Abstract Introduction/objectives Systemic lupus erythematosus (SLE) is a classic prototype of the multisystem autoimmune disease and follows a relapsing and remitting course. Triptolide is a diterpene triepoxide extracted from Chinese medicine Tripterygium wilfordii Hook F, with potent immunosuppressive and anti-inflammatory properties. Our previous work observed that triptolide alleviated lupus in MRL/lpr lupus mice with the upregulation of regulatory T cells (Treg) proportion in previous study. In this study, we explored the proportion of follicular T regulatory (Tfr), follicular T helper (Tfh) and germinal center (GC) B cells in lupus mice and evaluated the efficacy of triptolide for lupus treatment in vivo. Methods 20 female MRL/lpr mice were randomly divided into 2 treatment groups and treated orally with vehicle or triptolide. C3H mice were all housed as controlled group and treated orally with vehicle. The percentage of Tfr cells, Tfh cells and GC B cells in spleen of mice were detected by Flow cytometric analysis and immunohistochemistry after 13 weeks of treatment. Results We found that the percentage of Tfr cells decreased in MRL/lpr mice compared with controlled mice. The percentage of Tfh cells in MRL/lpr mice was significantly higher compared with that in controlled mice. The ratio of Tfr/Tfh is also decreased in lupus mice. After treated with triptolide in MRL/Lpr mice in vivo, the percentage of Tfr cells and ratio of Tfr/Tfh increased. The proportion of GC B cells also decreased in mice treated with triptolide by FACS and immunohistochemistry. Conclusions Our results demonstrate that the effect of triptolide in alleviating lupus is partly by reversing immune imbalance with increased percentage of Tfr cells and ratio of Tfr/Tfh. Triptolide might also has effect on immune response through inhibiting proliferating GC B cells.

Neuroblastoma is one of the most common tumors in children. Cases where an isolated soft-tissue metastasis mass is the initial symptom are rare, with only four such cases reported to date. We describe the imaging findings of ten cases of neuroblastoma patients in our hospital with superficial soft tissue mass (SSTM) as the primary symptom. The main ultrasound finding of SSTM was hypoechoic masses or scattered speck-like hyperechoic masses. However, when this type of SSTM is caused by soft tissue metastasis, the location is often atypical, and ultrasound findings are difficult to distinguish from other benign diseases. Therefore, this research should remind clinicians to recognize atypical presentations of this common childhood malignant tumor. ABSTRACT Purpose: To explore the role and mechanisms of octreotide in neurofunctional recovery in the traumatic brain injury (TBI) model. Methods: Rats were subjected to midline incision followed by TBI in the prefrontal cortex region. After 72 hours, the behavioural and neurological deficits tests were performed, which included memory testing on Morris water maze for 5 days. Octreotide (15 and 30 mg/kg i.p.) was administered 30 minutes before subjecting to TBI, and its administration was continued for three days. Results: In TBI-subjected rats, administration of octreotide restored on day 4 escape latency time (ELT) and increased the time spent in the target quadrant (TSTQ) on day 5, suggesting the improvement in learning and memory. It also increased the expression of H2S, Nrf2, and cystathionine-γ-lyase (CSE) in the prefrontal cortex, without any significant effect on cystathionine-β-synthase. Octreotide also decreased the TNF-α levels and neurological severity score. However, co-administration of CSE inhibitor (D,L-propargylglycine) abolished octreotide-mediated neurofunctional recovery, decreased the levels of H2S and Nrf2 and increased the levels of TNF-α. Conclusions: Octreotide improved the neurological functions in TBI-subjected rats, which may be due to up-regulation of H2S biosynthetic enzyme (CSE), levels of H2S and Nrf2 and down-regulation of neuroinflammation.

