Resumo Com aproximadamente 1800 indivíduos, distribuídos em quatro aldeias, o povo Maxakali é a segunda maior população indígena aldeada no Estado de Minas Gerais. A avaliação da qualidade da água utilizada para consumo e lazer nesta população geralmente é realizada por meio de critérios de palatabilidade e visuais. Dessa forma, realizou-se um estudo descritivo incluindo amostras de água superficial e subterrânea consumida em quatro aldeias, considerando a coleta em três períodos sazonais durante o ano de 2015. Foram mensurados o pH (potencial de hidrogeniônico), turbidez, concentração de oxigênio dissolvido, condutividade, nitrato, coliformes totais e termotolerantes. As aldeias com maior número de amostras com valores superiores aos toleráveis foram a Aldeia Verde (100%), seguido da Aldeia Água Boa (85,7%) e Pradinho (71,4%). O oxigênio dissolvido e os coliformes totais e termotolerantes estiveram alterados em todas as aldeias, com percentuais superiores a 50% das amostras. A turbidez e a condutividade tiveram alterações detectadas em três das quatro aldeias analisadas. Assim, a água consumida por essa comunidade, in natura, conforme a tradição local, apresenta elevado risco para a ocorrência de doenças de veiculação hídrica nesse grupo populacional.

Abstract Distributed in four villages with approximately 1800 individuals, the Maxakali people have the second largest indigenous population in the state of Minas Gerais, Brazil. This community’s water quality is usually evaluated according to its palatability, and whether it meets certain visual criteria. Thus, a descriptive study was carried out including samples of surface and groundwater consumed in four villages. One survey was completed in each of the three seasonal periods in 2015. Hydrogen potential (Ph), turbidity, dissolved oxygen concentration, conductivity, nitrate, and total and thermotolerant coliforms were measured. The villages with the largest number of samples with values higher than tolerable were Verde Village (100%), followed by Água Boa Village (85.7%) and Pradinho (71.4%). All villages sampled showed higher-than-acceptable levels of dissolved oxygen, and total and fecal coliforms in more than 50% of the samples. The turbidity and conductivity changes were detected in three of the four villages. Thus, the water consumed by this community, untreated as per local tradition, presents a high risk for the occurrence of waterborne diseases in this population group.

Resumo O povo Maxakali é a segunda maior população indígena aldeada no Estado de Minas Gerais, Brasil, tendo nas doenças infecciosas e parasitárias, sua principal causa de morte. Problemas com a qualidade da água consumida por essa população, somados à ausência dos serviços públicos de saneamento, agravam o risco de adoecimento por diversas patologias de veiculação hídrica. Assim, um estudo descritivo de desenho transversal, com amostras de água coletadas em locais de captação para consumo, foi realizado nas aldeias em três períodos sazonais, com objetivo de avaliar a água consumida, através da quantificação de metais tóxicos em relação aos valores máximos permitidos na legislação brasileira. As aldeias com maior número de medidas alteradas de metais na água foram, a Aldeia Pradinho (100%), seguida de Água Boa (92%). Os menores números de alterações foram encontrados nas Aldeias Verde e Rafael (27%). Os metais que apareceram em maior número de amostras com valores acima do recomendado foram o Ferro (50%), seguido do Arsênio (46%), Alumínio (36%), Cádmio (22%) e o Mercúrio (20%). Dessa forma, este estudo identificou altas concentrações de metais tóxicos na água consumida pelas comunidades indígenas de Maxakali no Brasil.

Abstract The Maxakali is the second largest indigenous population in the state of Minas Gerais, Brazil; and parasitic diseases are its main cause of death. Problems related to the quality of water consumed by this population, added to the absence of public sanitation services, aggravate the risk of illnesses due to several water-borne pathologies. Thus, the main purpose of this paper was to evaluate the water quality consumed in natura by the Maxakali community, quantifying toxic metals in relation to the maximum values allowed by Brazilian law. A descriptive cross-sectional study was carried out with samples of water collected in surface- and groundwater in the Maxakali villages, including three seasonal periods. Villages with the greatest number of altered measures of metals in water were Aldeias Pradinho (100%) and Água Boa (92%). The smallest number of changes were found in Aldeias Verde and Rafael (27%). The metals that appeared in the largest number of collections with values higher than recommended were Iron (50%), followed by Arsenic (46%), Aluminum (36%), Cadmium (22%) and Mercury (20%), respectively. The study identified high concentrations of toxic metals in the water consumed by the Maxakali indigenous community in Brazil.

