ABSTRACT Ecological restoration mitigates the negative effects of fragmentation and habitat loss. This practice allows the conservation of key species, such as Ocotea porosa, a tree native to the Araucaria Forest and extremely endangered. A key point in restoration projects is the source of seeds, as well as guidelines for collection. When carried out under technical criteria, the collection allows the maintenance of genetic diversity and adaptive potential in restoration plantations. Given the importance of seed source, genetic diversity, and adaptive potential, this study aimed to define areas and criteria for collecting seeds by characterizing the demography, genetics, and reproductive phenology of an O. porosa population. A plot of 16 hectares was installed in the municipality of Passos Maia, Santa Catarina, Brazil, and a demographic survey of trees with diameter at breast height (DBH) > 15 cm was carried out. Indices of diversity and internal genetic structure (IGS) were estimated using allozyme markers. The reproductive phenology of 67 individuals was evaluated during 8 months. The studied population showed a high density of individuals (10.7 ind. ha-1) with normal diametric distribution. The phenological pattern of the species is regular, seasonal, and annual. The evaluated population showed high genetic diversity, high fixation index, and significant IGS up to 80 meters away. Based on these results, the evaluated fragment can be used as a seed collection area. It has high genetic diversity, density, and area size sufficient to contain several demes. In addition, it is highly recommended that the matrices be at least 80 meters apart to avoid the effects of significant IGS.

Abstract: Few twinning events have been recorded in the West Indian manatee (Trichechus manatus, Sirenia: Trichechidae) and no previous published study has provided confirmation of this phenomenon based in molecular tools. Here we investigate a possible case of twinning in an endangered Brazilian population of T. manatus using molecular tools. We analyzed two male neonates found stranded in Ceará State, on the northeastern coast of Brazil. The DNA of both individuals was isolated, and 10 microsatellite loci were amplified and genotyped. Following the identification of the alleles, the probabilities of identity by descent (∆7 and ∆8) and relatedness (rxy) were calculated using estimators that evaluate inbreeding. The two individuals shared most of the alleles, with differences in the genotypes being identified in only two loci. All the estimators identified a level of relatedness compatible with that found between siblings (selfed or outbred), indicating they were dizygotic twins. This is the first confirmed case of fraternal twins in free-ranging West Indian manatees in South America. The recognition of this type of twinning provides elements to improve actions for the rehabilitation of stranded animals and their subsequent release to the environment.

Abstract The objective of the study was to compare the richness and diversity of ant assemblages in an agroecological system under peach orchard, conventional system under peach orchard cultivation and native vegetation in rural properties located in a Pampa Biome. The study was conducted in four samplings in 2017: 1st and 09th March (summer); 24th and 31st July (winter); and four samplings in 2018: 23rd and 30th January (summer); 31st July and 07th August (winter). Pitfall traps were used. The assemblages were characterized and compared using richness, number of occurrences of ants, Shannon diversity (H’), equitability, rarefaction analysis and Chao 1. The association of the species with the samples was evaluated by a Principal Component Analysis (PCA). The agroecological system had the highest number of occurrences, while the conventional orchard the lowest number. Richness and abundance were greatest during the summer. The conventional peach orchard obtained the lowest H’ for both seasons when compared to the agroecological orchard and native vegetation. The PCA explained 77.40% of the occurrence of ants in the environments and in the seasons. The results found demonstrated that conservationist systems tend to harbor greater wealth and diversity of ant assemblages, as well as occurring in native áreas.

OBJECTIVES: To prospectively evaluate demographic, anthropometric and health-related quality of life (HRQoL) in pediatric patients with laboratory-confirmed coronavirus disease 2019 (COVID-19) METHODS: This was a longitudinal observational study of surviving pediatric post-COVID-19 patients (n=53) and pediatric subjects without laboratory-confirmed COVID-19 included as controls (n=52) was performed. RESULTS: The median duration between COVID-19 diagnosis (n=53) and follow-up was 4.4 months (0.8-10.7). Twenty-three of 53 (43%) patients reported at least one persistent symptom at the longitudinal follow-up visit and 12/53 (23%) had long COVID-19, with at least one symptom lasting for >12 weeks. The most frequently reported symptoms at the longitudinal follow-up visit were headache (19%), severe recurrent headache (9%), tiredness (9%), dyspnea (8%), and concentration difficulty (4%). At the longitudinal follow-up visit, the frequencies of anemia (11% versus 0%, p=0.030), lymphopenia (42% versus 18%, p=0.020), C-reactive protein level of >30 mg/L (35% versus 0%, p=0.0001), and D-dimer level of >1000 ng/mL (43% versus 6%, p=0.0004) significantly reduced compared with baseline values. Chest X-ray abnormalities (11% versus 2%, p=0.178) and cardiac alterations on echocardiogram (33% versus 22%, p=0.462) were similar at both visits. Comparison of characteristic data between patients with COVID-19 at the longitudinal follow-up visit and controls showed similar age (p=0.962), proportion of male sex (p=0.907), ethnicity (p=0.566), family minimum monthly wage (p=0.664), body mass index (p=0.601), and pediatric pre-existing chronic conditions (p=1.000). The Pediatric Quality of Live Inventory 4.0 scores, median physical score (69 [0-100] versus 81 [34-100], p=0.012), and school score (60 [15-100] versus 70 [15-95], p=0.028) were significantly lower in pediatric patients with COVID-19 at the longitudinal follow-up visit than in controls. CONCLUSIONS: Pediatric patients with COVID-19 showed a longitudinal impact on HRQoL parameters, particularly in physical/school domains, reinforcing the need for a prospective multidisciplinary approach for these patients. These data highlight the importance of closer monitoring of children and adolescents by the clinical team after COVID-19.

