Resumo A hipótese da síndrome da polinização geralmente não se refere exatamente à diversidade de fenótipos florais ou ajuda a predizer os polinizadores da maioria das espécies de plantas. Em Bromeliaceae, podemos encontrar uma ampla gama de visitantes florais, tornando suas espécies ideais para testar uma correlação entre características florais e do néctar com síndromes de polinização. Neste estudo, analisamos características florais, padrões de produção de néctar, polinizadores e visitantes florais em três populações naturais da Mata Atlântica de Vriesea gigantea, e discutimos seu potencial adaptativo e significado ecológico. A espécie apresentou protoginia e hercogamia, e sua antese ocorreu em diferentes períodos entre as populações. Vriesea gigantea tem uma taxa constante de produção de néctar durante o dia, com uma quantidade reduzida durante a noite. Flores recém-abertas possuem um considerável acúmulo de néctar. Embora a espécie tenha sido classificada como quiropterófila com base na morfologia das flores e observação de polinizadores, nossos resultados mostraram que os beija-flores são os polinizadores prováveis nas populações estudadas de V. gigantea.

Abstract The pollination syndrome hypothesis usually does not successfully apply to the diversity of floral phenotypes or help predict the pollinators of most plant species. In Bromeliaceae, there is a wide range of floral visitors, making its species ideal to test for a correlation between nectar and floral traits with pollination syndrome. In this study, we analyzed the floral features, nectar production patterns, pollinators and floral visitors of Vriesea gigantea, and discussed its potential adaptive and ecological significance. We study three natural populations from the Atlantic Forest, Southern Brazil. The species presented protogyny and herkogamy, and its anthesis occurred at different periods among different populations. Vriesea gigantea has a relatively constant rate of nectar production during the day that continues overnight but at a reduced rate. Newly opened flowers already have around 80.0 μl of nectar. Although classified as chiropterophilous, based on flower morphology and pollinator observations, our results show that hummingbirds are effective pollinators in the studied populations of V. gigantea.

Abstract The Southern Brazilian Highlands are composed by a mosaic of Mixed Ombrophilous Forest (MOF) and grassland formations, an interesting landscape for the study of population structure. We analyzed the genetic diversity within and among populations of the MOF-endemic bromeliad Vriesea reitzii by genotyping seven nuclear microsatellite loci in 187 individuals from six populations. We characterized levels of genetic diversity and assessed the genetic structure among populations. Vriesea reitzii populations showed high levels of genetic variation (number of alleles 28 - 43, allelic richness 3.589 - 5.531) and moderate levels of genetic differentiation (FST = 0.123, RST = 0.096). The high levels of genetic diversity may be explained by species life-history traits, such as habit and mating system. The moderate structure may be a product of the combination of ancient and contemporary gene flow, resulting from the expansion of the forest in the Holocene, and/or due to facilitated dispersal mediated by the MOF’s mosaic landscape. The genetic results indicated no imminent threat to this bromeliad. However, the species is highly associated with the MOF, putting landscape conservation at the center of conservation efforts for the species’ maintenance.

Bromeliaceae is a morphologically distinctive and ecologically diverse family originating in the New World. Three centers of diversity, 58 genera, and about 3,140 bromeliad species are currently recognized. We compiled all of the studies related to the reproductive biology, genetic diversity, and population structure of the Bromeliaceae, and discuss the evolution and conservation of this family. Bromeliads are preferentially pollinated by vertebrates and show marked variation in breeding systems, from predominant inbreeding to predominant outcrossing, as well as constancy in chromosome number (2n = 2x = 50). Autogamous or mixed mating system bromeliads have a high inbreeding coefficient (F IS), while outcrossing species show low F IS. The degree of differentiation among populations (F ST) of species ranges from 0.043 to 0.961, which can be influenced by pollen and seed dispersal effects, clonal growth, gene flow rates, and connectivity among populations. The evolutionary history of the Bromeliaceae is poorly known, although some studies have indicated that the family arose in the Guayana Shield roughly 100 Mya. We believe that genetic, cytogenetic, and reproductive data will be essential for diagnosing species status and for assisting conservation programs.

