Resumo O objetivo deste trabalho foi avaliar a modulação imunológica precoce e tardia da infecção experimental de larvas de T. canis em camundongos. Estes foram infectados com 100 larvas infectantes e eutanasiados em diferentes períodos: 24 e 48 horas pós infecção (HPI), 15 e 30 dias após a infecção (DPI). A resposta humoral foi avaliada por ELISA indireto. RT-PCR quantitativo (qPCR) foi usado para quantificar a transcrição de mRNA das citocinas IL4, IL10, IL12 e Ym1 nos períodos de infecção precoce e tardia. A infecção por T. canis foi capaz de gerar IgG total específico aos 15 e 30 DPI. A análise do isótipo IgG revelou uma diferenciação significativa para IgG1 em comparação com IgG2a, IgG2b e IgG3, caracterizando uma resposta Th-2. Avaliando-se a transcrição gênica na fase inicial da infecção, foram observados níveis mais elevados de transcrição de IL10, IL4 e Ym1 e a regulação negativa de IL12. Na fase tardia, foram observados níveis aumentados de transcrição de IL4, Ym1 e IL12, e foi observada regulação negativa da transcrição de IL-10. Os dados obtidos sugerem, que durante a infecção experimental com T. canis, a participação das citocinas IL4, IL10, IL12 e Ym1 podem desempenhar um papel importante na imunomodulação de T. canis.

Abstract The objective of this work was to evaluate the early and late immunological modulation of an experimental infection of T. canis larvae in mice. Mice were infected with 100 infective larvae and euthanized at different period: 24, 48 hours post infection (HPI), 15- and 30 days post infection (DPI). The humoral response was evaluated by indirect ELISA. Quantitative RT–PCR (qPCR) was used to quantify the mRNA transcription of cytokines IL4, IL10, IL12 and Ym1 in the early and late infection periods. Infection with T. canis was able to generate specific total IgG at 15- and 30- DPI. Analyzing the IgG isotype revealed a significant differentiation for IgG1 compared with IgG2a, IgG2b and IgG3, characterizing a Th-2 response. Evaluating the gene transcription at the early phase of infection, higher transcription levels of IL10, IL4 and Ym1 and a downregulation of IL12 were observed. By the late phase, increased transcription levels of IL4, Ym1 and IL12 were observed, and downregulation of IL-10 transcription was observed. The data obtained suggest that during experimental infection with T. canis, the participation of the IL4, IL10, IL12 cytokines and Ym1 can play an important role in T. canis immunomodulation.

ABSTRACT Human toxocariasis consists of chronic tissue parasitosis that is difficult to treat and control. This study aimed to evaluate the action of the probiotic Lactobacillus acidophilus ATCC 4356 on larvae of Toxocara canis and the effect of IFN-γ cytokine on parasite-host in vivo (1.109 CFU) and in vitro (1.106, 1.107, 1.108, 1.109 CFU) interactions. Four groups of six BALB/c mice were formed: G1 - L. acidophilus supplementation and T. canis infection; G2 - T. canis infection; G3 - L. acidophilus supplementation; and G4 - PBS administration. Mice were intragastrically suplemented with probiotics for 15 days before inoculation and 48 h after inoculation with 100 T. canis eggs. The inoculation of T. canis was also perfomed intragastrically. The recovery of larvae took place through digestion of liver and lung tissues; the evaluation of IFN-γ gene transcription in leukocytes was performed by qPCR. The in vitro test consisted of incubating the probiotic with T. canis larvae. The supplementation of probiotics produced a reduction of 57.7% (p = 0.025) in the intensity of infection of T. canis larvae in mice, whereas in the in vitro test, there was no larvicidal effect. In addition, a decrease in the IFN-γ gene transcription was observed in both, T. canis-infected and uninfected mice, regardless of whether or not they received supplementation. The probiotic L. acidophilus ATCC 4356 reduced T. canis infection intensity in mice, however, the probiotic did not have a direct effect on larvae, demonstrating the need of interaction with the host for the beneficial effect of the probiotic to occur. Yet, the proinflammatory cytokine IFN-γ did not apparently contributed to the observed beneficial effect of probiotics.

