Abstract Searching for improvements in semen cryopreservation, natural substances are commonly studied focusing to improve the sperm quality. The aim of this study were evaluated the effect of adding orange, pineapple, and beet juices in different concentrations and combinations to the ram semen cryopreservation extender. Five ejaculates from five adult rams were used. The semen pool was diluted in egg yolk-based extender and mixed with the following 15 treatments (at a final concentration of 400.106 sptz/mL): orange 10% (O10) and 15% (O15); pineapple 10% (P10) and 15% (P15); beet 10% (B10) and 15% (B15); pineapple + orange 10% (PO10) and 15% (PO15); pineapple + beet 10% (PB10) and 15% (PB15); beet + orange 10% (BO10) and 15% (BO15); pineapple + beet + orange 10% (PBO10) and 15% (PBO15); and the control group (CON). Post-thaw in 0.25 mL straws semen quality analysis of cryopreserved semen was performed by CASA and flow cytometry. Analysis of variance (PROC GLM) was carried out and the averages were compared using the SNK test. Pearson's correlation test was also performed. No effect was noted in the addition of juices to the semen extender prior to cryopreservation. Post-thawed, although, statistically similar to the control group, the total motility of the B10 group reached acceptable standards of total motility. In addition, B10 group showed the highest values (p<0.05) of progressive motility than control group or other treatments. The addition of 10% beet juice to the ram semen extender can improve the cryopreservation of sperm motility.

Resumo Em busca de melhorias na criopreservação do sêmen, substâncias naturais são comumente estudadas com o objetivo de melhorar a qualidade do sêmen. O objetivo deste estudo foi avaliar o efeito da adição de sucos de laranja, abacaxi e beterraba em diferentes concentrações e combinações ao diluidor de criopreservação de sêmen ovino. Foram utilizados cinco ejaculados de cinco carneiros adultos. O pool de sêmen foi diluído em diluente à base de gema de ovo e misturado com os seguintes 15 tratamentos (na concentração final de 400x10⁶ sptz/ml): laranja 10% (O10) e 15% (O15); abacaxi 10% (P10) e 15% (P15); beterraba 10% (B10) e 15% (B15); abacaxi + laranja 10% (PO10) e 15% (PO15); abacaxi + beterraba 10% (PB10) e 15% (PB15); beterraba + laranja 10% (BO10) e 15% (BO15); abacaxi + beterraba + laranja 10% (PBO10) e 15% (PBO15); e o grupo controle (CON). Pós-descongelação em palhetas de 0,25 ml a análise da qualidade do sêmen criopreservado foi realizada pelo CASA e citometria de fluxo. A análise de variância foi realizada e as médias comparadas pelo teste SNK. O teste de correlação de Pearson também foi realizado. Nenhum efeito foi observado na adição de sucos ao diluidor de sêmen antes da criopreservação. Após o descongelamento, embora estatisticamente semelhante ao grupo controle, a motilidade total do grupo B10 atingiu padrões aceitáveis de motilidade total. Além disso, o grupo B10 apresentou os maiores valores (p<0,05) de motilidade progressiva que o grupo controle ou os outros tratamentos. A adição de 10% de suco de beterraba ao diluente de sêmen ovino pode melhorar a criopreservação da motilidade espermática.

Abstract Coenzyme Q-10 (CoQ-10) is a cofactor for mitochondrial electron transport chain and may be an alternative to improve sperm quality of cryopreserved equine semen. This work aimed to improve stallion semen quality after freezing by adding CoQ-10 to the cryopreservation protocol. Seven saddle stallions were utilized. Each animal was submitted to five semen collections and freezing procedures. For cryopreservation, each ejaculate was divided in three treatments: 1) Botucrio® diluent (control); 2) 50 μmol CoQ-10 added to Botucrio® diluent; 3) 1 mmol CoQ-10 added to Botucrio® diluent. Semen batches were analyzed for sperm motility characteristics (CASA), plasma and acrosomal membranes integrity and mitochondrial membrane potential (by fluorescence probes propidium iodide, Hoechst 33342, FITC-PSA and JC-1, respectively), alterations in cytoskeletal actin (phalloidin-FITC) and mitochondrial function (diaminobenzidine; DAB). The 1 mmol CoQ-10 treatment presented higher (P<0.05) amount (66.8%) of sperm cells with fully stained midpiece (indicating high mitochondrial activity) and higher (P<0.05) amount (81.6%) of cells without actin reorganization to the post-acrosomal region compared to control group (60.8% and 76.0%, respectively). It was concluded that the addition of 1 mmol CoQ-10 to the freezing diluent was more effective in preserving mitochondria functionality and cytoskeleton of sperm cells submitted to cryopreservation process.

