O objetivo deste estudo foi avaliar a relação entre a atividade total das esterases e a resistência de indivíduos de M. persicae a inseticidas em populações coletadas em plantas de batata, em Contenda e Piraí do Sul, PR e Ibicoara, BA. No bioensaio, os afídeos foram colocados em recipientes de plástico transparente com o fundo coberto por uma tela de malha fina, em cinco repetições de 10 indivíduos para cada concentração. O conjunto foi mergulhado em concentrações (%) de pirimicarbe a 0,0031; 0,0062; 0,0125; 0,025; 0,05 e 0,1%, durante 10 segundos e depois secos em papel filtro. Após uma hora em câmaras a aproximadamente 25°C, os afídeos foram examinados sob microscópio estereoscópico, anotando-se o número de insetos mortos. A atividade de carboxilesterase foi medida através de ensaio colorimétrico in vitro. De acordo com os níveis de absorbância, os afídeos foram classificados em S; S/R1 (S ou R1); R1; R1/R2 (R1 ou R2); R2 e R3 (suscetível; suscetível/ levemente resistente; levemente resistente; levemente resistente/ altamente resistente; altamente resistente e extremamente resistente, respectivamente). Foi efetuada uma correlação simples entre os resultados das CL50 obtidas no bioensaio e a freqüência desses indivíduos. Encontrou-se uma correlação positiva entre a CL50 e R2 , mas negativa entre a CL50 e S e S/R1. O bioensaio, apoiado pelo teste colorimétrico, mostrou-se como uma ferramenta capaz de estimar com precisão tanto uma população com alta freqüência de indivíduos suscetíveis quanto outra com elevada proporção de indivíduos resistentes e altamente resistentes.

The objective of this research was to evaluate the relationship between the total activity of carboxylesterase and the resistance of M. persicae populations to pirimicarb. The aphids were collected on potato plants in Contenda and Piraí do Sul, PR and Ibicoara, BA, Brazil. For the bioassay, five replicates for each concentration were prepared with 10 aphids placed in plastic vials with a fine fabric mesh on the bottom, and dipped into pirimicarb concentrations (%) at 0.0031; 0.0062; 0.0125; 0.025; 0.05, and 0.1%, during 10 seconds, then dried on filter paper. After 1h inside a 25°C chamber, the aphids were examined under a stereomicroscope, recording the number of dead insects. The carboxylesterase activity was assessed by in vitro colorimetric technique. A simple correlation was established between the results of the LC50 bioassay and the frequency of M. persicae resistant variants. According to the absorbance level, the aphids were classified as variants S; S/R1 (S or R1); R1; R1/R2 (R1 or R2); R2 and R3, respectively susceptible; partially resistant, resistant and highly resistant. It was found a significant positive correlation between the LC50 and the frequency of R2 forms and a significant negative correlation between LC50 and the frequency of S and S/R1 forms. The colorimetric data supported the bioassay results, indicating that the bioassay was precise enough to determine either populations with high frequency of susceptible individuals or population with high proportion of resistant and highly resistant individuals.

Resultados de bioesnsaios com pirimicarbe, associados aos níveis de esterases totais obtidos através de testes colorimétricos, foram utilizados nesta pesquisa para determinar concentrações diagnostica para a avaliação da resistência de populações de Myzus persicae a inseticidas em cultura de batata Coletas de M. persicae foram feitas ao acaso em plantas de batata de plantações comerciais, na região de Curitiba, Piraí do Sul e Contenda, no Paraná, e Ibicoara na Bahia, durante o período de 1994 a 1996. A CL50 foi estimada submetendo-se os afídeos a bioensaios com concentrações (g/100 ml) de 0.0125; 0.025; 0.05 e 0,1% foram congelados uma hora após o tratamento e submetidos à avaliação das esterases totais, através de ensaio colorimétrico. Na concentração de 0,0125%, a mortalidade foi muito baixa, enquanto que a 0,1% a mortalidade atingiu 100% da população, indicando que a concentração diagnostica encontra-se dentro desta faixa. A diferença foi significativa entre as mortalidades nas concentrações de 0,025 e 0.05% em cada população estudada, mas não entre as populações coletadas nas quatro regiões. A concentração de 0,05% de pirimicarbe causa a mortalidade de aproximadamente 50% dos indivíduos altamente resistentes (R2), 72% dos levemente resistentes (R1) e 89,7% dos suscetíveis (S), podendo ser utilizada para diagnosticar populações resistentes de M. persicae. A determinação desta concentração diagnóstica é importante nos programas de manejo da resistência da praga em cultivo de batata, pois os indivíduos podem ser avaliados quanto à resistência nesta única concentração e, assim, baixa freqüência de resistência podem ser detectadas.

