ABSTRACT This paper proposes a conceptual, theoretical and analytical map, aiming to help understand the contemporary process of digitization of industrial companies in developing economies. It examines the relevance of the Schumpeterian hypothesis that technical progress in the digital age can drive a 4th long cycle of industrial dynamism or the 4th Industrial Revolution. It also highlights the complementarity of this hypothesis with the Keynesian view of the business calculation of the rate of return on investment in innovation under uncertainty, changing expectations, and conventions, pointing out the key role of the entrepreneurial drive (animal spirits) in capitalist development. The paper also analyzes the growing challenges that the digital age poses to developing countries, based on the structuralist and institutionalist views of Furtado and Abramovitz, respectively. Resumo O estudo objetivou realizar o levantamento florístico-taxonômico e a distribuição dos representantes da família Rhamnaceae, como parte do projeto “Flora do Ceará: conhecer para conservar”. O trabalho foi embasado na análise de caracteres morfológicos de amostras obtidas em campo, coleções de herbário e bibliografias especializadas. No território cearense, Rhamnaceae está representada por dez espécies pertencentes a cinco gêneros: Colubrina (2), Crumenaria (1), Gouania (3), Rhamnidium (1) e Sarcomphalus (3). As espécies foram registradas em diferentes fitofisionomias, mas ocorrem preferencialmente na Floresta Estacional Semidecídua (Mata Seca) e no Complexo Vegetacional da Zona Litorânea (Mata de Tabuleiro). O táxon com o maior número de registros foi Sarcomphalus joazeiro, ocorrendo em 62 municípios. Sete espécies foram registradas em 14 Unidades de Conservação do Ceará. Colubrina cordifolia, Rhamnidium molle e Crumenaria decumbens ocorrem exclusivamente no domínio da Caatinga e as duas primeiras espécies são endêmicas da região Nordeste. Chaves de identificação, descrições morfológicas, comentários sobre distribuição, ecologia, fenologia e usos, mapas e ilustrações das espécies são apresentados.

Abstract We present the floristic-taxonomic and distribution survey of the family Rhamnaceae as part of the “Flora of Ceará: knowing to conserve” project. The survey was based on the analysis of morphological characters of specimens gathered during field expeditions and from herbaria collections, as well as the specialized literature. Rhamnaceae is represented by ten species in Ceará State belonging to five genera: Colubrina (2), Crumenaria (1), Gouania (3), Rhamnidium (1), and Sarcomphalus (3). The species were encountered in various phytophysiognomies, but especially occur in Seasonal Semideciduous Forests (Mata Seca) and the Coastal Zone Vegetation Complex (Lowland Semideciduous Forest). The species with the highest number of records was Sarcomphalus joazeiro, occurring in 62 municipalities. Seven species were encountered in 14 legally protected conservation areas in Ceará. Colubrina cordifolia, Rhamnidium molle, and Crumenaria decumbens occur exclusively in the Caatinga domain, with the first two being endemic to northeastern Brazil. Identification keys, morphological descriptions, comments on distributions, ecology, phenology, and uses of the species are provided, as well as maps and illustrations.

RESUMO CONTEXTO: Um local comum de tumores neuroendócrinos (TNEs) é o reto. A técnica mais utilizada é a ressecção endoscópica da mucosa com injeção de solução salina. No entanto, as margens profundas costumam ser difíceis de ressecar porque a invasão da submucosa é comum. A ressecção endoscópica sob imersão d’água (RESI) é uma técnica em que o lúmen intestinal é preenchido com água em vez de ar, evitando a necessidade de elevação submucosa. OBJETIVO: Este estudo teve como objetivo avaliar a eficácia e segurança da RESI para a remoção de pequenos TNEs retais (rTNEs). MÉTODOS: Estudo retrospectivo com pacientes que realizaram RESI em dois centros. RESI foi realizada usando um colonoscópio padrão. Nenhuma injeção submucosa foi realizada. Patologistas certificados conduziram avaliação histopatológica. RESULTADOS: RESI foi realizada para pequenos rTNEs em 11 pacientes (nove mulheres) com média de idade de 55,8 anos e 11 lesões (tamanho médio de 7 mm, variando de 3-12 mm). Havia 9 (81%) pacientes com G1 rTNEs e dois pacientes com G2, sendo que todos os tumores invadiam a submucosa sendo apenas um restrito a mucosa. Nenhum caso mostrou invasão vascular ou perineural. Todas as lesões foram removidas em bloco. Nove (81%) ressecções tiveram margens livres. Dois pacientes tiveram envolvimento de margens profundas; um teve biópsias negativas por meio de vigilância endoscópica e o outro perdeu o acompanhamento. Não ocorreram perfurações ou sangramento tardios. CONCLUSÃO: A RESI parece ser uma alternativa eficaz e segura para o tratamento de pequenos rTNEs sem eventos adversos e com altas taxas de ressecção em bloco e R0. Mais estudos prospectivos são necessários para comparar as intervenções endoscópicas disponíveis e para elucidar a técnica endoscópica mais adequada para ressecção de rTNEs.