Resumo Este estudo avaliou a ação in vitro e in vivo de extratos de Eugenia uniflora em nematódeos gastrintestinais de ovinos. No teste de inibição da eclodibilidade dos ovos, extratos aquosos e hidroalcoólicos foram testados nas concentrações de 40 a 1,25 mg/mL. O efeito citotóxico foi mensurado através do ensaio de MTT em células VERO nas concentrações de 4000 a 1,95 µg/mL. A composição química foi analisada através da marcha fitoquímica qualitativa. No teste in vivo, foram administrados 100 mg/kg, por via oral, do extrato hidroalcoólico por três dias em ovinos naturalmente infectados e realizada coleta de fezes para estimar a redução de ovos por grama de fezes (OPG). Diferentes extratos de E. uniflora inibiram a eclodibilidade dos ovos, com percentual de inibição variando de 14,56 a 99,75%, sendo os hidroalcoólicos mais promissores que os aquosos. Na marcha fitoquímica, compostos com ação anti-helmíntica comprovada foram observados, como flavonoides, saponinas, taninos e triterpenos. Nas condições testadas, os extratos apresentaram citotoxicidade nula a moderada. In vivo, a redução do OPG foi observada apenas no grupo controle (anti-helmíntico). Os extratos de E. uniflora mostraram-se promissores com ação in vitro, necessitando estudos que avaliem outras concentrações e formas de administração in vivo.

Abstract: This study evaluated the in vitro and in vivo action of Eugenia uniflora extracts on sheep gastrointestinal nematode. In the egg hatchability inhibition assay, aqueous and hydroalcoholic extracts were tested in the concentrations ranging from 40 to 1.25 mg/mL. The cytotoxic effect was measured using the MTT assay on VERO cells at concentrations ranging from 4000 to 1.95 μg/mL. The chemical composition was analyzed through the qualitative phytochemical screening. In the in vivo test, 100 mg/kg of the hydroalcoholic extract was administered orally for three days in sheep naturally infected and fecal collection was performed to estimate the egg count per gram of faeces (EPG). Different extracts of E. uniflora inhibited hatchability of eggs with a percentage of inhibition ranging from 14.56 to 99.75%, being the hydroalcoholic most effective than the aqueous. In phytochemical analysis, compounds with anthelmintic action were observed, such as flavonoids, saponins, tannins and triterpenes. Extracts showed moderate to null cytotoxicity under the conditions tested. Reduction of EPG was observed only in control group (anthelmintic). The extracts of E. uniflora proved promising with in vitro action, requiring studies that assess other concentrations and forms of in vivo administration.

ABSTRACT Due to the growing population of pets, especially homeless dogs and cats, zoonoses still represent a significant public health problem. Toxoplasma gondii and Toxocara spp. are epidemiologically important zoonotic agents as they are etiological factors of human toxoplasmosis and toxocariasis, respectively. These parasites remain neglected even though they are substantially prevalent in rural areas. The aim of this study was to investigate T. gondii and T. canis seroprevalence and risk factors of seropositivity in a rural population in Pelotas municipality, Brazil. The study participants (n=344) were patients of a Basic Healthcare Unit (BHU) located in Cerrito Alegre. Blood samples were collected and tested for T. gondii antibodies by indirect immunofluorescence and T. canis antibodies by an indirect ELISA that targets an excreted-secreted antigen (TES). T. gondii seropositivity was 53.2%, with higher titers (1:256 - 1:1,024) in individuals who habitually eat pork, beef, or chicken, while T. canis seropositivity was 71.8% and concomitant T. gondii and T. canis seropositivity was 38.3%. Among the seropositivity risk factors assessed, only habitual undercooked meat consumption was significant (p = 0.046; OR = 3.7) for T. gondii and none of them were associated with T. canis seropositivity. Both parasites have a high prevalence in rural areas, which reinforces the need to invest in rural community education and health.

