence in the emergency setting. The possibility of underlying medical etiologies must be considered first and foremost. Particular attention should be paid to the patient’s appearance and behavior, physical signs, and mental state. If agitation is severe, rapid tranquilization with medications is recommended. Finally, if verbal measures fail to contain the patient, physical restraint should be performed as the ultimate measure for patient protection, and always be accompanied by rapid tranquilization. Healthcare teams must be thoroughly trained to use these techniques and overcome difficulties if the verbal approach fails. It is important that healthcare professionals be trained in non-pharmacological management of patients with psychomotor agitation as part of the requirements for a degree and graduate degree. Conclusion: The non-pharmacological management of agitated patients should follow the hierarchy of less invasive to more invasive and coercive measures, starting with referral of the patient to an appropriate environment, management by a trained team, use of verbal techniques, performance of physical and mental assessment, use of medications, and, if unavoidable, use of the mechanical restraint. The scientific publication landscape is changing quickly, with an enormous increase in options and models. Articles can be published in a complex variety of journals that differ in their presentation format (online-only or in-print), editorial organizations that maintain them (commercial and/or society-based), editorial handling (academic or professional editors), editorial board composition (academic or professional), payment options to cover editorial costs (open access or pay-to-read), indexation, visibility, branding, and other aspects. Additionally, online submissions of non-revised versions of manuscripts prior to seeking publication in a peer-reviewed journal (a practice known as pre-printing) are a growing trend in biological sciences. In this changing landscape, researchers in biochemistry and molecular biology must re-think their priorities in terms of scientific output dissemination. The evaluation processes and institutional funding for scientific publications should also be revised accordingly. This article presents the results of discussions within the Department of Biochemistry, University of São Paulo, on this subject.

RESUMO: Este estudo objetivou avaliar aspectos comportamentais de ovinos Santa Inês criados em pasto em clima tropical úmido. Para tanto, foram utilizados nove carneiros Santa Inês, mantidos a pasto, expostos diretamente ao clima, sem sombra, durante 24 dias, sendo 12 dias alternados em cada período (seco e chuvoso) em 2015. Os parâmetros comportamentais avaliados foram consumo de alimento, ócio em pé, ócio deitado e ruminação. As observações ocorreram das 08:00h às 17h:55min. O delineamento experimental foi o inteiramente casualizado (DIC) com dois tratamentos (período chuvoso e período seco), nove repetições, um animal por unidade experimental. Para entender as correlações dos parâmetros comportamentais com as variáveis climáticas foi realizada a correlação de Pearson. Foram observados valores médios gerais aproximados entre os períodos chuvoso e seco para as variáveis comportamentais, porém, houve maior ingestão no volume de água no período seco (9,59L/ovino). Também, ocorreu diferença no comportamento dos animais ao longo do dia. No horário mais quente do dia, das 14h às 15h, registrou-se ócio em pé de 25,83min. e ócio deitado de 1,21min., e os animais passaram menor tempo consumindo alimento (27,08min.) em relação aos demais horários. Podemos concluir que os ovinos apresentaram parâmetros comportamentais semelhantes nos períodos chuvoso e seco, porém, quando observamos os horários dos dias, os comportamentos são diferentes. A principal atividade registrada foi o pastejo. As variáveis temperatura e a umidade relativa do ar influenciam de forma diferente o comportamento dos ovinos entre as estações do ano, sendo mais marcantes no período seco.
ABSTRACT: This study aimed to evaluate behavioral aspects of Santa Inês sheep in pasture in humid tropical climate. In order to do so, nine Santa Inês sheep were kept on pasture, exposed to the climate directly, without shade, for 24 days, with 12 alternate days in each period (dry and raining) in 2015. The behavioral parameters evaluated were: food consumption, standing idleness, lying idle and rumination. Observations occurred from 08:00 A.M. to 05:55 P.M. The experimental was a completely randomized design (CRD) with two treatments (rainy season and dry period), nine replications, one animal per experimental unit. To understand the correlations of the behavioral parameters with the climatic variables, Pearson’s correlation was performed. Approximate average values between the rainy and dry periods for the behavioral variables were observed, but there was a greater intake in the volume of water in the dry period (9.59L/sheep). Also, there were significant differences in the behavior of the animals throughout the day. In the hottest time of the day, at 02:00 P.M., there was standing idle of 25.83min., lying idle of 1.21min., and spent less time-consuming food (27.08min.) in relation to other schedules. We concluded that the sheep had similar behavioral parameters in the rainy and dry periods, but when we observe the times of the days, the behaviors are different. The main activity recorded was grazing, and the variables, temperature and relative humidity of the air influence differently between the seasons, being more marked in the dry period.

