Objective: To assess the adherence to a set of evidence-based recommendations to support mental health during the coronavirus disease 2019 (COVID-19) pandemic and its association with depressive and anxiety symptoms. Methods: A team of health workers and researchers prepared the recommendations, formatted into three volumes (1: COVID-19 prevention; 2: Healthy habits; 3: Biological clock and sleep). Participants were randomized to receive only Volume 1 (control), Volumes 1 and 2, Volumes 1 and 3, or all volumes. We used a convenience sample of Portuguese-speaking participants over age 18 years. An online survey consisting of sociodemographic and behavioral questionnaires and mental health instruments (Patient Health Questionnaire-9 [PHQ-9] and Generalized Anxiety Disorder-7 [GAD-7]) was administered. At 14 and 28 days later, participants were invited to complete follow-up surveys, which also included questions regarding adherence to the recommendations. A total of 409 participants completed the study – mostly young adult women holding university degrees. Results: The set of recommendations contained in Volumes 2 and 3 was effective in protecting mental health, as suggested by significant associations of adherence with PHQ-9 and GAD-7 scores (reflecting anxiety and depression symptoms, respectively). Conclusion: The recommendations developed in this study could be useful to prevent negative mental health effects in the context of the pandemic and beyond.

Abstract The extensive marine biodiversity has proved to be a promising source of substances with biomedical potential. In this study, the cytotoxicity of the Brazilian octocoral Phyllogorgia dilatata (Gorgoniidae) was evaluated against two tumor cell lines and three bacterial strains. The methanol/dichloromethane crude extract presented no antibacterial activity up to the highest concentration tested (512 µg/mL), however it revealed a noteworthy antiproliferative effect against HCT-116 (80%) and MCF-7 (54%) cell lines at 50 μg/mL. Therefore, guided by the cytotoxic activity, a multistep chemical fractionation of the extract provided the subfraction 5 (PDPH2-5) with IC50 values of 3.18 and 17.80 μg/mL against HCT-116 and MCF-7, respectively. The LC-HRMS/MS analysis of PDPH2-5 showed ions of m/z 219.1742 and 219.1743, characterized as (E,E) and (Z,E) germacrone, after a LC-DAD-SPE/NMR analysis of the hexanic fraction and comparisons of NMR data with the literature. Previously reported assessments to the cytotoxic activity of the (E,E)-diastereoisomer disclosed higher IC50 values than that obtained for the PDPH2-5 fraction, suggesting, herein, a potentiated effect of the diastereoisomeric mixture. Such remark encourage further bioactivity studies with stereoisomer mixtures and reduce the urge for compound isolation.

In recent years, great advances have been observed in the proteomic area regarding the different approaches adopted as top-down, middle-down and bottom-up. Regarding middle-down and bottom-up approaches, a previous step for protein digestion is necessary for further analysis of the generated peptides. Classical digestion protocols employing proteases are usually carried out in solution, which can lead to a number of disadvantages such as high digestion time, enzymatic autolysis and impossibility to reuse the enzymes. In view of these drawbacks, different approaches have been studied and applied for improving enzymatic efficiency, inhibition of autolysis, reduction of digestion time, and to minimize the number of missed cleavages. In the literature, great efforts have been made in order to achieve maximum efficiency in enzymatic digestion, with emphasis on proteases immobilization. Phosphoenolpyruvate carboxykinase (PEPCK) is a ubiquitous enzyme found in all known groups of organisms, acting in the reversible conversion of oxaloacetate (OAA) to phosphoenolpyruvate (PEP) in the presence of divalent metal ion, and dependent of adenosine 5’-triphosphate (ATP) or guanosine-5’-triphosphate (GTP). PEPCK is an important enzyme in the metabolism of some organisms, such as Trypanosoma cruzi, being suggested as a potential drug target to treat Chagas’ disease. Its catalytic activity is, classically, measured by coupled assays. Herein, a direct assay by liquid chromatography-tandem mass spectrometry (LC-MS/MS) capable of quantifying PEP, in co-elution with OAA by the differentiation obtained by the mass spectra, is reported. The developed assay was used throughout the purification protocol in order to measure the activity of PEPCK of T. cruzi, which was expressed in Escherichia coli. The purified enzyme was kinetically characterized by the developed method with Michaelis-Menten constant (KMapp) values of 96 ± 4 and 275 ± 18 µmol L-1 to OAA and ATP as substrates, respectively. The developed assay was also used for ligand screening and proved to be able to identify very low inhibitions for small molecules (50 µmol L-1).

