Resumo Os microRNAs (miRNAs) são reconhecidos como biomarcadores do diabetes mellitus tipo 2 (DM2), úteis para a compreensão do metabolismo da doença, e possuem grande potencial como alvos terapêuticos. O aumento da expressão de BDNF e IGF1 está altamente envolvido nos benefícios as vias de insulina e glicose, porém, são regulados negativamente em condições de resistência à insulina, enquanto seu aumento de expressão está correlacionado com a melhora do metabolismo da glicose e da insulina. Estudos sugerem a regulação desses genes por microRNA em vários contextos diferentes, proporcionando uma nova abordagem de investigação para compreender o metabolismo do DM2 e revelar potenciais alvos terapêuticos. No presente estudo, investigamos em diferentes modelos animais (humanos, ratos e camundongos) miRNAs que têm como alvo BDNF e IGF1 em tecido muscular esquelético com condições fisiológicas de DM2. As análises foram realizadas utilizando ferramentas de bioinformática e bancos de dados para predição de miRNA, homologia molecular, validação experimental de interações, expressão na condição fisiológica estudada e interação em rede. Os resultados mostraram três candidatos a miRNAs para IGF1 (miR-29a, miR-29b e miR-29c) e um para BDNF (miR-206). As avaliações experimentais e a busca pela expressão no músculo esquelético de indivíduos com DM2 confirmaram a interação prevista entre miRNA-mRNA para miR-29b e miR-206 através de modelos humanos, ratos e camundongos. Essa interação foi reafirmada em múltiplas análises de rede. Em conclusão, nossos resultados mostram a relação de regulação entre miR-29b e miR-206 com os genes investigados, em diversos tecidos, sugerindo um padrão de inibição. Contudo, esses dados mostram um grande número de possíveis processos fisiológicos de interação para perspectivas biotecnológicas.

Abstract microRNAs (miRNAs) are recognized as diabetes mellitus type 2 (T2DM) biomarkers useful for disease metabolism comprehension and have great potential as therapeutics targets. BDNF and IGF1 increased expression are highly involved in the benefits of insulin and glucose paths, however, they are down-regulated in insulin resistance conditions, while their expression increase is correlated to the improvement of glucose and insulin metabolism. Studies suggest the microRNA regulation of these genes in several different contexts, providing a novel investigation approach for comprehending T2DM metabolism and revealing potential therapeutic targets. In the present study, we investigate in different animal models (human, rat, and mouse) miRNAs that target BDNF and IGF1 in skeletal muscle tissue with T2DM physiological conditions. Bioinformatics tools and databases were used to miRNA prediction, molecular homology, experimental validation of interactions, expression in the studied physiological condition, and network interaction. The findings showed three miRNAs candidates for IGF1(miR-29a, miR-29b, and miR-29c) and one for BDNF (miR-206). The experimental evaluations and the search for the expression in skeletal muscle from T2DM subjects confirmed the predicted interaction between miRNA-mRNA for miR-29b and miR-206 through human, rat, and mouse models. This interaction was reaffirmed in multiple network analyses. In conclusion, our results show the regulation relationship between miR-29b and miR-206 with the investigated genes, in several tissues, suggesting an inhibition pattern. Nevertheless, these data show a large number of possible interaction physiological processes, for future biotechnological prospects.

Resumo A leishmaniose visceral é uma doença tropical negligenciada. Foram avaliadas a distribuição espacial dos casos de leishmaniose visceral no estado de Alagoas. Todos os casos de LV, registrados pela secretaria de saúde, foram analisados e georreferenciados. Entre 2008 e 2017, 97,1% dos municípios apresentaram classificação esporádica de transmissão. Com a evolução temporal, a incidência de casos de leishmaniose visceral se concentrou na maioria dos municípios da microrregião de Santana do Ipanema-AL. A análise espaço-tempo, se considerada, pode promover o aprimoramento das ações de vigilância e controle da leishmaniose visceral.