Insecticide bioassays associated with colorimetric testes were used to determine the diagnostic concentrations of pirimicarb for insecticide resistance detection in Myzus persicae populations from potato crops. The aphids were randomly collected on potato plants of commercial crops in Curitiba, Piraí do Sul and Contenda, in Paraná State and Ibicoara in Bahia state, from 1994 to 1996. The LC50 was estimated by submitting the aphids to bioassay with increasing concentrations of pirimicarb. Dead and live aphids from the 0.0125; 0.025; 0.05 and 0.1% (g/100ml) insecticide treatments were frozen after one hour of the bioassay and submitted to the evaluation of total esterase by colorimetric assays. Mortality was low at 0.0125%, but at 0.1% it reached 100% of the populations; thus, indicating that the diagnostic concentration is within this range. A significant difference was found between mortality caused by 0.025 and 0.05% pirimicarb in the same population, but not between populations collected from different areas. The 0.05% primicarb concentrations caused mortality of nearly 50% of high resistant individuals (R2), 72.0%of the moderately resistant populations (R1) and 89.7% of susceptible ones (S). The determination of this diagnostic concentration is useful for M. persicae resistance management programs because all individuals can be tested at this single appropriate concentration and low resistance frequencies can be detected in the population.

A freqüência e a intensidade da resistência de Myzus persicae (Sulz.) a inseticidas foram avaliadas em duas áreas localizadas no município de Ibicoara, BA, onde as aplicações com inseticida falharam no controle do afídeo. Em uma das áreas com a cultura em sua fase final, foram feitas duas aplicações de pirimicarbe, com intervalo de quatro dias, enquanto a outra não recebeu pulverizações. Foram feitas seis coletas ao acaso, de 700 afídeos cada, antes e após as aplicações e também na área não tratada e em plantas voluntárias (plantas que nasceram espontaneamente em áreas cultivas com batata em safras anteriores) de batata. No laboratório, foi estimada a CL50 das populações de campo, que foram posteriormente submetidas à avaliação de esterases totais através de teste colorimétrico. Após as aplicações de pirimicarbe, a freqüência da população suscetível (S), moderadamente resistente (R1) e das populações mistas (S/R1) reduziu-se de 36,6 para 9,9; 12 para 7,5 e 11,4 para 5,9%, respectivamente. As populações altamente resistente (R2), extremamente resistente (R3) e a mista R1/R2, entretanto tiveram a sua freqüência elevada após as aplicações (17,7 para 36,7; 2,3 para 9,1 e 20 para 30,8%, respectivamente). A sobrevivência de indivíduos S foi devida, possivelmente, ao incremento da movimentação para evitar o contato com o inseticida. Todas as mudanças na freqüência de indivíduos resistentes e suscetíveis de M. persicae refletiram-se na CL50 e na razão de resistência (RR), demonstrando a sensibilidade do bioensaio. Na área não tratada com pirimicarbe observou-se alta freqüência de R2 + R3 (86,9) e R1/R2. A alta freqüência de populações de M. persicae resistentes a inseticidas e a elevada intensidade da resistência observadas nesta pesquisa podem estar ligadas à seleção de indivíduos resistentes e à entrada de migrantes alados resistentes provenientes das plantas voluntárias , que apresentaram freqüência de R2 + R3 (81,4%).

