Abstract The Santos Basin (SB) is the main petroliferous basin in the Brazilian continental margin and one of the most studied marine areas in Brazil. However, historical data suggest that new efforts should be carried out to acquire quantitative biological data, especially in the deep sea, to establish the baseline of essential ocean variables in different ecosystems for future monitoring programs. The Brazilian energy company Petrobras planned and executed 24 oceanographic cruises over a period of 2 years to assess the benthic (SANSED cruise) and pelagic (SANAGU cruise) systems of the SB (356 days at sea in 2019 and 2021/2022). These efforts were part of the Santos Project, which comprised a comprehensive environmental study aimed at investigating benthic and pelagic variables to characterize ecology, biogeochemistry, thermohaline properties of water masses, and ocean circulation patterns, geomorphology, and sedimentology, as well as organic and inorganic chemistry. Here we present the detailed sampling designs and the field methods employed on board, during the SB scientific cruises. All sampling protocols were based on standardized approaches. For the benthos analyses, triplicate sediment samples were performed using a GOMEX-type box corer (0.25 m²) or a large modified Van Veen grab (0.75 m²) at 100 stations ranging from 25 to 2400 m depth. At each station, 25 geochemical and physico-chemical parameters were analyzed in addition to micro-, meio-, and macrofauna and living foraminifera samples. For the pelagic system, 60 stations were selected to investigate the plankton community, ranging in size from pico- to macroplankton, through vertical, horizontal, and oblique net hauls (20, 200, and 500 μm mesh size), as well as 25 biogeochemical parameters collected with an aid of a CTD-rosette sampler. Part of this scientific information also serves the Regional Environmental Characterization Project (PCR-BS) in support of Petrobras’ Santos Basin drilling licensing process led by the Brazilian Environmental Agency – IBAMA. This project contributes to the sustainable development of the SB, in line with the guidelines of the United Nations Decade of Ocean Science for Sustainable Development.

ABSTRACT The Atlantic Forest is a biome constantly threatened by human disturbances. The suppression of vegetation by road works highlights the need for conservation projects for the species that occur there. The success of rescuing and relocating plants, such as the Syagrus romanzoffiana palm, can contribute to the conservation of these species. The objective of this work was to study the growth of S. romanzoffiana seedlings rescued from areas of suppression of the “Rodoanel Mário Covas” - North Section - and transplanted in natural areas of the Cantareira State Park, São Paulo, SP state. Measurements of height, stem diameter, number of leaves and clod volume of seedlings were obtained. The transplant was carried out in three light conditions after being separated into three clod volume classes and three crown diameter classes and observed for one year. The percentage of survival increase in height, crown diameter, number of leaves and photosynthetic pigments were evaluated. The experimental design was completely randomized in a 3 x 3 x 3 factorial scheme (luminosity x crown diameters x clod volumes) with three replications of five plants each. The data obtained were subjected to analysis of variance and the means compared to each other using the Tukey test (5%). Survival was over 80% in all environments. There was greater growth, mainly regarding the stem diameter, in the small clearing. The quantification of chlorophyll a, b and carotenoids, showed higher pigments content for plants kept in the shade. As for the chlorophyll a/b ratio, the results showed a greater relationship in plants kept in shade, with increasing values up to 12 months. It can then be concluded that S. romanzoffiana has little restriction on the relocation environment, but with preference for small clearings, and it is unnecessary that they are relocated with large clod.

Abstract The construction of highways causes several impacts on the environment, including, the suppression of vegetation, which can lead to reduced biodiversity. One mitigating measure involves reallocating plant to protected areas and orchids might be important for this use. An example could be the terricolous orchid Sauroglossum nitidum. In an attempt to optimize the survival of S. nitidum following reallocation, the aim of this study was to investigate whether the fertilization can increase the survival of this orchid. For this, five lots with ten plants per lot were randomly separated, with five plants submitted to soluble fertilization with NPK 10-52-10 (1 g.L-1), spraying each plant with 1 mL of the fertilizer, and the other five sprayed with the same volume of distilled water. This treatment was repeated fortnightly for two months before reallocation. The plants were relocated in September 2015 and monitored monthly until November 2016. Overall orchid survival was 92%. Additionally, flowering was observed to be significantly higher in fertilized plants. In conclusion, reallocation of S. nitidum with previous NPK fertilization is recommended.