Abstract INTRODUCTION: This study aimed to evaluate the prevalence of intestinal parasitosis and to identify risk factors associated therewith in hospitalized children. METHODS: Three fecal samples from each patient were evaluated using three different techniques. The patients’ nutritional and socioeconomic status and hematologic profiles were evaluated. RESULTS: Of 106 children, 32.1% tested positive for intestinal parasitosis. The associated risk factors were low parental education levels and children's nail-biting habit. Eosinophilia, observed in 15 cases, was not associated with parasitosis. CONCLUSIONS: We recommend routine fecal parasitologic examination for hospitalized children and implementation of educational campaigns on the prevention of these diseases.

Colectinas e galectinas são proteínas da família das lectinas que possuem a capacidade de reconhecer padrões moleculares associados aos patógenos. Estudos em bovinos têm demonstrado a alta expressão dessas proteínas durante a infecção por nematoides gastrintestinais. O objetivo deste estudo foi investigar se o nível de infecção de Haemonchus contortus altera a expressão de colectinas (SPA e CGN) e galectinas (Gal11 e Gal14) de ovinos. Doze ovinos da raça Corriedale expostos a infecção natural com nematoides foram separados em dois grupos: grupo 1 (G1, n=7) com menor grau de parasitismo; e grupo 2 (G2, n=5) com maior grau, a partir da contagem do número de parasitos recuperados do abomaso e OPG. A contagem de OPG e de parasitos recuperados do abomaso dos grupos G1 e G2 apresentaram diferença estatística (p<0,05). A expressão dos genes de colectinas e galectina foi observada em todas as amostras de abomaso dos ovinos, porém animais com menor grau de infecção apresentaram menor expressão dos genes de Gal14, SPA e CGN (p<0,05). A expressão de lectinas foi associada ao número de H. contortus encontrados no abomaso de ovinos, indicando um possível papel dessas proteínas no controle da infecção.

Galectins and collectins are proteins classified in the lectin family that have the ability to recognize molecular patterns associated with pathogens. Studies on cattle have demonstrated high expression of these proteins during infection with gastrointestinal nematodes. The aim of this study was to investigate whether the level of Haemonchus contortus infection would alter the expression of galectins (Gal11 and Gal14) and collectins (SPA and CGN) in sheep. Twelve Corriedale sheep exposed to natural infection with nematodes were divided into two groups: group 1 (G1, n = 7) and group 2 (G2, n = 5), with low and high parasite burdens, respectively, based on fecal egg counts and abomasal parasite counts. The fecal egg counts and abomasal parasite counts were significantly different (p < 0.05) between the groups. Galectin and collectin gene expression was observed in all sheep abomasal samples. However, animals with lower infection levels showed lower expression of the genes Gal14, SPA and CGN (p < 0.05). Expression of lectins was associated with the abomasal H. contortus burden, thus suggesting that these proteins may have a role in controlling of this infection.

A neosporose é uma doença causada pelo protozoário Neospora caninum que leva a perdas econômicas importantes em muitos países. No presente estudo, é descrita a utilização da proteína recombinante NcSRS2 de N. caninum expressa em Pichia pastoris em um ensaio imunoenzimático indireto (ELISA) para o diagnóstico de infecção por Neospora em ovelhas e cães. Observou-se, que utilizando-se um ELISA, o teste produziu uma especificidade de 94,5% e uma sensibilidade de 100% para ovinos; e uma especificidade de 93,3% e sensibilidade de 100% para cães. Uma maior sensibilidade foi observada em relação à IFI que foi confirmada por Western blot. Os resultados deste estudo sugerem que a proteína recombinante expressa em P. pastoris é bom antígeno para ser utilizado no diagnóstico imunológico para detectar N. caninum em duas espécies importantes expostas a esta parasitose

Neosporosis is a disease caused by the protozoon Neospora caninum that leads to significant economic losses in many countries. In the present study, we report on use of the recombinant protein NcSRS2 of N. caninum expressed in Pichia pastoris in an indirect immunoenzymatic assay (ELISA) for diagnosing neosporosis infection in sheep and dogs. We observed that the ELISA test yielded specificity of 94.5% and sensitivity of 100% for sheep and specificity of 93.3% and sensitivity of 100% for dogs. We observed that the sensitivity was higher than shown by the indirect fluorescent antibody test, and this was confirmed by means of Western blot. The results from this study suggest that the recombinant protein expressed in P. pastoris is a suitable antigen for use in immunodiagnosis to detect N. caninum in two important species exposed to this parasitosis.

