A survey of Microsporum gypseum was conducted in soil samples in different geographical regions of Brazil. The isolation of dermatophyte from soil samples was performed by hair baiting technique and the species were identified by morphology studies. We analyzed 692 soil samples and the recuperating rate was 19.2%. The activities of keratinase and elastase were quantitatively performed in 138 samples. The sequencing of the ITS region of rDNA was performed in representatives samples. M. gypseum isolates showed significant quantitative differences in the expression of both keratinase and elastase, but no significant correlation was observed between these enzymes. The sequencing of the representative samples revealed the presence of two teleomorphic species of M. gypseum (Arthroderma gypseum and A. incurvatum). The enzymatic activities may play an important role in the pathogenicity and a probable adaptation of this fungus to the animal parasitism. Using the phenotypical and molecular analysis, the Microsporum identification and their teleomorphic states will provide a useful and reliable identification system.

INTRODUÇÃO: Isolar e identificar Candida spp. do ambiente, dos profissionais de saúde e de pacientes com diagnóstico presuntivo de candidíase, em Unidade de Pediatria do Hospital Universitário da Faculdade de Medicina de Jundiaí, verificar a produção de enzimas consideradas fatores de virulência e identificar a resistência das cepas isoladas dos pacientes aos antifúngicos. MÉTODOS: Foram feitas 283 coletas no período entre março e novembro de 2008 de forma randomizada do ambiente, das mãos de profissionais de saúde e de todos os pacientes com casos suspeitos de infecção hospitalar por Candida spp., triados pelo Comitê de Infecção Hospitalar. Todo o material coletado foi processado e as leveduras do gênero Candida foram isoladas e identificadas pelas características macroscópicas, microscópicas e fisiológicas. RESULTADOS: A incidência de Candida spp. no meio-ambiente e funcionários foi de 19,2%. Destes, as espécies mais frequentes entre os funcionários foram C. parapsilosis e C. tropicalis; no ar ambiente, C. guilliermondii e C. tropicalis; nas superfícies dos ambientes, C. lusitanae e nos sistemas de climatização, C. tropicalis e C. guilliermondii. No período de estudo, foram feitas 320 internações, e 13 (4%) apresentaram infecção hospitalar por Candida spp, sendo que 3 pacientes foram a óbito, dois apresentando infecção por C. tropicalis e um por C. albicans. Todas as espécies de Candida spp. isoladas dos pacientes foram sensíveis à anfotericina B, nistatina e fluconazol. CONCLUSÕES: Verificamos um aumento crescente de infecções hospitalares por Candida não albicans, o que sugere a necessidade de maiores precauções em relação a um controle ambiental destas espécies.

INTRODUCTION: This study aimed to isolate and identify Candida spp. from the environment, health practitioners, and patients with the presumptive diagnosis of candidiasis in the Pediatric Unit at the Universitary Hospital of the Jundiaí Medical College, to verify the production of enzymes regarded as virulence factors, and to determine how susceptible the isolated samples from patients with candidiasis are to antifungal agents. METHODS: Between March and November of 2008 a total of 283 samples were taken randomly from the environment and from the hands of health staff, and samples of all the suspected cases of Candida spp. hospital-acquired infection were collected and selected by the Infection Control Committee. The material was processed and the yeast genus Candida was isolated and identified by physiological, microscopic, and macroscopic attributes. RESULTS: The incidence of Candida spp. in the environment and employees was 19.2%. The most frequent species were C. parapsilosis and C. tropicalis among the workers, C. guilliermondii and C. tropicalis in the air, C. lusitanae on the contact surfaces, and C. tropicalis and C. guilliermondii in the climate control equipment. The college hospital had 320 admissions, of which 13 (4%) presented Candida spp. infections; three of them died, two being victims of a C. tropicalis infection and the remaining one of C. albicans. All the Candida spp. in the isolates evidenced sensitivity to amphotericin B, nystatin, and fluconazole. CONCLUSIONS: The increase in the rate of hospital-acquired infections caused by Candida spp. indicates the need to take larger measures regarding recurrent control of the environment.