Espécies silvestres são fontes importantes de variabilidade genética e podem ser exploradas pelos programas de melhoramento. Cruzamentos entre teosinto e milho ocorrem naturalmente, e o teosinto pode ser utilizado como fonte de caracteres agronômicos para introdução em milho. O objetivo deste estudo foi estimar a variabilidade genética entre e dentro de populações de milho e teosinto (Zea mays mexicana). Duas populações de milho doce (BR400 e BR402), duas de milho comum (Suwan e Pampa) e uma de teosinto foram analisadas utilizando-se marcadores microssatélites. Os resultados indicaram que 64,5% da variação foi detectada dentro das populações, sugerindo a possibilidade de obtenção de progresso genético através da seleção dentro de cada população. A análise de 25 locos microssatélites permitiu identificar 92 alelos, com uma média de 3,7 alelos por loco. O percentual de locos polimórficos variou de 64% nas populações BR400 e Pampa a 80% na população de teosinto. A distância genética estimada confirmou a similaridade genética entre milho e teosinto.

Wild species are important sources of genetic variability and may be exploited by breeding programs. Crosses between teosinte and maize occur freely and teosinte serves as genetic source of agronomic traits for introduction in maize. The objective of this study was to estimate genetic variability among and within maize and teosinte populations (Zea mays mexicana). Two sweet maize populations (BR400 and BR402), two common maize populations (Suwan and Pampa) and one teosinte population were analyzed using microsatellites markers. Results indicated that 64,5% of the variation was detected within the populations, suggesting the possibility of obtaining genetic progress by selection within each population. The analysis with 25 microsatellites loci enabled the identification of 92 alleles with a mean of 3.7 alleles per locus. The average Polymorphism Information Content (PIC) was 0.52. The percentage of polymorphic loci varied from 64% in the BR400 and Pampa populations to 80% in the teosinte population. The estimated genetic distance confirmed the genomic similarity of maize and teosinte.

A utilização do milho (Zea mays) na cultura de anteras é limitada devido ao baixo número de genótipos com alto potencial androgenético. A obtenção de micrósporos no estádio de desenvolvimento apropriado no início da cultura é um dos fatores cruciais para o sucesso do processo androgenético. Em genótipos brasileiros de milho não existem estudos relatando a correlação entre características citológicas e parâmetros morfológicos. Neste estudo, parâmetros morfológicos foram avaliados e associados com estádios específicos da microsporogênese em dois genótipos brasileiros de milho os quais foram semeados em diferentes locais (campo e câmara de crescimento). Para ambos os genótipos, as plantas crescidas na câmara de crescimento apresentaram atraso no desenvolvimento. O comprimento da espigueta e a razão comprimento da antera/comprimento da espigueta não são bons parâmetros uma vez que podem ser muito influenciados pelo ambiente. O comprimento da antera foi o melhor parâmetro para indicar o estádio de desenvolvimento do micrósporo. Todavia, variações entre genótipos e ambiente foram detectadas.

The use of maize in anther culture has been limited because only few genotypes presented a high androgenetic potential. Obtaining the proper stage of microspore development at culture initiation is one of the most crucial factors for success in the androgenesis. For Brazilian maize genotypes there are no studies reporting a correlation between cytological features and morphological parameters. In this study, morphological parameters were recorded and associated with cytological specific stages of the the microsporogenesis in two Brazilian maize genotypes that were sowed in different places (field and growing chamber). For both genotypes, the plants of the growing chamber presented a delay in development. Spikelets length and anther length/spikelet length ratio are not good parameters since they can be greatly influenced by the environment. The anther length was the more reliable parameter to correlate with a specific developmental stage. Nevertheless, variations between genotypes and environment were detected.