ABSTRACT: Among soil microorganisms, the genus Cupriavidus has garnered particular scientific, economic and ecological interest because of its ability to fix nitrogen and tolerate high concentrations of metals. The aim of this study was to analyze four strains of Cupriavidus necator for their ability to tolerate and bioaccumulate cadmium and zinc. The tolerance of these strains to these metals was assessed in liquid culture medium containing different concentrations of Zn + Cd and in soil solutions and soils contaminated with multiple elements including Zn, Cd, Cu and Pb. The four strains showed high tolerance to Zn and Cd, both in culture medium and when inoculated into contaminated soil solutions or multi-element contaminated soil. The UFLA02-71 strain displayed the highest ability to bioaccumulate these metals. It was able to accumulate 93.76 µmol g cell−1 of Zn and 16.03 µmol g cell−1 of Cd when cultured in liquid medium with a total heavy metal concentration of 9,140 µmol L−1 (9,000 Zn + 140 Cd) and was able to accumulate 16.98 µmol g cell−1 of Cd in the soil solution. An increase in the pH of the culture medium resulting from the growth of the C. necator strains reduced the Zn2+ and Cd2+ ions in the medium and increased the concentrations of the ZnHPO4 and CdHPO4 species in solution. Thus, we concluded that they show great potential for use in the bioremediation of HM-contaminated areas.

Resumo Os fungos têm uma grande capacidade para produzir enzimas que são capazes de degradar compostos. De acordo com esse fato, os corantes solúveis são frequentemente utilizados como maneira de avaliação e seleção de linhagens com potencial de biodegradação. O presente estudo verificou a capacidade de linhagens de Pleurotus sp. na degradação do corante azul de metileno, a fim de identificar linhagens com potencial para processos de biorremediação. Desse modo, um total de sete linhagens de Pleurotus sp. foram cultivadas em meio líquido de batata com o corante azul de metileno nas concentrações de 0,02 g L-1 e 0,04 g L-1. Os resultados das absorvâncias mostraram que todos os isolados foram visivelmente capazes de reduzir a coloração do meio, após sete dias de crescimento fúngico. Observou-se ainda, uma redução de aproximadamente 80% na coloração para as linhagens denominadas ERY, HI e SB em comparação com o controle. Assim, foi possível constatar que essas linhagens podem ser utilizadas em bioprocessos de remoção de cor e na biodegradação de compostos poluentes.

Abstract Fungi have a large capacity to produce enzymes that are capable of degrading compounds. In this regard, soluble dyes are often used as a means of evaluating and selecting strains with potential for biodegradation. This study verified the capacity of the strains of Pleurotus sp. in degrading methylene blue dye, in order to identify the strains with potential for the bioremediation. For this reason, a total of seven strains of Pleurotus sp. were grown in a potato liquid medium with the methylene blue dye at the concentrations of 0.02 g L-1 and 0.04 g L-1. The results of the absorbances showed that all the isolates were visibly able to reduce the media staining following seven days of growth. However, a reduction of approximately 80% in the staining was observed for the strains named ERY, HI, and SB compared to the control. Thus, it was observed that these strains could be used in the color removal bioprocesses and in the biodegradation of pollutant compounds.

Anthracnose, caused by Colletotrichum sublineolum Henn. ex Sacc. & Trotter, is one of the most important sorghum [Sorghum bicolor (L.) Moench] diseases in Brazil. This fungus exhibits conidial dimorphism, producing either falcate or oval conidia on solid and liquid media, respectively. We compared patterns of the initial infection events by these two types of conidia on sorghum leaves using light microscopy and scanning electron microscopy. The infection events during the first 24 h were similar for both oval and falcate conidia. Globose and melanized apressoria were formed at 24 h after inoculation (hai) regardless of the conidia type. Dense mycelium and oval conidia developed from germinated falcate conidia at 32 hai. Hyphal mass displaying acervuli filled with falcate conidia and surrounded by setae, developed from germinated oval conidia at 48 hai. Oval conidia were as capable as falcate conidia of infecting sorghum leaves. The inherent ability to grow faster and the easeness with which oval conidia can be produced in vitro as compared to falcate, make the former a preferred choice for studies on the C. sublineolum-sorghum interaction. It would be instructive to further investigate the potential role of the oval conidia in epidemics.

