Resumo O peixe-espada, Trichiurus lepturus, é um predador oportunista, voraz e piscívoro. Os estudos do comportamento alimentar dos peixes com base na análise do conteúdo estomacal são limitados pelo potencial de identificação visual do material ingerido. No entanto, ferramentas moleculares, em particular o DNA barcode, têm sido utilizadas com sucesso para identificar o conteúdo do estômago. Quando as análises morfológicas não são possíveis, essas ferramentas moleculares podem identificar com precisão os componentes da dieta de um peixe com base em seu conteúdo estomacal. Este estudo utilizou o mini barcode (uma sequencia parcial do gene COI do DNA mitocondrial) para identificar alimentos ingeridos por T. lepturus no litoral Norte do estado de São Paulo, Brasil. Quarenta e seis sequências foram obtidas e combinadas com seis espécies diferentes de peixes: Pimelodus maculatus, Paralonchurus brasiliensis, Isopisthus parvipinnis, Opisthonema oglinum, Harengula clupeola e Pellona harroweri ou como pertencente aos gêneros Lycengraulis e Sardinella. Trichiurus lepturus é um predador oportunista que explora qualquer presa disponível que possua tamanho apropriado. Os resultados indicam que esses peixes migram para águas mais quentes em determinadas épocas do ano, como as encontradas em ambientes estuarinos, onde exploram espécies que se reproduzem neste ambiente. Um exemplo foi Pimelodus maculatus, sendo a espécie mais explorada por T. lepturus, a partir da análise do material coletado.

Abstract The largehead hairtail, Trichiurus lepturus, is an opportunistic, voracious, and piscivorous predator. Studies of fish feeding behavior based on the analysis of stomach contents are limited by the potential for the visual identification of the ingesta. However, molecular tools, in particular DNA barcoding, have been used successfully to identify stomach contents. When morphological analyses are not possible, molecular tools can precisely identify the components of the diet of a fish based on its stomach contents. This study used mini barcoding to identify food items ingested by T. lepturus off the northern coast of São Paulo State, Brazil. Forty-six sequences were obtained and were diagnosed as belonging to six different fish species: Pimelodus maculatus, Paralonchurus brasiliensis, Isopisthus parvipinnis, Opisthonema oglinum, Harengula clupeola, and Pellona harroweri or as belonging to the genera Lycengraulis and Sardinella. Trichiurus lepturus is an opportunistic predator that will exploit an available prey of an appropriate size. The results indicate that these fish migrate to warmer waters, such as those found in estuarine environments, at certain times of the year, where they exploit prey species that reproduce in this environment. One example was Pimelodus maculatus, which was the prey species most exploited based on the analysis of the material collected.

Resumo Prochilodus lineatus é uma espécie de peixe migratório amplamente distribuído na bacia do rio Paraná, principalmente nos rios Grande, Pardo e Mogi-Guaçu localizados em uma região bem desenvolvida do estado de São Paulo. Este estudo analisou a diversidade genética e a estrutura populacional em cardumes de P. lineatus com base na análise temporal de espécimes amostrados ao longo dos anos de 2003, 2005, 2006, 2009, 2010 e 2015 na Cachoeira de Emas no rio Mogi-Guaçu, São Paulo, Brasil. A análise genética realizada com o marcador D-Loop e sete microssatélites revelou variabilidade genética significativa em todos os grupos amostrados. Níveis moderados de estruturação entre os grupos foram identificados com os marcadores microssatélites (Fst = 0.14), enquanto o marcador mitocondrial não revelou padrões de estruturação genética (Fst = 0.01). A variabilidade genética identificada no estoque oscilou ao longo do tempo, caracterizando padrões de estruturação entre as amostras analisadas. A ocorrência de alterações ambientais resultando em aumento das taxas de mortalidade, bem como mudanças no nível de água no ecossistema, entre outros fatores, podem determinar mudanças no comportamento reprodutivo das espécies. A falta de condições ambientais favoráveis para a reprodução na bacia, pode ter resultado na diferenciação das populações de P. lineatus ao longo do tempo.