Abstract Glucocorticoids are produced and released by the adrenal gland and become elevated in response to stress. Although glucocorticoids are well known for their immunosuppressive effects, less is known about their effects on B cells. ABCB1 is an efflux pump expressed in both cancer and normal cells, modulating the gradient of various metabolites, including hydrocortisone. Our goal was to evaluate the effect of this glucocorticoid on murine B cell differentiation and whether sensitivity to hydrocortisone could be related to ABCB1 activity in vivo. C57BL/6 mice received one or three consecutive i.p. injections of hydrocortisone (70, 140 and 200 mg/kg/day). ABCB1 activity was evaluated via the rhodamine-123 transport and inhibited by cyclosporin A in hydrocortisone-treated and control mice. Cells from bone marrow, spleen and blood were counted, incubated with antibodies and analyzed by flow cytometry. A single hydrocortisone injection did not alter the number of bone marrow subsets. Conversely, three daily injections were able to reduce the cell number of most bone marrow subsets, excepting c-kit-sca-1+ and mature B cells. This treatment reduced marginal zone, follicular and transitional B cells, though splenic subsets were more resistant than bone marrow B cells. Recirculating follicular B cells in the blood were resistant to hydrocortisone. With the exception of follicular B cells, all subpopulations exhibited ABCB1 activity. However, hydrocortisone treatment did not affect ABCB1 activity in most subsets analyzed. Results suggest that hydrocortisone is able to regulate B cell lymphopoiesis although ABCB1 activity is not related to the susceptibility to that glucocorticoid in B cell subsets.

Abstract: Amaranthus spp. are nephrotoxic plants popularly known as "pigweed". In cases of poisoning by these plants, the main histopathological alteration is found in the kidneys as toxic tubular nephrosis; however, in some cases, there may be cardiac changes. ECG changes associated with hyperkalemia have been described in pigs poisoned by Amaranthus retroflexus. Degeneration and necrosis of myocytes have been described in pigs poisoned by A. caudatus and sheep poisoned by A. spinosus. There are doubts regarding cardiac changes, since in most cases they are incipient and don't exhibit inflammatory reaction, which can lead to misinterpretation. For this study, paraffin blocks with tissues from a poisoning outbreak by A. spinosus in southeastern Brazil were used. The objective of the study was to detect the presence of incipient regressive changes in the myocardium of sheep poisoned by A. spinosus using anti-troponin C antibody-based immunohistochemistry. Fragments of hearts from 8 adult sheep and 2 fetuses naturally poisoned by A. spinosus were used. In the immunohistochemistry evaluation of the 8 hearts from the adult sheep there were several groups of myocytes with significant decrease or absence of immunoreactivity for anti-troponin C antibody. In most cases, these same areas on Hematoxylin and Eosin (HE) staining exhibited changes that varied from mild cellular tumefaction to increased eosinophilia, as well as loss of striation, cell lysis and karyolysis, sometimes accompanied by inflammatory infiltrate. In four cases, it was possible to notice that several small groups of myocytes exhibited decreased immunoreactivity corresponding to cells with undetectable or very subtle alterations on HE staining. The hearts of both fetuses did not exhibit areas with loss or decreased immunoreactivity for the anti-troponin C antibody. The results of this immunohistochemical evaluation confirmed the presence of incipient regressive changes and confirmed the occurrence of well-established necrotic lesions in the myocardium of sheep poisoned by Amaranthus spinosus. There was correspondence between areas where immunostaining for troponin was absent and areas with the most marked lesions observed after HE staining. Additionally, the technique was able to detect lesions at a very early stage (myocardial areas with no morphological signs of insult), and demonstrated that the most insulted/injured areas were larger than estimated by observation of HE-stained samples. This study demonstrates the need to better investigate the possible participation of hyperkalemia and other metabolic alterations present in acute renal lesion in the genesis of rapid-onset cardiac lesions, i.e. those not related to vascular lesions caused by long-term uremia.