Abstract Establishment of pregnancy after embryo transfer is the ultimate goal of an embryo transfer program and increasing pregnancy rates and reducing pregnancy loss are mandatory. The utilization of treatments to improve conception rates in recipient mares has been the focus of several research groups over the last years and the results are controversial. Some studies using human chorionic gonadotrophin (hCG) found promising results. Our hypothesis was that hCG administration would cause an additional stimulation on luteal function, uterine and luteal vascularization and progesterone concentration, and the mares would have increased uterine and cervix tone. Therefore, in the present study the effects of hCG administration to induce ovulation, on day 0 (day of ovulation) or day 5 postovulation were evaluated on corpus luteum characteristics, reproductive tract vascularization, and serum progesterone concentration from ovulation until day 15 postovulation. Groups were: G1: (control) - no hCG; G2: 2500 IU of hCG to induce ovulation when a follicle greater than 35mm and uterine edema were detected; G3: 2500 IU hCG on day 0; G4: 2500 IU hCG on day 5 postovulation. Twelve mares were randomly assigned to each group, during consecutive cycles, in a Latin Square experimental design, in a total of 48 cycles. Doppler ultrasound evaluations were performed daily from day 0 until day 15 postovulation, including mesometrial vascularity, endometrial vascularity and corpus luteum vascularity. Blood samples were collected for serum progesterone concentration. Data was analyzed using the Proc Glimmix SAS Procedure for nonparametric variables and Proc Mixed for parametric parameters. There was no treatment effect for all variables studied (P > 0.05). Characteristics were only affected by day (P < 0.05). It can be concluded that hCG administration at the time points suggested in the current study did not alter the characteristics evaluated.

The objectives of this study were to validate a non-extractive RIA for seminal testosterone and quantify the hormone using a solid-phase commercial kit, and study the correlation between testosterone in seminal plasma and sperm characteristics. Parallelism showed a correlation index r = 0.992 (Y = -5.47 + 1.073X; R² = 0.985), indicating that the non-extractive method presented is indicated particularly for assessment of testosterone when establishing comparisons between samples. Overall mean (±SD) of testosterone level was 0.60±0.65 ng/mL. Correlation was only found between the seminal concentrations of testosterone and pH of the semen.

Para validar uma técnica prática de avaliação simultânea das membranas plasmática, acrossomal e mitocondrial em espermatozóides eqüinos três sondas fluorescentes (PI, FITC-PSA e MITO) foram associadas. Quatro ejaculados de três garanhões (n=12) foram diluídos em meio TALP e divididos em duas alíquotas, uma alíquota foi submetida a flash frozen em nitrogênio líquido para induzir danos nas membranas celulares. Três tratamentos foram preparados com as seguintes proporções de sêmen fresco: sêmen flash frozen: 100:0 (T100), 50:50 (T50), e 0:100 (T0). Uma amostra de 150 µL de sêmen diluído de cada tratamento foi adicionada de 2 µL de PI, 2 µL de MITO e 80 µL de FITC-PSA; incubadas à 38,5ºC/8 min, e as células espermáticas foram avaliadas por microscopia de epifluorescência. Baseados na análise de regressão esta é uma técnica eficiente e prática para determinar danos em espermatozóides eqüinos, capaz de determinar a porcentagem de espermatozóides mais representativa do potencial fertilizante do ovócito.

