Chitosan Imparts Better Biological Properties for Poly(ε-caprolactone) Electrospun Membranes than Dexamethasone

Poly(ε-caprolactone), an aliphatic polyester with biodegradability and cytocompatibility, has been used to create scaffolds for tissue engineering purposes. However, the hydrophobicity and low water absorptivity of poly(ε-caprolactone) reduce cell anchorage on their membranes. Here, poly(ε-caprolactone)-based scaffolds were prepared by electrospinning of poly(ε-caprolactone)-chitosan blend, and poly(ε-caprolactone)-dexamethasone solution. Chitosan and dexamethasone play an essential role to increase the scaffolding performance of poly(ε-caprolactone)-based electrospun membranes. A poly(ε-caprolactone) membrane without chitosan and dexamethasone did not provide satisfactory results to promote cell culture of adipose mesenchymal stem cells (AMSCs). Compared to the poly(ε-caprolactone)-dexamethasone surface, poly(ε-caprolactone)-chitosan membrane imparts better cytoskeletal reorganization, and cell spreading, increasing the strength of cell attachment. Also, poly(ε-caprolactone)-chitosan composite provides strong antimicrobial activity against Pseudomonas aeruginosa (ca. 90% inhibition). Therefore, the poly(ε-caprolactone)-chitosan composite is a better alternative to treat skin diseases and promote skin regeneration than conventional approaches based on dexamethasone.

Resumo O controle higiênico e sanitário nas Unidades de Alimentação e Nutrição (UAN) é considerado um procedimento padrão para produzir refeições adequadas e reduzir o risco de doenças transmitidas pelos alimentos e infecções hospitalares. Este estudo teve como objetivo isolar e identificar bactérias de equipamentos e superfícies de contato com alimentos em uma UAN hospitalar, bem como avaliar a condição sanitária. Do mesmo modo, analisou-se a adesão dos micro-organismos em tábuas de corte de polietileno. A presença de micro-organismos aeróbios mesófilos, leveduras, fungos, Sthapylococcus coagulase-positivos, coliformes, coliformes fecais e Escherichia coli foi analisadas na superfície de mesas do refeitório, superfícies de bancada e tábuas de corte usadas para manuseio de carne ou vegetais e, em equipamentos como micro-ondas e refrigeradores. A identificação molecular foi feita pelo sequenciamento do gene 16S rRNA. A adesão dos micro-organismos (formação de biofilmes) em tábuas de corte de carne e de vegetais também foi avaliada por microscopia eletrônica de varredura. Os resultados mostraram elevada contagem para todos os micro-organismos analisados, exceto para E. coli, a qual não foi observada nas amostras. A análise molecular identificou espécies da família Enterobacteriaceae e Pseudomonadaceae. A análise de microscopia eletrônica de varredura revelaram adesão bacteriana nas superfícies das placsa de corte. Os resultados obtidos neste estudo indicaram que as condições higiênicas das superfícies e de equipamentos nesta UAN hospitalar estavam inadequadas. A aplicação de procedimentos operacionais padrão poderia auxiliar positivamente na padronização do controle higiênico-sanitário, reduzindo a contaminação microbiana e fornecendo um alimento seguro para pacientes hospitalizados.
Abstract The hygienic and sanitary control in Food and Nutrition Units (FNU) is considered a standard procedure to produce adequate meals and reduce the risk of foodborne diseases and hospital infections. This study aimed to evaluate the isolation and identification of bacteria from equipment and food contact surfaces in a hospital FNU as well as to evaluate the sanitary condition. Likewise, it was analyzed the adhesion of the microorganisms on polyethylene cutting boards. The presence of aerobic mesophilic microorganisms, yeasts, molds, coagulase-positive staphylococci, coliform and fecal coliform, and Escherichia coli were analyzed on eating tables, countertop surfaces and cutting boards used for meat or vegetable handling, and equipment such as microwaves and refrigerators. The molecular identification it was done by 16S rRNA gene sequencing. The adhesion of the microorganisms (biofilm formation) on meat and vegetable cutting boards was also evaluated by scanning electron microscopy. The results showed high numbers of all microorganisms, except for E. coli , which was not observed in the samples. The molecular analysis identified species of the Enterobacteriaceae family and species of the Pseudomonadaceae family. Scanning electron microscopy analyses revealed bacterial adhesion on the cutting board surfaces. The results obtained in this study indicated that the hygienic conditions of surfaces like plastic cutting boards and equipment in this hospital FNU were inadequate. The achievement and application of standard operating procedures could positively help in the standardization of sanitary control, reducing the microbial contamination and providing a safe food to hospitalized patients.