Abstract Prochilodus lineatus is a species of migratory fish widely distributed in the Paraná River basin, found mainly in the Grande, Pardo and Mogi-Guaçu rivers located in a well-developed region of the state of São Paulo. This study analyzes the genetic diversity and population structure in shoals of P. lineatus based on temporal analysis of specimens sampled over the years 2003, 2005, 2006, 2009, 2010, and 2015 in the Mogi-Guaçu River, São Paulo, at the region of Cachoeira de Emas. Genetic analysis performed using the D-Loop and seven microsatellite marker revealed significant genetic variability in all sampled groups. Moderate levels of structuring between groups were identified with the microsatellite markers (Fst = 0.14), while the mitochondrial marker did not reveal patterns of genetic structuring (Fst = 0.01). The genetic variability fluctuated over time, characterizing patterns of structuring among the analyzed samples. The occurrence of environmental alterations resulting in increased mortality rates, as well as changes in the water level in the ecosystem, among other factors, could determine changes in the reproductive behavior of species. The lack of favorable environmental conditions for reproduction in the basin, as reflected by tests of population bottlenecks, could have resulted in the differentiation of populations of P. lineatus over time.

Abstract The chromosomes of two freshwater stingrays, Potamotrygon motoro and Potamotrygon sp., from the Amazon River basin in Brazil were investigated using integrated molecular (cytochrome c oxidase subunit 1) and cytogenetic analyses. Potamotrygon motoro presented intraspecific variation in the diploid number, with 2n=66 in the females and 2n=65 in the males, while Potamotrygon sp. had a karyotype with 66 chromosomes, in both sexes. The C-banding revealed the presence of heterochromatic blocks accumulated in the centromeric region of all the chromosomes in both species. The FISH assays with 18S DNA probes highlighted the terminal region of three or four chromosome pairs in P. motoro and seven chromosomes in Potamotrygon sp. The rDNA 5S sequences were found in only one chromosomal pair in both species. The interspecific genetic distance based on the COI sequences, between P. motoro and Potamotrygon sp. from Amazon River was 10.8%, while that between the Amazonian P. motoro and Potamotrygon amandae from the Paraná River was 2.2%, and the genetic distance between Potamotrygon sp. and P. amandae was 11.8%. In addition to the new insights on the cytogenetics of the study species, the results of the present study confirmed the existence of heteromorphic sex-linked chromosomes in P. motoro.

RESUMO Rhinoptera bonasus é uma espécie bento-pelágica e altamente migratória, que ocorre do sul dos Estados Unidos ao norte da Argentina. Devido aos efeitos da sobrepesca, R. bonasus está atualmente em risco, classificada pela Lista Vermelha da IUCN como vulnerável. Considerando a falta de dados moleculares disponíveis para R. bonasus, este estudo teve como objetivo descrever a variabilidade genética e estrutura populacional de espécimes amostrados em três ecorregiões do litoral brasileiro (Ecorregião Amazônica, Pará; Ecorregião Nordeste, Pernambuco e Ecorregião Sudeste, Rio de Janeiro, São Paulo e Santa Catarina), por meio de cinco marcadores microssatélites polimórficos. Assim, testaremos as hipóteses de panmixia e filopatria natal. Um total de 69 espécimes analisados revelou diferenciação genética individual e significativa entre os locais amostrados. As análises de Φ ST (0,12), PCA, DAPC e Bayesiana revelaram pelo menos dois agrupamentos genéticos distintos de R. bonasus. Os testes de IBD foram significativos, indicando uma correlação entre a distância genética e geográfica entre as populações, o que pode ser explicado pelo comportamento filopátrico reprodutivo. O comportamento filopátrico associado à mobilidade de R. bonasus pode influenciar os valores de diferenciação observados para todos os loci nas amostras investigadas.