To validate a practical technique of simultaneous evaluation of the plasma, acrosomal and mitochondrial membranes in equine spermatozoa three fluorescent probes (PI, FITC-PSA and MITO) were associated. Four ejaculates from three stallions (n=12) were diluted in TALP medium and split into 2 aliquots, 1 aliquot was flash frozen in liquid nitrogen to induce damage in cellular membranes. Three treatments were prepared with the following fixed ratios of fresh semen: flash frozen semen: 100:0 (T100), 50:50 (T50), and 0:100 (T0). A 150-µL aliquot of diluted semen of each treatment was added of 2 µL of PI, 2 µL of MITO and 80 µL of FITC-PSA; incubated at 38.5ºC/8 min, and sperm cells were evaluated by epifluorescent microscopy. Based in regression analysis, this could be an efficient and practical technique to assess damage in equine spermatozoa, as it was able to determine the sperm percentage more representative of the potential to fertilize the oocyte.

A seleção de galos para a reprodução pelo desenvolvimento da crista vem sendo empregada rotineiramente na avicultura, sendo necessários estudos para verificar a eficiência deste método de seleção. Este trabalho teve por objetivo comparar as características seminais de galos selecionados para a reprodução pelo desenvolvimento da crista. Foram utilizados 65 galos matrizes da linhagem AgRoss, selecionados na 20ª semana de idade e divididos em dois grupos: grupo A, animais sem crista desenvolvida (n=33), e grupo B, animais com crista desenvolvida (n=32). Foram realizadas colheitas semanais de sêmen dos galos de 24 a 71 semanas de idade, para avaliar as características seminais. Foram realizados os testes t de Student e de Wilcoxon para comparar os grupos e análises de perfis para verificar efeitos do grupo, idade e interação grupo x idade. Os valores de volume seminal, concentração espermática, motilidade e vigor foram maiores (p < 0,01) para o grupo B no período de 24-31 semanas de idade. A percentagem de defeitos espermáticos para o grupo A foi maior nos períodos de 24-27 (p < 0,01) e de 32-35 semanas de idade (p < 0,05). O peso corporal foi maior (p < 0,05) para os animais do grupo B no período entre 24-39 semanas de idade e para os galos do grupo A no período de 60-71 semanas de idade. Foram observados efeitos da idade e interação grupo x idade para as características seminais, exceto para vigor. Em conclusão, a observação do desenvolvimento da crista na 20ª semana de idade é um método eficiente na seleção de galos para a reprodução.

The selection of cocks for reproduction by comb development has been employed usually in the aviculture; however, studies have been necessary to establish the efficiency of this method of selection. The objective of this paper was to compare the seminal characteristics in cocks selected to reproduction by comb development. Sixty-five broiler breeder males (AgRoss strain) were selected to breed at 20th week old and divided in two groups, A: birds without comb (n=33) and B: birds with comb (n=32). To evaluate seminal characteristics, semen was collected weekly between 24 to 71 weeks of age. Group means were compared by the Student's t and Wilcoxon tests while profile analysis were performed for to verify effects of group, age, and group x age. Seminal volume, sperm concentration, motility and vigor were greater (p < 0.01) for group B from 24-31 weeks of age. Percentage of sperm morphological defects was greater in group A during 24-27 (p < 0.01) and 32-35 (p < 0.05) weeks of age. Mean body weight was higher for group B between 24-39 weeks of age (p < 0.05) and for group A between 60-71 weeks of age. There were no main effects of group for seminal characteristics or body weight (p > 0.05), but effects of age (p < 0.001) and group x age interaction were significant (p < 0.05), except for vigor (p > 0.05). Low or no correlations were observed between seminal characteristics and body weight. In conclusion, the method to select cocks by observation of comb development at 20 weeks of age showed efficient.