Resumo Flutuações na densidade populacional de planorbídeos hospedeiros do S. mansoni são influenciadas por fatores climáticos. O conhecimento sobre a interferência de alterações na temperatura da água nestas populações é um aspecto importante da epidemiologia da esquistossomose. Neste experimento avaliou-se a influência de diferentes temperaturas no desenvolvimento de Schistosoma mansoni em Biomphalaria glabrata variantes melânica e albina. Os resultados indicaram uma relação intrínseca entre temperatura e desenvolvimento do parasito na fase intramolusco, independente da pigmentação do manto dos moluscos. Quanto mais elevada a temperatura, menor o período necessário para desenvolvimento do parasito e maior mortalidade dos moluscos infectados. Conclui-se que, na presença de alterações climáticas, o aumento da temperatura em regiões frias e alagadas poderá favorecer o estabelecimento de novos focos de transmissão da esquistossomose alterando a extensão geográfica e ampliando o potencial epidemiológico da transmissão. Em regiões de clima quente, o aumento da temperatura, por sua vez, poderá comprometer a transmissão da doença em virtude do estresse biológico sofrido por parasito e hospedeiro. Nestas condições, poderá ocorrer a morte do parasito ou uma alteração na sua habilidade de infectar novas espécies de moluscos hospedeiros em novas áreas. Padrões de pigmentação do manto nos moluscos não demonstraram interferência significativa no desenvolvimento do parasita.
Abstract Fluctuations in population density of planorbid hosts of S. mansoni are influenced by climatic factors. The knowledge about interference from changes in water temperature in these populations is an important aspect of the epidemiology of schistosomiasis. In this experiment, it is explored the influence of different temperatures on the development of Schistosoma mansoni in Biomphalaria glabrata melanic and albino variants. The results indicated an intrinsic relationship between temperature and development of the parasite in the intramollusc phase, independent of the pigmentation of the mantle of the molluscs. The higher the temperature, the shorter the period necessary for the development of the parasite was while the higher the mortality of infected mollusks. It is concluded that, in the presence of climate change, the increasement of temperature in cold and flooded regions may encourage the establishment of new foci of transmission of schistosomiasis by changing the geographic extent and extending the epidemiological transmission potential. In warm climates, higher temperatures, however, could compromise the transmission of the disease because of biological stress suffered by parasite and host. Under these conditions, it can result in the death of the parasite or a change in their ability to infect new host species of molluscs in new areas. Mantle pigmentation patterns in molluscs have not shown significant interference in the development of the parasite.

