RESUMO: O fenômeno quorum sensing corresponde a um processo de comunicação intra e interespécies microbianas e é mediado por sinais químicos extracelulares, denominados moléculas sinalizadoras ou auto indutoras (AI). A molécula AI2 está envolvida na comunicação interespécies, denominada sistema “universal” de comunicação. Este estudo teve como objetivo avaliar a capacidade de Enterococcus faecium, Enterococcus faecalis e Bacillus cereus isolados do processamento de ricota em produzir moléculas sinalizadoras de Quorum sensing (AI-2). Os isolados foram avaliados quanto à presença do gene luxS utilizando a reação em cadeia da polimerase (PCR). As moléculas sinalizadoras (AI-2) foram medidas em unidades relativas de luz (RLU) através de um luminômetro. Um total de 74% dos isolados de E. faecium, 91% de E. faecalis e 95% de B. cereus foram positivos para o gene luxS. Além disso, todos os isolados apresentaram capacidade de induzir o fenômeno de bioluminescência em Vibrio harveyi BB170, indicando a presença de auto indutores AI-2.

ABSTRACT: The quorum sensing phenomenon is a process of intra- and inter-species microbial communication involving the production and detection of extracellular signaling molecules. The autoinducer AI-2 has been proposed to serve as a ‘universal signal’ for interspecies communication. This study aimed to evaluate the capability of Enterococcus faecium, Enterococcus faecalis, and Bacillus cereus strains isolated from ricotta processing to produce quorum sensing signalling molecules (AI-2). The strains were evaluated for the presence of the luxS gene using the polymerase chain reaction. AI-2 quorum sensing signalling molecules were measured in relative light units (RLUs) using a luminometer. A total of 74% of E. faecium, 91% of E. faecalis, and 95% of B. cereus isolates were positive for luxS gene. In addition, the induced bioluminescence in Vibrio harveyi BB170 was observed in all strains, indicating the presence of the AI-2 autoinducer.

RESUMO: A população brasileira tem apresentado um elevado consumo de peixes, no entanto pescados são alimentos altamente perecíveis e podem ser contaminados por uma diversidade de microrganismos. A situação ainda é mais alarmante quando se trata da formação de biofilmes. Para verificação da formação de biofilme, as cepas foram testadas dentro de cultivos mono e multi-espécies a 25°C durante o tempo total de 48h, onde as análises foram realizadas no tempo 0 e nos intervalos de 4, 8, 24 e 48h em cupom de aço inoxidável medindo 1,0x1,0cm, imersos em caldo TSB (Tryptic Soy Broth).O comportamento dos biofilmes formados, diante da sanitização por imersão dos cupons em solução de hipoclorito de sódio a 200ppm por 10 minutos, também foi avaliado, nas mesmas condições citadas acima, com algumas particularidades. A avaliação estrutural foi procedida por microscopia de varredura eletrônica (MEV). Nos resultados de biofilmes mono-espécie, foi verificado que todas as cepas foram capazes de formar biofilme, nos diferentes tempos, sem diferenças estatisticamente significativas. Entretanto, comparando-se o crescimento de culturas mono e múltiplas espécies observou-se formação de biofilme, estatisticamente maior, entre as culturas puras. Nas culturas multi-espécies, Salmonella spp. apresentou crescimento, estatisticamente menor, em todos os tempos, entre os microrganismos testados, enquanto S. aureus apresentou maior crescimento que E. coli até 8 horas de incubação, deste tempo em diante a diferença não foi significativa. Quanto ao comportamento frente sanitização com hipoclorito, o sanitizante foi eficaz, uma vez que não foi observado crescimento celular nas análises dos cupons tratados com a solução de hipoclorito a 200ppm por 10 minutos. Este estudo demonstrou a capacidade dos microrganismos isolados em formar biofilmes, reforçando, a importância dos estabelecimentos manipuladores de alimentos investirem em treinamento de Boas Práticas de Fabricação, desenvolvimento de protocolos adequados de limpeza e desinfecção de áreas e equipamentos envolvidos na produção de alimentos e manutenção e troca desses sempre que se fizer necessário.

