O objetivo deste estudo foi avaliar as mudanças na atividade de pectinases (pectinesterase e poligalacturonase), teor de fibras dietéticas (sólidos insolúveis em álcool, pectina e fibra total) e índices físico-químicos relacionados ao sabor (pH, acidez titulável, sólidos solúveis, relação Brix/acidez, açúcares redutores e não-redutores) de frutos de cubiu (Solanum sessiliflorum Dunal) em diferentes estádios de maturação (verde, de vez, maduro e muito maduro). Os sólidos insolúveis em álcool e pectina apresentaram perfis muito similares, com teores maiores nos estádios verde e de vez, e menores no estádio muito maduro. Teores de fibra total mostraram-se uniformes nos estádios verde, de vez e maduro, diminuindo no estádio muito maduro. Essas mudanças correlacionaram-se com a atividade das pectinases, cujos perfis assemelharam-se àqueles de outras espécies da família Solanaceae durante a maturação dos frutos. No entanto, os frutos de cubiu apresentaram teores significativos de fibras dietéticas em todos os estádios. O conteúdo de açúcares redutores atingiu teores maiores no estádio de vez, sendo a glicose o açúcar principal. O conteúdo de açúcares não-redutores, como a sacarose, permaneceu baixo em todos os estádios. Os demais índices físico-químicos apresentaram aumento durante a maturação, caracterizando o cubiu como um fruto muito ácido com pequeno grau de doçura.

The objective of this study was to evaluate the changes in the activity of pectinases (pectinesterase and polygalacturonase), dietary fiber content (alcohol-insoluble solids, pectin, and total fiber), and physicochemical indices related to the flavor (pH, titratable acidity, soluble solids, Brix/acid ratio, and reducing and nonreducing sugars) of cubiu fruits (Solanum sessiliflorum Dunal) at different stages of ripening (green, turning, ripe, and fully ripe). Alcohol-insoluble solids and pectin were very similar, with the highest levels detected at the green and turning stages, and the lowest levels occurring at the fully ripe stage. The amount of total fiber was consistent at the green, turning, and ripe stages, but declined at the fully ripe stage. These changes correlated with the pectinase activities, the profiles of which resembled those of other species of the Solanaceae family during fruit ripening. However, cubiu fruits were significant sources of dietary fiber at all stages. The reducing sugar content reached the highest level at the turning stage, with glucose as the major sugar. The content of nonreducing sugars, such as sucrose, remained low at all stages. The other physicochemical indices displayed increases during ripening, characterizing cubiu as a very acidic fruit with a small degree of sweetness.

A desidratação osmótica é uma operação unitária importante para transformar os frutos perecíveis em novos produtos com valor agregado e com maior vida de prateleira. A aplicação dos modelos matemáticos confiáveis permite prever o comportamento dos diversos fenômenos que ocorrem durante o processo de secagem, o que implica na redução do custo operacional. A polpa do cupuaçu é comumente usada para preparar sucos e sorvetes, mas durante a safra é produzida em grande quantidade, que muitas vezes, é descartada devido ao custo de produção ser mais alto do que o da comercialização. O objetivo desse estudo foi investigar o efeito da solução binária e da ternária na taxa da secagem por convecção e avaliar a adequação dos modelos matemáticos na descrição das curvas da secagem do cupuaçu. No experimento, a pré-desidratação foi feita com solução binária (sacarose a 40 ºBrix) e ternária (80% de sacarose/20% de cloreto de sódio), durante três horas em temperatura ambiente, seguida pela secagem convencional na estufa (ar circulante) a 65 ºC, até peso constante. Os dados experimentais foram ajustados utilizando os modelos de Page e Midilli, Kucuk e Yapar. A solução binária usada na pré-secagem reduziu o tempo da subseqüente secagem convencional. Com variação de 99,64 a 99,77% no coeficiente de determinação (R²), o modelo de Page apresentou o melhor ajuste aos dados experimentais.