O objetivo deste trabalho foi avaliar a tolerância de estirpes de Cupriavidus necator a zinco, cádmio, cobre e chumbo, além de determinar a eficiência simbiótica das estirpes mais tolerantes em associação a espécies leguminosas com potencial para revegetação. A tolerância foi testada em meio LB, suplementado com 2,5; 5,0; 7,5; 10; 12,5 e 15 mmol L-1 de ZnSO4.7H2O, CdSO4.8H2O, CuSO4.5H2O e PbCl2, respectivamente, em comparação ao controle sem adição de metal. Determinou-se a eficiência simbiótica das quatro estirpes de C. necator mais tolerantes aos metais avaliados (UFLA02-71, UFLA02-73, UFLA01-659 e UFLA01-663), as quais foram inoculadas nas espécies: Leucaena leucocephala, Enterolobium contortisiliquum, Acacia mangium, Mimosa caesalpiniifolia, M. pudica, M. pigra e M. acutistipula. Em vasos com solos, avaliaram-se L. leucocephala, M. pudica e M. caesalpiniifolia e as estirpes UFLA01-659 e UFLA02-71, selecionadas na avaliação de eficiência simbiótica. A estirpe UFLA02-71 proporcionou incrementos de matéria seca da parte aérea de 870% em M. caesalpiniifolia, enquanto que UFLA01-659 proporcionou 885% em M. pudica e 924% em L. leucocephala. As estirpes UFLA01-659 e UFLA02-71, além da alta tolerância a metais pesados, apresentaram eficiência em fixar nitrogênio, em simbiose com essas leguminosas, em solos com rizóbios nativos capazes de nodulá-las, e devem ser avaliadas quanto ao seu potencial de utilização em programas de recuperação de áreas degradadas.

The objective of this work was to evaluate the tolerance of Cupriavidus necator strains to zinc, cadmium, copper and lead, as well as to determine the symbiotic efficiency of the most tolerant ones in legume species suited for use in revegetation. Tolerance was evaluated in LB medium supplemented with 2.5, 5.0, 7.5, 10, 12.5 and 15 mmol L-1 of ZnSO4.7H2O, CdSO4.8H2O, CuSO4.5H2O and PbCl2, respectively, in comparison to a control without metal. The symbiotic efficiency of the four C. necator most metal-tolerant strains (UFLA02-71, UFLA02-73, UFLA01-659 and UFLA01-663) was determined, and these strains were inoculated in the species: Leucaena leucocephala, Enterolobium contortisiliquum, Acacia mangium, Mimosa caesalpiniifolia, M. pudica, M. pigra and M. acutistipula. Leucaena leucocephala, M. pudica and M. caesalpiniifolia and the strains UFLA02-71 and UFLA01-659, which presented the high symbiotic efficiency, were evaluated in pots with soil. UFLA02-71 provided increments of 870% in shoot dry matter of M. caesalpiniifolia, and UFLA01-659 provided 885% in M. pudica and 924% in L. leucocephala. These strains should be assessed for potential use in programs to restore degraded areas, since they showed high efficiency in nitrogen fixation and were competitive with indigenous rhizobia populations, besides being highly tolerant to heavy metals.

O processo de fixação biológica de nitrogênio (FBN), realizado por bactérias fixadoras de N2 simbióticas de leguminosas, comumente denominados α e β rizóbios, proporciona alta sustentabilidade aos ecossistemas. Seu manejo como uma biotecnologia é bem sucedido para aumentar a produtividade das culturas. Um exemplo notável desse sucesso é a inoculação da soja com estirpes de Bradyrhizobium.Os rizóbios produzem grande diversidade de estruturas químicas dos exopolissacarídeos (EPS). Embora o papel dos EPS seja relativamente bem estudado no processo de FBN, o seu potencial econômico e ambiental ainda não é explorado. Esses EPS são principalmente heteropolissacarídeos espécie-específicos, que podem variar de acordo com a composição dos açúcares, as suas ligações em uma única subunidade, o tamanho da unidade repetitiva e o grau de polimerização. Estudos mostram que os EPS produzidos por essas bactérias exercem importante papel no processo de invasão, formação do cordão de infecção, desenvolvimento do bacteroide e do nódulo e resposta de defesa da planta. Esses EPS também conferem proteção a essas bactérias quando submetidas a diversos estresses ambientais. Em geral, estirpes de rizóbios que produzem maior quantidade de EPS são mais tolerantes às condições adversas, quando comparadas com estirpes que produzem menor quantidade. Além disso, sabe-se que os EPS produzidos por microrganismos são amplamente utilizados em vários segmentos industriais. Esses compostos, também denominados biopolímeros, fornecem uma alternativa válida para a substituição das gomas comumente usadas na indústria de alimentos, por meio do desenvolvimento de produtos com propriedades praticamente idênticas ou com melhores características reológicas, que podem ser usados para novas aplicações. Os EPS microbianos também são capazes de aumentar a adesão de partículas do solo, favorecendo a estabilidade mecânica dos agregados, além de aumentarem os níveis de retenção de água e fluxo de ar nesse ambiente. Diante da importância dos EPS, na presente revisão discute-se o papel desses compostos no processo de fixação biológica de N2, na adaptação dos rizóbios a estresses ambientais, bem como no processo de agregação do solo. As possíveis aplicações desses biopolímeros na indústria também são discutidas.

