Abstract This study evaluated relations among nutrients, bioactive compounds and antioxidant activity of commercial tomato varieties grown in field conditions in order to identify their biological potential by the Principal Component Analysis (PCA). Four commercial tomato varieties (Long Life, Italian, Khaki and Cherry), homogenized by their maturation stage (color, sugar/acidity ratio and pH) were evaluated regarding macro components (sugars, pectin and minerals) and functional ones (carotenoids, phenolic compounds and ascorbic acid). Cherry tomatoes stood out for their pectin and lycopene contents and their sugar/acidity ratio. Tomato samples were good sources of Cu and Ca. The PCA showed that the antioxidant potential of phenolic compounds was related to protocatecoic acid and quercetin concentrations. Italian and Cherry varieties stood out for their carotenoid contents. In field conditions, Cherry and Italian tomatoes were the best sources of functional compounds among the most common varieties found in the human diet.

The present study aimed to evaluate the lipid content and fatty acid profiles of 9 marine species (Nannochloropsis oculata, Thalassiosira pseudonana, Phaeodactylum tricornutum, Isochrysis galbana, Tetraselmis suecica, Tetraselmis chuii, Chaetoceros muelleri, Thalassiosira fluviatilis and Isochrysis sp.) and 1 freshwater microalga species (Chlorella vulgaris) that were cultivated in Erlenmeyer flasks with 800 mL of culture medium under stationary autotrophic conditions while exposed to a continuous photon flux density of approximately 175 umol m-2 s-1, a temperature of 25±2 ºC and constant airflow. N. oculata and T. pseudonana exhibited the highest lipid content, although they had a high proportion of monounsaturated fatty acids relative to polyunsaturated fatty acids. C. vulgaris and Isochrysis sp. exhibited high levels of polyunsaturated fatty acids, whereas P. tricornutum, N. oculata and T. suecica exhibited the highest eicosapentaenoic acid levels, and I. galbana and C. muelleri had the highest docosahexaenoic fatty acid levels. The freshwater species C. vulgaris had the highest levels of linoleic and linolenic acids, followed by the marine species T. chuii.

El presente estudio pretende como objetivo evaluar el perfil de ácidos grasos y contenido de lípidos de nueve microalgas marinas (Nannochloropsis oculata, Thalassiosira pseudonana, Phaeodactylum tricornutum, Isochrysis galbana, Tetraselmis suecica, Tetraselmis chuii, Chaetoceros muelleri, Thalassiosira fluviatilis e Isochrysis sp.) y una de agua dulce (Chlorella vulgaris) y la cultura autotrófica estacionaria, usando frascos que contienen 800 ml de medio de cultivo, proporcionando constante la densidad del flujo de fotones de iluminación en los cultivos de alrededor de 175 mol m-2 s-1, temperatura 25 ± 2 °C y aireación. La especie N. oculata y T. pseudonana mostraron los niveles más altos de lípidos, pero con una alta proporción de ácidos grasos monoinsaturados en relación con poliinsaturados. La especie C. vulgaris e Isochrysis sp. mostraron altos niveles de ácidos grasos poliinsaturados; P. tricornutum, N. oculata y T. suecica revelaron los niveles más altos de ácido eicosapentaenoico y C. muelleri e I. galbana alcanzaron los más altos niveles del ácido graso docosahexaenoico. C. vulgaris reveló un mayor contenido de ácidos linoleico y linolénico, seguido de las especies marinas T. chuii.

Este trabalho investigou o conteúdo de compostos fenólicos totais e a atividade antioxidante em farelo de arroz fermentado, com o objetivo de avaliar o efeito da fermentação em estado sólido nessas propriedades. O processo foi realizado com o fungo Rhizopus oryzae CTT 1217 em reatores de bandeja a 30 °C por 120 horas. Amostras de farelo de arroz fermentado foram coletadas a cada 24 horas. A capacidade antioxidante foi avaliada pelo método do sequestro do radical livre difenil-1-picrilhidrazil e também por meio da inibição da oxidação enzimática e da peroxidação lipídica do óleo de oliva. O extrato metanólico produzido com a biomassa de 96 horas sequestrou 50% do radical livre em 15 minutos. O mesmo extrato reduziu o índice de peróxido no óleo em 57% após 30 dias de armazenamento. O extrato aquoso obtido da biomassa de 120 horas foi o inibidor mais eficiente da reação de escurecimento enzimático catalisada pela peroxidase.