The osmotic dehydration is an important unitary operation to transform the perishable fruits in new products with added value and longer shelf life. The application of reliable mathematical models allows prediction the behavior of various phenomena that occur during the drying process, which implies in reduction of operating cost. The cupuaçu pulp is commonly used to prepare juices and ice creams, but during the crop it is produced in large quantity, which is often discarded because of the cost of production is higher than the selling price. The aim of this study was to investigate the effect of binary and ternary solutions at the rate of convective drying and evaluate the adequacy of mathematical models to describe the drying curves of the cupuaçu. In the experiment, osmotic dehydration was carried out with binary (sucrose at 40 ºBrix) and ternary (80% sucrose/20% sodium chloride) solutions for three hours at room temperature, followed by conventional drying in an oven (air-circulating) at 65 °C to constant weight. The experimental data were adjusted using the models of Page and Midilli, Kucuk and Yapar. The binary solution used during pre-drying, reduced the time of the subsequent conventional drying. Varying from 99.64 to 99.77% in the coefficient of determination (R²), the Page model showed a better adjustment to experimental data.

Cubiu (Solanum sessiliflorum Dunal) é um fruto amazônico cujo interesse decorre do alto teor de pectina. Atualmente, tecnologias de processamento são necessárias para a transposição do sistema tradicional (pequeno cultivo e processamento artesanal) para o de escala maior e, assim, aumentar o uso da biodiversidade e viabilizar os Arranjos Produtivos Locais e o agronegócio na Amazônia. Esta pesquisa tem o objetivo de avaliar os métodos para o descascamento do cubiu. Frutos maduros foram divididos em lotes (150 frutos cada) e submetidos aos seguintes tratamentos: imersão em solução (fervente) de NaOH a 2,5%, exposição ao vapor de água e imersão em água a 96 ºC por 5, 10, 15 e 20 minutos. A casca liberada durante os tratamentos térmicos foi removida sob a água corrente. No controle, os frutos foram descascados (sem aquecimento) com faca de aço inoxidável. Os tratamentos foram avaliados pela totalidade e facilidade do descascamento, integridade dos tecidos, textura e atividade da peroxidase. O tratamento em solução fervente de NaOH 2,5% (5 minutos) destacou-se como o melhor tratamento uma vez que inibiu o escurecimento enzimático, intensificou a cor natural amarela do fruto, descascou inteira e facilmente todos os frutos e não danificou a integridade dos tecidos. Este tratamento foi escolhido e recomendado para o processamento industrial do cubiu por promover simultaneamente o descascamento e branqueamento do fruto.

Cubiu (Solanum sessiliflorum Dunal) is an Amazonian Basin native fruit. Its importance comes from its high contents of pectin. Currently, processing technologies are necessary for the substitution of the traditional system (small crops and small-scale processing) for a larger scale system and thus increase the use of biodiversity and promote the implementation of Local Productive Arrangements of agribusiness in the Amazon. This research aims to evaluate the methods of peeling cubiu. Ripe fruits were divided into lots (150 each) and subjected to the following treatments: immersion in 2.5% NaOH boiling solution for 5 minutes, exposure to water vapor, and immersion in water at 96 ºC for 5, 10, 15 and 20 minutes. The peel released during heat treatment and immediately removed under running tap water. In the control treatment, the fruits were manually peeled (unheated) with a stainless steel knife. The treatments were evaluated for completeness and ease of peeling, tissue integrity, texture, and peroxidase activity. The immersion in 2.5% NaOH boiling solution (5 minutes) stood out as the best treatment since it inhibited the enzymatic browning and intensified the natural yellow color of the cubiu fruit and easily and fully peeled the whole fruit more rapidly without damaging its tissues. This treatment was chosen as the most advantageous because it can promote simultaneous peeling and bleaching. Therefore, it is recommended for cubiu industrial processing.