Butia capitata (Mart.) Becc. lamina anatomy as a tool for taxonomic distinction from B. odorata (Barb. Rodr.) Noblick comb. nov (Arecaceae)

A distinção de Butia capitata e de B. odorataé baseada em poucos caracteres morfológicos, o que motiva a busca por mais dados que corroborem a separação. Como dados da anatomia foliar são relevantes na circunscrição de taxa em Arecaceae, objetivou-se descrever a anatomia foliar de B. capitatae comparar com a de B. odorata. Amostras da porção mediana das pinas foram coletadas e processadas de acordo com técnicas usuais em Anatomia Vegetal. A epiderme é unisseriada e possui cutícula espessa e ceras epicuticulares em formato de filamentos em anzol. As células subsidiárias arqueiam-se sobre as células-guarda localizadas no nível da hipoderme. O mesofilo é isobilateral e compacto. Os feixes vasculares são colaterais com extensão de bainha esclerenquimática alcançando a hipoderme. Stegmatas possuem corpos silicosos esféricos e drusóides. A nervura mediana é adaxialmente projetada, apresentando espessa camada fibrosa que circunda os feixes vasculares, adjacente ao parênquima clorofiliano. Tecido de expansão estratificado ocorre na face abaxial, nos limites do mesofilo com a nervura mediana. Ráfides ocorrem apenas em B. capitata. Os feixes diminutos da nervura mediana circundam totalmente o anel fibroso somente em B. odorata. Além de diagnósticos, esses caracteres são úteis para corroborar a separação atualmente proposta.
The distinction between Butia capitata and B. odorata is based only on a few morphological characteristics, therefore there is a need for additional studies for supporting the separation of the species. As lamina anatomy characteristics are relevant in circumscribing Arecaceae taxa, this work aimed to describe B. capitata lamina anatomy and compare it with B. odorata. Samples from the middle portion of the pinnae were collected and processed in accordance with standard plant anatomy techniques. The epidermis is uniseriate and composed of a thick cuticle and epicuticular waxes into like hooked filaments. The subsidiary cells that arch over the guard cells are located at the hypodermis. The mesophyll is isobilateral and compact. The vascular bundles are collateral with a sclerenchymatic sheath extension that reaches the hypodermis. The stegmata cells have spherical and druse-like silica bodies. The midrib faces the adaxial surface with a thick fibrous layer surrounding the vascular bundles adjacent to the chlorenchyma. The stratified expansion tissue is on the abaxial surface, within the boundary between the mesophyll and midrib. Raphides are only found in B. capitata. Small bundles of the midrib fully surround the fibrous cylinder only in B. odorata. These characteristics are diagnostic and useful for supporting the proposed separation.

