Abstract We developed poly-ε-caprolactone (PCL)-based nanoparticles containing D-α-tocopherol polyethylene glycol-1000 succinate (TPGS) or Poloxamer 407 as stabilizers to efficiently encapsulate genistein (GN). Two formulations, referred to as PNTPGS and PNPol, were prepared using nanoprecipitation. They were characterized by size and PDI distribution, zeta potential, nanoparticle tracking analysis (NTA), GN association (AE%), infrared spectroscopy (FT-IR), and differential scanning calorimetry (DSC). PNTPGS-GN exhibited a particle size of 141.2 nm, a PDI of 0.189, a zeta potential of -32.9 mV, and an AE% of 77.95%. PNPol-GN had a size of 146.3 nm, a better PDI than PNTPGS-GN (0.150), a less negative zeta potential (-21.0 mV), and an AE% of 68.73%. Thermal and spectrometric analyses indicated that no new compounds were formed, and there was no incompatibility detected in the formulations. Cellular studies revealed that Poloxamer 407 conferred less toxicity to PCL nanoparticles. However, the percentage of uptake decreased compared to the use of TPGS, which exhibited almost 80% cellular uptake. This study contributes to the investigation of stabilizers capable of conferring stability to PCL nanoparticles efficiently encapsulating GN. ABSTRACT In Brazil, Dengue, Zika and Chikungunya viruses constitute a major threat to the public health system. Simultaneous circulation of these arboviruses occurs in many regions of the world due to the expansion of transmission vectors. The infection by these arboviruses triggers similar symptoms during their acute phase. However, in some cases, severe symptoms may occur, leading to different types of disabilities and even death. In this context, considering the similarity of the symptoms, the problems caused by the infection of these arboviruses, and the increasing risk of coinfection in humans, the differential diagnosis of these infections is essential for clinical management and epidemiological investigation. Thus, this study aimed to identify, through diagnosis via Quantitative Polymerase Chain Reaction with Reverse Transcription, arbovirus coinfection in patients from the Tocantins state (Northern Brazil). A total of 495 samples were analyzed, three from which were determined to be a coinfection of Dengue and Chikungunya viruses. The data obtained here indicate the co-circulation and coinfection by Dengue and Chikungunya viruses in the Tocantins state. These results highlight the importance of monitoring the circulation of these arboviruses for the development of health actions that aim their prevention and combat, as well as their clinical and therapeutic management. ABSTRACT The limited temporal completeness and taxonomic accuracy of species lists, made available in a traditional manner in scientific publications, has always represented a problem. These lists are invariably limited to a few taxonomic groups and do not represent up-to-date knowledge of all species and classifications. In this context, the Brazilian megadiverse fauna is no exception, and the Catálogo Taxonômico da Fauna do Brasil (CTFB) (http://fauna.jbrj.gov.br/), made public in 2015, represents a database on biodiversity anchored on a list of valid and expertly recognized scientific names of animals in Brazil. The CTFB is updated in near real time by a team of more than 800 specialists. By January 1, 2024, the CTFB compiled 133,691 nominal species, with 125,138 that were considered valid. Most of the valid species were arthropods (82.3%, with more than 102,000 species) and chordates (7.69%, with over 11,000 species). These taxa were followed by a cluster composed of Mollusca (3,567 species), Platyhelminthes (2,292 species), Annelida (1,833 species), and Nematoda (1,447 species). All remaining groups had less than 1,000 species reported in Brazil, with Cnidaria (831 species), Porifera (628 species), Rotifera (606 species), and Bryozoa (520 species) representing those with more than 500 species. Analysis of the CTFB database can facilitate and direct efforts towards the discovery of new species in Brazil, but it is also fundamental in providing the best available list of valid nominal species to users, including those in science, health, conservation efforts, and any initiative involving animals. ABSTRACT Objective: Previous studies indicated that face masks reduce the probability of infection by SARS-CoV-2 but did not examine the relationship between SARS-CoV-2 viral load and mask usage. This study