This study investigated the content of total phenolic compounds and antioxidant activity in fermented rice bran in order to evaluate the effect of solid state fermentation on these properties. The process was performed with the fungus Rhizopus oryzae CTT 1217 in tray reactors at 30 °C for 120 hours. Samples of fermented rice bran were collected every 24 hours. Antioxidant property was evaluated by the diphenyl-1-picrylhydrazyl radical scavenging method and through the inhibition of enzymatic oxidation and lipid peroxidation of olive oil. The methanol extract of the biomass obtained at 96 hours of fermentation inactivated 50% of free radical in 15 minutes. The same extract reduced the peroxide value in the olive oil by 57% after 30 days of storage. The aqueous extract of the biomass obtained at 120 hours was the most efficient inhibitor of the darkening reaction catalyzed by peroxidase.

Este trabalho teve como objetivo estudar a utilização de películas, à base de compostos proteicos e fenólicos provenientes de farelo de arroz fermentado, em tomates (Lycopersicum esculentum). Foram realizados testes com: glicerol 3% (v/v); glicerol com extrato fenólico e proteico do farelo de arroz (5%); glicerol com extrato fenólico e proteico da biomassa gerada em 96 hours (5%), e um controle (sem a película). Os tomates revestidos foram mantidos à temperatura ambiente durante 28 dias, sendo determinados, a cada 96 horas, os seguintes aspectos: a perda de massa, o pH e a acidez, os sólidos solúveis totais e os carotenoides. A película elaborada com os extratos da biomassa reduziu os níveis de carotenoides e acidez dos frutos estudados em 17 e 21,1%, respectivamente, em relação ao controle. A película também foi eficiente como barreira ao vapor de água; assim, com perda de massa 57% inferior à do controle, sugere-se que esta poderá ser utilizada como alternativa para conservação desse tecido vegetal.

The aim of this study was to investigate the use of protein-phenolic based coating made from fermented rice bran on cherry tomatoes (Lycopersicum esculentum). Tests were performed with glycerol 3% (v/v), glycerol with protein-phenolic rice bran extract (5%), glycerol with protein-phenolic extract after 96 hours of fermentation (5%), and a control (without coating). The coated cherry tomatoes were kept at room temperature for 28 days. Mass loss, pH and acidity, total soluble solids, and carotenoids were determined every 96 hours. The coating made from the biomass extract reduced the carotenoid and acidity levels in the fruits studied by 17 and 21.1%, respectively, compared to the control. The coating proved an efficient barrier to water vapor with mass loss of 57% less than the control suggesting that it can be used as an alternative for vegetable tissue conservation.

The work herewith investigated the production of yeast biomass as a source of protein, using Yarrowia lipolytica NRRL YB-423 and raw glycerol from biodiesel synthesis as the main carbon source. A significant influence of glycerol concentration, initial pH and yeast extract concentration on biomass and protein content was observed according to the 2v5-1 fractional design. These factors were further evaluated using a central composite design and response surface methodology, and an empirical model for protein content was established and validated. The biomass of Yarrowia lipolytica NRRL YB-423 reached 19.5 ± 1.0 g/L in shaken flasks cultivation, with a protein content of 20.1 ± 0.6% (w/w).

An evaluation of the pesticides extraction from onion using a modern sample preparation method (QuEChERS) and determination by liquid chromatography tandem mass spectrometry was carried out. All the calibration curves showed r>0.99. The recoveries ranged between 61.8 and 120.0% with relative standard deviation lower than 20% for all compounds. Due to the occurrence of matrix effect, the quantification was performed using matrix-matched calibration. The limits of quantification of the method were between 0.0005 and 0.05 mg kg-1. The method shows the advantages of not require the clean-up step and consume low volume of organic solvents, decreasing time, costs and residues.

O objetivo deste trabalho foi relacionar os níveis de fenóis totais em cebola, farelo de arroz e microalga Chlorella phyrenoidosa com a atividade antifúngica testada contra o fungo Rhyzopus oryzae. Os compostos fenólicos da cebola foram extraídos em três sistemas solventes: aquoso, metanólico e com acetato de etila, os do farelo de arroz e da microalga Chlorellaphyrenoidosa extraídos com metanol. Os extratos foram quantificados colorimetricamente com reagente de Folin-Ciocalteau. O método para avaliar a atividade antifúngica empregou a técnica de Ágar diluído. O Rhyzopus oryzae teve seu desenvolvimento inibido em presença de extratos metanólico e acetoetílicos de cebola e metanólico de farelo de arroz, que continham as concentrações de compostos fenólicos testadas, 86; 2,6; e 46 µg fenóis.mL-1meio, respectivamente. O extrato aquoso de cebola e metanólico de Chlorella phyrenoidosa não inibiram o crescimento fúngico em nenhuma das concentrações fenólicas testadas.