ABSTRACT BACKGROUND: A common site of neuroendocrine tumors (NETs) is the rectum. The technique most often used is endoscopic mucosal resection with saline injection. However, deep margins are often difficult to obtain because submucosal invasion is common. Underwater endoscopic mucosal resection (UEMR) is a technique in which the bowel lumen is filled with water rather than air, precluding the need for submucosal lifting. OBJECTIVE: This study aimed to evaluate the efficacy and safety of UEMR for removing small rectal neuroendocrine tumors (rNETs). METHODS: Retrospective study with patients who underwent UEMR in two centers. UEMR was performed using a standard colonoscope. No submucosal injection was performed. Board-certified pathologists conducted histopathologic assessment. RESULTS: UEMR for small rNET was performed on 11 patients (nine female) with a mean age of 55.8 years and 11 lesions (mean size 7 mm, range 3-12 mm). There were 9 (81%) patients with G1 rNET and two patients with G2, and all tumors invaded the submucosa with only one restricted to the mucosa. None case showed vascular or perineural invasion. All lesions were removed en bloc. Nine (81%) resections had free margins. Two patients had deep margin involvement; one had negative biopsies via endoscopic surveillance, and the other was lost to follow-up. No perforations or delayed bleeding occurred. CONCLUSION: UEMR appeared to be an effective and safe alternative for treatment of small rNETs without adverse events and with high en bloc and R0 resection rates. Further prospective studies are needed to compare available endoscopic interventions and to elucidate the most appropriate endoscopic technique for resection of rNETs.

RESUMO A criopreservação é uma técnica que pode potencialmente conservar o germoplasma de espécies do gênero Coffea por tempo indeterminado. O objetivo neste estudo foi avaliar os aspectos fisiológicos, bioquímicos e ultraestruturais de sementes criopreservadas de Coffea arabica L., cultivar Catuaí amarelo IAC 62, submetidas a diferentes protocolos de desidratação, pré-resfriamento, resfriamento, reaquecimento e uso de água catódica. De acordo com cada protocolo as sementes foram submetidas à secagem rápida ou lenta, até 17 ou 20% de umidade (base úmida), resfriadas por diferentes maneiras e, em seguida, foram imersas em nitrogênio líquido por 24 horas. Diferentes tempos de reaquecimento em banho-maria também foram utilizados. Análises fisiológicas, bioquímicas e ultra-estruturais foram realizadas nas sementes após as etapas de criopreservação. O teor de umidade a 17% bu é o fator chave para a criopreservação de Coffea arabica L., umidade em que as sementes apresentam melhor qualidade fisiológica e estruturas celulares mais preservadas. O pré-resfriamento das sementes de café antes da imersão em nitrogênio líquido é dispensado por não apresentar vantagens em relação à imersão direta. Os tempos de reaquecimento testados (2, 4 e 6 minutos) e o uso de água catódica não causam alterações na qualidade fisiológica e bioquímica, bem como nas estruturas celulares das sementes criopreservadas de Coffea arabica L. O padrão de estrutura celular observado em todas as sementes indica que o dano da criopreservação é menos drástico nas células dos embriões do que nos endospermas, sendo este último menos tolerante aos estresses de desidratação, pré-resfriamento e reaquecimento.