analyzed this relationship. Methods: This cross-sectional study evaluated patients admitted to a public Emergency Care Unit in Belo Horizonte, MG, Brazil, between October 2020 and March 2021. Convenience samples were recruited during the study period. Adult patients with COVID-19 symptoms were invited to participate after being examined by a physician according to the Emergency Care Unit routine. This study included 441 adults with mild symptoms diagnosed with COVID-19 via RT-PCR, who were tested until the 14th day after symptom onset. Participants were interviewed about mask usage, oropharyngeal swabs were collected, and the SARS-CoV-2 viral load was measured using RT-PCR. The difference between the SARS-CoV-2 viral load in the groups of individuals who regularly used and did not use masks was subjected to a hypothesis test. Results: Of the patients who had swabs collected 1-5 days after symptom onset, 64.9% of those not using masks regularly and 45.5% of those using masks regularly presented a high SARS-CoV-2 viral load. This difference was statistically significant (p=0.0073). Considering only the patients who had swabs collected 4-8 days after symptom onset, 44.1% of those not using masks regularly and 30.8% of those using masks regularly presented a high SARS-CoV-2 viral load; this difference was statistically significant (p=0.0364). Conclusion: When people who regularly wear a face mask contract COVID-19, they have a significantly lower probability of having a high viral load of SARS-CoV-2 compared to those who do not regularly wear masks. Abstract Although the terms "rare diseases" (RD) and "orphan diseases" (OD) are often used interchangeably, specific nuances in definitions should be noted to avoid misconception. RD are characterized by a low prevalence within the population, whereas OD are those inadequately recognized or even neglected by the medical community and drug companies. Despite their rarity, as our ability on discovering novel clinical phenotypes and improving diagnostic tools expand, RD will continue posing a real challenge for rheumatologists. Over the last decade, there has been a growing interest on elucidating mechanisms of rare autoimmune and autoinflammatory rheumatic diseases, allowing a better understanding of the role played by immune dysregulation on granulomatous, histiocytic, and hypereosinophilic disorders, just to name a few. This initiative enabled the rise of innovative targeted therapies for rheumatic RD. In this review, we explore the state-of-the art of rare RD and the critical role played by rheumatologists in healthcare. ABSTRACT The objective of this study was to evaluate, through principal component analysis, the effect of different feedlot structures on climatic conditions, nutrient intake, and performance of Nellore bulls during the feeding period in the semi-arid region. Data from 225 non-castrated Nellore bulls with initial mean body weight (BW) of 294 ± 19 kg and age of 24 months were used. This was done in a completely randomized design with three feedlot structures (no shading, natural shade, and artificial shade with 50% light interception), resulting in 75 repetitions (each animal serving as an experimental unit). The feeding period for the animals in the feedlot was 107 days. The first two components explained 99.65 (no shade), 97.32 (artificial shade), and 99.89 (natural shade) of total data variation. The variables, namely the black globe temperature index (0.88), radiant heat load (0.96, 0.99, 0.76), black globe temperature (0.96), and hot carcass weight (0.64) exerted the greatest influence on the feedlot structures. Climatic and intake variables were identified as having the most significant impact on feedlot structures. Abstract In the present work, the electrical properties of BaFe12O19 (BaM) M-type hexaferrite were investigated in the low-temperature regime, something new in the literature on this material. The technique used was impedance spectroscopy, with measurements performed in the temperature range of 300 to 50 K, with the frequency of the oscillating external electric field varying from 1 Hz to 1 MHz. Resumo No presente trabalho foram investigadas as propriedades elétricas da hexaferrita do tipo M BaFe12O19 (BaM) no regime de baixas temperaturas, algo novo na literatura deste material. A técnica utilizada foi a espectroscopia de impedância, sendo as medidas realizadas no intervalo de temperatura de 300 a 50 K, com a frequência do campo