Abstract Visceral leishmaniosis is a neglected tropical disease. We evaluated the spatial distribution of cases of visceral leishmaniosis in the state of Alagoas, Brazil. All cases of VL, registered by the health department, were analyzed and georeferenced. Results: Between 2008 and 2017, 97.1% of the municipalities presented sporadic classification of transmission. With temporal evolution, the incidence of cases of visceral leishmaniosis was concentrated in most municipalities in the microregion of Santana do Ipanema-AL. Space-time analysis, if considered, may promote the improvement of surveillance and control actions of visceral leishmaniosis.

Resumo Nematoides entomopatogênicos (NEP) das famílias Heterorhabditidae e Steinernematidae são amplamente utilizados no controle de insetos-pragas. No Brasil, os estudos relacionados a caracterização e identificação destes entomopatógenos são recentes e escassos. Nesse sentido, o objetivo deste estudo foi isolar NEP de amostras de solos coletadas em diferentes áreas no estado do Paraná, Brasil. Um isolado Heterorhabditis (UEL 08) detectado em amostra de solo em área de pastagem cultivada com braquiária, localizada em Londrina, Paraná, Brasil (23º34´311´´S, 050º58´298´´W), utilizando o método de “inseto-isca” com lagartas de Galleria mellonella. Para a identificação foram realizados estudos de morfometria e identificação molecular a partir da amplificação da região ITS. Algumas diferenças foram encontradas em termos de morfometria em comparação com a descrição original, entretanto, os dados moleculares demonstraram que a sequência obtida para o isolado UEL 08 é idêntica à sequência de referência de H. amazonensis (DQ665222), com a qual apresentou 100% de similaridade. Os estudos das características biológicas de H. amazonensis (UEL 08) revelaram que o isolado tem dois ciclos de vida distintos, um curto (216 h) e outro longo (288 h), sendo que ocorrem duas gerações em ambos os ciclos. O isolado UEL 08 H. amazonensis foi patogénico e virulento sobre os três hospedeiros avaliados. Notadamente, as larvas de G. mellonella e Alphitobius diaperinus foram consideradas mais susceptíveis do que os adultos do percevejo Dichelops (Diacereus) melacanthus, com percentagens de mortalidade de 100%, 85% e 46% de mortalidade, respectivamente. O ensaio de produção in vivo revelou um rendimento médio diário de 3,4 × 103 juvenis infectantes/g de larva hospedeira.

Abstract Entomopathogenic nematodes (EPNs) from Heterorhabditidae and Steinernematidae families are extensively used to control insect pests. In Brazil, however, relatively few studies have identified and characterized these entomopathogens. The objective of this study was to identify and characterize an EPN isolate obtained from soil samples collected in the state of Paraná, Brazil. An isolate (UEL 08) of Heterorhabditis was detected in a soil sample collected from a pasture area cultivated with Brachiaria grass in Londrina, state of Paraná, Brazil (23°34ʹ311ʹʹS, 050°58ʹ298ʹʹW), using the insect-baiting technique with Galleria mellonella larvae as hosts. The nematode was identified through morphometric studies and molecular analyses based on amplification of the rDNA ITS region. Although we identified certain morphometric differences compared with the original description, the molecular data indicated that the ITS sequence obtained for the UEL 08 isolate is identical to the reference sequence of H. amazonensis (DQ665222) and presented 100% similarity. Thus, the findings of our morphological and molecular studies confirmed that the isolated nematode is H. amazonensis, which is the first time this species has been registered in Paraná. Study of the biological characteristics of H. amazonensis (UEL 08) revealed that the isolate has two distinct life cycles – one short (216 h) and the other long (288 h) – and produces two generations in both cycles. We observed that H. amazonensis (UEL 8) was pathogenic and virulent to the three evaluated hosts, although with different virulence against these hosts. The larvae of G. mellonella and Alphitobius diaperinus were more susceptible than adult Dichelops (Diacereus) melacanthus, with 100%, 85%, and 46% mortality, respectively. Furthermore, an in vivo production assay revealed a mean daily yield of 3.4 × 103 infective juveniles/g host larvae.

