ABSTRACT Aiming at a more sustainable food production, the reuse of effluent from fish farming in hydroponics can be a viable option. Thus, the objective of present study was to evaluate the effects of the use of effluent from fish farming on the development of ‘Folha Larga’ watercress and the possibility of reducing the use of fertilizer in a hydroponic system. The assay was carried out in a greenhouse from October 2021 to February 2022 (two cycles). The treatments were distributed in randomized blocks, with four replications, with split plots, consisting of two times of recirculation of the solution in the plots (T1 = 15 min operating by 15 min off and T2 = 15 min operating by 30 min off) and five solutions, in the subplots, obtained by mixing different proportions (0, 25, 50, 75, and 100%) of nutrient solution and effluent from fish farming. The variables analyzed were: plant height, number of leaflets, leaf area, SPAD index, fresh and dry weight of the aerial part and roots. The use of effluent from fish farming reduces the amount of fertilizers for the yield of ‘Folha Larga’ watercress. Nutrient solution recirculation time of 15 minutes is suitable for hydroponic watercress.

RESUMO Visando uma produção de alimentos mais sustentável, a reutilização de efluente da piscicultura na hidroponia pode ser uma opção viável. Assim, objetivo desta pesquisa foi avaliar os efeitos do uso de efluente da piscicultura no desenvolvimento do agrião ‘Folha Larga’ e a possibilidade de redução do uso de fertilizantes em sistema hidropônico. A pesquisa foi realizada em casa de vegetação entre outubro de 2021 e fevereiro de 2022 (dois ciclos). Os tratamentos foram distribuídos em blocos ao acaso, com quatro repetições, com parcelas subdivididas, consistindo em dois tempos de recirculação da solução nas parcelas (T1 = 15 min operando por 15 min desligado e T2 = 15 min operando por 30 min desligado) e cinco soluções, nas subparcelas, obtidas pela mistura em diferentes proporções (0, 25, 50, 75 e 100%) de solução nutritiva e efluente da piscicultura. As variáveis analisadas foram: altura de plantas, número de folíolos, área foliar, índice SPAD, massa fresca e massa seca da parte aérea e raiz. O uso de efluente da piscicultura reduz a quantidade de fertilizantes para a produção de agrião ‘Folha Larga’. O tempo de recirculação da solução nutritiva de 15 minutos é adequado para o agrião hidropônico.

ABSTRACT Plant growth-promoting bacteria emerge as an alternative to attenuate abiotic stresses in plants such as salinity. The objective of this study was to evaluate the effects of Bacillus sp. strains 186 and 109 on the acclimatization of banana cv. ‘Prata Catarina’ irrigated with saline water. The experiment was conducted in a completely randomized design in a 4 × 2 factorial scheme, with 4 growth-promotion treatments, subjected to 2 levels of electrical conductivity of irrigation water. Irrigation with saline water decreased the number of leaves, leaf area, transpiration rate, stomatal conductance, and CO2 assimilation rate in banana leaves, regardless of inoculation with bacterial strains. The use of Bacillus sp. strain 109 promoted increments in phosphorus and iron contents in banana plants irrigated with low-salinity water.

RESUMO Bactérias promotoras de crescimento vegetal surgem como alternativa para atenuar estresses abióticos em plantas como a salinidade. O objetivo deste estudo foi avaliar os efeitos do Bacillus sp. cepas 186 e 109 na aclimatização da cv. ‘Prata Catarina’ irrigada com água salina. O experimento foi conduzido em delineamento inteiramente casualizado em esquema fatorial 4 × 2, com 4 tratamentos promotores de crescimento, submetidos a 2 níveis de condutividade elétrica da água de irrigação. Irrigação com água salina diminuiu o número de folhas, a área foliar, a taxa de transpiração, a condutância estomática e a taxa de assimilação de CO2 nas folhas de bananeira, independentemente da inoculação com cepas bacterianas. O uso de Bacillus sp. cepa 109 promoveu incrementos nos teores de fósforo e ferro em bananeiras irrigadas com água de baixa salinidade.