A toxocaríase é zoonose amplamente difundida e considerada importante problema de saúde pública. O objetivo deste estudo foi avaliar a frequência da transmissão transmamária de Toxocara canis em camundongos BALB/c neonatos amamentados por fêmeas experimentalmente infectadas com 1.200 ovos logo após o parto. Após 50 dias de idade, foi avaliada a presença de larvas em diferentes órgãos dos neonatos. A infecção por via transmamária foi confirmada em 73,9% dos camundongos amamentados por fêmeas infectadas. Estes dados demonstram elevada transmissão transmamária de T. canis e confirmam a importância desta via de transmissão em hospedeiros paratênicos.

Toxocariasis is a widespread zoonosis and is considered an important worldwide public health problem. The aim of this study was to investigate the frequency of trans-mammary Toxocara canis infection in newborn BALB/c mice nursed by females experimentally infected with 1,200 eggs after delivery. After 50 days of age, the presence of larvae in different organs of the offspring was investigated. Trans-mammary infection was confirmed in 73.9% of the mice that had been nursed by infected females. These data show a high trans-mammary transmission of T. canis and confirm the significance of this transmission route in paratenic hosts.

A toxocaríase é uma zoonose na qual os títulos de IgM podem permanecer elevados por longos períodos, tornando difícil a determinação do estágio em que a doença se encontra. O objetivo deste estudo foi investigar a aplicabilidade de um teste indireto de ELISA, associado com ureia, para fazer a discriminação entre as fases aguda e crônica da toxocaríase. A avidez de IgG foi avaliada em 25 camundongos BALB/c experimentalmente infectados com 1000 ovos embrionados de Toxocara canis. A cada duas semanas, amostras de sangue foram coletadas, o soro tratado com ureia 6M e realizado o ensaio pela técnica de ELISA. O percentual de avidez de IgG foi determinado, usando-se a média das absorbâncias dos soros tratados com ureia dividida pela média das absorbâncias dos soros não tratados. Nos primeiros 15 dias pós-inoculação, foi observado um baixo percentual de avidez de IgG, entre 7,25 e 27,5%, característico da fase aguda da infecção. Após 60 dias de infecção, todos apresentaram avidez de IgG entre 31,4 e 58%, indicando a fase crônica da infecção.

Toxocariasis is a zoonotic disease in that IgM titers can remain high for long periods making difficult to determine the stage of the disease. The aim of this study is to investigate the applicability of indirect ELISA, associated with urea, to discriminate between the acute and chronic toxocariasis. IgG avidity was evaluated in 25 BALB/c mice experimentally infected with 1000 Toxocara canis eggs. Blood samples were collected, and sera treated with 6 M urea and assayed by ELISA every two weeks. The percent IgG avidity was determined using the mean absorbance of sera treated with urea, divided by the mean absorbance of untreated sera. In the first 15 days post-inoculation, was observed a low percentage, between 7.25 and 27.5%, IgG avidity, characteristic of an acute infection. After 60 days of infection, all the mice showed between 31.4 and 58% IgG avidity, indicating a chronic infection.

Em virtude da resistência dos nematódeos gastrintestinais aos antihelmínticos, diversas pesquisas têm sido desenvolvidas buscando-se alternativas de controle para essas parasitoses. Nesse contexto, o estudo avaliou a ação in vitro de Origanum vulgare sobre ovos de nematódeos gastrintestinais de bovinos. Para avaliar a capacidade de inibição da eclodibilidade dos ovos, diferentes formas de extratos das folhas secas desta planta foram testadas como tintura, extrato hidroalcoólico e extrato aquoso nas concentrações de 0,62 a 80 mg/mL. Cada ensaio foi acompanhado de controle contendo cloridrato de levamisol (0,2 mg/mL), água destilada e álcool de cereais 70° GL nas mesmas concentrações dos produtos. Os resultados dos testes demonstraram que os diferentes extratos de O. vulgare inibiram a eclodibilidade dos ovos de nematódeos gastrintestinais de bovinos com percentual de inibição variando de 8,8 a 100%, sendo a tintura e o extrato hidroalcoólico as formas mais promissoras. Frente a esta propriedade ovicida, o O. vulgare pode representar uma importante fonte de compostos antiparasitários viáveis para o controle das nematodioses em ruminantes.