Angiostrongilíase abdominal é uma zoonose causada pelo Angiostrongylus costaricensis, nematódeo que se localiza no interior de vasos mesentéricos. Nosso objetivo foi de abordar vários aspectos da história natural da parasitose, num estudo longitudinal clínico-sorológico. Um total de 179 indivíduos residentes em área rural no sul do Brasil, com transmissão ativa, foram seguidos por cinco anos. Neste período foram registradas prevalências de 28,2%, 4,2%, 10%, 20,2% e 2,8% e incidências de 0%, 5,9%, 8% e 1,5%. Tanto o sexo masculino quanto o feminino foram afetados com maiores frequências na faixa etária dos 30 aos 49 anos. Em 32 indivíduos, amostras de soro foram coletadas em todas as etapas e a reatividade de IgG detectada por ELISA foi variável e geralmente não persistindo mais do que um ano. Alguns padrões individuais foram sugestivos de re-infecção. Não houve associação com a ocorrência nem de dor abdominal nem com outras enteroparasitoses e não houve nenhum caso com diagnóstico confirmado (histopatológico) da infecção. Moluscos foram encontrados portando larvas infectantes de terceiro estadio, em algumas moradias, com uma prevalência geral de 16% e baixas cargas parasitárias. Em conclusão, a angiostrongilíase abdominal no sul do Brasil pode ser uma infecção frequente, porém com baixa morbidade e reatividade sorológica de gradual declínio.

Abdominal angiostrongyliasis is a zoonotic infection caused by Angiostrongylus costaricensis, a nematode with an intra-vascular location in the mesentery. Our objective was to address several aspects of the natural history of this parasitosis, in a longitudinal clinical and seroepidemiological study. A total of 179 individuals living in a rural area with active transmission in southern Brazil were followed for five years (1995-1999) resulting in yearly prevalence of 28.2%, 4.2%, 10%, 20.2% and 2.8% and incidences of 0%, 5.9%, 8% and 1.5%, respectively. Both men and woman were affected with higher frequencies at age 30-49 years. In 32 individuals serum samples were collected at all time points and IgG antibody reactivity detected by ELISA was variable and usually persisting not longer than one year. Some individual antibody patterns were suggestive of re-infection. There was no association with occurrence of abdominal pain or of other enteroparasites and there was no individual with a confirmed (histopathologic) diagnosis. Mollusks were found with infective third-stage larvae in some houses with an overall prevalence of 16% and a low parasitic burden. In conclusion, abdominal angiostrongyliasis in southern Brazil may be a frequent infection with low morbidity and a gradually decreasing serological reactivity.

A caracterização de variabilidade genética é essencial para qualquer programa de melhoramento. O conhecimento dos genótipos permite que o melhorista planeje seus cruzamentos objetivando a ampliação da variância genética. O objetivo deste trabalho foi caracterizar e agrupar o germoplasma de trigo disponível para o melhorista do Sul do Brasil através dos caracteres adaptativos ciclo e estatura. Foram avaliados 53 genótipos e evidenciada uma redução significativa dos dois caracteres analisados no decorrer das últimas décadas. O agrupamento dos genótipos revelou a formação de cinco grupos principais, constituídos, basicamente, de cultivares da mesma época de recomendação para cultivo.

Identification of genetic variability is crucial in any breeding program. The knowledge of genotypes helps the breeder in planning crosses aiming to increase genetic variance. The objective of this study was to identify and cluster useful wheat germplasm through the adaptive characters heading date and plant height. Fifty-three wheat genotypes which revealed a significant decrease of both characters in the last years were evaluated. The dendrogram clustered five groups composed mainly of cultivars of the same era of release.