Abstract The effect of synthesis parameters on the microstructural behavior and morphology during the yttrium oxide (Y2O3) formation is reported. Y2O3 crystals were produced by a modified sol-gel route assisted by polyvinyl alcohol solution varying the calcination temperature and solution pH. The crystalline phase formation was investigated using X-ray powder diffraction combined with the Rietveld refinement method. The microstructural properties were analyzed by using the Williamson-Hall formalism. The calcination temperature followed the thermal events observed in the differential thermal analysis combined with the thermogravimetric analysis of the precursor xerogel. It was seen that the combination of PVA and pH variation can be used to minimize the calcination time and temperature. The morphological analysis showed samples with different sizes and appearances depending on pH and calcination temperature. Therefore, it was shown that the parameters in the synthesis process can be used to tailor the properties of Y2O3, such as crystallite size, degree of structural ordering, and morphology, and consequently, improve the desired application. Abstract The present study sought to evaluate the antibacterial activity of trans-anethole against food-borne strains of Enterobacter cloacae and Enterococcus faecalis. The study was performed using Minimum Inhibitory Concentration (MIC), and Minimum Bactericidal Concentration (MBC) methods, in addition, disc diffusion technique was used to evaluate the association of trans-anethole with synthetic antimicrobials. Minimum Inhibitory Concentration for Adherence (MICA) testing was also performed. The results revealed that trans-anethole presents no antibacterial activity at any of the concentrations used against the E. cloacae strains tested. However, trans-anethole presented antibacterial effect against five of the six E. faecalis bacterial strains tested, with MIC values ranging from 500 μg/mL to 1000 μg/mL. Further, when analyzing the MBC results against E. faecalis, it was observed that the compound presented values ranging from 500 μg/mL to 1000 μg/mL. As for the associations, it was observed that trans-anethole when combined with the antimicrobials ampicillin, gentamicin, ciprofloxacin, and ceftriaxone presented synergistic effect against most strains of E. faecalis. However, both trans-anethole and the control chlorhexidine (0.12%) presented no antibiofilm effects against strains of E. faecalis. In short, trans-anethole presented potential antibacterial against E. faecalis strains of food origin, and may upon further study, it may be used alone or in association with synthetic antimicrobials to combat infections caused by this bacterium.

Resumo O presente estudo procurou avaliar a atividade antibacteriana do trans-anetol contra cepas de Enterobacter cloacae e Enterococcus faecalis de origem alimentar. O estudo foi realizado utilizando métodos de Concentração Inibitória Mínima (CIM), e Concentração Bactericida Mínima (CBM), além disso, foi utilizada a técnica de difusão de disco para avaliar a associação do trans-anetol com antimicrobianos. O teste de Concentração Inibitória Mínima de Aderência (CIMA) também foi realizado. Os resultados revelaram que o trans-anetol não apresentou atividade antibacteriana em nenhuma das concentrações utilizadas contra as cepas de E. cloacae testadas. No entanto, o trans-anetol apresentou efeito antibacteriano contra cinco das seis cepas bacterianas de E. faecalis testadas, com valores de CIM variando de 500 μg/mL a 1000 μg/mL. Além disso, ao analisar os resultados da CBM contra E. faecalis, observa-se que o composto apresentou valores variando de 500 μg/mL a 1000 μg/mL. Quanto às associações, observou-se que o trans-anetol quando combinado com os antimicrobianos ampicilina, gentamicina, ciprofloxacino, e ceftriaxona apresentou efeito sinérgico contra a maioria das cepas de E. faecalis. No entanto, tanto o trans-anetol quanto o controle clorexidina (0,12%) não apresentaram efeito antibiofilme contra a cepa de E. faecalis. Em suma, o trans-anetol apresentou potencial antibacteriano contra cepas de E. faecalis de origem alimentar, podendo, mediante estudos mais aprofundados, ser utilizado isoladamente ou em associação com antimicrobianos sintéticos para combater infecções causadas por esta bactéria.

Abstract This study evaluated the antimicrobial efficacy of a new intracanal drug based on Pentaclethra macroloba extract, a plant of Amazonian origin, against Enterococcus faecalis using macrodilution test and intratubular evaluation with Confocal Laser Scanning Microscopy (CLSM). The minimum inhibitory concentration (MIC) and the minimum bactericidal concentration (MBC) of the pure extract of Pentaclethra macroloba andits association with calcium hydroxide and ultracall were determined. Then, thirty-three dentin cylinders were prepared and inoculated with E. faecalis, to evaluate the antimicrobial effect of the medications on the dentinal tubules with CLSM. The data was analyzed using the Kruskal-Wallis and Dunn tests. The extract in association with calcium hydroxide showed a lower CBM, and in the intratubular test all tested medications were effective against E. faecalis (P >0.05). The new intracanal drug based on P. macroloba extract has an antimicrobial effect against E. faecalis and further studies are needed for its clinical use.