Due to anthelmintic resistance in nematodes, several research studies have been developed seeking control alternatives to these parasites. This study evaluated the in vitro action of Origanum vulgare on gastrointestinal nematode eggs of cattle. In order to evaluate the ability to inhibit egg hatch, different dried leaves extracts of this plant were tested, such as dye, hydroalcoholic and aqueous extracts at concentrations varying from 0.62 to 80 mg/mL. Each assay was accompanied by control containing levamisole hydrochloride (0.2 mg/mL), distilled water and 70º GL grain alcohol at the same concentration of the extracts. Test results showed that the different O. vulgare extracts inhibited egg hatch of cattle gastrointestinal nematodes at a percentage that varied from 8.8 to 100%; dye and hydroalcoholic extract were the most promising inhibitors. In view of this ovicidal property, O. vulgare may be an important source of viable antiparasitic compounds for nematodiosis control in ruminants.

A toxocaríase visceral é uma zoonose negligenciada causada por larvas de Toxocara canis em hospedeiros não usuais. Em cães, os hospedeiros definitivos, a infecção ocorre normalmente por transmissão transplacentária e através do colostro. Estudos com modelos experimentais têm demonstrado a ocorrência de transmissão vertical durante a infecção aguda. Considerando que a toxocaríase é caracterizada como uma infecção crônica, com uma possível reativação das larvas presentes no cérebro, este estudo avaliou a presença de larvas no cérebro de camundongos Balb/C fêmeas e suas proles com infecção crônica durante três gestações sucessivas. Para avaliar os níveis de anticorpos foi utilizado ELISA-TES. Larvas de T. canis foram detectadas no encéfalo dos animais durante as três gerações sucessivas avaliadas. O nível de IgG das proles foi diminuindo gradualmente e as médias de absorbâncias (ABS) acima do ponto de corte (0,070) foram evidenciadas somente aos 30 (0,229) e 50 dias (0,096) de vida, enquanto que não foi detectada IgM. Infecções das proles confirmam a transmissão vertical de larvas de T. canis durante a toxocaríase crônica em três gerações sucessivas de camundongos.

Visceral toxocariasis is a neglected zoonosis caused by Toxocara canis larvae in unusual hosts. In dogs, the definitive host, the infection occurs mainly through transplacental and transcolostral transmission. Studies on experimental models have shown that vertical transmission may result from acute infections. Considering that toxocariasis is characterized as a chronic infection, with possible reactivation of larvae present in the brain, this study evaluated the presence of larvae in the brain of female BALB/c mice and their offspring with chronic infection during three successive pregnancies. ELISA-TES was used to evaluate the antibody levels. T. canis larvae were detected in the brain tissue of the mice during the three successive generations evaluated. The offspring's IgG level gradually decreased, and mean absorbance (ABS) above the cutoff point (0.070) was observed only at 30 (0.229) and 50 (0.096) days of age, while IgM was not detected. The infections in the offspring confirmed that vertical transmission of T. canis larvae occurred during chronic toxocariasis in three successive generations of mice.