O objetivo do presente estudo foi verificar a eficácia do acaricida maleato de estanho tri-n-butílico, aplicado industrialmente em amostras de carpetes, tecidos de revestimentos de móveis e de calçados, assim como de espumas de colchão. Aproximadamente 100 ácaros adultos da espécie Dermatophagoides pteronyssinus foram inoculados em placa de Petri contendo a amostra (pedaço de colchão, tecido ou carpete), tratada com o produto acaricida, coletados aleatoriamente. Foi acrescentado sobre a amostra, meio de cultivo para a manutenção dos ácaros. Cada placa foi examinada após 1, 2, 3, 7 e 30 dias de incubação a 25 ºC e 75% de U.R.A. (umidade relativa do ar), sob microscópio estereoscópico com 40X de aumento. O acaricida maleato de estanho tri-n-butílico apresentou 100% de eficácia acaricida após 30 dias da aplicação, em condições ótimas para a manutenção dos ácaros.

The aim of this study was to determine the effectiveness of the acaricide tri-n-butyl tin maleate, industrially applied to samples of carpets, mattress foam, and fabrics used for furniture upholstery, soft toys and shoe uppers. Approximately 100 adult house dust mites of the species Dermatophagoides pteronyssinus were inoculated into a Petri dish containing the sample (a piece of carpet, mattress foam, or fabric), treated with the acaricide, randomly collected. Mite-maintenance culture medium was added on top of each sample. After one, two, three, seven and 30 days of incubation at 25 ºC and 75% relative humidity, each dish was examined using a 40X stereoscopic microscope (40X). One hundred percent acaricide effectiveness was obtained in treated materials by the end of the 30th-day postinoculation period, under optimal conditions for mite maintenance.

Cryptococcus neoformans é um fungo que ocasiona micose de alta morbidade e mortalidade, especialmente em pacientes com Aids. Muitos reservatórios de C. neoformans têm sido relatados, mas a ecologia desta levedura deve ser ainda elucidada para se estabelecer programas de vigilância e prevenção desta infecção. O objetivo deste estudo foi o de avaliar a presença de C. neoformans no Rio de Janeiro, RJ, Brasil. Dez igrejas foram selecionadas para este estudo, coletando-se fezes de pombo, amostras de ar, das torres das igrejas e de áreas vizinhas, durante um ano. Os resultados revelaram que C. neoformans estava presente em todas as igrejas e em 37,8% das 219 amostras das excretas das aves. Ao mesmo tempo, o fungo foi isolado do solo, insetos, ovos e ninhos de pombos. Quinze (4,9%) do total das amostras de ar foram positivas. O crescimento no meio de CGB revelou que todas as amostras pertenciam a C. neoformans var. neoformans, e 98,8% destas amostras pertenciam ao sorotipo A.

Cryptococcosis has been a significant cause of morbidity and mortality in patients with Aids. Many reservoirs of the agent Cryptococcus neoformans have been reported, but the ecology of this yeast must be elucidated in order to establish surveillance programs and to prevent infections. The objective of this study was to evaluate the presence of C. neoformans in Rio de Janeiro City, RJ, Brazil. Ten churches were selected for sampling and detection of the yeast collecting pigeon dropping, air samples from church towers and neighboring areas during one year. The data demonstrated that C. neoformans has been present in every church selected and was present in 37.8% of 219 pigeon dropping samples. As well as, the yeast was isolated from soil, insects, eggs, pigeon nests and feathers. Fifteen air samples (4.9%) were positive. The growth on C.G.B. medium showed that all strains belonged to C. neoformans var. neoformans, with 98.8% of the strains belonging to serotype A.

