ABSTRACT Introduction: Surface treatment is an important technique to increase adhesion between implants and bones, improving its mechanical characteristics and consequently the patient’s comfort. Objectives: Ni-Cr alloys were the object of study in this work, with the purpose of analyzing and evaluating the effect of thin films deposition of titanium nitride via plasma, on its surface and comparing with cathodic cage (CC) and hollow cathode (HC) methods, for dental applications. Methods: Eighteen samples were prepared and the experiment was conducted in two steps: the pre-sputtering (1h, 350 °C, gases: Ar and H) and sputtering (4h, 450 °C, gases: Ar, Ni, and H). To characterize and compare the samples with those of reference, the Scanning Electron Microscopy (SEM), X-Ray Diffraction (XRD), Vickers Microhardness and wettability tests were used. Results: The results presented by SEM showed that during the surface treatment using CC, voids were formed, and using HC, the samples showed a more homogenous behavior. In microhardness tests, using a 25gf load, it was possible to observe that the HC method allowed an increase of 87% when compared to the reference and treated samples. Significance: Lastly, it can be concluded that both methods are suitable for Ni-Cr alloy surface treatment, and the HC technique, a method already used by dental professionals, presents better results due to the formation of a thicker film layer.

Abstract The aim of this study was to determine the resistance of gastrointestinal nematodes in goats and sheep to the anthelmintic drugs levamisole, ivermectin, and albendazole in the metropolitan region of São Luís Island, Maranhão, Brazil. Fecal samples were collected from 150 animals across four different farms; two farms had goats, and the other two had sheep. The samples were then randomly divided into three to four groups of 10 animals: Group I: control, without treatment; Group II: ivermectin treatment; Group III: levamisole treatment; and Group IV: albendazole treatment. Stool samples were collected from the rectal ampulla one day before treatment and 10 days after anthelmintic treatment. Individual coproparasitological examinations were performed using the modified McMaster technique at the Animal Health Laboratory of the Federal Institute of Maranhão, Campus São Luís-Maracanã. The efficacies of the anthelmintic drugs against gastrointestinal nematodes in goats and sheep were: 14.28%, and 13.6% for ivermectin; 0% and 79.4% for levamisole; and 59.8% and 3.43% for albendazole, respectively Gastrointestinal nematodes demonstrated multiple anthelmintic resistance, as the percentage reduction in egg count was less than 95% and the lower limit of the confidence interval was less than 90%.

Resumo Objetivou-se determinar a resistência de nematoides gastrintestinais aos anti-helmínticos levamisol, ivermectina e albendazol em caprinos e ovinos da região metropolitana da Ilha de São Luís, Maranhão, Brasil. Foram coletadas amostras de fezes de 150 animais de quatro propriedades diferentes, sendo 2 com caprinos e 2 com ovinos, e aleatoriamente distribuídos de três a quatro grupos de 10 animais: Grupo I: grupo controle, sem tratamento. Grupo II: tratado com anti-helmíntico à base de ivermectina, administrado oralmente na dose de 200 mcg/kg; Grupo III: tratado com anti-helmíntico à base de levamisol, administrado oralmente na dose de 7,5mg/kg e Grupo IV: tratado com anti-helmíntico à base de albendazol administrado oralmente na dose de 3mg/kg. Amostras de fezes foram colhidas da ampola retal um dia antes do tratamento e 10 dias após o tratamento anti-helmíntico. Foram feitos exames coproparasitológicos individuais, pela técnica de McMaster modificada, no Laboratório de Sanidade Animal, do Instituto Federal do Maranhão (IFMA), Campus São Luís-Maracanã. A ivermectina, na espécie caprina, mostrou eficácia de 14,28%, enquanato para ovina, 13,6 e 52,2%. Considerando o levamisol na espécie caprina, não apresentou eficácia contra os nematoides gastrintestinais, enquanto para ovinos, apresentou eficácia de 79,4%. Já o albendazol, apresentou eficácia de 59,8% para caprinos, e 3,43% para ovinos. Os nematoides gastrintestinais demonstraram resistência múltipla (RAM), visto que a percentagem de redução da contagem de ovos foi inferior a 95% e o limite inferior do intervalo de confiança menor do que 90%.