Results of Chagas' disease diagnosis show disagreement. The aim of this study was to compare commercial tests for Chagas' disease serodiagnosis in southern Brazil. A total of 161 samples were evaluated. Three enzyme-linked immunosorbent assays, one indirect hemagglutination and one indirect immunofluorescence were assessed. Trypomastigote excreted-secreted antigen-blot was a confirmatory method. From 161 samples, 65.84% were positive in all tests, while 34.16% presents mismatch result in at least one of the tests. All techniques tested presented false-positive and/or false-negative results as follows: Enzyme-linked immunosorbent assay 1 had more false-positive results (lower specificity), indirect immunofluorescence had the highest rate of false-negative results (lower sen sitivity), enzyme-linked immunosorbent assays had fewer false-negative results (higher sensitivity), while indirect hemagglutination showed no false-positive result (higher specificity). Knowing the characteristics of techniques make it possible to combine them and obtain more reliable diagnosis. Therefore, it seems useful to combine techniques for diagnosing this infection.

A frequência de contaminação parasitária de áreas públicas de Pelotas, Rio Grande do Sul, Brasil, foi avaliada entre junho de 2010 e maio de 2011, com coletas mensais de amostras de solo de oito praças. Das 400 amostras submetidas à técnica de centrífugo-flutuação em solução de dicromato de sódio com densidade de 1,35, 176 (44%) apresentaram pelo menos uma forma parasitária e, das amostras positivas, 29 (16,5%) estavam poliparasitadas. Os resultados demonstraram relevante índice de contaminação do solo em todas as praças avaliadas, com maiores índices de positividade para ovos de ancilostomídeos (13,5%) e ovos deToxocara (8,8%), sendo também identificados ovos de Trichuris, Ascaris eCapillaria. O estudo demonstrou a contaminação ambiental de praças públicas e os riscos a que a população está exposta em relação a doenças causadas por geoparasitos zoonóticos e sugere que medidas de saneamento e educação em saúde devem ser implementadas no município.

The frequency of parasitic contamination of public areas in the municipality of Pelotas, state of Rio Grande do Sul, Brazil, was studied between June 2010 and May 2011, when soil samples were collected from eight city squares. Out of 400 samples submitted to centrifugal floatation technique in solution of sodium dichromate with density of 1.35, 176 (44%) proved positive for at least one parasite; 29 (16.5%) samples were multi-infested. The results showed that there was a significant soil contamination rate in all the parks included in the study. The positivity rate was higher for hookworms eggs (13.5%) andToxocara eggs (8.8%);Trichuris, Ascaris andCapillaria eggs were also detected. This study shows the risks to which the population is exposed in relation to zoonotic geohelminths, and suggests that sanitation and health education measures should be implemented in the municipality.

A contaminação ambiental por formas parasitárias foi estudada na área do campus universitário no município de Pelotas, Rio Grande do Sul. Amostras de solo foram analisadas para identificação de formas parasitárias e a presença de ovos de nematóides foi verificada em 62% das amostras. Entre os parasitos observados destacam-se a presença de ovos de Toxocara spp. e de ancilostomatídeos em todo período estudado. Os resultados demonstram que há contaminação ambiental significativa, o que representa risco de infecção zoonótica aos humanos que frequentam a região estudada.

Environmental contamination by parasite forms was studied on a university campus in the municipality of Pelotas, Rio Grande do Sul. Soil samples were analyzed using the modified Caldwell & Caldwell technique to identify parasite forms. Nematode eggs were detected in 62% of the samples. Among the parasites detected, eggs of Toxocara spp. and Ancylostomatidae were the most prevalent parasites in the studied area throughout the study period. The results demonstrated that there is significant environmental contamination, thereby representing a risk of zoonotic infection for humans frequenting the study area.

Anticorpos antiTrypanosoma cruzi no cordão umbilical de 351 parturientes da Cidade de Pelotas, RS foram pesquisados a fim de investigar a prevalência da doença de Chagas em gestantes. Um (0,3%) caso foi identificado, não sendo detectada transmissão congênita. Salienta-se a importância da investigação da doença de Chagas em gestantes de zonas endêmicas ou provenientes destas.

Anti-Trypanosoma cruzi antibodies in the umbilical cord of 351 parturients in the city of Pelotas, Rio Grande do Sul were investigated to determine the prevalence of Chagas disease among pregnant women. One case was identified (0.3%), without detection of congenital transmission. This highlights the importance of investigating Chagas disease among pregnant women living in or originating from endemic areas.