Candida dubliniensis é uma nova espécie recentemente descrita. Este patógeno oral emergente compartilha muitas características fenotípicas e bioquímicas com C. albicans dificultando assim a diferenciação entre elas. As mesmas, porém, mostram-se genotipicamente distintas. Este trabalho tem como objetivo identificar, pela técnica de PCR (Polymerase Chain Reaction), a possível presença de C. dubliniensis dentre amostras isoladas de candidose oral eritematosa, provenientes de pacientes HIV positivos e HIV negativos. Em um total de 37 amostras identificadas anteriormente, por método clássico, como C. albicans encontramos duas amostras de C. dubliniensis (5,4%) utilizando a técnica do PCR. Esta técnica mostrou-se útil, prática e com identificação taxonômica mais acurada.

Candida dubliniensis is a new, recently described species of yeast. This emerging oral pathogen shares many phenotypic and biochemical characteristics with C. albicans, making it hard to differentiate between them, although they are genotypically distinct. In this study, PCR (Polymerase Chain Reaction) was used to investigate the presence of C. dubliniensis in samples in a culture collection, which had been isolated from HIV-positive and HIV-negative patients with oral erythematous candidiasis. From a total of 37 samples previously identified as C. albicans by the classical method, two samples of C. dubliniensis (5.4%) were found through the use of PCR. This study underscores the presence of C. dubliniensis, whose geographical and epidemiological distribution should be more fully investigated.

Com frequência o dermatófito Microsporum canis está envolvido na etiologia da dermatofitose, podendo ser transmitido para o homem por cães e gatos. Neste estudo foi procedida inoculação experimental de M. canis em cobaias resultando em lesões em 100 % dos animais e o curso clínico consistiu de período de incubação, fase inflamatória e fase de resolução das lesões. O exame histopatológico de biópsias cutâneas revelou presença de infiltrado neutrofílico e edema; acantose, hiperqueratose e espongiose. Estas lesões tornaram-se menos acentuadas no 30.0 dia pós-inoculação. Esporos e hifas de M. canis foram detectadas em cortes histológicos corados com PAS (Ácido Periódico de Schiff). A combinação dos corantes fluorescentes diacetato de fluoresceína (DF) e brometo de etídio (BE), possibilitou uma adequada visualização das células fúngicas viáveis e não viáveis com evidenciação de seus caracteres morfológicos. Este modelo experimental representa um valioso instrumento para o estudo da patogênese da infecção por dermatófitos, para a avaliação da eficácia de drogas antifúngicas, podendo também ser utilizado em estudos sobre a imunologia das dermatofitoses e na determinação da morfogênese de dermatófitos.

Dermatophytosis is a zoonosis in whose etiology the dermatophyte Microsporum canis is frequently involved. The fungus can be transmitted to man by dogs and cats. In the present study, guinea pigs were experimentally inoculated with M. canis and the course of the lesions was evaluated. Inoculation resulted in lesions in 100% of the animals, with a clinical course consisting of an incubation period, an inflammatory and a phase of lesion resolution. The histopathologic evaluation of the skin biopsies revealed the presence of acanthosis, hyperkeratosis, spongiosis and marked dermal edema. M. canis spores and hyphae were detected in histologic sections stained with periodic acid-Schiff. The combination of the fluorescent dyes FD and EB permitted the adequate visualization of viable and dead fungal cells in skin fragments of guinea pigs inoculated with M. canis and the identification of the morphologic characteristics of the cells. This experimental model represents a valuable instrument for the study of the pathogenesis of dermatophytic infection with respect to the evaluation of the efficacy of antifungal drugs, and may also be used for the study of the immunology of dermatophytoses and of dermatophyte morphogenes

A aderência é considerada como um importante fator de virulência de Candida albicans por determinar a colonização dessas leveduras em diversas regiões do hospedeiro. É a causa inicial da infecção. Neste trabalho estudamos a aderência de C. albicans (sorotipos A e B) em culturas de células Hela e Vero, com leveduras cultivadas por 18 e 30 horas em meio YNB adicionado de fontes de carboidrato como glicose, sacarose, maltose, galactose e manose (500 mM). Cada ensaio foi realizado em triplicata. Os melhores resultados foram obtidos no meio adicionado de glicose. A aderência foi maior em culturas de 18 horas, incubadas a 37ºC no tempo de contato com as células de 1 hora. O estudo da interação entre as leveduras e a cultura de células foi realizado utilizando-se a microscopia eletrônica de varredura.