The frequency and intensity of Myzus persicae (Sulz.) resistance were studied using bioassays and biochemical tests in two potato crops in Ibicoara, Bahia. Both areas received several insecticide applications which failed to control M. persicae. By the end of the crop season, one of the areas received two applications of pirimicarb and the other was not sprayed. Six samples of 700 aphids each were randomly collected before and after the pirimicarb applications, with an interval of four days. Specimens of M. persicae were also collected from isolated potato plants. At the laboratory, the samples were characterized by the CL50 based on insecticide bioassays and by total esterase activity using colorimetric assays. After the pirimicarb applications, the susceptible (S), the moderate resistant (R1) and mixed S/R1populations presented decreasing resistance frequencies (from 36.6 to 9.9; 12,0 to 7.5 and 11.4 to 5.9%, respectively). On the other hand, high resistant individuals (R2), extremely resistant (R3), and mixed populations of R1/R2 had increasing frequencies (from 17.7 to 36.7%; 2.3 to 9.1%, and 20.0 to 30.8%, respectively). The survival of S individuals was probably due to their wandering behavior on the plants to avoid sprayed areas. All changes in frequency were reflected in the LC50 and in the resistance ratio (rr). In the pirimicarb untreated area, high frequencies of R2, R3 and R1/R2 were observed. High frequency and resistance intensity of M. persicae in areas under intensive insecticide applications can be related to the selection of resistant populations and due to the entrance of winged migrants from spontaneous plants, where the frequency of R2+R3 was 81.4%.

The efficiency of insecticides against Sitophilus oryzae (Linnaeus, 1763) (Coleoptera, Curculionidae) and Rhyzopertha dominica (Fabricius, 1792) (Coleoptera, Bostrichidae) was determined exposing them to treated wheat in laboratory. Each plot, which was composed by 1.2 kg of wheat grain, was treated with 5 ml of the concentration/kg of wheat using a propelling air atomizer. Fifteen days after treatment 50 g samples of the grain were taken, from each plot, placed in plastic jars into which 10 adult insects of each species were released. This was repeated every 30 days until 165 days after treatment. The number of dead insects was counted 15 days after each infestation thus the evaluations were made at 30, 60, 90, 120, 150 and 180 days after the insecticide application. Esfenvalerate+fenitrothion+pyperonil butoxide (0,375+7,5+3,0 and 0,5+10,0+4,0 g a.i./t), fenitrothion+esfen-valerate (7,5+0,375 and 10,0+0,375 g a.i./t), fenitrothion+synergised deltamethrin (7,5+0,375 g a.i./t) and fenitrothion (7,5 g a.i./t) were effective against Sitophilus oryzae until 180 days after treatment. Esfenvalerate+fenitrothion+pyperonil butoxide (0,5+10,0+4,0 g a.i./t), fenitrothion+esfenvalerate (7,5+0,375 and 10,0+0,375 g a.i./t), fenitrothion+synergised deltamethrin (7,5+0,375 g a.i./t) were effective against R. dominica until 180 days after treatment. Synergised deltamethrin (0,375 g a.i./t) was effective against S. oryzae until 150 days and against R. dominica until 120 days after treatment. Esfenvalerate (0,5 g a.i./t) was ineffective against S. oryzae and effective until 60 days after treatment against R. dominica. Fenitrothion (7,5 g a.i./t) was ineffective against R. dominica. The use of mixtures of organophosphorus plus pyrethroids were more effective against S. oryzae and R. dominica when compared with these insecticides used alone.