Resumo Biomphalaria amazonica é uma espécie de planorbídeo considerada vetora potencial do Schistosoma mansoni. É amplamente distribuída na zona neotropical, especialmente no Norte e Centro-Oeste do Brasil e Norte da Bolívia. O presente trabalho teve por objetivo estudar a relação parasito-hospedeiro entre B. amazonica e S. mansoni (linhagens BH e SJ). Espécimes de B. amazonica e sua água de condicionamento foram examinados em relação à sua capacidade de atração miraxonal. A infectividade dos moluscos foi testada expondo-os a 20 miracídios de ambas as linhagens. A viabilidade dos esporocistos e o desenvolvimento de reações amebocitárias foram estudados após cada molusco ser exposto a 100 miracídios. Apesar de não eliminarem cercárias, B. amazonica provou ser capaz de atrair 77% dos miracídios a que foram expostos. Esporocistos viáveis sem reação amebócitaria foram encontrados 96 horas após a exposição aos miracídios. Esses resultados indicam a suscetibilidade de B. amazonica às linhagens BH e SJ de S. mansoni e, portanto, demonstram a importância desta espécie de planorbídeo como um vetor potencial do trematodeo na área onde ele ocorre.
Abstract Biomphalaria amazonica is a planorbid species considered a potential host of Schistosoma mansoni. It is widely distributed in the Neotropical zone, particularly in the North and Centre-West of Brazil and in the North of Bolivia. The aim of the present study was to determine the host-parasite relationship between B. amazonica and S. mansoni (BH and SJ strains). Specimens of B. amazonica and their snail-conditioned water were examined in terms of their ability to attract miracidia. The infectivity of the mollusks was determined by exposing them to 20 miracidia of both strains. Sporocyst development and amebocyte reactions were studied after each mollusk specimen was exposed to 100 miracidia. Although no cercariae were eliminated, specimens of B. amazonica proved capable of attracting 77% of the miracidia they were exposed to. Viable sporocysts with no amebocyte reaction were found 96 hours after the exposure to miracidia. These results indicate the susceptibility of B. amazonica to the BH and SJ strains of S. mansoni, and therefore demonstrate the importance of this planorbid species as a potential vector of the trematode in the areas where it occurs.

Spatial distribution of southern brown shrimp (Farfantepenaeus subtilis) on the Amazon continental shelf: a fishery, marine geology and GIS integrated approach

Resumo A distribuição espacial do camarão-rosa Farfantepenaeus subtilis (Pérez-Farfante, 1967) foi estudada a partir de dados oriundos da pesca industrial, os quais foram associados às características geológicas e oceanográficas dos ambientes bentônicos da plataforma continental do Amazonas. A partir do uso de um sistema de informações geográficas (SIG), este trabalho teve como objetivo calcular a abundância relativa do camarão rosa baseada na captura por unidade de esforço (CPUE) e compará-la com dados batimétricos, tipo de estrutura sedimentar, taxa de sedimentação e salinidade de fundo. Como resultado, nós podemos afirmar que a abundância relativa (em CPUE) não está uniformemente distribuída. A análise espacial indica que o esforço do arrasto da pesca industrial foi realizado na região frontal do delta submerso (foreset) do Amazonas, entre 40 e 60 m de profundidade. Nesta região, os camarões são responsáveis pela bioturbação dos sedimentos e geração de uma estrutura sedimentar chamada lama mosqueada. Na região do foreset, as taxas de sedimentação foram de até 10 cm/yr; o processo de ressuspensão foi reduzido e a salinidade de fundo mostrou-se alta (~ 36). Parece que todos estes fatores definem uma área estável de lama com intensa bioturbação. Esta notável atividade biológica é explicada pela ocorrência de alta abundância de F. subtilis que parece ter sido originada pela alça microbiana. A partir da combinação de dados pesqueiros com dados ambientais em um SIG, é possível identificar um padrão de distribuição de abundância do camarão rosa e outros recursos pesqueiros de importância econômica, assim como compreender como eles variam espacialmente.
Abstract The spatial distribution of the southern brown shrimp Farfantepenaeus subtilis (Pérez-Farfante, 1967) was studied based on industrial fishing fleet activities and is associated with geological and oceanographic characteristics of the benthonic environments on the Amazon continental shelf. Using a geographical information system (GIS) this paper sought to calculate the relative abundance of brown shrimp based on catch per unit effort (CPUE) and compare it with bathymetry, type of sedimentary structure, sedimentation rate and bottom salinity. As a result, we have concluded that the relative abundance (in terms of CPUE) is not uniformly distributed in space. Spatial analysis indicates that commercial trawling efforts were made in the (foreset) region of the subaqueous Amazon delta at depths of 40 to 60 m. In this region, prawn are responsible for the bioturbation of the sediments and the creation of a sedimentary structure called mottled mud. In the foreset region, sedimentation rates progressively increased up to 10 cm.yr-1; re-suspension was reduced and bottom salinity was high (~ 36). It appears that all of these factors define a stable muddy area with intense bioturbation. This notable biological activity is to be explained by the occurrence of a high F. subtilis abundance that appears to originate in a microbial loop. We concluded that by combining fishery information with environmental data from a GIS, it was possible to identify abundance distribution patterns for southern brown shrimp and other economically important fishery resources and to understand how they change on a large spatial-scale.