Abstract: Sodium monofluoroacetate (MF) is the toxic principle of several plants that cause "sudden death" of cattle in Brazil. Groups of cardiomyocites with high cytoplasmic eosinophilia are sometimes observed in animals poisoned by MF. However, this cardiac alteration is difficult to interpret, as there is no inflammatory reaction and it must be differentiated from artifacts. The present study had the objective to detect the presence of early regressive lesions in the myocardium of sheep and cattle experimentally poisoned by MF through immunohistochemistry with troponin C (cTnC). Fragments of the heart muscle from six cattle (three received, orally, single doses of 0.5mg/kg and the others, single doses of 1.0mg/kg) and five sheep (one received, orally, single dose of 0.5mg/kg, the other two received single doses of 1.0mg/kg, one received sublethal daily doses of 0.1mg/kg for four days, and another received daily sublethal doses of 0.2mg/kg for six days) were submitted to immunohistochemistry with antibody anti-cTnC. In the cardiomyocites of cattle and sheep, it was possible to observe reduction of the expression levels for cTnC in the cytoplasm of groups of cardiac muscle fibers. Significant reduction of immunoreactivity ocurred overall in cardiomyocites that presented high cytoplasmic eosinophilia. The decrease or absence of expression for cTnC in animals poisoned by MF allowed to estabilish the difference between coagulative necrosis of cardiomyocites and artifacts caused by fixation. This indicates that this method can be used safely to identify any lesions, early regressive or not, in the myocardium independently of the cause. It is also possible to affirm that poisoning by MF as well as the one caused by "sudden death" causing plants can progress with necrotizing myocardial lesions.

Summary Objective: it is broadly accepted, but little explored, that obese children practice less physical activity and eat more. This study has the objective of comparing feeding habits and physical activity between eutrophic and overweight/obese children and adolescents. Methods: 126 students with ages ranging from 6 to 18 years were evaluated. Eutrophic and overweight/obese students were compared according to calorie intake, macro and micronutrients, prevalence of physical inactivity and ingestion of micronutrients. Results: differences were observed in the amount of calories ingested per unit of BMI (eutrophic, 97.6, and overweight/obese, 70.5, p=0.0061), as well as in calcium intake (eutrophic, 546.2, and overweight/obese, 440.7, p=0.0366). Both groups presented sedentarism, as well as a high prevalence of micronutrient intake deficiency, especially calcium and vitamins A, E, and C, but with no difference observed between eutrophic and overweight/obese subjects. Conclusion: energy and macronutrients consumption, as well as physical activity, were similar between eutrophic and overweight/obese. Calcium intake was lower in the overweight/obese group and the ingestion of vitamin C was lower in the eutrophic group. These results demonstrate the importance of considering all etiologic factors that may lead to obesity, so that new strategies for prevention and control may be added to traditional interventions.

Background:Recent studies have suggested that B-type Natriuretic Peptide (BNP) is an important predictor of ischemia and death in patients with suspected acute coronary syndrome. Increased levels of BNP are seen after episodes of myocardial ischemia and may be related to future adverse events. Objectives:To determine the prognostic value of BNP for major cardiac events and to evaluate its association with ischemic myocardial perfusion scintigraphy (MPS).Methods:This study included retrospectively 125 patients admitted to the chest pain unit between 2002 and 2006, who had their BNP levels measured on admission and underwent CPM for risk stratification. BNP values were compared with the results of the MPS. The chi-square test was used for qualitative variables and the Student t test, for quantitative variables. Survival curves were adjusted using the Kaplan-Meier method and analyzed by using Cox regression. The significance level was 5%.Results:The mean age was 63.9 ± 13.8 years, and the male sex represented 51.2% of the sample. Ischemia was found in 44% of the MPS. The mean BNP level was higher in patients with ischemia compared to patients with non-ischemic MPS (188.3 ± 208.7 versus 131.8 ± 88.6; p = 0.003). A BNP level greater than 80 pg/mL was the strongest predictor of ischemia on MPS (sensitivity = 60%, specificity = 70%, accuracy = 66%, PPV = 61%, NPV = 70%), and could predict medium-term mortality (RR = 7.29, 95% CI: 0.90-58.6; p = 0.045) independently of the presence of ischemia. Conclusions:BNP levels are associated with ischemic MPS findings and adverse prognosis in patients presenting with acute chest pain to the emergency room, thus, providing important prognostic information for an unfavorable clinical outcome.