Resumo O presente estudo avaliou os métodos de controle empregados por médicos veterinários clínicos para o controle de parasitos de cães no Brasil. Vinte e uma perguntas foram feitas a 403 veterinários de diferentes regiões do país. O uso de associações de compostos ativos em intervalos de três meses foi o mais recomendado para profilaxia de helmintos gastrointestinais, repetido após 15 dias. A profilaxia de rotina contra ectoparasitos foi recomendada por 82,4% dos veterinários, e 46,6% mudam rotineiramente o composto indicado. Tratamentos profiláticos mensais para ectoparasitos, com produtos sistêmicos, tópicos e / ou impregnados com colar, foram recomendados por 21,5% dos veterinários. Os efeitos colaterais das coleiras impregnadas com ectoparasiticidas foram relatados por 58% dos médicos veterinários. As isoxazolinas foram o grupo químico mais utilizado para tratar ectoparasitos em cães. A baixa eficácia do fipronil no controle de carrapatos foi suspeitada por 79,5% dos médicos veterinários. As isoxazolinas e a associação de compostos anti-helmínticos são os medicamentos mais comuns para prevenir ou tratar ectoparasitos e nematoides gastrointestinais, respectivamente. A suspeita da ineficácia dos antiparasitários é compartilhada entre os médicos veterinários de algumas regiões do Brasil. Orientações são necessárias, especificamente para o controle de helmintos e ectoparasitos gastrointestinais em cães no Brasil.

Abstract The present study attempted to evaluate the practical experience and methods employed by Brazilian veterinary practitioners for control of parasites. Twenty-one questions were asked of 403 veterinary practitioners based in different climatic zones with reference to parasite epidemiology from the country. Administration of a combination of drugs at three-month intervals was the most common regime recommended for prophylaxis against gastrointestinal helminths, with a single treatment repeated after 15 days. Routine prophylaxis against dog ectoparasites was recommended by 82.4% veterinary practitioners, and 46.6% changed the drug compound used. Monthly prophylaxic treatments for ectoparasites, using systemic, topical and/or collar-impregnated drugs, was recommended by 21.5% veterinary practitioners. Side-effects of ectoparasiticide-impregnated collars were suspected by 58% of the veterinary practitioners. Isoxazolines were the most frequently used chemical group to treat ectoparasites in dogs. Poor efficacy of fipronil in controlling ticks was suspected by 79.5% of the veterinary practitioners. The isoxazolines and combination of anthelmintic compounds are the most common drugs to prevent or treat ectoparasites and gastrointestinal nematodes, respectively. The suspect of the inefficacy of antiparasitic drugs is shared among the veterinary practitioners from part of Brazil. Guidelines are needed, specifically for the control of gastrointestinal helminths and ectoparasites in Brazilian dogs.

Resumo O objetivo deste trabalho foi avaliar o desenvolvimento e a sobrevivência de larvas de terceiro estágio (L3) de Haemonchus contortus em forrageiras na Amazônia Oriental. Foram utilizados quatro canteiros compostos por Megathyrsus maximus 'Massai', M. maximus 'Mombaça', Urochloa brizantha 'Marandu' e Urochloa humidicola, os quais foram divididos em 13 parcelas cada um. Fezes de ovinos com cerca de 10.000 ovos de H. contortus foram depositadas em cada parcela. Amostras de capim, fezes e solo foram coletadas no no sétimo, no décimo quinto e no trigésimo dia pós-contaminação (DPC) e, sequencialmente, a cada 30 dias até o trecentésimo trigésimo DPC. Foram determinados os seguintes dados das gramíneas: microclimáticos, como temperatura, umidade relativa do ar, umidade do solo e luminosidade do capim, e macroclimáticos, como pluviosidade, temperatura, umidade relativa do ar e radiação solar. As larvas do terceiro estágio foram recuperadas em todas as amostras de gramíneas e solo, em todas as parcelas, do sétimo ao trecentésimo trigésimo DPC. Os parâmetros microclimáticos mostram correlações entre a recuperação de L3 na grama e no solo, e os parâmetros macroclimáticos, entre a recuperação de L3 nas fezes e na grama. Urochloa humidicola e M. maximus 'Massai' favorecem o desenvolvimento e a sobrevivência de L3 de H. contortus, enquanto U. brizantha 'Marandu' e M. maximus 'Mombaça' apresentam menor biodisponibilidade dessas larvas.