O corpo lúteo (CL) é a glândula produtora de progesterona (P4), hormônio cuja secreção contínua é essencial para o início e a manutenção da gestação em fêmeas eqüinas, e, conseqüentemente, para a aplicabilidade de inúmeras biotécnicas de reprodução. Considerando-se a importância do CL para a manutenção de uma gestação normal e suas características anatomofisiológicas, objetivou-se determinar por ultra-sonografia (US) o tamanho e a morfoecogenicidade (ME) do CL em receptoras de embriões eqüinos desde a ovulação (D0) até nove dias após (D9), bem como os níveis plasmáticos de P4 produzida no mesmo período. Para tanto, 57 éguas receptoras de um programa de transferência de embriões foram examinadas diariamente por US transretal desde a primeira detecção dos sinais de estro até o D9. A cada exame, os CL foram mensurados e sua ME registrada segundo escore de 1 a 6 (1=anecóico; 6=hiperecóico). Amostras de sangue foram colhidas diariamente e a P4 dosada por radioimunoensaio. O diagnóstico de gestação foi realizado por US aos 13 e 25 dias após a ovulação. Houve uma tendência de os corpos lúteos apresentarem ME crescente (de 1 a 5) desde o dia da ovulação até o D9. Os níveis de P4 foram < 2,16 ng/ml até o D3, com conseqüente elevação e manutenção em níveis de diestro entre D4 e D9 (3,41 a 4,33 ng/ml). O tamanho luteínico não diferiu, com exceção das médias extremas durante o período (D2 = 31,54 mm versus D8 = 25,95mm; p < 0,05). Assim, o aumento da ME média dos CLs avaliados por US é acompanhado por aumento na concentração plasmática de P4 em receptoras de embriões, mas este evento parece não ser dependente do tamanho da glândula luteínica. Não existe diferença na ME, no tamanho dos corpos lúteos, nem nos níveis de P4 circulante do D0 ao D9 em receptoras de embriões eqüinos que se tornaram gestantes ou não após a transferência de embriões.

Corpus luteum (CL) synthesizes progesterone (P4), which has a major function in maintenance of pregnancy in equine females and also enables the application of biotechnologies of reproduction. Considering the importance of the CL and its anatomical and physiological features to achieve normal pregnancy, our aims were to determine the size and morphoechogenicity (ME), as well as plasma P4 concentrations of corpus luteum from recipient mares for nine days after ovulation (D0). Therefore, 57 recipient mares were examined daily by transrectal ultrasonography (US) from early signals of estrus to D9. CLs were measured and their ME classified according to a 1 to 6 scale (1=anechogenic; 6=hiperechogenic). Blood samples were collected daily and progesterone concentrations assessed by radioimmunoassay. Pregnancies were checked by US 13 and 25 days after ovulation. Corpus luteum echogenicity had a tendency to increase from D0 to D9. Progesterone concentrations were < 2,16 ng/ml until D3, but there was a significant elevation from D4 to D9 (3,41 to 4,33 ng/ml). There were no differences in CL size, except between D2 (31,54 mm) and D8 (25,95 mm); p < 0,05). Thus, an increase in mean luteal ME is accompanied by an increase in plasmatic P4 concentration, but this event seems independent of luteal size. There were no differences between ME, size and P4 levels from D0 to D9 in recipient mares that became pregnant or not after embryo transfer.

Muitos fatores podem afetar as taxas de gestação após a transferência de embriões em eqüinos. Este estudo teve como objetivo avaliar, em eqüinos da raça Mangalarga e em duas estações reprodutivas, a influência do garanhão e de três diferentes técnicas de inseminação (monta natural, inseminação artificial com sêmen diluído fresco ou resfriado) sobre os índices de recuperação de embriões e de prenhez. No primeiro ano, foram encontradas diferenças significativas (p < 0,05) na recuperação de embriões entre cinco garanhões estudados (28,6; 65,0; 62,9; 66,7 e 84,2%), mas não foram encontradas diferenças significativas (p > 0,05) nos índices de prenhez (50,0; 80,7; 70,6; 70,0 e 62,5%). No segundo ano, verificou-se uma tendência à significância (p = 0,057), na recuperação de embriões entre quatro garanhões estudados (44,4; 56,2; 71,4 e 73,7%) e não houve diferença significativa (p > 0,05) nos índices de prenhez (75,0; 55,6; 86,7 e 71,4%). Não houve diferença significativa (p > 0,05) nos índices de embriões recuperados e de prenhez comparando-se a monta natural (50,0 e 87,5%) e a inseminação artificial com sêmen fresco (60,6 e 73,7%) ou refrigerado (63,8 e 80,3%).