elétrico externo oscilante variando de 1 Hz a 1 MHz. Abstract This study aimed to assess the influence of streptozotocin (STZ)-induced diabetes on the nociceptive behavior evoked by the injection of hypertonic saline (HS) into the masseter muscle of rats. Forty male rats were equally divided into four groups: a) isotonic saline control, which received 0.9% isotonic saline (IS), (Ctrl-IS); b) hypertonic saline control, which received 5% HS (Ctrl-HS); c) STZ-induced diabetic, which received IS, (STZ-IS); d) STZ-induced diabetic, which received HS (STZ-HS). Experimental diabetes was induced by a single intraperitoneal injection of STZ at dose of 60 mg/kg dissolved in 0.1 M citrate buffer, and 100 μL of HS or IS were injected into the left masseter to measure the nociceptive behavior. Later on, muscle RNA was extracted to measure the relative expression of the following cytokines: cyclooxygenase-2 (COX-2), tumor necrosis factor (TNF-α), and interleukins (IL)-1β, -2, -6, and -10. One-way analysis of variance (ANOVA) was applied to the data (p < 0.050). We observed a main effect of group on the nociceptive response (ANOVA: F = 11.60, p < 0.001), where the Ctrl-HS group presented the highest response (p < 0.001). However, nociceptive response was similar among the Ctrl-IS, STZ-IS, and STZ-HS group (p > 0.050). In addition, the highest relative gene expression of TNF-α and IL-6 was found in the masseter of control rats following experimental muscle pain (p < 0.050). In conclusion, the loss of somatosensory function can be observed in deep orofacial tissues of STZ-induced diabetic rats. Abstract Hypostomus is distributed by Central and South America basins, with diverse species with taxonomic conflicts. This way, the integration of auxiliary techniques contributes to understanding the systematics and phylogeny of the group. Thus, this study aimed to investigate the Hypostomus cochliodon and H. boulengeri from the Onça stream (Paraguai River basin) by allozyme and cytogenetic techniques. Hypostomus boulengeri showed a diploid number of 68 chromosomes (14m+14sm+18st+22a), multiple NOR revealed by Ag-NOR and 18S rDNA FISH, a polymorphism of heterochromatin in acrocentrics and the presence of B microchromosome. Hypostomus cochliodon showed a diploid number of 64 chromosomes (16m+26sm+14st+8a); despite the single NOR, some individuals showed NOR in both telomeres detected by Ag-NOR and 18S rDNA FISH. Isozyme identified two diagnostic loci (Idh-A and Gdh-A) between the two species and multiple loci with unique alleles in H. boulengeri. The genetic variability indicated by the mean heterozygosity (He) was 0.2461 and 0.0309 in H. boulengeri and H. cochliodon,respectively.Thus, this study reports the first cytogenetic data for H. boulengeri and the first isozymatic data for H. boulengeri and H. cochliodon. Desta forma, a integração de técnicas auxiliares contribui para a compreensão da sistemática e filogenia do grupo. Assim, este estudo teve como objetivo investigar Hypostomus cochliodon e H. boulengeri do riacho Onça (bacia do rio Paraguai) por meio de técnicas aloenzimáticas e citogenéticas. Abstract: The influenza-like illness (ILI) sentinel surveillance operates in Brazil to identify respiratory viruses of public health relevance circulating in the country and was first implemented in 2000. Recently, the COVID-19 pandemic reinforced the importance of early detection of the circulation of new viruses in Brazil. Therefore, an analysis of the design of the ILI sentinel surveillance is timely. To this end, we simulated a sentinel surveillance network, identifying the municipalities that would be part of the network according to the criteria defined in the design of the ILI sentinel surveillance and, based on data from tested cases of severe acute respiratory illness (SARI) from 2014 to 2019, we drew samples for each sentinel municipality per epidemiological week. The draw was performed 1,000 times, obtaining the median and 95% quantile interval (95%QI) of virus positivity by Federative Unit and epidemiological week. According to the ILI sentinel surveillance design criteria, sentinel units would be in 64 municipalities, distributed mainly in capitals and their metropolitan areas, recommending 690 weekly samples. The design showed good sensitivity (91.65% considering the 95%QI) for qualitatively detecting respiratory viruses, even those with low circulation. However, there was important uncertainty in the quantitative