Resumo Este estudo avaliou a eficácia antimicrobiana de uma nova droga intracanal à base de extrato de Pentaclethra macroloba, uma planta de origem amazônica, contra Enterococcus faecalis por meio do teste de macrodiluição e avaliação intratubular em Microscopia Confocal de Varredura a Laser (CLSM). Foram determinadas a concentração inibitória mínima (CIM) e a concentração bactericida mínima (CBM) do extrato puro de Pentaclethra macroloba, sua associação com hidróxido de cálcio e ultracall. Em seguida, trinta e três cilindros de dentina foram preparados e inoculados com E. faecalis, para avaliar o efeito antimicrobiano de medicamentos nos túbulos dentinários em CLSM. Os dados foram analisados por meio dos testes de Kruskal-Wallis e Dunn. O extrato em associação com hidróxido de cálcio apresentou menor CBM. E no teste intratubular, todas as medicações testadas foram efetivas contra E. faecalis (P >0,05). A nova droga intracanal à base de extrato de P. macroloba tem efeito antimicrobiano contra E. faecalis e mais estudos são necessários para seu uso clínico.

Abstract Potential breeding containers for Aedes aegypti and Aedes albopictus mosquitoes are found in different environments, these places are considered by the National Dengue Control Program in Brazil as strategic points (SP), which have favorable conditions for the development of those insects. The aim of this study is to identify potential breeding containers for A. aegypti and A. albopictus at strategic points in the city of Codó, Maranhão. This study was conducted in five districts, one in each administrative area of the city. A survey of the types and quantity of existing strategic points in each neighborhood was carried out, and surveys of these properties were carried out to inspect the potential breeding containers, with the collection of the vectors’ number of immatures found at them. 125 properties were inspected of which 76.91% and 81.25% were vacant lots in the dry and rainy seasons, respectively. There was a difference between the median of the different strategic points (H = 21.96; p < 0.0001), For A. aegypti, there was no difference between the medians of number of immatures regarding the strategic points (H = 3.71; p = 0.28). The total number of immature A. albopictus was higher in vacant lots than in workshops (H = 9.41; p = 0.02, p < 0.05). 4,356 and 4,911 potential breeding containers were found in the dry and rainy seasons, respectively. Regarding the potential breeding containers, 7 (0.16%) and 47 (0.96%) were found with immature vectors, in the dry and rainy seasons, respectively. There were more positive containers in the rainy season for which there was a difference between the median of the number of mosquitoes per positive container (H = 15.66; p = 0.01), the number of immatures for the tires group was greater than Vases and Fixed groups (p < 0.05); the same result was observed in the analyses of the species in the separate form. The impressive number of potential breeding containers found in the SP in both seasons highlights the importance of developing control strategies for these vectors, with emphasis on places such as vacant lots, workshops, tire shops and junkyards, which are serving as dispersal urban sites of vectors.

Resumo Os potenciais criadouros dos mosquitos Aedes aegypti e Aedes albopictus são encontrados em diversos ambientes, esses locais são considerados pelo Programa Nacional de Controle da Dengue como pontos estratégicos (PE), os quais possuem condições favoráveis ao desenvolvimento desses insetos. Propõe-se nesse estudo identificar os potenciais criadouros de A. aegypti e A. albopictus em pontos estratégicos na cidade de Codó, Maranhão. Este estudo foi realizado em cinco bairros, um em cada zona administrativa da cidade. Foi realizado o levantamento dos tipos e quantidade de pontos estratégicos existentes em cada bairro, e vistorias desses imóveis para inspeção dos recipientes potencial criadouro, com coleta das formas imaturas encontradas nestes. Foram inspecionados 125 imóveis, para os quais houve diferença entre a mediana dos diferentes pontos estratégicos (H = 21,96; p < 0,0001), sendo que os terrenos baldios foram mais frequentes com 76,91% e 81,25% nos períodos seco e chuvoso, respectivamente. Para A. aegypti não houve diferença entre as medianas de imaturos em relação aos pontos estratégicos (H = 3,71; p = 0,28). A quantidade de imaturos de A. albopictus foi maior em terrenos baldios do que em oficinas (H = 9,41; p = 0,02, p < 0,05). Foram encontrados 4.356 e 4.911 recipientes potenciais criadouros nos períodos seco e chuvoso, nesta ordem. Em relação aos recipientes positivos, 7 (0,16%) e 47 (0,96%) foram encontrados com imaturos dos vetores, nos períodos seco e chuvoso, respectivamente. Só houve predominância de recipiente positivo no período chuvoso, para o qual houve diferença entre as medianas de imaturos das duas espécies (H = 15,66; p = 0,01), sendo que a quantidade de imaturos para o grupo Pneus foi maior que Vasos e Fixos (p < 0,05), o mesmo resultado foi observado nas análises das espécies da forma separada. O número expressivo de recipientes potenciais criadouros encontrados nos PE nos dois períodos, evidencia a importância de se desenvolver estratégias de controle a esses vetores com ênfase em locais como terrenos baldios, oficinas, borracharias e ferros-velhos, os quais estão servindo de local de dispersão dos vetores em área urbana.