ABSTRACTThis experiment was conducted to evaluate the combination effect of low dietary non-phytate phosphorus (NPP) concentrations, phytase (PHY) levels, and 25-hydroxycholecalciferol (25-OH-D3) levels on the growth performance and meat quality of broilers. Two levels of NPP, two levels of PHY, and two levels of 25-OH-D3 resulted in a 2�2�2 factorial arrangements, with eight treatments (TRT). The birds on TRT 1-4 were fed diet 1 (NRC NPP was reduced by 0.1) and the birds on TRT 5-8 were fed with diet 2 (NRC NPP was reduced by 0.2). Each diet was mixed with different levels PHY and 25-OH-D3. Performance and meat quality parameters were measured. Results showed that during entire experiment the most advantageous effects were obtained with TRT 3 (NRC NPP reduced by 0.1 + 600 U/kg phytase + 34.5�g/kg 25-OH-D3) and TRT 4 (NRC NPP reduced by 0.1 + 600 U/kg phytase + 69�g/kg 25-OH-D3). The lowest body weight gain (BWG) and feed intake(FI) were observed with TRT 5 (NRC NPP reduced by 0.2 + 300 U/kg phytase + 34.5�g/kg 25-OH-D3). Lowering NRC NPP by 0.1 to 0.2 significantly reduced weight gain (WG) (p< 0.05) and FI (p< 0.05) during the starter phase (ST), while during grower phase (GF) lowering NRC NPP by 0.1 to 0.2 did not affect WG (p>0.05) and produced small decrease in FI. BWG, FI and feed conversion ratio were not influenced (p>0.05) by different PHY or 25-OH-D3 levels. In addition, the meat color, pH, and shear force were not affected by the different NPP, PHY or 25-OH-D3levels.

The influence of nitrogen (N) limitation on the cell growth, chlorophyll content, intracellular lipid production and expression levels of two pathway genes in Chlorella sorokiniana was investigated in mixotrophic culture in this study. The maximum biomass concentration of C. sorokinianacultured in modified BG11 medium containing 3, 6, 9, 12 and 15 mM N were 1.60, 2.21, 2.74, 3.18 and 3.21 g/L of dry cell weight, respectively with maximum specific growth rate of 0.180, 0.198, 0.201, 0.203, 0.206 day−1 during culture with an initial N feed of 3, 6, 9, 12 and 15 mM in the first eight days, respectively. The maximum lipid content was 51 % with 3 mM N. However, the maximum lipid productivity of 0.00884 g L−1 day−1 was achieved with 6 mM N. Expression levels of accD (heteromeric acetyl-CoA carboxylase beta subunit) and accI (homomeric acetyl-CoA carboxylase) genes in C. sorokiniana were studied by real-time PCR. Increased expression levels of accD reflected the increased lipid content in stationary phase. In contrast, expression of the acc1 gene always remained low, showing that the gene might not be critical for lipid accumulation.

Three fully optimized structures of 3,6-diamino-1,2,4,5-tetrazine-1,4-dioxide (LAX-112) dimers have been obtained with the density functional theory (DFT) method at the B3LYP/6-311++G level. Vibrational frequency calculations were carried out to ascertain that each structure is a minimum (no imaginary frequencies). The intermolecular interaction energy is calculated with the basis set superposition error (BSSE) correction and zero point energy (ZPE) correction. The greatest corrected binding energy among the three dimers is -42.38 kJ mol-1. The charge redistribution mainly occurs on the adjacent O(N)......H atoms between submolecules and the charge transfer between two subsystems is very small. Natural bond orbital (NBO) analysis was performed to reveal the origin of the interaction. Based on the vibrational analysis, the standard thermodynamic functions (heat capacities (cºp), entropies (S°m) and enthalpies (H°m)) and the changes of thermodynamic properties from the monomer to dimer with the temperature ranging from 200.00 K to 800.00 K have been obtained using statistical thermodynamics. The results show that the strong hydrogen bonds dominantly contribute to the dimers, while the bonding energies are not only determined by the hydrogen bonding. The dimerization process of dimer II can occur spontaneously at room temperature.