Arbustos de cubiu (Solanum sessiliflorum Dunal) têm atraído a atenção de pesquisadores por sua versatilidade biológica (heliófilos preferenciais ou ombrófilos facultativos), sua capacidade de crescimento em terra firme ou várzea e seus frutos de boa qualidade tecnológica para a indústria alimentícia. O objetivo deste estudo foi verificar mudanças físico-químicas em frutos de cubiu durante a maturação. Os frutos foram colhidos na estação experimental de olericultura do Instituto Nacional de Pesquisas da Amazônia (INPA), Brasil. As análises foram realizadas em frutos integrais de cubiu (casca, polpa e placenta) em quatro estádios de maturação tradicionais (verde, de vez, maduro, muito maduro), compondo-se de peso, umidade, sólidos totais, carotenoides totais, proteínas, lipídios e cinza. Os frutos de cubiu apresentaram grande variação de peso, com distribuição amodal. O estádio maduro foi crítico na manutenção da umidade e, a partir desse estádio, a perda de água tornou-se evidente. Os lipídios aumentaram progressivamente ao longo dos quatro estádios, mantendo, todavia, níveis insignificantes em termos calóricos. Os carotenoides totais, as proteínas e as cinzas alcançaram níveis máximos no estádio muito maduro. Com exceção do peso e da umidade, todas as mudanças físico-químicas exibiram o mesmo comportamento geral, i.e., aumentaram conforme os frutos amadureceram nos quatro estádios investigados.

Cubiu shrubs (Solanum sessiliflorum Dunal) have drawn the attention of researchers for their biological versatility (preferential heliophilous or facultative ombrophilous shrubs), their capacity to grow in upland or lowland areas, and the good technological quality of their fruits for the food industry. The aim of this study was to verify physicochemical changes in cubiu fruits during maturation. The fruits were harvested from the experimental station of olericulture of the Instituto Nacional de Pesquisas da Amazônia (INPA), Brazil. The analyses were performed in whole cubiu fruits (peel, pulp, and placenta) at four traditional ripening stages (green, turning, ripe, and fully ripe) for the determination of weight, moisture, total solids, total carotenoids, proteins, lipids, and ash. Cubiu fruits showed large weight variation, with amodal distribution. The ripe stage was critical to maintain moisture, and from that stage on, water loss became evident. The lipids increased steadily over the four ripening stages, maintaining, however, insignificant calorie content. Total carotenoids, proteins, and ash reached the maximum level at the fully ripe stage. With the exception of weight and moisture, all physicochemical changes exhibited the same general behavior, i.e. they increased as the fruits ripened at the four investigated stages.

Duas estirpes de rizóbia isoladas de solos de várzea da Amazônia Central produziram grandes quantidades de proteases alcalinas extracelulares, usando fontes baratas de carbono e nitrogênio. Os extratos brutos de proteases foram ativos em pH 9,0-11,0. As temperaturas ótimas foram de 35 ºC para a enzima do Rhizobium R-986 e de 55 ºC para a do Bradyrhizobium R-993. As atividades proteolíticas aumentaram na presença de 5 mM dos íons Na+, Ca2+ , Mg2+ e Mn2+ . As proteases secretadas pelos rizóbios foram fortemente inibidas por PMSF, um inibidor de serina protease. As enzimas foram ativas na presença de surfactantes (SDS e Triton X-100), e estáveis na presença de agentes oxidantes (H2O2) e redutores (β-mercaptoetanol) e solventes orgânicos (acetona, hexano, metanol, 1-propanol e tolueno).

Two rhizobia strains isolated from soils of the Central Amazonian floodplain produced appreciable quantities of crude alkaline protease extracts with inexpensive carbon and nitrogen sources. These protease crude extracts were optimally active at pH 9.0-11.0. The optimum temperatures were 35 ºC for Rhizobium sp. strain R-986 and 55 ºC for Bradyrhizobium sp. strain R-993. Protease activities in the crude extracts were enhanced in the presence of 5 mM metal ions, such as Na+, Ca2+, Mg2+ and Mn2+. Rhizobia proteases were strongly inhibited by PMSF, a serine-protease inhibitor. The enzymes were active in the presence of surfactants (SDS and Triton X-100) and stable in oxidizing (H2O2) and reducing agents (β-mercaptoethanol), and organic solvents (acetone, hexane, methanol, 1-propanol and toluene).