estimate of positivity, reaching at least 20% in 11.34% of estimates. The results presented here aim to assist in evaluating and updating the ILI sentinel surveillance design. Strategies to reduce uncertainty in positivity estimates need to be evaluated, as does the need for greater spatial coverage. Resumo: A vigilância sentinela de síndrome gripal atua no Brasil identificando os vírus respiratórios de importância para a saúde pública circulantes no país, e começou a ser implementada em 2000. Recentemente, a pandemia de COVID-19 reforçou a importância da detecção precoce de novos vírus em circulação no território brasileiro. Assim, se faz oportuna uma análise do desenho da vigilância sentinela de síndrome gripal. Para tal, simulamos uma rede sentinela, identificando os municípios que fariam parte da rede segundo os critérios definidos no desenho da vigilância sentinela de síndrome gripal, e, a partir dos dados de casos testados de síndrome respiratória aguda grave (SRAG) de 2014 a 2019, sorteamos amostras para cada município sentinela por semana epidemiológica. Resumen: La vigilancia centinela de la enfermedad tipo infuenza (ETI) funciona en Brasil para identificar los virus respiratorios de importancia para la salud pública que circulan en el país y comenzó a ser implementada en 2000. Recientemente, la pandemia de COVID-19 ha reforzado la importancia de la detección temprana de la circulación de nuevos virus en el territorio brasileño. Así, se hace oportuno un análisis del diseño de la vigilancia centinela de la ETI. Para ello, simulamos una red centinela identificando los municipios que formarían parte de la red según los criterios definidos en el diseño de la vigilancia centinela de la ETI y, a partir de los datos de casos testados de infección respiratoria aguda grave (IRAG) de 2014 a 2019, se extrajeron muestras para cada municipio centinela por semana epidemiológica. El sorteo se repitió 1.000 veces y se obtuvo la mediana y el intervalo cuantílico del 95% (IC95%) de la positividad por virus, por Unidad Federativa y semana epidemiológica. Abstract Several studies have shown COVID-19 has affected the mental health and quality of life of students. This study aimed to assess the quality of life of students in two Brazilian public universities in 2020, using a cross-sectional study. The profile of the participants was delineated from a sociodemographic questionnaire, and the quality of life was assessed by the WHOQOL-bref. A total of 144 students were assessed, in which most of them were under 23 (57%), female (78%), self-declared brown (46%) and without a partner (87%). Most of them declared to sleep less than 8 hours a day, considered their quality of life to be good, and were satisfied with their health. The social relationships domain had the highest average, followed by the environment, physical and psychological domains. The perception of the student's quality of life was considered good, and the factors associated were sex, family income, and number of sleeping hours. Resumo Diversos estudos têm demonstrado que a pandemia do COVID-19 tem prejudicado a saúde mental e a qualidade de vida dos estudantes. Assim, o objetivo deste estudo foi avaliar a qualidade de vida dos estudantes em duas universidades públicas brasileiras em 2020, utilizando um estudo transversal. O perfil dos participantes foi delineado a partir de um questionário sociodemográfico e a qualidade de vida foi avaliada pelo WHOQOL-bref. Um total de 144 estudantes participaram, sendo a maioria com idade inferior a 23 anos (57%), do sexo feminino (78%), autodeclarada pardos (46%) e sem companheiro(a) (87%). A maioria declarou dormir menos de 8 horas/dia, considerou sua qualidade de vida como sendo boa e estavam satisfeitos com a sua saúde. O domínio relações sociais teve maior média, seguido pelos domínios meio ambiente, físico e psicológico. A percepção sobre a qualidade de vida dos estudantes foi considerada boa e os principais fatores associados foram sexo, renda familiar e número de horas dormidas. Resumen La salud mental y la calidad de vida de los estudiantes se vieron perjudicadas por los efectos de la pandemia de COVID-19. El objetivo de este estudio es evaluar la calidad de vida de los estudiantes universitarios durante la pandemia. Se trata de un estudio transversal realizado en das universidades públicas brasilehas en 2020. La información se recolectó, mediante la utilización de dos cuestionarios: cuestionario sociodemográfico y WHOQOL-bref. Participaron un total de 144 estudiantes, la mayoría menores de 23 ahos (57%), mujeres (78%), autodeclaradas morenas (46%) y sin pareja (87%). La mayoría declaró dormir menos de 8 horas diarias, consideró buena su calidad de vida y se mostró satisfecha con su salud. El dominio de las relaciones sociales tuvo el mayor promedio, seguido por los dominios medio ambiente, físico y psicológico. Drug repurposing and personalized treatment strategies for bipolar disorder using transcriptomics: an exploratory study transcriptomics