ABSTRACT: Foot diseases are the main causes of lameness in small ruminants, causing great impact on animal welfare and important economic losses due to depreciation and culling of affected animals. The aim of the present study was to characterize foot diseases found in eight flocks of sheep with a history of the ailment, that belonged to eight municipalities in the state of Bahia. At each visit, inspection of the flock, survey questionnaire, and characterization of diseases were performed. Altogether 600 sheep were inspected through foot examination in sick animals to characterize the injuries and record the diseases present per animal, as affected hooves and the number of affected limbs. The occurrence of foot disease was 23.3% (140/600) ranging from 12.8 to 55.5% per farm. Two hundred and twenty cases involving nine diseases were recorded. Foot rot was the most common disease with 51.3% of cases (113/220), 70% corresponding to advanced and terminal stages. Interdigital dermatitis represented 27.7% of the cases, excessive hoof growth 8.6%, white line disease 3.2%, double soles 3.2%, toe granuloma 2.3%, interdigital hyperplasia 2.3%, sole ulcers 0.9% and 0.4% foot abscess. The main predisposing factors involved in the genesis of diseases were the high rainfall precipitation, soil moisture conditions, pens and pastures, poor sanitary management and lack of preventive measures (footbath, quarantine, isolation and vaccination), treatment failures and low infrastructure of most properties.

ABSTRACT The objective of this study was to determine the sample size necessary to estimate the mean and coefficient of variation in four species of crotalarias (C. juncea, C. spectabilis, C. breviflora and C. ochroleuca). An experiment was carried out for each species during the season 2014/15. At harvest, 1,000 pods of each species were randomly collected. In each pod were measured: mass of pod with and without seeds, length, width and height of pods, number and mass of seeds per pod, and mass of hundred seeds. Measures of central tendency, variability and distribution were calculated, and the normality was verified. The sample size necessary to estimate the mean and coefficient of variation with amplitudes of the confidence interval of 95% (ACI95%) of 2%, 4%, ..., 20% was determined by resampling with replacement. The sample size varies among species and characters, being necessary a larger sample size to estimate the mean in relation of the necessary for the coefficient of variation.

Abstract Objective: To evaluate prevalence, clinical manifestations, laboratory abnormalities and treatment in a multicenter cohort study including 847 childhood-onset systemic lupus erythematosus (cSLE) patients with and without diffuse alveolar hemorrhage (DAH), as well as concomitant parameters of severity. Methods: DAH was defined as the presence of at least three respiratory symptoms/signs associated with diffuse interstitial/alveolar infiltrates on chest x-ray or high-resolution computer tomography and sudden drop in hemoglobin levels. Statistical analysis was performed using Bonferroni correction (p < 0.0022). Results: DAH was observed in 19/847 (2.2%) cSLE patients. Cough/dyspnea/tachycardia/hypoxemia occurred in all cSLE patients with DAH. Concomitant parameters of severity observed were: mechanical ventilation in 14/19 (74%), hemoptysis 12/19 (63%), macrophage activation syndrome 2/19 (10%) and death 9/19 (47%). Further analysis of cSLE patients at DAH diagnosis compared to 76 cSLE control patients without DAH with same disease duration [3 (1–151) vs. 4 (1–151) months, p = 0.335], showed higher frequencies of constitutional involvement (74% vs. 10%, p < 0.0001), serositis (63% vs. 6%, p < 0.0001) and sepsis (53% vs. 9%, p < 0.0001) in the DAH group. The median of disease activity score(SLEDAI-2 K) was significantly higher in cSLE patients with DAH [18 (5–40) vs. 6 (0–44), p < 0.0001]. The frequencies of thrombocytopenia (53% vs. 12%, p < 0.0001), intravenous methylprednisolone (95% vs. 16%, p < 0.0001) and intravenous cyclophosphamide (47% vs. 8%, p < 0.0001) were also significantly higher in DAH patients. Conclusions: This was the first study to demonstrate that DAH, although not a disease activity score descriptor, occurred in the context of significant moderate/severe cSLE flare. Importantly, we identified that this condition was associated with serious disease flare complicated by sepsis with high mortality rate.