ABSTRACT Cryopreservation is a technique that may potentially conserve the germplasm of species of the Coffea genus for an indeterminate time. The aim of this study was to evaluate the physiological, biochemical and ultrastructural characteristics of cryopreserved seeds of Coffea arabica L., cultivar Catucaí amarelo IAC 62, which was subjected to different protocols regarding dehydration, precooling, cooling, rewarming and cathode water use. According to each protocol, the seeds were subjected to fast or slow drying to moisture contents of 17 or 20% (wet basis), cooled in different ways, and then immersed in liquid nitrogen for 24 hours. Different rewarming times in a water bath were also used. Physiological, biochemical and ultrastructural analyses were performed on the seeds after the cryopreservation steps. Moisture content at a 17% wb is the key factor for the cryopreservation of Coffea arabica L. seeds, which have better physiological quality and better preserved cell structures. Precooling of coffee seeds before immersion in liquid nitrogen does not provide advantages compared to direct immersion. The rewarming times tested (2, 4, and 6 minutes) and cathode water use did not cause changes in the physiological and biochemical quality or in the cell structures of Coffea arabica L. cryopreserved seeds. The pattern of cell structure observed in all seeds indicates that the damage from cryopreservation is less drastic in the cells of the embryos than in those of the endosperm, with the latter less tolerant to the stresses of dehydration, precooling, and rewarming.

RESUMO O café é umas das principais commodities agrícolas do país, sendo importante a conservação do material vegetal para os programas de melhoramento genético. A criopreservação é uma alternativa promissora para conservar em longo prazo o germoplasma de espécies consideradas recalcitrantes. Entretanto, estudos devem ser conduzidos para alcançar a máxima sobrevivência de plântulas após imersão em nitrogênio líquido. Objetivou-se neste trabalho estabelecer um protocolo de criopreservação para armazenar sementes de Coffea canephora Pierre, aplicando-se dois métodos de criopreservação, resfriamento lento e rápido. As sementes foram submetidas à secagem em sílica gel, até o teor de água de 0.25 g g-1. No primeiro experimento, as sementes secas foram submetidas a tratamentos de resfriamento lento nas velocidades -1 ºC min.-1, -3 ºC min.-1 e -5 ºC min.-1 até as temperaturas finais de -40 ºC, -50 ºC, e -60 ºC, por meio de um biocongelador, e posteriormente imersas em nitrogênio líquido. No segundo experimento, foi selecionado o melhor resultado do primeiro experimento e comparado com o resfriamento rápido, em que as sementes secas, com 0.25 g g-1 de umidade, foram imersas diretamente em nitrogênio líquido. As alterações fisiológicas e bioquímicas ocorridas nas sementes após criopreservação foram avaliadas. As sementes de Coffea canephora respondem melhor a criopreservação por resfriamento rápido quando comparado ao resfriamento lento. A secagem das sementes de Coffea canephora até o teor de água de 0.25 g g-1 não prejudica a viabilidade das sementes. As enzimas catalase e esterase são bons marcadores bioquímicos para sementes de café criopreservadas e sua atividade é maior nas sementes de maior qualidade fisiológica.

ABSTRACT Coffee is one of the main agricultural commodities in the country, and it is important to conservation of plant material for breeding programs. Cryopreservation is a promising alternative for preserving in the long-term the germplasm of species considered recalcitrant. However, studies should be performed to achieve maximum survival of seedlings after immersion in liquid nitrogen. The objective of this work was to find a cryopreservation protocol for storing seeds of Coffea canephora, studying two methods of cryopreservation, slow and fast cooling. Seeds were subjected to drying in silica gel up to the water content of 0.25 g g-1. In the first experiment, dried seeds were subjected to treatments of slow cooling at speeds of -1 ºC min-1,-3 ºC min-1 and -5 ºC min-1 until the end temperatures of -40 ºC, -50 ºC and -60 ºC, by means of a bio freezer and subsequently immersed in liquid nitrogen. In the second experiment, the best result was selected of the first experiment and compared with the rapid cooling, in which dried seeds, with 0.25 g g-1 of water content, were immersed directly into liquid nitrogen. Physiological and biochemical alterations occurring in the seeds after cryopreservation were evaluated. Coffea canephora seeds respond better to cryopreservation by rapid cooling, when compared to slow cooling. Drying, one of the cryopreservation steps does not affect the viability of Coffea canephora Pierre seeds, when these seeds are dried to 0.25 g g-1 of water content. Catalase and esterase enzymes are good biochemical markers for cryopreserved coffee seeds and their activity is greater in larger seed physiological quality.