The objective of this work was to relate the levels of total phenolic in onion, rice bran, and Chlorellaphyrenoidosa with the antifungal activity tested against the fungus Rhyzopus oryzae. The phenolic compounds of onion were extracted in three solvent systems: aqueous, methanolic, and with ethila acetate, whereas the phenolic compounds of rice bran and Chlorella phyrenoidosa were extracted with methanol. The extracts were quantified with the Folin-Ciocalteau reagent. The method to evaluate the antifungal activity employed was the diluted Ágar technique. The Rhyzopus oryzae had its development inhibited in the presence of onion methanolic and aceto-ethylic extracts and rice bran methanolic extracts at the following phenolic concentrations, 86; 2.6, and 46 µg phenolic.mL-1agar, respectively. The aqueous extracts from onion and the methanolic extract from Chlorellaphyrenoidosa did not inhibit fungal growth under the phenolic concentrations tested.

O objetivo deste trabalho foi avaliar a disponibilização de nutrientes no farelo de arroz através de um processo de fermentação utilizando Saccharomyces cerevisiae como inóculo, bem como caracterizar os farelos não fermentado e fermentado química e nutricionalmente. Os parâmetros definidos para a fermentação foram 3% de levedura, 30% de umidade e um intervalo de 6 horas a 30 °C. A caracterização físico-química foi realizada segundo AOAC (2000), a digestibilidade in vitro e metionina disponível por método enzimático, cálcio por complexometria, açúcares redutores por espectrofotometria e micotoxinas pelo multimétodo de TANAKA (2001). Observou-se, que os açúcares redutores variaram de 3,4 a 4,8% durante 6 horas de fermentação. Comparando-se o farelo não fermentado com o fermentado, o pH diminuiu de 6,5 para 5,8, a acidez, os minerais e as fibras variaram de 2,1 a 4,7%, de 10,5 a 11,9% e de 9,4 a 9,9%, respectivamente. A digestibilidade in vitro e os teores de lipídios, proteínas e cálcio não variaram significativamente com a fermentação.

The objective of this work was to evaluate the availability of nutrients in rice bran through a solid-state fermentation process, using Saccharomyces cerevisiae as the inoculum, and to characterize the fermented and non-fermented bran physicochemically and nutritionally. The parameters defined for the fermentation were 3% yeast, 30% humidity and an interval of 6 hours at 30 °C. The physicochemical characterization was conducted according to an official methodology (AOAC, 2000), the pH and acidity were determined by the Adolfo Lutz Institute (1985) method, in vitro digestibility and available methionine by the enzymatic method, calcium by complexometry, reducing sugars by spectrophotometry and mycotoxins by TANAKA's multimethod (2001). The reducing sugars were found to vary from 3.4 to 4.8% during a 6 hours fermentation period. A comparison of the non-fermented and fermented bran indicated that the pH, available methionine and carbohydrates decreased, respectively, from 6.5 to 5.8, from 4.4 to 4.2 mg.g-1 and from 46.3 to 44.1%, while acidity, ashes and fibers varied from 2.1 to 4.7%, 10.5 to 11.9% and 9.4 to 9.9%, respectively. The in vitro digestibility and lipids, protein and calcium contents did not vary significantly with the fermentation process.

Farelo de arroz desengordurado e farelo de trigo foram submetidos à fermentação em estado sólido pelos fungos Rhizopus sp. e Aspergillus oryzae, visando a obtenção de um produto enriquecido em proteínas, reaproveitando um subproduto da indústria beneficiadora de cereais. Os farelos fermentados obtidos foram caracterizados quanto aos seus compostos nitrogenados. Foi determinado o conteúdo protéico, de aminoácidos totais e digeríveis e de metionina disponível. Os maiores aumentos nos teores protéicos dos farelos foram observados com o microrganismo Rhizopus sp.. Os teores de aminoácidos totais, aminoácidos digeríveis e metionina disponíveis foram aumentados nos farelos fermentados por ambos os fungos, sendo que o escore químico, após 72 horas, variou entre 37,5 e 54,3%.

Defatted rice bran and wheat bran were subjected to solid-state fermentation by the fungi Rhizopus sp. and Aspergillus oryzae to increase their nutritional value, especially their protein content. The fermented bran was analyzed to determine the protein, total amino acids, digestible amino acids and disposable methionine content. The highest increase in protein content in fermented bran occurred with the use of Rhizopus sp., while total amino acids, digestible amino acids and available methionine increased with both fungi. After 72 hours of fermentation the chemical score varied from 37.5 to 54.3%.