Abstract The objective of this work was to evaluate the survival of third-stage larvae (L3) of Haemonchus contortus in forage species in the Eastern Amazon. Four paddocks composed of Megathyrsus maximus 'Massai', M. maximus 'Mombaça', Urochloa brizantha 'Marandu', and Urochloa humidicola were used and divided into 13 plots each. Sheep feces containing about 10,000 eggs of H. contortus were deposited in each plot. Grass, feces, and soil samples were collected on the seventh, fifteenth, and thirtieth day post-contamination (DPC), and, then, they were sequentially collected every 30 days until the three-hundredth and thirtieth DPC. The following data were determined for the grass species: microclimatic, such as temperature, relative humidity, soil moisture, and grass luminosity, as well as macroclimatic data for rainfall, temperature, relative humidity, and solar radiation. L3 were recovered on all grasses and soil samples, in all plots, from the seventh to the three-hundredth and thirtieth DPC. The microclimatic parameters show correlations between the L3 recovery on grass and in the soil, and the macroclimatic parameters, between the L3 recovery in feces and on grass. Urochloa humidicola and M. maximus 'Massai' favor the development and survival of L3 of H. contortus, while U. brizantha 'Marandu' and M. maximus 'Mombaça' show a lower bioavailability of these larvae.

Resumo Este estudo relata a ação de óleos essenciais de cinco plantas na atividade de acetilcolinesterases (AChE) nativas e recombinantes de Rhipicephalus microplus. A atividade enzimática do extrato de acetilcolinesterase nativa suscetível (S.AChE) e resistente (R.AChE) e da enzima recombinante (rBmAChE1) foi determinada. Um teste de inibição da AChE foi utilizado, para verificar o efeito dos óleos essenciais sobre a atividade enzimática. Óleos essenciais de Eucalyptus globulus, Citrus aurantifolia, Citrus aurantium var. dulcis inibiram a atividade de S.AChE e R.AChE. Os óleos das duas espécies de Citrus inibiram S.AChE e R.AChE de maneira semelhante, mas mostraram maior inibição sobre R.AChE. O óleo de E. globulus inibiu a AChE nativa, mas sem diferença entre a S.AChE e a R.AChE; no entanto, 71% de inibição para rBmAChE1 foi observada. O óleo de Mentha piperita também inibiu S.AChE e R.AChE, mas houve inibição significativa apenas nas concentrações mais altas testadas. O óleo de Cymbopogon winterianus não inibiu a AChE. Estudos adicionais são necessários com os óleos das duas espécies de Citrus que inibiram a R.AchE, devido ao problema de R. microplus resistente aos organofosforados ter como alvo AChE. O óleo de C. winterianus pode ser usado contra populações de R. microplus, que são resistentes a organofosforados, porque suas propriedades acaricidas agem por mecanismos diferentes.

Abstract This study reports the action of essential oils (EO) from five plants on the activity of native and recombinant acetylcholinesterases (AChE) from Rhipicephalus microplus. Enzyme activity of native susceptible AChE extract (S.AChE), native resistant AChE extract (R.AChE), and recombinant enzyme (rBmAChE1) was determined. An acetylcholinesterase inhibition test was used to verify the effect of the EO on enzyme activity. EO from Eucalyptus globulus, Citrus aurantifolia, Citrus aurantium var.dulcis inhibited the activity of S.AChE and R.AChE. Oils from the two Citrus species inhibited S.AChE and R.AChE in a similar way while showing greater inhibition on R.AChE. The oil from E. globulus inhibited native AChE, but no difference was observed between the S.AChE and R.AChE; however, 71% inhibition for the rBmAChE1 was recorded. Mentha piperita oil also inhibited S.AChE and R.AChE, but there was significant inhibition at the highest concentration tested. Cymbopogon winterianus oil did not inhibit AChE. Further studies are warranted with the oils from the two Citrus species that inhibited R.AChE because of the problem with R. microplus resistant to organophosphates, which target AChE. C. winterianus oil can be used against R. microplus populations that are resistant to organophosphates because its acaricidal properties act by mechanism(s) other than AChE inhibition.