The adherence of Candida albicans is considered an important virulence factor since it determines the colonization of the yeast in various regions of the host, being the initial cause of the infection. In this work we studied the adherence of C. albicans (serotypes A and B) to HeLa and Vero cells after culture for 18 h and 30 h in chemically defined medium (YNB) plus carbohydrate sources such as glucose, sucrose, maltose, galactose and mannose (500 mM). Each assay was performed in triplicate. The best results were obtained with the medium containing glucose. The adherence was greatest in 18 h cultures at 37ºC and at 1 h contact time. The interaction of yeast cells with the host cells was studied by scanning microscopy.

Cinqüenta felinos hígidos, adultos - vinte e seis machos e vinte e quatro fêmeas - sem definição racial, com distintas idades, reunidos em dois grupos (GRUPO 1- animais domiciliados e GRUPO 2 - animais que rendados) de vinte e cinco animais cada. Após anamnese e prévia sedação, os gatos foram submetidos a exame físico e à otoscopia, comprovando-se a ausência de lesões meatais e timpânicas, com cureta de Buck e wragatoas estéreis, foi colhido material meatal, que foi então submetido a exames bacteriológico e micológico. No Grupo 1, foi evidenciado presença de Otodectes cynotis (4% das amostras), em 72% dos casos de bolores e leveduras (Cladosporium sp 66,6%, Malassezia sp 40%, Penicillium sp 33%, Aspergillus sp 33,3%, Rhodotorula sp 20,0%, Mycelia sp 13,3% e Alternaria sp, Aureobasidium sp, Ryzopus sp, Trichosporon sp, todos com 6,6%) e, finalmente, em 64% da amostragem, bactérias dos gêneros Staphylococcus spp (81,2%), Pseudomonas sp (12,5%), Klebsiella sp (12,5%), Acinetobacter sp, Bacilos difteróides, Enterobacter sp, Lactobacillus spp (todos com 6,2%). No Grupo 2, o Otodectes sp foi identificado em 36% das amostras, em 96% daquelas isolaram-se fungos dos gêneros: Malassezia sp - 54,1%, Aspergillus e Penicillium sp, ambos com 33,3%, Microsporum sp - 29,1%, Cladosporium sp - 16,6%, Trichoderma sp - 12,5%, Alternaria e Phoma sp, ambos com 8,3% e Epicoccum sp, Neurospora sp. Mycelia sp, Rhodotorula sp, todos com 4,1% e, por fim, em 20 das 25 amostras (80%) isolaram-se pelo menos uma cepa bacteriana (Staphylococcus spp 75%, Klebsiella sp 20,8%, Bacilos difteróides 12,5%, Pseudomonas sp, 8,3%) e Acinetobacter sp, Enterobacter sp e Escherichia sp, todos com 4,1% cada um em cultivo monoespecífico ou em associação.