A produção e parcial caracterização de extratos brutos de amilase de duas estirpes de rizóbio (R-926 e R-991) foram avaliadas. Para ambas as estirpes, as máximas atividades amilolíticas foram obtidas no início/meio da fase exponencial de crescimento. As amilases rizobiais foram ativas numa variação de pH de 4,5 a 8,5 e temperatura de 30 a 50 ºC. Nenhum dos íons testados (K+, Na+, Ca2+, Hg2+, Mg2+, Mn2+, Cu2+ e Zn2+) foi exigido para a atividade catalítica da estirpe R-926. A amilase produzida pelo R991 foi estimulada na presença de K+, Hg2+ e Zn2+. Os surfactantes SDS, Triton X-100 e Tween-80 não exerceram um pronunciado efeito inibitório sobre as atividades enzimáticas, e SDS e Tween-80 causaram os maiores efeitos estimulatórios. A atividade amilolítica rizobial foi reduzida em até 30% na presença de EDTA; a amilase produzida pela estirpe R-926 também foi inibida pelo HgCl2, sugerindo, ao menos em parte, a importância de Ca2+ e resíduos de cisteína na atividade amilolítica dessa estirpe.

Amylase production and partial characterization of crude enzyme preparations from two rhizobia strains (R-926 and R-991) were evaluated. For both the strains, maximal amylase activities were achieved during the early-to-mid- exponential growth phase; both were active over a pH range from 4.5 to 8.5 and temperature from 30 to 50 ºC. None of the ions studied (K+, Na+, Ca2+, Hg2+, Mg2+, Mn2+, Cu2+ and Zn2+) was required for the catalytic activity of strain R-926; amylase activity of strain R-991 was stimulated in the presence of K+, Hg2+ and Zn2+. The surfactants SDS, Triton X-100 and Tween-80 did not have a pronounced inhibitory effect on enzyme activities; SDS and Tween-80 caused the highest stimulatory effects. Amylase activities from the rhizobia strains were reduced by up to 30% in the presence of EDTA; amylase activity of R-926 was also inhibited by HgCl2, suggesting that Ca2+and cysteine residues could be important for activity of this strain.

Os fungos têm sido bastante usados como produtores de diferentes substâncias de interesse econômico, tais como: enzimas, antibióticos, vitaminas, aminoácidos e esteróides. Este estudo teve como objetivo detectar a produção de enzimas por linhagens de Basidiomycetes, oriundas de áreas de floresta da Amazônia. Para a produção de enzimas, os fungos foram cultivados em meio líquido adicionado de substrato indutor (0,5%), pH ajustado para cada enzima e incubados a 28 °C, sob agitação a 140 rpm, durante 96 ou 120 horas. A massa micelial foi separada for filtração e os filtrados foram inoculados em cup plates de 6 mm de diâmetro, perfurados na superfície de meios de cultura sólidos, adequados para a detecção das enzimas amilases, proteases, celulases, fenoloxidases e pectinases em placa de Petri. As placas foram incubadas à temperatura de 28 °C por 24 horas, e reveladas para observação dos halos indicativos da atividade enzimática. Foi verificada também a atividade da amilase e protease produzida pelos fungos, crescidos em meio líquido, com diferentes fontes nutricionais. Foi possível detectar a produção de celulases e proteases por todos os isolados, 40% produziram amilases, 50% produziram fenoloxidases e 10% produziram pectinases. Quanto à atividade da amilase, o substrato farelo de trigo foi o que proporcionou os maiores halos de degradação, destacando-se os fungos Daedalea sp. 4E6 e Daedalea sp. 1A, Stereaceae 22B e Pycnoporus sanguineus 12B. Considerando os substratos testados para produção de proteases, o substrato concentrado protéico de peixe se destacou como a melhor fonte protéica. Os fungos P. sanguineus 12B, Stereaceae 22B e Cantharellus guyanensis 4Bl foram os melhores produtores de protease.