Resumo Um levantamento atualizado das plantas com sementes e análises relevantes acerca desta biodiversidade são apresentados. Este trabalho se iniciou em 2010 com a publicação do Catálogo de Plantas e Fungos e, desde então vem sendo atualizado por mais de 430 especialistas trabalhando online. O Brasil abriga atualmente 32.086 espécies nativas de Angiospermas e 23 espécies nativas de Gimnospermas e estes novos dados mostram um aumento de 3% da riqueza em relação a 2010. A Amazônia é o Domínio Fitogeográfico com o maior número de espécies de Gimnospermas, enquanto que a Floresta Atlântica possui a maior riqueza de Angiospermas. Houve um crescimento considerável no número de espécies e nas taxas de endemismo para a maioria dos Domínios (Caatinga, Cerrado, Floresta Atlântica, Pampa e Pantanal), com exceção da Amazônia que apresentou uma diminuição de 2,5% de endemicidade. Entretanto, a maior parte das plantas com sementes que ocorrem no Brasil (57,4%) é endêmica deste território. A proporção de formas de vida varia de acordo com os diferentes Domínios: árvores são mais expressivas na Amazônia e Floresta Atlântica do que nos outros biomas, ervas são dominantes no Pampa e as lianas apresentam riqueza expressiva na Amazônia, Floresta Atlântica e Pantanal. Este trabalho não só quantifica a biodiversidade brasileira, mas também indica as lacunas de conhecimento e o desafio a ser enfrentado para a conservação desta flora.
Abstract An updated inventory of Brazilian seed plants is presented and offers important insights into the country's biodiversity. This work started in 2010, with the publication of the Plants and Fungi Catalogue, and has been updated since by more than 430 specialists working online. Brazil is home to 32,086 native Angiosperms and 23 native Gymnosperms, showing an increase of 3% in its species richness in relation to 2010. The Amazon Rainforest is the richest Brazilian biome for Gymnosperms, while the Atlantic Rainforest is the richest one for Angiosperms. There was a considerable increment in the number of species and endemism rates for biomes, except for the Amazon that showed a decrease of 2.5% of recorded endemics. However, well over half of Brazillian seed plant species (57.4%) is endemic to this territory. The proportion of life-forms varies among different biomes: trees are more expressive in the Amazon and Atlantic Rainforest biomes while herbs predominate in the Pampa, and lianas are more expressive in the Amazon, Atlantic Rainforest, and Pantanal. This compilation serves not only to quantify Brazilian biodiversity, but also to highlight areas where there information is lacking and to provide a framework for the challenge faced in conserving Brazil's unique and diverse flora.

Eficiência na produção de frutos e alocação relativa de nutrientes em cultivares de cafeeiro

Considerando a baixa produtividade das plantas em solos de menor fertilidade natural e o alto custo dos insumos agrícolas, torna-se necessária a seleção de cultivares mais eficientes na absorção e utilização dos nutrientes minerais. Foram avaliados quatro cultivares de cafeeiro arábica (Acaiá IAC 474 19, Icatu Amarelo IAC 3282, Rubi MG 1192 e Catuaí Vermelho IAC 99) quanto à eficiência na produção de frutos e alocação relativa de nutrientes. O experimento foi conduzido em Viçosa MG, em condições de campo, no delineamento experimental em blocos ao acaso, envolvendo quatro cultivares, quatro repetições e três níveis de adubação (baixo, normal e alto). As parcelas úteis constituíram-se de nove plantas espaçadas de 2 x 1 m. O cultivar Icatu Amarelo IAC 3282 foi o mais produtivo no ambiente com restrição de nutrientes, enquanto Rubi MG 1192 e Catuaí Vermelho IAC 99 mostraram-se mais produtivos em ambientes com alto suprimento de nutrientes. A eficiência de produção de café em coco por unidade de P, Ca, Mg e B acumulados na planta foi maior no nível alto de adubação. Os cultivares Rubi MG-1192 e Catuaí Vermelho IAC 99 apresentaram maior eficiência de utilização de nutrientes para produção de frutos no nível alto de adubação. Considerando a média de alocação relativa de nutrientes nos frutos para os quatro cultivares, no nível normal de adubação, verificou-se que eles possuem 38,1% do N, 46,34% do P, 40,19% do S, 42,68% do K, 13,19% do Ca, 25,04% do Mg, 40,63% do Cu, 19,49% do Zn e 17,73% do B.
Considering the low productivity of plants in soil of lower natural fertility and the high cost of fertilizers, the selection of cultivars more efficient in the absorption and use of mineral nutrients is a necessity. Four coffee varieties were evaluated for mineral nutrient use efficiency in the production of beans, as well as the relative allocation of these nutrients in reproductive organs. The experiment was carried-out in field conditions in an experimental area of the Universidade Federal de Viçosa, Viçosa, Minas Gerais State, Brazil. The treatments were distributed in randomized blocks design, in a factorial arrangement (4 x 3) x 4, consisting of four coffee varieties (Acaiá IAC-474-19, Icatu Amarelo IAC-3282, Rubi MG-1192 and Catuaí Vermelho IAC-99), three fertilization levels (lower, adequate and high), with four replications. The plot useful area consisted of nine plants with spacing of 2 x 1 m. The variety Icatu Amarelo IAC-3282 was the most productive in the condition of low nutrient input, while Rubi MG-1192 and Catuaí Vermelho IAC-99 were more productive with high nutrient input. The production of dry coffee per unit of P, Ca, Mg and B accumulated in the plant was larger at the high fertilization level. The varieties Rubi MG-1192 and Catuaí Vermelho IAC-99 showed greater efficiency of nutrient use for production of beans at the high level of fertilization. Considering the mean of relative allocation of nutrients in the beans, of the four varieties, at the usual level of fertilization, it was found that they have 3 8.1 % of N, 46.3% of P, 40.2% of S, 42.7% of K, 13.2% of the Ca, 25.0% of Mg, 40.6% of the Cu, 19.5% of Zn and 17.7% of B.

