ABSTRACT We investigate convective storms over the Sabana de Bogotá, a high-altitude and densely populated area in the Colombian tropical Andes. Convective events are identified using infrared satellite images and in-situ precipitation data. As expected, convection shows a strong early-afternoon peak during the two rainy seasons. Previous studies hypothesize that early-afternoon westerly winds and their moisture advection from the warmer Magdalena valley are the main explanatory mechanism for intense storms. We find that early-afternoon westerlies are present in 78% of rainy season days, but convective events develop in only 26% of them. Thus, although westerlies seem necessary for convection due to the convergence they generate, they only occasionally generate storms and are therefore not a good predictor. Furthermore, reanalysis data indicate that precipitable water vapor (PWV) at the Magdalena valley is anomalously low during convective days, suggesting that moisture converges locally instead of being advected from the west. Based on composites of surface wind speed, air temperature, surface pressure, and GPS-derived PWV, we identify the most prominent signals associated with deep convection: a weaker than average wind speed throughout the morning, higher than normal values of surface air temperature towards noon, followed by an anomalous steep increase of PWV and wind speed. These features indicate that convection results from a strong diurnal forcing facilitated by convergence of westerly winds, combined with sufficient water vapor convergence, with a timescale of about 3 h. This highlights the relevance of high temporal resolution monitoring of PWV offered by Global Navigational Satellite System stations.

RESUMEN Con el fin de estudiar tormentas convectivas en la Sabana de Bogotá, un área densamente poblada de gran elevación en los Andes tropicales colombianos, identificamos eventos convectivos con imágenes satelitales y datos de precipitación. Los eventos son más frecuentes en las primeras horas de la tarde de las dos estaciones lluviosas. Estudios previos sugieren que los vientos del oeste de las primeras horas de la tarde y su advección de humedad desde el Valle del Magdalena son la principal causa de estas tormentas. Sin embargo, encontramos que estos vientos están presentes en el 78% de los días en temporada de lluvias, pero las tormentas se desarrollan solo en 26% de ellos. Así, aunque los vientos del oeste parecen necesarios debido a la convergencia que generan, sólo en ocasiones producen tormentas, de manera que no son un buen predictor. Datos de reanálisis indican que el vapor de agua precipitable (PWV, por su sigla en inglés) en el Valle del Magdalena es anormalmente bajo durante días convectivos, lo que sugiere que la humedad converge localmente en lugar de ser transportada desde el oeste. Usando análisis de compuestos, identificamos las señales más prominentes asociadas con la convección: la rapidez del viento es menor que la media en la mañana, mientras que la temperatura es superior al promedio al mediodía, seguida de un anómalo incremento del PWV y la rapidez del viento. Estas características indican que la convección resulta de un fuerte forzamiento diurno, facilitado por convergencia de los vientos del oeste y del vapor de agua, en una escala de tiempo de alrededor de 3 h.

RESUMEN Objetivos. Determinar los cambios en las características clínicas y desenlaces intrahospitalarios de los pacientes hospitalizados por COVID-19 en un hospital privado de Caracas durante dos años de pandemia. Materiales y métodos. Estudio retrospectivo, observacional, de pacientes hospitalizados por COVID-19. Se investigó la correspondencia entre las olas de ingresos hospitalarios con las variantes circulantes del SARS-CoV-2 en la población general del Distrito Capital y estado Miranda. Resultados. Se incluyeron 1025 pacientes (569 hombres y 456 mujeres), con edad promedio de 62,9 DE: 16,2 años. Cuatro olas de ingresos hospitalarios fueron identificadas: primera (marzo-noviembre 2020) 150/1025 (14,6%) casos; segunda (diciembre-2020 a mayo-2021) 415/1025 (40,5%) casos; tercera (junio-diciembre 2021) 344/1025 (33,6%) casos; cuarta (enero-febrero 2022) 116/1025 (11,3%) casos. La edad promedio fue mayor en la cuarta ola (primera 64,0±15,7, segunda 61,4±15,8, tercera 62,1±16,5, y cuarta ola 68,5±16,4), mientras que la proporción de pacientes masculinos (primera 66,7%, segunda 58,8%, tercera 50,3%, y cuarta 44,8%), los pacientes con enfermedad grave-crítica (primera 65,3%, segunda 57%, tercera 51,7% y cuarta 44,8%), la estadía intrahospitalaria (primera 9,1±6,0, segunda 9,0±7,3, tercera 8,8±7,7, y cuarta 6,9±5,0 días), los ingresos a la UCI (primera 23,3%, segunda 15,7%, tercera 14,0%, y cuarta 11,2%; p=0,027) y la mortalidad (primera 21.8%, segunda 10,7%, tercera 9,1%, y cuarta 7,1%; p<0,001) disminuyeron progresivamente con el tiempo. Conclusiones. Los resultados muestran menor frecuencia de casos severos y mejoría de los desenlaces intrahospitalarios en dos años de pandemia. Es probable que los cambios en las variantes circulantes, las mejoras del manejo de la enfermedad y la vacunación hayan influido sobre estos resultados.