Toads of the Rhinella genus have a pair of paratoid glands that store biological secretions of high toxicity and varied chemical composition, rich in biologically active compounds. The present work aimed to carry out the investigation of the metabolomic profile and evaluation of the biological potential of the secretion paratoid glands (PGS) from Rhinella granulosa. The paratoid secretion was collected in the Piauí state (Brazil), extracted with methanol and the extract was analyzed by ultra-performance liquid chromatography with quadrupole time-of-flight mass spectrometry. Fifty chemical constituents were identified. The extract showed cytotoxicity against tumor cell lines of the central nervous system (half maximal inhibitory concentration (IC50) = 1.9 µg mL-1) and prostate (IC50 = 1.6 µg mL-1), unsatisfactory antimicrobial potential (minimal inhibitory concentration (MIC) > 312 µg mL-1) and inhibited the enzyme acetylcholinesterase (IC50 = 5.119 mg mL-1). The results presented relevant information about the PGS and contributed to the understanding of the metabolomic and biological potential of R. granulosa.

ABSTRACT Nutritional management can be an efficient strategy to mitigate the effects of salinity on fruit production and quality. This study aimed to evaluate the production and quality of fruits of mini watermelon crop, Sugar Baby cultivar, grown in a protected environment, under salt stress and K+/Ca2+ ratios. The study was carried out in a greenhouse, using a randomized block design, 1 + 5 scheme, resulting in six treatments and four replications. Six nutrient solutions were employed, one using low-salinity water, with NaCl (0.5 dS m-1) and standard nutrient solution (S1, control treatment), and the others using salinized water at 5.0 dS m-1 (S2) and extra addition of K (S3 = 50% and S4 = 100%) and Ca (S5 = 50% and S6 = 100%). The production (fruit production, longitudinal and transverse diameter of fruit, rind thickness, pulp firmness) and postharvest quality (soluble solids, vitamin C, titratable acidity, and maturation index), in addition to the variables related to the color of the pulp (lightness, chroma index, and Hue angle) were evaluated. The addition of NaCl reduced fruit production, longitudinal, and transverse diameter of fruit, rind thickness, and vitamin C content. Among saline nutrient solutions, extra addition of 50% K (S3) attenuates the deleterious effects of salt stress on the production variables and vitamin C content. Under salt stress, extra addition of 50% K (S3) and 100% Ca (S6) intensifies the red color of the pulp.

RESUMO O manejo nutricional pode ser uma estratégia eficiente para mitigar os efeitos da salinidade na produção e qualidade de frutos. O objetivo deste trabalho foi avaliar a produção e qualidade de frutos de mini melancia, cultivar Sugar Baby, cultivado em ambiente protegido, sob estresse salino e relações K+/Ca2+. O estudo foi realizado em casa de vegetação, em delineamento de blocos casualizados, em esquema 1 + 5, resultando em seis tratamentos e quatro repetições. Foram aplicadas seis soluções nutritivas, uma utilizando água de baixa salinidade (0,5 dS m-1) e solução nutritiva padrão (S1, tratamento controle) e as demais utilizando água salinizada com NaCl, a 5,0 dS m-1 (S2) e adição extra de K (S3 = 50% e S4 = 100%) e Ca (S5 = 50% e S6 = 100%). Foram avaliadas as variáveis de produção (produção de frutos, diâmetro longitudinal do fruto, diâmetro transversal do fruto, espessura da casca, firmeza da polpa) e qualidade pós-colheita (sólidos solúveis, vitamina C, acidez titulável e índice de maturação), além das variáveis referentes a cor da polpa (luminosidade, croma e ângulo de Hue). A adição de NaCl reduziu a produção de frutos, diâmetro longitudinal dos frutos, diâmetro transversal dos frutos, espessura da casca e vitamina C. Entre as soluções nutritivas salinas, a adição extra de K a 50% (S3) atenua os efeitos deletérios do estresse salino nas variáveis de produção e o teor de vitamina C. Sob estresse salino, a adição extra de K em 50% (S3) e Ca em 100% (S6) intensifica a coloração vermelha da polpa.