RESUMO Objetivou-se, com este trabalho, estimar parâmetros genéticos para características do crescimento de animais da raça Nelore Mocho. Foram utilizados pesos corporais (Kg) ajustados aos 205 (P205), 365 (P365) e 550 (P550) dias. Adotou-se modelo multicaraterísticas ajustado para o efeito fixo dos grupos de contemporâneos, idade da vaca ao parto (covariável) e, para os efeitos aleatórios genético direto, genético materno, ambiente permanente materno e residual. As variâncias genéticas diretas (98,80, 276,38 e 547,52 Kg2) e maternas (61,77, 64,63 e 79,85 Kg2), elevaram-se com o avanço das idades. Observou-se redução das variâncias de ambiente permanente materno de P205 para P365 e aumento para P550. As herdabilidades diretas, e seus erros padrão, para P205, P365 e P550 foram respectivamente de 0,21 ± 0,01; 0,38 ± 0,00 e 0,39 ± 0,00. As herdabilidades maternas foram maiores no início da vida dos animais (0,13 ± 0,01, 0,08 ± 0,00 e 0,05 ± 0,00). As correlações genéticas diretas foram superiores a 0,94. As correlações fenotípicas apresentaram valores de moderados a altos (0,50 a 0,87). As correlações genéticas maternas e de ambiente permanente materno foram superiores a 0,91. Ganhos genéticos são esperados, ao se usarem as características do crescimento como critérios de seleção. A existência de variabilidade genética permite a seleção em idades menores, possibilitando a redução do intervalo de gerações e aumento do ganho genético.

ABSTRACT We aimed to estimate genetic parameters for growth traits of Polled Nellore beef cattle. Adjusted weights at 205 (W205), 365 (W365) and 450 (W450) days were used. The effects of contemporary groups (fixed), dam age at calving (covariate), and additive genetic, maternal genetic, maternal permanent environmental and residual (random) were fit under a multi-trait model. The additive genetic (98.80, 276.38 and 547.52 Kg2) and maternal genetic (61.77, 64.63 and 79.85 Kg2) variances estimates increased as the age of the animals increased. Unlikely, a decrease was observed from W205 to W365 for the maternal permanent environmental effect. Direct heritability estimates were, respectively, 0.21 ± 0.01; 0.38 ± 0.00 and 0.39 ± 0.00 for W205, W365 and W450. The maternal heritabilities were higher in lower ages (0.13 ± 0.01, 0.08 ± 0.00 and 0.05 ± 0.00). Genetic correlations were higher than 0.94 whereas phenotypic were between 0.50 and 0.87. Maternal correlations were higher than 0.91 for genetic and permanent environmental effects. Potential genetic gain might be expected by using any of these growth traits as selection criterion. The verified genetic variability may allow the selection of animals in lower ages (e.g., weaning), enabling higher genetic gain and generation interval reduction.

RESUMO A Campomanesia rufa é uma espécie frutífera nativa do Brasil considerada como vunerável pela IUCN (International Union for Conservation of Nature) e poucas informações sobre a propagação são encontradas. Neste contexto, o objetivo do presente estudo foi desenvolver protocolos de germinação in vitro, micropropagação e calogênese para a espécie. Para germinação in vitro, as sementes foram inoculadas em meio MS suplementado com GA3 (Ácido Giberelico), para a indução da parte aérea, o meio de cultura foi suplementado com BA (benzilamina), BAP (6-Benzilamina Purina) ou TDZ (Thidiazuron). Para o crescimento da parte aérea, o meio de cultura contendo BA, BAP e TDZ foi mantido sob lâmpadas fluorescentes brancas e sob diodos emissores de luz (LED). O GA3 também foi testado para elongação de brotações in vitro. Para o controle da oxidação, o meio foi suplementado com PVP (polivinilpirrolidona) e para a indução de calos, 2,4-D (Ácido 2,4-diclorofenoxiacético) . Os resultados mostraram 68% de germinação, independentemente da concentração de GA3. No estágio de propagação, BAP a 4,5 µM induziu maior número de brotações (4,53) e lâmpadas de LED como fonte de luz combinada com o meio de cultura com 1 μM de BAP induziu maior número de brotações (4,08). A maior média de comprimento de C. rufa (31,9 mm) foi obtida usando 8,8 µM de GA3. O uso de PVP (584,3 µM) controla até 27,3% de oxidação em explantes de folhas jovens. O uso de 10 µM de 2,4-D leva a uma maior formação de calos (58,7%). Portanto, pode-se concluir que o uso de BAP é eficiente na indução de brotações, o PVP controla a oxidação de segmentos foliares e o 2,4-D induz calos em C. rufa.