Abstract: The systemic effect of chronic periodontitis (CP) has been suggested by several studies as an etiologic factor and modulator of diseases based on the changes in the inflammatory marker levels. This study aimed to investigate the relationship between the changes in clinical periodontal outcomes and serum biomarkers (CRP, IL-6, albumin and percentage of leukocytes) after non-surgical periodontal therapy in systemically healthy adults. An interventional study was conducted with a sample of 29 individuals without CP (control group) and 33 with CP (CP group). Periodontal clinical variables were recorded, and the serum levels of inflammatory markers were measured. Statistical analysis included the chi-square and Student’s t-tests and Pearson’s correlation analysis. After 90 days of non-surgical periodontal treatment, a reduction of periodontal parameters and IL-6 in both groups could be observed (P < 0.001). The correlation analysis revealed a directly proportional correlation between changes in the probing depth (r = 0.349, P = 0.049) and clinical attachment level (r = 0.374, P = 0.034) with CRP in the CP group. The findings suggest a reduction of IL-6 serum concentration and periodontal clinical measures 90 days after periodontal therapy in both groups.

Resumo Um levantamento atualizado das plantas com sementes e análises relevantes acerca desta biodiversidade são apresentados. Este trabalho se iniciou em 2010 com a publicação do Catálogo de Plantas e Fungos e, desde então vem sendo atualizado por mais de 430 especialistas trabalhando online. O Brasil abriga atualmente 32.086 espécies nativas de Angiospermas e 23 espécies nativas de Gimnospermas e estes novos dados mostram um aumento de 3% da riqueza em relação a 2010. A Amazônia é o Domínio Fitogeográfico com o maior número de espécies de Gimnospermas, enquanto que a Floresta Atlântica possui a maior riqueza de Angiospermas. Houve um crescimento considerável no número de espécies e nas taxas de endemismo para a maioria dos Domínios (Caatinga, Cerrado, Floresta Atlântica, Pampa e Pantanal), com exceção da Amazônia que apresentou uma diminuição de 2,5% de endemicidade. Entretanto, a maior parte das plantas com sementes que ocorrem no Brasil (57,4%) é endêmica deste território. A proporção de formas de vida varia de acordo com os diferentes Domínios: árvores são mais expressivas na Amazônia e Floresta Atlântica do que nos outros biomas, ervas são dominantes no Pampa e as lianas apresentam riqueza expressiva na Amazônia, Floresta Atlântica e Pantanal. Este trabalho não só quantifica a biodiversidade brasileira, mas também indica as lacunas de conhecimento e o desafio a ser enfrentado para a conservação desta flora.
Abstract An updated inventory of Brazilian seed plants is presented and offers important insights into the country's biodiversity. This work started in 2010, with the publication of the Plants and Fungi Catalogue, and has been updated since by more than 430 specialists working online. Brazil is home to 32,086 native Angiosperms and 23 native Gymnosperms, showing an increase of 3% in its species richness in relation to 2010. The Amazon Rainforest is the richest Brazilian biome for Gymnosperms, while the Atlantic Rainforest is the richest one for Angiosperms. There was a considerable increment in the number of species and endemism rates for biomes, except for the Amazon that showed a decrease of 2.5% of recorded endemics. However, well over half of Brazillian seed plant species (57.4%) is endemic to this territory. The proportion of life-forms varies among different biomes: trees are more expressive in the Amazon and Atlantic Rainforest biomes while herbs predominate in the Pampa, and lianas are more expressive in the Amazon, Atlantic Rainforest, and Pantanal. This compilation serves not only to quantify Brazilian biodiversity, but also to highlight areas where there information is lacking and to provide a framework for the challenge faced in conserving Brazil's unique and diverse flora.