The process of biological nitrogen fixation (BNF), performed by symbiotic nitrogen fixing bacteria with legume species, commonly known as α and β rhizobia, provides high sustainability for the ecosystems. Its management as a biotechnology is well succeeded for improving crop yields. A remarkable example of this success is the inoculation of Brazilian soybeans with Bradyrhizobium strains. Rhizobia produce a wide diversity of chemical structures of exopolysaccharides (EPS). Although the role of EPS is relatively well studied in the process of BNF, their economic and environmental potential is not yet explored. These EPS are mostly species-specific heteropolysaccharides, which can vary according to the composition of sugars, their linkages in a single subunit, the repeating unit size and the degree of polymerization. Studies have showed that the EPS produced by rhizobia play an important role in the invasion process, infection threads formation, bacteroid and nodule development and plant defense response. These EPS also confer protection to these bacteria when exposed to environmental stresses. In general, strains of rhizobia that produce greater amounts of EPS are more tolerant to adverse conditions when compared with strains that produce less. Moreover, it is known that the EPS produced by microorganisms are widely used in various industrial activities. These compounds, also called biopolymers, provide a valid alternative for the commonly used in food industry through the development of products with identical properties or with better rheological characteristics, which can be used for new applications. The microbial EPS are also able to increase the adhesion of soil particles favoring the mechanical stability of aggregates, increasing levels of water retention and air flows in this environment. Due to the importance of EPS, in this review we discuss the role of these compounds in the process of BNF, in the adaptation of rhizobia to environmental stresses and in the process of soil aggregation. The possible applications of these biopolymers in industry are also discussed.

Uma bactéria identificada como Pantoea ananatis foi recentemente isolada de lesões jovens da doença mancha branca do milho de plantas naturalmente infectadas. Esta bateria reproduziu sintomas semelhantes aos da doença quando inoculada em plantas de milho em casa de vegetação. Estudos anteriores realizados por outros autores demonstraram que o controle desta doença em condições de campo foi obtido pelo uso de fungicidas, principalmente o Mancozeb, nas fases iniciais de seu desenvolvimento. O objetivo deste estudo foi avaliar a freqüência de isolamento da bactéria P. ananatis a partir de plantas infectadas coletadas na região de Londrina, Estado do Paraná, e reproduzir sintomas da doença através de inoculações artificiais em plantas de milho em casa de vegetação. Utilizando os produtos químicos testados anteriormente por outros autores para o controle desta doença a campo, foi também objetivo deste trabalho avaliar o potencial destes produtos na inibição da bactéria tanto em condições de laboratório como em condições de infecção natural. Os resultados mostraram que P. ananatis foi isolada em 40% das lesões jovens coletadas a campo e quando inoculada em casa de vegetação sob condições controladas reproduziu sintomas semelhantes aos observados a campo. Entre os produtos químicos testados, o fungicida Mancozeb mostrou-se eficiente no controle da doença a campo, em concordância com os relatos anteriores. Este produto inibiu completamente o crescimento da bactéria em laboratório, explicando os resultados obtidos a campo. Os demais produtos não foram eficientes no controle a campo e eles também não inibiram a bactéria em laboratório. Estes resultados representam evidências adicionais de que a bactéria P. ananatis é o agente causal da doença mancha branca do milho.

A bacterium identified as Pantoea ananatis was recently isolated from young lesions of the disease maize white spot from infected plants in the field. This bacterium produced similar symptoms of the disease when inoculated on maize plants in greenhouse. Previous studies carried out by other authors demonstrated that the control of this disease in field conditions has been achieved through the use of fungicides mainly with the Mancozeb in the initial phases of its development. The aim of this study was to evaluate the frequency of isolation of P. ananatis from infected plants in the region of "Londrina, Paraná State" and to reproduce symptoms of the disease through artificial inoculation of maize plants in greenhouse. Using the chemical products tested previously by other authors for the control of this disease in the field, it was also objective of this work to evaluate the potential of these products to inhibit the bacterium, at both in laboratory and in condition of natural infection. The results showed that P. ananatis was successfully isolated from 40% of the young lesions collected in the field and when inoculated in greenhouse (controlled conditions), it reproduced symptoms similar to what was observed in the field. Among the tested chemical products, the fungicide Mancozeb was the best in the control of the disease under field conditions, in agreement with previous reports. This product inhibited completely the bacterium in laboratory, explaining the results found in the field. The others products were not efficient to control the disease and they did not also inhibit the bacterium in laboratory. The results represent additional evidences that the bacterium P. ananatis is the causal agent of the maize white spot disease.