ABSTRACT Management of nitrogen fertilization and light interception of pastures contributes to forage production in regions with low water availability. Therefore, the objective was to evaluate the effect of different nitrogen doses and light interception levels on the growth of Panicum maximum cv. Tanzania under different irrigation managements. The experiment was carried out in a greenhouse, using a factorial scheme (5 x 2 x 2), corresponding to five irrigation depths (40, 60, 80, 100, and 120% of actual evapotranspiration), two canopy light interception levels (90 and 95%), and two nitrogen doses (300 and 600 kg N ha-1 year-1). The highest biomass production occurs under irrigation with 120% of evapotranspiration. However, the best structural characteristics (tillering and number of leaves) occur with 100% of evapotranspiration, regardless of nitrogen dose and light interception level. Interruption of growth with 95% of incident light and fertilization with 600 kg N ha-1 year-1 promoted greater tillering, number of leaves and leaf length, regardless of the irrigation level. Tanzania grass management with an interruption of growth with 95% of incident light, 600 kg N ha-1 year-1, and irrigation with 80% of actual evapotranspiration led to tillering and number of leaves only 11% lower than the values found in the other treatments irrigated with 100% of actual evapotranspiration. Interruption of growth with 95% of incident light and fertilization with 600 kg N ha-1 year-1 improve the performance of Tanzania grass under deficit irrigation.

RESUMO O manejo da adubação nitrogenada e interceptação luminosa das pastagens contribuem para produção de forragem em regiões com baixa disponibilidade de água. Portanto, objetivou-se avaliar o efeito de diferentes doses de nitrogênio e níveis de interceptação luminosa no crescimento do capim Panicum maximum cv. Tanzânia sob diferentes manejos de irrigação. O experimento foi conduzido em casa de vegetação, em esquema fatorial (5 x 2 x 2), relativo a cinco lâminas de irrigação (40, 60, 80, 100 e 120% da evapotranspiração real), dois níveis de interceptação de luz pelo dossel (90 e 95%) e duas doses de nitrogênio (300 e 600 kg N ha-1 ano-1). A maior produção de biomassa ocorre com 120% da evapotranspiração. Porém as melhores características estruturais (perfilhamento e número de folhas) ocorrem com 100% da evapotranspiração, independente da dose de nitrogênio e nível de interceptação luminosa. A interrupção do crescimento com 95% de luz incidente e a adubação com 600 kg N ha-1 ano-1 proporcionaram maior perfilhamento, número e comprimento de folhas do capim, independentemente da lâmina de irrigação. O capim Tanzânia com interrupção do crescimento com 95% da luz incidente, 600 kg N ha-1 ano-1 e irrigação com 80% da evapotranspiração obteve perfilhamento e número de folhas apenas 11% inferior aos outros tratamentos irrigados com 100% da evapotranspiração. A interrupção do crescimento com 95% da luz incidente e adubação com 600 kg N ha-1 ano-1 melhoram o desempenho do capim Tanzânia sob irrigação deficitária.

ABSTRACT Introduction: One of the most critical complications in myelodysplastic syndromes (MDS) is the progression to acute myeloid leukemia (AML). The dynamics of clonal evolution in MDS and how acquired mutations can be used as biomarkers to track disease progression remains under investigation. Objective and method: Herein, we investigated the frequency of common myeloid clonal mutations (FLT3, NPM1, JAK2, IDH1 and IDH2) in 88 patients with MDS and 35 AML patients with myelodysplasia-related changes, followed at a single reference center in northeastern Brazil. Results: Overall, 9/88 (10%) ofthe MDSpatients and 9/35 (26%) of the secondary AML patients had at least one mutation. While the JAK2 V617F mutation was the most frequent in the MDS patients, the FLT3, NPM1, IDH1 and IDH2 mutations were more frequently found in the secondary AML group. Furthermore, there was a higher frequency of FLT3, NPM1, IDH1 and IDH2 mutations in MDS patients classified as high-risk subtypes than in those of lower risk. Conclusion: Despite the limited sample size, our data suggest that mutations in FLT3, NPM1, IDH1 and IDH2 genes could be potential biomarkers to detect early disease progression in MDS.