Redescription of Campoletis sonorensis (Cameron, 1886) (Hymenoptera, Ichneumonidae, Campopleginae), parasitoid of Spodoptera frugiperda (J. E. Smith, 1797) (Lepidoptera, Noctuidae) in Brazil

Resumo Spodoptera frugiperda é uma praga voraz de diversas culturas de importância econômica no Novo Mundo. No Brasil, suas larvas podem ser atacadas por diversas espécies de vespas parasitóides que são candidatos a agentes de controle biológico contra essa praga. Pesquisando os parasitóides da fauna de Spodoptera frugiperda em cultivos de milho no Brasil foram encontradas duas espécies do gênero Campoletis, muito similares morfologicamente. Este trabalho apresenta uma redescrição para Campoletis sonorensis usando caracteres morfológicos e DNA Barcoding (Hebert et al., 2003) com o objetivo de evitar erros de identificação desse grupo, aprimorando estudos de controle biológico.
Abstract The fall armyworm Spodoptera frugiperda (Lepidoptera; Noctuidae) is a voracious pest of numerous crops of economic importance throughout the New World. In Brazil, its larvae are attacked by several species of parasitoid wasps, making them potential candidate as biological control agents against this pest. A survey of the parasitoid fauna on S. frugiperda in maize crops throughout Brazil reveals two species of Campoletis, which are morphologicaly very similar species. In this paper we combine these data with pictures from the type material of C. sonorensis and C. flavicincta, as well as their descriptions to provide a redescription to Campoletis sonorensis (Cameron, 1886) using for this both morphological characters and DNA Barcoding (Hebert et al., 2003) information, in an attempt to help with the correct identification of the taxa to improve biological control studies.

Resumo Um levantamento atualizado das plantas com sementes e análises relevantes acerca desta biodiversidade são apresentados. Este trabalho se iniciou em 2010 com a publicação do Catálogo de Plantas e Fungos e, desde então vem sendo atualizado por mais de 430 especialistas trabalhando online. O Brasil abriga atualmente 32.086 espécies nativas de Angiospermas e 23 espécies nativas de Gimnospermas e estes novos dados mostram um aumento de 3% da riqueza em relação a 2010. A Amazônia é o Domínio Fitogeográfico com o maior número de espécies de Gimnospermas, enquanto que a Floresta Atlântica possui a maior riqueza de Angiospermas. Houve um crescimento considerável no número de espécies e nas taxas de endemismo para a maioria dos Domínios (Caatinga, Cerrado, Floresta Atlântica, Pampa e Pantanal), com exceção da Amazônia que apresentou uma diminuição de 2,5% de endemicidade. Entretanto, a maior parte das plantas com sementes que ocorrem no Brasil (57,4%) é endêmica deste território. A proporção de formas de vida varia de acordo com os diferentes Domínios: árvores são mais expressivas na Amazônia e Floresta Atlântica do que nos outros biomas, ervas são dominantes no Pampa e as lianas apresentam riqueza expressiva na Amazônia, Floresta Atlântica e Pantanal. Este trabalho não só quantifica a biodiversidade brasileira, mas também indica as lacunas de conhecimento e o desafio a ser enfrentado para a conservação desta flora.
Abstract An updated inventory of Brazilian seed plants is presented and offers important insights into the country's biodiversity. This work started in 2010, with the publication of the Plants and Fungi Catalogue, and has been updated since by more than 430 specialists working online. Brazil is home to 32,086 native Angiosperms and 23 native Gymnosperms, showing an increase of 3% in its species richness in relation to 2010. The Amazon Rainforest is the richest Brazilian biome for Gymnosperms, while the Atlantic Rainforest is the richest one for Angiosperms. There was a considerable increment in the number of species and endemism rates for biomes, except for the Amazon that showed a decrease of 2.5% of recorded endemics. However, well over half of Brazillian seed plant species (57.4%) is endemic to this territory. The proportion of life-forms varies among different biomes: trees are more expressive in the Amazon and Atlantic Rainforest biomes while herbs predominate in the Pampa, and lianas are more expressive in the Amazon, Atlantic Rainforest, and Pantanal. This compilation serves not only to quantify Brazilian biodiversity, but also to highlight areas where there information is lacking and to provide a framework for the challenge faced in conserving Brazil's unique and diverse flora.