Many factors can affect pregnancy rates after equine embryo transfer. This study had the objective of evaluating the influence of stallion and of three different insemination techniques (natural breed, artificial insemination with fresh or cold semen), on pregnancy and on embryos recovery rates. In the first year, differences were observed (p < 0.05) in embryo recovery rates among the five stallions studied (28.6, 65.0, 62.9, 66.7 and 84.2%), but no statistical differences (p > 0.05) were detected in pregnancy rates (50.0, 80.7, 70.6, 70.0 and 62.5%). In the second year, it was observed a tendency to statistical significance (p = 0.057) in embryos recovery rates between the stallions (44.4, 56.2, 71.4 and 73.7%), and no statistical differences (p > 0.05) were detected in pregnancy rates (75.0, 55.6, 86.7 and 71.4%). There were no statistical difference (p > 0.05) in the number of embryos transferred and in pregnancy rates comparing natural breed (50.0 and 87.5%) and artificial insemination with fresh (60.6 and 73.7%) or cold semen (63.8 and 80.3%).

Os resultados das taxas de fertilidade e dos tamanhos das leitegadas foram analisados na granja de suínos da Faculdade de Medicina Veterinária e Zootecnia da Universidade de São Paulo no Campus de Pirassununga, durante o período de 4 anos (1992 a 1995), com o objetivo de comparar a inseminação artificial (IA) e a cobertura natural (CN) em diferentes épocas do ano, procurando estudar as influências sazonais sobre os índices reprodutivos. Foram avaliadas 799 coberturas em fêmeas das raças Landrace (L), Large White (LW) e mestiças, sendo 539 de IA e 260 de CN. Reprodutores (L e LW) de comprovada fertilidade foram utilizados tanto para CN quanto para IA. As doses de sêmen apresentavam concentração mínima de 3 bilhões de espermatozóides em volume de 100 ml. As IA foram realizadas às 12 e às 24 horas após reflexo positivo de tolerância ao macho, enquanto as montas foram realizadas no momento e às 24 horas após o diagnóstico do cio. Os índices de fertilidade foram de 72,9% e 75,8% e o número de leitões nascidos de 12,4 e 12,1, respectivamente, para inseminação artificial e monta natural, não mostrando diferença significativa. A estação climática influenciou a taxa de parição (71,2%, 81,4%, 76,9% e 66,4%, para verão, outono, inverno e primavera, respectivamente; p < 0,05), mas não mostrou efeito sobre o tamanho da leitegada. Não houve influência do tipo de cobertura (IA ou CN) e da interação entre estação climática e tipo de cobertura sobre as taxas de parições e os tamanhos das leitegadas. As médias de temperatura máxima e mínima se correlacionaram negativamente com a taxa de parição.

The data of parturition rate and litter size from the herd of the Veterinary School of São Paulo University were analysed during a four years period (1992 to 1995) in order to compare the data from artificial insemination (IA) and natural mating (CN) in different seasons, trying to study the seasonal influences and define which of these methods would be better in order to reach adequate reproductive rates. Data from 799 breeding (539 IA and 260 CN) in Landrace (L), Large White (LW) females and the cross-breed from both were analysed. Boars (L and LW) with known fertility were used either for IA or for CN. The semen doses used had at least 3 billions spermatozoa in 100 ml. The inseminations were performed 12 and 24 hours after the positive back pressure test in response to the boar, and natural mating were performed at the moment of positive back pressure test and 24 hours later. The fertility rates were 72.9% and 75.8% and the litter size was 12.4 and 12.1, for artificial insemination and natural mating, respectively, which were not different. The season influenced the parturition rate (71.2%, 81.4%, 76.9% and 66.4%, for summer, autumn, winter and spring, respectively; p < 0.05) but not the litter size.

O objetivo deste trabalho foi avaliar a influência do folículo dominante nas fases de crescimento, estática e de regressão sobre a resposta superovulatória em 18 ciclos estrais de 9 novilhas da raça Nelore. Avaliaram-se os ovários entre o dia 6 e o dia 10 do ciclo estral pela técnica de ultra-sonografia, medindo-se o diâmetro do maior folículo e contando-se o número de folículos subordinados. Os animais foram superovulados com 400 ou 500 UI de FSH/LH, iniciando-se no dia 10 do ciclo estral, em subdoses decrescentes, por 4 dias consecutivos. Aplicou-se PGF2alfa concomitante com a quinta subdose de FSH/LH e realizaram-se inseminações artificiais às l2 horas e às 24 horas após o início dos sintomas de estro. Os embriões foram colhidos no dia 6,5 após a primeira inseminação artificial, obtendo-se 3,00; 3,83 e 10,17 embriões, respectivamente, nas fases de crescimento, estática e regressão. Observaram-se melhores resultados quanto ao número de embriões quando o folículo dominante se encontrava em fase de regressão.