ABSTRACT Rhinoptera bonasus is a bento-pelagic and highly migratory species occurring from southern United States to northern Argentina. Due to overfishing effects, R. bonasus is currently at risk, classified by the IUCN Red List as vulnerable. Considering the lack of molecular data available for R. bonasus, this study aimed to describe the genetic variability and population structure of specimens sampled from three Brazilian coast ecoregions (Amazon ecoregion, Pará; Northeastern ecoregion, Pernambuco and Southeastern ecoregion, Rio de Janeiro, São Paulo and Santa Catarina), through five polymorphic microsatellite markers. Here testing the panmixia hypothesis for Brazilian ecoregions and test natal philopathy. A total of 69 analyzed specimens revealed individual and significant genetic differentiation between the sampled locations. Φ ST (0.12), PCA, DAPC and Bayesian analyses of the genetic population structure revealed at least two distinct genetic R. bonasus groupings. IBD tests were significant, indicating a correlation between genetic and geographical distance among populations, which can be explained by reproductive philopatric behavior. Philopatric behavior associated with R. bonasus mobility may influence the differentiation values observed for all loci in the investigated samples.

Resumo As raias violas Pseudobatos horkelii e Pseudobatos percellens, são listados como “Criticamente em Perigo” (CR) e “Em Perigo” (EN), respectivamente. Ambas as espécies ocorrem no Sul do Oceano Atlântico. Considerando a falta de dados sobre a estrutura genética dessas espécies, o presente estudo avaliou a variabilidade genética e a estrutura populacional de P. horkelii e P. percellens na região sudeste do Brasil e litoral norte da Argentina, com base em sequências de DNA mitocondrial, região de controle (D-loop). Amostras de 135 indivíduos de P. horkelii analisados em seis localidades, situadas no norte da Argentina e ao longo da costa dos estados brasileiros. A média da diversidade nucleotídica foi de 0.0053, o índice ΦST foi de 0.4277 e a análise demográfica de P. horkelii, indicou a existência de estabilidade das populações, com D = 0.9929, Fus = 2.0155, SSD = 0.0817, R = 0.2153. Em 101 exemplares de P. percellens, foram analisados em seis localidades brasileiras ao longo do litoral de Santa Catarina, Paraná e São Paulo. A diversidade nucleotídica média foi de 0.0014 e o valor ΦST de 0.2921, a análise demográfica indicou uma alta taxa de migração de P. percellens entre as localidades analisadas, com D = 0.5222, FS = 0.3528, SSD = 0.01785, R = 0.3890.

Abstract The guitarfishes Pseudobatos horkelii and Pseudobatos percellens meet the criteria for threatened status as Critically Endangered (CR) and Endangered (EN), respectively. Both species occur in the Southern Atlantic Ocean. Considering the lack of data on the genetic structure of these species, the present study evaluated the genetic variability and population structure of the P. horkelii and P. percellens in the southern region of Brazil and the northern coast of Argentina, based on sequences of mitochondrial DNA, Control Region (D-loop). Samples of P. horkelii (n = 135) were analyzed in six localities situated in Northern Argentina, along the Brazilian states’ coast. The mean of nucleotide diversity was 0.0053, the ΦST was 0.4277 and demographic analysis of P. horkelii suggests the existence of stability of the populations, with D = 0.9929, FS = 2.0155, SSD = 0.0817, R = 0.2153. In P. percellens (n = 101) were analyzed from six Brazilian localities along the coast of Santa Catarina, Paraná, and São Paulo. The mean nucleotide diversity was 0.0014 and ΦST value of 0.2921, the demographic analysis indicates a high migration rate of P. percellens among the localities evaluated, with D = 0.5222, FS = 0.3528, SSD = 0.01785, R = 0.3890.