The characterization of genetic variability and an estimate of the genetic relationship among varieties are essential to any breeding program, because artificial crosses among less similar parents allow a larger segregation and the combination of different favorable alleles. Genetic variability can be evaluated in different ways, including the Coefficient of Parentage (COP), which estimates the probability of two alleles in two different individuals being identical by descent. In this study, we evaluated the degree of genetic relationship among 53 wheat genotypes, and identified the ancestor genotypes which contributed the most to the current wheat germplasm, as a prediction of the width of the genetic base of this cereal. The results revealed a mean COP of 0.07 and the formation of 22 similarity groups. The ancestor genotypes Ciano 67 and Mentana were those which contributed the most to the current wheat germplasm. According to the COP analyses, the genetic base of wheat rests on a small number of ancestral genotypes.

A helmintosporiose é uma moléstia que afeta a cultura da aveia, reduzindo seu rendimento e qualidade de grão. A seleção de genótipos resistentes com utilização de filtrados tóxicos do fungo Helminthosporium sativum é eficaz, pois delimita a variabilidade do patógeno e reduz a interferência do ambiente na expressão do genótipo. Contudo, a obtenção dos filtrados tóxicos deste fungo é um processo lento e delicado. Dessa forma, a possibilidade do uso de substâncias sintéticas que simulem seu efeito, inibindo o transporte de elétrons da cadeia respiratória, é de grande interesse. O inseticida Methomyl é eficaz para simular o efeito da toxina do fungo que causa a helmintosporiose em milho, tendo o presente trabalho visado testar sua eficiência na cultura da aveia. Para isso, foi avaliado o crescimento de calos e raízes de aveia expostas ao Methomyl, quando crescimentos maiores indicaram maior resistência ao produto, e possível resistência à moléstia. Os resultados indicam que o Methomyl afeta o crescimento de raízes e calos de aveia e pode ser utilizado para separar os grupos de genótipos com e sem resistência à helmintosporiose. Assim, UFRGS 14, com maior sensibilidade aos filtrados tóxicos de H. sativum em outros estudos, também apresentou crescimento mais afetado pelo Methomyl em todos os experimentos aqui conduzidos.

Spot Blotch is a plant disease which causes yield losses in oat and other cereals. Selection for resistant genotypes using Helminthosporium sativum toxic filtrates is an efficient technique, which reduces the pathogen variability and the influence of the environment over the genotype expression. The filtrates extraction is time consuming and a difficult process, and the possibility of using a synthetic product to simulate its action of inhibiting the cell electron transport chain would be useful. The Methomyl insecticide is an efficient product to simulate the effects of the fungus that causes spot blotch in corn. The objective of this study was to check its efficiency in oat. Oat roots and callus growth were evaluated in the insecticide presence. Growth values that were less inhibited by the insecticide were used to correlate with spot blotch resistance. The results indicate that Methomyl reduces the growth of oat root and calli and may be used to differentiate genotypes more and less resistant to Spot Blotch. Therefore, UFRGS 14, which was the most affected genotype in the presence of the fungus toxic filtrates, had also the most reduced root and calli growth in the presence of Methomyl.

Na implantação de um programa de melhoramento, uma das principais necessidades do melhorista é a capacidade de identificar genótipos superiores em uma população segregante. O conhecimento das relações genéticas e a capacidade geral e específica de combinação entre os indivíduos é essencial para a seleção de genitores. Os marcadores genéticos poderão auxiliar na identificação de indivíduos através de suas diferenças genéticas. Estes marcadores podem ser divididos em morfológicos e moleculares (enzimáticos e de DNA). Os marcadores de DNA como o RFLP e o RAPD poderão contribuir para incrementar a eficiência do melhoramento de plantas através do mapeamento de espécies de interesse e de caracteres agronômicos. Além disto, diversos autores têm comprovado a sua eficiência em caracterizar e agrupar genótipos diferentes de várias espécies com bastante precisão.

One of the most important factor in a plant breeding program is the capacity of the breeder to identify superior genotypes in a segregant population. The understanding of genetic relationships and combining ability among individuals is essential for parental selection. Genetic markers are divided in morphological and molecular (Enzyme or DNA). They may help in the identification of individuals based on their genetic similarities. DNA markers, as RFLP and RAPD, contribute to increase efficiency in plant breeding through linkage mapping and agronomic traits mapping. In addition, several researchers have pointed out the efficiency of these markers to characterize genotypes and to generate clusters of genetic similarity in germplasm from different species.

