Abstract This study aimed to investigate the effects of the combined injectable contraceptive (CIC) containing estradiol valerate (EV) and norethisterone enanthate (NET-EN) on aorta function and morphology, as well as on redox status, of female Wistar rats. Female rats (9-10 weeks of age) received intramuscular injections of CIC (0.1 mg EV plus 1 mg NET-EN) or castor oil (control group, CTL) for 8 weeks, once a week. Food intake, body weight and systolic blood pressure were measured during the treatment period. Thoracic aortic segments were prepared for isometric tension recording and morphological analysis. Redox status was evaluated by total oxidant status (TOS) and lipid peroxidation (LP) on plasma and reduced glutathione (GSH) on whole blood. CIC group presented lower food intake and lower total weight gain compared to CTL group. There was no change in systolic blood pressure, vascular response of aorta to phenylephrine and acetylcholine and aorta thickness. Plasma TOS and LP values were reduced in CIC group, although GSH was not altered. It was shown that the long-term treatment with the CIC containing EV plus NET-EN does not induce endothelial dysfunction and histomorphometric changes of vascular wall, as well as improves redox status on female Wistar rats.

O objetivo deste estudo é analisar, a partir da manifestação de piora ou melhora de capacidade para o trabalho, os aspectos condicionantes de mudanças na relação entre trabalho e voz, segundo o discurso de docentes da rede municipal de ensino de São Paulo. Participaram as professoras que, em comparação com pesquisa anterior, apresentaram maior diferença tanto para piora (Grupo A) como para melhora (Grupo B) nos resultados do Índice de Capacidade para o Trabalho. Elas foram convidadas a discutir quais aspectos poderiam explicar a melhora ou piora desses resultados, em um contexto de grupo focal. Os relatos foram transcritos e analisados qualitativamente, segundo a recorrência dos enunciados. Constatou-se que o grupo A apresentou condição mais adoecida e maior necessidade de falar sobre as dificuldades no trabalho. O Grupo B apresentou mais força para enfrentar os problemas referentes ao trabalho, inclusive propostas criativas. A favor do Grupo B, também foram registradas melhores relações no trabalho quanto ao apoio social e autonomia.

This study aims to analyze, by means of manifestation of worsening or improvement in work ability, the aspects conditioning changes in the relation between work and voice, according to the discourse of teachers working at the municipal education network of São Paulo, Brazil. The participants were female teachers who, when comparing with a previous research, had greater difference both in terms of worsening (Group A) and improvement (Group B) in the results of the Work Ability Index. They were invited to discuss which aspects could explain the improvement or worsening in these results, in a focus group context. The reports were transcribed and qualitatively analyzed, according to the recurrence of utterances. We found out that group A showed a worse health status and it had a greater need to talk about difficulties at work. Group B showed more strength to face problems related to work, including creative proposals. In favor of Group B, better work relationships were also registered concerning social support and autonomy.

INTRODUCTION: Sepsis is a systemic inflammatory response related to high mortality rates in the hospital environment. Delayed etiological diagnosis and inadequate antimicrobial therapy are associated with treatment failures. Molecular tests based on polymerase chain reaction are regarded as faster and more accurate procedures than culture techniques for microbial identification, providing a higher rate of therapeutic success. OBJECTIVE: To develop a panel of primers for DNA fragments of sepsis-related microorganisms. METHODS: Primers for amplification of Enterobacter spp., Escherichia coli, Pseudomonas aeruginosa, Staphylococcus aureus and Candida spp. were designed and tested for sensitivity and specificity on the basis of their respective standard strains. RESULTS: The intended specificity was obtained for P. aeruginosa, S. aureus and Candida spp primers. Sensitivity tests showed a threshold for detection from 5 ng to 500 fg in blood samples contaminated with microbial DNA. CONCLUSIONS: The molecular panel presented offers the advantage of a flexible 'open' system when compared to other multiplex detection methods.

INTRODUÇÃO: A sepse é uma resposta inflamatória sistêmica relacionada com altas taxas de mortalidade no meio hospitalar. O diagnóstico etiológico tardio e terapia antimicrobiana inadequada se associam a falhas do tratamento. Exames moleculares baseados na reação em cadeia da polimerase são considerados métodos mais rápidos e precisos do que técnicas de hemocultura para identificação microbiana, proporcionando uma taxa mais elevada de sucesso terapêutico. OBJETIVO: Desenvolver um painel de seqüências iniciadoras (primers) para fragmentos de DNA de microrganismos associados à sepse. MÉTODOS: Seqüências iniciadoras para amplificação de Enterobacter spp., Escherichia coli, Pseudomonas aeruginosa, Staphylococcus aureus e Candida spp. foram desenvolvidos e testados quanto a sensibilidade e especificidade com base em suas respectivas cepas padrão. RESULTADOS: A especificidade pretendida foi obtida para os primers de P. aeruginosa, S. aureus e Candida spp. O teste de sensibilidade mostrou um limite de detecção de 5 ng a 500 fg em amostras de sangue contaminado com DNA microbiano. CONCLUSÕES: O painel molecular apresentado oferece a vantagem de constituir um sistema flexível "aberto" em comparação a outros métodos de detecção múltipla.

Cross-infection with Pseudomonas aeruginosa among cystic fibrosis (CF) patients is a rare occurrence. However, the emergence of transmissible strains has been reported between unrelated individuals. We analyzed the genetic relationship among P. aeruginosa isolates from Brazilian CF patients and transmissible clones which are worldwide spread. The data does not indicate the presence of closely related variant clones.