Objective: The present study combined transcriptomic data and computational techniques based on gene expression signatures to identify new bioactive compounds or Food and Drug Administration-approved drugs for the treatment of bipolar disorder (BD). Methods: Five transcriptomic datasets containing 165 blood samples from individuals with BD were selected from the Gene Expression Omnibus (GEO). The number of participants varied from six to 60, with a mean age between 35 and 48 years and a gender difference between them. Most of these patients were receiving pharmacological treatment. Master regulator analysis (MRA) and gene set enrichment analysis (GSEA) were performed to identify genes that were significantly different between patients with BD and healthy controls and their associations with mood states in patients with BD. In addition, molecules that could reverse the transcriptomic profiles of BD-altered regulons were identified from the Library of Network-Based Cellular Signatures Consortium (LINCS) and the Broad Institute Connectivity Map Drug Repurposing Database (cMap) databases. Results: MRA identified 59 candidate master regulators (MRs) that modulate regulatory units enriched with BD-altered genes. In contrast, GSEA identified 134 enriched genes and 982 regulons whose activation state was determined. Both analyses revealed genes exclusively associated with mania, depression, or euthymia, and some genes were shared among these three mood states. We identified bioactive compounds and licensed drug candidates, including antihypertensives and antineoplastic agents, as promising candidates for the treatment of BD. However, experimental validation is essential to confirm these findings in further studies. Conclusion: Although our data are still preliminary, they provide some insights into the biological patterns of different mood states in patients with BD and their potential therapeutic targets. The strategy of transcriptomics plus bioinformatics offers a way to advance drug discovery and personalized medicine by using gene expression information. ABSTRACT BACKGROUND AND OBJECTIVES: Klippel-Trenaunay syndrome (KTS) is a rare congenital disease that manifests at birth or in childhood and has an unknown etiology, with no ethnic or gender predilection. Pain management, common in many cases, is challenging due to the complexity of the factors involved. The aim of this report is to demonstrate the effective use of a multimodal approach, including medicinal cannabis, in the treatment of pain in a child with KTS. CASE REPORT: this is the case of a child suffering from CTS who had severe chronic pain, significantly compromising his quality of life. The pain was managed with pharmacological therapies, including the use of medicinal cannabis as a therapeutic alternative when conventional treatments proved insufficient. The multimodal approach, which included the use of tetrahydrocannabinol, led to a reduction in pain and an improvement in the patient’s quality of life. Medical cannabis therapy proved to be an effective option, especially in cases of pain refractory to other interventions. CONCLUSION: The treatment of pain in patients with KTS should be multimodal, with an emphasis on non-invasive approaches. The use of medicinal cannabis is a viable and safe alternative, particularly in situations where conventional treatments do not provide adequate relief. RESUMO JUSTIFICATIVA E OBJETIVOS: A síndrome de Klippel-Trenaunay (KTS) é uma doença congênita rara que se manifesta no nascimento ou na infância e apresenta etiologia desconhecida, sem predileção étnica ou de gênero. O manejo da dor, comum em muitos casos, é desafiador devido à complexidade dos fatores envolvidos. O objetivo deste relato foi demonstrar o uso eficaz de uma abordagem multimodal, incluindo cannabis medicinal, no tratamento da dor em uma criança com KTS. RELATO DO CASO: Trata-se do caso de uma criança portadora