ABSTRACT: The consumption of fish by the Brazilian population is increasing. However, fish and seafood are highly perishable and can be contaminated with several microorganisms. In addition, the possibility of biofilm formation is a greater cause for concern. In this study, biofilm formation was evaluated in single- and multispecies cultures at 25°C for incubation periods of 0, 4, 8, 24, and 48h in stainless steel coupons (size, 1.0×1.0cm) immersed in tryptic soy broth. The characteristics of the formed biofilms after sanitizing by immersing the coupons in 200ppm sodium hypochlorite solution for 10min were also evaluated under the same experimental conditions but with some modifications. Biofilm structure was evaluated using scanning electron microscopy. Analysis of single-species biofilms indicated that all bacterial strains formed biofilms at different intervals without any statistically significant difference. However, comparison of the growth of single- and multispecies cultures indicated a significantly higher biofilm formation by the pure cultures. In multispecies biofilms, compared with the other microorganisms, growth of Salmonella spp. was significantly lower for all tested incubation periods; whereas, of Staphylococcus aureus was significantly higher than that of E. coli until 8h of incubation; the differences in growth were not significantly different after this incubation period. Sanitizing with sodium hypochlorite was effective because no cell growth was observed in the coupons that were treated with 200ppm sodium hypochlorite for 10min. This study demonstrated the ability of isolated microorganisms to form biofilms, reinforcing the need for food handling establishments to adopt good manufacturing practices, developing adequate protocols for cleaning and disinfecting surfaces and equipment used in food production, maintaining and replacing equipment when necessary.

RESUMO A prática de balneotarapia em paciente queimado foi abandonada em muitas partes do mundo, mas no Brasil ainda é comum. O objetivo deste estudo foi verificar se a balneoterapia é um fator de risco para a colonização por Pseudomonas aeruginosa em pacientes queimados. Dezoito pacientes internados em um Centro de Queimadura (CQ) foram acompanhados por 14 semanas. Amostras foram coletadas do exsudato de feridas, antes e depois do banho dos pacientes e também da mesa onde a balneoterapia foi realizada. A relação genética entre as cepas de P. aeruginosa foi determinada pela electroforese em gel de campo pulsado. Trinta e sete cepas foram detectadas a partir de 292 swabs coletados de área de superfície das feridas dos pacientes e da mesa de balneoterapia. Análise de fragmentação do DNA das 37 P. aeruginosa mostrou a existência de 10 clones. O tipo A foi o clone mais prevalente, com 23 cepas distribuídas em oito subtipos. Estas estavam presentes nas lesões dos pacientes antes e após o banho e na mesa onde o banho foi realizado, sugerindo contaminação cruzada inter e intra-pacientes e pacientes e mesa de banho. Este trabalho mostra que a balneoterapia é um fator de risco para colonização por P. aeruginosa, no CQ estudado, pois um mesmo clone da bactéria foi encontrado nos isolados coletados em vários pontos e épocas diferentes.

ABSTRACT The practice of immersion in burn patient has been abandoned in many parts of the world but in Brazil it is still common. The aim of this study was to ascertain if balneotherapy is a risk factor for Pseudomonas aeruginosa colonization in thermally injured patients. Eighteen patients from a Burn Center were studied for 14 weeks for Pseudomonas aeruginosa. Samples were collected by swabbing the exudate of wounds, before and after giving bath to the patients and from balneotherapy table. Pulsed-field gel electrophoresis was used to determine bacterial genetic relatedness. Thirty-seven P. aeruginosa isolates were detected from 292 swabs collected from patients' burn surface area and from the balneotherapy table. Profile analysis of P. aeruginosa DNA fragmentation showed 10 clones among the 37 strains analyzed. Type A is the most prevalent clone, with 23 strains distributed into eight subtypes. These were present in the swabs collected, before and after the patients' bath, from the surface of the bath table, suggesting that there was cross-contamination between the patients in different ways. This work demonstrates that balneotherapy is a risk factor in the Burn Center studied, because the same clone was found among P. aeruginosa isolates collected at various points and times.

