Foi realizado estudo comparativo na produção de diferentes antígenos virais usando sistema de microcarregador e sistema tradicional. Células Vero, BHK e MA-104 foram cultivadas em microcarregadores (2mg/ml) utilizando-se biorreatores com capacidade de 3,7 litros e, em paralelo, no sistema convencional com garrafas Roux. Após quatro dias de cultura para as células BHK e sete dias para as células Vero e MA-104, as células foram infectadas com 0,1 MOI (multiplicidade de infecção) de vírus da raiva, vírus do sarampo, poliovírus e rotavírus. Foi determinado o rendimento das células e dos vírus em microcarregadores e sistema convencional. Foi observado no sistema de microcarregador um aumento médio obtido de vinte vezes mais células/ml em relação à cultura convencional em monocamadas, usando garrafas Roux. Por outro lado, as células que cresceram em garrafas Roux apresentaram 1,6 a 6,7 mais vírus/ml em culturas do que no sistema de microcarregador. Contudo o total das amostras em termos de vírus por grupo foi estatisticamente similar para ambos os sistemas (p > 0,05). O rendimento na produção de antígeno viral pode depender não somente da concentração das células, mas também de outros fatores da cultura, como características do suporte no crescimento. Assim, o estudo deste parâmetro pode proporcionar uma linha de base para um futuro melhoramento e estratégias para se estabelecer um aumento em escala na produção de vírus, já que, dependendo do tipo de vírus, a ótima condição encontrada para uma produção de vírus em pequena escala pode não ser adequada para a produção em grande escala, requerendo novas padronização e avaliação.

A comparative study was performed in the production of different viral antigens by using microcarrier systems and traditional systems. Vero, BHK and MA 104 cells were cultivated in microcarriers (2mg/ml) using a bioreactor with a working capacity of 3.7 liters, in parallel with conventional Roux bottles. After four days (BHK cells), and seven days of culture (Vero and MA-104 cells), the cells were infected with 0.1 MOI (multiplicity of infection) of rabies virus, measles virus, poliovirus and rotavirus. The yields of the cells and virus in microcarriers and in the conventional system were determined. It was observed that in the microcarrier system, an average increase of twenty-fold more cells/ml was obtained in relation to the conventional monolayer culture, using Roux bottle. On the other hand, cells grown in Roux bottles presented 1.3 to 6.7 more viruses/ml culture than those in the microcarrier systems. However, the overall data showed that yieldings, in terms of viruses per batch, were statistically similar for both systems (p > 0.05). The amount of viral antigen production seems to depend not only on cell concentration, but also on other culture factors such as the characteristic of the cell-growth surface. Thus, the present findings provide a baseline for further improvements and strategies to be established for a scaling-up virus production since depending on the type of virus the optimal conditions found for a small-scale virus production seem unsuitable for large-scale production, requiring new standardization and evaluation.

OBJETIVOS: investigar a prevalência Streptococus pyogenes em secreção de orofaringe de escolares procedentes de duas escolas públicas da cidade Recife. MÉTODOS: estudo epidemiológico e clínico-microbiológico descritivo no qual foram examinados 753 escolares. A cultura bacteriana da secreção orofaringeana foi realizada em ágar-sangue de carneiro 5% e as cepas SBGA identificadas através dos testes de bacitracina, Pyr e aglutinação em látex. RESULTADOS: a faixa etária dos 753 escolares analisados variou de cinco a 19 anos, sendo 54,3% do sexo masculino e 45,7% do sexo feminino. Seis eram portadores assintomáticos de SBGA e foram submetidos ao tratamento com penicilina. Após o tratamento, realizou-se a dosagem da antiestreptolisina o (ASLO), cujos títulos séricos foram inferiores a 200UT. CONCLUSÕES: uma prevalência de SBGA de 0,8% foi estimada em portadores assintomáticos, considerada baixa, quando comparado a outros resultados em estudos semelhantes. Os autores sugerem a realização de outros estudos para estimar a prevalência de SBGA em crianças com faringite e sua relação com a febre reumática aguda.

OBJECTIVES: assess the prevalence of Streptococcus pyogenes in throat secretions of students from two public elementary schools in Recife. METHODS: an epidemiological and clinical-microbiological study was performed. From two schools, 753 individuals were examined. Throat secretion cultures were performed in blood sheep ágar 5% and SBGA strains identified by bacitracin, Pyr and latex agglutination tests. RESULTS: the school children were aged from five to 19 years old, 54,3% of the sample were male and 45,7% female. Six asymptomatic SBGA carriers were identified. Following treatment with penicillin, ASLO sera titration was performed. All carriers had antibody titers below 200UT. CONCLUSIONS: SBGA prevalence rate of 0,8% was estimated in asymptomatic carriers, this percentage was low when compared with other results from similar studies. The authors suggest that epidemiological studies be accomplished to estimate SBGA prevalence in children with pharyngitis and correlation with acute rheumatic fever.

De 40 (35,7%) deles a bactéria em questão foi isolada de, pelo menos, um dos nichos estudados. Entre estes portadores, 27 (67,5%) foram positivos em apenas um dos quatro nichos, 8 (20,0%) em dois e 5 (12,5%) em três. Das 113 cepas de S. aureus identificadas, 28 (24,8%) isoladas de 9 (22,5%) dos portadores revelaram-se tox +. Entre essas cepas, 7 (25,0%) produziram enterotoxina do tipo A, 6 (21,4%) do tipo B, 11 (39,3%) do tipo C e 4 (14,3%) dos tipos A e C simultaneamente. A fagotipagem revelou a predominância de cepas sensíveis aos fagos do Grupo I/III/NC (16,8%) . Os resultados obtidos não demonstraram a ocorrência simultânea de cepas de Staphylococcus aureus em amostras colhidas de boca, mãos e fezes do grupo estudado.

Paulo (Brazil). A total of 40 (35.7%) individuals were detected as carriers of this microorganism. Among these carriers, 27 (67.5%) were positive in only one of the four niches studied, 8 (20.0%) in two and 5 (12.5%) in three niches. They were identified 113 Staphylococcus aureus strains, and 28 (24.8%) of these strains, isolated from 9 (22.5%) carriers, produced enterotoxin. Of these strains, 7 (25.0%) produced type A enterotoxin, 6 (21.4%) of type B, 11 (39.3%) type C and 4 (14.3%) produced both type A and C. The phage typing of the 113 Staphylococcus aureus strains revealed the predominance of the strains lysed by phages belonging to the Group I/III/NC (16.8%). The results obtained did not demonstrate the simultaneous incidence of Staphylococcus aureus strains in samples collected from mouth, hands and stools of the group studied.
