Abstract The study was carried out on an agroecological property located in Cologne São Manoel, 8th district of Pelotas, RS, Brazil. Anochetus neglectus Emery, 1894 was collected in an area of peach orchard that has been under an agroecological system for 18 years. This record expands the area of occurrence and distribution of this rare species in the Neotropical Region.

Abstract The spatial variability of physical properties, such as bulk density, penetration resistance and gravimetric moisture, obtained by applying geostatistics in precision agriculture, can effectively indicate the physical behavior of agricultural soils in longitudinal profiles. In this way, the spatial dependence of physical properties in streets of coffee plantations with different lengths was evaluated in the southern Minas Gerais, Brazil. For this purpose, five longitudinal profiles were measured in streets, each one with depths ranging from 0 to 0.60 m, in six layers of 0.10 m, being the database composed of 432 property, 144 by property, submitted to the ordinary kriging geostatistical method in order to obtain spatial variability maps using the R software. They were evaluated by the lower mean cross-validation error of theoretical models fitted by ordinary least squares (OLS), being detected in higher superficial layers, from 0 to 0.30 m, lower bulk density and lower penetration resistance, with variable gravimetric moisture in the length direction of some streets of coffee plantations, being that these properties presented different structures of spatial dependence for each street.

Subcutaneous injection of an immunologically tolerated protein up to 5 days before skin injuries improves wound healing

Oral tolerance blocks the development of specific immune responses to proteins ingested by the oral route. One of the first registries of oral tolerance showed that guinea pigs fed corn became refractory to hypersensitivity to corn proteins. Mice fed with chow containing corn are tolerant to zein, and parenteral injection of zein plus adjuvant blocks immunization to unrelated proteins injected concomitantly and reduces unspecific inflammation. Extensive and prolonged inflammatory infiltrate in the wound bed is one of the causes of pathological wound healing. Previous research shows that intraperitoneal injection of zein concomitant with skin injuries reduces the inflammatory infiltrate in the wound bed and improves wound healing. Herein, we tested if one subcutaneous injection of zein before skin injury improves wound healing. We also investigated how long the effects triggered by zein could improve skin wound healing. Mice fed zein received two excisional wounds on the interscapular skin under anesthesia. Zein plus Al(OH)3 was injected at the tail base at 10 min, or 3, 5, or 7 days before skin injuries. Wound healing was analyzed at days 7 and 40 after injury. Our results showed that a zein injection up to 5 days before skin injury reduced the inflammatory infiltrate, increased the number of T-cells in the wound bed, and improved the pattern of collagen deposition in the neodermis. These findings could promote the development of new strategies for the treatment and prevention of pathological healing using proteins normally found in the common diet.

Homeopathic products (HP) are a preventive and therapeutic alternative in pig farming. This study evaluated the effect of the concentration of dietary HP on performance, carcass traits, meat quality, and plasma metabolites of growing-finishing pigs. A total of 60 crossbred male pigs (Agroceres PIC * DanBred), immunocastrated at 90 and 120 days old, with an initial BW of 30.71 ± 2.60 kg were allocated in a completely randomized design to three treatments: i) control diet (CT), without HP), ii) CT + HPi (Figotonus® and Sanoplus®) (1 g/kg of diet), and iii) CT + HP2 (Figotonus and Sanoplus) (2 g/kg of diet). The results indicate that pigs fed diets containing HP2 had a greater average daily gain compared with those fed the control diet, although a higher feed efficiency was observed when pigs were fed the HP1 diet. Animals fed the HP2 diet showed a slight increase in initial postmortem pH in the Longissimus dorsi muscle compared to the control group. Pigs fed HP2 showed a higher initial temperature in the L. dorsi than those fed the control diet. However, there was no effect on plasma metabolite concentration. Based on the results of the present study, the HP1 diet enhanced the performance of finishing pigs without negative effects on meat traits and plasma metabolites.