ABSTRACT The Campomanesia rufa is a fruitful species native from Brazil considered as endangered by the IUC (International Union for Conservation of Nature) and low information regarding its propagation is available. In this context, the aim of the present study was to develop in vitro germination, micropropagation and callogenesis protocols for the species. For in vitro germination, seeds were inoculated in MS medium supplemented with GA3 (gibberellic acid) and for shoot induction, the medium was supplemented with three different cytokinins BA (benzyladenine), BAP (6-Benzylaminopurine) or TDZ (Thidiazuron). For shoot growth, culture medium containing BA, BAP, TDZ were maintained under white fluorescent lamps and blue:red light-emitting diodes (LED). GA3 was also tested on in vitro shoot elongation. For oxidation control, the medium was supplemented with PVP (polyvinylpyrrolidone) and for callus induction, 2.4-D (dichlorophenoxyacetic acid). The results showed 68% germination regardless of GA3 concentration. In the propagation stage, BAP at 4.5 µM induced a higher number of shoots (4.53) and LED lamps as the light source combined with the culture medium with 1 μM BAP induced a higher number of shoots (4.08). The highest average of C. rufa length (31.9 mm) was obtained using 8.8 µM GA3. The use of PVP (584.3 uM) controls up to 27.3% oxidation in young leaf explants. The use of 10 µM 2.4-D leads to a higher callus formation (58.7 %). Therefore, it can be concluded that the use of BAP is efficient in the induction of shoots, PVP controls oxidation leaf segments, and 2.4-D induces callus in C. rufa.

Resumo: A criopreservação é uma opção viável para a conservação de germoplasma de café. Entretanto, para que essa técnica apresente total sucesso é de fundamental importância realizar pesquisas que garantam a manutenção da integridade celular antes e após a imersão em NL. Portanto, o objetivo neste estudo foi investigar o teor de água, velocidade e temperatura final de resfriamento mais adequados para criopreservar sementes de Coffea arabica L. As sementes foram secadas em sílica gel até teores de água de 5, 10, 15, 20, 30 e 40% bu, submetidas a tratamentos de resfriamento lento nas velocidades -1, -3 e -5 ºC min-1 até às temperaturas finais de -40, -50 e -60 ºC e diretamente imersas em NL. Após armazenamento as sementes foram reaquecidas por dois minutos à 40 ºC. A sobrevivência e a viabilidade de sementes e embriões foram avaliadas por meio do teste de tetrazólio e de germinação. Por meio do teste de tetrazólio, observa-se que embriões excisados das sementes criopreservadas são menos sensíveis à criopreservação quando comparados às sementes inteiras. O teor de água de 20% bu permite maior sobrevivência das sementes de Coffea arabica L., bem como de embriões zigóticos após a criopreservação, dependendo das velocidades e temperaturas finais de pré-resfriamento.

Abstract: Cryopreservation is a viable option for conservation of coffee germplasm. However, for this technique to be completely successful, it is of fundamental importance to carry out studies that ensure maintenance of cell integrity before and after immersion in liquid nitrogen (LN). Therefore, the aim in this study was to investigate the water content, cooling rate, and final temperature most suitable for cryopreservation of Coffea arabica L. seeds. The seeds were dried by silica gel to water contents of 5, 10, 15, 20, 30 and 40 % wb, subjected to slow cooling treatments at speeds of -1, -3 and -5 °C min.-1 to final temperatures of -40, -50 and -60 °C and then directly immersed in LN. After storage, seeds were rewarmed at 40 °C for two minutes. The survival rate and viability of the seeds and embryos were evaluated by the tetrazolium and germination tests. Results of the tetrazolium test indicate that embryos excised from cryopreserved seeds are less sensitive to cryopreservation than whole seeds are. The water content of 20% wb and the use of zygotic embryos led to the highest survival rate of the coffee seeds, depending on the cooling rate and the final temperature of precooling.

