Abstract In this study, we aimed to assess the spatial variability of microclimate inside a closed compost-bedded pack barn (CBP) with a negative ventilation system during summer and winter. The research was carried out in a CBP located in the Zona da Mata region, Minas Gerais, Brazil. For each of the stations analyzed, the following environmental mean variables observed inside a CBP were measured: air dry-bulb temperature (tdb), air relative humidity (RH), and windspeed, Temperature-Humidity index, and specific enthalpy. The kriging maps showed that the most critical housing conditions in the thermal environment were found, mainly, from the central part of the CBP, close to the exhaust fans. The analyses also pointed out that the system presented temperature gradients along the length, up to 3°C. During the summer afternoon, the entire region of the CBP was in a discomfort situation (tdb>26°C; RH>75%). During the winter, the measured environmental data remained within the comfort zone throughout the facility. However, probably due to the lack of thermal insulation of the material used to close the sides of the CBP, it did not allow spatial thermal uniformity for both seasons. It was also inefficient to keep the animals within the comfort zone for lactating cattle during the critical summer period.

The scientific publication landscape is changing quickly, with an enormous increase in options and models. Articles can be published in a complex variety of journals that differ in their presentation format (online-only or in-print), editorial organizations that maintain them (commercial and/or society-based), editorial handling (academic or professional editors), editorial board composition (academic or professional), payment options to cover editorial costs (open access or pay-to-read), indexation, visibility, branding, and other aspects. Additionally, online submissions of non-revised versions of manuscripts prior to seeking publication in a peer-reviewed journal (a practice known as pre-printing) are a growing trend in biological sciences. In this changing landscape, researchers in biochemistry and molecular biology must re-think their priorities in terms of scientific output dissemination. The evaluation processes and institutional funding for scientific publications should also be revised accordingly. This article presents the results of discussions within the Department of Biochemistry, University of São Paulo, on this subject.

RESUMO: O hábito de pastejo praticado pelos bovinos pode ser positiva ou negativamente influenciado pela estrutura da pastagem onde esse animal está inserido. Vários são os fatores que determinam a estrutura da pastagem, e a presença de plantas daninhas pode ser considerada um dos principais. O presente trabalho teve por objetivo avaliar o efeito de espécies com e sem estruturas contundentes sobre o comportamento de pastejo de bovinos em áreas de pastagens. O experimento foi conduzido em faixa no delineamento de blocos completos casualizados com quatro repetições, onde os tratamentos foram dispostos em faixas 4 x 3: quatro espécies de plantas daninhas (Zanthoxylum rhoifolium, Cnidoscolus urens, Dasyphyllum brasiliensis e Luehea divaricata) associados a três faixas de proximidade das plantas daninhas: 0-50, 50-100 e 100-150 cm de distância em relação ao caule principal da planta daninha estudada. O consumo de forragem pelos animais foi mensurado determinando-se a oferta real de forragem aos 0, 3, 6, 9 e 15 dias após o início de pastejo animal (DAP). A presença de arbustos nocivos influencia o comportamento de pastejo de bovinos. A influência no consumo é mais evidente quando existem plantas que promovem desconforto animal. Z. rhoifolium, C. urens e D. brasiliensis e foram as daninhas de maior influência negativa sobre o acesso ao alimento entre as espécies que possuem unidades contundentes. A influência negativa no pastejo é maior na faixa mais próxima à planta, ou seja, 0-50 cm de distância do caule principal da daninha.

ABSTRACT: The grazing behavior practice by bovines can be positive or negatively influenced by the pasture structure where the animal is inserted. Several factors determine the pasture structure and the presence of weeds is considered one of the most important. This study aimed to assess the effect of species with and without stiff structures over the grazing behavior of bovines in pasture areas. The experimental design was a split block design with four replications, in which treatments were arranged in a 4 x 3 factorial design: four weed species (Zanthoxylum rhoifolium, Cnidoscolus urens, Dasyphyllum brasiliensis, and Luehea divaricata) associated with three proximity strips of weeds: 0-50, 50-100, and 100-150 cm in relation to the main stem of the studied weed. Forage intake by animals was measured by determining the real forage offers at 0, 3, 6, 9, and 15 days after the beginning of animal grazing (DAP). The presence of noxious shrubs influenced bovine grazing behavior. The influence on the intake is most evident in the presence of plants that promote animal discomfort. The species Z. rhoifolium, C. urens, and D. brasiliensis were the weeds with the greatest influence on feed access among the species that have stiff structures. The negative influence on grazing behavior is higher in the proximity strip closest to the plant, i.e. 0-50 cm from the main stem of the weed.