The study aimed to find out the frequency of microrganisms in the external ear canal in healthy cats. Fifty adult male (26) and female (24) short hair cats were utilized for the purpose, and they were divided according to living enviroment (Group l - indoor and Group 2 - outdoor). All animals were submitted to sedation prior to physical and otoscopic examinations in order to confirm to be healthy. Samples were obtained from ear canal by use of Buck curetes and sterile cotton sticks for microbiological investigation. In cats of Group 1, Otodects cynotis were observed in 4%. In 72% of the animals yeasts and molds were found (Cladosporium sp 66.6%, Malassezia sp 40%, Penicillium sp 33%, Aspergillus sp 33.3%, Rhodotorula sp 20%, Mycelia sp 13.3%, and 6.6% for Alternaria sp, Aureobasidium sp, Ryzopus sp and Trichosporon sp), finally, m 64% of the samples, was detected Staphylococcus spp (81.2%), Pseudomonas sp (12.5%), Klebsiella sp (12.5%), and 6,2% for Acinetobacter sp. Bacilos difteroides, Enterobacter sp, Lactobacillus spp. In Group 2, O. cynotis was isolated in 36% of cats, and in 96% of the animals wasfound Malassezia sp (54.1%), Aspergillus sp (33.3%), Penicillium (33.3%), Microsporum sp (29.1%), Cladosporium sp (16.6%), Trichoderma sp (12.5%) Alternaria sp (8.3%), Phoma sp (8.3%), Epicoccum sp (4.1%), Neurospora sp (4.1%), Mycelia sp (4.3%), Rhodotorula sp (4.1%). In 20 out of 25 (80%), at least one bacterial microrganism was isolated (Staphylococcus sp 75%, Klebsiela sp 20.8%, Bacilos difteroides 12.5%, Pseudomonas sp 8.3%, and 4.1% for Acynetobacter sp, Enterobacter and Escherichia sp).

Foi realizada a análise bioquímica, antigênica e alergênica de extratos de esporos e micélios de Pisolithus tinctorius. Os esporos foram coletados de basidiocarpos em plantações de Eucaliptus spp e os micélios obtidos de cultura em meio de MNM. Baseado na curva de crescimento do fungo, obteve-se massa miceliana aos 10, 20, 30 e 40 dias de incubação, que correspondem respectivamente ao início, meio, final da fase log e início da fase de declínio. A massa miceliana, juntamente com os esporos, foram submetidos a ação de três líquidos extratores (Coca, Tris-HCl e bicarbonato de amônio). Determinou-se os teores de carboidratos e proteínas. A análise eletroforética por SDS-PAGE revelou, além de frações comuns, frações distintas nestes extratos, em função do tempo de cultivo e métodos de extração. Os extratos selecionados para os testes alérgicos foram aqueles com maior número de frações. Realizou-se o "prick-test" em trabalhadores rurais, trabalhadores de áreas de plantações de Eucaliptus spp e em estudantes universitários, totalizando 374 pacientes. A positividade ao "prick-test" com extrato antigênico de P. tinctorius foi, respectivamente, 3,78%, 28,20% e 6,40%. A maioria dos pacientes positivos ao "prick-test" (82,75%) apresentava também sintomas de alergia respiratória (asma e/ou rinite). Não houve diferença de reatividade quando do emprego de extratos de esporos e de micélio do fungo. A análise dos soros dos pacientes positivos, revelou a presença de IgE específica aos antígenos do P. tinctorius. Considerando que o Pisolithus tinctorius é encontrado como micorriza de Eucaliptus spp e que o reflorestamento, na maioria dos países, é feito com esta planta, deve ser relevada a importância deste fungo como possível causador de alergias respiratórias, principalmente em trabalhadores ocupacionalmente expostos

The antigenic and allergenic chemical analysis of spore and mycelia extracts of Pisolithus tinctorius was carried out. The spores were collected from basidiocarps in plantations of Eucalyptus spp and the mycelia from culture in MNM medium. With basis on the fungus growth curve, the mycelia masses were obtained after 10, 20, 30, and 40 days of incubation, which correspond, respectively, to the beginning, middle and end of the log phase, and beginning of the decline phase. The mycelia masses, together with the spores, were submitted to the action of three extractors (Coca, Tris-HCl, and ammonium bicarbonate). The contents of carbohydrates and proteins were determined. The SDS-PAGE electrophoretical analysis revealed separate fractions in these extracts, besides common fractions, in function of cultivation time and extraction methods. The selected extracts for the allergic tests were the ones with the highest number of fractions. The prick-tests were conducted in 374 patients – rural workers, eucalyptus plantation works and college students. The positivity to the "prick test" with the antigenic extract of P. tinctorius was, respectively, 3.78%, 28.20% and 6.40%. Most prick-test positive patients (82.75%) also presented symptoms of respiratory allergy (asthma and rhinitis). There was no reactivity difference when the spore and mycelia extracts were employed. The analysis of the positive patients’ sera revealed the presence of IgE specific to the P. tinctorius antigens. Since Pisolithus tinctorius is found as mycorrhyza of Eucalyptus spp, and this plant is used in reforestation in most countries, the importance of that fungus should be regarded as a possible cause of respiratory allergies, especially in occupationally exposed workers