RESUMO: A capacidade de produzir fatores antimicrobianos é considerada uma importante característica dos microrganismos probióticos. Bacteriocinas, peróxido de hidrogênio, ácido acético e ácido lático, são exemplos destas substâncias. Com o presente trabalho, objetivou-se desenvolver sobremesas lácteas (SL) probióticas, acrescidas de Lactobacillus acidophilus e avaliar a viabilidade desta cepa, além da ação frente a patógenos alimentares. Foram produzidos tratamentos com e sem interações entre o L. acidophilus e bactérias patogênicas Gram negativas (Salmonella sp e Escherichia coli O157:H7) e Gram positivas (Bacillus cereus e Staphylococcus aureus). Os produtos foram armazenados em temperatura de 8ºC e analisados nos tempos 24, 48, 72 horas, 7 dias e 28 dias (apenas T1 por deterioração dos demais produtos neste tempo). A sobremesa láctea com L. acidophilus foi considerada produto probiótico, verificando o potencial de desenvolvimento de um novo produto. Analisando as contagens dos patógenos nas sobremesas lácteas com e sem adição de L. acidophilus, observaram-se diferentes comportamentos diante dos patógenos, o que poderia ser justificado por uma possível ação de bacteriocinas ou competição microbiana, porém não houve uma redução total ou até um nível considerado seguro. Conclui-se que os produtos probióticos desenvolvidos reduziram os patógenos alimentares de importância, porém não a níveis considerados seguros. Dessa forma, ressalta-se a importância de ferramentas de qualidade no desenvolvimento e monitoramento de sobremesas lácteas.

ABSTRACT: The ability to produce antimicrobial factors is considered an important feature of probiotic microorganisms. Bacteriocins, hydrogen peroxide, acetic acid and lactic acid are examples of these substances. The present research aimed to develop probiotic dairy desserts (DD) with Lactobacillus acidophilus and evaluate the viability of this strain, as well as its action on food pathogens. Treatments with and without interactions between L. acidophilus and pathogenic Gram-negative bacteria (Salmonella sp. and Escherichia coli O157:H7) and Gram positive (Bacillus cereus and Staphylococcus aureus) were produced. The products were stored at a temperature of 8°C and analyzed at the times 24, 48, 72 hours, 7 days and 28 days (at 28 days, only T1 was analyzed because the other products were deteriorated). In an analysis of the potential for development of new products, the dairy dessert with L. acidophilus was considered a probiotic product. Assessment of the counts of pathogens in dairy desserts with or without L. acidophilus showed different behaviors of these products in response to pathogens, which could be justified by a possible action of bacteriocins or microbial competition, but there has been no overall reduction or reduction up to a safe level. It is concluded that the probiotic products developed reduced significant food pathogens, but not up to safe levels. Thus, we emphasize the importance of the use of quality tools in the development and monitoring of dairy desserts.

Shiga toxin-producing Escherichia coli (STEC) O157:H7 strains (isolated by cattle’s faeces and a reference strain, EDL933), were inoculated into pasteurized milk (102 and 103 cells.mL–1) to prepare the Minas frescal cheese. As control was used uninfected milk. Physicochemical and microbiological analyses were performed to milk and elaborated cheese. The O157:H7 strains were quantified in the stages of cheese processing and during 0, 2, 4, 5, 7, 10 and 15 storage days at 8 °C onto Sorbitol MacConkey Agar supplemented with potassium tellurite and cefixime (CT-SMAC). O157:H7 was not present in the pasteurised milk prior to the artificial inoculation. At the end of the processing the cheese had 10 to 100 times more STEC O157:H7 than the initial inoculum. During the storage, the Minas frescal cheese exhibited the largest population increase on the 4th and 5th day when inoculated with 102 and 103 cells.mL–1, respectively. Additionally, viable cells were found up to the 10th and 15th day, according to the amount of initial inoculum. This number of cells is able to cause infection in humans, and therefore, Minas frescal cheese, even when stored under refrigeration, is a potential vehicle of disease caused by STEC O157:H7.