Abstract This study aimed to analyze the phytochemical profile of essential oil obtained from the leaves of Coriandrum sativum L., and its antifungal activity against Candida spp. The research consisted of an in vitro study including collecting the vegetable product, analysis of its macronutrients, extraction, and chemical analysis of the essential oil, and assaying antifungal activity through minimum inhibitory concentration (MIC) and minimum fungicidal concentration (MFC), with growth inhibition kinetics, and the product’s effects on multi-species Candida biofilm. Nitrogen (47.08 g Kg-1), phosphorus (5.3 g Kg-1) and potassium (50.46 g Kg-1) levels were within the normal range. The major constituents were octanal, decanal, dec-(2E)-enal, and dodecanal. The MIC and MFC of the product evaluated against 11 tested Candida strains ranged from 31.25 to 250 μg/mL. There was inhibition of fungal growth during 24 hours of exposure at the 3 concentrations tested (250, 125, and 62.5 μg/mL). The concentration of 80 mg/mL promoted the greatest reduction in multispecies biofilm (70% reduction in biofilm). Coriandrum sativum L. essential oil extract is principally constituted of alcohols and aldehydes and presents fungicidal activity against Candida spp. in its in planktonic and biofilm forms.

Resumo Este trabalho teve como objetivo analisar o perfil fitoquímico do óleo essencial obtido das folhas de Coriandrum sativum L., e sua atividade antifúngica contra Candida spp. A pesquisa consistiu em um estudo in vitro incluindo a coleta do produto vegetal, análise de seus macronutrientes, extração e análise química do óleo essencial e ensaio da atividade antifúngica por meio da concentração inibitória mínima (CIM) e concentração fungicida mínima (MFC), com crescimento cinética de inibição e os efeitos do produto no biofilme de Candida multi-espécies. Os níveis de nitrogênio (47,08 g Kg-1), fósforo (5,3 g Kg-1) e potássio (50,46 g Kg-1) estavam dentro da normalidade. Os principais constituintes foram octanal, decanal, dec-(2E)-enal e dodecanal. A CIM e CFM do produto avaliado contra 11 cepas de Candida testadas variaram de 31,25 a 250 μg/mL. Houve inibição do crescimento fúngico durante 24 horas de exposição nas 3 concentrações testadas (250, 125 e 62,5 μg/mL). A concentração de 80 mg/mL promoveu a maior redução no biofilme multiespécies (redução de 70% no biofilme). O extrato do óleo essencial de Coriandrum sativum L. é constituído principalmente por álcoois e aldeídos e apresenta atividade fungicida contra Candida spp. em suas formas planctônicas e biofilme.

ABSTRACT A new species of Mahabalella B. Sutton & S. D. Patil was isolated from decaying leaves of an unidentified plant, during a survey in the Charles Darwin Ecological Refuge in Igarassu, Pernambuco, Brazil, in June 2018. The new species is morphologically distinguishable from the other Mahabalella species by the size of their conidia, conidiogenous cells and setae. In this study, the morphological descriptions, illustrations and cultural characteristics for Mahabalella pernambucana are provided, as well as a key to species of this genus. Total DNA from the isolate was extracted, and four genes were partially sequenced (nLSU, ITS cluster, TEF1 alpha, and beta tubulin). Based on the nLSU analysis, the phylogenetic position of the new species was determined within the Sordariomycetes, Ascomycota, as a sister genus to Neotracylla, in the Tracyllaceae family.

Abstract Heat treatment is used in the orange juice industry to neutralize the action of pathogenic microorganisms. However, it can reduce the nutritional value of the juice. Thus, our study assessed the cold plasma treatment as an alternative method against Escherichia coli and Candida albicans in 'Lima' orange juice. Both, plasma and heat treatments, reduced the amount of E. coli in the juice, inactivating 16.72 and 100%, respectively. Plasma did not inactivate C. albicans, but heat treatment inactivated 100%. Plasma and heat treatment increased Hue angle and luminosity (more yellowish juice). Plasma reduced vitamin C, carotenoids, and polyphenols content, while increased flavonoids. Heat treatment reduced the carotenoid content. However, neither heat nor plasma treatment altered the antioxidant activity. The plasma treatment reduced the intensity of color (chroma), the soluble solids content and the acidity ratio, total sugars, and the vitamin C content of juice compared to the heat-treated and control juices. Plasma-treated juice showed increased levels of yellow flavonoids, total phenolics and antioxidant activity until the 12th day of storage.