Resumo Cochliomyia hominivorax (Coquerel, 1858), a mosca-da-bicheira, causa miíase primária em animais silvestres e domésticos em regiões tropicais e subtropicais do Brasil. Embora esta espécie seja considerada de ocorrência em todo o país, informações organizadas sobre sua distribuição com base em registros não estavam disponíveis até o momento. Este artigo teve como objetivo fornecer uma revisão abrangente dos dados históricos e atuais publicados sobre o registro de estágios imaturos (miíase) e adultos de C. hominivorax, no Brasil. De 174 artigos encontrados, 141 relataram casos de miíase em bovinos (146 relatos), humanos (68 relatos) e outros mamíferos hospedeiros (40 relatos) e capturas de adultos de C. hominivorax foram registradas em 33 artigos. C. hominivorax encontra-se amplamente distribuída no Brasil, tendo sido registrada em 208 municípios brasileiros em todos os principais biomas do país.

Abstract Cochliomyia hominivorax (Coquerel, 1858), the New World screwworm, causes primary myiasis in wild and domestic animals in tropical and subtropical regions of Brazil. Although this species is considered to occur throughout the country, organized information about its recorded distribution has not been available until now. This article aimed to provide a comprehensive review of the historical and current data published on both immature (myiasis) and adult stages of C. hominivorax in Brazil. A total of 174 articles were found; of these, 141 articles reported myiasis cases in cattle (146 records), humans (68 records), and other mammalian hosts (40 records), and captures of adult flies were reported in 33 articles. C. hominivorax is widespread in Brazil, having been recorded in 208 municipalities in all major biomes of the country.

ABSTRACT The objective of this study was to evaluate the acaricidal activity of the volatile oils of three species of Croton, Euphorbiaceae, against the cattle tick Rhipicephalus microplus. The volatile oils were obtained by hydrodistillation, analyzed by GC-MS and GC-FID and their acaricidal activity was evaluated by the larval packet test and adult immersion test. The volatile oils from Croton conduplicatus Kunth, Croton pulegiodorus Baill., and two different collections of Croton grewioides Baill. (CG1 and CG2) showed eucalyptol (24.09%), p-cymene (23.13%) and methyl chavicol (83.59% and 95.38%) as the major compounds, respectively. All the volatile oils tested in this study showed efficacy against larvae and engorged females of Rhipicephalus microplus. Therefore, Croton pulegiodorus volatile oil is promising for a potential acaricidal formulation because of the best activity against both stages of the cattle tick.

Abstract BRS FC104 is a carioca seeded common bean cultivar with super-early maturity (mean of 65 days; emergence to physiological maturity) and disease resistance superior to other early maturity carioca seeded cultivars. Is is recommended for growing in 21 Brazilian states and has high yield potential (3792 kg ha-1), a mean yield of 2186 kg ha-1, and 6.9% higher mean of the relative yield compared to the control cultivars.

Resumo A infecção por nematoides gastrintestinais é uma importante causa de grandes perdas econômicas na pecuária. O controle de nematoides com compostos químicos sintéticos é considerado insustentável devido ao aumento da resistência anti-helmíntica. Alternativas de controle, como o uso de produtos naturais, estão se tornando relevantes do ponto de vista ambiental e econômico. As proteínas são macromoléculas com várias propriedades que podem ser obtidas de uma ampla gama de organismos, incluindo plantas e fungos. Proteínas pertencentes a diferentes classes têm mostrado grande potencial para o controle de nematoides. A ação das proteínas pode ocorrer em estágios específicos do ciclo de vida do nematoide, dependendo da composição das camadas externas do parasito e do sítio ativo da proteína. Avanços na biotecnologia resultaram no surgimento de numerosas terapias de proteínas e peptídeos; no entanto, pouco foi discutido com foco no controle de nematoides parasitos de animais. Na presente revisão foi discutido o uso de proteínas exógenas e peptídeos no controle de nematoides gastrintestinais, os mecanismos sugeridos de ação, e os desafios e perspectivas para o uso dessas biomoléculas como uma classe de anti-helmínticos.