Abstract The objective of the study was to compare the richness and diversity of ant assemblages in an agroecological system under peach orchard, conventional system under peach orchard cultivation and native vegetation in rural properties located in a Pampa Biome. The study was conducted in four samplings in 2017: 1st and 09th March (summer); 24th and 31st July (winter); and four samplings in 2018: 23rd and 30th January (summer); 31st July and 07th August (winter). Pitfall traps were used. The assemblages were characterized and compared using richness, number of occurrences of ants, Shannon diversity (H’), equitability, rarefaction analysis and Chao 1. The association of the species with the samples was evaluated by a Principal Component Analysis (PCA). The agroecological system had the highest number of occurrences, while the conventional orchard the lowest number. Richness and abundance were greatest during the summer. The conventional peach orchard obtained the lowest H’ for both seasons when compared to the agroecological orchard and native vegetation. The PCA explained 77.40% of the occurrence of ants in the environments and in the seasons. The results found demonstrated that conservationist systems tend to harbor greater wealth and diversity of ant assemblages, as well as occurring in native áreas.

OBJECTIVE: The folate pathway is involved in hepatic carcinogenesis and angiogenesis. Polymorphisms in genes related to such processes, including methylene tetrahydrofolate reductase (MTHFR) and vascular endothelial growth factor (VEGF)] may play an important role in the development of hepatocellular carcinoma (HCC). The objective of this study was to evaluate MTHFR and VEGF polymorphisms in Brazilian patients with hepatitis C virus (HCV)-related HCC. METHODS: A total of 119 patients diagnosed with confirmed HCC and HCV were included in the study. SNP genotyping assays were performed using real-time PCR. VEGFA (rs2010963, rs3025039, and rs833061) and MTHFRC677T (rs1801133, rs1801131) polymorphisms were evaluated. RESULTS: The C alleles of MTHFR (rs1801131) and VEGF (rs2010963) were associated with protection against the development of multinodular HCC, while the T allele of MTHFR (rs1801133) was associated with a higher risk of multinodular presentation [p=0.04 OR 1.835 CI (1.022-3.297)]. Multivariate analysis revealed that the GG/GC genotypes of VEGF rs2010963 were independently associated with multinodular tumors at diagnosis (p=0.013; OR 4.78 CI (1.38-16.67)]. CONCLUSION: Our results suggest that these polymorphisms may increase the risk of rapid tumor progression in patients with HCV infection. This subgroup of patients with HCC and who present polymorphism is more likely to be diagnosed with multinodular disease and not be amenable to receiving curative treatments. These data must be validated in larger cohorts, and the screening intervals can be customized based on genetic history.

RESUMO Este trabalho apresenta uma análise de clusterização de Áreas Mínimas Comparáveis (AMC’s) para traçar um mapa de agrupamentos homogêneos a partir de uma combinação de variáveis climáticas, de características do solo e de produção agropecuária. A metodologia permite a visualização de interações entre as diversas variáveis utilizadas, identificando-se, por exemplo, padrões de coexistência, no nível municipal, de diferentes culturas agrícolas. A discussão apresenta os algoritmos tradicionais sem contiguidade (aglomerativo hierárquico e k-means) e o algoritmo aglomerativo hierárquico com imposição de contiguidade. Busca-se, dessa forma, explorar diferenças entre as tipologias construídas com diferentes abordagens, além de prover configurações alternativas de agrupamentos. Ainda, as metodologias discutidas permitem a incorporação de critérios tradicionais de escolha do número de clusters, tais como estatísticas CCC, pseudo-F e pseudo-t 2.
ABSTRACT This paper presents a clustering analysis of Minimum Comparable Areas(MCAs) to draw a map of homogeneous grouping from a combination of climatic variables, soil characteristics and agricultural production. The methodology allows the visualization of interactions among the many different variables used, indentifying, for example, coexistence patterns, at the municipal level, of different crops. The discussion presents the traditional algorithms with no contiguity (hierarchical algorithm and k-means) and the agglomerative hierarchical algorithm with contiguity. Therefore, this paper seeks to explore differences among the typologies built with different approaches, as well as, provide alternative configurations of grouping. Also, the methodologies discussed allow the incorporation of traditional criteria for choosing the number of clusters, such as the CCC, pseudo-F and pseudo- t 2 statistics.