Determinou-se em laboratório a eficiência de inseticidas contra Sitophilus oryzae (L.) (Coleoptera: Curculionidae) e Rhyzopertha dominica (Fab.) (Coleoptera: Bostrichidae) em milho (Zea mays) a granel tratado. Cada tratamento, constituído por 1,2 kg de milho, foi tratado por meio de um pulverizador equipado com bico pneumático usando-se 5 ml de calda/kg de milho. Quinze dias após os tratamentos 200 g de milho, de cada tratamento, foram infestadas com 40 insetos adultos de cada espécie, sendo este procedimento repetido a cada trinta dias até completar 180 dias. A contagem do número de insetos mortos foi realizada 15 dias após cada uma das infestações, desta forma foram feitas avaliações aos 30, 60, 90, 120, 150 e 180 dias após a aplicação dos inseticidas. Após 180 dias esfenvalerate + fenitrothion + butóxido de piperonila, esfenvalerate + fenitrothion e fenitrothion + deltametrina + butóxido de piperonila foram eficientes contra S. oryzae e R. dominica. Fenitrothion foi eficiente contra S. oryzae e deltametrina + butóxido de piperonila contra R. dominica. Esfenvalerate + butóxido de piperonila e esfenvalerate mostraram-se ineficientes contra S. oryzae, o mesmo sendo observado para fenitrothion contra R. dominica. O uso de misturas de organofosforados e piretróides foi mais eficiente contra S. oryzae e R. dominica quando comparado com o uso isolado destes inseticidas.

The number of dead insects was counted 15 days after each infestation, so the evaluations were made at 30, 60, 90, 120, 150 and 180 days after the insecticide application. After 180 days esfenvalerate + fenitrothion + piperonyl butoxide, esfenvalerate + fenitrothion and fenirothion + deltamethrin + pyperonil butoxide were effective against S. oryzae and R. dominica. Fenitrothion was effective against S. oryzae and deltamethrin + piperonyl butoxide against R. dominica. Esfenvalerate + pyperonil butoxide were ineffective against S. oryzae and the same was observed with fenitrothion against R. dominica. The use of mixtures of organophosphorus plus pyrethroids were more effective against S. oryzae and R. dominica when compared with these insecticides used alone.

Determinou-se a eficiência de inseticidas contra Sitophilus oryzae (L.) (Coleoptera: Curculionidae) e Rhyzopertha dominica (Fab.) (Coleoptera: Bostrichidae), em arroz (Oryza sativa) armazenado tratado. Os tratamentos foram feitos usando um pulverizador equipado com bico pneumático, sendo utilizado para todos os tratamentos 5 ml de calda/kg de arroz, exceto na testemunha tratada apenas com água. Para cada tratamento foram usados 1,2 kg de arroz, que foram tratados em sacos plásticos e em seguida acondicionados em sacos de ráfia e armazenados em sala climatizada (25 ± 2ºC). Quinze dias após os tratamentos, 200 g de arroz de cada um dos tratamentos, foram infestados com 40 insetos adultos de cada espécie, sendo este procedimento repetido a cada 30 dias. A contagem do número de insetos mortos foi realizada 15 dias após cada infestação aos 30, 60, 90, 120, 150 e 180 dias após a aplicação dos inseticidas. Após 180 dias da aplicação o Sumigran plus (15 e 20 ml/t) foi eficiente no controle de S. oryzae. No controle de R. dominica, até os 180 dias após a aplicação, foram eficientes: Sumigran plus (15 e 20 ml/t), Sumigran ® + Sumidan ® (15 + 15 e 20 + 20 ml/t), Sumigran ® + K-Obiol ® (15 + 15 ml/t).

The insecticides were applied using a propelling air atomizer (5 ml of mixture/kg of rice); for the control only water was used. For each treatment 1.2 kg of stored rice, were treated in plastic bags and stored in raffia bags in a climatized room (25 ± 2ºC). Fifteen days after treatment, 200 g of the treated rice, from each treatment, were infested with 40 adults of each species. This procedure was repeated every 30 days. The number of dead insects was recorded 15 days after infestations, at 30, 60, 90, 120, 150 and 180 days after the insecticide application. Sumigran plus (15 and 20 ml/t) was effective against S. oryzae until 180 days after treatment. Sumigran plus (15 and 20 ml/t), Sumigran ® + Sumidan ® (15 + 15 and 20 + 20 ml/t), Sumigran ® + K-Obiol ® (15 + 15 ml/t), were effective against R. dominica up to 180 days after treatment.