The mean age was higher in the fourth wave (first: 64.0±15.7, second: 61.4±15.8, third: 62.1±16.5, and fourth wave: 68.5±16.4), while the proportion of male patients (first: 66.7%, second: 58.8%, third: 50.3%, and fourth wave: 44.8%), patients with severe-critical illness (first: 65.3%, second: 57%, third: 51.7%, and fourth wave: 44.8%), in-hospital stay (first: 9.1±6.0, second: 9.0±7.3, third: 8.8±7.7, and fourth wave: 6.9±5.0 days), ICU admissions (first: 23.3%, second: 15.7%, third: 14.0%, and fourth wave: 11.2%; p=0.027) and mortality (first: 21. 8%, second: 10.7%, third: 9.1%, and fourth wave: 7.1%; p<0.001) progressively decreased over time. Conclusions. The results show lower frequency of severe cases and improvement of in-hospital outcomes in two years of the pandemic. Changes in circulating variants, improvements in disease management and vaccination are likely to have influenced these results.

RESUMEN Objetivo . Explorar la viabilidad de desarrollar un modelo de neurocisticercosis (NCC) de oveja mediante infección intracraneal de oncosferas de T. solium. Materiales y métodos. Se realizó un modelo de infección experimental de NCC en ovejas. Se inocularon aproximadamente 10 posoncósferas de T. solium cultivadas previamente por 30 días por vía intracraneal en diez ovejas. Las oncósferas, en 0,1 mL de solución salina fisiológica, se inyectaron en el lóbulo parietal a través de una aguja de calibre 18. Resultados. Después de tres meses, en dos ovejas se encontraron granulomas y en una tercera identificó un quiste de 5 mm de diámetro en el ventrículo lateral derecho y la evaluación histológica confirmó que el quiste corresponde a una larva de T. solium. También se utilizó inmunohistoquímica con anticuerpos monoclonales dirigidos contra componentes de membrana y antígenos excretorios/secretorios del quiste de T. solium para confirmar la etiología de los granulomas encontrados. Uno de ellos mostro reactividad ante los anticuerpos monoclonales utilizados, confirmando así que se trató de un cisticerco. Conclusión. Este experimento es la prueba de concepto de que es posible infectar ovejas con cisticercosis por inoculación intracraneal.

ABSTRACT Objective. To explore the feasibility of developing a sheep model of neurocysticercosis (NCC) by intracranial infection with T. solium oncospheres. Materials and methods. We carried out an experimental infection model of NCC in sheep. Approximately 10 T. solium oncospheres previously cultured for 30 days were inoculated intracranially into ten sheep. The oncospheres, in 0.1 mL of physiological saline, were injected into the parietal lobe through an 18-gauge needle. Results. After three months, granulomas were found in two sheep. In a third sheep we identified a 5 mm diameter cyst in the right lateral ventricle and histological evaluation confirmed that the cyst corresponded to a T. solium larva. Immunohistochemistry with monoclonal antibodies directed against membrane components and excretory/secretory antigens of the T. solium cyst was also used to confirm the etiology of the found granulomas. One of them showed reactivity to the monoclonal antibodies used, thus confirming that it was a cysticercus. Conclusion. This experiment is the proof of concept that it is possible to infect sheep with cysticercosis by intracranial inoculation.