Objetivou-se avaliar a inclusão de ureia e glicerina bruta como aditivos na ensilagem da cana-de-açúcar, na composição químico-bromatológica, pH, N-amoniacal (N-NH3) e digestibilidade in vitro (DIV). Os tratamentos foram quatro doses de ureia, 0, 10, 20 e 30 g de ureia por kg de cana-de-açúcar na ensilagem, e cinco doses de glicerina bruta, 0, 10, 20, 30 e 40g de glicerina bruta por kg de cana-de-açúcar na ensilagem. As silagens foram armazenadas por 180 dias. O tratamento com ureia afetou a maioria das variáveis relacionadas ao valor nutritivo, aumentando os teores de matéria seca (MS) e proteína (PB) (2,58; 7,76; 18,70 e 19,31%), reduzindo os teores de fibra em detergente neutro (FDN) e melhorando a DIV da MS (42,61; 48,53; 50,69 e 51,18%) e FDN (38,81; 39,23; 41,06 e 43,46%), e as características fermentativas da silagem, apresentando valores de pH de 3,49; 3,86; 4,18 e 3,93 e de N-NH3 de 1,72; 3,80; 7,88 e 9,00, para as dose de 0, 10, 20 e 30 g, respectivamente. A glicerina bruta aumentou os teores de MS e extrato etéreo (1,45; 3,03; 3,62; 3,41 e 4,38%), melhorou a DIV da MS com valores de 49,61; 52,24; 53,28; 55,60 e 56,09% e reduziu perdas por gases durante o processo de fermentação, apresentando médias de 6,69; 5,97; 5,89; 5,51 e 5,48% da MS para as doses 0, 10, 20, 30 e 40g, respectivamente. Assim, a ureia e a glicerina bruta podem ser utilizadas como aditivos na ensilagem da cana-de-açúcar.
The aim of this study was to evaluate the inclusion of urea and crude glycerin as an additive in ensiling of sugar cane, in chemical composition, pH, ammonia-N (N-NH3) and in vitro digestibility (IVD). The treatments were four doses of urea 0, 10, 20 and 30 g per kg of sugar cane ensiling and five doses of crude glycerin 0, 10, 20, 30 and 40g per kg of crude glycerin sugar cane ensiling. The silages were stored for 180 days. Treatment with urea affected most variables related to nutritional value, increasing the concentrations of dry matter (DM) and protein (CP) (2.58, 7.76, 18.70 and 19.31%) and reduced levels of neutral detergent fiber (NDF) and improved IVDDM (42.61, 48.53, 50.69 and 51.18%) and NDF (38.81, 39.23, 41.06 and 43.46%) and fermentation characteristics of silage, with pH values of 3.49, 3.86, 4.18 and 3.93 and NH3 1.72, 3.80, 7.88 and 9.00 for the dose of 0, 10, 20 and 30 g, respectively. The crude glycerin increased in DM and ether extract (1.45, 3.03, 3.62, 3.41 and 4.38%), improved IVDDM with values of 49.61, 52.24, 53 28; 55.60 and 56.09% and reduced gas losses during the fermentation process with mean of 6.69, 5.97, 5.89, 5.51 and 5.48% of DM for the doses 0, 10 , 20, 30 and 40g, respectively. Urea and crude glycerin can be used as an additive in ensiling of sugar cane.

Enteropathogenic Escherichia coli (EPEC) are important human gastroenteritis agents. The prevalence of six non-LEE genes encoding type 3 translocated effectors was investigated. The nleC, cif and nleB genes were more prevalent in typical than in atypical EPEC, although a higher diversity of genes combinations was observed in atypical EPEC.