Supercritical CO2 extraction of raw propolis and its dry ethanolic extract

Three types of propolis extract were prepared and analyzed with respect to their global extraction yields and with respect to the concentration of the following markers: 3,5-diprenyl-4-hydroxycinnamic acid; 3-prenyl-4-hydroxycinnamic acid; 4-hydroxycinnamic acid and 4-methoxy-3,5,7-trihydroxyflavone. The extract EEP (ethanolic extract of propolis) was obtained by the conventional method from raw propolis using ethanol as solvent. The extracts (SFE) were obtained by supercritical solvent extraction from the raw propolis using supercritical carbon dioxide (sc-CO2), with and without the addition of ethanol as a co-solvent. The fractionated supercritical extracts (FSCE) were obtained by fractionation (extract and raffinate) of the dry EEP with sc-CO2. EEP yields of 39.5% were obtained and maximum global extraction yields were 7.3% for SFE with no co-solvent, 51% for SFE with 15% ethanol and 18% for the FSCE extract fraction. The concentrations of the markers in the different extracts differed as a function of the operational parameters, indicating that the addition of co-solvent and the selectivity of sc-CO2 could be manipulated so as to obtain extracts with the yields and concentrations of interest.

Molecular variability in the maize grey leaf spot pathogens in Brazil

Isolates of Cercospora species from leaves displaying symptoms of grey leaf spot were collected in maize-producing areas of south-central Brazil in 2001 and 2002. Restriction digests of the internal transcribed spacer region of rDNA detected the presence of the same two Cercospora species described on maize in the United States, namely C. zeae-maydis and the recently described species, C. zeina . Genetic variability among isolates was assessed by analysing 104 amplified fragment length polymorphism loci. Cluster analysis confirmed the genetic separation of isolates into two species with a mean similarity of 35%. Similarity levels within species were high, averaging 93% and 92% among isolates of C. zeae-maydis and C. zeina , respectively. The mean genetic similarity between C. zeae-maydis and C. zeina and two isolates of C. sorghi f. sp. maydis was 45% and 35%, respectively. Results of this study showed that populations of the grey leaf spot pathogens in Brazil are similar to those in the United States regarding species composition and that C. zeina is also present in Brazil.

Efeito da porosidade nas propriedades mecânicas de uma alumina de elevada pureza

Uma alumina de elevada pureza dopada com MgO foi sinterizada entre 1300 e 1700 °C para preparação de corpos com diferentes porosidades. Foram determinadas a resistência à flexão em quatro pontos, a tenacidade à fratura pelo método SEVNB, a dureza Vickers, as velocidades sônicas (longitudinal e transversal) e as constantes elásticas (ni, E, G e K) pelo método do pulso-eco ultrassônico. Os resultados foram comparados com os modelos baseados em concentração de tensão (CT) e modelos baseados na área sólida mínima (ASM). A porosidade das amostras variou entre 0,8% e 35%. Em geral, as propriedades diminuíram com o aumento da porosidade, sendo que a diminuição foi menos acentuada nas velocidades sônicas e mais acentuada na dureza. O coeficiente de Poisson só diminuiu em porosidade acima de 19%. O modelo analítico (CT) que melhor se ajustou aos resultados dos módulos E e G foi o de Hashin-Rosen para configuração de poros cilíndricos alinhados transversalmente à tensão aplicada. A análise pelos modelos de ASM indicou que, até 19% de porosidade, o modelo de poros esféricos em arranjo cúbico foi predominante e, acima desta porosidade, atuou também o modelo de partículas esféricas em arranjo cúbico.
A high purity alumina doped with MgO was sintered between 1300 and 1700 °C in order to prepare samples with different porosities. The following properties were determined: four-point flexural strength, fracture toughness, by SEVNB method, Vickers hardness, (transversal and longitudinal) sonic velocities, and elastic constant (nu, E, G e K), by ultrasonic pulse-echo method. The results were compared with models based on stress concentration (SC) and models based on minimum solid area (MSA). The porosity of the samples varied between 0.8% and 35%. Generally, the properties lowered with increasing porosity. This lowering was weaker in sonic velocities and stronger in hardness. The Poisson's ratio lowered only above 19% porosity. The analytical model (SC) which best fitted the results of E and G moduli was the Hashin-Rosen model for cylindrical pores aligned transversally to the applied stress. Analysis by ASM models indicated that, until 19% porosity, the model for spherical pores in cubic stacking was predominant and, above this porosity, the model for spherical particles in cubic stacking also acted.