Abstract An updated inventory of Brazilian seed plants is presented and offers important insights into the country's biodiversity. This work started in 2010, with the publication of the Plants and Fungi Catalogue, and has been updated since by more than 430 specialists working online. Brazil is home to 32,086 native Angiosperms and 23 native Gymnosperms, showing an increase of 3% in its species richness in relation to 2010. The Amazon Rainforest is the richest Brazilian biome for Gymnosperms, while the Atlantic Rainforest is the richest one for Angiosperms. There was a considerable increment in the number of species and endemism rates for biomes, except for the Amazon that showed a decrease of 2.5% of recorded endemics. However, well over half of Brazillian seed plant species (57.4%) is endemic to this territory. The proportion of life-forms varies among different biomes: trees are more expressive in the Amazon and Atlantic Rainforest biomes while herbs predominate in the Pampa, and lianas are more expressive in the Amazon, Atlantic Rainforest, and Pantanal. This compilation serves not only to quantify Brazilian biodiversity, but also to highlight areas where there information is lacking and to provide a framework for the challenge faced in conserving Brazil's unique and diverse flora.

The South American water rat Nectomys squamipes is a wild mammal reservoir of Schistosoma mansoni in Brazil. In the present study, wild rodents were collected in the field and categorized into two groups: infected and uninfected by S. mansoni. Blood was collected to analyze changes in the serum glucose level (mg/dL) and liver fragments were used to determine the hepatic glycogen content (mg of glucose/g tissue). The histological examination showed inflammatory granulomatous lesions in different phases of development in the liver of rodents naturally infected with S. mansoni, in some cases with total or partial occlusion of the vascular lumen. Early lesions were characterized by the presence of inflammatory infiltrate around morphologically intact recently deposited eggs. Despite the significance of these histological lesions, the biochemical changes differed in extent. N. squamipes naturally infected by S. mansoni showed no variation in hepatic glycogen reserves. These findings were accompanied by a significant increase in plasma glucose contents, probably as a consequence of amino acids deamination, which are degraded, resulting in the formation of intermediates used as precursors for the glucose formation, without compromising the reserves of liver glycogen. In the wild, naturally infected N. squamipes can maintain S. mansoni infections without undergoing alterations in its carbohydrate metabolism, which minimizes the deleterious effects of S. mansoni.

Quatorze lectinas foram utilizadas (Con-A, DBA, SBA, PNA, SJA, RCA-I, UEA-I, WGA, SWGA, GSL, PSA, PHA-L, PHA-E e LCA).
Inspite that Brachiaria spp. are most important forage grasses for beef cattle raising in Brazil, they can under certain conditions be toxic, cause outbreaks of hepatogenic photosensibilization and determine severe economic losses. Animals grazing toxic Brachiaria spp. commonly show in their liver foamy macrophages, isolated or grouped together, and crystals inside the biliary ducts. Steroidal saponins have been identified in these crystals and are liable for damage the liverleading to accumulation of phylloerytrin. Occasionally negative images of the crystals may be seen in the cytoplasm of foamy macrophages. The pathogenesis of formation and the type of material stored in the foamy cells is still unknown. The lectin histochemistry aims to assist in the detection of foamy macrophages and consequently in the diagnosis of the poisoning. Lectin histochemistry detects what kind of specific sugar residues exists in the cytoplasm of the foamy cells. The purpose of the present study was to identify the most suitable lectins the detection of steroidal saponins in the liver and kidney of sheep with photosensitization caused by Brachiara decumbens. Fragments of liver and kidney of fifteen sheep of both sexes and variable age, from Conceição do Mato Dentro, Minas Gerais, southeastern Brazil, and one sheep kept free grazing Brachiaria spp. were evaluated by lectin histochemistry. Fourteen lectins were used (Con-A, DBA, SBA, PNA, SJA, RCA-I, UEA-I, WGA, SWGA, GSL, PSA, PHA-L, PHA-E and LCA). It was found that the PNA lectin detected in the liver of sheep with photosensitization, caused by ingestion of Brachiaria decumbens, showed specificity and great binding reactivity for foamy macrophages, and also specificity and slight binding reactivity for hepatocytes. The WGA lectin showed specificity and moderate binding reactivity for foamy macrophages of the liver, and also specificity and slight binding reactivity for hepatocytes. The SBA, GSL and LCA lectins showed specificity and moderate binding reactivity for foamy macrophages, but were not specific for hepatocytes. The PNA lectin showed greater binding reactivity in the kidney of the sheep. Lectin histochemistry evaluation had not yet been performed in liver and kidney of sheep with photosensitization caused by Brachiaria spp.