de KTS que apresentava dor crônica intensa, comprometendo significativamente sua qualidade de vida. A dor foi manejada com terapias farmacológicas, incluindo uso de cannabis medicinal como alternativa terapêutica, quando os tratamentos convencionais se mostraram insuficientes. A abordagem multimodal, que incluiu o uso de tetraidrocanabinol, levou à redução da dor e à melhora na qualidade de vida do paciente. A terapia com cannabis medicinal demonstrou ser uma opção eficaz, especialmente em casos de dor refratária a outras intervenções. CONCLUSÃO: O tratamento da dor em pacientes com KTS deve ser multimodal, com ênfase em abordagens não invasivas. O uso de cannabis medicinal é uma alternativa viável e segura, particularmente em situações em que tratamentos convencionais não proporcionam alívio adequado. ABSTRACT Objective: to report the strategic planning of a stricto sensu postgraduate program in nursing. Method: an experience report on implementing the strategic planning of the Professional Master’s Degree in Nursing Care at the Fluminense Federal University, from 2019 to 2023. It was developed from face-to-face workshops with professors and students of the program. The workshops included: awareness raising, definition of the mission, vision and values, self-assessment, action plan and presentation of results. Results: based on the workshops, the mission, vision and values of the program were updated and an institutional self-assessment was conducted, which enabled listing the strengths, weaknesses, threats and opportunities. A total of 12 actions were listed in the program axis, 11 actions in the training axis and 11 actions in the impact axis. All actions were implemented and the goals were achieved throughout the planning. The program achieved a score of 5 in the CAPES evaluation; an international cooperation agreement was established with the University of Florida, in the United States. The program won funding notices and off-site classes in the states of Acre (funding notice CAPES/Federal Nursing Council) and Minas Gerais (partnership between the Regional Council of Minas Gerais and the Fluminense Federal University). The program is currently graduating its second class in both states. Conclusion: all actions that constituted the strategic planning were implemented in the proposed period with the active participation of the actors involved and culminated in the achievement of a score of 5 and approval of the Professional Doctorate by the Coordination for the Improvement of Higher Education Personnel, demonstrating success in the planned actions and cohesion of the team. RESUMO Objetivo: relatar o planejamento estratégico de um programa de pós-graduação stricto sensu em enfermagem. Método: relato de experiência sobre a realização do planejamento estratégico do Mestrado Profissional em Enfermagem Assistencial da Universidade Federal Fluminense, no período de 2019 a 2023. Desenvolveu-se a partir de oficinas presenciais com docentes e discentes do programa. As oficinas compreenderam: sensibilização, definição de missão, visão e valores, autoavaliação, plano de ação e apresentação dos resultados. Resultados: a partir das oficinas foi atualizada a missão, visão e valores do programa e realizada a autoavaliação institucional que possibilitou elencar as forças, fraquezas, ameaças e oportunidades. Foram elencadas 12 ações no eixo programa, 11 ações no eixo formação e 11 ações no eixo impacto. Ao longo do planejamento todas as ações foram realizadas e metas alcançadas. RESUMEN Objetivo: informar la planificación estratégica de un programa de posgrado stricto sensu en enfermería. Método: relato de experiencia en la realización de la planificación estratégica de la Maestría Profesional en Atención de Enfermería de la Universidad Federal Fluminense, de 2019 a 2023. Se desarrolló a partir de talleres presenciales con profesores y estudiantes del programa. Los talleres incluyeron: sensibilización, definición de misión, visión y valores, autoevaluación, plan de acción y presentación de resultados. Resultados: a partir de los talleres se actualizó la misión, visión y valores del programa y se realizó una autoevaluación institucional que permitió enumerar las fortalezas, debilidades, amenazas y oportunidades. Se enumeraron 12 acciones en el eje programa, 11 acciones en el eje formación y 11 acciones en el eje impacto. A lo largo de la planificación, se llevaron a cabo todas las acciones y se alcanzaron los objetivos.