Abstract Gastrointestinal nematode infection is an important cause of high economic losses in livestock production. Nematode control based on a synthetic chemical approach is considered unsustainable due to the increasing incidence of anthelmintic resistance. Control alternatives such as the use of natural products are therefore becoming relevant from an environmental and economic point of view. Proteins are macromolecules with various properties that can be obtained from a wide range of organisms, including plants and fungi. Proteins belonging to different classes have shown great potential for the control of nematodes. The action of proteins can occur at specific stages of the nematode life cycle, depending on the composition of the external layers of the nematode body and the active site of the protein. Advances in biotechnology have resulted in the emergence of numerous protein and peptide therapeutics; however, few have been discussed with a focus on the control of animal nematodes. Here, we discuss the use of exogenous proteins and peptides in the control of gastrointestinal.

Abstract BRS Sublime is a common bean cultivar with carioca grains, high nutritional grain quality, and high yield potential (4667 kg ha-1). It has a normal cycle, upright growth, low loss rates in the mechanical harvest, and is recommended for planting in 19 Brazilian states, in the three main common bean cultivation regions. It is the first cultivar developed in the world with angular leaf spot resistance.

Resumo Haemonchus contortus é um nematoide gastrintestinal, responsável por altas taxas de mortalidade em rebanhos de pequenos ruminantes. A resistência dos nematoides aos anti-helmínticos sintéticos está generalizada e requer uma busca contínua por novos compostos bioativos, como as proteínas. O objetivo deste trabalho foi avaliar o potencial anti-helmíntico da protease purificada do látex de Ficus benjamina contra H. contortus . O látex fresco foi coletado das plantas por pequenas incisões nas hastes verdes e o extrato proteico de látex (EPL) foi obtido. Após o fracionamento do EPL com sulfato de amônio e cromatografia da fração contendo a maior atividade proteolítica da CM-Celulose, uma protease cisteínica (FbP) foi purificada. A FbP tem massa molecular de cerca de 23,97 kDa, a atividade proteolítica foi estável entre pH 6,0 e pH 10 e ao longo de uma ampla faixa de temperatura, com atividade ótima a 60 °C. A FbP inibiu tanto o desenvolvimento quanto o desembainhamento das larvas de H. contortus, com 50% de inibição nas concentrações de 0,26 e 0,79 mg/mL, respectivamente. Concluímos que esta protease cisteínica do látex de F. benjamina, com ação anti-helmíntica contra H. contortus, pode ser uma alternativa promissora para o desenvolvimento de produtos a serem utilizados em programas de controle de parasitos.

Abstract Haemonchus contortus is a gastrointestinal nematode that is responsible for high mortality rates in ruminant herds. The resistance of nematodes to synthetic anthelmintics is widespread and requires a continuous search for new bioactive molecules, such as proteins. The objective of this study was to evaluate the anthelmintic potential of a protease purified from the latex of Ficus benjamina against H. contortus . Fresh latex was collected from plants via small incisions in the green stems, the rubber was removed by centrifugation, and the latex protein extract (LPE) was obtained. After LPE fractionation with ammonium sulfate and chromatography of the fraction containing the highest proteolytic activity on CM-cellulose, a cysteine protease (FbP) was purified. FbP has a molecular mass of approximately 23.97 kDa, and its proteolytic activity was stable between pH 6.0 and pH 10 and over a broad temperature range, with optimum activity at 60 °C. FbP inhibited both the development and exsheathment of H. contortus larvae, with 50% effective concentrations of 0.26 and 0.79 mg/mL, respectively. We conclude that this cysteine protease from F. benjamina latex with anthelmintic activity against H. contortus could be a promising alternative for the development of products for use in parasite control programmes.

Resumo O objetivo deste estudo foi avaliar a atividade acaricida de óleos essenciais de três espécies de plantas com concentrações intermediárias de 1,8-cineol contra o carrapato Rhipicephalus (Boophilus) microplus . Dessa forma, cinco concentrações diferentes (100,0; 50,0; 25,0; 12,5; 6,2 mg/mL) de óleos essenciais de Mesosphaerum suaveolens (L.) Kuntze , Ocimum gratissimum L. e Alpinia zerumbet (Pers.) B. L. Burtt & R. M. Sm. foram avaliadas pelos testes de pacote de larvas e de imersão de adultos. Os óleos essenciais foram analisados pela cromatografia gasosa acoplada a espectometria de massa (GC/MS) e cromatografia gasosa acoplada a detector de ionização de chama (GC-FID), sendo detectados 35,8, 24,7 e 24,0% de 1,8-cineol nos óleos de M. suaveolens, O. gratissimum e A. zerumbet , respectivamente. A concentração letal (CL50) de cada óleo essencial para larvas e fêmeas ingurgitadas foi calculada por meio da análise de Probit. Todos os óleos essenciais na concentração de 100,0 mg/mL apresentaram elevada eficácia (≥ 95,0%) sobre fêmeas ingurgitadas. Com relação as larvas, o óleo essencial de O. gratissimum (CL50 = 11,9 mg/mL) foi o mais potente, seguido pelos óleos de A. zerumbet (LC50 = 19,7 mg/mL) e M. suaveolens (LC50 = 51,6 mg/mL). Estes resultados demonstram que outros compostos interferem na eficácia de 1,8-cineol.