Resumo Plectranthus barbatus Andrews (Lamiaceae) é amplamente distribuída no mundo e com uma série de indicações terapêuticas populares. Este trabalho teve como objetivo avaliar a caracterização fitoquímica de dois extratos da folha de P. barbatus e seu potencial antimicrobiano, antineoplásico e imunomodulador. Após coleta, herborização e obtenção do extrato aquoso (PBA) e acetona: água 7: 3 (orgânico) (PBO) de P. barbatus, a caracterização fitoquímica foi realizada por cromatografia líquida de alta eficiência (CLAE). A atividade antimicrobiana foi realizada para determinar a concentração inibitória mínima (CIM) contra oito cepas bacterianas usando o teste de microdiluição e o fungo Trichophyton rubrum por ensaio de difusão em disco e teste de microdiluição. A citotoxicidade foi avaliada por métodos MTT e azul de tripan em células normais mononucleares do sangue periférico (CMSP) em concentrações variadas entre 0,1 a 100 µg.mL-1 e nas linhagens celulares neoplásicas Toledo, K562, DU-145 e PANC-1 em 1, 10 e 100 µg.mL-1 . A atividade imunomoduladora foi avaliada por ELISA sanduíche de citocinas pró-inflamatórias em sobrenadante de culturas de esplenócitos de camundongos BALB/c. Ambos os extratos apresentaram flavonoides, derivados cinâmicos, esteróides e ácido elágico. O PBO mostrou atividade bacteriostática contra Acinetobacter baumannii (CIM = 250 µg.mL-1) e atividade fungistática do PBA contra Trichophyton rubrum (CIM = 800 µg.mL-1). Os extratos não apresentaram toxicidade para CMSP e células neoplásicas (IC50 > 100 µg.mL-1). Além disso, o PBO a 100 µg.mL-1 inibiu significativamente as citocinas IFN-γ e IL-17A (p = 0,03). Plectranthus barbatus é um candidato potencial para uso terapêutico devido à sua baixa toxicidade em células humanas saudáveis e exibe atividade de interesse médico como bacteriostática, fungistática e imunomoduladora.

Abstract Plectranthus barbatus Andrews (Lamiaceae) is widely distributed in the world and has a range of popular therapeutic indications. This work aimed to evaluate the phytochemical characterization of two leaf extracts of P. barbatus, and their antimicrobial, antineoplastic and immunomodulatory potential. After collection, herborization and obtainment of the P. barbatus aqueous extract (PBA) and acetone:water 7:3 P. barbatus organic extract (PBO), the phytochemical characterization was performed by high-performance liquid chromatography (HPLC). The antimicrobial activity was performed to determine the minimum inhibitory concentration (MIC) against eight bacterial strains using the microdilution test and the fungus Trichophyton rubrum by disc diffusion assay and microdilution test. Cytotoxicity was assessed by MTT and trypan blue methods in normal peripheral blood mononuclear cells (PBMCs) at concentrations ranged between 0.1 to 100 µg.mL-1 and in neoplastic cell lines Toledo, K562, DU-145 and PANC-1 at 1, 10 and 100 µg.mL-1 . Immunomodulatory activity, was evaluated by sandwich ELISA of proinflammatory cytokines at BALB/c mice splenocytes cultures supernatant. Both extracts presented flavonoids, cinnamic derivatives, steroids and ellagic acid. PBO showed bacteriostatic activity against Acinetobacter baumannii (MIC = 250 µg.mL-1) clinical isolate and PBA fungistatic activity against Trichophyton rubrum (MIC = 800 µg.mL-1). The extracts did not exhibit toxicity to PBMCs and neoplastic cells (IC50 > 100 µg.mL-1). Additionally, PBO at 100 µg.mL-1 significantly inhibited IFN-γ and IL-17A cytokines (p = 0.03). Plectranthus barbatus is a potential candidate for therapeutic use due to its low toxicity in healthy human cells and exhibits biological activities of medical interest as bacteriostatic, fungistatic and immunomodulatory.