Mushrooms, edible basidiomycetes, have been extensively used as producers of different substances of economical interest, such as enzymes, antibiotics, vitamins, amino acids, and steroids. The objective of this study is to detect the production of enzymes such as amylases, proteases, cellulases, phenoloxidases and pectinases for lineages of basidiomycetes originating from the Amazonian forest. For the production of enzymes, mushrooms were cultivated in a liquid medium with substratum (0,5%), pH adjusted for each enzyme, and incubated at 28 °C, under agitation and 140 rpm for 96 or 120 hours. After this period, the samples were filtered for the separation of the mycelial mass. The filtrates were inoculated in solid medium surface perforated cup-plates of 6 mm diameter, appropriate for the detection of enzymes in Petri dishes. The plates were incubated at 28 °C for 24 hours, for observation to reveal enzymatic halos. It was verified the activity of the amylase and protease produced by the mushrooms grown in the liquid medium, with different nutritive sources. It was possible to detect the cellulase and protease production for all the ones that were isolated, 40% produced amylases, 50% produced phenoloxidases, and 10% produced pectinases. Regarding the activity of the amylase, the wheat bran substrate presented the largest degradation halos revealing Daedalea sp.4E6 and Daedalea sp. 1A, Stereaceae 22B and Pycnoporus sanguineus 12B mushrooms. Considering the substrata tested for protease production, it was observed that the fish protein concentrate was the best. The P. sanguineus 12B, Stereaceae 22B and Cantharellus guyanensis 4Bl mushrooms were the best protease producers.

As amilases estão entre as mais importantes enzimas industriais e são de grande interesse na biotecnologia atual. Embora elas possam ser derivadas de diversas fontes, as de origem microbiana são geralmente as mais procuradas pelas indústrias. As espécies do gênero Bacillus são consideradas as principais fontes de amilases. Apesar disso, a busca por novas fontes microbianas vem crescendo em todo o mundo. O presente estudo objetivou avaliar a produção de amilase por rizóbios nativos, usando farinha de pupunha como substrato. Neste estudo, foi adotado o delineamento experimental inteiramente casualizado, com três repetições. Foram determinados ainda os coeficientes de correlação de Pearson entre as variáveis pH do meio, proteína extracelular, biomassa celular, diâmetro médio da colônia (DMC), diâmetro médio do halo (DMH), índice enzimático (IE) e atividade amilolítica das bactérias selecionadas. Dos 19 isolados com atividade amilolítica em meio YMA modificado, sete (36,8%) exibiram "IE" > 2,1, o que permite considerá-los bons produtores de amilase. Os "IE" apresentados pelos isolados INPA R-987, 950 e 915B foram significativamente inferiores (p < 0,01) aos mostrados pelas bactérias INPA R-926, 975 e 957. A atividade amilolítica variou significativamente (p < 0,01) entre as bactérias investigadas. Os isolados INPA R-975 e R-926 apresentaram, respectivamente, a maior (1,00 U.min-1.mL-1) e a menor (0,31 U.min-1.mL-1) média de atividade. Em termos gerais, a proteína extracelular correlacionou-se positivamente com "IE" (r = 0,52*; p < 0,05) e "DMH" (r = 0,55*; p < 0,05). A biomassa celular apresentou correlações positivas com atividade amilolítica (r = 0,55*; p < 0,05) e "DMH" (r = 0,54*; p < 0,05) e negativa com o pH final do meio de cultivo (r = 0,93**; p < 0,01).