Abstract The aim of this study was to evaluate the acaricidal activity of essential oils from three species of plants with intermediary concentrations of 1,8-cineole against the tick species Rhipicephalus (Boophilus) microplus. For this purpose, five serial concentrations (100.0, 50.0, 25.0, 12.5, 6.2 mg/mL) of essential oils from Mesosphaerum suaveolens (L.) Kuntze, Ocimum gratissimum L. and Alpinia zerumbet (Pers.) B. L. Burtt & R. M. Sm. were used on larval packet and adult immersion tests. The essential oils were analysed by gas chromatography-mass spectrometry (GC/MS) and gas chromatography-flame ionization detection (GC-FID), being detected 35.8, 24.7 and 24.0% of 1.8-cineol in the oils of M. suaveolens, O. gratissimum and A. zerumbet, respectively. The lethal concentration (LC 50) of each oil for larvae and engorged females was calculated through Probit analysis. All essential oils showed high efficacy (≥ 95.0%) on engorged females at the 100.0 mg/mL concentration. In regards to larvae, O. gratissimum (LC 50 = 11.9 mg/mL) was the most potent, followed by the A. zerumbet (LC50 = 19.7 mg/mL) and the M. suaveolens (LC50 = 51.6 mg/mL) essential oils. These results show that other compounds interfere with 1,8-cineole action.

Resumo Este estudo teve como objetivo determinar a dinâmica da infecção natural na transmissão de Babesia spp. em bovinos de uma área de instabilidade enzoótica no Nordeste do Brasil. Foram coletadas amostras de sangue de 30 bezerras, proveniente de duas propriedades leiteiras para determinação do volume globular e da primo-infecção por meio da reação em cadeia da polimerase associando aos fatores climáticos e medidas de manejo. Na fazenda A, o período médio da primo-infecção para B. bigemina, determinado por meio da PCR, foi de 249,4 (±24,42) dias de idade, enquanto que para B. bovis foi aos 252,6 (±17,07) dias de idade, não existindo diferença estatística. A infecção coincidiu com o período de alta precipitação pluviométrica na região. Não houve infecção por Babesia spp. na fazenda B, na qual o uso intensivo de acaricidas determinou ausência de carrapatos. Não houve diferença significativa entre médias de VG dos animais das fazendas A e B. O manejo adotado nas fazendas estudadas, associado às condições climáticas, interferem na exposição dos animais aos vetores, podendo favorecer a manutenção de uma área de instabilidade enzoótica no Nordeste do Brasil.

Abstract This study aimed to determine the dynamics of natural infection in the transmission of Babesia spp. to cattle in an enzootic instability area in Northeastern Brazil. Blood samples were collected from 30 calves located on two dairy farms to determine the packed cell volume (PCV) and the timing of the primo-infection using polymerase chain reaction (PCR) and their association with climatic factors and management practices. On Farm A, the determination of primo-infection was observed on average at 249.4 (±24.42) days of age for B. bigemina and at 252.6 (±17.07) days of age for B. bovis; there was no significant difference between the times of infection (P> 0.05). The infection coincided with a period of high rainfall in the region. On Farm B, primo-infection infection was not observed. There was no infection by Babesia spp. on Farm B due to the intensive use of acaricides that led to an absence of ticks. There was no significant difference between the average PCV of animals from Farms A and B (P> 0.05). The management practices on the properties, in addition to the weather conditions influenced the exposure of the animals to disease vectors and may have contributed to the maintenance of this enzootic area in Northeastern Brazil.