RESUMO O dendezeiro é uma monocotiledônea, lenhosa e oleaginosa de elevada importância econômica, devido à alta produção de óleo por seus frutos. A germinação das sementes é, atualmente, o método convencional mais utilizado para a propagação do dendezeiro, porém, o sucesso por meio dessa via é limitado pela demora e a baixa porcentagem de germinação. Uma alternativa para a propagação em larga escala do dendezeiro é a técnica biotecnológica de embriogênese somática, mas o enraizamento das plantas germinadas a partir dos embriões somáticos é uma etapa difícil e de grande importância para posterior aclimatização e sucesso na propagação. Este trabalho foi realizado com o objetivo de avaliar o efeito das auxinas ácido indol acético (AIA) e ácido indol-butírico (AIB) no enraizamento de embriões somáticos de dendezeiro híbrido Tenera. Plantas obtidas via embriogênese somática foram inoculadas em meio MS modificado, com 10% de sacarose e 0,6% de ágar e suplementado com AIA ou AIB, nas concentrações 5 µM, 10 µM, 15 µM e na ausência de regulador de crescimento. Depois de 120 dias foram analisados os parâmetros presença de raiz, tipo de raiz, comprimento da maior raiz, número de folhas e comprimento da parte aérea. A presença de reguladores de crescimento foi favorável ao enraizamento, tendo as plantas cultivadas com o regulador de crescimento AIB na concentração de 15 µM apresentado maior porcentagem de enraizamento (87%) e melhores resultados para os parâmetros número de raízes (1,33) e comprimento da parte aérea (9,83).

ABSTRACT Oil palm is a woody monocot of economic importance due to high oil production from its fruits. Currently, the conventional method most used to propagate oil palm is seed germination, but success is limited by long time requirements and low germination percentage. An alternative for large-scale propagation of oil palm is the biotechnological technique of somatic embryogenesis. The rooting of plants germinated from somatic embryos is a difficult step, yet it is of great importance for later acclimatization and success in propagation. The aim of this study was to evaluate the effect of the auxins indole acetic acid (IAA) and indole butyric acid (IBA) on the rooting of somatic embryos of Tenera hybrid oil palm. Plants obtained by somatic embryogenesis were inoculated in modified MS medium with 10% sucrose and 0.6% agar and supplemented with IAA or IBA at concentrations of 5 µM, 10 µM, and 15 µM, and the absence of growth regulators. After 120 days, the presence of roots, root type, length of the longest root, number of roots, number of leaves, and shoot length were analyzed. Growth regulators were favorable to rooting; plants cultivated with IBA growth regulator at 15 µM showed higher rooting percentage (87%) and better results for the parameters of number of roots (1.33) and shoot length (9.83).

RESUMO A conservação dos recursos genéticos vegetais é uma medida importante para prevenção do processo de erosão genética. Bancos de sementes são as formas mais comuns de conservação ex situ, entretanto, sementes de cafeeiro não podem ser armazenadas por métodos convencionais. A criopreservação é uma alternativa viável para conservação em longo prazo de espécies, como o cafeeiro, que produzem sementes intermediárias ou recalcitrantes. O objetivo deste trabalho foi criopreservar embriões zigóticos de Coffea arabica L. cv Catuaí Vermelho avaliando o efeito da desidratação antes da criopreservação, e da reidratação osmótica após o descongelamento. Antes da criopreservação foram testados e determinados os teores de umidade em diferentes tempos de desidratação (0, 15, 30, 60 e 120 min). Após a desidratação os embriões foram criopreservados por uma hora em nitrogênio líquido, e após o descongelamento foram reidratados utilizando solução osmótica. Para avaliação da viabilidade de embriões desidratados e/ou criopreservados, utilizou-se o teste com o cloreto de 2,3,5-trifenil tetrazólio. Embriões não desidratados não sobreviveram ao congelamento. Embriões desidratados até cerca de 20% de umidade que não passaram por uma reidratação osmocondicionada, não germinaram. Embriões desidratados até cerca de 20% de umidade, criopreservados e reidratados lentamente após o descongelamento, apresentaram 98% de germinação. A máxima viabilidade obtida através do teste de tetrazólio foi 75% após 60 minutos de desidratação. Portanto, embriões zigóticos de cafeeiro podem ser criopreservados utilizando desidratação física em sílica gel por 60 minutos (23% de teor de umidade), seguida de reidratação com solução osmótica após o descongelamento. Esse método permitiu a germinação de 98% dos embriões criopreservados.