Amylases are among the most important industrial enzymes and are of great significance in present-day biotechnology. Although they can be derived from various sources, enzymes from microbial sources are generally the most required by industry. Species of the genus Bacillus are considered to be the main sources of amylases, although screening for new microbial sources is increasing all over the world. The objective of the present study was to evaluate the production of amylase by indigenous rhizobia, using peach palm flour as substrate. In this study, a completely randomized experimental design was adopted with three replicates. Pearson's correlation coefficients were calculated for medium pH, extracellular protein, cellular biomass, mean colony diameter (MCD), mean halo diameter (MHD), enzymatic index (EI) and amylolytic activity variables from the bacteria selected. Out of the 19 rhizobia strains with amylolytic activity on modified YMA, seven (36.8%) strains showed "EI" > 2,1, and they were considered as good producers of amylase. The "IE" presented by bacteria INPA R-987, R-950 and R-915B were significantly lower (p < 0.01) than those shown by the INPA R-926, R-975 and R-957. The amylolytic activity varied significantly (p < 0.01) among the rhizobia strains. INPA R-975 and R-926 strains presented, respectively, the highest (1.00 U.min-1.mL-1) and the lowest (0.31 U.min¹.mL-1) activity average. In general terms, the extracellular protein was correlated positively with "EI" (r = 0.52*; p < 0.05) and "MHD" (r = 0.55*; p < 0.05). The cellular biomass showed positive correlations with amylolytic activity (r = 0.55*; p < 0.05) and "MHD" (r = 0.54*; p < 0.05), and a negative with final pH medium (r = 0.93**; p < 0.01).

A associação rizóbia x leguminosa contribui para enriquecer o solo com nitrogênio por meio da fixação biológica. Entretanto, pouco se conhece a respeito do perfil enzimático desses microrganismos. Nesse contexto, a presente investigação propõe avaliar a produção de enzimas hidrolíticas extracelulares por isolados de rizóbia nativos da Amazônia Central. Essa triagem constitui o primeiro passo na seleção de microrganismos nativos que são potencialmente exploráveis como produtores de enzimas. Foram testados 67 isolados nativos de rizóbia para as atividades amilolítica, celulolítica, lactolítica, lipolítica, pectinolítica e proteolítica, em meio YMA modificado. A atividade ureolítica foi detectada em meio ágar-uréia. As bactérias isoladas dos nódulos de feijão caupi mostraram maior capacidade em produzir enzimas do que os isolados bacterianos de soja. De todas as enzimas hidrolíticas avaliadas, apenas a pectinase não foi detectada neste estudo. Amilase (32,8%), protease (28,4%), urease (20,9%) e carboximetilcelulase (9,0%) foram as enzimas mais freqüentes produzidas pelos isolados. Neste trabalho, apenas as enzimas amilase e protease variaram significativamente entre os isolados de rizóbia. Os isolados INPA R-926 e INPA R-915 exibiram os maiores índices amilolíticos (IE = 3,1) e proteolíticos (IE = 6,6), respectivamente. Este estudo revelou alguns isolados de rizóbia nativos da Amazônia Central como fontes promissoras de enzimas de importância industrial para uso biotecnológico.

Legumes enrich the soil by contributing nitrogen through symbiotic biological nitrogen fixation by rhizobia bacteria. However, very little is known about the extracellular enzymatic profile of these microorganisms. In this context, the production of extracellular hydrolytic enzymes by indigenous strains of rhizobia in Central Amazonia was evaluated. This screening constitutes the first step in selecting indigenous microorganisms that are potentially exploitable as enzyme producers. Indigenous strains of rhizobia were screened for extracellular amylolytic, carboxymethylcellulolytic, lactolytic, lipolytic, pectinolytic and proteolytic activities on modified YMA. Ureolytic activity was detected on a urea-agar slant. Rhizobia strains isolated from cowpea nodules produced more enzymes than those isolated from soybean nodules. Out of all the extracellular hydrolytic enzymes evaluated, only pectinase was not detected in this study. The most frequent rhizobia enzymes were amylase (32.8%), protease (28.4%), urease (20.9%) and carboxymethylcellulase (9.0%). In this study, only amylase and protease enzymes varied significantly among rhizobia strains. INPA strains R-926 and R-915 showed the highest enzymatic levels for amylase (EI = 3.1) and protease (EI = 6.6), respectively. This paper showed some indigenous strains of rhizobia from Central Amazonia as promising sources of industrially relevant enzymes for biotechnological purposes.