Biologia e não-preferência para oviposição por Bemisia tabaci (Gennadius) biótipo B (Hemiptera: Aleyrodidae) em cultivares de algodoeiro

Os objetivos deste trabalho foram avaliar alguns aspectos biológicos das fases de ovo e de ninfa de Bemisia tabaci (Gennadius) biótipo B e avaliar a não-preferência para oviposição em testes com e sem chance de escolha e sua correlação com o número e tipo de tricomas nas cultivares de algodoeiro (BRS Ipê, BRS 186-Precoce 3, BRS Acala, BRS Verde, BRS 200-Marrom, BRS Cedro, BRS Ita 90-2 e BRS Aroeira). Os experimentos foram realizados em câmaras climatizadas (28 ± 2ºC, 70 ± 10% UR e fotofase de 14h), e em casa-de-vegetação. A viabilidade dos ovos não foi afetada pelas cultivares de algodoeiro, porém, a sobrevivência no período de ovo a adulto foi influenciada pelo hospedeiro. Não houve efeito das cultivares na duração da fase de ovo, do segundo, terceiro e quarto ínstares nem no período de ovo a adulto. As ninfas que se desenvolveram na cultivar BRS Ipê tiveram o primeiro ínstar alongado, diferentemente daquelas desenvolvidas nas demais cultivares. As cultivares BRS Aroeira, BRS Verde e BRS Ita 90-2 apresentaram baixo número de ovos nos testes com e sem chance de escolha, indicando um possível mecanismo de resistência, mas não foi possível estabelecer uma correlação entre a densidade de tricomas e a não-preferência para oviposição.
The purposes of this work were to evaluate some biological aspects of Bemisia tabaci (Gennadius) biotype B at egg and nymphal stages and to evaluate the non-preference for oviposition and its correlation with the number and type of trichomes on the cotton cultivars BRS Ipê, BRS 186-Precoce 3, BRS Acala, BRS Verde, BRS-200 Marrom, BRS Cedro, BRS Ita 90-2 and BRS Aroeira. The experiments were conducted in climatic chambers at 28 ± 2°C, 70% RH and photophase of 14h, and in greenhouse. Egg fertility was not affected by the cotton cultivars but survival in egg-adult period was influenced by the host plant. There was no influence of cultivars neither on the duration of egg stage, nymphs at 2nd, 3rd and 4th instars nor on the duration from egg to adult, but nymphs reared on the cultivar BRS Ipê had their 1st instar extended. Low number of eggs was detected on the cultivars BRS Aroeira, BRS Verde and BRS Ita 90-2 in both experiments with and without oviposition choice, indicating a possible mechanism of resistance, but no correlation could be established between trichome densisty and oviposition non-preference.