The aim of this study was to evaluate the influence of the dominant follicle during it’s growing, static and regression phases on the superovulatory response in 18 estral cycles of nelore heifers. The follicular development was monitored by ultrasound from Day 6 to Day 10 (Day 0=estrus) of the oestrus cycle, when the diameter of the largest follicle was measured and the number of subordinate follicles counted. The animals where superovulated with 400 or 500IU of FSH/LH twice daily for 4 days begining on day 10 of the oestrus cycle I was injected PGF2alpha concomitantly with the fifth dose of FSH/LH. Artificial insemination was done 12 and 24 hours after the begining of the estrus. Embryos were recovered on day 6.5 after the first insemination. If a dominant follicle was present before superovulation, it was observed subordinate follicles atretic and low response to superovulatory treatment. The best result of transferable and total embryos was observed when the dominant follicle was in regression phase (3.67 and 10.17) at the beginning of the superovulatory treatment.

Este trabalho teve como objetivo identificar a dose mais eficiente de FSH/LH (300, 400 e 500 UI) no tratamento superovulatório de novilhas da raça Nelore, assim como avaliar o método one-step no processo de congelação de embriões. A variação da resposta superovulatória tem sido muito grande, o que explica o interesse de diversos pesquisadores em encontrar novos hormônios, doses e momentos para realizar a estimulação ovariana. Foram empregadas doses de 300 (n = 20), 400 (n = 21) ou 500 UI (n = 21) de FSH/LH, iniciando-se no décimo dia do ciclo estral, em 8 aplicações decrescentes, durante quatro dias consecutivos. Foi aplicado PGF2alfa concomitante com a quinta subdose de FSH/LH e realizadas duas inseminações artificiais às 12 e às 24 horas após o início dos sintomas de estro. As colheitas dos embriões foram realizadas 6,5 dias após a primeira inseminação artificial. Pelo exame ultra-sonográfico, avaliaram-se os números de folículos no momento da inseminação artificial (15,12; 15,76; e 14,94) e de corpos lúteos (10,68; 11,55; e 10,81) no dia da colheita, encontrando 5,20; 1,81; e 2,76 embriões viáveis, respectivamente, para 300 UI, 400 UI e 500 UI de FSH/LH. O grupo de 300 UI de FSH/LH apresentou os melhores resultados em relação aos embriões viáveis. Dos 106 embriões congelados pelo método one-step em 1,5 M de etilenoglicol e transferidos pelo método não-cirúrgico, 8 resultaram em prenhez (7,5%). A dose de 300 UI de FSH/LH apresentou melhor resposta superovulatória em comparação com as de 400 e 500 UI para novilhas da raça Nelore. A transferência de embriões Bos taurus indicus congelados pelo método one-step em 1,5 M de etilenoglicol não foi eficiente.

The objetive of this study was to identify the better dose between 300 (n = 20), 400 (n = 21) and 500 IU (n = 21) of FSH/LH to stimulate Nelore heifers. The superovulation treatment started on day 10 (D0 = estrous) of the estrous cycle in 8 decreasing aplications for 4 days. The embryo recovery was achieved on day 6.5 after the first artificial insemination. The superovulatory response for 300, 400 and 500 IU FSH/LH was follicles (15.12, 15.76 and 14.94); corpus luteum (10.68, 11.55 and 10.81) and transferable embryos (5.20, 1.81 and 2.76). The 300 IU of FSH/LH group presented the best results in regard to transferable embryos. The transferable embryos were cryopreserved by one-step method with 1.5 M of ethylene-glycol, resulting in 8 pregnancies (7.5%) of 106 embryos transferred by non-curgical method. The 300 IU of FSH/LH presented better superovulatory response in comparison with 400 and 500 IU in Nelore heifers. The transfer of Bos taurus indicus embryos cryopreserved by one-step method in 1.5 M of ethilene glycol was not efficient.