Resumo O tratamento térmico é utilizado na indústria de suco de laranja para neutralizar a ação de microrganismos patogênicos. No entanto, pode reduzir o valor nutricional do suco. Assim, o estudo avaliou o tratamento com plasma frio como método alternativo contra Escherichia coli e Candida albicans em suco de laranja 'Lima'. Ambos os tratamentos, plasma e calor, reduziram a quantidade de E. coli no suco, inativando 16.72 e 100%, respectivamente. O plasma não inativou C. albicans, mas o tratamento térmico inativou 100%. Plasma e tratamento térmico aumentaram o ângulo Hue e a luminosidade do suco (tom mais amarelo). O plasma reduziu o conteúdo de vitamina C, carotenóides e polifenóis, enquanto aumentou os flavonóides. O tratamento térmico reduziu o teor de carotenóides. No entanto, nem o calor nem o tratamento com plasma alteraram a atividade antioxidante. O tratamento com plasma reduziu a intensidade da cor (croma), o teor de sólidos solúveis e a razão de acidez, açúcares totais e o teor de vitamina C do suco em comparação com os sucos tratados termicamente e controle. O suco tratado com plasma apresentou maiores teores de flavonóides amarelos, fenólicos totais e atividade antioxidante até o 12º dia de armazenamento.

The production of conductive and organic devices from a 3D printer represents a promising strategy for several areas. In particular, the synthesis of polypyrrole-coated acrylonitrile butadiene styrene (ABS) composites can be considered an important step to produce conductive supports for 3D printing. Herein, it is reported the production of ABS samples through the additive manufacturing process (3D printing) accordingly to the Fused Deposition Modeling (FDM) method. The hydrophilic behavior was controlled by the surface treatment using air plasma for the following step of coating with polypyrrole (PPy) via an in situ polymerization, using two different oxidants: ferric chloride (FeCl3.6H2O) and ammonium persulfate (APS). The chemical, optical, surface, and electrical properties of these materials were characterized through Fourier Transform infrared spectroscopy (FTIR), contact angle measurements, cyclic voltammetry, Scanning Electron Microscopy (SEM), 4-probe electrical measurement, and mechanical tensile testing. The ABS/PPy (FeCl3) composite exhibited a low electrical contact resistance and better performance for applications that require electrodes with a good conductance level.

Abstract Pseudomonas aeruginosa is a non-lactose fermenting Gram-negative bacteria responsible for causing numerous nosocomial infections. The present research aimed to analyze the anti-Pseudomonas aeruginosa potential of 2-Chloro-N-(4-fluoro-3-nitrophenyl)acetamide (A8). The antibacterial potential of A8 was evaluated from the Minimum Inhibitory Concentration (MIC), Minimum Bactericidal Concentration (MBC) and Association using the checkerboard method. MIC and MBC values were 512 µg/mL for all P. aeruginosa strains evaluated, demonstrating predominantly bactericidal activity. Furthermore, when A8 was associated with the drug ceftriaxone, pharmacological additivity and indifference were evidenced. In this sense, the synthetic amide was interesting, since it demonstrates the potential to become a possible candidate for an antimicrobial drug.

Resumo Pseudomonas aeruginosa é uma bactéria Gram-negativa não fermentadora de lactose, responsável por causar inúmeras infecções nosocomiais. A presente pesquisa teve como objetivo analisar o potencial anti-Pseudomonas aeruginosa da molécula 2-Cloro-N-(4-fluoro-3-nitrofenil)acetamida (A8). O potencial antibacteriano do A8 foi avaliado a partir da Concentração Inibitória Mínima (CIM), Concentração Bactericida Mínima (CBM) e Associação pelo método de checkboard. Os valores de CIM e CBM foram de 512 µg/mL para todas as cepas de P. aeruginosa avaliadas, demonstrando atividade predominantemente bactericida. Além disso, quando o A8 foi associado ao fármaco ceftriaxona, evidenciou-se aditividade e indiferença farmacológica. Nesse sentido, a amida sintética se mostrou interessante, pois demonstra potencial para se tornar um possível candidato a fármaco antimicrobiano.