RESUMO O objetivo do presente estudo foi avaliar o efeito do óleo de orégano sobre a morfometria intestinal e hepática do lambari-do-rabo-amarelo, Astyanax altiparanae. Os peixes (1,46 ± 0,09 g) foram distribuídos em aquários contendo 60 L, na densidade de 0.5 peixes L-1. Foram avaliadas seis dietas práticas contendo quantidades variadas de óleo de orégano (0,0; 0,5; 1,0; 1,5; 2,0 e 2,5 g de óleo de orégano kg-1). Ao fi nal de 90 dias, os peixes foram eutanasiados. Foi realizada a coleta de 4 intestinos e 4 fígados por tratamento, que foram fi xados em Bouin e incluídos em resina. Foi observado efeito linear crescente do óleo de orégano sobre a altura das vilosidades, largura das vilosidades, área de superfície de absorção e espessura da camada muscular. Também foi observado efeito linear decrescente sobre o número total de células caliciformes. Houve efeito linear crescente da porcentagem citoplasmática e glicogênio hepático. Para a porcentagem de núcleos dos hepatócitos e capilares houve efeito linear decrescente. Dessa forma, o óleo essencial de orégano promove o aumento da área de absorção e modula a quantidade de células caliciformes envolvidas na proteção da mucosa intestinal e promove o aumento citoplasmático, com maior deposição de glicogênio hepático no lambari-do-rabo-amarelo.

ABSTRACT This study aimed to evaluate the effect of oregano oil on the intestinal and liver morphometry of yellow tail tetra, Astyanax altiparanae. Fish (1.46 ± 0.09 g) were kept in a 60-L aquaria, at a stocking density of 0.5 fi sh L-1. Six diets containing varying amounts of oregano oil were evaluated (0.0; 0.5; 1.0; 1.5; 2.0 and 2.5 g of oregano oil kg-1). At the end of 90 days, the fi sh were euthanised. Four intestines and four livers were collected per treatment, which were fi xed in Bouin and embedded in resin. For height and width folds, the absorption surface area and thickness of the muscular layer a positive linear effect of oregano oil was observed. A decrescent linear effect on the total number of goblet cells was also observed. For the cytoplasmic percentage of hepatocytes and liver glycogen, a positive linear effect of oregano oil was observed. There was a decreasing linear effect on the percentage of nuclei in the hepatocytes and capillaries. Thus, the oregano essential oil promotes increased absorption areas, modulates the amount of goblet cells involved in protecting the intestinal mucosa and promotes cytoplasmic increase with greater deposition of liver glycogen in yellow tail tetra.

Austrodiplostomum compactum e Ithyoclinostomum dimorphum são dois trematódeos comumente encontrados em traíras, contudo, esses parasitas nunca foram relatados em traíras do Rio Doce. O objetivo do estudo é descrever a ocorrência de metacercárias de A. compactum e I. dimorphum, e presença de ovos em metacercárias de I. dimorphum em traíras provenientes do médio curso do Rio Doce. Os parasitas foram analisados e identificados utilizando métodos descritos anteriormente. Foram encontradas 10 metacercárias de A. compactum no humor aquoso dos olhos em quatro traíras e 12 metacercárias de I. dimorphum encistadas na cavidade peritoneal de cinco traíras. A maceração de metacercárias de I. dimorphum revelou a presença de ovos nestes parasitos. Esses resultados demonstram a ampla distribuição desses trematódeos e este é o primeiro relato destes parasitos em traíra do Rio Doce.

Austrodiplostomum compactum and Ithyoclinostomum dimorphum are two trematodes commonly found in trahira, but these parasites were never reported in trahiras from Rio Doce. Thus, the aim of this study is to describe the occurrence of A. compactum and I. dimorphum metacercariae in trahira from the middle course of the Rio Doce and to record the presence of eggs in I. dimorphum metacercariae. The parasites were identified and analyzed using methods described previously. There were found 10 A. compactum metacercariae in the aqueous humor of eyes in four of the trahiras and 12 I. dimorphum metacercariae encysted in the peritoneal cavity in five of the trahiras. Maceration of the I. dimorphum metacercariae revealed the presence of eggs. These results demonstrate the broad distribution of these parasites and the first report of these parasites in trahira from Rio Doce.