It is currently accepted that superoxide anion (O2•−) is an important mediator in pain and inflammation. The role of superoxide anion in pain and inflammation has been mainly determined indirectly by modulating its production and inactivation. Direct evidence using potassium superoxide (KO2), a superoxide anion donor, demonstrated that it induced thermal hyperalgesia, as assessed by the Hargreaves method. However, it remains to be determined whether KO2 is capable of inducing other inflammatory and nociceptive responses attributed to superoxide anion. Therefore, in the present study, we investigated the nociceptive and inflammatory effects of KO2. The KO2-induced inflammatory responses evaluated in mice were: mechanical hyperalgesia (electronic version of von Frey filaments), thermal hyperalgesia (hot plate), edema (caliper rule), myeloperoxidase activity (colorimetric assay), overt pain-like behaviors (flinches, time spent licking and writhing score), leukocyte recruitment, oxidative stress, and cyclooxygenase-2 mRNA expression (quantitative PCR). Administration of KO2 induced mechanical hyperalgesia, thermal hyperalgesia, paw edema, leukocyte recruitment, the writhing response, paw flinching, and paw licking in a dose-dependent manner. KO2 also induced time-dependent cyclooxygenase-2 mRNA expression in the paw skin. The nociceptive, inflammatory, and oxidative stress components of KO2-induced responses were responsive to morphine (analgesic opioid), quercetin (antioxidant flavonoid), and/or celecoxib (anti-inflammatory cyclooxygenase-2 inhibitor) treatment. In conclusion, the well-established superoxide anion donor KO2 is a valuable tool for studying the mechanisms and pharmacological susceptibilities of superoxide anion-triggered nociceptive and inflammatory responses ranging from mechanical and thermal hyperalgesia to overt pain-like behaviors, edema, and leukocyte recruitment.

Resumo Um levantamento atualizado das plantas com sementes e análises relevantes acerca desta biodiversidade são apresentados. Este trabalho se iniciou em 2010 com a publicação do Catálogo de Plantas e Fungos e, desde então vem sendo atualizado por mais de 430 especialistas trabalhando online. O Brasil abriga atualmente 32.086 espécies nativas de Angiospermas e 23 espécies nativas de Gimnospermas e estes novos dados mostram um aumento de 3% da riqueza em relação a 2010. A Amazônia é o Domínio Fitogeográfico com o maior número de espécies de Gimnospermas, enquanto que a Floresta Atlântica possui a maior riqueza de Angiospermas. Houve um crescimento considerável no número de espécies e nas taxas de endemismo para a maioria dos Domínios (Caatinga, Cerrado, Floresta Atlântica, Pampa e Pantanal), com exceção da Amazônia que apresentou uma diminuição de 2,5% de endemicidade. Entretanto, a maior parte das plantas com sementes que ocorrem no Brasil (57,4%) é endêmica deste território. A proporção de formas de vida varia de acordo com os diferentes Domínios: árvores são mais expressivas na Amazônia e Floresta Atlântica do que nos outros biomas, ervas são dominantes no Pampa e as lianas apresentam riqueza expressiva na Amazônia, Floresta Atlântica e Pantanal. Este trabalho não só quantifica a biodiversidade brasileira, mas também indica as lacunas de conhecimento e o desafio a ser enfrentado para a conservação desta flora.
Abstract An updated inventory of Brazilian seed plants is presented and offers important insights into the country's biodiversity. This work started in 2010, with the publication of the Plants and Fungi Catalogue, and has been updated since by more than 430 specialists working online. Brazil is home to 32,086 native Angiosperms and 23 native Gymnosperms, showing an increase of 3% in its species richness in relation to 2010. The Amazon Rainforest is the richest Brazilian biome for Gymnosperms, while the Atlantic Rainforest is the richest one for Angiosperms. There was a considerable increment in the number of species and endemism rates for biomes, except for the Amazon that showed a decrease of 2.5% of recorded endemics. However, well over half of Brazillian seed plant species (57.4%) is endemic to this territory. The proportion of life-forms varies among different biomes: trees are more expressive in the Amazon and Atlantic Rainforest biomes while herbs predominate in the Pampa, and lianas are more expressive in the Amazon, Atlantic Rainforest, and Pantanal. This compilation serves not only to quantify Brazilian biodiversity, but also to highlight areas where there information is lacking and to provide a framework for the challenge faced in conserving Brazil's unique and diverse flora.