Candidina, constituída de suspensão de células leveduriformes mortas, é comumente usada em provas intradérmicas de hipersensibilidade retardada, principalmente na avaliação da competência imunológica do paciente, quando usada conjuntamente com outras provas intradérmicas do mesmo tipo. Considerando-se o estudo histopatológico de reação positiva com este tipo de antígeno, é possível obter uma reação positiva não específica na leitura da prova intradérmica. Esta pesquisa apresenta os resultados obtidos a partir da comparação entre o antígeno de suspensão celular e o antígeno polissacarídico, ambos obtidos a partir das mesmas amostras de Candida albicans. As diferenças observadas no estudo histopatológico de reações positivas com antígeno polissacarídico e com antígeno de suspensão celular podem explicar as diferenças observadas nas porcentagens entre reações intradérmicas positivas com o antígeno de suspensão celular (69,0%) e com o antígeno polissacarídico (61,0%), nas provas intradérmicas realizadas em 100 indivíduos. A coincidência de resultados positivos e negativos nesta pesquisa foi obtida em 82,0% dos indivíduos. A conclusão da pesquisa: é possível utilizar antígeno polissacarídico como candidina em provas de hipersensibilidade tipo tardio

A candidin, which is a suspension of killed yeast cells, is commonly used for intradermal tests of delayed hypersensitivity, to evaluate the immunological cellular competence of the patient, when the test is applied along with other similar tests. When working with a cellular antigen, the histopathology of positive skin tests reveals a cellular infiltrate which not only presents a characteristic hypersensitivity reaction but also a neutrophilic abscess in the central part. This research presents the results of a comparison between the yeast cell suspension and the polysaccharide antigens, both obtained from the same strains of Candida albicans. The results obtained by skin tests in one hundred individuals were 61.0% with the polysaccharide antigen and 69.0% with the yeast cell suspension antigen. Concordant results concerning the two antigens were observed in 82.0% of the individuals. The discussion section presents an assumption to explain the differences of positivity obtained with the two antigens. We conclude that the polysaccharide antigen can be utilized in the intradermal test of delayed hypersensitivity to Candida albicans.

Padronizou-se método de fluorescência (solução de diacetato de fluoresceína DF e brometo de etídio BE) para análise de viabilidade de células fúngicas, em 40 amostras de liquor, provenientes de casos comprovados de neurocriptococose. A utilização de solução aquosa de saponina a 0,3% eliminou fluorescências interferentes emitidas por hemácias e leucócitos. Após o processamento dos materiais biológicos, foram retiradas alíquotas de 0,1 ml das supensões obtidas e misturadas a volumes iguais da solução DF-BE preparada pouco antes do uso. O tempo de coloração ideal foi de 30 minutos, resultando perfeita diferenciação entre microrganismos viáveis (fluorescência verde) e não viáveis (fluorescência vermelha).

The utilization of the fluorescent method (fluorescein diacetate DF and ethidium bromide BE), to verify the viability of fungal cells, was studied in 40 samples of liquor, from patients with neurocryptococcosis. For removing leukocytes and red blood cells, which produce interfering fluorescence, good results were obtained with 0.3% saponin solution. After processing of liquor, 0.1 ml aliquots of resulting suspension were mixed to equal volumes of fresh DF-BE solution. The best incubation period for staining was 30 minutes, resulting in good differentiation between viable (green fluorescence) and non viable (red fluorescence) cells.