We compared two electroretinography (ERG) electrodes in dogs using ERG standards of the International Society for Clinical Electrophysiology of Vision (ISCEV). Ten healthy Yorkshire terrier dogs (mean age, 2.80 ± 1.42 years; 6 females) weighing 5.20 ± 1.56 kg were evaluated using an ERG system for veterinary use. Dark- and light-adapted ERG responses were recorded using an ERG-Jet electrode and a fiber electrode prototype. The examinations were performed during 2 visits, 3 weeks apart. Both electrodes (ERG-Jet or fiber prototype) were used on each animal and the first eye to be recorded (OD × OS) was selected randomly. Three weeks later the examination was repeated on the same animal switching the type of electrode to be used that day and the first eye to be examined. The magnitude and waveform quality obtained with the two electrode types were similar for all ERG responses. ERG amplitudes and implicit times obtained from dogs using the fiber electrode prototype were comparable to those obtained with the ERG-Jet electrode for rod, maximal rod-cone summed, cone, and 30-Hz flicker responses. The fiber electrode prototype is a low-cost device, available as an alternative instrument for clinical veterinary ERG recording for retinal function assessment.

Visando a redução da capacidade de absorção de água de fibras ligno-celulósicas, ciclos de molhagem e secagem são usualmente utilizados na indústria de papel e celulose. Esse procedimento enrijece a estrutura polimérica das fibro-células (processo conhecido como hornificação) resultando assim em maior estabilidade dimensional da fibra. No presente estudo foi avaliada a influência da hornificação de fibras de sisal no seu comportamento físico (variações dimensionais e absorção de água), mecânico (comportamento sob cargas de tração direta) e microestrutural (modificações superficiais da fibra e da estrutura das fibro-células). Ensaios de arrancamento da fibra de sisal em matriz de cimento portland foram realizados, utilizando comprimentos de embebimento de 25 mm e 50 mm, com o objetivo de verificar se a possível estabilidade dimensional decorrente da hornificação aumentava a adesão fibra-matriz. Os resultados indicaram maior estabilidade dimensional, redução na capacidade de absorção de água, aumento na resistência à tração e capacidade de deformação e redução no módulo de elasticidade da fibra de sisal com a hornificação. Acréscimos na carga de arrancamento foram observados indicando uma maior aderência da fibra hornificada à matriz de cimento.

Aiming a reduction in the water absorption capacity of lingo-cellulosic fibers, wetting and drying cycles are usually performed in the industry of paper and cellulose. This procedure stiffens the polymeric structure of the fiber-cells (process known as hornification) resulting in a higher dimensional stability. In the present work it was investigated if the dimensional stability, obtained from the hornification process, can imporove the adhesion of the sisal fiber in a Portland cement matrix. In order to study the fiber-matrix bond adhesion pull-out tests were performed in embedment lengths of 25mm and 50mm. Furthermore, it was investigated the influence of the hornification in the physical behavior (dimensional variation and water absorption), mechanical (behavior under tensile loading) and microstructural (surface modifications of the fiber and changes in the fiber-cell structure). The results indicate a higher dimensional stability, reduction in the water absorption capacity, increase in the tensile strength and strain capacity and reduction in the modulus of elasticity with the hornification. The pull-out load increased which indicates a higher bond strength in the hornified fiber with the cementitious matrix.

This is a descriptive character search with a qualitative approach based on the perception and preparation of the nurse faced with patients for whom there is no therapy possible. The data reveal that there is a concern on the part of interviewees with preparedness to deal with death and dying, an area that still suffers many limitations. Nevertheless, care is the only practice that does not end with the various situations in which patients findthemselves.

Trata-se de uma pesquisa de caráter descritivo de abordagem qualitativa, tendo como base a percepção e preparo do enfermeiro frente ao paciente fora de possibilidade terapêutica. Os dados permitiram observar que há uma preocupação por parte dos entrevistados com o preparo para lidar com a morte e o morrer, fato este que ainda sofre muitas limitações. Apesar destas o cuidado é a única prática que não cessa diante das diversas situações em que o paciente se encontra.

Se trata de una investigación de carácter descriptivo de enfoque cualitativo, basado en la percepción y la preparación del enfermero frente al paciente sin posibilidad terapéutica. Los datos permitieron notar que existe una preocupación de los entrevistados con la preparación para lidiar con la muerte y el morir, hecho que aún posee muchas limitaciones. Pese a ello, el cuidado es la única práctica que no termina frente a las distintas situaciones en las cuales el paciente se encuentra.