Resumo O objetivo deste estudo foi avaliar os padrões de expressão gênica dos receptores de LH (lhr) e de estrogênio (er) e dos níveis de esteróides plasmáticos durante o desenvolvimento testicular de Genyatremus luteus. Os machos foram classificados histologicamente em imaturos, em maturação e maduros, de acordo com a estrutura celular dos testículos. A concentração plasmática de 11-KT apresentou um pico na fase de maturação final (P<0.05). As maiores concentrações plasmáticas de 17α-OHP foram encontradas no estádio imaturo (P<0.05), com consequente diminuição nos estádios em maturação e maturo. O 17β-estradiol (E2) apresentou maiores níveis de concentração no estádio em maturação (P<0.05). A expressão de er aumentou significativamente ao longo do desenvolvimento maturacional dos testículos (P<0.05). O mRNA para o receptor de LH diminuiu do estádio imaturo para o estádio em maturação (P<0.05) com consequente pico de expressão no estádio maduro. Houve alta relação entre a expressão gênica dos receptores e os níveis de esteróides plasmáticos, especialmente com E2. Em conclusão, este estudo caracterizou pela primeira vez, sob os aspectos celular, endócrino e molecular, os diferentes estádios de maturação reprodutiva em machos de G. luteus, demonstrando que o perfil da expressão gênica para os receptores er e lhr, bem como as concentrações plasmáticas de 11-KT e E2 foram diretamente relacionados à maturação testicular, apesar de não se relacionarem necessariamente com o índice gonadossomático.

Abstract The aim of the current study is to evaluate gene expression patterns of LH (lhr) and estrogen (er) receptors and plasma steroid levels during testicular development in Genyatremus luteus. Males were histologically classified as immature (n=7), maturing (n=7) and mature (n=7), based on the cellular structure of their testes. Plasma 11-KT concentration recorded peak at the final maturation stage. The highest plasma 17α-OHP concentrations were observed at the immature stage; they decreased at the maturation and mature stages. On the other hand, 17β-estradiol (E2) recorded higher concentrations at the maturation stage. Er expression has significantly increased along the maturational development of animals’ testes. The mRNA observed for the LH receptor has decreased from immature to maturing stage; it presented expression peak at the mature stage. There was high association between receptor gene expression and plasma steroid levels, mainly E2. The current study was the first to feature different reproductive maturation stages in male G. luteus specimens, based on cellular, endocrine and molecular aspects. In addition, it has shown that the gene expression profile for er and lhr receptors, as well as plasma 11-KT and E2 concentrations, are directly linked to testicular maturation, although they are not necessarily associated with the gonadosomatic index.

The development of a method based on extraction induced by emulsion breaking (EIEB) aiming copper determination in gasohol using graphite furnace atomic absorption spectrometry (GF AAS) is reported. Emulsification of the gasohol samples was carried out using nitric acid solution, (1,1,3,3-tetramethylbutyl)phenyl-polyethylene glycol surfactant (Triton X-114), and ultrasound energy. The variables related to the extraction efficiency were optimized by multivariate approaches (two-level factorial fractional design and Doehlert design). The best results were obtained when the emulsion was formed with 5.0 mL of the sample, 0.200 mL of 2.4 mol L-1 HNO3, 0.100 mL of 10% Triton X-114 and 10 min of sonication. A fast and efficient emulsion breaking was performed at 90 ºC. A volume of 6.65 mL of the copper-rich hydroalcoholic phase was extracted, separated from the organic phase and analyzed. Under these conditions, the following analytical characteristics were obtained: 0.61 and 2.0 µg L-1 for limits of detection and quantification, respectively; precision (percentage of relative standard deviation, 10 µg L-1) of 2.8%; and analytical sensitivity of 0.011 L µg -1. Accuracy was assessed by spike tests, and values in the range of 95-110% were achieved. The method was applied in the determination of copper in Brazilian gasoline samples (gasohol with 27% ethanol).