The postharvest conservation of cut Heliconia flowers is an important factor to the success of commercialization, especially with regard to exportation. In the present study, the maximal storage time of cut inflorescences of Heliconia bihai cv. Lobster Claw at two different temperatures (12 and 19 °C) was evaluated and compared to laboratory conditions (25 °C, control treatment). Changes in visual quality, fresh weight and bract color (L*, a* and b*) were determined. The visual quality of the inflorescences and fresh weight decreased with time in all treatments. Symptoms of chilling injury were observed on the inflorescences stored at 12 °C for six and eight days. Bract color was not affected by temperature, storage time or the senescence process. The results indicate that a temperature of 12 °C is not recommended for a storage time longer than four days, whereas 19 °C can be used for a storage time of up to eight days for cut inflorescences of H. bihai cv. Lobster Claw.

BACKGROUND: Few studies have focussed on the transepidermal elimination of parasites in Jorge Lobo's disease (lobomycosis). OBJECTIVE: To identify the morphological features of the transepidermal elimination of parasites in lobomycosis. METHODS: Sections were obtained from paraffin-embedded biopsy specimens of patients with lobomycosis and stained with hematoxylin-eosin for microscopic examination. Only the presence of parasites in epidermal structures was considered to constitute transepidermal elimination. RESULTS: Forty biopsies from 37 patients were included in the study (31 males and 6 females). The mean age of patients was 51.03 years (range 29-80 years). Biopsies performed over a period of 37 years (1967-2003) were used, from which 511 sections were obtained (a mean of 12.77 sections per case; range 2-39 sections per case). Transepidermal elimination of parasites was found in 110/511 (21.52%) and was absent in 401/511 sections (78.48%) (p<0.0001). Features consistent with the phenomenon were found in 15/37 patients (40.5%) and were absent in 22/37 (59.5%) (p>0.05). Parasites in the epidermis were detected within hyperplastic infundibula, either connected in chains or as isolated units, associated or not with inflammatory cells. CONCLUSION: Features consistent with transepidermal elimination of parasites were found in a statistically nonsignificant number of patients in the sample (p>0.05), suggesting that in Jorge Lobo's disease, this phenomenon invariably occurs through the infundibular epithelium. Future studies are required to evaluate the significance of this finding in the epidemiology of mycosis.

Pernambuco State produces tropical flowers (Heliconiaceae, Zingiberaceae, Costaceae, Araceae) mainly in the rain forest zone and coastal lands. The climatic conditions of the region favour the blooming and the obtaining of flowers with good color, size and quality. The flower industry in Pernambuco is organized in associations and cooperatives and has increased in the last 10 years, due to private and governmental investments, giving to the farmers financial aid, research support and capacitation for exporting. This work describes the procedures used during harvest and postharvest manipulation of tropical flowers, with the objective of helping growers to reach new market niches.

The volatile constituents obtained from a static cryogenic headspace of Caesalpinia echinata Lam. (Leguminosae) showed E-beta-ocimene as the major compound (57.2%), beside other monoterpenes, C6 derivatives like n-hexanal and (E)-2-hexenal and nitrogen compounds such as indole and methyl anthranilate. From the essential oil of the leaves obtained by hydrodistillation in a Clevenger apparatus, (E)-3-hexen-1-ol was identified as the major constituent while phenolic compounds were the most representative class of secondary metabolites.