Development of a larvicidal nanoemulsion with Copaiba (Copaifera duckei) oleoresin

Copaiba (Copaifera duckei Dwyer, Fabaceae) oleoresin is an important Amazonian raw material. Despite its insecticidal potential, poor water solubility remains a challenge for the development of effective and viable products. Nanotechnology has emerged as a promising area to solve this problem, especially oil-in-water nanoemulsions. On this context, the aim of the present study was to develop oil-in-water nanoemulsions using copaiba oleoresin dispersed through a high internal phase; and evaluate its potential insecticidal action against Aedes aegypti larvae. Overall, 31 formulations were prepared, ranging from 11.5 ± 0.2 to 257.3 ± 4.1 nm after one day of manipulation. Some of them reached small mean droplet sizes (< 200 nm) and allowed achievement of a nanoemulsion region. The formulation consisted of 5% (w/w) of copaiba oil, 5% (w/w) of surfactant and 90% (w/w) of water, which presented mean droplet size of 145.2 ±0.9 nm and polidispersity of 0.378 ± 0.009 after one day of manipulation, and these were evaluated for larvicidal potential. According to mortality level (250 ppm - 93.3 after 48 h), this nanoemulsion was classified as a promising insecticidal agent against Aedes aegypti larvae. The present study allowed the development of low-cost ecofriendly green natural-based nanoformulations with potential larvicidal activity, using a nanobiotechnology approach.