A expressão de RNAm para leptina, receptor de leptina (obRb), adiponectina, receptor de adiponectina (AdipoR1) e resistina foi avaliada por meio da técnica de PCR em tempo real, em tecidos ovariano, hipofisário, adiposo do omento e da região perirrenal, em ovelhas alimentadas sem farelo de mamona ou com farelo de mamona detoxificada durante 14 meses. O tipo de dieta não afetou os níveis de RNAm para leptina, obRb, adiponectina, AdipoR1 e resistina nos diferentes tecidos avaliados (P>0,05). Nos tecidos ovariano e hipofisário, não foi verificada a expressão da adiponecina e da resistina, respectivamente. Como consequência, pode-se concluir que o farelo de mamona detoxificada pode ser utilizado como fonte proteica na dieta de ovelhas, sem afetar a expressão do gene resistina e dos genes leptina e adiponectina, bem como de seus receptores.
The expression of leptin, leptin receptor (obRb), adiponectin, adiponectin receptor (AdipoR1) and resistin was assessed by real-time PCR technique in ovarian, pituitary, and the omental adipose perirenal tissue in sheep feed without castor meal or with detoxified castor meal. The type of diet did not affect mRNA levels for leptin, obRb, adiponectin, resistin AdipoR1 evaluated in different tissues (P>0.05). However, in pituitary and ovarian tissues there was no expression of resistin and adiponectin, respectively. The detoxified castor meal can be used in sheep diets as alternative food protein without affecting the expression of leptin and adponectin as well as their receptors and resistin.

Descreve-se a ocorrência de Aviadenovirus (FAdV) na avicultura mineira. Foram amostrados fígados de poedeiras jovens (n=25) e velhas (n=25) e de frangos de corte (n=300). Em matrizes pesadas foram amostrados fígados (n=25) e fezes (n=25). O estudo envolveu as principais regiões avícolas do Estado de Minas Gerais. O DNA de FAdV foi pesquisado por PCR universal, descrito para a amplificação do gene que codifica o hexon de Aviadenovirus, usando FAdV Phelps como referência. Foi demonstrada a presença do DNA de FAdV em 100% (25/25) das poedeiras velhas (78 semanas de idade) e em 36% (9/25) das jovens (18 dias). Em frangos de corte, a detecção variou entre 24 e 86%. Embora as fezes das matrizes tenham sido negativas, foi obtido o amplicon específico em 4/25 dos fígados dessas mesmas aves, indicando menor sensibilidade para detecção nas fezes. Em amostras da avicultura familiar (fígado), colhidas de uma das regiões de avicultura intensificada, foi detectado o genoma de FAdV em 100% das galinhas (n=12). A constatação de alta disseminação de FAdV em aves da avicultura industrial e familiar de Minas Gerais contribui para o entendimento da epidemiologia de Aviadenovirus. As sequências nucleotídicas dos produtos de PCR alinharam com FAdV da espécie D de Aviadenovirus. A demonstração de FAdV em reprodutores indica risco de transmissão vertical e reforça a necessidade de biosseguridade estrita nesses plantéis. A presença de FAdV em diversos setores avícolas, incluindo poedeiras comerciais, frangos de corte, reprodutores e galinhas da avicultura familiar, recomenda a biosseguridade estrita entre as criações de mesmo tipo e de tipos diferentes de aves. A detecção em matrizes pode indicar a continuada geração de progênies infectadas.
The occurrence of Aviadenovirus (FAdV) was investigated in chickens from the poultry industry of Minas Gerais state, Brazil. The investigation was conducted due to the scarcity of recent data in the country and its description in neighboring countries. For this purpose, livers were collected from layer chicks (n=25), older layers (n=25), broilers (n=300), and livers (n=25) and stool (n=25) samples from broiler breeders, representing the major poultry regions of the state. FAdV DNA was demonstrated using a previously described PCR protocol for amplifying part of the hexon gene encoding sequence. FAdV was found in layer chicks (36%), widespread (100%) in older layers, and with regional differences in broilers (24-86%). Although all broiler breeder stools were negative, FAdV DNA was detected in livers (16%, 4/25) of stool-negative birds. In order to obtain additional information on the circulation of the infection, livers of subsistence chickens collected from one poultry intensive region, were evaluated (n = 12), with FAdV being detected in all samples. FAdV was found in young and old layers, broilers, broiler breeders and free-range chickens, and results suggest the circulation of FAdV among different types of chickens. The detection in older layer chickens may indicate an extended risk of horizontal transmission in regions of Minas Gerais with mixed activity of egg and meat type chickens and poor biosecurity strategies. The infection in breeders may indicate vertical transmission and the continuous production of infected progenies. The hexon-gene-targeted PCR amplicon sequences aligned with FAdV of species D of Aviadenovirus. Results indicate the necessity for biosecurity, especially for breeders, separating flocks according to origin, age and health status, which will be an advantage regarding any pathogen.