ABSTRACT The goal of this study was to estimate the leaf area of Crotalaria juncea according to the linear dimensions of leaves from different ages. Two experiments were conducted with C. juncea cultivar IAC-KR1, in the 2014/2015 sowing seasons. At 59, 82, 102, 129 days after sowing (DAS) of the first and 61, 80, 92, 104 DAS of the second experiment, 500 leaves were collected, totaling 4,000 leaves. In each leaf, the linear dimensions were measured (length, width, length/width ratio and length × width product) and the specific leaf area was determined through Digimizer and Sigma Scan Pro software, after scanning images. Then, 3,200 leaves were randomly separated to generate mathematical models of leaf area (Y) in function of linear dimension (x), and 800 leaves for the models validation. In C. juncea, the leaf areas determined by Digimizer and Sigma Scan Pro software are identical. The estimation models of leaf area as a function of length × width product showed superior adjustments to those obtained based on the evaluation of only one linear dimension. The linear model Ŷ=0.7390x (R2=0.9849) of the real leaf area (Y) as a function of length × width product (x) is adequate to estimate the C. juncea leaf area.

Abstract Streptococcus agalactiae is one of the most common pathogens leading to mastitis in dairy herds worldwide; consequently, the pathogen causes major economic losses for affected farmers. In this study, multilocus sequence typing (MLST), genotypic capsular typing by multiplex polymerase chain reaction (PCR), and virulence gene detection were performed to address the molecular epidemiology of 59 bovine (mastitis) S. agalactiae isolates from 36 dairy farms located in the largest milk-producing mesoregions in Brazil (Minas Gerais, São Paulo, Paraná, and Pernambuco). We screened for the virulence genes bac, bca, bibA, cfb, hylB, fbsA, fbsB, PI-1, PI-2a, and PI-2b, which are associated with adhesion, invasion, tissue damage, and/or immune evasion. Furthermore, five capsular types were identified (Ia, Ib, II, III, and IV), and a few isolates were classified as non-typeable (NT). MLST revealed the following eight sequence types (STs): ST-61, ST-67, ST-103, ST-146, ST-226, ST-314, and ST-570, which were clustered in five clonal complexes (CC64, CC67, CC103, CC17, and CC314), and one singleton, ST-91. Among the virulence genes screened in this study, PI-2b, fbsB, cfb, and hylB appear to be the most important during mastitis development in cattle. Collectively, these results establish the molecular epidemiology of S. agalactiae isolated from cows in Brazilian herds. We believe that the data presented here provide a foundation for future research aimed at developing and implementing new preventative and treatment options for mastitis caused by S. agalactiae.

ABSTRACT Grasses and legumes are two groups of plants physic and chemically different (amount of crude protein and fiber, mostly), very used in animal feed, whose differences can determine variation in intake, digestibility and animal behavior. We aimed to evaluate performance, feeding behavior and the serum biochemical profile of sheep. The treatments were four levels of substitution of Bermuda grass (Cynodon dactylon [L. ] Pers.) hay for alfalfa (Medicago sativa L.) hay (0, 330, 660 and 1000 g kg-1 DM). The randomized block design was used being two blocks and four treatments with 10 replicates). There were no effects on the performance of animals. On the other hand, there was a positive linear effect (P < 0.05) for intake of DM, organic matter, crude protein (CP), total carbohydrates, and non-fibrous carbohydrates. However, the use of neutral detergent fiber (NDF) showed linear decrease with the increase in alfalfa hay (P < 0.05). CP and NDF decreased linearly (P < 0.05) with the replacement of Bermuda grass hay for alfalfa hay. There were decreasing linear effects (P < 0.05) when compared to the time of ingestion and chewing of feed, and increasing linear effects regarding total resting time. Estimated passage rate, density of particles and blood glucose showed a positive linear effect (P < 0.05), while weight of gastrointestinal contents decreased according to alfalfa levels (P < 0.05). The replacement of Bermuda grass hay by alfalfa hay to promote improvement in the performance is not recommended.