OBJETIVO: Descrever a densidade mineral óssea (DMO) e conteúdo mineral ósseo (CMO) de crianças e adolescentes que vivem com o vírus da imunodeficiência humana e comparar com os dados do National Health and Nutrition Examination Survey IV (NHANES IV). MÉTODO: Participaram do estudo48 crianças e adolescentes (sete a 17 anos de idade) com infecção pelo vírus da imunodeficiência humana adquirida por transmissão vertical. A DMO e o CMO foram mensurados pela absorciometria por dupla emissão de raios-X, calculando-se escores-z com base nos dados do NHANES IV. Nos prontuários médicos foram obtidas as informações dos parâmetros clínicos e laboratoriais da infecção pelo vírus da imunodeficiência humana. Foram ainda avaliada a atividade física, a ingestão de cálcio e a maturação esquelética. Utilizaram-se procedimentos da estatística descritiva e inferencial, estabelecendo níveis de significância de 5%. RESULTADOS :Os pacientes soropositivos demonstraram valores inferiores comparados aos dados do NHANES IV em todos os escores-z da massa óssea (média = -0,52 a -1,22, dp = 0,91 e 0,84, respectivamente). Com base no z-DMOsubtotal, há uma prevalência de 16,7% de crianças e adolescentes com massa óssea reduzida para a idade. Indivíduos que utilizaram inibidores de protease apresentaram um z-DMOtotal inferior, comparado ao grupo que não utilizou (-1,31 vs. -0,79; p = 0,02). Não foram encontradas diferenças na massa óssea em relação ao nível de atividade física e ingestão de cálcio. CONCLUSÕES: Na presente amostra, crianças e adolescentes que vivem com o vírus da imuno deficiência humana possuem baixa massa óssea para idade, e o uso de inibidores de protease parece estar relacionado a tais reduções.
OBJECTIVE: To describe bone mineral density (BMD) and bone mineral content (BMC) in children and adolescents infected with the human immunodeficiency virus (HIV), and to compare them with data from the National Health and Nutrition Examination Survey IV (NHANES IV). METHOD: The study included 48 children and adolescents (7 to 17 years old) infected with HIV through vertical transmission. BMC and BMD were measured by dual energy absorptiometry X-ray, by calculating z-scores based on data from NHANES IV. The information on clinical and laboratory parameters of infection by HIV was obtained from medical records. Physical activity, calcium intake, and skeletal maturation were also assessed. Descriptive and inferential statistical procedures were used, with levels of significance set at 5%. RESULTS: Seropositive patients presented lower values compared to data from NHANES IV in all z-scores of bone mass (mean = -0.52 to -1.22, SD = 0.91 and 0.84, respectively). Based on the subtotal z-BMD, there was a prevalence of 16.7% of children and adolescents with low bone mass for age. Individuals using protease inhibitors presented a lower total z-BMD when compared to the group that did not use (-1.31 vs. -0.79, p = 0.02). There were no bone mass differences in relation to physical activity and calcium intake. CONCLUSIONS: In the present sample children and adolescents living with HIV have low bone mass for age, and the use of protease inhibitors appears to be related to such decreases.

Verificaram-se os efeitos da associação de furosemida e fenilbutazona sobre variáveis hidroeletrolíticas de cavalos antes e após a corrida. Dezenove equinos foram distribuídos em três grupos, de acordo com os protocolos de tratamento. O primeiro grupo, de cinco animais, não recebeu medicação (grupo-controle); o segundo grupo, de sete animais, foi tratado com furosemida, na dose de 1mg/kg, por via intramuscular, até quatro horas antes do páreo; o terceiro, de sete animais, recebeu furosemida, por via intramuscular, e fenilbutazona, por via intravenosa, nas doses de 1,0 e 4,4mg/kg, respectivamente, até quatro horas antes da corrida. Amostras de sangue foram colhidas antes, imediatamente após e duas horas após o páreo, para avaliação da osmolalidade plasmática e das concentrações plasmáticas de sódio, potássio e cloreto. A utilização de furosemida e da associação furosemida e fenilbutazona até 4h antes dos páreos nas dosagens descritas alterou (P<0,05) a osmolalidade plasmática dos equinos, mas não alterou (P>0,05) as concentrações de sódio, potássio e cloreto. Os páreos alteraram de forma fisiológica a osmolalidade plasmática e a concentração sanguínea de K+ devido ao exercício de alta intensidade.
The objective of this study was to verify the effects of furosemide and phenylbutazone association on fluid and electrolyte balance characteristics of horses before and after a race. Nineteen horses were divided into three groups according to treatment protocols. The first group (five animals - control) was not medicated. A second group (seven animals) was treated with furosemide (1mg/kg, intramuscular up to four hours before the race). A third group (seven animals) received furosemide (1mg/kg) and phenylbutazone (4.4mg/kg), both intramuscular, up to four hours before race. Blood samples were collected before, immediately after and two hours after a race to evaluate the plasma osmolality and sodium, potassium and chloride concentrations. The use of furosemide and furosemide plus phenylbutazone up to four hours before the race altered (P<0.05) the plasma osmolality but did not change (P>0.05) the sodium, potassium and chloride concentrations. It was not possible to determine an antagonist effect of phenylbutazone on furosemide, based on fluid and electrolyte balance. Due to the high intensity exercise, the increase in plasma osmolality and potassium concentration was attributed to the race effect.