A importância das bactérias conhecidas como rizóbia no estabelecimento de leguminosas tem sido amplamente reconhecida. Porém, poucas são as informações referentes ao perfil enzimático dessas bactérias benéficas. O estudo objetivou investigar a influência do pH do meio sólido sobre a atividade enzimática de rizóbios nativos da Amazônia Central. Essa triagem constitui o primeiro passo no processo de seleção de microorganismos benéficos, como produtores de enzimas de aplicação biotecnológica. Nesse estudo, 64 isolados de rizóbia foram testados para a produção extracelular de amilase, lipase, pectinase e protease, em meio YMA modificado. Excetuando a atividade pectinolítica, todas as outras enzimas (amilase, lipase e protease) foram detectadas nos isolados investigados. Dois isolados (INPA R-975 e INPA R-926) exibiram atividades amilolíticas, lipolíticas e proteolíticas. Os índices enzimáticos amilolíticos e proteolíticos variaram significativamente entre os isolados e as condições de pH do meio de cultura. De maneira geral, as maiores atividades amilolíticas e proteolíticas foram exibidas pelos isolados INPA R-957, INPA R-915B e INPA R-991 em pH 6,5. O isolado INPA R-957 também se mostrou amilolítico e proteolítico nos pHs 5,0 e 8,0. Esse estudo mostrou que alguns rizóbios nativos da Amazônia representam fontes promissoras de amilase e protease de uso biotecnológico, sobretudo na tecnologia de alimentos.

The importance of rhizobia bacteria in the establishment of legume plants has been widely recognized. However, information is scarce regarding the enzymatic profiles of these beneficial bacteria. The objective of this study was to investigate the influence of pH of solid medium on enzymatic activity of native Central Amazonian rhizobia strains. This screening constitutes the first step in selecting beneficial microorganisms as enzyme producers. In this study, 64 strains of rhizobia were screened for extracellular amylolytic, lipolytic, pectinolytic and proteolytic activities on modified YMA. With the exception of pectinolytic activity, the other enzymatic activity groups (amylase, lipase and protease) were found in the strains investigated. Two isolates (INPA R-975 and INPA R-926) exhibited amylolytic, lipolytic and proteolytic activities. The activity levels of the amylases and proteases varied significantly among strains of rhizobia and among pH conditions. In general, the highest amylase and protease activities were presented by isolates INPA R-957, INPA R-915B and INPA R-991 at pH 6.5. The INPA R-957 isolate also showed amylolytic and proteolytic activities at pH 5.0 and 8.0. This study revealed several Central Amazonian rhizobia strains as promising sources of amylase and protease for biotechnological applications, especially in food technology.

O elevado teor de ácido ascórbico no camu-camu (Myrciaria dubia McVaugh, Myrtaceae) desperta o interesse de extrativistas, agricultores e consumidores, e leva à necessidade de desenvolvimento de tecnologias adequadas para produção em terra firme e aproveitamento industrial do fruto. Este trabalho teve por objetivo verificar a adequação do camu-camu para a produção de bebida alcoólica fermentada, assim como o efeito do branqueamento do fruto e da incorporação da casca à polpa nas características nutricionais e sensoriais da bebida. Os frutos foram separados em quatro lotes, sendo dois branqueados (90 ºC por 7 min). Após a despolpa, as cascas de um lote de cada tratamento (com e sem branqueamento) foram incorporadas às respectivas polpas e avaliadas quanto à composição química (umidade, pH, acidez, sólidos solúveis, açúcares, ácido ascórbico, compostos fenólicos, antocianinas e flavonóides). Após a correção do mosto com açúcar, pasteurização, fermentação (25 dias), trasfega, pasteurização (70 ºC por 15 min), filtragem e clarificação, as bebidas foram avaliadas quanto a composição química, edulcoradas e submetidas à análise sensorial. O branqueamento reduziu a concentração de ácido ascórbico das polpas (33 %) e a agregação da casca aumentou os teores de matéria seca (39 % polpa), ácido ascórbico (33 % na polpa, 23 % no mosto e 50 % na bebida) e fenólicos (50 % bebida). O perfil sensorial e a aceitabilidade sugerem que o camu-camu é adequado para a produção de bebida alcoólica fermentada e que a agregação da casca à polpa contribuiu positivamente para a aceitabilidade (6,7 com casca e 6,2 sem casca, na escala de 9 pontos). As bebidas apresentaram flavor característico do fruto, limpidez, coloração vermelho-laranjada e sabor agradável.