Resumo: O estado de Goiás, no Brasil central, é coberto principalmente pelo domínio do Cerrado, com a Mata Atlântica do Alto Paraná ocupando sua porção centro-sul. Goiás é uma das 20 unidades federativas brasileiras que ainda não possui uma lista de espécies de mamíferos. Neste estudo, apresentamos a primeira lista de mamíferos para o estado. Registramos as espécies de mamíferos com base principalmente na análise de espécimes depositados em coleções científicas, bem como na literatura apresentando material testemunho associado. Listamos 191 espécies pertencentes a 125 gêneros, 31 famílias e 10 ordens, as quais representam 25,2% das espécies de mamíferos que ocorrem no Brasil. As ordens mais especiosas foram Chiroptera (90 spp.), seguida pelas ordens Rodentia (43 spp.), Carnivora (19 spp.) e Didelphimorphia (17 spp.), com as demais ordens respondendo por uma porção menor da diversidade: Cetartiodactyla (7 spp.), Cingulata (7 spp.), Primates (4 spp.), Pilosa (2 spp.), Lagomorpha (1 sp.) e Perissodactyla (1 sp.). Um total de 28 espécies (14,7%), principalmente representadas por mamíferos de médio e grande porte, estão ameaçadas nacionalmente e 12 (6,3%) encontram-se globalmente ameaçadas. Nossos resultados indicaram grandes porções do estado ainda não devidamente pesquisadas em relação aos seus mamíferos, com informações escassas e fragmentadas, principalmente no que diz respeito à sua porção noroeste. Discutimos a riqueza de espécies, a distribuição e o estado de conservação dos mamíferos do estado de Goiás nos cenários nacional e regional. Ressaltamos a importância da realização de inventários que utilizem técnicas complementares de amostragem, incluindo a coleta de material testemunho, proporcionando a obtenção de dados cariotípicos, moleculares, morfológicos, parasitológicos e ecológicos. Estas informações são a base de estudos integrativos, os quais aumentam nossa compreensão a respeito da riqueza e diversidade atual dos mamíferos. O conhecimento a respeito da distribuição da riqueza de espécies em Goiás é essencial para embasar estratégias de conservação, tão necessárias em áreas que vem sofrendo com a perda e fragmentação de seus hábitats naturais, como a porção centro-sul do estado.

Abstract: The state of Goiás, in central Brazil, is covered mainly by the Cerrado domain, with the Alto Paraná Atlantic Forest occupying its central-southern portion. Goiás is one of the 20 Brazilian federative units without a mammal checklist. In this study, we provide the first checklist of mammals from Goiás state. We recorded mammal species based primarily on the analysis of specimens housed in scientific collections as well as on literature with associated voucher material. We listed 191 mammalian species belonging to 125 genera, 31 families and 10 orders, which represents 25.2% of the mammal species occurring in Brazil. The most speciose orders were Chiroptera (90 spp.), followed by Rodentia (43 spp.), Carnivora (19 spp.) and Didelphimorphia (17 spp.). The following orders accounted for a smaller portion of the state diversity: Cetartiodactyla (7 spp.), Cingulata (7 spp.), Primates (4 spp.), Pilosa (2 spp.), Lagomorpha (1 sp.), and Perissodactyla (1 sp.). A total of 28 species (14.7%), mainly represented by medium and large-sized mammals, are nationally threatened while 12 (6.3%) are globally threatened. Our results indicate great portions of the state lacking a proper survey of mammals, especially the northwestern portion. We discuss species richness, distribution and conservation status of the mammals of Goiás state in national and regional scenarios. We highlight the need for mammal inventories based on complementary survey techniques with the collection of vouchers in order to provide karyologic, molecular, morphologic, parasitologic, and ecological data. These informations are the basis for integrative studies that lead to the understanding of current mammalian richness and diversity. Indeed, knowledge on species richness distribution in the state will guide conservation strategies, especially in areas undergoing habitat loss and fragmentation, such as the central-southern portion of Goiás.

RESUMO: O plasma rico em plaquetas (PRP) é uma alternativa terapêutica promissora, pois as plaquetas são ricas em fatores de crescimento com ação na regeneração de tecidos. No entanto, o PRP fresco não pode ser armazenado por longos períodos. Esse trabalho teve como objetivo desenvolver um protocolo de obtenção de PRP liofilizado canino capaz de manter a viabilidade pós reconstituição. Portanto, foram testados diversos protocolos de extração e liofilização. Para validação do PRP canino liofilizado foi analisada a concentração dos fatores de crescimento VEGF e TGF-β antes e após o processo de liofilização, a estrutura tridimensional do PRP liofilizado reconstituído em forma de gel por microscopia eletrônica e seu efeito in vitro na proliferação de células-tronco mesenquimais. Os resultados demonstraram que o PRP liofilizado apresentou concentrações terapêuticas adequadas dos fatores de crescimento VEGF e TGF- β (9,1pg/ml e 6161,6pg/ml, respectivamente). O gel de PRP reconstituído após liofilização apresentou uma durabilidade in vitro de 10 dias, sua estrutura tridimensional mostrou-se semelhante ao PRP fresco e proporcionou intensa proliferação de células-tronco mesenquimais durante o cultivo. O teste de imunogenicidade não demonstrou evidências de reação imune. Os achados foram promissores, sugerindo a possibilidade de uso de PRP canino liofilizado para o mercado. Novos estudos in vivo e in vitro deverão ser conduzidos para comprovação terapêutica.