ABSTRACT Mitochondria are complex organelles that play a critical role within the cell; mitochondrial dysfunction can result in significant cell damage or death. Previous studies have demonstrated the promising therapeutic effects of autologous mitochondria transplantation into ischemic cardiac tissue; however, few studies have examined the in vivo effects of mitochondria infusion into the brain. The aim of this study is to report a procedure for carotid infusion of autologous mitochondria into porcine brains. By using this infusion technique, we propose that a selective and minimally invasive administration is feasible and may provide benefits in the treatment of various central nervous system disorders.

RESUMEN Las mitocondrias son organelas complejas que desempeñan un papel fundamental en la célula, la disfunción mitocondrial puede ocasionar daños celulares significativos o la muerte. Estudios previos han demostrado los prometedores efectos terapéuticos del trasplante de mitocondrias autólogas a un tejido cardiaco isquémico, sin embargo, pocos estudios han evaluado los efectos in vivo de la infusión de mitocondrias en el cerebro. El presente trabajo tiene como objetivo dar a conocer el procedimiento para la infusión vía carótida de mitocondrias autólogas en cerebros porcinos. Mediante esta técnica de infusión, proponemos que una administración selectiva y mínimamente invasiva es factible y puede proporcionar beneficios en el tratamiento de diversas patologías del sistema nervioso central.

Resumo Os objetivos primários do estudo foram determinar a validade convergente e discriminante do construto da Escala Breve de Otimismo Interativo-G (EBOI-G), em participantes de seis estados do México e calcular sua consistência interna. Participaram 3 289 mexicanos, sendo 2 028 homens e 1 243 mulheres. A média de idade foi = 30.43 anos, DP = 10.52. Foram utilizadas análises fatoriais confirmatórias e análises de regressão múltipla. Bons indicadores médios de qualidade de ajuste foram encontrados (por exemplo, CFI = .99; RMSEA = .07.) A validade convergente foi avaliada, r (3 289) = .52 (p = <.01; d = mediana), com a Escala de satisfação com a vida. A validade discriminante, r (3 289) = -.19 foi estimada com a Escala Breve de Disposição à Raiva (p = <.01; d = quase pequeno). O alfa = 0,70 (3 289); p = <0,01; o ômega = 0,76. Conclui-se que há evidências nacionais parciais que apoiam o uso da EBOI-G, devido à falta de uma medida desse tipo no México, sendo útil pelo menos para fins de pesquisa.

Resumen Los objetivos primarios del presente estudio fueron determinar la validez de constructo, convergente y discriminante de la Escala Breve de Optimismo Interactivo-G (EBOI-G), en participantes de seis estados de México y calcular su consistencia interna. Participaron 3 289 mexicanos, 2 028 hombres y 1 243 mujeres (18 casos no contestaron cuál era su género). Su edad promedio = 30.43 años, DE = 10.52. Se usó el análisis factorial confirmatorio y análisis de regresión múltiple, y se encontraron buenos indicadores promedio de bondad de ajuste (e. g., CFI = .99; RMSEA = .07.). Se evaluó la validez convergente, r (3 289) = .52 (p = < .01; d = mediano), con la Escala de Satisfacción con la Vida. Se estimó la validez discriminante, r (3 289) r = -.19, con la Escala Breve de Disposición a la Ira (p = < .01; d = casi pequeño). El alfa = .70 (3 289); p = < .01; el omega = .76. Se concluye que hay evidencia parcial nacional que apoya el uso de la EBOI-G, debido a la carencia de una medida de este tipo en México, útil cuando menos para propósitos de investigación.

Abstract The primary objectives of this study were determining the construct, convergent, and discriminant validity of the Brief Interactive Optimism Scale-G (BIOS-G) in participants from six states of Mexico and estimating its internal consistency. In this study 3289 Mexicans participated (2028 men and 1243 women). The average age was = 30.43 years and SD = 10.52. Confirmatory factor analysis (CFA) and multiple regression analysis (MRA) were applied. There were appropriate fit indexes (e. g., CFI = .99; RMSEA = .07.). Convergent validity showed an r (3289) = .52 (p = < .01; d = medium), with the Satisfaction with Life Scale (SWLS) and the estimation of discriminant validity was r (3 289) r = -.19 with the Brief Scale for Assessing Anger Proneness (APS-G) (p = < .01; d = almost small), Alpha = .70 (3 289); p = < .01; omega = .76. The conclusion is that there is partial national evidence supporting the use of BIOS-G, because Mexico lacks a measure of this kind, being useful, at least, for research purposes.