Pneumonectomy is associated with high rates of morbimortality, with postpneumonectomy pulmonary edema being one of the leading causes. An intrinsic inflammatory process following the operation has been considered in its physiopathology. The use of corticosteroids is related to prevention of this edema, but no experimental data are available to support this hypothesis. We evaluated the effect of methylprednisolone on the remaining lungs of rats submitted to left pneumonectomy concerning edema and inflammatory markers. Forty male Wistar rats weighing 300 g underwent left pneumonectomy and were randomized to receive corticosteroids or not. Methylprednisolone at a dose of 10 mg/kg was given before the surgery. After recovery, the animals were sacrificed at 48 and 72 h, when the pO2/FiO2 ratio was determined. Right lung perivascular edema was measured by the index between perivascular and vascular area and neutrophil density by manual count. Tissue expression of vascular endothelial growth factor (VEGF) and transforming growth factor-beta (TGF-β) were evaluated by immunohistochemistry light microscopy. There was perivascular edema formation after 72 h in both groups (P = 0.0031). No difference was observed between operated animals that received corticosteroids and those that did not concerning the pO2/FiO2 ratio, neutrophil density or TGF-β expression. The tissue expression of VEGF was elevated in the animals that received methylprednisolone both 48 and 72 h after surgery (P = 0.0243). Methylprednisolone was unable to enhance gas exchange and avoid an inflammatory infiltrate and TGF-β expression also showed that the inflammatory process was not correlated with pulmonary edema formation. However, the overexpression of VEGF in this group showed that methylprednisolone is related to this elevation.

RESUMO O mamoeiro Carica papaya (L.) é acometido por diversos problemas fitossanitários, sendo o ácaro Tetranychus urticae (Koch, 1836) (Acari: Tetranychidae) uma das pragas-chave. A utilização de fungos entomopatogênicos é uma alternativa ao controle por agrotóxicos, reduzindo os riscos de resistência e também a carga de substâncias tóxicas ao ambiente. Este trabalho foi realizado para verificar se as diferentes cultivares de mamoeiro interferem na patogenicidade de Beauveria bassiana, Metarhizium anisopliae e Lecanicillium longisporum. No bioensaio foram utilizadas fêmeas adultas de T. urticae mantidas por pelo menos uma geração sobre folhas de quatro cultivares de mamão, Calimosa, Golden, Sunrise e Tainung 01, e também de uma testemunha de feijão-de-porco. Foram então transferidas dez fêmeas para placas de Petri com discos de folhas das respectivas cultivares de origem e de feijão-de-porco. Em seguida, os ácaros foram pulverizados com suspensões 5 x 107 conídios.mL-1 em Torre de Potter. As suspensões de conídios foram preparadas a partir das formulações comerciais: Boveril® B. bassiana; Metarril® M. anisopliae; Vertirril® L. longisporum. Foi avaliada a mortalidade corrigida e confirmada. Foram observadas diferenças (Tukey, P < 0,05) entre as cultivares na resposta patogênica de T. urticae. Para o 3° DAP, uma melhor eficiência em porcentagem de mortalidade foi observada para o formulado Vertirril® aplicado nas cultivares Calimosa, Golden e Sunrise, e para Metarril® na cultivar Tainung 01. Para o 5° DAP, as eficiências entre as cultivares foram muito próximas.
ABSTRACT Papaya Carica papaya (L.) is affected by various phytosanitary problems, and the mite Tetranychus urticae (Koch, 1836) (Acari: Tetranychidae) is a key pest. The use of entomopathogenic fungi is an alternative to the use of pesticides, lowering the risk of resistance while reducing the the load of toxic substances in the environment. This study was conducted to determine whether the different cultivars of papaya interfere with the pathogenicity of Beauveria bassiana. Metarhizium anisopliae and Lecanicillium longisporum. The bioas say was carried out using adult T. urticae females maintained for at least one generation on leaves of four papaya cultivars (Calimosa, Golden, Sunrise, Tainung 01) or on jack bean leaves (control). Ten females were transferred to Petri dishes with leaf disks of the origin cultivars and jack beans. The mites were then sprayed with a 5 x 107 conidia.mL-1 suspension in a Potter Tower. Conidial suspensions were prepared from the commercial formulations Boveril® B. bassiana, Metarril® M. anisopliae, and Vertirril® L. longisporum. The corrected and confirmed mortalities were evaluated. Differences were observed (Tukey, P < 0.05) among the papaya cultivars in terms of the T. urticae pathogenic response. On the 3rd day after spraying (DAS), a better efficiency in mortality percentage was observed for the Vertirril® formulation applied to the Calimosa, Golden and Sunrise cultivars, and for Metarril® applied to the Tainung 01cultivar. On the 5th DAS, the efficiencies among the cultivars were very close.