A existência de um reservatório animal é de grande importância na transmissão de Escherichia coli, produtora de toxina shiga (STEC) aos humanos. Epidemiologicamente, o sorotipo O157:H7 tem sido o mais envolvido em surtos de doença humana causada por STEC, porém surtos envolvendo STEC não pertencentes ao sorogrupo O157 (STEC não-O157) têm sido descritos. Inúmeros trabalhos constatam uma elevada ocorrência destes microrganismos em fezes de bovinos no Brasil, entretanto, pouco se sabe sobre a transmissão destes aos produtos de origem animal em nosso país. Neste trabalho, foi avaliada a viabilidade de E.coli O153:H25; O113:H21 e O111:H8 em Queijo Minas Frescal (QMF), produzido com inóculos de STEC não O157: H7 e armazenados a 8ºC. Realizaram-se contagens de E. coli e psicrotróficos totais após o processamento do queijo e com intervalos de sete e quinze dias. Foi observado aumento nas contagens de E. coli STEC não O157: H7 e psicrotróficos totais logo após o processamento do QMF, bem como durante o armazenamento a 8ºC, temperatura máxima recomendada pela legislação brasileira. Demonstra-se que, caso haja contaminação da matéria-prima com STEC não O157: H7 (deste estudo), o processamento do QMF não elimina os microrganismos e a temperatura máxima recomendada pela legislação não inibe a multiplicação bacteriana, mantendo-se o risco à população. Reforça-se, portanto, a atenção à qualidade da matéria-prima, das ferramentas de qualidade no campo e na indústria de alimentos para garantir a inocuidade do produto final

The existence of an animal reservoir is of great importance in the transmission of shiga toxin-producing E. coli (STEC) to humans. Epidemiologically serotype O157:H7 has been the most involved in human disease outbreaks caused by STEC, however STEC not belonging to serogroup O157 (STEC non-O157) have been described in outbreaks. Studies have revealed a risk in occurrence of these organisms in feces of cattle in Brazil, but little is known about the transmission to animal's products origin in our country. This study evaluated the viability of E. coli O153:H25, O113:H21 and O111:H8 in Minas Frescal Cheese (MFC), produced with non- STEC O157:H7 and stored at 8ºC. Counts of E. coli and total psychrotrophic were performed after processing and at intervals seven and fifteen days. There were an increase in the counts of E. coli and total psychrotrophic just after MFC processing as well as during storage at 8°C. The results demonstrated that, if the raw material (milk) is contaminated with STEC non O157:H7 (this study), the MFC processing does not eliminate the microorganisms and the maximum temperature recommended does not eliminate bacterial growth, keeping the risk to the population. The results reinforces, the attention to the quality of the raw material, the quality tools in the field and in the food industry to ensure the safety of final products

Staphylococcus epidermidis resistente à meticilina (MRSE) e Staphylococcus haemolyticus resistente à meticilina (MRSHa) são importantes estafilococos coagulase negativa. São frequentemente isolados em bacteremia humana, principalmente devido à capacidade de formar biofilmes nas superfícies de dispositivos médicos introduzidos no organismo. A papaína é mistura complexa de enzimas proteolíticas e peroxidases extraídas do látex de Carica papaya, reconhecida por acelerar os processos de cura de feridas. Este estudo teve como objetivo avaliar a capacidade dos MRSE e MRSHa em produzir biofilmes e analisar a capacidade da papaína em inibir a formação de biofilme ou desintegrar biofilmes já formados por essas bactérias. Observou-se que a papaína tem capacidade de reduzir a formação de biofilme por MRSE (p < 0,06) e MRSHa (p = 0,0005). Além disso, a papaína foi capaz de desintegrar biofilme maduro formado por MRSE (p = 0,014). Nenhuma atividade antibacteriana da papaína foi observada para qualquer das duas espécies de bactérias testadas. A papaína mostrou-se produto potencial para reduzir biofilme.

Methicillin-resistant Staphylococcus epidermidis (MRSE) and methicillin-resistant Staphylococcus haemolyticus (MRSHa) are important coagulase-negative staphylococci. They are often isolated from bacteremia in humans mainly due to their ability to form biofilm on the surfaces of medical devices. Papain is a complex mixture of proteolytic enzymes and peroxidases extracted from the latex of Carica papaya and it is recognized by accelerating the healing process of wounds. This study aimed to evaluate the ability of the MRSE and MRSHa isolates to produce biofilms. Besides this, the ability of papain to inhibit the formation of biofilms or to disrupt the ones already formed by those bacteria was analyzed. Thirty MRSHa and 30 MRSE were isolated from bacteremia and used in this study. It was observed that papain has ability to reduce biofilms formed by MRSE (p < 0.06) and by MRSHa (p = 0.0005). In addition, papain was able to disrupt mature biofilms made by MRSE (p = 0.014). No antibacterial activity of papain was observed for any isolates of MRSE and MRSHa tested. Papain has been demonstrated as a potential product for reducing biofilm.