SUMMARY: The Viperidae venoms are composed of a mixture of constituents with enzymatic and non-enzymatic actions, which act on ultrastructural components of cells and tissues. Here, the number of mitochondria, mitochondrial area and the number of mitochondrial cristae from adrenal glands cortex treated with snake venoms were tested after 3, 6 and 24 hours of venom injections. The mitochondria quantitative changes showed a statistically significant decrease, in the number of mitochondria past 3, 6 and 24 h. There was an increase in the mitochondrial area after 6 h, where Crotalus vegrandis venom did not present significant differences with Crotalus pifanorum or Bothrops venezuelensis venoms. After 24 h, there was an escalation of mitochondrial area in all tested venoms. The number of mitochondrial cristae after 3 h did not present important differences with the control treatment. After 6 h, the number of mitochondrial cristae initiated to decrease under the activities of the 3 venoms action, until 24 h of observation. In the qualitative observations it was possible to witness an intense damage of the mitochondria, with loss and swelling of membranes, disappearance of cristae and the appearance of myelin figures, which started at 3 h after the Crotalus and Bothrops venoms injections. These damages probably were due to cytotoxic effects of phospholipases, metalloproteases and/or other proteolytic activities present in Viperidae snake venoms, being more evident in Crotalus venoms. As far as we know, these results define a novel finding that suggest that Viperidae snake venoms are extremely toxic to mammalian mitochondria.

RESUMEN: Los venenos de Viperidae tienen acciones enzimáticas y no enzimáticas, que actúan sobre la estructura celular. Aquí se probaron, a las 3, 6 y 24 horas de la inyección del veneno, el número de mitocondrias, el área mitocondrial y el número de crestas mitocondriales de la corteza de las glándulas adrenales. Los cambios cuantitativos de las mitocondrias mostraron una disminución en el número de mitocondrias a las 3, 6 y 24 h. Hubo un aumento en el área mitocondrial a las 6 h, donde el veneno de la serpiente Crotalus vegrandis no presentó diferencias significativas con los venenos de Crotalus pifanorum o Bothrops venezuelensis. Después de 24 h, hubo un aumento del área mitocondrial en todos los venenos. El número de crestas mitocondriales a las 3 h no presentó alteraciones o diferencias importantes con el tratamiento de control. Después de 6 h, el número de crestas mitocondriales comenzó a disminuir bajo la acción de los 3 venenos, hasta las 24 h de observación. En las observaciones cualitativas se observó un daño intenso de las mitocondrias, con pérdida y edema de las membranas, desaparición de las cristae y aparición de figuras mielínicas, que comenzó a las 3 h después de las inyecciones de veneno de Crotalus y Bothrops. Estos daños se debieron factiblemente a los efectos citotóxicos de componentes proteolíticos de los venenos. Creemos que estos resultados definen un nuevo y original hallazgo, que sugiere que los venenos de serpiente Viperidae son extremadamente tóxicos para las mitocondrias de mamíferos.

ABSTRACT The use of L protein coupled magnetic particles for the concentration and purification of immunoglobulin M (mIgM) monoclonal antibodies against Taenia solium was evaluated. Three concentration methods and different elution times were evaluated and the ratio of particles to the ratio of mIgM was optimized. It is demonstrated that: 1) with the use of magnetic particles, a previous concentration of mIgM is not required, which reduces the manipulation of the antibodies and improves the recovery, 2) the use of a binding buffer can be omitted, since the pH of most cell culture supernatants are neutral, and 3) longer elution times (~ 45 minutes) are needed to increase recovery to a level greater than 80%. The study demonstrates that the use of L protein-coupled magnetic particles is a simple and efficient tool for mIgM concentration and purification.