Abstract An updated inventory of Brazilian seed plants is presented and offers important insights into the country's biodiversity. This work started in 2010, with the publication of the Plants and Fungi Catalogue, and has been updated since by more than 430 specialists working online. Brazil is home to 32,086 native Angiosperms and 23 native Gymnosperms, showing an increase of 3% in its species richness in relation to 2010. The Amazon Rainforest is the richest Brazilian biome for Gymnosperms, while the Atlantic Rainforest is the richest one for Angiosperms. There was a considerable increment in the number of species and endemism rates for biomes, except for the Amazon that showed a decrease of 2.5% of recorded endemics. However, well over half of Brazillian seed plant species (57.4%) is endemic to this territory. The proportion of life-forms varies among different biomes: trees are more expressive in the Amazon and Atlantic Rainforest biomes while herbs predominate in the Pampa, and lianas are more expressive in the Amazon, Atlantic Rainforest, and Pantanal. This compilation serves not only to quantify Brazilian biodiversity, but also to highlight areas where there information is lacking and to provide a framework for the challenge faced in conserving Brazil's unique and diverse flora.

The objective of the study was to isolate, cultivate and characterize equine peripheral blood-derived multipotent mesenchymal stromal cells (PbMSCs). Peripheral blood was collected, followed by the isolation of mononuclear cells using density gradient reagents, and the cultivation of adherent cells. Monoclonal mouse anti-horse CD13, mouse anti-horse CD44, and mouse anti-rat CD90 antibodies were used for the immunophenotypic characterization of the surface of the PbMSCs. These cells were also cultured in specific media for adipogenic and chondrogenic differentiation. There was no expression of the CD13 marker, but CD44 and CD90 were expressed in all of the passages tested. After 14 days of cell differentiation into adipocytes, lipid droplets were observed upon Oil Red O (ORO) staining. Twenty-one days after chondrogenic differentiation, the cells were stained with Alcian Blue. Although the technique for the isolation of these cells requires improvement, the present study demonstrates the partial characterization of PbMSCs, classifying them as a promising type of progenitor cells for use in equine cell therapy.

The predatory capacity of one isolate of nematode-trapping fungus Duddingtonia flagrans (AC001) on infective larvae of cyathostomes was evaluated in laboratorial conditions in medium water-agar 2% (WA 2%). There was significant reduction (p < 0. 01) of 93.64% in the average of infective larvae of cyathostomes recovered of medium WA 2% at seven day. These results show that the isolated AC001 could be used in the biological control of cyathostomes of horses.

It was studied the effect of Xylella fastidiosa infection, causal agent of Citrus Variegated Chlorosis (CVC), on the xylem sap ingestion rate of citrus plants by two sharpshooters species (Hemiptera: Cicadellidae). Seedlings of sweet orange Pera and Valencia (Citrus sinensis) were used and infected by X. fastidiosa, strain 9a5c, obtained by mechanical inoculation. The insects used in the experiments were collected in the field, one from Cicadellini Tribe (Dilobopterus costalimai) and another from Proconiini Tribe (Oncometopia facialis). The xylem sap ingestion rate by O. facialis was quantified in seedling twigs, and by D. costalimai in leaves and twigs, by means of evaluation of the liquid volume (honeydew) excreted by time unity. The consumption by O. facialis sharpshooter in diseased plants was inferior to healthy plants. In the diseased Pera variety the consumption was lower, not permitting a xylem sap quantification. In healthy Pera and healthy and diseased Valencia, O. facialis showed expressive rate of consumption, with high feeding during the day. In the healthy plants of both varieties, the consumption by D. costalimai sharpshooter was higher than in CVC diseased plants. Comparing both varieties, the rate was higher in the Valencia variety, and concerning parts of the plant, leaves and twigs, the ingestion rate was higher in the twigs of both varieties, with higher consumption during the day.

The objective of this research was to know whether the consequences of practicing professions such as lawyer and musician are relevant to obtain motivational aims inherent to the individual’s axiological priorities. Schwartz’s theory of values which emphasizes the motivational bases of values has been used. A sample of 122 subjects, 71 lawyers and 51 professional musicians, was given Schwartz’s Inventory of Values. The average service time in a professional area was 9.3 years (SD = 9.42). A 2x2 Anova (sex and profession) was calculated according to each of the ten motivational types of values and to each of the four second order factors. The musicians, when compared to the lawyers, gave more importance to hedonism and stimulation. The lawyers emphasized more than the musicians three motivational types of values: conformity, security, and power. The musicians’ profile was marked by the search for changes, new sensations and pleasures, which constitute the pole for individualism, for intellectual and emotional autonomy, and for the propensity for change. In the opposite pole of this dimension is the tendency to conservatism, to collectivism, to preserve the status quo, which characterizes the lawyers’ profile. So, these two professions are based on opposite motivations: on one hand, the tendency to create, to innovate, and to seek for new ways of thinking and feeling, even risking mistakes and, on the other hand, the motivation to conserve and to respect society’s rules, traditions, and uses.