Nove genótipos de aveia foram cultivados in vitro com o objetivo de formação de calos e posterior avaliação de embriogênese somática. Os calos foram testados em três protocolos (meio MS- MURASHIGE & SKOOG (1962) com diferentes dosagens de hormônios). Após um mês em meio de subcultivo, os calos foram avaliados quanto à porcentagem de embriogênese somática. Os protocolos testados revelaram respostas diferentes na indução de embrióides, proporcionando a escolha do que fosse mais adequado para esta função. No primeiro experimento não foi obtida diferença entre os genótipos, por outro lado, no experimento 2, a UFRGS 7 e UFRGS 8 possuíram médias significativamente maiores. A análise de covariância realizada com o objetivo de verificar se o tamanho de embrião estava envolvido na indução de embriogênese evidenciou que para alguns genótipos esta medida é importante.

Nine oat genotypes were cultivated in vitro to evaluate callus initiaton and subsequent somatic embryogenesis. The immature embryo were submited to different protocola (MURASHIGE & SKOOG (1962) médium with differents hormones dosages). The protocols tested caused differences in somatic embryogenesis, and the best of them was selected to continue the research. After a month in subculture médium the cali were evaluated concerning embryoid porcentage, and showed differences according to genotype in experiment 2, UFRGS 7 and UFRGS 8 presented superior means. The covariance analysis revealed that in some genotypes this parameter is important.

Existem variações entre genótipos de trigo quanto à regeneração de plantas in vitro. Em trabalho precursor com o genótipo Maringá obteve-se 100% de regeneração de plantas. Este trabalho visou investigar a capacidade de regeneração de plantas do Maringá e duas linhas isogênicas com um e dois genes de estatura reduzida (Rht1 e Rht2). Foram utilizados embriões imaturos de Maringá, das linhas isogênicas e das F2's dos cruzamentos entre estes genótipos, que foram colocados em meio de cultura MS modificado (MILACH et al., 1991) com sacarose, ágar e doses decrescentes de 2,4D conforme a etapa de cultivo (2mg/l; 0,5mg/l; 0,1mg/l e sem 2,4D). Também foi avaliada a possibilidade de supressão de um subcultivo, quando os calos não passariam pelo meio com 0,1mg/l de 2,4D. As análises estatísticas demonstraram que os genótipos diferiram quanto à regeneração de plantas, evidenciando a influência do gene Rht, que reduziu o número de plantas regeneradas por calo. A retirada do meio com 0,1mg/l de 2,4D não alterou a regeneração, podendo-se eliminar esta etapa de cultivo. Os genótipos responderam da mesma forma nas diferentes sequências de subcultivo, não havendo interação entre estes dois caracteres.

Different genotypes show different responses for in vitro plant regeneration. In previous work with lhe Maringá genotype a result of 100% of in vitro plant regeneration was obtained. The purpose of this study was to investigate the capacity of plant regeneration of Maringá genotype and two of it's isogenic lines with one and two shorts heigth genes (Rht1 and Rht2). Imature embryos from: (a) Maringá, (b) the isogenic lines and (c) the crosses between these genotypes were placed on MS medium (modified by MILACH et al., 1991) with saccharose, agar and decreasing doses of 2,4D (2mg/l; 0.5mg/l; 0.1mg/l and 0.0mg/l) according to the stage of culture. The possibility of suppressing one of the stages of culture, (which uses 0.1mg/1 of 2.4D) was also evaluated. The statistic analisys demonstrated that the different genotypes gave different responses, therefore this indicates that the Rht genes had influence on plant regeneration rates. The suppression of the stage of culture which uses 0.1mg/l of 2,4D did not have any influence on regeneration, showing that this stage can be eliminated. Different genotypes had the same responses on the different culture medium sequences. The intercection between genotype x medium was not significam.