Avaliaram-se o papel do óxido nítrico (NO) por meio da inibição da enzima óxido nítrico sintase induzível (iNOS), após a adição da aminoguanidina (AG), na motilidade, no vigor e na integridade da membrana plasmática nos tempos de 15, 60, 120, 180, 240 e 300min e a atividade mitocondrial e a capacitação de espermatozoides bovinos após 300min de cultivo. Adicionaram-se diferentes concentrações (0,001, 0,01 e 0,1M) de AG durante a capacitação induzida pela heparina e 500μM de nitroprussiato de sódio (SNP, doador de NO) à concentração deletéria. A adição de 0,1M de AG diminuiu a motilidade e o vigor espermático e a integridade da membrana (P<0,05). A adição de SNP ao meio de cultivo com 0,1M de AG somente reverteu a integridade da membrana após 300min. A inibição da síntese de NO pela adição de AG não alterou a atividade mitocondrial. A percentagem de oócitos penetrados com espermatozoides tratados com 0,01 e 0,1M de AG diminuiu 20,3 e 100%, respectivamente, em relação aos não tratados (controle) (P<0,05), contudo houve aumento de 15% na percentagem de oócitos desnudados penetrados com espermatozoides capacitados em presença de 0,1M de AG. Conclui-se que a inibição da síntese de NO pela AG diminuiu a qualidade espermática durante a capacitação de espermatozoides bovinos in vitro, exceto a atividade mitocondrial. Somente a integridade da membrana foi revertida após adição de NO, sugerindo diferentes vias de ação do NO na qualidade espermática ao longo da capacitação in vitro de espermatozoides bovinos.
The role of nitric oxide (NO) was evaluated by inhibition of inducible nitric oxide synthase (iNOS), with aminoguanidine (AG) on motility, vigor, and plasmatic membrane integrity of bovine spermatozoa culture after 15, 60, 120, 180, 240, and 300min and on mitochondrial activity and capacitation after 300min, respectively. Different concentrations, 0.001, 0.01, and 0.1M of AG were added during the heparin induced capacitation and sodium nitroprusside (SNP, NO donor-500μM) to the deleterious concentration. The addition of 0.1M of AG diminished progressive motility, spermatic vigor, and membrane integrity (P<0.05). SNP addition to the 0.1M of AG did revert only plasmatic membrane integrity after 300min. Mitochondrial activity was not influenced by addition of AG. Percentage of penetrated oocytes after addition of 0.01 and 0.1M of AG diminished, 20.3 and 100%, respectively, in relation to the control oocytes (P<0.05). However, an increase of 15% was observed when denuded oocytes were used with 0.1M AG treated sperm (P<0.05). It was concluded that the inhibition of NO synthesis with aminoguanidine diminished sperm quality during in vitro capacitation of bovine spermatozoa, except the mitochondrial activity. Only membrane integrity was reverted with the addition of NO to culture medium, suggesting different pathways of NO action on bovine sperm quality during in vitro capacitation.