N-benzoyl-3,3-dinitroazetidine(BDNAZ) is a derivative of 3,3-dinitroazetidine (DNAZ). Its thermal behaviour was studied by DSC methods. The results show that there are one melting process and two exothermic decomposition processes. The kinetic parameters of the intense exothermic decomposition process were obtained from the analysis of the DSC curves. The apparent activation energy, pre-exponential factor and the mechanism function are 170.77 kJ mol-1,10(14.12) s-1 and f(α) = (1-a)-1/2, respectively. The specific heat capacity of BDNAZ was determined with a continuous Cp mode of a micro-calorimeter. The standard mole specific heat capacity of BDNAZ was 286.31 J mol¹ K-1 at 298.15 K. Using the relationship between Cp and T with the thermal decomposition parameters, the time of the thermal decomposition from initialization to thermal explosion (adiabatic time-to-explosion, tTIAD), the self-accelerating decomposition temperature (TSADT), thermal ignition temperature (T TIT), critical temperatures of thermal explosion (Tb) and period of validity (t0.9) were obtained to evaluate its thermal safety. The detonation velocity (D) and pressure (P) of BDNAZ were estimated by using the nitrogen equivalent equation according to the experimental density.

Ramie (Boehmeria nivea and Boehmeria tenacissima) is a widely used fiber crop. Traditional water retting or chemical boiling method performed in order to extract ramie fiber seriously pollute the environment and severely damage the fiber, so biological method is the general trend of the fiber-extracting industry. Some strains (687), involving 26 genera and 43 species, were collected from the three samples, which produce hydrolyzed circles in the selective culture medium in order to detect the degumming effect and to compare the enzyme activity. Among these strains, 13 of them did not produce cellulase and had a ramie decreasing weight rate above 25 %, which were regarded as efficient ramie-degumming strains named from R1 to R13. R1 to R13 belonged to Amycolata autotrobutylicun, Bacillus subtilis, Clostridium acetobutylicum, Bacillus subtilis, Rhizobium leguminosarum, Bacteroides finegoldii, Streptomyces lividans, Bacillus amyloliquefaciens, Clostridium acetobutylicum, Pseudomonas brassicacearum, Bacillus pumilus, Bacillus licheniformis, Pectobacterium wasabiae respectively. Bacteroides sp., Rhizobium sp. and Pseudomonas sp. were firstly reported to be used in ramie-degumming. At the same time, the pectinase was the key enzyme in the ramie-degumming process.

Thymosin alpha 1 (Tα1) has been shown to have beneficial effects on numerous immune system parameters, but little is known about the effects of Tα1 on patients with gastric carcinoma. The objective of this study was to determine the effect of Tα1 on subpopulations of Th1, Th2, Th17, and regulatory T cells (Tregs) in vitro, and to evaluate its efficacy as an immunoregulatory factor in patients with gastric carcinoma. We compared the effect of Tα1 on the frequency of CD4+ and CD8+ T cells, especially the CD4+CD25+Foxp3+ Tregs in peripheral blood mononuclear cells (PBMCs) from gastric carcinoma patients (N = 35) and healthy donors (N = 22). We also analyzed the changes in the proliferation of PBMCs in response to treatment with Tα1, and examined the production of Th1, Th2, and Th17 cytokines by PBMCs and tumor-infiltrating lymphocytes. The treatment of PBMCs from gastric cancer patients, with Tα1 (50 µg/mL) alone increased the percentage of CD4+CD25+Foxp3+ (suppressive antitumor-specific Tregs) from 1.68 ± 0.697 to 2.19 ± 0.795% (P < 0.05). Our results indicate that Tα1 increases the percentage of Tregs and IL-1β, TNF-α, and IL-6 in vitro.