Com o objetivo de avaliar a disponibilidade de nitrogênio do solo para a aveia e o milho, realizou-se um experimento em microparcelas com vinte solos do Rio Grande do Sul. As microparcelas foram constituídas por recipientes com 20 L de solo, com drenagem livre, mantidas em céu aberto e com irrigação quando necessária. Foi determinado o N absorvido por plantas de aveia preta (Avena strigosa ) e por três cultivos sucessivos de milho (Zea mays ). Foram determinados, inicialmente, os teores de N-total e de matéria orgânica dos solos. Antes de cada cultivo, foram também determinados os teores extraíveis de N por KMnO4 0,05 mol L-1, tampão fosfato-borato e por KCl 2 mol L-1 às temperaturas de 100 e 95°C, durante 4 e 16 horas, respectivamente. O N do solo extraído por KCl 2 mol L-1 a 95°C por 16 h apresentou os maiores coeficientes de correlação com o somatório do N acumulado pelas plantas nos quatro cultivos. Os coeficientes de correlação entre o N extraído por todos os métodos químicos e o N acumulado pelas plantas aumentaram com a correção da acidez do solo a pH 6,0.
In order to evaluate organic N availability to oats and corn, a microplot experiment was carried out using 20 types of soil from Rio Grande do Sul State, Brazil. Microplots consisted of 20 L soil plastic pots with free drainage, maintained in an open area, with supplemental irrigation when necessary. N taken up by oats (Avena strigosa) and three corn (Zea mays) crops grown in successions was evaluated. Soil total-N and organic matter contents were initially determined as well as N extracted by acid 0.05 mol L-1 KMnO4, phosphate-borate buffer and by 2 mol L-1 KCl at 100°C and 95°C temperature for 4 and 16 hours, respectively, on samples taken before each crop. N extracted by 2 mol L-1 KCl at 95°C for 16 hours showed the highest correlation coefficient with total-N taken up by plants from all crops. Liming soil to pH 6.0 increased the correlation coefficients for all the tested methods.

Population dynamics and net primary production of the aquatic macrophite Nymphaea rudgeana C. F. Mey in a lotic environment of the Itanhaém River basin (SP, Brazil)

Neste trabalho, foi estudada a dinâmica populacional e estimada a produção primária líquida da macrófita aquática Nymphaea rudgeana em um braço do rio Itanhaém (Estado de São Paulo, Brasil). Esta espécie apresenta uma ampla variação anual de biomassa, em função da estação do ano, no local estudado. A partir do mês de novembro (13,1 g PS/m²), pode ser observado um aumento gradual da biomassa, atingindo o máximo em fevereiro (163,1 g PS/m²). Posteriormente, a biomassa diminui, mantendo-se em níveis baixos até um novo período de crescimento. A diminuição de biomassa está associada ao desenvolvimento de macrófitas flutuantes (Pistia stratiotes e Salvinia molesta) e, subseqüentemente, às condições ambientais desfavoráveis (valores de salinidade elevados) ao seu desenvolvimento. A produção primária líquida de N. rudgeana foi obtida a partir dos dados de biomassa e seu valor é estimado entre 3,02 e 3,82 t/ha/ano.
In this paper we evaluated the population dynamics and obtained estimates of the net primary production of the aquatic macrophyte Nymphaea rudgeana in an arm of the Itanhaém River (São Paulo State, Brazil). This species presents, in the studied area, a broad seasonal variation of biomass. As from November (13.1 g DW/m²) we observed a gradual increase of biomass that reached a maximum in February (163.1 g DW/m²). Then, the biomass decreased, maintaining low levels until a new growth period. The reduction of biomass is associated to the development of floating aquatic macrophytes (Pistia stratiotes and Salvinia molesta) and, subsequently to environmental factors (higher salinity values) that are unfavorable to their development. The net primary production of N. rudgeana was estimated from the biomass data, and the annual productivity value was estimated between 3.02 and 3.82 t/ha/year.