Abstract Background: Endemic systemic mycoses remain a health challenge, since these opportunistic diseases are increasingly infecting immunosuppressed patients. The simultaneous use of antifungal compounds and other drugs to treat infectious or non-infectious diseases has led to several interactions and undesirable effects. Thus, new antifungal compounds should be investigated. The present study aimed to evaluate the activity of liriodenine extracted from Annona macroprophyllata on agents of systemic mycoses, with emphasis on the genus Paracoccidioides. Methods: The minimum inhibitory concentration (MIC) and minimum fungicide concentration (MFC) were determined by the microdilution method. The cellular alterations caused by liriodenine on a standard P. brasiliensis (Pb18) strain were evaluated by transmission and scanning electron microscopy. Results: Liriodenine was effective only in 3 of the 8 strains of the genus Paracoccidioides and in the Histoplasma capsulatum strain, in a very low concentration (MIC of 1.95 μg.mL-1); on yeasts of Candida spp. (MIC of 125 to 250 μg.mL-1), including C. krusei (250 μg.mL-1), which has intrinsic resistance to fluconazole; and in Cryptococcus neoformans and Cryptococcus gattii (MIC of 62.5 μg.mL-1). However, liriodenine was not effective against Aspergillus fumigatus at the studied concentrations. Liriodenine exhibited fungicidal activity against all standard strains and clinical isolates that showed to be susceptible by in vitro tests. Electron microscopy revealed cytoplasmic alterations and damage to the cell wall of P. brasiliensis (Pb18). Conclusion: Our results indicate that liriodenine is a promising fungicidal compound that should undergo further investigation with some chemical modifications.

Abstract: A polymerase chain reaction quantitative method (qPCR) followed by melting curve analysis was used for fast and simultaneous detection of botanical composition of honey samples from Yucatán Peninsula, Mexico. Ten honey samples from 2013 and 2014 production were collected directly from beekeepers and analyzed for Viguiera dentata, Gymnopodium floribundum, Piscidia piscipula, Acacia angustissima and Mimosa bahamensis content. Seven primers from generic genes (Adh1, Hmg2, Brass lip, Plant 1, Plant nest, Act1, and Helli-all) were used to amplify plant species and honey samples DNA. Comparisons of melting curves among plant and honey samples for each primer amplification, revealed a variable taxonomic content M-1 (V. dentata), M-3 (M. bahamensis y G. floribundum), M-4 (G. floribundum), M-8 (M. bahamensis) y M-13 (V. dentata y G. floribundum). M-7, M-11 and M-12 did not have evidence of presence for any of the plant species under study, whilst M-14 and M-15 showed a different plant species amplification pattern. These results correlate to melissopalynological analysis for most cases. P. piscipula was not detected in any honey sample; however, according to melissopalynological analysis A. angustissima was present in M-3 and M-4 even though it was unable to detect it, possibly due to a low or no similarity with generic genes sequence.

Resumen: El método cuantitativo de reacción en cadena de la polimerasa (qPCR), seguido por análisis de curvas de disociación, fue desarrollado para la detección rápida y simultánea de la composición botánica en mieles de la Península de Yucatán, México. Cinco muestras de miel ciclo apícola 2013 y cinco del 2014, se caracterizaron para definir el contenido de cinco especies de plantas de importancia apícola; Viguiera dentata, Gymonopodium floribundum, Piscidia piscipula, Acacia angustissima y Mimosa bahamensis. Siete iniciadores de genes genéricos (Adh1, Hmg2, Brass lip, Plant 1, Plant nest, Act1, y Helli-all) se emplearon para caracterizar las especies vegetales y las muestras de miel. Al finalizar la amplificación se obtuvo una curva de disociación por reacción representando productos específicos de amplificación. Los resultados obtenidos indicaron que el contenido taxonómico en las muestras de miel fue diferencial M-1 (V. dentata), M-3 (M. bahamensis y G. floribundum), M-4 (G. floribundum), M-8 (M. bahamensis) y M-13 (V. dentata y G. floribundum). M-7, M-11 y M-12 no revelaron tener ninguna de las especies analizadas, mientras que M-14 y M-15 presentaron un patrón de amplificación diferente a las especies incluidas en este estudio; concordando con los análisis palinológicos. P. piscipula no mostró ningún patrón de amplificación con ninguno de los iniciadores de este estudio y A. angustissima no fue identificada en ninguna muestra de miel, aun cuando el análisis palinológico reveló presencia de esta especie en M-3 y M-4, posiblemente derivado de la ausencia de similitud con los genes de estudio.

