RESUMO A elevada variabilidade do HIV-1, bem como, a ausência de mecanismos eficientes de reparo durante os estágios da replicação viral, contribuem para a rápida emergência de cepas de HIV-1 resistentes aos antirretrovirais. A pressão seletiva exercida pelas drogas, leva à fixação de mutações capazes de conferir graus variados de resistência. A presença dessas mutações constitui um dos fatores mais importantes na falha da resposta terapêutica aos medicamentos. Assim, é de fundamental importância compreender os padrões de resistência e os mecanismos a eles associados, possibilitando a escolha de um esquema terapêutico apropriado que considere a frequência e outras características das mutações. Utilizando a Rede Neural Artificial Paraconsistente, assentada na Lógica Paraconsistente Anotada Et que tem a capacidade de mensurar incertezas e inconsistências, obtivemos níveis de concordância acima de 90% quando comparado à metodologia proposta com a metodologia atual empregada para classificar os subtipos do HIV-1. Os resultados obtidos demonstram que a Rede Neural Artificial Paraconsistente pode servir como ferramenta promissora de análise.

ABSTRACT The high variability of HIV-1 as well as the lack of efficient repair mechanisms during the stages of viral replication, contribute to the rapid emergence of HIV-1 strains resistant to antiretroviral drugs. The selective pressure exerted by the drug leads to fixation of mutations capable of imparting varying degrees of resistance. The presence of these mutations is one of the most important factors in the failure of therapeutic response to medications. Thus, it is of critical to understand the resistance patterns and mechanisms associated with them, allowing the choice of an appropriate therapeutic scheme, which considers the frequency, and other characteristics of mutations. Utilizing Paraconsistents Artificial Neural Networks, seated in Paraconsistent Annotated Logic Et which has the capability of measuring uncertainties and inconsistencies, we have achieved levels of agreement above 90% when compared to the methodology proposed with the current methodology used to classify HIV-1 subtypes. The results demonstrate that Paraconsistents Artificial Neural Networks can serve as a promising tool of analysis.

Background Several studies suggest that metabolic disorders and nutrition imbalance during prenatal life may induce adaptations that program cardiovascular diseases and hypertension. We have previously shown that moderate zinc restriction during prenatal life, lactation and/or growth leads to the development of hypertension and renal dysfunction in adulthood. Objectives To evaluate the presence of early cardiovascular alterations in rats exposed to a moderate zinc deficient diet during prenatal life and lactation, and to determine whether there are differences between males and females. Material and Methods Female Wistar rats received low zinc diet or control diet from the beginning of pregnancy up to weaning. Four experimental groups were established at birth: males and females born from low-diet mothers, and males and females born from control-diet mothers. Male and female offspring were sacrificed at 6 and 21 days of life to evaluate body weight, heart weight, cardiovascular morphometric parameters and nitric oxide synthase activity in the cardiovascular system and cardiac oxidative status. Results The insufficient zinc intake during prenatal life and lacta-tion induced a remodeling process of the cardiomyocyte which was different in males and females, increased cardiac oxidative stress, produced a hypotrophic remodeling of the thoracic aorta and reduced nitric oxide synthase activity in the cardiovascular system. Conclusions This study shows that zinc deficiency induces cardiovascular abnormalities in early stages of development, which are different in males and females that may contribute to programming of diseases in adulthood.

Introducción Numerosos estudios sugieren que trastornos metabólicos y desequilibrios nutricionales durante la vida intrauterina pueden inducir adaptaciones que programen enfermedades cardiovasculares e hipertensión arterial. En trabajos previos mostramos que la restricción moderada de cinc durante la vida fetal, la lactancia y/o el crecimiento conduce al desarrollo de hipertensión arterial y disfunción renal en la adultez. Objetivos Evaluar la presencia de alteraciones cardiovasculares tempranas en ratas sometidas a una deficiencia moderada de cinc durante la vida fetal y la lactancia y si existen diferencias respecto del sexo. Material y métodos Ratas Wistar hembras recibieron durante la preñez hasta el destete una dieta control o baja en cinc. En el momento del nacimiento se conformaron cuatro grupos experimentales: machos y hembras nacidos de madres bajas y machos y hembras nacidos de madres controles. A los 6 y a los 21 días de vida se sacrificaron y se determinaron el peso corporal, el peso del corazón, parámetros morfométricos cardiovasculares, la actividad de la óxido nítrico sintasa en el sistema cardiovascular y el estado oxidativo cardíaco. Resultados El aporte insuficiente de cinc durante la vida fetal y la lactancia indujo un proceso de remodelación del cardiomiocito, diferente en machos que en hembras, un aumento del estrés oxidativo cardíaco, una remodelación hipotrófica de la aorta torácica y una disminución de la actividad de la óxido nítrico sintasa en el sistema cardiovascular. Conclusiones Este trabajo demuestra que la deficiencia de cinc induce alteraciones cardiovasculares, distintas en machos que en hembras, tempranas en el desarrollo, que podrían contribuir a la programación de enfermedades en la vida adulta.

O objetivo do presente estudo foi avaliar a carga viral no soro de pacientes cronicamente infectados pelo vírus da Hepatite B (HBV) e investigar a distribuição de genótipos HBV na cidade de São Paulo. PCR quantitativo do HBV e genotipagem ganharam importância para a previsão de progressão da doença, empregada para avaliar a infectividade, para tratamento e acompanhamento e para detectar o aparecimento de resistência aos anti-retrovirais. Vinte e nove pacientes brasileiros com suspeita de hepatite B crônica foram estudados, utilizando PCR em tempo real para a determinação da carga viral e seqüenciamento direto para determinação do genótipo. A sorologia revelou que 22 estavam, de fato, cronicamente infectados pelo HBV. O HBV-DNA foi positivo em 68% das amostras (15/22). Em sete casos, HBV-DNA foi indetectável por PCR quantitativo. A análise filogenética mostra que onze pacientes foram infectados com hepatite B genótipo A, dois com genótipo F e dois com genótipo D. Desta forma, o genótipo A foi o mais prevalente em nosso estudo.

The objective of the present study was to evaluate the serum viral load in chronically infected Hepatitis B virus (HBV) patients and to investigate the distribution of HBV genotypes in São Paulo city. Quantitative HBV-DNA assays and HBV genotyping have gained importance for predicting HBV disease progression, have been employed for assessing infectivity, for treatment monitoring and for detecting the emergence of drug resistance. Twenty-nine Brazilian patients with suspected chronic hepatitis B were studied, using real time PCR for viral load determination and direct DNA sequencing for the genotyping. The serology revealed chronic HBV infection in 22 samples. The HBV-DNA was positive in 68% samples (15/22). The phylogenetic analysis disclosed that eleven patients were infected with HBV genotype A, two with genotype F and two with genotype D. Thus, the genotype A was the most prevalent in our study.