RESUMEN Se evaluó el uso de partículas magnéticas acopladas a proteína L para la concentración y purificación de anticuerpos monoclonales inmunoglobulina M (mIgM) contra Taenia solium. Se evaluaron tres métodos de concentración y diferentes tiempos de elución y se optimizó la proporción de partículas a la proporción de mIgM. Demostramos que: 1) con el uso partículas magnéticas no se requiere de una concentración previa de mIgM, lo que disminuye la manipulación de los anticuerpos y mejora la recuperación, 2) se puede omitir el uso de un tampón de unión, ya que el pH de la mayoría de los sobrenadantes de cultivo celular son neutros, y 3) se necesitan tiempos de elución más largos (~45 minutos) para aumentar la recuperación a un nivel mayor a 80%. El estudio demuestra que el uso de partículas magnéticas acopladas a proteína L es una herramienta simple y eficiente para la concentración y purificación de mIgM.

Abstract The ovicidal effect of hydro-alcoholic extracts from mature fruits and dried leaves of the Caesalpinia coriaria tree, against Haemonchus contortus and H. placei was evaluated by the egg hatching inhibition test (EHIT). Secondary compounds within the extracts were identified by high performance liquid chromatography. Five extract concentrations were assessed in the inhibition tests (25.0, 12.5, 6.2, 3.1 and 1.5 mg/mL), using distilled water and Thiabendazole (0.5 mg/mL) as negative and positive controls, respectively. The results were analysed under a 2 × 2 × 6 completely randomized factorial design, and mean differences were established by the Tukey’s post hoc test. The 50% (EC50) and 90% (EC90) effective concentrations for each extract were determined using a Probit analysis. A concentration-dependent ovicidal effect of hydro-alcoholic extracts from both leaves and fruits was observed, reaching a 100% efficacy against both nematode species with the 25.0 mg/mL concentration (p < 0.05). The EC50 for the fruit and leaves extracts were established as 1.63 and 3.91 mg/mL, and as 3.98 and 11.68 mg/mL, against H. contortus and H. placei, respectively. The most important secondary compounds identified in extracts were free-condensed tannins, gallic acid and methylgallate. In conclusion, hydro-alcoholic extracts from C. coriaria mature fruits and dried leaves inhibit in vitro egg hatching of H. contortus and H. placei.

Abstract This study presents the first results of Ni-rich coating growth by the Pechini method on 304 stainless steels fractures, in order to healing fracture surfaces of metallic parts. A series of tests were performed to establish the calcination temperature by differential scanning calorimetry (DSC) at 1300°C using a heating rate of 10°C/min in mixtures of (CH3COO)2Ni•4H2O (Nickel Acetate Tetrahydrate) with HOC(COOH)(CH2COOH)2•H2O (Citric Acid Monohydrate) and precursors: HOCH2CHOHCH2OH (Glycerol) and HOCH2CH2OH (Ethyleneglycol). Results indicate that the Ethyleneglycol precursor is better than the Glycol because it shows a thermal stability at lower temperature of calcination. Subsequently, 304 stainless steel cylindrical fractures (0.6 cm in diameter, 1 cm height) were immersed in a solution of Ethyleneglycol and these samples were gelated at 110°C for 24 h in a muffle furnace. Additionally, these samples were calcined at 660°C for 60 min under an Ar flow of 200 ml/min, using a heating/cooling rate of 5°C/min in order to synthesize the nickel nanoparticles on the fractures. Coating growth on the fractures was carried out at an Ar flow of 220 ml/min at 1100°C for 60 min at a heating/cooling rate of 10°C/ min. By optical and scanning microscopy MEB), it was observed that the coating has a thickness of ~700 μm with a series of cuboidal precipitates larger than 1 μm, as well as laminar phases rich in Ni, Fe and Cr.