We investigated the somatic maturation of neonate rats treated during the suckling period with citalopram, a selective serotonin reuptake inhibitor. Groups with 6 male neonates were randomly assigned to different treatments 24 h after birth. Each litter was suckled by one of the dams until the 21st postnatal day. Body weight, head axis and tail length were measured daily from the 1st to the 21st postnatal day. Time of ear unfolding, auditory conduit opening, incisor eruption, and eye opening was determined. Pups received 5 mg (Cit5), 10 mg (Cit10) or 20 mg/kg (Cit20) citalopram sc, or saline (0.9% NaCl, w/v, sc). Compared to saline, body weight was lower (24.04%, P < 0.01) for Cit10 from the 10th to the 21st day and for Cit20 from the 6th to the 21st day (38.19%, P < 0.01). Tail length was reduced in the Cit20 group (15.48%, P < 0.001) from the 8th to the 21st day. A reduction in mediolateral head axis (10.53%, P < 0.05) was observed from the 11th to the 21st day in Cit10 and from the 6th to the 21st day in Cit20 (13.16%, P < 0.001). A reduction in anteroposterior head axis was also observed in the Cit20 group (5.28%, P < 0.05) from the 13th to the 21stday. Conversely, this axis showed accelerated growth from the 12th to the 21stday in the Cit5 group (13.05%, P < 0.05). Auditory conduit opening was delayed in the Cit5 and Cit20 groups and incisor eruption was delayed in all citalopram groups. These findings show that citalopram injected during suckling to rats induces body alterations and suggest that the activity of the serotoninergic system participates in growth mechanisms.

Cytochrome P450 (CYP) 2A enzymes are involved in the metabolism of numerous drugs and hormones and activate different carcinogens. Human CYP2A6, mouse CYP2A5 and rat CYP2A3 are orthologous enzymes that present high similarity in their amino acid sequence and share substrate specificities. However, different from the human and mouse enzyme, CYP2A3 is not expressed in the rat liver. There are limited data about expression of CYP2A3 in extrahepatic tissues and its regulation by typical CYP inducers. Therefore, the objective of the present study was to analyze CYP2A3 mRNA expression in different rat tissues by RT-PCR, and to study the influence of 3-methylcholanthrene, pyrazole and ß-ionone treatment on its expression. Male Wistar rats were divided into four groups of 5 rats each, and were treated ip for 4 days with 3-methylcholanthrene (25 mg/kg body weight), pyrazole (150 mg/kg body weight), ß-ionone (1 g/kg body weight), or vehicle. Total RNA was extracted from tissues and CYP2A3 mRNA levels were analyzed by semiquantitative RT-PCR. CYP2A3 mRNA was constitutively expressed in the esophagus, lung and nasal epithelium, but not along the intestine, liver, or kidney. CYP2A3 mRNA levels were increased in the esophagus by treatment with 3-methylcholanthrene and pyrazole (17- and 7-fold, respectively), in lung by pyrazole and ß-ionone (3- and 4-fold, respectively, although not statistically significant), in the distal part of the intestine and kidney by 3-methylcholanthrene and pyrazole, and in the proximal part of the intestine by pyrazole. CYP2A3 mRNA was not induced in nasal epithelium, liver or in the middle part of the intestine. These data show that, in the rat, CYP2A3 is constitutively expressed in several extrahepatic tissues and its regulation occurs through a complex mechanism that is essentially tissue specific.

In this communication we review the results obtained with the confocal laser scanning microscope to characterize the interaction of epimastigote and trypomastigote forms of Trypanosoma cruzi and tachyzoites of Toxoplasma gondii with host cells. Early events of the interaction process were studied by the simultaneous localization of sites of protein phosphorylation, revealed by immunocytochemistry, and sites of actin assembly, revealed by the use of labeled phaloidin. The results obtained show that proteins localized in the interaction sites are phosphorylated. The process of formation of the parasitophorous vacuole was monitored by labeling the host cell surface with fluorescent probes for lipids (PKH26), proteins (DTAF) and sialic acid (FITC-thiosemicarbazide) before interaction with the parasites. Evidence was obtained indicating transfer of components of the host cell surface to the parasite surface in the beginning of the interaction process. We also analyzed the distribution of cytoskeletal structures (microtubules and microfilaments visualized with specific antibodies), mitochondria (visualized with rhodamine 123), the Golgi complex (visualized with C6-NBD-ceramide) and the endoplasmic reticulum (visualized with anti-reticulin antibodies and DIOC6) during the evolution of intracellular parasitism. The results obtained show that some, but not all, structures change their position during evolution of the intracellular parasitism.