No presente estudo foram investigadas as respostas fisiológicas da alga vermelha Porphyra haitanesis às elevadas concentrações de cobre (acima de 50 μM) e de zinco (acima de 100 μM). Os resultados mostram que os efeitos de Cu2+ e Zn2+ sobre o crescimento, pigmentos fotossintéticos (clorofilas e carotenóides), ficobiliproteína e metabolismo (o espectro de emissão de fluorescência e as atividades do fotossistema) não seguem o mesmo padrão. A taxa de crescimento relativo foi inibida por diferentes concentrações de Cu2+ e, em presença de Zn2+, aumentou ligeiramente em baixas concentrações (abaixo de 10 μM) e foi inibida em altas concentrações. Por outro lado, os teores de ficoeritrina apresentaram leve aumento em concentrações relativamente baixas de Cu2+ e Zn2+ (até 1 μM Cu2+ e até 20 μM Zn2+, respectivamente) e foram inibidas por altas concentrações. Além disso, tanto a fotossíntese quanto a respiração mostraram aumento nas trocas de oxigênio em resposta às concentrações relativamente baixas de Cu2+ (até 1 μM) e de Zn2+ (até 10 μM), além da redução em concentrações relativamente altas desses metais. Adicionalmente, as atividades fotoredutoras e as emissões de fluorescência do fotossistema II (PSII) foram incrementadas em baixas concentrações de Cu2+ (até 0,1 μM) e de Zn2+ (até 10 μM) e inibidas por altas concentrações. Desta forma, a intensidade da fluorescência da clorofila-a e dos centros de reação ativa PSII seguiram um padrão semelhante em resposta às elevadas concentrações de Cu2+ e Zn2+. Esses resultados sugerem que baixas concentrações de Cu2+ e Zn2+ afetam o metabolismo de P. haitanesis, que se torna inibido por altas concentrações desses metais.

In the present study, several physiological responses of the red marine alga Porphyra haitanesis to elevated concentrations of copper (up to 50 μM) and zinc (up to 100 μM) were investigated. Our results showed that the effects of Cu2+ and Zn2+ on growth, photosynthetic pigments (chlorophylls and carotenoids), phycobiliprotein and metabolism (the fluorescence emission spectra and the activities of photosystemII) did not follow the same pattern. The relative growth rate was inhibited by different concentrations of Cu2+, and was slightly increased at lower concentrations (up to 10 μM) and inhibited at higher Zn2+concentrations. On the other hand, the phycoerythrin contents were slightly increased at relatively low concentrations (up to 1 μM Cu2+ or 20 μM Zn2+) and inhibited by high Cu2+ and Zn2+ concentrations. Moreover, photosynthesis and respiration showed an increase in the amount of oxygen exchange in response to relatively low Cu2+ (up to 1 μM) and Zn2+ concentrations (up to 10 μM), and a reduction to relatively high Cu2+ and Zn2+ concentrations. Oxygen evolution was more sensitive than oxygen uptake to Cu2+ and Zn2+. In addition, the photoreductive activities and fluorescence emission of photosystem II (PS II) were enhanced by lower concentrations of Cu2+ (up to 0.1 μM) and Zn2+ (up to 10 μM) and inhibited by higher concentrations. Furthermore, the intensity of chlorophyll a fluorescence and the active PSII reaction centers followed a similar pattern in response to elevated concentrations of Cu2+ and Zn2+. These results suggest that lower concentrations of Cu2+ and Zn2+ affected the metabolism of P. haitanesis, which was inhibited by higher concentrations of these metals.

The retrotransposon known as long interspersed nuclear element-1 (L1) is 6 kb long, although most L1s in mammalian and other eukaryotic cells are truncated. L1 contains two open reading frames, ORF1 and ORF2, that code for an RNA-binding protein and a protein with endonuclease and reverse transcriptase activities, respectively. In this work, we examined the effects of full length L1-ORF2 and ORF2 fragments on green fluorescent protein gene (GFP) expression when inserted into the pEGFP-C1 vector downstream of GFP. All of the ORF2 fragments in sense orientation inhibited GFP expression more than when in antisense orientation, which suggests that small ORF2 fragments contribute to the distinct inhibitory effects of this ORF on gene expression. These results provide the first evidence that different 280-bp fragments have distinct effects on the termination of gene transcription, and that when inserted in the antisense direction, fragment 280-9 (the 3' end fragment of ORF2) induces premature termination of transcription that is consistent with the effect of ORF2.