A cianobactéria Spirulina tem sido comercializada e estudada devido as suas propriedades nutricionais e terapêuticas no tratamento de doenças como o câncer, a hipercolesterolemia e a aterosclerose. Determinados compostos presentes na Spirulina, como os compostos fenólicos, a ficocianina e o tocoferol, podem apresentar atividade antioxidante. O principal objetivo deste trabalho foi avaliar o efeito da temperatura de cultivo e da concentração de nitrogênio no meio de cultivo, sobre o potencial antioxidante da microalga Spirulina (Arthospira) platensis. Quando a microalga foi cultivada a 35ºC e concentrações de nitrato de sódio de 1,875 g.L-1 ou 2,5 g.L-1, o potencial antioxidante dos extratos obtidos a partir da biomassa, sobre o escurecimento enzimático causado pela peroxidase, foi de 29% e 35%, respectivamente, sendo a redução no escurecimento relacionada com as quantidades de compostos fenólicos presentes nos extratos.

The main aim of this work was to investigate the effects of temperature and nitrogen concentration on the antioxidant potential of extracts from Spirulina (Arthospira) platensis biomass. S. platensis biomass obtained at 35ºC and with 1.875 g.L-1 or 2.5 g.L-1 of sodium nitrate in the culture medium presented higher concentrations of phenolic compounds. The antioxidant potential of methanol extracts of biomass on the enzymatic browning caused by peroxidase were 29 and 35%, respectively, being the reduction related to the amount of phenolic compounds present in this extract.

The aim of the study was to investigating the antihypertensive effect of the aqueous extract from skin fish Stephanolepis hispidus (AqeSh) in normotensive and N G-nitro-L-arginine methyl ester (L-NAME)-induced hypertension. The treatment of rats with L-NAME (60 mg/l, drinking water) caused a sustained increased in rat blood pressure (BP). AqeSh restored rat BP to normal levels in the L-NAME-treated hypertensive rats. Moreover, AqeSh was able to prevent the increase of BP induced by L-NAME. At the end of the treatment no difference in body weight and vital organs were noted between the control and treated groups. Hematological and biochemical studies revealed no differences in treated animal except AST and ALT profile, may due to the presence of L-NAME treatment. These results suggest that oral administration of AqeSh prevent the increase of BP in L-NAME-induced hypertension that may be mediated partially by nitrite and nitrate present in the skin.

Eso estudio ha tenido como objetivo investigar el efecto antihipertensivo del extracto acuoso del piel del pez Stephanolepis hispidus en el normotenso y N G-nitro-L-arginine metilo ester (L-NAME) hipertensión-inducido ratos. El tratamiento de ratas con el L-NAME (60 mg/l) causó un persistente aumento en la tensión arterial (TA). El extracto acuoso de Stephanolepis hispidus (EacSh) restauró la TA a los niveles normales en las ratas hipertensas tratadas con L-NAME. Además, EacSh pudo prevenir el aumento de TA inducido por el L-NAME. Al final del tratamiento no se observó ninguna diferencia en el peso del cuerpo y órganos vitales entre los grupos controles y tratados. Estudios hematológico y bioquímicos no revelaron diferencias entre los grupos controles y tratados, excepto los perfiles de AST y ALT, lo que puede haber sido por la presencia de L-NAME. Estos resultados sugieren que la administración oral de EacSh puede previnir el aumento de TA en ratas con hipertensión inducida por L-NAME, lo que puede estar mediado parcialmente por la presencia del nitrite y nitrato en lo EacSh.

O objetivo deste estudo foi o de investigar o efeito antihipertensivo do extrato aquoso da pele do peixe Stephanolepis hispidus (AqeSh) em ratos normotensos e hipertensos induzidos pelo tratamento com N G-nitro-L-arginina metil ester (L-NAME). O tratamento dos animais de experimentação com L-NAME (60 mg/l, água de beber) provocou um aumento sustentado na pressão arterial. O tratamento com AqeSh restaurou a pressão sanguínea para os níveis normais nos animais hipertensos. Além disso, o AqeSh preveniu o aumento da pressão sanguínea induzida pelo L-NAME. Ao final dos tratamentos não foram observadas diferenças significativas nos pesos corporal e dos órgãos vitais entre o grupo controle e os tratados. A avaliação hematológica e bioquímica não revelou diferenças entre os grupos exceto para os valores de AST e ALT no grupo hipertenso, provavelmente devido à presença do tratamento com L-NAME. Estes resultados sugerem que a administração oral do AqeSh previne o aumento da pressão sanguínea induzida pelo L-NAME. Este efeito pode ser mediado parcialmente por nitrito e nitrato presentes na pele do peixe.

