Abstract The seasonal forest formations of the Atlantic Forest are a threatened and poorly known habitat. We present here a list of the non-volant mammals occurring in a 515-ha forest fragment known as Santa Maria and located in the Brazilian state of São Paulo. Our surveys are based on live trap captures, camera traps, and active searches for footprints, as well as secondary data. We list 29 species of non-volant mammals in the fragment, recorded between 1996 and 2021. One species found in the fragment, Leontopithecus chrysopygus, is globally endangered. Two species are classified as “vulnerable” in the global red list: Myrmecophaga tridactyla, and Tapirus terresris. One species, Panthera onca, is classified as “critically endangered” in São Paulo state. The red howler, Alouatta guariba, was not recorded after 1999 and has probably been extirpated in the fragment. We show that the number of non-volant mammal species in Santa Maria fragment is high, in relation to its size. The fragment is also in a strategic position, between the Morro do Diabo state park and the Black Lion Tamarin Ecological station, the two largest protected areas in the region. Considering its high mammal richness and its possible role as stepping stone for the local fauna, we recommend that the fragment become a protected area.

RESUMO Nove locos de microssatélites polimórficos foram isolados e caracterizados para Cryptocarya mandioccana Meisner, uma árvore da Mata Atlântica cujas sementes são dispersas por grandes animais. Os locos foram caracterizados usando 48 indivíduos de duas populações e o sucesso de genotipagem testado em quatro tecidos: folhas de adultos e plântulas, e dois tecidos maternos de diásporos. Análises de maternidade foram também realizadas em diásporos e folhas de nove árvores adultas. O número de alelos por loco variou de nove a 15 e as heterozigosidades observada e esperada de 0,214 a 0,864 e 0,745 a 0,892, respectivamente. O sucesso de genotipagem dos locos não diferiu entre os tecidos e variou de 56 a 96%. Os microssatélites mostraram polimorfismo suficiente para atribuir as nove árvores aos seus diásporos. O sucesso de genotipagem em todos os tecidos e a identificação das árvores mães mostraram que os microssatélites são adequados para estudos como estrutura genética espacial e análise de maternidade.

ABSTRACT Nine polymorphic microsatellite loci were isolated and characterized for Cryptocarya mandioccana Meisner, a tree from the Atlantic Rainforest with seeds dispersed by large animals. The loci were characterized using 48 individuals from two populations and their genotyping success tested in four tissues: leaves from adults and seedlings, and two diaspore maternal tissues. Maternity analyses were also performed on diaspores and leaves from nine adult trees. The number of alleles per locus ranged from nine to 15 and the observed and expected heterozygosities ranged from 0.214 to 0.864 and 0.745 to 0.892, respectively. The loci genotyping success did not significantly differ between tissues and varied from 56 to 96%. The microsatellites showed enough polymorphism to assign the nine adult trees to their diaspores. The successful genotyping in all tissues and identification of mother trees show that the microsatellites are suitable for studies such as spatial genetic structure and maternity analyses.

A Mata Atlântica é o bioma melhor conhecido em relação à mastofauna no Brasil, contudo ainda assim apresenta uma série de lacunas de conhecimento sobre a persistência e distribuição de espécies, o que representa um risco adicional à conservação de mamíferos. Neste trabalho, através da coleta de dados por diferentes métodos - armadilhas fotográficas, censos diurnos, registro de vestígios, armadilhas de queda e captura viva - ao longo de oito anos (2004-2012), foi realizado o inventário da mastofauna não voadora do Parque Estadual Carlos Botelho (SP). No total foram registradas 53 espécies, sendo 12 espécies ameaçadas regionalmente e uma exótica (Lepus europaeus), com a presença da maioria dos mamíferos esperados para o Continuum Florestal da Serra do Paranapiacaba. A comunidade de mamíferos não voadores quase completa, e a presença de espécies ameaçadas reforçam o papel desta área protegida para a conservação de mamíferos na Mata Atlântica. Embora, a extinção local de uma espécie, Tayassu pecari, alerte para a necessidade de medidas efetivas de proteção.

The Atlantic Forest is one of the most studied Brazilian biomes in relation to its mammalian fauna. However, there is still a series of gaps of knowledge about the distribution and persistence of some of these species which prevents taking adequate conservation measures to better protect the mammals. In order to make the inventory of the non-volant mammalian fauna of the Carlos Botelho State Park (SP), we compiled data of camera trapping, diurnal census, track records, pitfall and live trapping collected over 8 years (2004-2012). We registered a total of 53 species, of which 12 are regionally threatened and one is an exotic species (Lepus europaeus), including the presence of most mammal species expected for the Paranapiacaba Forest Continuum. The high non-volant mammals species richness allied to the presence of threatened species, strengthen the role of this protected area for mammal conservation in the Atlantic Forest. Although, the local extinction of one species, Tayassu pecari, alert to the need for effective measures of protection.