A atividade metabólica do M. tuberculosis sob diversas condições experimentais foi estudada utilizando um sistema radiométrico automático, capaz de quantificar o 14CO2 produzido pela oxidação de substâncias marcadas com carbono-14. As experiências realizadas incluíram: a) vias metabólicas; b) determinação dos tempos de detecção para inoculações de diversas magnitudes; c) efeito da filtração sobre a reprodutibilidade dos resultados; d) influência de meio hostil; e) determinação das concentrações inibitórias mínimas para hidrazida, estreptomicina, etambutol e rifampicina f) tempo de duplicação para o M. tuberculosis e M. bovis. Estes microorganismos metabolizaram até 14CO2 o 14c-formato, (U-14C) acetato, (U-14C)glicerol, ácido palmítico, (1-14C) ácido láurico, ((U-14C) L-ácido málico, (U-14C) D glicose e (1-14C) D-glicose, mas não (1-14C) L-glicose, (U-14C) glicina ou (U-14C) piruvato. Usando 14C-formato, (1-14C) ácido palmítico ou (1-14C) ácido láurico foi possível detectar 10 bacilos/frasco em 24-48 horas e até 10 bacilos/ frasco em 16-20 dias. Resultados reprodutíveis foram obtidos sem filtrar a suspensão de bactérias, desde que cultivadas em meio 7H9 líquido com 0,05% de polissorbato 80 e homogenizadas antes da experiência. Drogas que bloqueiam a síntese protêica apresentaram concentração inibitória mínima menor com o método radiométrico do que com o convencional. O tempo médio de duplicação para o M. bovis e várias cepas de M. tuberculosis com diversas substâncias marcadas foi 9 ± 1 horas.
An in vitro assay system that included automated radiometric quantification of 14CO2 released as a result of oxidation of 14C- substrates was applied for studying the metabolic activity of M. tuberculosis under various experimental conditions. These experiments included the study of a) mtabolic pathways, b) detection times for various inoculum sizes, c) effect of filtration on reproducibility of results, d) influence of stress environment e) minimal inhibitory concentrations for isoniazid, streptomycin, ethambutol and rifampin, and f) generation times of M. tuberculosis and M. bovis. These organisms were found to metabolize 14C-for-mate, (U-14C) acetate, (U-14C) glycerol, (1-14C) palmitic acid, 1-14C) lauric acid, (U-14C) L-malic acid, (U-14C) D-glucose, and (U-14C) D-glucose, but not (1-14C) L-glucose, (U-14C) glycine, or (U-14C) pyruvate to 14CO2. By using either 14C-for-mate, (1-14C) palmitic acid, or (1-14C) lauric acid, 10(7) organisms/vial could be detected within 24 48 hours and as few as 10 organisms/vial within 16-20 days. Reproducible results could be obtained without filtering the bacterial suspension, provided that the organisms were grown in liquid 7H9 medium with 0.05% polysorbate 80 and homogenized prior to the study. Drugs that block protein synthesis were found to have lower minimal inhibitory concentrations with the radiometric method when compared to the conventional agar dilution method. The mean generation time obtained for M. bovis and different strains of M. tuberculosis with various substrates was 9 ± 1 hours.