The objective of this paper was to determine nitrate and nitrite anions in toasted fish skin tea, which the population of southern Rio Grande do Sul believes to have pharmacological properties for trating asthma. Two methods were compared, an official and one described by Guozhen et al., that has the advantage of not needing reduction of nitrate to nitrite on a Cd column. The samples were the skin of the fish species Balistes capriscus (peixe porco) and Menticirrhus littoralis (papa-terra), both crude and toasted, as traditionally used. The method of Guozhen et al. was used due to its performance (detection limit 0.005 µmol/L NO3- and 0.02 µmol/L - NO2- and mean recuperation of 100 and 97% respectively for each anion) and results similar to those of the official methods under conditions used by the population, the estimated daily intake, of the teas is equivalent to 0.03 to 0.05 mg of nitrate and 0.007 to 0.014 mg of nitrite. These values are in accord with safety recommendations and don't represent a health risk.

SUMMARY. Evaluation of the AOAC 985.29 enzimic gravimetric method for determination of dietary fiber in oat and corn grains. The precision attributes and use of the enzymatic-gravimetric method of Prosky et al. (1992) (AOAC 985.29) were evaluated using corn (BR 5202 Pampa) and oat (UFRGS 15) samples. The effect of laboratory batches carried out in different days were evaluated in six laboratory batches, using for each material one duplicate for total fiber (FT) determination, one duplicate for insoluble fiber (FI) determination and blank ones for FT and for FI (both in duplicate). In order to characterize repetitive aspects, five other FT and FI determinations added to each sample were evaluated, summing up 11 data. The low coefficients of variation in the first six batches were considered acceptable as an expression of expected total intralaboratory variation. The repetitive of the method was considered good for FT determinations (CVs < 10%). However, in the FI determination a high frequency of negative values of ash and blanks was found, impairing the repetitive aspects evaluation. The magnitude of the total gravimetric corrections varies with the kind of the sample and is especially influenced by the protein content.

RESUMO. Foram avaliados os atributos de precisão e o uso do método enzímico-gravimétrico descrito por Prosky et al. (1992) (AOAC, 985.29) para a determinação de fibra alimentar em grãos de cereais, utilizando-se milho (BR 5202 Pampa) e aveia (UFRGS 15). Para determinar o efeito de corridas laboratoriais executadas em diferentes dias, organizou-se seis corridas para cada amostra, cada qual formada por uma duplicata para a determinação de fibra total (FT), uma para fibra insolúvel (FI) e provas em branco, também em duplicata, para FT e FI. A fim de caracterizar a repetibilidade, outras cinco determinações de FT e FI foram realizadas para cada uma das amostras, totalizando 11 dados. Os coeficientes de variação de FT e FI obtidos nas 6 primeiras corridas foram baixos, indicando uma variação intralaboratorial total aceitável. Os CVs (<10%) encontrados para FT de milho e aveia indicaram alta repetibilidade do método para essa fração da fibra alimentar. Nas determinações de FI, observou-se alta freqüência de valores negativos para correções de cinzas e provas em branco, que diminuíram a confiabilidade dos resultados finais. A magnitude do total das correções gravimétricas variou com a natureza da amostra e foi grandemente influenciada pela correção de proteína.

Os tricotecenos são metabólitos produzidos por diversos fungos, principalmente os do gênero Fusarium. As condições climáticas de diversos países, como o Brasil, favorecem o crescimento desses fungos e a produção dos tricotecenos, os quais podem estar presentes em vários cereais, como por exemplo a cevada, que pode ser utilizada em processos fermentativos visando a obtenção de álcool. Com o objetivo de estudar a influência que essas micotoxinas apresentam na produção de álcoois, um processo fermentativo em pequena escala foi simulado. Malte foi contaminado com deoxinivalenol e toxina T-2 em diferentes quantidades (até 500 ppb), de acordo com um planejamento fatorial 3², e os álcoois obtidos após a fermentação foram determinados por cromatografia gasosa. Os resultados mostraram que os tricotecenos podem influenciar o perfil dos álcoois produzidos pela levedura Saccharomyces cerevisiae durante o processo fermentativo do malte, especialmente dos álcoois metílico e isoamílico.

In order to study the influence of mycotoxins on the production of alcohols, a fermentative process on a laboratorial scale was simulated. Malt was contaminated with deoxynivalenol and T-2 in different ratios (up to 500 ppb), according to a 3² factorial design, and the alcohols obtained after the fermentation were determined through gas chromatography. The results showed that trichothecenes influenced the profile of the alcohols produced by Saccharomyces cerevisiae during the fermentative process of malt, especially the profile of methyl and isoamyl alcohols.