The high levels of ascorbic acid in camu-camu (Myrciaria dubia McVaugh, Myrtaceae) have stimulated interest of extractivists, farmers and consumers. This has led to a need to develop adequate technology for it's production on non-flooded land and the industrial use of this fruit. This study had as its main objective to verify if camu-camu is adequate for the production of fermented alcoholic beverages, measuring the effect of blanching the fruit and the incorporation of the fruit peel with the fruit pulp on the nutritional and sensory characteristics of the drink. The fruits were separated into 4 groups, two being blanched (90 ºC for 7 minutes). After the pulp was removed, the peels of one group from each blanching treatment were incorporated into the respective pulps and their chemical composition evaluated. After sugar correction of the must, pasteurisation, fermentation (25 days), decanting, pasteurisation (70 ºC for 15 minutes), filtering and clarification, the beverages were evaluated as to their chemical composition, sweetened and submitted to sensory analysis. Blanching reduced the concentration of ascorbic acid in the pulps (33 %) and the addition of the peel increased the amount of dry matter (39 % in pulp), ascorbic acid (33 % in pulp, 23 % in must and 50 % in drink) and phenolic compounds (50 % in drink). The sensory profile and acceptability suggest that camu-camu is adequate for the production of fermented alcoholic beverages and that the addition of the peel to the pulp contributes positively to it's acceptability (6.7 with versus 6.2 without, of 9 points possible). The beverage had flavour characteristic of the fruit, a orangish-red color and agreeable taste.

O cubiu (Solanum sessiliflorum Dunal) é uma hortaliça da Amazônia, domesticada pelos índios, que pode produzir até 100 t/ha de frutos ricos em sais minerais e vitaminas. Os frutos são utilizados pelas populações tradicionais da região nas formas de sucos, sorvetes, doces e molhos para carnes bovina, de frango e de peixes. Treze caracteres morfológicos e químicos dos frutos de 24 etnovariedades da Amazônia brasileira, peruana e colombiana foram utilizadas para estimar os coeficientes de correlações fenotípica (rf), genética (rg) e de ambiente (ra) entre pares de caracteres. Na maioria dos casos, as correlações genéticas apresentaram valores superiores aos das fenotípicas e de ambientes, indicando que o ambiente teve pouca influência. Entre os caracteres morfológicos, as dimensões dos frutos são estreitamente relacionadas com o teor de umidade. Não foram observadas correlações entre caracteres morfológicos e químicos que pudessem ser úteis ao melhoramento do cubiu. Entre os caracteres químicos, as correlações entre brix e acidez e açúcares redutores e açúcares totais foram altas e positivas (rg = 0,62 e rg = 0,83), respectivamente. Entre vários caracteres químicos será difícil praticar seleção simultânea sem perda de genótipos.

The cubiu (Solanum sessiliflorum Dunal) is a fruiting shrub domesticated by indians in Amazonia, and produces 40 to 100 t/ha of fruit rich in minerals and vitamins. The fruits are traditionally used for juices, ice-creams, sweets and in sauces for meat, chicken and fish. Thirteen chemical and morphological characters of 24 etnovarieties of cubiu from Brazilian, Peruvian and Colombian Amazonia were used to estimate the coefficients of phenotypic (r), genetic (rg) and environmental (rc) correlation between pairs of characters. In most cases, the genetic correlations were higher than the phenotypic and environmental correlations, indicating that the environment had less influence. Among the morphological characters, fruit dimensions were closely correlated with moisture. There were no correlations between morphological and chemical characters that could be useful in cubiu improvement. Among the chemical characters, the correlations between brix and acidity and reducing sugars and total sugars were high and positive (r = 0,62 and rg = 0,83, respectively). Thus, it will be difficult to simultaneously select for these without losing genotypes.