Abstract Parkia platycephala is the only species of the genus Parkia that is endemic to the brazilian Cerrado and the tree symbol of the state of Tocantins, but there are still few studies regarding its bioprospecting. In this study, we aimed to investigate the phytochemical composition, toxicity and bioactivities of the bark and flower of Parkia platycephala. Hot sequential extractions (Soxhlet) were performed using methanol and hydroethanolic solution (70%), after degreasing the sample (hexane). The presence of flavonoids, tannins, steroids and alkaloids was detected in the preliminary screening. Trilinolein, (Z)-9-octadecenamide, 3-O-methyl-d-glucose were detected by Gas Chromatography coupled to Mass Spectrometry (GC-MS). In the Liquid Chromatography with Diode Array Detector (LC-PDA) analysis, it was detected exclusively ferulic acid (bark) and ellagic acid (flower). The ethanolic extract of the bark (IC50=10.69 ± 0.35 µgmL-1) has an antioxidant potential (DPPH• radical) higher than that of the rutin standard (IC50=15.85 ± 0.08 µgmL-1). All extracts showed excellent anticholinesterase potential (Ellman), with emphasis on the ethanol extract of the flower (IC50 =5.34 ± 0.12 µgmL-1). Regarding toxicity (Artemia salina), the methanolic extract of the bark and the ethanolic extract of the flower presented high and moderate levels, respectively. Such results limit the concentrations of biological activities in this study, however, the antioxidant and anticholinesterase indices fall short of toxicity. The results demonstrated promising antioxidant and anticholinesterase activities of both the bark and the flower of Parkia platycephala.

Resumo A Parkia platycephala é a única espécie do gênero Parkia, endêmica do Cerrado brasileiro e a árvore símbolo do estado do Tocantins, porém ainda com pouco estudos com relação a sua bioprospecção. Neste estudo, objetivamos realizar a caracterização fitoquímica, avaliar a toxicidade e as atividades antioxidante e anticolinesterásica dos extratos da casca e da flor da Parkia platycephala. Foram realizadas extrações sequenciais a quente (Soxhlet), utilizando hexano, metanol e solução hidroetanólica (70%). A presença de flavonoides, taninos, esteroides e alcaloides foi detectada na triagem preliminar. A análise por Cromatografia Gasosa acoplada a Espectrometria de Massa (GC-MS) indicou a presença de variados compostos, tais como trilinoleina, (Z)-9-octadecenamida, 3-O-methyl-d-glucose e methylsulfinyl(methylthio)-methane. Na análise por Cromatografia Líquida com Detector de Arranjo de Diodo (LC-PDA), destacam-se o ácido elágico, presente apenas nos extratos da flor, e o ácido ferúlico, presente apenas nos extratos da casca. Os extratos da casca predominaram quanto ao potencial antioxidante (radical DPPH•), sendo que o extrato etanólico (IC50=10,69 ± 0,35 µgmL-1) superou o padrão rutina (IC50=15,85 ± 0,08 µgmL-1). Todos os extratos apresentaram excelentes potenciais anticolinesterásicos (teste de Ellman), com ênfase no extrato etanólico da flor (IC50=5,34 ± 0,12 µgmL-1). Em relação à toxicidade (Artemia salina), o extrato metanólico da casca e o extrato etanólico da flor, apresentaram níveis elevado e moderado, respectivamente. Tais resultados limitam as concentrações das atividades biológicas deste estudo, entretanto, os índices antioxidante e anticolinesterásico ficam aquém da toxicidade. Os resultados mostraram as promissoras atividades antioxidante e anticolinesterásica da casca e flor da Parkia platycephala.