ABSTRACT: Platelet-rich plasma (PRP) has been considered a promising therapeutic alternative, since platelets are rich in growth factors that are used in the Regenerative Medicine field. However, fresh PRP cannot be stored for long periods. This study aimed to develop a protocol for obtaining lyophilized canine PRP capable of maintaining viability after its reconstitution. For that purpose, canine PRP extraction and lyophilization protocols were initially tested. Subsequently, assays were carried out to quantify the growth factors VEGF and TGF-β, before and after the lyophilization process, gelation test and the three-dimensional gel structure analysis of the reconstituted lyophilized PRP by electron microscopy, as well as in vitro cell proliferation test in lyophilized PRP gel. Additionally, the immunogenicity test was performed, using allogeneic samples of lyophilized PRP. The results showed that the lyophilized PRP had adequate therapeutic concentrations of growth factors VEGF and TGF-β (9.1pg/mL and 6161.6pg/mL, respectively). The reconstituted PRP gel after lyophilization showed an in vitro durability of 10 days. Its electron microscopy structure was similar to that of fresh PRP. In the cell proliferation test, an intense division process was verified in mesenchymal stem cells (MSCs) through the three-dimensional mesh structure of the lyophilized PRP gel. The immunogenicity test showed no evidence of an immune reaction. The findings were promising, suggesting the possibility of having a lyophilized canine PRP that can be marketed. New in vivo and in vitro studies must be carried out for therapeutic confirmation.

Resumo Este estudo avaliou a diversidade beta de peixes em seis riachos de cabeceira localizados em uma área afetada pelo maior centro de aqüicultura ornamental do Brasil, localizado em Minas Gerais. Amostramos assembleias de peixes em 2017 e 2018 para investigar mudanças na estrutura (riqueza de espécies e diversidade beta), comparando esses dados com tendências históricas de composição de comunidades. Registramos 60 espécies de peixes, sendo 16 nativos e 44 não nativos: 19 translocados e 25 exóticos. Os exóticos Poecilia reticulata, Xiphophorus maculatus, X. variatus, Danio rerio e Misgurnus anguillicaudatus foram os mais distribuídos nos riachos. O Índice de Contaminação mostrou que a maioria dos riachos apresentou alta contaminação proporcional por espécies exóticas (acima de 60%). A diversidade beta aumentou do período histórico para o contemporâneo em todos os riachos devido à introdução e pressão de colonização de várias espécies não-nativas translocadas e exóticas, indicando diferenciação biótica. Temperatura da água e número de tanques de piscicultura foram os principais fatores de mudança na diversidade beta dos riachos no período contemporâneo. Os não-nativos induziram mudanças em nível de comunidades e sob influência de variáveis ambientais. Os resultados mostram que a aquicultura é um dos principais vetores da introdução de peixes não-nativos e perda de biodiversidade nativa nos Neotrópicos.

Abstract This study evaluated fish beta diversity in six headwater creeks located in the area affected by the largest ornamental aquaculture center implemented in the Minas Gerais State, southeastern Brazil. We sampled fish assemblages in 2017 and 2018 to investigate changes in assemblage structure (species richness and beta diversity), comparing these data with the historic species pool. We recorded 60 fish species, of which 16 were native and 44 non-native with 19 translocated, and 25 exotic. The exotics Poecilia reticulata, Xiphophorus maculatus, X. variatus, Danio rerio, and Misgurnus anguillicaudatus were the most widely distributed in the headwater creeks. The Contamination Index showed that most creeks had high proportional contamination by exotic species (above 60%). Beta diversity increased from historical to contemporary periods in all creeks due to the introduction and differential colonization pressure of several non-native translocated and exotic species, indicating biotic differentiation. Temperature and number of ponds were the main preditors of change in beta diversity in the headwater creeks during the contemporary period. In summary, we observed that invaders have induced substantial changes to fish communities under influence of environmental filters. Our results support the hipothesis that aquaculture is a main driver of fish non-native fish introduction and native biodiversity loss in the Neotropics.