ABSTRACT Conservation of plant genetic resources is important to prevent genetic erosion. Seed banks are the most common method of ex situ conservation; however, coffee seeds can not be stored by conventional methods. Cryopreservation is a viable alternative for long-term conservation of species that produce intermediate or recalcitrant seeds, as coffee. The aim of this work was to cryopreserve Coffea arabica L. cv Catuaí Vermelho IAC 144 zygotic embryos, and analyse the effects of dehydration prior cryopreservation and osmotic rehydration after thawing, in embryos germination and seedlings formation after cryopreservation. Prior to cryopreservation, different dehydration times (0, 15, 30, 60 and 120 min) were tested. Dehydrated embryos were cryopreserved in liquid nitrogen for 1 hour, and after thawing were rehydrated by osmotic solutions. Dehydrated and non-cryopreserved embryos were also analysed. The test with 2,3,5 triphenyl tetrazolium chloride (TTC) was used to evaluate the embryos viability. Non-dehydrated embryos did not survive after freezing. Embryos that were dehydrated until 20% of the moisture content did not germinate when osmotic rehydration was not performed. In contrast, cryopreserved embryos with the same moisture content presented 98% germination when they were rehydrated slowly in osmotic solution. According to tetrazolium tests, embryos presented maximum viability (75%) after dehydration for 60 minutes (23% moisture content). Therefore, coffee zygotic embryos (Coffea arabica L. cv. Catuaí Vermelho) can be successfully cryopreserved using physical dehydration in silica gel for 60 minutes (23% moisture content), followed by osmotic rehydration after thawing. This method allowed a germination of 98% of cryopreserved zygotic embryos.

In this paper, Genetic Algorithm (GA) is used to search for combinations of learning algorithms and associated parameters with maximum accuracy. An important feature of the approach is that the GA initial population is formed by using parameter values gathered from ExpDB (a public database of data mining experiments). The proposed approach was implemented in a tool called EMiner, built on top of a grid based software infrastructure for developing collaborative applications in medicine and healthcare domains (ECADeG project). Experiments on 16 datasets from the UCI repository were performed. The results obtained have shown that the strategy of combining the data from ExpDB via GA is effective in finding classification models with good accuracy.

O objetivo deste trabalho foi avaliar a eficiência da droplet vitrification e vitrificação na criopreservação de ápices caulinares de mangabeira. Foi avaliada a retomada de crescimento de ápices de mangabeira criopreservados conforme a técnica de droplet vitrificationutilizando diferentes tempos de tratamento com a solução de vitrificação (15, 30, 45 e 60 min.) e submetidos a diferentes períodos de pré-cultivo (ausência, 24 ou 48h) em meio com 0,3 M de sacarose antes da criopreservação. Além desta técnica, foi avaliado o efeito da técnica clássica de vitrificação utilizando quatro tempos de exposição à solução de vitrificação (15, 30, 45 e 60 min.). Foram observadas diferenças significativas no tempo de exposição para a técnica de vitrificação e droplet vitrification.Para ambas as técnicas o tempo de imersão de 60 minutos promoveu uma retomada de crescimento superior a 70% nos ápices criopreservados. O pré-cultivo promoveu o aumento na sobrevivência dos explantes criopreservados. As técnicas de criopreservação de ápices caulinares por droplet vitrification e vitrificação mostraram-se viáveis para a conservação em tempo prolongado de mangabeira.

The aim of this study was to evaluate the efficiency of vitrification and droplet vitrification for the cryopreservation of Hancornia speciosa shoot tips. The shoot tips were subjected to four different periods of exposure (15, 30, 45 and 60 min.) to plant vitrification solution 2 (PVS2) before plunging into liquid nitrogen. We evaluated the regrowth of H. speciosa shoot tips that were cryopreserved by the classical vitrification technique and by the droplet vitrification technique. Shoot tips were submitted to different periods of pre-culture (absence, 24 or 48h) in a medium containing 0.3 M sucrose prior to cryopreservation. With a PVS2 exposure period of 60 min., significant differences were observed between the two techniques used in this study with respect to regrowth rates; however, both cryopreservation methods allowed for a shoot tip regrowth rate of over 70%. Use of a preculture step increased the regrowth rate of cryopreserved shoot tips. Shoot tip cryopreservation techniques employing vitrification and droplet vitrification proved to be viable for the long-term storage of mangaba tree samples.