ABSTRACT: Leptospirosis is a disease that causes economic and social impact, as it affects wild and domestic animals and humans. There may be peculiarities in the epidemiology of this disease in the Caatinga biome, Brazil, where the environment is adverse and the etiologic agent, Leptospira spp., requires alternative transmission routes. Considering that in bovine leptospirosis the genital carrier is constantly neglected and the lack of reports on the role of bulls in the epidemiology of the bovine genital leptospirosis (BGL) syndrome, mainly in semiarid conditions such as Caatinga biome, this study aimed to investigate bulls maintained in Caatinga biome conditions as genital carriers of leptospires. Urinary tract (urine, bladder, and kidney) and reproductive tract (vas deferens, cauda epididymis, and vesicular gland) samples were collected from 42 slaughtered bulls. Microscopic agglutination test (MAT), polymerase chain reaction (PCR), and microbiological isolation were included as diagnostic methods. Anti-Leptospira spp. antibodies were found in 17 (40.48%) animals, while 26 animals (61.90%) had at least one organ or urine with leptospiral DNA, and 10 animals (23.81%) were positive at bacteriological culture. Sequenced samples targeting the LipL32 gene showed 99% similarity with Leptospira borgpetersenii. Molecular analysis of samples from the vas deferens and cauda epididymis is recommended for the diagnosis of genital leptospirosis in bulls and, if it is impossible to collect these tissues, semen can be used. In conclusion, this study provides important information relating to bulls from the Caatinga biome, Brazil, as carriers of genital leptospirosis. The results indicate that, even under adverse environmental conditions, leptospires may survive and propagate, mainly due to the characteristic of genital carriers for the sexually spreading of adapted Leptospira species without influence by external variables. Thus, prevention and control strategies for bovine leptospirosis need to include actions aimed at the genital carrier.

RESUMO: A leptospirose é uma doença que causa impacto econômico e social, pois afeta animais silvestres, domésticos e humanos. É possível que existam peculiaridades na epidemiologia desta doença no bioma Caatinga, Brasil, onde o ambiente é adverso e o agente etiológico, Leptospira spp., requer vias alternativas de transmissão. Considerando que na leptospirose bovina o portador genital é constantemente negligenciado e a falta de relatos sobre o papel dos touros na epidemiologia da síndrome da leptospirose genital bovina (BGL), principalmente em condições semiáridas como no bioma Caatinga, este estudo teve como objetivo investigar touros mantidos em condições do bioma Caatinga como portadores genitais de leptospiras. Amostras do trato urinário (urina, bexiga e rim) e do trato reprodutivo (ducto deferente, cauda do epidídimo e glândula vesicular) foram coletadas de 42 touros abatidos. Teste de soroaglutinação microscópica (SAM), reação em cadeia da polimerase (PCR) e isolamento microbiológico foram incluídos como métodos de diagnóstico. Anticorpos anti-Leptospira spp. foram encontrados em 17 (40,48%) animais, enquanto 26 animais (61,90%) apresentaram pelo menos um órgão ou urina com DNA leptospírico, e 10 animais (23,81%) foram positivos na cultura bacteriológica. As amostras sequenciadas a partir do gene LipL32 apresentaram 99% de similaridade com Leptospira borgpetersenii. Recomenda-se a análise molecular de amostras de ducto deferente e cauda do epidídimo para o diagnóstico de leptospirose genital em touros e, na impossibilidade da coleta desses tecidos, o sêmen pode ser utilizado. Em conclusão, este estudo fornece informações importantes sobre touros do bioma Caatinga, Brasil, como portadores de leptospirose genital. Os resultados indicam que, mesmo em condições ambientais adversas, as leptospiras podem sobreviver e se propagar, principalmente devido à característica de os portadores genitais disseminarem sexualmente espécies adaptadas de Leptospira sem influência de variáveis externas. Assim, as estratégias de prevenção e controle da leptospirose bovina precisam incluir ações voltadas para o portador genital.

ABSTRACT Perineal hernia results from weakening and/or the inability of the pelvic diaphragm to support the rectal wall, promoting caudal displacement of abdominal or pelvic organs in the perineum. Unneutered male dogs aged between 7 and 10 years are the most affected, being rare in females. The main clinical signs are unilateral or bilateral swelling of the perineal region, associated or not with tenesmus, dysuria, urinary and/or fecal incontinence, anuria and pain during defecation. The diagnosis is based on clinical history, anamnesis, physical examination, and complementary tests such as radiography and ultrasound. Clinical treatment can be done in some cases, but surgical intervention is required to resolve the problem. The present work aims to report the case of a patient treated at the FullPet Veterinary Clinic in Guarulhos, São Paulo, Brazil, a 7-year-old male Yorkshire canine, unneutered, weighing 4.5 kg. The same was submitted to surgery by the traditional method of suture with nylon thread and enteropexy. When there was recurrence, surgical reintervention was necessary to correct the hernia using a polypropylene mesh.