Resumen En este estudio se llevó a cabo el crecimiento de recubrimientos ricos en Ni por el método Pechini sobre fracturas de acero inoxidable 304, para sanar superficies de fractura de piezas metálicas. Una serie de pruebas se llevaron a cabo para establecer la temperatura de calcinación por calorimetría de barrido diferencial (DSC) a 1300°C por 10°C/min en mezclas de (CH3COO)2Ni•4H2O (acetato de níquel tetrahidratado) con HOC(COOH)(CH2COOH)2•H2O (ácido cítrico monohidratado) y precursores: HOCH2CHOHCH2OH (glicerol) y HOCH2CH2OH (etilenglicol). Los resultados mostraron que el uso de Etilenglicol es mejor que el Glicerol debido a que muestra una estabilidad térmica a menor temperatura para la calcinación. Posteriormente, fracturas cilíndricas de 0.6 x 1 cm de acero inoxidable 304 se sumergieron en una solución con Etilenglicol y, se gelaron en una mufla a 110°C por 24 h. Adicionalmente, las muestras se calcinaron a 660°C por 60 min bajo un flujo de 200 ml/ min de Ar, a velocidad de calentamiento/enfriamiento de 5°C/min para sintetizar las nanopartículas de Ni sobre las fracturas. El crecimiento del recubrimiento sobre las fracturas se llevó a cabo en un flujo de Ar de 220 ml/min a 1100°C por 60 min a una velocidad de calentamiento/enfriamiento de 10°C/min. Mediante microscopía óptica y de barrido, se observó que el recubrimiento presenta un espesor de hasta 700 μm con una serie de precipitados cuboidales mayores a 1 μm, así como fases laminares ricos en Ni, Fe y Cr.

ABSTRACT Neurocysticercosis is a nervous system infection caused by the larvae (cysticercus) of the pork tapeworm (Taenia solium). Neurocysticercosis is the primary cause of acquired epilepsy worldwide and, therefore, a global public health problem. On the other hand, T. solium taeniasis/cysticercosis is one of very few infectious diseases considered to be potentially eradicable. Recently, a large-scale elimination program in Tumbes, Peru, demonstrated the feasibility of interrupting transmission. Based on these advances, a series of initial guidelines are proposed aimed at setting out the foundations for regional and national taeniasis/cysticercosis control programs, with simple and feasible local interventions as a starting point.

RESUMEN La neurocisticercosis es la infección del sistema nervioso por la larva (cisticerco) de la tenia del cerdo (Taenia solium). La neurocisticercosis es la mayor causa de epilepsia adquirida a nivel mundial y por ende un problema de salud pública global. De otro lado, la teniasis/cisticercosis por T. solium es una de las pocas infecciones consideradas potencialmente erradicables. Recientemente, un programa de eliminación de transmisión a gran escala en Tumbes, Perú, demostró la factibilidad de interrumpir la transmisión. Sobre la base de estos avances, se proponen una serie de lineamientos iniciales para poner las bases de programas de control regionales y nacionales de la teniasis/cisticercosis, partiendo de intervenciones simples y factibles a nivel local.

The magnitude of air velocity at the fan outlet of agricultural sprayers influences the aerodynamic airflow and is a key factor in the flow structure. The effect of varying the fan output speed in aerodynamic airflow during work at different speeds of movement of the sprayer, using computer simulation (CFD) is analyzed in this work. Simulations of output speeds were conducted in the diffuser fan of 40, 45 and 50 m/s with a fan output width of 115 mm and considering the sprayer without motion and working speeds of 2.18, 4.5 and 6.35 km / h. The aerodynamic behavior of the airflow for each variant was analyzed. The increase in the fan output speed produces an accelerated wear on the core of the current; therefore the use of lower speeds enhances aerodynamic flow

La magnitud de la velocidad del aire a la salida del ventilador de los pulverizadores agrícolas influye en la aerodinámica del flujo de aire y constituye un factor fundamental en la estructura del flujo. En el trabajo se analiza mediante el uso de la simulación por computadora (CFD), el efecto de la variación de la velocidad de salida del ventilador en la aerodinámica del flujo de aire durante el proceso de trabajo a distintas velocidades de movimiento del pulverizador. Se realizaron simulaciones de velocidades de salida en el difusor de ventilador de 40, 45 y 50 m/s con un ancho de salida del ventilador de 115 mm y considerando el pulverizador sin movimiento y a velocidades de trabajo de 2,18, 4,5 y 6,35 km/h, se analizó el comportamiento de la aerodinámica del flujo de aire para cada variante. El aumento de la velocidad de salida en el ventilador produce un desgaste acelerado en el núcleo de la corriente, por lo que la utilización de velocidades menores favorece la aerodinámica del flujo