Análises citogenéticas de Potamotrygon aff. motoro e P. falkneri identificaram a ocorrência de um sistema múltiplo de cromossomos sexuais do tipo X1X1X2X2/X1X2Y, em ambas as espécies, com 2n = 66 cromossomos em fêmeas e 2n = 65 cromossomos nos machos. As regiões organizadoras de nucléolos (RONs) identificadas pela reação Ag-RON, evidenciaram marcações múltiplas em ambas as espécies (com variações de 5 a 7 RONs). A técnica de bandamento C, revelou a presença de blocos heterocromáticos localizados nas regiões centromérica em quase todos os cromossomos nas duas espécies em estudo. Através do presente estudo foi evidenciada uma heterogeneidade nos cariótipos, permitindo sugerir que rearranjos cromossômicos, como inversões e/ou translocações, ocorreram durante a evolução cromossômica nas duas espécies desse gênero.

Cytogenetic analysis of Potamotrygon aff. motoro and P. falkneri indicated the occurrence of an X1X1X2X2/X1X2 Y multiple sex chromosome system in both species, with 2n = 66 chromosomes for females and 2n = 65 chromosomes for males. The nucleolus organizer regions (NORs) identified using Ag-NOR technique showed that both species have multiple Ag-NORs (5 to 7 chromosomes stained). C-banding technique indicated the presence of heterochromatic blocks in the centromeric regions of almost all chromosomes in both species. Through this study there was evidence of heterogeneity in the karyotypes, which suggests that chromosomal rearrangements such as inversions and/or translocations occurred during the chromosomal evolution in two species of this genus.

O objetivo deste trabalho foi avaliar a atividade in vitro do resíduo em pelo de cobertura de cães tratados com dinotefuran sobre larvas e adultos de Ctenocephalides felis felis. Foram utilizadas três cadelas da Raça Beagle: a nº 1 foi tratada com uma formulação spray de dinotefuran a 0,834%, a nº 2 com uma formulação "strip-on" de dinotefuran a 30% e a nº 3 foi mantida sem tratamento. Pequenas áreas foram tricotomizadas nos dias 2, 9, 16, 23, 30, 37 e 44 após o tratamento. Para avaliação adulticida e larvicida, foram utilizados pulgas de uma colônia mantida em laboratório. Seis repetições com cada uma contendo 10 exemplares de C. f. felis, acondicionados em tubos de ensaio, da etapa correspondente foram utilizados por dia de desafio. Em cada repetição foi adicionado 0,02 gramas de pelo de cada área tricotomizada e dieta na etapa de larvas. O material da etapa adulto foi avaliado num período de 24 horas, o da etapa larvas foi avaliado após 20 dias de cada desafio. O resíduo no pêlo de cães tratados com ambas as formulações de dinotefuran foi eficaz no controle de larvas por um período de 44 dias, e no controle de adultos as formulações spray e a "strip-on" foram eficazes por um período de 16 e 23 dias, respectivamente.

The aim of this study was to evaluate the residual activity of treated dog's hair coat with dinotefuran on larvae and adults of Ctenocephalides felis felis. Three female Beagle dogs were used. One female dog was treated with 0.834% dinotefuran spray, the second was treated with 30% dinotefuran strip-on and the third was not treated. Some areas of dog's hair were clipped on days 2, 9, 16, 23, 30, 37 and 44 after treatment. For the evaluation of adulticidal and larvicidal activities, flea adults and larvae from the laboratory colony were used. Six repetitions were used with 10 samples of each flea stage per day, placed in assay tubes. In each repetition we added 0.02 g of treated or untreated dog's hair and larval diet for the immature stage. Flea adult assay was evaluated with 24 hours and the flea larvae 20 days after treatment. The residue on hair coat of both treated dogs showed efficacy on the flea larvae control for 44 days. Regarding the flea adults control the spray and strip-on formulations showed efficacy until days 16 and 23, respectively.