RESUMEN La hernia perineal resulta del debilitamiento y/o incapacidad del diafragma pélvico para sostener la pared rectal, lo que promueve el desplazamiento caudal de los órganos abdominales o pélvicos en el perineo. Los perros machos no castrados de entre 7 y 10 años son los más afectados, siendo raro en las hembras. Los principales signos clínicos son tumefacción unilateral o bilateral de la región perineal, asociada o no a tenesmo, disuria, incontinencia urinaria y/o fecal, anuria y dolor durante la defecación. El diagnóstico se basa en la historia clínica, la anamnesis, el examen físico y pruebas complementarias como la radiografía simple y la ecografía. El tratamiento clínico se puede hacer en algunos casos, pero se requiere una intervención quirúrgica para resolver el problema. El presente trabajo tiene como objetivo relatar el caso de un paciente atendido en la Clínica Veterinaria FullPet de Guarulhos, São Paulo, Brasil, un canino Yorkshire macho de 7 años, sin castrar, con un peso de 4,5 kg. Fue intervenido quirúrgicamente por el método tradicional de sutura con hilo de nylon y enteropexia. Cuando hubo recidiva, fue necesaria la reintervención quirúrgica para corregir la hernia mediante una malla de polipropileno.

The interest of researchers in understanding the stage of development of specific areas of knowledge is a way to understand their scientific maturity and their capacity for consolidation. This “popularization” of the knowledge of researchers working in a specific area of knowledge also allows a greater ability to recognize where science is produced and which actors participate in this effort from a series of evaluation parameters, indicative of the productivity of researchers through the number of articles published and the consistency of production and its relationship with the impact of this in the scientific community through citations of articles. Chemometrics, a science that relates measurements obtained in chemical systems or processes through the application of statistical and mathematical methods, an area created 50 years ago, still does not have a study evaluating the evolution of the area in qualitative and quantitative terms. In this sense, this article aims to present data from the profile of Brazilian scientific production in Chemometrics, as well as the training of human resources, in its scientometric aspects to allow an understanding of the dynamics and contemporary reality of the area. In terms of national scientific production is growing proportionally faster than the rest of the world, placing Brazil among the leading producers of knowledge in the area. Researchers in Chemometrics working in Brazil not only publish a significant number of articles, but these publications have a large number of citations, in addition to the training of human resources spread throughout Brazil, distributed among most graduate courses in chemistry, but not restricted to this area. Finally, the area of Chemometrics, which is about fifty years old, is mature, with numbers of publications and training of human resources, but it is noted that a plateau has not been reached in these two numbers, because the critical mass of researchers working in the area is still increasing.

A procedure using disposable pipette tips adapted for solid-phase extraction, known as DPX SPE, was used as an alternative way for the chromium determination in wastewater samples. DPX-SPE consists of a device which allows a dynamic contact between the sample and the solid-phase. A residue obtained from the processing of coffee beans, denominate coffee silverskin, was used as a new adsorbent. Characterization techniques revealed properties of lignocellulosic materials with potential application for chromium adsorption. The parameters including adsorbent mass (25.00 mg), elution solvent (0.1 mol L-1 HCl, 200 μL) and pH (2.0) were optimized. Thus, 1.00 mL of deionized water for conditioning, 4.00 mL of sample, 2 extraction cycles and 1 elution cycle were the employed conditions. Enrichment factor of 12, limit of detection of 6.00 µg L-1 and relative standard deviation (RSD) 1.3% (n = 3) were obtained. The method proved to be fast, cheap, environmentally friendly, and simple, providing good recoveries (104-113%), and it was satisfactorily applied in real samples.
